Bacillus cereus strain K-22 produced two distinct omega-amino acid transaminases, one catalyzing the transamination between beta-alanine and pyruvic acid and the other that between gamma-aminobutyric acid and alpha-ketoglutaric aic. The two enzymes were partially purified and separated from each other by various chromatographies. beta-Alanine:pyruvic acid transaminase and gamma-aminobutyric acid:alpha-ketoglutaric acid transaminase were induced by the addition of beta-alanine and gamma-aminobutyric acid, respectively, to the growth medium. beta-Alanine transaminase showed an optimum pH of 10.0 and optimum temperature of 35 degrees C, and its Km values for beta-alanine and pyruvic acid were both 1.1 mM. gamma-Aminobutyric acid, epsilon-aminocaproic acid, 2-aminoethylphosphonic acid, and propylamine showed about 30-40% of the activity of beta-alanine as amino donors, and oxalacetic acid was as good an amino acceptor as pyruvic acid. The optimum pH and temperature of gamma-aminobutyric acid transaminase were 9.0 and 50 degrees C, respectively, and its Km value for gamma-aminobutyric acid was 2.8 mM, while that for alpha-ketoglutaric acid was 2.3 mM. gamma-Aminobutyric acid and delta-aminovaleric acid were good amino donors but other omega-amino acids were virtually inactive with gamma-aminobutyric acid transaminase; alpha-ketoglutaric acid, and to a lesser extent glyoxylic acid, were active amino acceptors. 不同种源山桐子果实脂肪酸组成变异分析   总被引:1,自引:0,他引:1  
以采自11个种源的山桐子为材料,测定其果实脂肪酸的组成及其变异情况,结果表明:山桐子果实中不饱和脂肪酸含量较高,尤以亚油酸含量最高,11个种源的平均值为63.58%,且种源间差异显著,分宜、宜昌种源亚油酸相对含量明显高于其他9个种源;饱和脂肪酸以棕榈酸为主,11个种源山桐子果实棕榈酸差异显著,且以平武种源最高;其余脂肪酸含量均较低,变异幅度较大;种子中棕榈酸和棕榈烯酸含量明显高于果肉,而亚油酸、亚麻酸及硬脂酸含量明显低于果肉,油酸含量相近:果实不饱和脂肪酸含量依次为果肉〉全果〉种子,且变异系数及相对极差均较小,尤以果肉中最小;除硬脂酸外,山桐子果实中其他4种主要脂肪酸组分受海拔等地理环境的影响均较小。  相似文献

以219份枣种质脆熟期果实为试材,采用超高效液相色谱(UPLC)法检测不同枣种质果实中的山楂酸、桦木酸、齐墩果酸和熊果酸4种三萜酸组分含量,并进行描述统计分析、相关性分析和聚类分析,进而筛选三萜酸含量较高的优特异种质。结果表明:(1)枣果中总三萜酸(TTA)含量的变化范围95.72~737.82μg·g-1,平均值306.83μg·g-1,变异系数为29.02%,含量最高的为‘大荔小圆枣’。4种组分以山楂酸含量最高,其次为桦木酸,齐墩果酸和熊果酸含量较低,各组分在不同枣种质中都存在丰富的遗传多样性,其中熊果酸变异系数最高(60.75%)。(2)正态性检验结果表明,各种质的山楂酸、桦木酸和总三萜酸含量服从正态分布,齐墩果酸和熊果酸含量不服从正态分布。(3)相关性分析结果显示,总三萜酸含量与4个组分均呈极显著正相关关系,组分间也存在相关性,山楂酸含量与桦木酸呈极显著正相关关系,桦木酸含量与齐墩果酸呈极显著正相关关系,齐墩果酸含量与熊果酸呈极显著正相关关系。(4)聚类分析将219份枣种质划分为4大类群,分别为熊果酸含量最高的类群,齐墩果酸和熊果酸含量较高类群,各组分中低含量类群以及山楂酸、桦木酸和总三萜酸含量较高类群。(5)筛选出13份各组分或总三萜酸含量较高的优特异种质,为枣三萜酸相关功能产品的开发提供原料选择。  相似文献

