This report describes a case of peritoneal larval cestodiasis caused by tetrathyridia of Mesocestoides sp. in an adult female squirrel monkey. The monkey had lived in a zoological garden in Japan and had a clinical history of wasting. At necropsy, numerous whitish oval masses were found in the liver and peritoneal cavity. These masses contained larval cestodes. Morphological observation and molecular analyses of the mitochondrial 12S ribosomal RNA gene and cytochrome c oxidase subunit 1 gene sequences allowed us to identify the larva as the tetrathyridium of Mesocestoides sp. This is the first report of Mesocestoides larvae in a squirrel monkey in Japan.  相似文献

Mesocestoides corti is a suitable in vitro model for studying the development of human endoparasitic platyhelminthes. Treatment with trypsin, supplemented with fetal bovine serum (FBS), induces M. corti development from larvae (tetrathyridia) to segmented adult worm; however, the role of this protease and of FBS in post-larval development induction remains unknown. To characterize the participation of trypsin enzymatic activity and of FBS in the induction of tetrathyridia growth and development, both stimuli were added to the larvae either together or sequentially. Additionally, specific inhibition of trypsin activity was also monitored. Finally, the effect of the enzyme on the parasite tegument as well as the proliferative activity and location of proliferating cells after induction of tetrathyridia development were also studied. We conclude that trypsin-induced tetrathyridia development to adult worm is FBS-dependent and that the effect of serum factors is dependent upon a previous trypsin-induced reversible damage to the larva tegument. In dividing and non-dividing tetrathyridia, proliferative activity of cells is mainly located within the apical massif in the anterior region and nerve cords of larvae, respectively. In tetrathyridia stimulated to develop to adult worms, an intense proliferative activity is evident along the nerve cords. Our results suggest that in natural infections the tetrathyridia tegument is temporally made permeable to growth factors by proteolytic enzyme activity in the intestine juice of the definitive host, thus leading to development to adult worms.  相似文献

The function of calcareous bodies, commonly found in the parenchyma of cestodes and trematodes, is relatively poorly understood. The present histochemical ultrastructural study of the proliferative tetrathyridia of Mesocestoides vogae revealed that calcareous corpuscles begin to form as organic (lipid-protein) masses that coalesce in parenchymal (calcareogenic) cells. Concentric accretion of organic and inorganic crystalline material then leads to the formation of typical refractile calcareous bodies. The precise composition of such bodies, determined by x-ray diffraction, revealed that their major inorganic constituent is indeed calcium, with significant amounts of phosphorus, silicon, and zinc as well. Emission of calcareous bodies through the tegument was observed by scanning electron microscopy, explaining their accumulation in the intracapsular spaces around worms embedded in liver tissue of the host. Following their emission, the crystalline substance of corpuscles dissolves, leaving only nonrefractile, membrane-bound cytoplasmic debris. These observations do not preclude the possibility that calcareous bodies may play some role as buffers or reservoirs of inorganic ions. However, it is difficult to accept such a function for unencapsulated worms in the coelom or intestinal lumen; we suggest that excretion is a more likely role in such sites.  相似文献

With the aim of identifying genes involved in development and parasite adaptation in cestodes, four coding sequences were isolated from the cyclophyllidean Mesocestoides corti larval stage (tetrathyridium). Genes showed significant similarity to the cysteine-rich secreted protein (CRISP) encoding genes, a large family that includes stage and tissue-specific genes from diverse organisms, many associated with crucial biological processes. The full-length McCrisp2 cDNA encodes a predicted protein of 202 residues in length, containing 10 cysteines and a putative signal peptide. The expression level of McCrisp2 was estimated by Real-time PCR, relative to GAPDH, showing an increase of 75% in segmented worms compared to tetrathyridia. By in situ hybridization, McCrisp2 expression was localized mainly at the larvae apical region of tetrathyridia and in the proglottids of segmented worms. Taken together our results suggest a possible role for M. corti CRISP proteins as ES products, potentially involved in differentiation processes as proposed for homologs in other organisms.  相似文献

