T and B mouse spleen lymphocytes were separated by density gradient electrophoresis on the basis of their surface charge. In all strains examined, the T lymphocytes were found in the high mobility fractions and the B in the low. The T and B cells were separated completely in most fractions, with some overlapping in the middle. Significant differences were found in the electrophoretic distribution profiles between the strains: C57BL/6j, C57BL/10j, (BALB/cXC57BL/6j)F1, and all the following: B6·C-H-2d/cBy (congenic to C57BL/6j), BALB/c, CBA/H/T6j, C57BL/10Sn, and C3H. The C57BL/6j and the (BALB/cXC57BL/6j)F1 cells appear more heterogeneous as far as electrophoretic mobility is concerned. Almost all the other strains give two major peaks. Moreover, the high mobility areas are less populated in the C57BL/6j and the (BALB/cXC57BL/6j)F1 animals than in all the others. The above differences were found consistently when cells prepared by different methods were electrophoresed. It is concluded that the surface charge of lymphocytes may be genetically determined. Possible dependency on the H-2 complex or non-H-2 areas is discussed.  相似文献

Summary The surfaces of developing erythrocytic cells and macrophages have been examined in normal human bone marrow by means of the pyroantimonate-osmium, ruthenium red and Thorotrast techniques for inorganic cations, surface glycoprotein-phospholipid complexes and surface anionic binding sites, respectively. No differences in the degree of surface coat reactivity were noted in the erythrocytic cells at different stages of maturation while pyroantimonate binding to the plasmalemma was not evident developmentally until the final stages of erythrocytic development. Rhopheocytotic invaginations proved to be chemically distinct from the remainder of the cell surface since they did not bind Thorotrast or pyroantimonate and gave more staining with ruthenium red. Pyroantimonate does not bind to the surface of macrophages and the binding of Thorotrast by these cells is less. Macrophage-erythrocytic cell contact zones did not stain with Thorotrast but stained with ruthenium red. 通过RT-PCR扩增957bp的MAGE-3全长编码序列,将该片段克隆至Pgex-4T-2原核表达载体,转化大肠杆菌BL-21,经IPTG诱导表达,并经12%SDSPAGE凝胶电泳,考马斯亮蓝染色及Western blot鉴定,证明了目的基因的有效表达,目的蛋白高达细菌总蛋白的32%。表达产物经Glutathione Sepharose 4B 纯化后,每100mL菌液最终可获得3mg的目的蛋白,蛋白纯度在90%以上。纯化的GST-MAGE-3蛋白在体外冲击树突状细胞,能诱导特异性CTL杀伤肿瘤细胞活性。  相似文献

Vicia ser. Vicia, which includes the Vicia sativa aggregate, was studied by observing morphological variation. The study is an attempt to resolve the taxonomic relationships within the Vicia sativa aggregate by novel investigations along with a synthesis of the results obtained by previous researchers. The study has included some accessions and characters used previously, but has drawn accessions from a wider geographical range than has been attempted previously. Cluster analysis on the basis of 53 morphological characters justified the delimitation of four species within the series and six subspecies within the Vicia sativa aggregate. Accessions from South Asia could easily be incorporated in the classification, but some of the accessions from North Africa showed morphological differences. The morphometric data were coded using the gap weighting method of Thiele (1993) and used in a phylogenetic analysis to study the relationships between the taxa. The phylogenetic analysis, using 33 variables, showed Vicia pyrenaica closely related to Vicia sativa subsp. amphicarpa. A dichotomous key and a multi-access key to aid the identification of the taxa in the series are presented. Magnetic filtration of surface-labeled cells has been applied to the fractionation of leukocytes in a model system, using a colloidal magnetite reagent to label mouse spleen cells. This reagent was completely free from problems of aggregation or settling. Since the individual submicron particles were invisible under the microscope, cells were not visibly altered by labeling. Viability also was unaffected by either labeling or magnetic filtration. Using a 10-kG magnet and a 5-mL filter column, 50 million cells were fractionated in less than 10 min, with 99% removal of labeled T lymphocytes. The efficiency of the magnetic method is limited at present by the fact that cells that do not have the surface target antigen of interest, and so are not antibody coated, may adsorb a small amount of label nonspecifically. These then have a nonzero chance of being captured in the filter along with the labeled cells.  相似文献

