This investigation is aimed to study an impact of habitat degradation on proximate composition and amino acid (AAs) profile of Catla catla, Labeo rohita and Cirrhinus mrigala collected from polluted, non-polluted area (upstream) and a commercial fish farm. The amino acid profile was estimated by the amino acid analyzer. C. catla collected from the polluted environment had highest lipid, protein and ash contents (12.04?±?0.01, 13.45?±?0.01 and 0.93?±?0.03%, respectively). The high protein content (14.73?±?0.01 and 14.12?±?0. 01%) was recorded in C. catla procured from non-polluted (upstream) wild habitat of River Chenab and controlled commercial fish farm. Farmed fish species showed comparatively higher moisture contents followed by upstream and polluted area fishes. C. mrigala showed significant differences in amino acid and proximate composition collected from a polluted site of the river Chenab. C. catla collected from non-polluted site of the river showed an excellent nutrient profile, followed by L. rohita (wild and farmed) and C. mrigala (polluted area), respectively. All fishes from the polluted areas of the River Chenab indicated a significant decrease in the concentration of some AAs when compared to farmed and wild (upstream) major carps. Omitting of some important AAs was also observed in the meat of fish harvested from polluted habitat of this river. C. mrigala and L. rohita exhibited a significant increase in the concentration of some of non-essential amino acids such as cysteine in their meat. The results indicated that wild fish (upstream) and farmed fish species had highest protein contents and amino acid profile and hence appeared to be the best for human consumption. The proximate composition and AAs profiles of fish harvested from the polluted area of the river clearly indicated that efforts shall be made for the restoration of habitat to continue the requirement of high quality fish meat at a low cost to the human population.  相似文献

Owing to white meat production Labeo rohita have vast economic importance, but its population has been reduced drastically in River Chenab due to pollution. Atomic absorption spectrophotometry showed a merciless toxicity level of Cd, Cu, Mn, Zn, Pb, Cr, Sn and Hg. Comet assay results indicated significant (p?<?.05) DNA fragmentation in Labeo rohita as 42.21?±?2.06%, 31.26?±?2.41% and 21.84?±?2.21% DNA in comet tail, tail moment as 17.71?±?1.79, 10.30?±?1.78 and 7.81?±?1.56, olive moment as 13.58?±?1.306, 8.10?±?1.04 and 5.88?±?0.06, respectively, from three different polluted sites on the river. Micronucleus assay showed similar findings of single micronucleus induction (MN) as 50.00?±?6.30‰, double MN 14.40?±?2.56‰, while nuclear abnormalities (NA) were found as 150.00?±?2.92‰. These higher frequencies of MN induction and NA were found to be the cause of reduction of 96% of the population of this fish species in an experimental area of the River Chenab. This fish species has been found near extinction through the length of the river Chenab and few specimens in rainy seasons if restored by flood, may die in sugarcane mill season. Due to sweeping extinction Labeo rohita showed the highest sensitivity for pollution and could be used as bioindicator and DNA fragmentation in this column feeder fish species as a biomarker of the pollution load in freshwater bodies.  相似文献

Various metabolites exist in the medicinal plants have lot of potential to cure various diseases and disorders. Plants such as, Vetiveria zizanioides, Trichosanthes cucumerina, and Mollugo cerviana were collected from Western Ghats, Tamilnadu, India. Phytochemicals were extracted from these plants using various organic solvents and tested against Gram-positive and Gram-negative bacteria. The phytochemicals such as, carbohydrate, alkaloids, steroids, saponins, flavonoids and tannin were detected from these medicinal plants. Among the extracts, methanol showed potent activity and this solvent was used to extract polyherbal medicinal plants. Methanol extract of V. zizanioides was found to be highly active against E. coli (27 ± 2 mm), P. mirabilis (19 ± 3 mm) and B. subtilis (18 ± 2 mm). Ethyl acetate extract showed high activity against E. coli (24 ± 2 mm), P. mirabilis (22 ± 3 mm) and B. subtilis (20 ± 1 mm). These three plants were taken at 1:1:1 ratio and extracted with methanol at 1:10 ratio and synergistic activity was tested against bacterial pathogens. Synergistic activity of polyherbal extract was analyzed. The extracted crude herbal medicine was found to be effective against Staphylococcus aureus, E. coli, Enterbacter sp., Pseudomonas aeruginosa, Bacillus subtilis and Proteus mirabilis. The zone of inhibition was 33 ± 3 mm, 17 ± 2 mm, 22 ± 2 mm, 40 ± 2 mm, 33 ± 1 mm and 38 ± 2 mm zone of inhibition against E. coli, S. aureus, P. aeruginosa, P. mirabilis, B. subtilis and Enterobacter sp. Polyherbal extract was found to be highly effective against P. mirabilis and Enterobacter sp. MIC values of polyherbal extract ranged from 29 ± 2.5 µg/ml to 34 ± 2.5 µg/ml. MIC value was found to be less against P. mirabilis and was high against S. aureus. Antioxidant property varied between 49 ± 3% and 95.3 ± 2%. At 20 µg/ml antioxidant activity was reported as 49 ± 3% and it was increased at higher concentrations of polyherbal extract. Two cell lines (HeLa and MCF cell lines) were selected to analyze cytotoxic activity of polyherbal extract. The methanol extract of polyherbal fraction showed cytotoxicity against these two cell lines. The LC50 value was 467 ± 2.9 µg/ml against HeLa cell line and >800 µg/ml against MCF-7 cell lines. The polyherbal extract showed antibacterial, antioxidant and anticancer activities.  相似文献