1. Although Mycobacterium smegmatis could utilize a number of aromatic compounds as sole sources of carbon for growth, it did not appear to be able to use salicylic acid for growth or to metabolize it to any great extent. 2. When M. smegmatis was grown on shikimic acid as sole source of carbon, salicylic acid, anthranilic acid and 3,4-dihydroxybenzoic acid were released into the medium. When it was grown on quinic acid these compounds, together with p-hydroxybenzoic acid, p-hydroxyphenylacetic acid and a number of unidentified compounds, were formed. When it was grown on glucose only small amounts of salicylic acid could be detected. 3. When a washed suspension of cells with a normal iron content was incubated with shikimic acid, only small amounts of aromatic compounds were formed in the medium. When the cells were iron-deficient, substantial amounts of salicylic acid, 3,4-dihydroxybenzoic acid and catechol were formed, together with several other compounds not definitely identified. 4. When washed suspensions of cells, whether iron-sufficient or iron-deficient, were incubated with tryptophan no evidence of formation of salicylic acid, anthranilic acid or phenolic compounds was obtained. Washed suspensions did not convert anthranilic acid into salicylic acid. 5. When cell-free extracts of M. smegmatis were incubated with shikimic acid, or shikimic acid 5-phosphate, traces of anthranilic acid were formed under certain conditions. No formation of salicylic acid or other phenolic compound was observed even when a number of combinations of cofactors and coenzymes were tried.  相似文献

Metabolite profiles of lactic acid bacteria in grass silage   总被引:2,自引:0,他引:2  
The metabolite production of lactic acid bacteria (LAB) on silage was investigated. The aim was to compare the production of antifungal metabolites in silage with the production in liquid cultures previously studied in our laboratory. The following metabolites were found to be present at elevated concentrations in silos inoculated with LAB strains: 3-hydroxydecanoic acid, 2-hydroxy-4-methylpentanoic acid, benzoic acid, catechol, hydrocinnamic acid, salicylic acid, 3-phenyllactic acid, 4-hydroxybenzoic acid, (trans, trans)-3,4-dihydroxycyclohexane-1-carboxylic acid, p-hydrocoumaric acid, vanillic acid, azelaic acid, hydroferulic acid, p-coumaric acid, hydrocaffeic acid, ferulic acid, and caffeic acid. Among these metabolites, the antifungal compounds 3-phenyllactic acid and 3-hydroxydecanoic acid were previously isolated in our laboratory from liquid cultures of the same LAB strains by bioassay-guided fractionation. It was concluded that other metabolites, e.g., p-hydrocoumaric acid, hydroferulic acid, and p-coumaric acid, were released from the grass by the added LAB strains. The antifungal activities of the identified metabolites in 100 mM lactic acid were investigated. The MICs against Pichia anomala, Penicillium roqueforti, and Aspergillus fumigatus were determined, and 3-hydroxydecanoic acid showed the lowest MIC (0.1 mg ml(-1) for two of the three test organisms).  相似文献

本实验以华南主要野生蔬菜守宫木、土人参、一点红、白仔菜、紫背菜、鳄嘴花、藤三七、塘葛菜为材料,并以华南特产蔬菜菜心为对照,对8种野生蔬菜的脂肪酸成分进行了分析.结果表明:8种野菜共检出十四酸、软脂酸、棕榈油酸、硬脂酸、油酸、亚油酸、亚麻酸、二十二酸和二十四酸共9种脂肪酸,但不同野菜之间的脂肪酸组成与含量差异极大.8种野菜的饱和脂肪酸的总量都高于菜心,饱和脂肪酸种类最多的是藤三七,含4种.不饱和脂肪酸的变化与饱和脂肪酸相反,8种野菜都低于菜心,但其油酸、亚油酸远高过菜心.菜心富含亚麻酸,但不含亚油酸.可见8种野菜油营养价值较高.  相似文献

Bile acid profiles of bile, urine, and feces obtained from a patient with cerebrotendinous xanthomatosis on the same day have been analyzed by gas-liquid chromatography-mass spectrometry after fractionation into groups by mode of conjugation by an ion-exchange chromatography. The predominant biliary bile acid was cholic acid conjugated with glycine and taurine. Lesser amounts of the amino acid conjugates of chenodeoxycholic acid, ursodeoxycholic acid, 7-ketodeoxycholic acid, allocholic acid, and deoxycholic acid, and of unconjugated norcholic acid and allonorcholic acid were also present in the bile. The major fecal bile acid was 7-epicholic acid. Relatively large amounts of bile acids were excreted in the urine. Unconjugated 7-epicholic acid, norcholic acid, allonorcholic acid, and cholic acid predominated. The bile acid profiles of the patient were different from those of normal subjects and should be useful for the diagnosis.  相似文献