Melanoma is an aggressive malignancy with poor prognosis. Eradication of tumor cells requires an effective interaction between melanoma cells and different players of the immune system. As the most potent professional antigen‐presenting cells, dendritic cells (DCs) play a pivotal role in mounting a specific immune response where their intratumoral and peritumoral density as well as their functional status are correlated with clinical staging of the disease and with patients’ survival. Under steady‐state conditions, internalization of apoptotic cells by immature DCs designates a state of tolerance to self‐antigens. Nevertheless, pathogens and necrotic cells interacting with pattern recognition receptors trigger downstream signaling pathways that evoke maturation of DCs, leading to the production of pro‐inflammatory cytokines. These mature DCs are essential for T‐cell priming and subsequent development of a specific immune response. Altered functions of DCs have an impact on the development of various disorders including autoimmune diseases and cancers. Herein, we focus on the checkpoints created throughout DCs antigen capturing and presentation to T cells, with subsequent development of either tolerance or immune response, with an emphasis on the role played by DCs in melanoma tumorigenesis and their therapeutic potential.  相似文献

Ecosystem stress and health: an expansion of the conceptual basis   总被引:2,自引:0,他引:2  
The assessment of the ecosystem health and departures from it requires clarity of what the system, its structure, dynamics, and healthy conditions are. Available definitions provide inadequate tools to acquire this clarity and may lead to arbitrary diagnoses of ecosystem health but such diagnoses can be overturned on a variety of scientific, philosophical, or political grounds.Nested hierarchy of ecosystem structure compounds the difficulty in the assessment of stress and health because both states may occur simultaneously at different hierarchical levels: with stress at one level being a necessary condition of health at another.An approach based on a formal definition of system change is advanced. First, a conceptual model identifies a self-maintaining minimum interactive structure (MIS) at each level of ecosystem organization. Components of MIS are complementary, coordinated, and exchanging information — they are integrated. Function is defined as a contribution of a component to the maintenance of the whole. In this context health is viewed as persistence of the system at a given temporal and spatial scale. Impairment of the function is stress and is contrasted with change of system structure (loss, addition, or replacement of components of MIS) which is disturbance. Stress can be measured directly by changes of function or indirectly by changes in integration. Even though undesirable from the human point of view, a changed system may again be considered healthy.  相似文献

Morphometric and physiological studies were conducted to determine whether the wall ingrowths of transfer cells in the minor-vein phloem of Pisum sativum L. leaves increase the capacity of the cells for solute influx. Size and number of wall ingrowths are positively correlated to the photon flux density (PFD) at which the plants are grown. An analysis of plasmodesmatal frequencies indicated that numerous plasmodesmata are present at all interfaces except those between the sieveelement-transfer-cell complex (SE-TCC) and surrounding cells where plasmodesmata are present but few in number. Flux of exogenous sucrose into the SE-TCC was estimated from kinetic profiles of net sucrose influx into leaf discs, quantitative autoradiography, and measurements of sucrose translocation. Flux based both on the saturable (carrier-mediated) and the linear components of influx was 47% greater in leaves of plants grown at high PFD (1000 mol·m–2·s–1) than those grown in low PFD (200 mol·m–2·s–1) and was paralleled by a 47% increase in SE-TCC plasmalemma surface area. Flux of endogenous photosynthate across the SE-TCC plasmalemma was calculated from carbon balance and morphometric data. The increase in flux in high-light leaves over that in low-light leaves can be explained on the basis of an increase in plasmalemma surface area. 以南京市为研究区,基于ArcGIS、Erdas、GuidosToolbox和Conefor等软件平台,采用形态学空间格局分析(MSPA)和景观连通性分析方法,通过在MSPA中设置不同的粒度、边缘宽度和在景观连通性分析中设置不同的扩散距离阈值,对2013年南京市绿色基础设施网络格局变化的尺度效应、边缘效应与距离效应进行评价.结果表明: 基于MSPA获取的景观类型构成存在明显的尺度效应和边缘效应,且边缘效应对MSPA景观类型的影响较尺度效应更为明显.不同扩散距离对景观连通性的影响很大,对于南京市来说,2 km或2.5 km是关键的扩散距离阈值.当输入数据选择粒度30 m、边缘宽度30 m时,可以得到南京市城市绿色基础设施(UGI)网络更为详尽的景观信息.基于MSPA与景观连通性方法,分析尺度效应、边缘效应、距离效应对研究区UGI网络景观类型的影响,有助于选择合适的粒度、边缘宽度及扩散距离,并更好地理解UGI网络的空间格局和与生态过程相关的尺度效应和距离效应,从而使得UGI网络时空格局变化分析时的参数设置更为科学合理.研究结果可为中尺度范围内UGI景观网络时空格局分析时的参数设置提供重要的参考和依据,对其他地区UGI景观网络的分析也具有一定的借鉴意义.  相似文献