The increasing prevalence of antibiotic resistant bacteria is a significant healthcare crisis with substantial socioeconomic impact on global community. The development of new antibiotics is both costly and time-consuming prompting the exploration of alternative solutions such as nanotechnology which represents opportunities for targeted drug delivery and reduced MIC. However, concerns have arisen regarding genotoxic effects of nanoparticles on human health necessitating an evaluation of nanoparticle induced DNA damage.This study aimed to investigate the antibacterial potential of already prepared, characterized chitosan nanoparticles loaded with carvacrol and their potential synergism with Topoisomerase II inhibitors against S. aureus, E. coli and S. typhi using agar well diffusion, microdilution and checkerboard method. Genotoxicity was assessed through comet assay.Results showed that both alone and drug combinations of varying concentrations exhibited greater zones of inhibition at higher concentrations. Carvacrol nanoparticles combined with ciprofloxacin and doxorubicin significantly reduced MIC compared to the drugs used alone. The MIC50 values for ciprofloxacin were 35.8 µg/ml, 48.74 µg/ml, 35.57 µg/ml while doxorubicin showed MIC50 values of 20.79 µg/ml, 34.35 µg/ml, 25.32 µg/ml against S. aureus, E. coli and S. typhi respectively. The FICI of ciprofloxacin and doxorubicin with carvacrol nanoparticles found ≤ 0.5 Such as 0.44, 0.44,0.48 for ciprofloxacin and 0.45, 0.45, 0.46 for doxorubicin against S. aureus, E. coli and S. typhi respectively revealed the synergistic effect. The analysis of comet assay output images showed alteration of DNA at high concentrations.Our results suggested that carvacrol nanoparticles in combination with Topoisomerase inhibitors may prevent and control the emergence of resistant bacteria with reduced dose.  相似文献

In the present study the alkaline comet assay and the cytokinesis-block micronucleus cytome (CBMN Cyt) assay were used to evaluate the baseline frequency of cytogenetic damage in peripheral blood lymphocytes (PBLs) of 50 healthy children from the general population in Croatia (age, 11.62 ± 1.81 years). Mean values of tail length, tail intensity and tail moment, as comet assay parameters, were 12.92 ± 0.10, 0.73 ± 0.06 and 0.08 ± 0.01, respectively. The mean frequency of micronuclei (MN) for all subjects was 2.32 ± 0.28 per 1000 bi-nucleated cells, while the mean frequency of nucleoplasmic bridges (NPBs) was 1.72 ± 0.24 and of nuclear buds (NBUDs) 1.44 ± 0.19. The mean nuclear division index (NDI) was 1.70 ± 0.05. When comet-assay parameters were considered, higher mean values for all three were found for the female population. According to the Mann–Whitney U test applied on the results of the comet assay, the only statistically significant difference between the male and female populations was found for tail length. Similar to the results obtained by the comet assay, girls showed higher mean values of all three measured parameters of the CBMN Cyt assay. This difference was statistically significant for total number of NPBs only. In the case of the NDI, a higher mean value was also obtained in girls, but this difference was not statistically significant. The results obtained present background data that could be considered as normal values for healthy children living in urban areas, and can later on serve as baseline values for further toxicological monitoring. Additionally, the usefulness of both techniques in measuring cytogenetic damage during bio-monitoring of children is confirmed.  相似文献