The alterations by peroxisome proliferators of metabolism of linoleic acid in rat liver were studied. Administration of P-chlorophenoxyisobutyric acid (clofibric acid) enhanced in vivo conversion of linoleic acid to its desaturated and/or elongated metabolites, 6,9,12-octadecatrienoic acid, 8,11,14-eicosatrienoic acid, and arachidonic acid, whereas the formation of 11,14-eicosadienoic acid was decreased. These changes observed in vivo were confirmed in vitro to be due to the increases in activities of delta 6 desaturation of linoleic acid to 6,9,12-octadecatrienoic acid (18.4 times), delta 8 desaturation of 11,14-eicosadienoic acid to 8,11,14-eicosatrienoic acid (3.4 times), and delta 5 desaturation of 8,11,14-eicosatrienoic acid to arachidonic acid (4.1 times). No considerable changes in activities of chain elongation of either linoleic acid or 6,9,12-octadecatrienoic acid were observed. The increases in the activities of three desaturations by clofibric acid were prevented by the treatment of rats with cycloheximide. The inductions of delta 6 and delta 5 desaturations were brought about by the treatment of rats with 2,2'-(decamethylenedithio)diethanol or di-(2-ethylhexyl)-phthalate, peroxisome proliferators structurally unrelated to clofibric acid, as well. These changes in metabolism of linoleic acid by clofibric acid were consistent with the changes in mass proportion of omega 6 fatty acids in hepatic lipid. Physiological significance of the marked changes in linoleic acid metabolism by peroxisome proliferators was discussed.  相似文献

The urinary bile acid profile, obtained by capillary gas chromatography, of a patient suffering from cerebrotendinous xanthomatosis and treated with ursodeoxycholic acid demonstrated, besides the occurrence of 23-norcholic acid and (23R)-hydroxycholic acid (as a consequence of this disease), six additional unknown bile acids and three known bile acids, viz. ursodeoxycholic acid, hyocholic acid and omega-muricholic acid. The structure of two of the unknown bile acids were elucidated and proven by organic syntheses. These were 23-norursodeoxycholic acid and 3 beta-ursodeoxycholic acid. The structures of three bile acids were tentatively elucidated as being 1 beta-hydroxyursodeoxycholic acid, 21-hydroxyursodeoxycholic acid and 22-hydroxyursodeoxycholic acid, and the possibility that the structure of the remaining bile acid is that of 5-hydroxyursodeoxycholic acid is discussed. Two of these bile acids (1 beta-hydroxyursodeoxycholic acid and 5-hydroxyursodeoxycholic acid) also occurred in urine of a healthy individual during oral ursodeoxycholic acid treatment, whereas 23-norcholic acid, 23-norursodeoxycholic acid, (23R)-hydroxycholic acid, 21-hydroxyursodeoxycholic acid and 22-hydroxyursodeoxycholic acid were only present in urine of the patient suffering from cerebrotendinous xanthomatosis. The metabolism of ursodeoxycholic acid, both in the normal state and in the cerebrotendinous xanthomatosis, is discussed.  相似文献

Cellular fatty acids of 10 strains of lactic acid bacteria were analyzed. The purpose of this work was to find lactic acid bacteria with high lactobacillic acid contents. The bacteria studied were unable to synthesize oleic acid. Some strains did not synthesize lactobacillic acid, although all were able to form dihydrosterculic acid. Twenty-one to thirty-four percent of the fatty acid content of Lactobacillus fermentum and L. buchneri was lactobacillic acid, and these species were chosen for future studies of environmental factors affecting cyclopropane fatty acid synthesis.  相似文献

