In this study, we use barcoded pyrosequencing of the 16S rRNA gene to characterize the fecal microbiota of neonatal calves and identify possible relationships of certain microbiota profiles with health and weight gain. Fecal samples were obtained weekly from 61 calves from birth until weaning (seventh week of the calves'' life). Firmicutes was the most prevalent phylum, with a prevalence ranging from 63.84% to 81.90%, followed by Bacteroidetes (8.36% to 23.93%), Proteobacteria (3.72% to 9.75%), Fusobacteria (0.76% to 5.67%), and Actinobacteria (1.02% to 2.35%). Chao1 index gradually increased from the first to the seventh postnatal week. Chao1 index was lower during the third, fourth, and fifth week of life in calves that suffered from pneumonia and were treated with antibiotics. Diarrhea incidence during the first four weeks of the calves'' life was also associated with a reduction of microbial diversity during the third week of life. Increased fecal microbial diversity after the second week of life was associated with higher weight gain. Using discriminant analysis we were able to show differences in the microbiota profiles between different weeks of life, between high and low weight gain groups of calves, and between calves affected and not affected with diarrhea during the first four weeks life. The prevalence of Faecalibacterium spp. in the first week of life was associated with weight gain and the incidence of diarrhea, with higher prevalence being associated with higher weight gain and less diarrhea. Representative sequences from Faecalibacterium spp. were closely affiliated to Faecalibacterium prausnitzii. Results presented here provide new information regarding the intestinal microbiota of neonatal calves and its association with health and growth. Fecal microbial diversity was associated with calf age, disease status and growth rates. Results suggesting a possible beneficial effect of Faecalibacterium spp. on health and growth are promising.  相似文献

The ecology of Escherichia coli in calves reared as dairy-cow replacements   总被引:6,自引:4,他引:2  
A continual turn-over in the strains forming the majority of faecal Escherichia coli flora was demonstrated in 16 calves reared as dairy cow replacements. The incidence of antibiotic resistance among isolates, as measured by an Antibiotic Resistance Index (ARI), changed markedly with the age of the calf. The value was low initially, when the calves were 1–2 days old and housed with adult animals. It then rose rapidly during the first week after the animals had been weaned and moved into nursery pens. This change in ARI was associated with the isolation of strains resistant to four or five of the six drugs included in the sensitivity test. The ARI then fell from the third week to low levels by the time that the calves were five months of age. This fall was due to the isolation of an increasing proportion of sensitive E. coli strains. These differed from the sensitive strains which had colonized the calves in the early days of life so demonstrating that the change was not due to the reemergence of strains identified several weeks previously. The source of E. coli strains was presumed to be the calfs' environment but further investigations are required to prove this conclusively.  相似文献

The aetiology of abortions and calf mortality in 65 Danish cattle herds consisting of both dairy and beef breeds during a 1-year period is described. All observed aborted foetuses, stillborn calves, and calves dying before 6 months of age were necropsied, and relevant microbiological examinations were performed. A total of 240 calves and 66 abortions were submitted corresponding to a calf mortality rate of 7%. The abortion frequency could not be calculated. 43% of the calves died at day 0, while 22% were aborted, 15% died during the first week of life, 9% died from 1 to 4 weeks of age, and 11% died at the age of 1 to 6 months. The most common cause was neonatal pulmonic atelectasis (stillbirth) followed by foetal infections, pneumonia, and septicaemia.  相似文献

Failed transfer of passive immunity (FTPI) in dairy calves – which is often due to the low amount of colostrum provided within a few hours after birth – remains a crucial issue. Enabling dairy calves to nurse colostrum from their dams could be useful in increasing intake and thus avoiding FTPI, but further potential effects on the health and welfare of both calves and dams should also be considered. In this study, 107 calf-dam pairs from two Italian dairy farms were alternately assigned to one of the following colostrum provision methods (CPMs): ‘hand-fed method’ (HFM) – the calf was separated from the dam immediately after birth and colostrum was provided by nipple-bottle (n = 50); ‘nursing method’ (NM) – the calf nursed colostrum from the dam for the first 12 h of life without farmer assistance (n = 30); and ‘mixed method’ (MM) – the nursing calf received a supplementary colostrum meal by nipple-bottle (n = 27). Serum of calves (1 to 5 days of age) and samples of their first colostrum meal were analysed by electrophoresis to assess immunoglobulin (Ig) concentration. Additionally, behavioural indicators of separation distress (calf and dam vocalisations; calf refusal of the first meal after separation; undesirable dam behaviour at milking) in the following 24 h were recorded as binary variables (Yes/No), and the health status of calves (disease occurrence and mortality) and dams (postpartum disorders and mastitis occurrence) were monitored for the first 3 months of life and 7 days after parturition, respectively. The lowest FTPI occurrence (calf serum Ig concentration <10.0 g/l) was found in the MM (11.1%) and the HFM (22.0%) compared with the NM (60.0%) (P<0.05), and the highest percentage of calves with optimal transfer of passive immunity (serum Ig concentration ≥16.0 g/l) was observed in the MM (55.6%). The lowest calf–dam separation distress was observed in the HFM (P<0.05). The highest calf disease occurrence was recorded in the HFM (64.0%) and the lowest in the NM (33.3%), with an intermediate value for the MM (44.4%) (P<0.05). No effect of the CPM was observed on dam health or calf mortality (P>0.05). The results of this study indicated that providing calves with a supplementary colostrum meal in addition to nursing from the dam (MM) is truly effective in maximizing passive immunity transfer. Anyway, specific strategies should be studied to minimise calf-dam separation distress.  相似文献

