House flies, Musca domestica L., are important pests of dairy operations worldwide, with the ability to adapt wide range of environmental conditions. There are a number of insecticides used for their management, but development of resistance is a serious problem. Insecticide mixtures could enhance the toxicity of insecticides in resistant insect pests, thus resulting as a potential resistance management tool. The toxicity of bifenthrin, cypermethrin, deltamethrin, chlorpyrifos, profenofos, emamectin benzoate and fipronil were assessed separately, and in mixtures against house flies. A field-collected population was significantly resistant to all the insecticides under investigation when compared with a laboratory susceptible strain. Most of the insecticide mixtures like one pyrethroid with other compounds evaluated under two conditions (1∶1-“A” and LC50: LC50-“B”) significantly increased the toxicity of pyrethroids in the field population. Under both conditions, the combination indices of pyrethroids with other compounds, in most of the cases, were significantly below 1, suggesting synergism. The enzyme inhibitors, PBO and DEF, when used in combination with insecticides against the resistant population, toxicities of bifenthrin, cypermethrin, deltamethrin and emamectin were significantly increased, suggesting esterase and monooxygenase based resistance mechanism. The toxicities of bifenthrin, cypermethrin and deltamethrin in the resistant population of house flies could be enhanced by the combination with chlorpyrifos, profenofos, emamectin and fipronil. The findings of the present study might have practical significance for resistance management in house flies.  相似文献

Helicoverpa armigera (Hübner) is one of the most destructive pests of several field and vegetable crops, with indiscriminate use of insecticides contributing to multiple instances of resistance. In the present study we assessed whether H. armigera had developed resistance to Bt cotton and compared the results with several conventional insecticides. Furthermore, the genetics of resistance was also investigated to determine the inheritance to Cry1Ac resistance. To investigate the development of resistance to Bt cotton, and selected foliar insecticides, H. armigera populations were sampled in 2010 and 2011 in several cotton production regions in Pakistan. The resistance ratios (RR) for Cry1Ac, chlorpyrifos, profenofos, cypermethrin, spinosad, indoxacarb, abamectin and deltamethrin were 580-fold, 320-, 1110-, 1950-, 200-, 380, 690, and 40-fold, respectively, compared with the laboratory susceptible (Lab-PK) population. Selection of the field collected population with Cry1Ac in 2010 for five generations increased RR to 5440-fold. The selection also increased RR for deltamethrin, chlorpyrifos, profenofos, cypermethrin, spinosad, indoxacarb, abamectin to 125-folds, 650-, 2840-, 9830-, 370-, 3090-, 1330-fold. The estimated LC50s for reciprocal crosses were 105 µg/ml (Cry1Ac-SEL female × Lab-PK male) and 81 g µg/ml (Lab-PK female × Cry1Ac-SEL male) suggesting that the resistance to Cry1Ac was autosomal; the degree of dominance (DLC) was 0.60 and 0.57 respectively. Mixing of enzyme inhibitors significantly decreased resistance to Cry1Ac suggesting that the resistance to Cry1Ac and other insecticides tested in the present study was primarily metabolic. Resistance to Cry1Ac was probably due to a single but unstable factor suggesting that crop rotation with non-Bt cotton or other crops could reduce the selection pressure for H. armigera and improve the sustainability of Bt cotton.  相似文献

Two field populations of Spodoptera exigua (Hübner) (Lepidoptera: Noctuidae) from Dera Ghazi Khan (D. G. Khan) and Multan, Pakistan, were tested for resistance to the 10 most commonly used insecticides in Pakistan by using a standard leaf disc bioassay on the F1 progeny. For comparison, a susceptible strain was generated from the Multan strain, which displayed lower LC50 values for most of the insecticides, by either mass rearing without exposure to insecticides or single-pair crosses against selected insecticides. The single-pair crosses generated a more susceptible strain than mass rearing. The D. G. Khan field strain was highly resistant to cypermethrin, profenofos, spinosad, abamectin, and chlorpyrifos and moderately resistant to deltamethrin, indoxacarb, and methoxyfenozide compared with susceptible lab strain. The Multan strain was highly resistant to profenofos and indoxacarb. Both field populations were susceptible to emamectin benzoate and lufenuron. Rotating these two insecticides with others that show very low, low, or moderate levels of resistance and have different modes of action may be useful for the effective management of this pest.  相似文献