It was found that when Rhodotorula rubra IFO 0911 was grown in a phenylalanine medium, benzoic acid and p-hydroxybenzoic acid besides cinnamic acid were formed in the cultured both. The conversions of cinnamic acid into benzoic acid and of benzoic acid into p-hydroxybenzoic acid, and the degradation of p-hydroxybenzoic acid were demonstrated in intact cells of Rhodotorula rubra. These activities were observed in the cells grown on various media, including the medium containing no phenylalanine, and were found to be distributed widely in Rhodotorula. The cells of Rhodotorula rubra were also able to degrade p-coumaric acid, 3,4-dihydroxybenzoic acid (protocatechuic acid), p-hydroxyphenyl-acetic acid, 3-methoxy-4-hydroxycinnamic acid (ferulic acid) and 3-methoxy-4-hydroxybenzoic acid (vanillic acid). From these results, the metabolic pathways for phenylalanine and tyrosine in Rhodotorula were discussed.  相似文献

采用气质联用技术对宽叶独行菜中脂肪酸成分进行了分析,从其乙醇提取物的石油醚萃取部分共分离得到20个组分,采用面积归一化法测定了各组分的含量,其中棕榈酸(hexadecanoic acid)53.980%,硬脂酸(oc-tadecanoic acid)17.063%,二十二烷酸(docosanoic acid)4.769%,二十四烷酸(tetracosanoic acid)3.363%,二十八烷酸(octacosanoic acid)2.773%,花生酸(eicosanoic acid)2.719%,十五烷酸(pentadecanoic acid)2.476%,十七烷酸(heptadecanoic acid)2.291%。此外,三十烷酸(triacontanoic acid,十四烷酸(tetradecanoic acid),二十三烷酸(tricosanoic acid)和二十六烷酸(hexacosanoic acid)的含量均已超过了1%。  相似文献

The formation of ursodeoxycholic acid from chenodeoxycholic acid and the role of 7-ketolithocholic acid as an intermediate in this biotransformation were studied in vitro in fecal incubations as well as in vivo in the human colon. [24-14C]-Labeled 7-ketolithocholic and chenodeoxycholic acids were studied at various concentrations, and the biotransformation products were analyzed by thin-layer chromatography, gas-liquid chromatography, and mass spectrometry. There was rapid colonic conversion of 7-ketolithocholic acid to ursodeoxycholic acid and, to a lesser extent, to chenodeoxycholic acid. The reduction of 7-ketolithocholic to ursodeoxycholic acid proceeded significantly faster anaerobically and at acid pH than under aerobic and alkaline conditions. When chenodeoxycholic acid was incubated in vitro or instilled into the colon, various amounts of 7-ketolithocholic and ursodeoxycholic acids were formed. The formation of 7-ketolithocholic acid was favored by alkaline conditions. Isotope dilution studies, in which trace amounts of labeled 7-ketolithocholic acid were incubated with unlabeled chenodeoxycholic acid, indicate 7-ketolithocholic acid to be the major intermediate in the intestinal bacterial conversion of chenodeoxycholic to ursodeoxycholic acid.  相似文献

对甘肃省116份胡麻地方品种的重要品质指标———粗脂肪、硬脂酸、棕榈酸、油酸、亚油酸、亚麻酸含量和碘值进行测定,并根据这些品质指标对供试品种进行聚类分析。结果显示:(1)供试品种粗脂肪含量平均值为37.48%,变异系数3.8%;硬脂酸、油酸含量平均值分别为5.32%和29.05%,变异系数分别为19.5%和11.6%;棕榈酸、亚麻酸、亚油酸含量平均值分别为5.9%、48.76%、10.95%,变异系数分别为8.4%、8%、8%;平均碘价175.60,变异系数2.76%;(2)聚类分析结果显示,116个品种聚为7大类,其中:b亚组群硬脂酸和油酸含量最高,而亚麻酸含量最低;d亚组群品种亚麻酸含量最高;e亚组群粗脂肪含量和碘价最高,油酸含量最低;f亚组群硬脂酸含量最低;g亚组群棕榈酸含量最高,碘价最低。  相似文献