This study quantified the prevalence of holdfast aggregation (fusion of holdfasts) for the kelp Ecklonia radiata on subtidal reefs in southwestern Australia, and tested whether morphology, age, attachment or drag were different between kelps growing alone (solitary) or in aggregates. Wave-sheltered in-shore reefs consistently had fewer aggregates than wave-exposed off-shore reefs (15–20% versus 20–30%). On average, individual thalli from aggregates were longer (97.8 cm ± 2.2 S.E. versus 88.0 cm ± 2.0 S.E.) and had smaller holdfasts (32.9 g fresh wt ± 1.7 S.E. versus 45.8 g fresh wt ± 2.9 S.E.) than solitary thalli, whereas there were no significant differences in other morphological characters, including total biomass (805.1 g fresh wt ± 38.7 S.E. versus 831.5 g fresh wt ± 38.5 S.E.), stipe length (7.93 cm ± 0.47 S.E. versus 7.65 cm ± 0.40 S.E.) and stipe diameter (12.6 mm ± 0.23 S.E. versus 13.0 mm ± 0.25 S.E.). There was no difference in age between solitary (2.7–3.0 years) and aggregated (2.4–2.8 years) individuals. While the attachment force of whole aggregates (256.5 N ± 21.6 S.E.) was found to be significantly larger than attachment force for solitary individuals (162.5 N ± 12.9 S.E.), attachment areas were also larger for aggregates (90.7 cm2 ± 5.40 S.E. versus 64.3 cm2 ± 5.54 S.E.) and consequently there were no differences in attachment strength between aggregates (2.92 N cm−2 ± 0.26 S.E.) and solitary thalli (2.71 N cm−2 ± 0.22 S.E.). Aggregates had significantly smaller (17%) roughness factors (equivalent to drag coefficients) than solitary individuals and a negative relationship (r = −0.68) between roughness factors and biomass suggested that this was related to the scope for compaction and rearrangement of the thalli. Further, there was no relationship between roughness factors of solitary individuals and the aggregates they produced when combined, suggesting that roughness factors are not additive or multiplicative. The spatial distribution of holdfast aggregates, the morphological differences between solitary and aggregated individuals as well as their attachment and drag characteristics were all consistent with aggregation reducing the rate of fatal kelp dislodgment.  相似文献

In order to assess the level of ecological stress caused by the pollution from local disturbances in a stretch of the Garonne River, France, we applied the Abundance-Biomass Comparison (ABC) index, using fish assemblages. Data were collected in a 10-year span (1992–2002) in a reference site and in two pollution-exposed sites. The ABC index mean value in the reference site (S1) was 0.03 ± 0.002 (95% Confidence Interval – CI); for the polluted sites (S2 and S3), the values were −0.09 ± 0.002 (95% CI) and −0.12 ± 0.002 (95% CI), respectively. The ABC index showed that, besides flow variations, both downstream sites are statistically different (p < 0.05) from the reference site, but all three seem to be under moderate stress. Furthermore, we related our ABC scores to water quality and flow regime variables in the reference site and one of the polluted sites by means of a cluster analysis. The results showed that, in the reference site, the ABC scores are closely related to the flow regime, while in the polluted site, downstream a urban area, ABC is related to water quality variables such as phosphates and total phosphorous. We argue that ecological indicators can help decisions on environmental damage liability.  相似文献

Many water hyacinth infestations in South Africa are the symptom of eutrophication, and as a result, biological control of this weed is variable. This study examined the effects of herbivory by the mirid, Eccritotarsus catarinensis, on water hyacinth grown at high, medium and low nitrogen (N) and phosphorus (P) nutrient concentrations. Water nutrient concentration appears to be the overriding factor affecting plant growth parameters of water hyacinth plants—at high nutrient concentrations, leaf and daughter plant production were more than double than at low nutrient concentrations, while stem length was twice as great at high nutrient concentrations compared to low concentrations. Chlorophyll content was also twice as high at high nutrient concentrations than low concentrations. Conversely, flower production at high nutrient concentrations was less than half that at low concentrations. Herbivory by E. catarinensis did not have as great an effect on water hyacinth vigour as nutrient concentration did, although it significantly reduced the production of daughter plants by 23 ± 9%, the length of the second petiole by 13 ± 5%, and chlorophyll content of water hyacinth leaves by 15 ± 6%. In terms of insect numbers, mirids performed better on plants grown under medium nutrient conditions (99 ± 28 S.E.), compared to high nutrient concentrations (52 ± 27 S.E.), and low nutrient concentrations (25 ± 30 S.E.). Thus, these results suggest that the fastest and most significant reduction in water hyacinth proliferation would be reached by lowering the water nutrient concentrations, and herbivory by E. catarinensis alone is not sufficient to reduce all aspects of water hyacinth vigour, especially at very high nutrient concentrations.  相似文献

