Enterococci are widespread bacteria forming the third largest genus among lactic acid bacteria. Some possess probiotic properties or they can produce beneficial proteinaceous antimicrobial substances called enterocins. On the other hand, some enterococci produce biogenic amines (BAs), so this study is focused on the sensitivity to enterocins of biogenic amine-producing faecal enterococci from ostriches and pheasants. Altogether, 60 enterococci isolated from faeces of ostriches and pheasants were tested for production of BAs. This target of the identified enterococci involved 46 strains selected from 140 ostriches and 17 from 60 pheasants involving the species Enterococcus hirae, E. faecium, E. faecalis, and E. mundtii. Although BAs histamine, cadaverine, putrescine, and tryptamine were not detected in the enterococci tested, in general high BA production by the tested enterococci was noted. The species E. hirae formed the majority of the enterococcal strains from ostrichs faeces (34 strains). High production of tyramine (TYM) was measured with an average amount of 958.16 ± 28.18 mg/ml. Among the enterococci from pheasants, the highest was production of TYM compared to phenylethylamine, spermidine, and spermine. Enterococci featured high BA production; however, they were sensitive to seven enterocins with inhibition activity ranging from 100 up to 25,600 AU/ml.  相似文献

Jay, James M. (Wayne State University, Detroit, Mich.). Production of lysozyme by staphylococci and its correlation with three other extracellular substances. J. Bacteriol. 91:1804-1810. 1966.-Lysozyme production was determined on plates containing 1 mg/ml of Lysozyme Substrate in Heart Infusion Agar with incubation at 37 C for 48 hr. Its production was compared with that of alpha-hemolysin and sheep hemolysin and egg-yolk precipitation, by use of both coagulase-positive and coagulase-negative strains of staphylococci. Of 126 coagulase-positive strains tested, 120 or 95.2% produced lysozyme, 117 or 92.9% produced alpha-hemolysin, 108 or 85.7% precipitated egg yolk, and 102 or 81% produced sheep hemolysin. Of the 49 coagulase-negative strains (which included 22 pathogens), only 4 or 8.1% produced lysozyme, 14 or 28.6% produced alpha-hemolysin, 13 or 26.5% produced sheep hemolysins, and 5 or 10.2% precipitated egg yolk. Only two of the six coagulase-positive strains which failed to produce lysozyme showed any consistent patterns in relation to the four characteristics determined. The four coagulase-negative strains which produced lysozyme were inconsistent for the other characteristics measured. It is suggested that lysozyme production is more a property of coagulase-positive staphylococci, and therefore a better ancillary test of pathogenicity, than either production of alpha-hemolysin or egg-yolk precipitation, because the incidence of lysozyme producers is higher among this group than among those producing the other substances and because fewer coagulase-negative staphylococci produced lysozyme than hemolysins or egg-yolk precipitation. Of 16 other species of bacteria and yeasts tested, all were found negative except Bacillus subtilis. Lysozyme production by staphylococci in heavily contaminated foods was not inhibited on plates containing sodium azide, whereas media containing 7.5% salt and sorbic acid were unsuitable. The possible relationship of lysozyme production to staphylococcal pathogenicity is discussed.  相似文献

Abstract: Ring-necked pheasants (Phasianus colchicus) are able to store dietary calcium as medullary bone, which they may mobilize for future eggshell synthesis. We define this mechanism as calcium-loading. Previous experiments on pheasants conducted to document the importance of calcium in limiting distribution did not account for calcium-loading. We hypothesized that calcium-loading could override experimental calcium treatments of the diet. We measured egg production, egg characteristics, and femoral mineral content for pheasants that were not calcium-loaded on 7 diets differing in calcium from 0.2% to 4.5% and compared these results to a similar study on calcium-loaded pheasants. We predicted that calcium-loaded pheasants would produce more eggs than those that were not calcium-loaded. We also predicted that there would be no significant difference between femur ash fractions in non-calcium-loaded pheasants, but that the ash fraction in calcium-loaded pheasants would differ significantly between the beginning and end of the experiment. Egg production was higher in calcium-loaded pheasants above 2% dietary calcium. Femur ash fraction was not different in non–calcium-loaded pheasants but differed significantly before and after the experiment and between high (>2%) and low (<2%) dietary levels in calcium-loaded pheasants. Calcium-loading may account for short-term persistence of captive pheasants introduced on calcium-poor soils, followed by their eventual population failure. Managers may improve survival of captive pheasants before introduction by surveying habitat for adequate calcium and by calcium-loading.  相似文献

A total of 195 Gram positive, catalase positive cocci, isolated from ovine mastitis, abscesses in slaughtered animals and parasitic pulmonary lesions in lambs were tested for glucose fermentation, anaerobic growth in thioglycollate medium, coagulase production and susceptibility to the lytic action of lysostaphin. On the basis of lysostaphin sensitivity, 192 strains were classified as staphylococci. The number of cultures able to produce acid anaerobically from glucose, or giving a positive result in the test of Evans and Kloos was lower. A good correlation among these three tests was not observed. Ninety-seven of the strains tested gave a positive coagulase reaction. Sensitivity to lysostaphin could not be used as a criterion for the differentiation of coagulase positive and coagulase negative strains. The turbidimetric method employed for the assessment of lysostaphin sensitivity is discussed.  相似文献