The field population of Spodoptera litura from Huizhou, Guangdong Province, China was evaluated for resistance to 21 insecticides, including conventional and new chemistry insecticides. Extreme levels of resistance were observed to metaflumizone and emamectin benzoate with resistance factors of 234.1 and 183.3, respectively. Resistance to abamectin was also high (perhaps extremely high) and over 71.9-fold. The Huizhou population of S. litura possessed high resistance to deltamethrin (96.5-fold) and moderate resistance to beta cyfluthrin and lambda cyhalothrin but remained susceptible to bifenthrin. Moderate resistance to chlorantraniliprole (22.3-fold), endosulfan (22.2-fold), tebufenozide (10.7-fold) and thiodicarb (14.3-fold), and low-level resistance to fipronil, indoxacarb and spinosad were also reported in this population. This field population remained susceptible to acetamiprid, chlorfenapyr, chlorfluazuron, hexaflumuron, chlorpyrifos, pyridalyl and spinetoram. The stabilities of resistance to metaflumizone, emamectin benzoate, deltamethrin, chlorantraniliprole and endosulfan were evaluated, the resistance level decreased when the insecticide stress was removed, suggesting stop of the application of insecticides with high level resistance could be implemented into the resistance management. Because S. litura from Huizhou developed resistance to multiple insecticides, integration of different control practices, especially the rotation of insecticides with biocontrol agents, should be performed in the management of this pest. The results suggested the suspension of the application of insecticide to which S. litura had developed high level of resistance in order to mitigate the resistance status, and the use of the insecticides to which this pest remained sensitive, including spinetoram, pyridalyl, indoxacarb, hexaflumuron, chlorfluazuron, chlorfenapyr and bifenthrin, could be incorporated into the alternating application for resistance management.  相似文献

The success of chemical control depends on toxicity of insecticides against insect pests. Camponotus sericeus is an important urban pest with the ability to cause substantial damage to wooden structures, but there is a lack of information on toxicity of insecticides against C. sericeus. To determine the insecticide toxicity, workers of C. sericeus were exposed to 17 insecticides from different classes: carbamate (methomyl, bendiocarb), organophosphate (chlorpyrifos, profenofos, temephos), pyrethroid (bifenthrin, deltamethrin, permethrin), neonicotinoid (acetamiprid, imidacloprid, thiamethoxam), avermectin (abamectin, emamectin), pyrrole (chlorfenapyr), phenylpyrazole (fipronil), and spinosyn (spinosad and spinetoram), via residual bioassay method. The LC50 ranged from: 0.15 to 0.20 µg/vial for carbamates, 0.09 to 0.27 µg/vial for organophosphates, 0.09 to 0.44 µg/vial for pyrethroids, 0.02 to 0.67 µg/vial for neonicotinoids, 0.54 to 0.82 µg/vial for avermectins, 0.78 µg/vial for pyrrole, 0.62 µg/vial for phenylpyrazole, and 1.96 to 2.05 µg/vial for spinosyns. Overall, acetamiprid was the most toxic one among the tested insecticides followed by permethrin, temephos, profenofos, bendiocarb and methomyl, while spinosad and spinetoram were the least toxic insecticides. Considering the potential toxicity of different insecticides against C. sericeus, future studies could investigate the practical application of these insecticides in order to design an effective management plan.  相似文献