Microbial resistance and other emerging health risk problems related to the side effects of synthetic drugs are the major factors that result in the research regarding natural products. Fruits, leaves, seeds, and oils-based phyto-constituents are the most important source of pharmaceutical products. Plant extract chemistry depends largely on species, plant components, solvent utilized, and extraction technique. This study was aimed to compare the ethanolic extracts of a mangrove plant, i.e., Avicennia marina (1E: Lower half of A. marina‘s pneumatophores, 2E: A. marina‘s leaves, 3E: Upper half of A. marina‘s pneumatophores, and 4E: A. marina‘s shoots), with non-mangrove plants, i.e., Phragmites australis (5E: P. australis‘s shoot), and Moringa oleifera (6E: M. oleifera‘s leaves) for their antimicrobial activities, total phenolic contents, antioxidant activity, and cytotoxicity potential. The antimicrobial activity assays were performed on gram-positive bacteria (i.e., Bacillus subtilis and Staphylococcus aureus), gram-negative bacteria (i.e., Escherichia coli, and Pseudomonas aeruginosa), and fungi (i.e., Aspergillus niger, Candida albicans, and Rhizopus spp.). We estimated antioxidant activity by TAC, DPPH, and FRAP assays, and the cytotoxicity was evaluated by MTT assay. The results of antimicrobial activities revealed that B. subtilis was the most sensitive to the tested plant extracts compared to S. aureus, while it only showed sensitivity to 6E and Imipenem. 5E and 6E showed statistically similar results against P. aeruginosa as compared to Ceftazidime. E. coli was the most resistant bacteria against tested plant extracts. Among the tested plant extracts, maximum inhibition activity was observed by 6E against A. niger (22 ± 0.57 mm), which was statistically similar to the response of 6E against C. albicans and 3E against Rhizopus spp. 2E did not show any activity against tested fungi. We found that 6E (208.54 ± 1.92 mg g?1) contains maximum phenolic contents followed by 1E (159.42 ± 3.22 mg g?1), 5E (131.08 ± 3.10 mg g?1), 4E (i.e., 72.41 ± 2.96 mg g?1), 3E (67.41 ± 1.68 mg g?1), and 2E (48.72 ± 1.71 mg g?1). The results depict a significant positive correlation between the phenolic contents and the antioxidant activities. As a result, phenolic content may be a natural antioxidant source.  相似文献

Dietary fibres may have prebiotic effects mediated by promotion of beneficial bacteria. This study explores the possibility that soluble plant fibre may also improve health by inhibiting epithelial adhesion and translocation by pathogenic bacteria. We have focussed on soluble non-starch polysaccharide (NSP) from plantain bananas (Musa spp.) which previous studies showed to be particularly effective at blocking Escherichia coli epithelial adherence. In vitro and ex vivo studies assessed the ability of plantain NSP to inhibit epithelial cell adhesion and invasion of various bacterial pathogens, and to inhibit their translocation through microfold (M)-cells and human Peyer′s patches mounted in Ussing chambers. Plantain NSP showed dose-related inhibition of epithelial adhesion and M-cell translocation by a range of pathogens. At 5 mg/ml, a concentration readily achievable in the gut lumen, plantain NSP inhibited adhesion to Caco2 cells by Salmonella Typhimurium (85.0±8.2%, P<.01), Shigella sonnei (46.6±29.3%, P<.01), enterotoxigenic E.coli (56.1±23.7%, P<.05) and Clostridium difficile (67.6±12.3%, P<.001), but did not inhibit adhesion by enteropathogenic E.coli. Plantain NSP also inhibited invasion of Caco2 cells by S. Typhimurium (80.2 ± 9.7%) and Sh. sonnei (46.7±13.4%); P<.01. Plantain NSP, 5 mg/ml, also inhibited translocation of S. Typhimurium and Sh. sonnei across M-cells by 73.3±5.2% and 46.4±7.7% respectively (P<.05). Similarly, S. Typhimurium translocation across Peyer′s patches was reduced 65.9±8.1% by plantain NSP (P<.01). Soluble plantain fibre can block epithelial adhesion and M-cell translocation of intestinal pathogens. This represents an important novel mechanism by which soluble dietary fibres can promote intestinal health and prevent infective diarrhoea.  相似文献