Improving immune status in neonates is crucial to health and production. Gut active carbohydrates (GAC) have been associated with increasing immunoglobin levels and immonucompetence development in mammals. The objective of the following studies was to evaluate whether GAC (mannan-oligosaccharides) applied orally to progeny immediately following parturition, improved blood plasma immunoglobulin (Ig) type G concentrations in piglets and calves. Three trials were conducted comparing control groups with those receiving GAC orally. The first two trials used piglets that were monitored for blood IgG at 2 days of age and for changes in body weight (BW), and the third trial monitored calf IgG from birth to 21 days of age. Piglets in the experimental group received 0.75 g GAC in 10 ml saline at birth and 24 h of age. The calf trial compared the control group against calves that received 22.5 g GAC mixed into 4.5 l of colostrum (to give 5 g/l) in the first 24 h after parturition. Blood serum samples were taken at 2 days post partum in piglets, and at several time points from 6 h to 21 days of age in calves, and were analysed for IgG levels by radial immunodiffusion. In the first piglet trial, significantly higher levels (32%) of IgG were observed for piglets fed GAC (P < 0.001), and in the second, IgG concentration was elevated by 23% (P < 0.01) and BW increased by 9% (P = 0.023) with GAC supplementation. Significant improvements for calves were recorded at all time points in those fed GAC (P < 0.05), with an increase in serum IgG observed after the first day, which was maintained throughout the sampling period, resulting in a difference of 39% at the end of the trial (21 d). These findings form a basis for further studies, which are required to investigate possible modes of action involved in enhancing blood immunoglobulin concentrations in young animals, and the longer-term effects this may have on the development of the immune response.  相似文献

We examined spleen cells from newborn to 1-month-old rabbits for easily detectable surface immunoglobulin, complement receptors, and for in vitro proliferative responsiveness to anti-immunoglobulin antisera and several mitogens. From birth through the first month of life about 15% of the cells from rabbit spleens had easily detectable surface immunoglobulin while about 45% had C3 receptors. In adults as many as 77% of the spleen cells had easily detectable surface Ig but the proportion with C3 receptors remained about 45%. The proliferative response to anti-allotype antisera was present at birth, and was at adult levels by 1 month of age. The proliferative response to pokeweed mitogen was low when cells were obtained during the first week of life but was comparable in magnitude to the response of adult cells by 2 weeks of age. In vitro responsiveness to concanavalin A was present at low levels at birth and increased sharply during the first week. We did not observe significant stimulation of spleen cells from neonatal to 4-week-old rabbits by lipopolysaccharide from Salmonella typhosa. Our data suggest that lymphocyte surface markers and functional responses appear asynchronously in spleen cells of developing rabbits.  相似文献

The aim of this study was to determine the influence of age and ingested food (colostrum and mature milk) on the concentrations of selected blood biochemical components connected with nitrogen and mineral metabolism in dairy calves during their first week of life. The experiment was carried out on 13 Polish Black and White breed dairy calves. The animals were fed colostrum within the first 3 days of postnatal life and thereafter the mature milk of their dams until the end ofthe experiment (7 days). The obtained results showed that intensive catabolic and anabolic changes in nitrogen occur in the first week of life. These changes were particularly intense during the first 24-48 hours of life and may reflect dynamic tissue remodelling. The results of this experiment also show that healthy calves efficiently regulate water and electrolyte homeostasis.  相似文献