[目的]研究6种杀虫剂对南美番茄潜叶蛾卵、幼虫和成虫的毒力及其在温室番茄上的防治效果,为南美番茄潜叶蛾防治提供高效杀虫剂和施药技术。[方法]采用浸渍法和药膜法测定了6种杀虫剂对南美番茄潜叶蛾卵、幼虫和成虫的毒力,田间调查毒力较高杀虫剂对温室番茄上南美番茄潜叶蛾防效。[结果] 6种杀虫剂中的乙基多杀菌素、甲氨基阿维菌素苯甲酸盐和阿维菌素对南美番茄潜叶蛾卵有毒力作用,致死中浓度(LC50)分别为1.415、13.588和23.194 mg·L-1。6种杀虫剂对南美番茄潜叶蛾幼虫的LC50分别为:阿维菌素0.026 mg·L-1、四唑虫酰胺0.052 mg·L-1、甲氨基阿维菌素苯甲酸盐0.057 mg·L-1、乙基多杀菌素0.072 mg·L-1、氯虫苯甲酰胺0.484 mg·L-1和呋虫胺2.039 mg·L-1。对于南美番茄潜叶蛾成虫,24 h时,仅甲氨基阿维菌素苯甲酸盐和四唑虫酰胺对成虫有较高毒力;72 h时,6种杀虫剂对成虫的LC50分别为:甲氨基阿维菌素苯甲酸盐0.390 mg·L-1、乙基多杀菌素1.646 mg·L-1、四唑虫酰胺2.630 mg·L-1、呋虫胺5.577 mg·L-1、阿维菌素22.502 mg·L-1和氯虫苯甲酰胺39.636 mg·L-1。在成虫盛发期第4天施药,阿维菌素、四唑虫酰胺、甲氨基阿维菌素苯甲酸盐、乙基多杀菌素在南美番茄潜叶蛾危害严重的温室番茄上防效达80%以上。[结论]阿维菌素、四唑虫酰胺、甲氨基阿维菌素苯甲酸盐、乙基多杀菌素、氯虫苯甲酰胺和呋虫胺对南美番茄潜叶蛾卵、幼虫或成虫有较高毒力,其中阿维菌素、四唑虫酰胺、甲氨基阿维菌素苯甲酸盐和乙基多杀菌素田间防效较好。  相似文献

American bolloworm, Helicoverpa armigera Hubner (Noctuidae: Lepidoptera) is considered as a major pest of various crops all over the world. It is mainly controlled by indiscriminate use of synthetic insecticides in the world due to which this pest developed resistance to most of the available insecticides. Therefore, in the current study, the efficacy of virulent strain of HaNPV (0.5 × 109 PIB/ml) alone and in combination with recommended doses of spintoram (20 g/100 L of water) and emamectin benzoate (200 ml/100 L of water) was tested in field. The combination of HaNPV with spintoram and emamectin benzoate 100% reduced the larval population as compared to emamectin benzoate and HaNPV alone. This suggested that the combination of spintoram and emamectin benzoate with HaNPV could be used in field to manage the infestation of H. armigera.  相似文献

Nucleopolyhedrosis viruses can be utilized for effective management of agriculture pests. Their efficacy can be increased if they are mixed with certain insecticides. In the current study, HaNPV was mixed with two insecticides: spinetoram and emamectin benzoate in various combinations and applied to larvae of H. armigera in laboratory conditions. There were a total of 15 combinations of HaNPV with each of the two insecticides in addition to five doses of HaNPV and three doses of insecticides alone. The synergistic and antagonistic effects of combinations were explored. The results revealed that there was synergistic effect of HaNPV @ 0.5 × 109 PIB/ml × Spinetoram @ 40, 20, 10 ml/100 L of water. In case of emamectin benzoate, synergistic effects were recorded at 1 × 109 PIB/ml HaNPV × emamectin benzoate @ 100 ml/100 L of water. However, 0.5 × 109 PIB/ml HaNPV has synergistic effects with all three doses of emamectin benzoate. The results suggested that HaNPV can be used in combination with spinetoram and emamectin benzoate for the management of resistant population of H. armigera.  相似文献

Background

Diarrhea is an important cause of childhood mortality in developing countries like Pakistan because of unhygienic conditions, lack of awareness, and unwise use of preventive measures. Mechanical transmission of diarrheal pathogens by house flies, Musca domestica, is believed as the most effective route of diarrhea transmission. Although the use of insecticides as a preventive measure is common worldwide for the management of house flies, success of the measure could be compromised by the prevailing environmental temperature since it significantly affects toxicity of insecticides and thus their efficacy. Peaks of the house fly density and diarrheal cases are usually coincided and season specific, yet little is known about the season specific use of insecticides.