Gekkotaenia novaeguineaensis n. gen., n. sp. (Cestoda: Anoplocephalidae) from the small intestine and Cosmocerca zugi n. sp. (Nematoda: Cosmocercidae) from the large intestine of the ring-tailed gecko, Cyrtodactylus louisiadensis (Sauria: Gekkonidae) are described and illustrated. Gekkotaenia novaeguineaensis n. gen., n. sp. is unique among the acraspedote Linstowiinae in possessing a poral female reproductory system. Cosmocerca zugi n. sp. is the 22nd species to be assigned to the genus and differs from other species in the genus by possessing 4 pairs of rosette papillae on plectanes and having a gubernaculum longer than the spicules. It is the fifth species to be described from the Australian biogeographical region. Eight additional helminth species were found: the digenean, Allopharynx macallisteri; 2 cestodes, cysticercoids of Cyclophyllidea gen. sp. and tetrathyridia of Mesocestoides sp.; 5 nematodes, larvae in cysts of Abbreviata sp., Aplectana macintoshii, Oswaldocruzia bakeri, Parapharyngodon maplestonei, and an undescribed species of Physalopteroides. Cyrtodactylus louisiadensis represents a new host record for each of these helminths.  相似文献

The number of polycephalic tetrathyridia present in 150-day-old intraperitoneal larval populations of Mesocestoides corti was markedly increased in mice gonadectomized 1 month prior to infection. This effect was more pronounced in male hosts. In both sexes it was inversely correlated with the size of the populations: the smaller the population, the larger was the number of polycephalic tetrathyridia. These forms are probably produced when the separation of daughter individuals lags behind the growth and organogenesis of these peculiar cestodes.  相似文献

1. Natural abundance carbon-13 nmr spectra of several intact cestodes have been obtained. 2. All spectra show peaks assignable to triglycerides and the N(CH3)3 carbons of the choline moiety. 3. The olefinic region of the 13C nmr spectra indicated that the cestode larvae Mesocestoides corti and Echinococcus multilocularis have a larger concentration of polyunsaturated fatty acids than Hymenolepis adults. 4. The tetrathyridia of Mesocestoides corti produce lactate, succinate, acetate, and CO2 as major carbon-containing end products during in vitro incubation with glucose as the substrate. Differences in the rate of glucose consumption and lactate production under anaerobic or aerobic conditions were observed, but their significance could not be determined. However, succinate production was greatly decreased in the presence of oxygen.The relative activities and intracellular distribution of various enzymes involved in energy-supplying metabolism of the larvae appear to conform to the pathways observed in other parasitic helminths known to produce lactate, succinate, and volatile fatty acids as metabolic end products. Some common features found in this respect are the relatively low pyruvate kinase activity, the presence of a highly active cytoplasmic phosphoenolpyruvate carboxylase and the capability of mitochondrial membrane bound fumarate reductase to reduce fumarate by means of NADH. Although a stimulatory effect of fructose-1,6-diphosphate on the reaction velocity of pyruvate kinase occurred, the absolute activity of this enzyme is very low.Nearly all the enzymes required for Krebs cycle activity are available in the tetrathyridia. Under the assay conditions employed by us, only NAD-dependent isocitrate dehydrogenase could not be demonstrated. The small amounts of 14CO2 liberated from 6-14C-glucose suggest that the cycle in its classical form probably only functions at a very low rate. The incorporation of 14C from labeled glucose into glycogen indicates the presence of enzymes capable of glycogenesis. The incorporation rate was found to be higher in the presence of oxygen than under anaerobic conditions. On account of the very low NAD-linked glycerol-3-phosphate dehydrogenase activity the glycerolphosphate cycle may be of minor importance for the tetrathyridia.As a result of these studies a scheme for the main carbohydrate dissimilating pathways in the tetrathyridia is proposed and the significance of oxygen with respect to energy-supplying metabolism is discussed.  相似文献