This study was designed to determine the effects of the combined addition of different levels of certain sugars (trehalose, sucrose and raffinose) and antioxidants (vitamin E, C and taurine), in Tris-egg yolk extender on frozen-thawed ram semen parameters. Semen samples were collected from five healthy, mature and fertile Iranian Afshari rams, twice a week for 8 weeks. Selected samples were pooled and diluted with a Tris-egg yolk extender containing different levels of sugars and antioxidants. In Experiment 1, different levels of trehalose (0, 50 and 100 mM) were tested with different levels of taurine (0, 25 and 50 mM), vitamin E and C (0, 1 and 2 mM). In Experiment 2, different levels of sucrose (0, 60 and 80 mM) were tested with different levels of taurine (0, 25 and 50 mM), vitamin E and C (0, 1 and 2 mM). In Experiment 3, different levels of raffinose (0, 5, 10 mM) were tested with different levels of taurine (0, 25 and 50 mM), and vitamin E and C (0, 1 and 2 mM). In Experiment 4, the selected extenders of experiments 1, 2 and 3 were compared statistically with control (no selected sugar and antioxidant) extender. The results of experiments 1, 2 and 3 revealed that the highest frozen–thawed sperm parameters were recorded for the selected extenders containing 100 mM trehalose +2 mM vitamin E (T100E2), 60 mM sucrose + 2 mM vitamin E (S60E2) and 10 mM raffinose + 2 mM vitamin E (R10E2), respectively. The results of experiment 4 revealed that the post-thaw sperm total motility in T100E2 (62.41 ± 2.41%), S60E2 (59.52 ± 1.91%) and R10E2 (58.33 ± 2.00%) was higher than that of the control extender (46.00 ± 1.79%; P ≤ 0.05). Similarly, the progressive sperm motility in T100E2 (57.18 ± 1.96%), S60E2 (57.49 ± 1.94%) and R10E2 (55.03 ± 2.99%) was also higher than that of the control extender (41.20 ± 1.70%; P ≤ 0.05). Post-thaw sperm viability in selected extenders of T100E2 (65.20 ± 2.67%), S60E2 (62.00 ± 2.07%) and R10E2 (61.80 ± 2.46%) was higher than that of control extender (51.00 ± 1.88%; P ≤ 0.05). In conclusion, the addition of 100 mM trehalose, 60 mM sucrose and 10 mM raffinose combined with 2 mM vitamin E in Tris-egg yolk extender significantly improved frozen-thawed ram semen parameters.  相似文献

Biofilm producing clinical bacterial isolates were isolated from periodontal and dental caries samples and identified as, Lactobacillus acidophilus, Streptococcus sanguis, S. salivarius, S. mutansand Staphylococcus aureus. Among the identified bacterial species, S. aureus and S. mutansshowed strong biofilm producing capacity. The other isolated bacteria, Streptococcus sanguis, S. salivarius showed moderate biofilm formation. These pathogens were subjected for the production of extracellular polysaccharides (EPS) in nutrient broth medium and the strain S. aureus synthesized more amounts of EPS (610 ± 11.2 µg/ml) than S. sanguis (480 ± 5.8 µg/ml).EPS production was found to be less in S. salivarius (52 ± 3.8 µg/ml).The solvent extract of A. sativum bulb showed the phytochemicals such as, carbohydrate, total protein, alkaloids, saponins, flavonoids, tannins and sterioids. The solvent extract of A. sativum bulb showed wide ranges of activity against the selected dental pathogens. The difference in antibacterial activity of the solvent extract revealed differences in solubility of phytochemicals in organic solvents. Ethanol extract was highly active againstS. aureus (25 ± 2 mm). The Minimum Inhibitory Concentration (MIC) of crude garlic bulb varied widely and this clearly showed that bacteria exhibits different level of susceptibility to secondary metabolites. MIC value ranged between 20 ± 2 mg/ml and 120 ± 6 mg/ml and Minimum Bactericidal Concentration (MBC) value ranged from 60 ± 5 mg/l to 215 ± 7 mg/ml. To conclude, A. sativum bulb can be effectively used to treat periodontal and dental caries infections.  相似文献