Methodology/Principal Findings

To determine the temperature-toxicity relationship in house flies, the effect of post-bioassays temperature (range, 20–34°C) on the toxicity of seven insecticides from organophosphate (chlorpyrifos, profenofos), pyrethroid (cypermethrin, deltamethrin) and new chemical (emamectin benzoate, fipronil, spinosad) classes was evaluated by using a feeding bioassay method. From 20–34°C, the toxicities of chlorpyrifos, profenofos, emamectin and fipronil increased 2.10, 2.93, 2.40 and 3.82 fold (i.e. positive temperature coefficient), respectively. Whereas, the toxicities of cypermethrin, deltamethrin and spinosad decreased 2.21, 2.42 and 3.16 fold (i.e. negative temperature coefficient), respectively.

Conclusion/Significance

These findings suggest that for the reduction in diarrheal cases, house flies should be controlled with insecticides according to the prevailing environmental temperature. Insecticides with a positive temperature coefficient may serve as potential candidates in controlling house flies and diarrhea epidemics in hot season and vice versa.  相似文献

西花蓟马田间种群对常用杀虫剂的抗性现状及防治对策   总被引:5,自引:0,他引:5  
【目的】西花蓟马Frankliniella occidentalis在中国是一种严重危害温室蔬菜的入侵害虫。本研究旨在了解该害虫在中国的抗药性现状,为防治该害虫提供理论支持。【方法】采用Munger cell法测定了北京,山东寿光和青岛以及云南晋宁和呈贡等5个地区西花蓟马田间种群对多杀菌素、毒死蜱、阿维菌素、甲维盐、氟氯氰菊酯、溴虫腈、灭多威、吡虫啉和啶虫脒9种杀虫剂的抗药性水平,同时利用这些田间种群测定了多功能氧化酶抑制剂胡椒基丁醚(PBO)、谷胱甘肽S 转移酶抑制剂顺丁烯二酸二乙酯(DEM)和羧酸酯酶抑制剂三丁基三硫磷酸酯(DEF)对多杀菌素、吡虫啉和甲维盐的增效作用。【结果】生物测定结果表明,北京、晋宁及呈贡种群分别对多杀菌素产生了34.45, 47.45和64.45倍的高水平抗性;晋宁种群对灭多威和甲维盐分别产生了16.58和11.03倍的中等水平抗性;呈贡种群对甲维盐、啶虫脒、吡虫啉、阿维菌素、溴虫腈分别产生了24.17, 21.69, 20.05, 16.45和10.31的中等水平抗性;青岛种群对啶虫脒和吡虫啉产生了17.70和12.49倍的中等水平抗性;寿光种群没有对任何杀虫剂产生高等或中等水平抗性。增效剂生物测定结果表明,对于吡虫啉和甲维盐,多功能氧化酶抑制剂PBO在所有田间种群上均有显著的增效作用。谷胱甘肽S-转移酶抑制剂DEM在呈贡、寿光和青岛种群中对吡虫啉存在显著增效作用;在北京、呈贡和寿光种群中,DEM对甲维盐存在显著增效作用。羧酸酯酶抑制剂DEF在呈贡、晋宁和青岛种群中对吡虫啉存在显著增效作用;在北京、呈贡和晋宁种群中,DEF对吡虫啉存在显著增效作用。但所有增效剂在各田间种群中对多杀菌素均无显著增效作用。【结论】结果提示:在使用多杀菌素防治西花蓟马时,应与其他杀虫剂轮换使用;此外,可通过添加酶抑制剂来增强甲维盐和吡虫啉对西花蓟马的防效。  相似文献

A residual contact vial plus water (RCVpW) bioassay method, in which water was supplemented to minimize control mortality, was established to monitor insecticide resistance in field populations of the melon thrips, Thrips palmi. In the RCVpW, median lethal doses (LD50) of six insecticides commonly used in T. palmi control, were determined at 8 h post-treatment, using a susceptible RDA strain according to the RCVpW protocol. Diagnostic doses for on-site resistance monitoring of the six insecticides, which were determined as doses two-fold higher than required to achieve LD90 in the RDA strain, were in the range of 0.299 to 164.3 μg?1 cm2. Insecticide resistance levels in five field populations of T. palmi were evaluated to test the applicability of RCVpW in monitoring the pest. Although the RDA strain exhibited 100% mortality to diagnostic doses, field populations showed a reduced mortality in response to all test insecticides, indicating different degrees of resistance. In particular, all test field populations exhibited a significantly low mortality in response to spinosad, suggesting a wide distribution of spinosad resistance. Synergistic bioassay revealed that cytochrome P450-mediated metabolic factor is involved in spinosad resistance in the Korean population. Interestingly, an apparently reduced mortality to emamectin benzoate and chlofenapyr was observed in some field populations, perhaps suggesting uneven distribution of resistance to these insecticides in field populations. Our study showed that the RCVpW protocol can be employed both as an on-site resistance monitoring method for major thrip species, and in the selection of appropriate insecticides for their control.  相似文献