Bone marrow mesenchymal stem cells (BMSC) have the potential to differentiate into a variety of cell types like osteoblasts, chondroblasts, adipocytes, etc. It is well known that mechanical forces regulate the biological function of cells. The aim of this study was to investigate the effect of uniaxial stretching on the orientation and biological functions of BMSC. Rat BMSCs were harvested from femoral and tibial bone marrow by density gradient centrifugation. Cells from passages 1-6 were characterized by flow cytometry using monoclonal antibodies. The recovered cells were stably positive for the markers CD90 and CD44 and negative for CD34 and CD45. A cyclic 10% uniaxial stretching at 1Hz was applied on rat BMSC for different time-courses. The length, width, and orientation of the cells were subsequently determined. Expression of collagen types I and III and tenascin-C mRNAs was measured by real-time RT-PCR, and the synthesis of these receptors was determined by radioimmunoassay. Results showed that uniaxial stretching lengthened and rearranged the cells. Compared with control groups, expression of collagen types I and III mRNAs was up-regulated after 12-h of stretching, while significant increase in synthesis of the two collagen protein types was not observed until after 24-h stretching. The expression of tenascin-C mRNA was significantly increased after a 24-h stretching. These data suggest that cyclic stretching promotes the synthesis of collagen types I and III and tenascin-C by the rat BMSC.  相似文献

目的:探讨熊果酸对酒精所致骨质疏松大鼠骨形成、骨矿化的影响。方法:雄性Wistar大鼠60只,按体重随机分为空白对照 组、熊果酸对照组、模型组、熊果酸低、中、高剂量组,同时分别给予生理盐水、150 mg/kg 熊果酸、50%酒精,50 mg/kg 熊果酸,100 mg/kg 熊果酸,150 mg/kg 熊果酸灌胃。熊果酸对照组生理盐水剂量同空白组,熊果酸低、中、高剂量组酒精剂量同模型组。灌胃共 持续8 周。磷钼酸法检测血清磷(P)含量,比色法检测血清钙(Ca)含量,酶联免疫吸附(ELISA)法检测血清骨钙素(BGP)、骨形成蛋 白-2(BMP-2)浓度;HE 染色法观察股骨结构的病理学变化。结果:与空白对照组相比较,模型组血清BGP、BMP-2 和Ca、P 均明显 降低,且有统计学差异(P < 0.05),但熊果酸对照与空白对照组各项指标结果相近。熊果酸中、高剂量组大鼠血清BGP、Ca 和P 水 平均较模型组有显著升高,差异具有统计学意义(P < 0.05),但仅熊果酸高剂量组血清BMP-2 显著升高(P < 0.05)。股骨组织HE 染色结果显示,空白对照组骨小梁致密、规则且较粗,粗细均匀;模型组骨小梁稀松、不规则、粗细不均匀,甚至可见骨小梁断裂; 熊果酸中、高剂量组骨小梁致密、规则、较厚、粗细均匀,未见骨小梁断裂。结论:熊果酸能够促进酒精性骨质疏松大鼠的骨形成, 抑制骨矿物质的流失,在改善酒精致骨质疏松方面有一定的保护作用。  相似文献