Most Trichloroethylene (TCE) biodegradation reports refer to methanogenic conditions, however, in this work, enhanced sulfidogenesis and TCE biodegradation were achieved in an upflow anaerobic sludge blanket (UASB) reactor in which a completely sulfidogenic sludge, from hydrothermal vents sediments, was developed. The work was divided in three stages, (i) sludge development and sulfate reducing activity (SRA) evaluation, (ii) TCE biodegradation and (iii) SRA evaluation after TCE biodegradation. For (i) SR was 98 ± 0.1%, 84% as sulfide (H2S, 1200 ± 28 mg/L), sulfate reducing activity (SRA) was 188 ± 50 mg COD H2S/g VSS*d. For (ii) The reactor reached 74% of TCE removal, concentrations of vinyl chloride of 16 ± 0.3 μM (5% of the TCE added) and ethene 202 ± 81 μM (67% of the TCE added), SRA of 161 ± 7 mg COD H2S/g VSS*d, 68% of sulfide (H2S) production and 93% of COD removal. For (iii) SRA was of 248 ± 22 mg COD H2S/g VSS*d demonstrating no adverse effects due to TCE.Among the genera of the microorganisms identified in the sludge during TCE biodegradation were: Dehalobacter, Desulfotomaculum, Sulfospirillum, Desulfitobacterium, Desulfovibrio and Clostridium. To the best of our knowledge, this is the first report using a sulfidogenic UASB reactor to biodegrade TCE. The overall conclusions of this work are that the reactor is efficient on both, sulfate and TCE biodegradation and it could be used to decontaminate wastewater containing organic solvents and relatively high concentrations of sulfate.  相似文献

Saccharum spontaneum is a wasteland weed consists of 45.10 ± 0.35% cellulose and 22.75 ± 0.28% of hemicellulose on dry solid (DS) basis. Aqueous ammonia delignified S. spontaneum yielded total reducing sugars, 53.91 ± 0.44 g/L (539.10 ± 0.55 mg/g of substrate) with a hydrolytic efficiency of 77.85 ± 0.45%. The enzymes required for hydrolysis were prepared from culture supernatants of Aspergillus oryzae MTCC 1846. A maximum of 0.85 ± 0.07 IU/mL of filter paperase (FPase), 1.25 ± 0.04 IU/mL of carboxy methyl cellulase (CMCase) and 55.56 ± 0.52 IU/mL of xylanase activity was obtained after 7 days of incubation at 28 ± 0.5 °C using delignified S. spontaneum as carbon source under submerged fermentation conditions. Enzymatic hydrolysate of S. spontaneum was then tested for ethanol production under batch and repeated batch production system using “in-situ” entrapped Saccharomyces cerevisiae VS3 cells in S. spontaneum stalks (1 cm × 1 cm) size. Immobilization was confirmed by the scanning electron microscopy (SEM). Batch fermentation of VS3 free cells and immobilized cells showed ethanol production, 19.45 ± 0.55 g/L (yield, 0.410 ± 0.010 g/g) and 21.66 ± 0.62 g/L (yield, 0.434 ± 0.021 g/g), respectively. Using Ringer solution as extender, the present study examined the protective effect of dimethyl sulphoxide (Me2SO; 8-12%, v/v) on the cryopreservation of giant grouper (Epinephelus lanceolatus) sperm. The cryopreserved sperm was then successfully applied in interspecific hybridization with tiger grouper (E. fuscoguttatus). Higher motility (90.56 ± 6.58%) and fertilization rate (69.61 ± 4.83%) was achieved in 10% Me2SO with Ringer solution as extender (dilution ratio 1:1), which should no significant difference in comparison with fresh sperm (95.88 ± 1.64% and 73.10 ± 1.28%). There were no statistical differences in both fertilization and hatching rates between hybrid and non-hybrid tiger grouper by using cryopreserved sperm for fertilization, but malformation rate of the hybrid was higher than non-hybrid (17%) (P < 0.05). Survival rate of the hybrid was lower than that of the controls at 15 days post hatching (23% vs 48%). However, hybrids showed survival rate equal to the controls at the end of the 60-day study period. Hybridization of E. fuscoguttatus x E. lanceolatus was successfully achieved using cryopreserved sperm from giant grouper. The cryopreservation of giant grouper sperm and its application in hybridization provided a technical support for further grouper breeding work.  相似文献