Southern root-knot nematode (Meloidogyne incognita) is an obligate, sedentary endoparasite of more than 3000 plant species, that causes heavy economic losses and limit the development of protected agriculture of China. As a biological pesticide, emamectin benzoate has effectively prevented lepidopteran pests; however, its efficacy to control M. incognita remains unknown. The purpose of the present study was to test soil application of emamectin benzoate for management of M. incognita in laboratory, greenhouse and field trials. Laboratory results showed that emamectin benzoate exhibited high toxicity to M. incognita, with LC50 and LC90 values 3.59 and 18.20 mg L-1, respectively. In greenhouse tests, emamectin benzoate soil application offered good efficacy against M. incognita while maintaining excellent plant growth. In field trials, emamectin benzoate provided control efficacy against M. incognita and resulted in increased tomato yields. Compared with the untreated control, there was a 36.5% to 81.3% yield increase obtained from all treatments and the highest yield was received from the highest rate of emamectin benzoate. The results confirmed that emamectin benzoate has enormous potential for the control of M. incognita in tomato production in China.  相似文献

The role of insecticidal application and host plant resistance in managing Spodoptera exigua has been well documented, but the effect of different host plants, on which the pest cycles its population in the field, has seldom been investigated. Therefore, we have studied the vulnerability of S. exigua against commonly used insecticides (cypermethrin, chlorpyrifos, lufenuron, and emamectin benzoate) with different mode of actions when it switches its generations from natal to auxiliary hosts and vice versa. Different field populations being established on different host plants including castor, cauliflower, cotton, okra, and spinach were collected and reared in the laboratory before insecticidal bioassays. The role of larval diet and host plant switching on their response to tolerate applied insecticides was studied using leaf‐dip bioassay methods. Host switching demonstrated a significant role in altering the vulnerability of S. exigua populations to tested insecticides. Spodoptera exigua sourced from castor, when switched host to okra and spinach, exhibited 50% higher mortality when treated with emamectin benzoate. This trend in mortality was consistent upon complete host switch cycle (natal—auxiliary—natal host). However, the highest increase (92%) in vulnerability was recorded when the larvae were shifted to spinach from cotton. In general, chlorpyrifos and lufenuron had highest efficacies in terms of larval mortality. The findings of present studies provide insights to a better understanding the behavior of polyphagous pests and the role of different host plants in altering the susceptibility of these pests against applied insecticides. Ultimately the results warrant that due consideration should be given to cropping patterns and time of host switching by pest population during planning and executing chemical control.  相似文献

The toxicity of the most commonly used insecticides of organochlorine, organophosphate, pyrethroid, and carbamate groups were investigated against Spodoptera litura (F.) (Lepidoptera: Noctuidae) populations collected for three consecutive years (2004-2006). For a chlorocyclodiene and pyrethroids tested, the resistance ratios compared with Lab-PK were in the range of 10- to 92-fold for endosulfan, 5- to 111-fold for cypermethrin, 2- to 98-fold for deltamethrin, and 7- to 86-fold for beta-cyfluthrin. For organophosphates and carbamates, resistance ratios were in the range of 3- to 169-fold for profenofos, 18- to 421-fold for chlorpyrifos, 3- to 160-fold for quinalphos, 6- to 126-fold for phoxim, 7- to 463-fold for triazophos, and 10- to 389-fold for methomyl and 16- to 200-fold for thiodicarb. Resistance ratios were generally low to medium for deltamethrin and beta-cyfluthrin and high to very high for endosulfan, cypermethrin, profenofos, chlorpyrifos, quinalphos, phoxim, triazophos, methomyl, or thiodicarb. Pairwise comparisons of the log LC50 values of insecticides tested for all the populations showed correlations among several insecticides, suggesting a cross-resistance mechanism. Integration of timely judgment of pest problem, delimiting growing of alternate crops such as arum, rotation of insecticides with new chemicals, and insect growth regulators in relation to integrated pest management could help in manageable control of this important pest.  相似文献