To study the effect of high temperature on infectivity of Toxoplasma gondii tissue cysts, pork from infected pigs was mixed with infected mouse brains and homogenized thoroughly. Twenty-gram samples of infected homogenized meat were sealed in plastic pouches, pressed to a uniform thickness of 2 mm, and subjected to water-bath temperatures of 49, 52, 55, 58, 61, 64, and 67 C for 0.01, 3, 6, 9, 12, 24, 48, and 96 min. Treated samples were digested in HCl-pepsin solution and bioassayed in mice. Toxoplasma gondii tissue cysts remained viable at 52 C for 9.5 min but not for 9.5 min at 58 C; tissue cysts were generally rendered nonviable by heating to 61 C or higher temperature for 3.6 min. Tissue cysts survived once at 64 C for 3 min. These data demonstrate that T. gondii tissue cysts are less heat resistant than encysted Trichinella spiralis larvae.  相似文献

Tetrathyridia of the cestode Mesocestoides corti were isolated from the peritoneal cavity of infected mice. The parasites activated guinea pig and mouse complement (C) in vitro by both the classical and alternative pathways as shown by quantitative C fixation and crossed immunoelectrophoresis. The ability of tetrathyridia to activate mouse C was enhanced by preincubating the parasites in serum obtained from mice infected with M. corti. Antibodies of the IgG1 class, an immunoglobulin found in profoundly increased amounts in mice infected with M. corti, as well as IgM and IgG2 antibodies, bound to cultured tetrathyridia and facilitated deposition of the third component of C (C3) from dilute mouse serum, presumably via classical pathway activation. The results demonstrate that mouse IgG1 antibodies do not prevent the activation of C by the tetrathyridia or by C-fixing antibodies of other classes which become attached to the tetrathyridia. The activation of C in vitro by tetrathyridia did not affect their ability to grow in mice, even though C3-derived polypeptides could be detected by immunofluorescence on the surface of the parasites.  相似文献

Mesocestoides vogae is a cestode of the order Cyclophyllidea, and its second intermediate hosts are mammals, birds, amphibians, and reptiles. The parasite forms a tetrathyridium in the second intermediate host and multiplies asexually, sometimes to the point of filling the host's abdominal cavity. Proliferated tetrathyridium may cause lethal conditions in the host. During the asexual multiplication period, the scolex first replicates into two and then divides into two worms. In this study, to investigate the factors that promote the replication, tetrathyridia were cultured under various in vitro conditions. When several worms which already had two scolexes were cultured together, the division into two worm bodies was complete, but when single worm was cultured, the division hardly proceeded. The result indicates that the division progression of tetrathyridia with two scolexes requires the presence of other worms. In contrast, tetrathyridia with only one scolex did not initiate the division process, whether cultured together or alone. Then, the necessity of direct contact between the bodies of the worms to promote the division of tetrathyridia with two scolexes was assessed. For this, the well of the culture plate was partitioned into upper and lower parts using a mesh, and 20 worms in the upper part and single worm in the lower part were cultured. In all examined wells, worms in the upper part showed complete division, whereas the worms in the lower part rarely completed the division. Thus, direct contact between tetrathyridium promotes the division of tetrathyridia bearing two scolexes.  相似文献

The gross and histological morphology of the larvae of 4 ascaridoid nematodes, Toxascaris leonina, Baylisascaris procyonis, Lagochilascaris sprenti, and Hexametra leidyi, are described. The larvae of T. leonina, B. procyonis, and L. sprenti were recovered from experimentally infected mice at 32, 14, and 75 days of infection, respectively. Hexametra leidyi larvae used for morphological study were collected on day 159 postinfection from a rhesus monkey, Macaca mulatta, while H. leidyi larvae used for histological study were collected from mice when they had reached the same size as those found in the monkey, i.e., at 23 days postinfection. Larvae for morphological study were collected by pepsin digestion, fixed in glacial acetic acid, and cleared in glycerin. Tissues for histological study were fixed in 10% formalin or Bouin's fixative. All sections were stained with hematoxylin and eosin. The differences in the morphology of the larvae of these 4 species were found to be length of the body, shape of the anterior end, and shape of the tail. The major differences in the histological anatomy of these larvae were found to be the body diameter, form of lateral alae when present, presence or absence of internal cuticular bars, shape and internal patterns of the excretory columns, and size and number of intestinal cells. These 4 larvae are differentiated from other described species of ascaridoid larvae that may cause visceral larva migrans, and keys have been devised to aid in the making of a diagnosis.  相似文献