Ten microsatellite loci were developed for the northern spectacled salamander, Salamandrina perspicillata, and cross-amplification was obtained for the southern spectacled salamander, Salamandrina terdigitata. These two species are the sole representative of the genus Salamandrina, a threatened amphibian endemic to the Italian peninsula. The two salamanders show a parapatric distribution with a contact zone in Central Italy. All but one locus were polymorphic and six microsatellites were found diagnostic for species discrimination. We genotyped a total of 169 individuals. Allelic diversity was 5.1 ± 1.5 SE in S. perspicillata and 1.9 ± 0.31 SE in S. terdigitata. Mean observed and expected heterozygosity were 0.51 ± 0.06 SE and 0.61 ± 0.07 SE, respectively in the northern spectacled salamander and 0.35 ± 0.15 SE and 0.35 ± 0.09 SE, respectively in the southern spectacled salamander. These new markers will provide an excellent tool to assess population structure and genetic diversity in Salamandrina and will be used to investigate natural hybridization events between northern and southern spectacled salamanders.  相似文献

The aim of this study was to determine the level of contamination and genotoxic impact through micronucleus assay and histopathology in Wallago attu and Cirrhinus mrigala procured from the polluted site of the River Chenab at industrial and sewage waste disposal. The water sample was found viciously contaminated with heavy metals i.e. Ni, Cr, Mn, Co, Pb, Hg, Zn, Sn, Cu while all other physio-chemical variables crossed the suggested limits of WHO. The heavy metals load induced histopathological alterations were correlated to environmental degradation and the productivity of this biological system. W. attu and C. mrigala harvested from contaminated sites of the river indicated higher intensity of DNA damage through micronucleus induction and nuclear abnormalities with 5.46 ± 0.17, 1.23 ± 0.08 and 4.2 ± 0.11, 0.4 ± 0.04‰ respectively. Muscle sections of W. attu and C. mrigala harvested from the polluted section of river demonstrated the necrosis, degeneration of muscle fibers, intra-fibular edema and release of the blood into the tissues due to the bursting of blocked of the blood vessels. Dermal layers showed degeneration of the collagen bundles those were found loose or collapsed in some regions. Photomicrography also revealed vacuolar degeneration in muscle tissues and atrophy of muscle bundles. Intra fibular edema and splitting of muscle fibers were also seen along with bioaccumulation of toxicants. W. attu showed maximum incidence of alterations with highest histopathological alteration index related to environmental degradation. Control fish samples showed normal muscle tissues with normal equally spaced muscle bundles and myotomes.  相似文献

Predictive models of parasite life cycles increase our understanding of how parasite epidemiology is influenced by global changes and can be used to support decisions for more targeted worm control. Estimates of parasite population dynamics are needed to parameterize such models. The aim of this study was to quantify the main life history traits of Ostertagia ostertagi, economically the most important nematode of cattle in temperate regions. The main parameters determining parasite density during the parasitic phase of O. ostertagi are (i) the larval establishment rate, (ii) hypobiosis rate, (iii) adult mortality and (iv) female fecundity (number of eggs laid per day per female). A systematic review was performed covering studies from 1962 to 2007, in which helminth-naïve calves were artificially infected with O. ostertagi. The database was further extended with results of unpublished trials conducted at the Laboratory for Parasitology of Ghent University, Belgium. Overall inverse variance weighted estimates were computed for each of the traits through random effects models. An average establishment rate (±S.E.) of 0.269 ± 0.022 was calculated based on data of 27 studies (46 experiments). The establishment rate declined when infection dose increased and was lower in younger animals. An average proportion of larvae entering hypobiosis (±S.E.) of 0.041 (±0.009) was calculated based on 27 studies (54 experiments). The proportion of ingested larvae that went into hypobiosis was higher in animals that received concomitant infections with nematode species other than O. ostertagi (mixed infections). An average daily adult mortality (±S.E.) of 0.028 (±0.002) was computed based on data from 28 studies (70 experiments). Adult mortality was positively correlated with infection dose. A daily fecundity (±S.E.) of 284 (±45) eggs per female was found based on nine studies (10 experiments). The average female sex ratio of O. ostertagi based on individual animal data (n = 75) from six different studies was estimated to be 0.55. We believe that this systematic review is the first to summarise the available data on the main life history traits of the parasitic phase of O. ostertagi. In conclusion, this meta-analysis provides novel estimates for the parameterization of life cycle-based transmission models, explicitly reports measures of variance around these estimates, gives evidence for density dependence of larval establishment and adult mortality, shows that host age affects larval establishment and, to our knowledge, provides the first evidence for O. ostertagi of a female-biased sex ratio.  相似文献