Grapholita molesta is one kind of serious fruit-boring pests in the world, which caused huge economic losses to the fruit industry including pear and apple. The heavy usage of synthetic insecticides resulted in different degrees of resistance of G. molesta to some commonly used insecticides. However, the tolerance mechanisms of G. molesta to insecticides are still unclear. Glutathione S-transferases (GSTs) belong to the superfamily of detoxifying enzymes and mainly play important roles in detoxification metabolism and insecticide susceptibility of insects. In this study, we first evaluated the toxicity of three insecticides including emamectin benzoate, chlorantraniliprole and lambda-cyhalothrin on G. molesta at different developmental stages. Synergism study showed that the susceptibility of G. molesta larvae to these three insecticides obviously increased when the enzyme activity of GST was inhibited by diethyl maleate compared with the controls. Next, we screened out six GmGST genes which are upregulated after insecticides treatment from a total of 21 GmGST genes and analysed their sequence characteristics and expression profiles. RNAi and bioassay results further revealed that the mortality of 5th instar larvae significantly increased after exposure to LC30 of emamectin benzoate, chlorantraniliprole and lambda-cyhalothrin when silencing of GmGSTo2, GmGSTs1 and GmGSTz1 compared with the controls. In conclusion, this study indicated that GmGSTo2, GmGSTs1 and GmGSTz1 play important roles in insecticides susceptibility of G. molesta to three commonly used insecticides, which is significance for its future integrated management.  相似文献

甜菜夜蛾Spodoptera exigua(Hübner)云南晋宁、上海奉贤和江苏六合种群对甲氨基阿维菌素苯甲酸盐抗性为45~437倍,对高效氯氰菊酯抗性为211~555倍,对其它药剂抗性不明显。这3个田间种群3龄幼虫多功能氧化酶、谷胱甘肽S-转移酶和酯酶的活力分别为室内敏感种群的2.7~8.4倍、1.9~8.6倍和1.6~5.7倍。多功能氧化酶抑制剂PBO和酯酶抑制剂DEF对甲氨基阿维菌素苯甲酸盐的增效比为1.2~4.3和1.3~7.7;PBO和DEF对高效氯氰菊酯的增效比为1.8~58和3.6~245;谷胱甘肽S-转移酶抑制剂DEM对这2种药剂均无增效作用。上述结果表明,解毒代谢增强可能是甜菜夜蛾田间种群对甲氨基阿维菌素苯甲酸盐和高效氯氰菊酯的重要抗性机理,与酯酶和多功能氧化酶活性升高有关,与谷胱甘肽S-转移酶活性升高无关。本文的研究结果还表明,对于代谢抗性机理复杂的多抗性田间种群,根据不同解毒酶抑制剂对药剂的增效作用判断不同解毒代谢酶在抗性形成中的作用更加可靠。  相似文献

Abstract:  To understand the risk of resistance and the possible mechanisms of resistance to abamectin in B-type Bemisia tabaci (Gennadius) better, a resistant strain of B. tabaci was selected in the laboratory and cross-resistance pattern and resistance mechanisms to abamectin were investigated. The NJ-Abm strain of B. tabaci was derived from a field population (NJ) collected in Nanjing, China in 2002 with 18 generations of selection with abamectin in the laboratory. Compared with the unselected NJ strain, the selected NJ-Abm strain developed 14.5-fold resistance to abamectin and showed significant cross-resistance to emamectin benzoate (4.4-fold) and imidacloprid (3.4-fold), but no cross-resistance to fipronil. The oxidase inhibitor piperonyl butoxide (PBO) and glutathione S -transferase inhibitor diethyl maleate (DEM) produced significant synergism on abamectin in the NJ-Abm strain (with synergistic ratios of 3.9- and 4.1-fold respectively); however, the esterase inhibitor triphenyl phosphate (TPP) did not act synergistically with abamectin. Biochemical analysis confirmed that P450 monooxygenase activity and glutathione S -transferase activity of the NJ-Abm strain were elevated to 2.1- and 2.0-fold, respectively, compared with that of the NJ strain. This indicated that enhanced metabolism mediated by P450 monooxygenase and glutathione S -transferase is likely to be involved in abamectin resistance and cross-resistance to imidacloprid and emamectin benzoate in the NJ-Abm strain.  相似文献