利用微生物发酵无花果开发特色香料,并采用同时蒸馏萃取装置收集挥发性成分并用气相色谱一质谱仪对生物技术制备的无花果香料挥发性成分进行分离和鉴定,经毛细管色谱分离出47种组分,确认了其中的45种成分,并用面积归一化法测定了各种成分的百分含量,其主要成分为:9,12-十八碳二烯酸乙酯(27.34%)、十六酸乙酯(23.99%)、邻苯二甲酸二丁酯(6.18%)、邻苯二甲酸二异丁酯(5.52%)、9,12-十八碳二烯酸甲酯(4.72%)、十六酸甲酯(4.67%)、9,12,15-十八碳三烯酸乙酯(4.48%)、9-十八碳烯酸乙酯(3.80%)、糠醛(2.53%)、9,12,15-十八碳三烯酸甲酯(1.85%)、十八酸乙酯(1.42%)、9-十八碳烯酸甲酯(1.26%)等。  相似文献

Fatty acid compositions of liver phospholipid, cholesterol ester and triacylglycerol fractions obtained from streptozotocin-induced diabetic rats were compared to those from control or from simple-acidotic rats. Significant reductions of arachidonic acid proportions in phospholipid and cholesterol ester were found on the 3rd day after the streptozotocin treatment. In triacylglycerol, arachidonic acid and the other desaturation and elongation products of linoleic acid except for gamma-linolenic acid were increased in the diabetic rats. Although essential fatty acid composition in liver phospholipid and cholesterol ester of simple-acidotic rats did not differ from control rats, dihomo-gamma-linolenic acid, arachidonic acid, adrenic acid and docosapentaenoic acid (22:5(n - 6] contents in liver TG were significantly increased over those in control rats and were similar to those in diabetic rats. These results suggest that metabolic acidosis may contribute to the fatty acid abnormalities observed in diabetic animals.  相似文献

采用索氏提取法提取木通科植物猫儿屎和三叶木通种子的脂溶性成分,甲酯化处理后用气相色谱-质谱联用技术(GC-MS)分离和鉴定其组成和含量。从猫儿屎种子油中鉴定出9种脂肪酸,占检出物总质量分数的94.67%(其中饱和脂肪酸占12.63%,不饱和脂肪酸占82.04%),主要成分为9-十六烯酸(47.22%)、9-油酸(27.13%)、棕榈酸(10.75%)、亚油酸(7.47%)和硬脂酸(1.61%)。从三叶木通种子油中鉴定出10种脂肪酸,占检出物总质量分数的99.75%(其中饱和脂肪酸占23.39%,不饱和脂肪酸占76.36%),主要成分为11-油酸(47.63%)、亚油酸(27.05%)、棕榈酸(20.14%)、16-甲基-十七烷酸(3.03%)和8-油酸(1.07%)。结果表明,猫儿屎和三叶木通种子油中脂肪酸含量丰富,在食用、医疗保健等方面具有较高的应用潜力和综合开发前景。  相似文献

该文研究了不同浓度的阿魏酸、对香豆酸、丁香酸、对羟基苯甲酸、香草酸5种酚酸类物质对三七幼苗生长和生理的影响。结果表明:处理后,三七幼苗的苗高、根长、可溶性蛋白质含量、根系活力、CAT以及POD活性均有所降低。其中,阿魏酸各处理组幼苗的苗高及POD活性均显著降低,50、100 mg·L~(-1)的对香豆酸以及100 mg·L~(-1)的香草酸处理组幼苗苗高也分别比对照显著降低16.19%、16.67%和29.29%;对香豆酸、丁香酸以及对羟基苯甲酸各处理组幼苗根长均显著低于对照;香草酸处理组幼苗的根系活力也显著低于对照,且幼苗的CAT活性在10、50、100 mg·L~(-1)丁香酸、对羟基苯甲酸以及香草酸处理下也达到了显著降低水平。此外,1 mg·L~(-1)阿魏酸以及100 mg·L~(-1)香草酸处理组幼苗的叶绿素含量也均显著降低;中高浓度的阿魏酸、对香豆酸、丁香酸、对羟基苯甲酸增加了三七幼苗的MDA含量,而香草酸在0.1、1、10、100 mg·L~(-1)浓度下显著降低幼苗的MDA含量;丁香酸、香草酸、对羟基苯甲酸以及中高浓度的对香豆酸增加了三七幼苗的SOD活性,且香草酸各处理组均达到了显著性水平。综上结果表明,5种酚酸类物质对三七幼苗均具有一定的化感抑制作用,但各酚酸物质的作用方式及强度并不完全一致,阿魏酸的化感影响较大,这为进一步研究三七的化感自毒作用提供了一定的理论参考。  相似文献