Population density and biomass of bacteria and meiofauna were investigated seasonally in the sediments of the north-western bank of Red Sea. Samples of sediments were collected seasonally from three different stations to determine microphytobenthic biomass (chlorophyll a), protein, lipid, carbohydrate, and total organic matter concentrations. These investigations revealed that microbial components tended to increase their dominancy, whereas sensitive meiofauna were extremely reduced during the entire study period. Thus a very low density of the total meiofauna (with an annual average of 109 ± 26 ind./10 cm2) was recorded whilst the benthic microbial population densities exhibited higher values (ranging from 0.31 ± 0.02 × 108 to 43.67 ± 18.62 × 108/g dry sediment). These changes in the relative importance analysis of benthic microbial components versus meiofaunal ones seem to be based on the impact of organic matter accumulation on the function and structure of these benthic communities. Proteins, lipids and carbohydrates showed very low concentration values, and the organic matter mostly consisted of carbohydrates, reflecting lower nutritional values for benthic fauna in general and meiofauna in particular. The distribution of microbial and meiofaunal communities seems to be dependent on the quality of the organic matter rather than on its quantity. Total organic matter concentrations varied between 5.8 and 7.6 mg/g, with organic carbon accounting for only 32% of the total organic matter. Chlorophyll a attained very low values, fluctuating between 0.11 and 0.56 μg/g, indicating the oligotrophy of the studied area. The very low concentration of chlorophyll a in the Red Sea sediment suggests that the sedimentary organic matter, heterotrophic bacteria and/or protozoa constitute an alternative resource that is consumed by meiofauna when algae are less abundant. Protozoa, therefore, represent the “missing link in bacteria–meiofauna interaction in the Red Sea marine sediment ecosystem.  相似文献

Present work elucidates the antioxidant and antibacterial activity of Woodwardia unigemmata (Makino) Nakai along with chemical characterization using its aqueous (AEW), methanol (MEW), and hexane (HEW) extracts. Chemical profile of different extracts was illustrated by using Gas chromatography and mass spectrometry (GC-MS) analysis. Antioxidant activities were tested using DPPH and FRAP assays, total phenolic and flavonoid content by Folin-Ciocalteu and aluminum chloride method, respectively. Further, antibacterial activity against six plant and four animal pathogenic bacteria was analyzed by employing the disc diffusion assay. GC-MS analysis revealed the presence of catechol (21.96%), glycerol (20.22%), n-pentadecanoic acid (6.95%), glyceryl monoacetate (6.35 %), ethyl acetimidate (5.39 %) and 3-hydroxy-2,3-dihydromaltol (5.36%) in AEW; β-sitosterol (17.39%), pentadecanoic acid (9.81%), vitamin E (7.82%) and glycerol (7.05%) in MEW; γ-sitosterol (33.45%), vitamin E (10.04%) and campesterol (7.32%) in HEW as major constituents. Maximum phenolics (873 ± 6.01 mgGAE/g dry extract) as well as flavonoids (151 ± 11.44 mgQE/g dry extract) content was found in MEW, which also showed remarkable antioxidant potential (IC50 6.07 ± 1.4 µg/ml for DPPH and 768 ± 10.4 mg AAE/g dry extract for FRAP assay. In antibacterial activity, maximum inhibition (15 ± 0.9 mm) was observed for HEW against R. solanacearum, followed by AEW against A. tumefaciens and X. phaseoli (11 ± 0.3 mm each). MEW was found positive only against A. tumefaciens. Significant minimum inhibitory concentration (MIC) value observed for AEW against L. monocytogenes (10 mg/ml). Polar extracts had remarkable antioxidant potential, while non-polar extract did show significant antibacterial activity. Further, GC- MS reports indicated that this traditionally useful fern species can be an excellent source of biologically active compounds.  相似文献