The evolution of insecticide resistance represents a global constraint to agricultural production. Because of the extreme genetic diversity found in insects and the large numbers of genes involved in insecticide detoxification, better tools are needed to quickly identify and validate the involvement of putative resistance genes for improved monitoring, management, and countering of field-evolved insecticide resistance. The avermectins, emamectin benzoate (EB) and abamectin are relatively new pesticides with reduced environmental risk that target a wide number of insect pests, including the beet armyworm, Spodoptera exigua, an important global pest of many crops. Unfortunately, field resistance to avermectins recently evolved in the beet armyworm, threatening the sustainable use of this class of insecticides. Here, we report a high-quality chromosome-level assembly of the beet armyworm genome and use bulked segregant analysis (BSA) to identify the locus of avermectin resistance, which mapped on 15–16 Mbp of chromosome 17. Knockout of the CYP9A186 gene that maps within this region by CRISPR/Cas9 gene editing fully restored EB susceptibility, implicating this gene in avermectin resistance. Heterologous expression and in vitro functional assays further confirm that a natural substitution (F116V) found in the substrate recognition site 1 (SRS1) of the CYP9A186 protein results in enhanced metabolism of EB and abamectin. Hence, the combined approach of coupling gene editing with BSA allows for the rapid identification of metabolic resistance genes responsible for insecticide resistance, which is critical for effective monitoring and adaptive management of insecticide resistance.  相似文献

Abstract:  To investigate fluctuation in susceptibility to insecticides in the field, natural populations of Helicoverpa armigera were collected from the same field in the region of Multan, Pakistan in 2002 and 2003. The populations were examined against pyrethroids (viz. cypermethrin, esfenvalerate and fenpropathrin) and new insecticides (viz. spinosad, abamectin and indoxacarb). In 2002, the resistance ratio (RR) of cypermethrin and esfenvalerate was significantly higher than fenpropahrin compared with susceptible population. The susceptibility to cypermethrin, esfenvalerate and fenpropathrin increased significantly in 2003; however, the RR for cypermethrin was about half the RR (38) of esfenvalerate (101) and fenpropathrin (89). The toxicity of spinosad, abamectin and indoxacarb was identical in both years. In the field experiments, abamectin was more effective than other compounds tested whereas fenpropathrin, cypermethrin and esfenvalerate had similar toxicity. These results might have important implications in resistance management and suggest that the rotational use of spinosad, abamectin and indoxacarb could help to avoid the development of multiple resistant in H. armigera .  相似文献

This study aimed at evaluating the toxicity of some insecticides (abamectin and deltamethrin) on the lethal time (LT50) and midgut of foragers honeybee workers of Apis mellifera jemenatica were studied under laboratory conditions. The bees were provided with water, food, natural protein and sugar solution with insecticide (concentration: 2.50 ppm deltamethrin and 0.1 ppm abamectin). The control group was not treated with any kind of insecticides. The mortality was assessed at 1, 2, 4, 6, 12, 24, 48, and 72 hour (h) after insecticides treatment and period to calculate the value of lethal time (LT50). But the samples the histology study of midgut collected after 24 h were conducted by Scanning Electron Microscope. The results showed the effects of insecticides on the current results show that abamectin has an adverse effect on honeybees, there is a clear impact on the lethal time (LT50) was the abamectin faster in the death of honeybee workers compared to deltamethrin. Where have reached to abamectin (LT50 = 21.026) h, deltamethrin (LT50 = 72.011) h. However, abamectin also effects on cytotoxic midgut cells that may cause digestive disorders in the midgut, epithelial tissue is formed during morphological alterations when digestive cells die. The extends into the internal cavity, and at the top, there is epithelial cell striated border that has many holes and curves, abamectin seems to have crushed the layers of muscle. Through the current results can say abamectin most toxicity on honeybees colony health and vitality, especially foragers honeybee workers.  相似文献