Cestode larvae spend one phase of their two-phase life cycle in the viscera of rodents, but cases of cestodes infecting subterranean rodents have only been rarely observed. To experimentally gain some insight into this phenomenon, we captured approximately 300 plateau zokors(Eospalax baileyi), a typical subterranean rodent inhabiting the Qinghai-Tibet Plateau, and examined their livers for the presence of cysts. Totally, we collected five cysts, and using a mitochondrial gene(cox1) and two nuclear genes(pepck and pold) as genetic markers, we were able to analyze the taxonomy of the cysts. Both the maximum likelihood and Bayesian methods showed that the cysts share a monophyly with Taenia mustelae, while Kimura 2-parameter distances and number of different sites between our sequences and T. mustelae were far less than those found between the examined sequences and other Taeniidae species. These results, alongside supporting paraffin section histology, imply that the cysts found in plateau zokors can be regarded as larvae of T. mustelae, illustrating that zokors are a newly discovered intermediate host record of this parasite.  相似文献

Two hundred and one whiptail lizards, Cnemidophorus spp., from Texas and Colorado (USA), were examined for Mesocestoides sp. tetrathyridia. Eleven (5%) were infected, including three of 58 (5%) C. dixoni, six of 70 (9%) C. gularis septemvittatus, one of 35 (3%) C. marmoratus, and one of 34 (3%) C. tesselatus; four C. inornatus heptagrammus were not infected. In addition, 41 non-cnemidophorine lizards from the same study area were not infected. Free tetrathyridia were found in the body cavity of lizards and encapsulated tetrathyridia were observed in the heart, liver, stomach, mesenteries, ovaries, intestines, and lungs. None of the Mesocestoides sp. exhibited any evidence of asexual proliferation such as multiple scoleces or buds. This note, the fifth in a series of reports on helminths of Cnemidophorus spp., represents the first time Mesocestoides sp. has been reported from these four taxa, and Colorado is a new geographic locality record for this parasite.  相似文献

Volumes and weights of populations of Mesocestoides tetrathyridia increased faster in SEC mice treated with cortisone, than in the controls. Cortisone did not alter the sex resistance of the host and the populations always grew faster in male than in female hosts. Larvae of the populations from cortisone treated mice were smaller than those from control mice, and those from males smaller than those from females.The average size of the larvae decreased as the population size increaed. Fast growing populations contained a higher percentage of larvae with 2 suckers. These forms may serve as indicators of accelerated division of larvae.  相似文献

Described are the characteristics of the polycystic larval cestodes found in an endemic area of echinococcosis in the Easter Plains of Colombia and the tissue reaction evoked in infected rodents. Of 848 free-ranging animals examined, polycystic hydatids were found in 44/93 Cuniculus paca and 1/369 Proechimys sp. None of 20 Dasyprocta fuliginosa examined was infected, but hunters provided a heart with hydatid cysts and information about two additional animals with infected livers. Recognition of an endemic area of polycystic echinococcosis provides a means to investigate the life cycle of the parasites and to study the histogenesis of the larval cestodes in susceptible laboratory animals.  相似文献

Modifications to the alkaline Comet assay by using lesion-specific endonucleases, such as formamidopyrimidine-DNA glycosylase (FPG) and endonuclease III (ENDOIII, also known as Nth), can detect DNA bases with oxidative damage. This modified assay can be used to assess the genotoxic/carcinogenic potential of environmental chemicals. The goal of this study was to validate the ability of this modified assay to detect oxidative stress-induced genotoxicity in Drosophila melanogaster (Oregon R(+)). In this study, we used three well known chemical oxidative stress inducers: hydrogen peroxide (H(2)O(2)), cadmium chloride (CdCl(2)) and copper sulfate (CuSO(4)). Third instar larvae of D. melanogaster were fed various concentrations of the test chemicals (50-200μM) mixed with a standard Drosophila food for 24h. Alkaline Comet assays with and without the FPG and ENDOIII enzymes were performed with midgut cells that were isolated from the control and treated larvae. Our results show a concentration-dependent increase (p<0.05-0.001) in the migration of DNA from the treated larvae. ENDOIII treatment detected more oxidative DNA damage (specifically pyrimidine damage) in the H(2)O(2) exposed larvae compared to FPG or no enzyme treatment (buffer only). In contrast, FPG treatment detected more oxidative DNA damage (specifically purine damage) in CuSO(4) exposed larvae compared to ENDOIII. Although previously reported to be a potent genotoxic agent, CdCl(2) did not induce more oxidative DNA damage than the other test chemicals. Our results show that the modified alkaline Comet assay can be used to detect oxidative stress-induced DNA damage in D. melanogaster and thus may be applicable for in vivo genotoxic assessments of environmental chemicals.  相似文献