目的构建小鼠CXC型趋化因子受体2(CXCR2)基因cxcr2过表达的骨髓间充质干细胞(Bone marrow mes-enchymal stem cell,BMSC)并进行鉴定。方法全骨髓贴壁法分离培养小鼠BMSC,采用流式细胞术检测干细胞抗原1(stem cell antigen-1,SCA-1)、CD44、CD43、CD45、IA/IE表达率,并诱导成骨分化。以含有小鼠cxcr2的质粒为模版进行PCR扩增,将获得的cxcr2克隆到慢病毒载体,命名为p Lenti-cxcr2-GZ;将其与慢病毒包装质粒共转染HEK-293T细胞,收获慢病毒后,通过离心法感染BMSC,经过1μg/mL zeocin压力选择建立了稳定表达CXCR2的小鼠BMSC(CXCR2-BMSC)。采用流式细胞术和RT-PCR分别检测其CXCR2蛋白和m RNA表达水平,Transwell趋化实验检测其迁移能力。结果 90%以上的第3代BMSC表达CD44、SCA-1,几乎不表达IA/IE、CD34、CD45,且成功诱导成骨分化。菌液PCR、质粒双酶切后,琼脂糖凝胶电泳鉴定结果得到特异、大小正确的条带及测序鉴定正确,表明成功构建了p Lenti-cxcr2-GZ表达质粒。流式细胞术和RT-PCR结果显示,CXCR2-BMSC的CXCR2蛋白和m RNA表达水平均明显高于对照组BMSC,差异有统计学意义(P<0.001)。Transwell结果显示,CXCR2-BMSC迁移能力高于对照组BMSC,差异有统计学意义(P<0.01)。结论利用慢病毒系统成功构建了稳定表达CXCR2的BM-SC,cxcr2基因修饰BMSC后可明显增加BMSC的迁移能力。  相似文献

Type I procollagen C-proteinase enhancer (PCPE) exists in hepatic stellate cells (HSCs) which can produce collagen. The deduced amino acid sequence of PCPE contains motifs specific for RNA-binding proteins. The effect of PCPE on the syntheses of collagen and noncollagenous protein was studied using an HSC clone derived from cirrhotic rat liver. When the cells were cultured in the presence of an antisense oligonucleotide (AS) against PCPE mRNA, the synthesis of noncollagenous protein as well as collagen was reduced compared to the cells cultured with addition of a nonsense oligonucleotide (NS). The extent of the reduction was similar in both syntheses. The total RNA content of the AS-treated cells and NS-treated cells did not differ. In the presence of actinomycin D, however, such total RNA content was decreased more rapidly in the AS-treated cells than in the NS-treated cells. PCPE may be involved in stabilization of RNA strands in noncollagenous protein synthesis as well as collagen synthesis.  相似文献