Age-related changes in testosterone, progesterone and estradiol 17-beta were determined by radioimmunoassay (RIA) in the serum of 155 male buffalo calves of varying ages. The calves were classified into 17 age groups. The mean weight of calves increased from 33.6 +/- 9.6 kg at one week of age to 531 +/- 41.4 kg at 42 months. The testosterone levels were less than 100 pg/ml from birth until 15 months of age, followed by peak concentrations of 422 +/- 79 pg/ml at 24 to 30 months and 793 +/- 193 pg/ml at 42 to 48 months (corresponding to puberty and maturity, respectively). The progesterone levels were higher in newly born calves and mature bulls. Otherwise, the levels continued to be low throughout the period of growth and development. Estradiol 17-beta was significantly higher in postnatal calves up to two months of age. The testosterone revealed a positive correlation with weight and age while E2 17-beta showed a negative correlation with age. These results do not support a direct role of peripheral progesterone and estradiol 17-beta in the onset of puberty and sexual maturity of buffalo bulls.  相似文献

Because impala are commonly exhibited and handreared in zoos and their natural nursing behavior had not previously been studied, we examined nursing and early development in five impala calves housed in a large, naturalistic enclosure. Calves were observed for the first 5 weeks of life during 12-hr continuous watches and 20-min focal animal samples. Total daily suckling time decreased while time grazing and feeding on concentrates increased from 1–5 weeks of age. An increasing proportion of nursing bouts were terminated by the dam as calves matured, with dams terminating almost 70% of bouts during the first week. Suckling success and maternal grooming also decreased after week I, suggesting that impala mothers cut back early on nursing and grooming of offspring. Rapid decline in mother-young spatial proximity and a concomitant increase in calf association with age-mates over time suggests that the mother-young bond is weak and ephemeral in impala. An example of the implications of our results for improving handrearing programs would be that newborn calves should be started on a daylight feeding schedule of one bottle-feeding every three hours, and that they not be allowed to gorge themselves at any one of these feedings.  相似文献

The birth of Sumatran rhino calves at the Cincinnati Zoo and Botanical Garden offered a unique opportunity to study early development and cow–calf behavior in this elusive, critically endangered species. Study goals were to: (1) record developmental milestones of newborn calves; (2) characterize cow and calf behavior during the first year following birth; (3) compare trends in the behavioral development of a male vs. female calf; and (4) record weight gain and footprint size throughout the first year. In addition to recording eight neonatal milestones, more than 450 hr of behavioral data were collected on each of the two calves (one male and one female) and their dam during the first 12 months of life. Neonatal milestones were achieved within 24 hr of birth with the exception of first defecation, which occurred at 16–18 days. Although nursing bouts decreased slightly in the second half of the year (from once every 90 min to once every 2 hr), they continued to occur frequently throughout the day and night. Therefore, calves grew rapidly from approximately 33 kg at birth to 400 kg at 12 months. Average daily weight gain for the first week was approximately 2.0 kg, whereas average daily weight gain for the remaining 12 months was slight (<1 kg) and did not differ between calves. Eating and resting occupied 70–80% of the Sumatran rhino cow and calves' time and no gender biases in either maternal investment or developmental parameters were noted. A survey of the mortality and morbidity affecting calves during the first 3 months of life was carried out. Results are reported from 131 herds with altogether 5,050 calvings. These herds kept individual calf cards for all calves born. The incidences of abortions and stillborn calves were 0.6% and 3.6%, respectively. During the period 0-90 days the mortality and morbidity were 2.6% and 11.0%, respectively. The frequencies of enteritis and pneumonia in calves were 7.2% and 0.8%. Mortality and morbidity were influenced by breed, season, age of the dam and time of first colostrum. The frequencies of abortions, stillborn calves, mortality and morbidity varied considerably between herds. Herd factors influencing mortality and morbidity during different periods of time were: herd size, yield, zero-grazing, whether the calf was allowed to feed by suckling, the design of the calf pens, and previous incidence of infectious enteritis in the herd. On average, both mortality and morbidity in calves were low, though in certain herds, temporarily high frequencies were registered.  相似文献

Cows fed 0.37 Kg crude protein per day for four months before calving showed no decrease in colostral IgG, IgA, or IgM from cows fed a normal ration. Similarly, no difference in serum immunoglobulin concentration was detected in their calves. Cows in the low protein group developed significantly lower (P < 0.01) titers to Salmonella pullorum antigen than did the normal group. The data show significant positive linear regressions of calf agglutination titer to S. pullorum on the dam's colostral titers. Serum levels of IgG measured at 24 hours were significantly lower in calves from second-calf heifers than from these of other age groups. The IgA levels of serums from calves from the six-year-old cows were significantly higher than the serum levels of calves from younger dams (P < 0.01).  相似文献