Evidence that damage decreases bone quality, increases fracture susceptibility, and serves as a remodeling stimulus motivates further study of what loading magnitudes induce damage in trabecular bone. In particular, whether damage occurs at the smaller strains characteristic of habitual, as opposed to traumatic, loading is not known. The overall goal of this study was to characterize damage accumulation in trabecular bone at small strains (0.20 - 0.45% strain). A continuum damage mechanics approach was taken whereby damage was quantified by changes in modulus and residual strain. Human vertebral specimens (n = 7) were tested in compression using a multi-cycle load - unload protocol in which the maximum applied strain for each cycle, epsilonmax, was increased incrementally from epsilonmax = 0.20% on the first loading cycle to epsilonmax = 0.45% on the last cycle. Modulus and residual strain were measured for each cycle. Both changes in modulus and residual strains commenced at small strains, beginning as early as 0.24 and 0.20% strain, respectively. Strong correlations between changes in modulus and residual strains were observed (r = 0.51 - 0.98). Fully nonlinear, high-resolution finite element analyses indicated that even at small apparent strains, tissue-level strains were sufficiently high to cause local yielding. These results demonstrate that damage in trabecular bone occurs at apparent strains less than half the apparent compressive yield strain reported previously for human vertebral trabecular bone. Further, these findings imply that, as a consequence of the highly porous trabecular structure, tissue yielding can initiate at very low apparent strains and that this local failure has detectable and negative consequences on the apparent mechanical properties of trabecular bone.  相似文献

Two populations, Duddingston and Feltwell, of potato cyst-nematode were inbred for seven and six generations respectively. The Duddingston population maintained a high fecundity in some lines for three generations, but in subsequent generations the fecundity as measured by cyst production declined in spite of selection for high fecundity. By selecting for fecundity, some lines of the Feltwell population still produced many cysts after five generations of inbreeding. In the Duddingston population it was possible to increase the number of cysts produced by an inbred line of poor fecundity by using paired cysts plus selection for fecundity. A similar change may have been obtained for unprolific lines of the Feltwell population. Selection for production of few cysts quickly produced lines with poor fecundity. One cause of poor fecundity was the small number of larvae in cysts used for inoculations. Yellow eggs were found in large numbers in some cysts but these were not a cause of loss of fecundity following repeated inbreeding.  相似文献

In a search for potential biocontrol agents for Acacia melanoxylon R. Br. (Mimosaceae), larvae of the beetle Diplocoelus dilataticollis Lea (Coleoptera; Biphyllidae) were found within damaged seeds of A. melanoxylon. The gut contents of larvae and adults were examined to determine whether their diet included seeds, in apparent contradiction to the known mycophagous diet of members of this family of beetles. Calcofluor M2R White, a plant cell-wall staining optical brightener was used to differentiate between plant cell fragments and fungal tissue in the gut content smears. Gut contents of adults of a known seed predator of A. melanoxylon, a weevil of the genus Melanterius, were examined in the same way to provide a benchmark. The gut contents of D. dilataticollis differed from those of Melanterius sp. Fungal structures and microbes were found in the gut of D. dilataticollis, in contrast to plant cell fragments found in the gut of the weevil and from scrapes made directly from seeds. We conclude that larvae of D. dilataticollis feed primarily on fungi associated with damaged seed and therefore may not be the proximate cause of seed damage.  相似文献