Indirect evidence for the site of stachyose biosynthesis has been provided by determining the occurrence and distribution of stachyose, raffinose and galactinol, the donor of the galactosyl moiety for stachyose synthesis, in Cucumis melo L. cv. Ranjadew. Studies of enzyme activities for the synthesis of these sugars and their distribution in different plant organs and isolates has led to the conclusion that stachyose is synthesized mainly in mature leaves and seeds. Nevertheless, stachyose-synthase activity varied with leaf age, the developmental stage of a plant, the growing season and the plant cultivar used. No stachyose or stachyose-synthase activity could be detected in isolated mesophyll protoplasts and chloroplasts, whereas both were found in a minor-vein-enriched fraction isolated from mature leaves. The conclusion that stachyose biosynthesis is associated with minor veins was confirmed by immunolocalization of the enzyme. Positive specific immunoreactivity of stachyose synthase with polyclonal anti-stachyose-synthase antibodies, labeled with protein A-gold, was detected in intermediary cells of leaf minor veins. Telocytes (TCs) are new cellular entities of mesenchymal origin described almost ubiquitously in human and mammalian organs ( www.telocytes.com ). Different subtypes of TCs were described, all forming networks in the interstitial space by homo‐ and heterocellular junctions. Previous studies analysed the gene expression profiles of chromosomes 1, 2, 3, 17 and 18 of murine pulmonary TCs. In this study, we analysed by bioinformatics tools the gene expression profiles of chromosome 4 for murine pulmonary TCs and compared it with mesenchymal stem cells (MSCs), fibroblasts (Fbs), alveolar type II cells (ATII), airway basal cells, proximal airway cells, CD8(+) T cells from bronchial lymph nodes (T‐BL) and CD8(+) T cells from lungs (T‐L). Key functional genes were identified with the aid of the reference library of the National Center for Biotechnology Information Gene Expression Omnibus database. Seventeen genes were up‐regulated and 56 genes were down‐regulated in chromosome 4 of TCs compared with other cells. Four genes (Akap2, Gpr153, Sdc3 and Tbc1d2) were up‐regulated between one and fourfold and one gene, Svep1, was overexpressed over fourfold. The main functional networks were identified and analysed, pointing out to a TCs involvement in cellular signalling, regulation of tissue inflammation and cell expansion and movement.  相似文献

Objectives:To investigate the effect of neurotrophin-3 (NT-3) on osteogenic/adipogenic differentiation of bone marrow mesenchymal stem cells (BMSCs).Methods:Osteogenic differentiation was detected by alkaline phosphatase (ALP) staining and alizarin red staining (ARS). Adipogenic differentiation was detected by oil red O (ORO) staining. The expression of bone-related genes (Runx2, Osterix, OCN, ALP) and lipogenic genes (FABP4, PPAR, CEBP, LPL) was detected by real-time quantitative polymerase chain reaction (real-time qPCR). The expression of p-Akt and Akt protein was detected by Western blot assay.Results:ALP staining and ARS staining showed that the overexpression of NT-3 could promote the differentiation into osteoblasts, while knockdown of NT-3 could inhibit that. Real-time qPCR showed that the overexpression of NT-3 could increase the expression of osteoblast genes, while knockdown of NT-3 could inhibit that. ORO staining showed that the overexpression of NT-3 could inhibit the differentiation into adipogenesis, while knockdown of NT-3 can promote that. Real-time qPCR showed that the overexpression of NT-3 could reduce the expression of lipogenic genes. while knockdown NT-3 could increase that. In addition, the overexpression of NT-3 increased p-Akt/Akt levels significantly, while knockdown NT-3 reduced that significantly.Conclusion:NT-3 could promote the differentiation of mouse BMSCs into osteoblasts and inhibit their differentiation into adipogenesis.  相似文献

The complete nucleotide sequences of the genomes of the type 2 ( P712 , Ch, 2ab ) and type 3 (Leon 12a1b ) poliovirus vaccine strains were determined. Comparison of the sequences with the previously established genome sequence of type 1 (LS-c, 2ab ) poliovirus vaccine strain revealed that 71% of the nucleotides in the genome RNAs were common, that the 5' and 3' termini of the genomes were highly homologous, and that more than 80% of the nucleotide differences in the coding region occurred in the third letter position of in-phase codons, resulting in a low frequency of amino acid difference. These results strongly suggested that the serotypes of poliovirus derived from a common prototype. A comparison of the amino acid sequences predicted from the genome sequences showed highest variation in the capsid protein region, whereas non-structural proteins are highly conserved. Initiation of polyprotein synthesis occurs in all three strains more than 740 nucleotides downstream from the 5' end. An analysis of the non-coding region suggests that small peptides that could potentially originate from this region are conserved. The amino acid sequences immediately surrounding the cleavage signals, however, show a higher than average degree of variation. The analysis of the amino acid sequences of the capsid protein VP1 of all serotypes has led to the prediction of potential antigenic sites on the virion involved in neutralization.  相似文献