In the investigations on five calves with fistulae in the abomasum and oesophagus the secretion of gastric juice as well as its content of proteolytic enzymes and hydrogen ions were measured. It was found that the gastric juice of calves possessed a fairly high proteolytic activity (about 6 UP/l) and hydrogen ion level already on the 3rd day of life. Sham feeding produced two different types of gastric secretion response, depending on the age of calves. In the first week of life sham feeding caused an inhibition of secretion which became more intensive only about 30 min following food administration. In the second week of life gastric juice secretion increased immediately after feeding.  相似文献

A better knowledge of chick survival rates is required to enable understanding of the population dynamics of gamebirds and to develop management measures to conserve their populations. The Red‐legged Partridge Alectoris rufa is a highly valued game species in Spain but its populations have declined in recent decades. A lack of appropriate monitoring methods has been a limitation in gaining information on the mortality of Red‐legged Partridge chicks. We developed methods for the effective radiotagging of chicks in captivity and applied these methods in the field in northern Spain to estimate survival during the first 5 months of life. The most effective method for radiotagging captive chicks between 3 and 8 days old involved gluing small tags directly to the skin in the interscapular space using cyanoacrylate adhesive. Backpack harness tags attached with elastic bands were the most effective method of radiotagging 4‐week‐old chicks. Predation was the main cause of chick mortality identified during the field experiments. Survival between hatching and 5 months of age was estimated to be 16–21%. The lowest survival rates occurred during the first 7 days of life (62–70% cumulative survival) and this period seems to be a major determinant in the life history of the species.  相似文献

The oxidative stress of birth in cattle (Bos taurus) was evaluated by measuring steady state concentration of free radicals in whole blood, rate of lipid peroxidation and activity of antioxidant enzymes in erythrocytes, antioxidant capacity of blood plasma in 14 calves at birth and four times after birth until 3 weeks of age and also in their mothers at calving. The same parameters were also measured in 58 dairy cows before calving, at parturition and after calving. Free radical concentration in the blood of newborn calves was higher than in cows confirming that birth means oxidative stress for calves. Red blood cell malondialdehyde in calves was the highest at birth and following the first solid feed intake at the third week. Superoxide dismutase activity increased in calves during the first three weeks of life. Ferric reducing ability of plasma was higher in calves at birth than in cows and decreased thereafter. Higher superoxide dismutase activity in red blood cells and lower ferric reducing ability of plasma in dairy cows was found at calving compared to the average of all pre- and post-calving results. We conclude that the blood of newborn calves is well prepared to deal with the oxidative stress of birth, and that such a stress is present even when some fingerprint markers of redox imbalance show no apparent alterations. Stress of calving has minor effects on the antioxidant system of cows.  相似文献

Colostrum ingestion by neonatal calves is widely recognized to provide passive transfer of immunity. In this study immunoglobulin absorption from colostrum was evaluated in 54 IVF-produced calves. The IVF calves were delivered by Cesarean section on Days 275 to 277 of gestation, 24 h after the dams had been administered 30 mg dexamethasone. The calves suckled bottles or were force-fed 6 L of colostrum in the first 12 h of life. Colostrum was obtained from the first post-calving milking of recipient dams or from frozen storage reserves if dam secretion was not adequate. Immunoglobulin type G (IgG) content of both sources of colostrum was determined. Serum samples from the calves were collected at 0, 12 and 24 h of age and analyzed for IgG. Twenty dairy calves born vaginally served as the controls and were subjected to the same colostrum management protocol except that the colostrum was obtained only from frozen post-calving milk of dairy cows from the same farm. The control calves were also subjected to the same sampling protocol. The IVF group of calves ingested more IgG (P < 0.0001) and absorbed more IgG by 24 h of age (P < 0.0001) than their control group counterparts. Absorption of IgG was analyzed by comparing the g/kg body weight of IgG with serum IgG values at corresponding times after birth. Colostrum absorption efficiency was the same for both IVF and control groups of calves at 12 and 24 h of age. There was a maximum IgG dose above which additional increases in serum IgG were not realized. The slightly premature, Cesarean delivered IVF calves absorbed IgG from colostrum similarly to control calves delivered vaginally.  相似文献

The hydrolysis of triglycerides and cholesteryl esters stored within cells is mediated by the enzyme, hormone-sensitive lipase. In adipose tissue and heart, hormone-sensitive lipase primarily hydrolyzes stored triglycerides to free fatty acids, while in steroidogenic tissues, it principally converts cholesteryl esters to free cholesterol for steroid hormone production. To determine whether hormone-sensitive lipase is under tissue-specific, developmental regulation, the steady state levels of hormone-sensitive lipase mRNA were determined in normal rats from late fetal life through 2 years of age. Hormone-sensitive lipase mRNA levels did not appear to vary in adipose tissue from epididymal fat pads obtained from animals between 3 weeks and 2 years of age. In heart, hormone-sensitive lipase mRNA levels were lowest in the fetus increased rapidly within the first day postnatally, and then gradually increased to stable adult levels by 2 months that were 3-fold higher than observed in fetal rats. Steady state mRNA levels of hormone-sensitive lipase in the adrenals were lowest in fetal rats, increased 4-fold during the first day and peaked at levels that were 9-fold higher by the end of the first week. Thereafter, levels fell and remained 3- to 4-fold higher than at birth throughout adult life. Hormone-sensitive lipase mRNA was undetectable in testes before 4 weeks of age and increased 25-fold to stable adult levels between 4 and 12 weeks. Thus, hormone-sensitive lipase is differentially expressed and regulated in a tissue-specific fashion during development and aging.  相似文献

LH and testosterone levels in bull calves were studied in the plasma samples collected sequentially at 15-min intervals every month during the first year of life. An episodic pattern of LH release occurred after birth and the frequency and magnitude of the LH peaks increased up to 4 months of age and decreased thereafter. A testicular response was not observed before this age. It is suggested that this episodic LH activity is responsible for the testicular development which then initiates puberty.  相似文献

The recent trend in the dairy industry towards elevated planes of milk feeding of young calves requires reconsideration of calf milk replacer (CMR) formulations. The fat:lactose ratio in CMR is typically lower than that of whole milk and effects of increasing fat inclusion at the expense of lactose in CMR on nutrient metabolism and gut function of rearing calves are not sufficiently understood. Therefore, the current study aimed to determine the effect of increasing replacement of lactose by fat on growth performance, nutrient digestibility and metabolism, and glucose/insulin sensitivity. A total of 40 male calves (1.7 ± 0.10 days of age, 46.7 ± 0.76 kg BW) were blocked based on arrival date and randomly assigned to one of four treatments containing differing levels of fat and lactose (18F: 18.8% and 47.6%; 22F: 22.3% and 42.8%; 26F: 26.0% and 38.6%; 30F: 30.1% and 33.8%, fat and lactose DM, respectively). Calves were individually housed for the duration of the 11 week study and received their CMR (150 g/L) twice a day (0700 and 1600 h) from a teat bucket. The CMR feeding schedule consisted of 6 L/d from d 2 to 14, 7 L/d from d 15 to 56, and then 4 L/d during gradual weaning from 56 to 63 days. Pelleted starter, chopped straw, and water were available ad libitum throughout the study. Measurements included feed intake, growth, nutrient digestibility, fecal composition, and blood parameters. A glucose tolerance test (GTT) was performed between 28 and 32 days of age. By design, metabolizable energy intake from CMR increased linearly with fat level, but this did not result in a difference in BW, ADG, or concentrate intake. Fecal composition remained unaffected by treatment except for higher fat content in 22F compared to 26F. Also, plasma non-esterified fatty acids and total bilirubin differences were limited to 22F having the highest values whereas 26F had the lowest values. Regarding the GTT, total area under the curve (AUC) for glucose was highest in 22F and 26F and lowest in 18F whereas the AUC between 30 and 60 min for glucose was highest in 26F and lowest in 18F and 22F. Overall, altering the lactose:fat ratio in CMR did not affect growth performance while having minor effects on nutrient metabolism, but future investigation should focus on physiological consequences and effects in adult life to understand biological impact of fat and lactose levels in CMR for rearing calves.  相似文献

An analysis of resource gain is provided for Ammotragus calves during their first year of age. Both grazing and suckling rates are analysed and potential effects caused by age, sex, mother's rank and weaning period studied. During their first day of life, calves devote their time mainly to suckling, grazing starting at the second week of age and exceeding suckling rate from the fourth week onwards. When suckling rate reaches a significant decrease (at 2 mo), male calves show a significantly higher grazing rate than female calves, suggesting a higher energetic demand for males at this age. When calves are 1 mo old, sons of high-ranking mothers devote less time grazing than those of low-ranking ones, which is in accordance with the higher maternal investment they receive. Grazing rate shows distinctive values for the different weaning periods distinguished in this species, but the resumption of mothers' sexual activity, when a mother–offspring conflict occurs, does not affect grazing behaviour. These results are consistent with the biased maternal investment recently found in the same study population but focusing on another aspect, i.e. grazing behaviour, which is determined by calves' own decisions.  相似文献