Our current knowledge of microbiota in wild ruminants is limited. The goal of this study was to evaluate staphylococcal species in red and roe deer for various attributes (haemolysis, DNase, and urease activities; lactic acid and biofilm production; and antibiotic profile) and their susceptibility to gallidermin. Sixteen staphylococcal strains were identified from faeces of 21 free-living animals (9 adult female Cervus elaphus—red deer and 12 young female Capreolus capreolus—roe deer) sampled by the Polish colleagues in the Strzałowo Forest District, Piska Primaeval Forest. The variability in the species of staphylococci was determined. Seven species (Staphylococcus capitis, S. epidermidis, S. haemolyticus, S. hominis, S. pseudintermedius, S. vitulinus and S. warneri) and five clusters/groups of coagulase-negative staphylococci (CoNS) were identified. The strains were generally not haemolytic and Dnase negative; did not form biofilms or only produced low-grade biofilms; exhibited high levels of lactic acid; were urease positive; and were generally susceptible to antibiotics (only two strains were resistant to multiple antibiotics). However, all of the strains were susceptible to the lantibiotic bacteriocin gallidermin, with a minimal inhibitory concentration of 0.0156 μg (up to 6400 AU/ml in arbitrary units). This is the first study to perform a detailed study of the properties of CoNS from roe and red deer.

A total of 504 clinical isolates of the family Micrococcaceae were tested for coagulase, deoxyribonuclease, clumping factor, and phosphatase to determine whether there is a correlation between the results of these tests and the pathogenicity of staphylococci. In the tests for coagulase production, it was found that either human or rabbit plasma could be used with broth cultures, whereas rabbit but not human plasma was satisfactory when microorganisms removed from solid culture medium were used. Deoxyribonuclease production correlated better than the fermentation of mannitol with coagulase production. The use of methyl green, Toluidine Blue O, or acridine orange offered no advantage over the use of HCl for detecting the production of deoxyribonuclease. Neither the presence of clumping factor nor the production of phosphatase correlated well with coagulase production. Strains of staphylococci that did not produce coagulase and deoxyribonuclease were isolated as frequently as, and from a greater variety of clinical sources than, strains which produced these substances. It is concluded that the production of coagulase and deoxyribonuclease are properties of staphylococci which are not necessarily indicative of potential pathogenicity of the organisms for man.  相似文献

【背景】魏斯氏菌广泛存在于发酵食品中,它们与食品发酵进程和风味物质的形成密切相关。酱油发酵过程酱醪中细菌的优势菌属是魏斯氏菌,研究魏斯氏菌的生理和代谢特性对于揭示菌株对环境的适应性和与酱油发酵相关的功能具有重要意义。【目的】从酱油酱醪中分离获得魏斯氏菌属中主要种的菌株,研究它们在酱油发酵过程的数量变化以及菌株的生理和生化特性,阐明菌株对酱油发酵体系的适应性和与酱油发酵相关的特性。【方法】通过菌株绝对数量的定量分析和耐受性比较,以及考察高盐条件下魏斯氏菌合成短链脂肪酸、胞外多糖、生物胺和氨基甲酸乙酯或其前体等特性,研究各类魏斯氏菌对酱油发酵和其安全性的影响。【结果】从高盐稀态酱油的酱醪中共分离得到16株魏斯氏菌,分别属于融合魏斯氏菌(Weissella confusa)、类肠膜魏斯氏菌(Weissellaparamesenteroides)和食窦魏斯氏菌(Weissellacibaria)。其中类肠膜魏斯氏菌可耐受高盐条件,是酱醪中魏斯氏菌属的主要菌种。它们合成短链脂肪酸的能力高于融合魏斯氏菌和食窦魏斯氏菌。酱醪来源的魏斯氏菌合成氨(胺)类危害物的特性区别较大,类肠膜魏斯氏菌的部分菌株产生物胺并可利用精氨酸积累瓜氨酸,食窦魏斯氏菌则能够降解多种生物胺。【结论】揭示了酱醪中主要魏斯氏菌的耐盐特性、在较低温度下生长情况和物质代谢规律,对于阐明魏斯氏菌与酱油发酵相关的功能和特性以及对酱油加工过程安全控制具有重要意义。  相似文献

Ostriches (Struthio camelus) achieve digesta retention times, digesta particle size reduction and digestibilities equal to similar-sized herbivorous mammals, in contrast to some other avian herbivores. The sequence of digestive processes in their gastrointestinal tract, however, is still unexplored. Using two groups of four ostriches (mean body mass 75.1 ± 17.3 kg) kept on fresh alfalfa, we tested the effect of two intake levels (17 and 42 g dry matter kg(-0.75)d(-1)) on the mean retention time (MRT) of a solute and three different-sized (2, 10, 20 mm) particle markers, mean faecal particle size (MPS), and digestibility. Intake level did not affect MRT, but MPS (0.74 vs. 1.52 mm) and dry matter digestibility (81 vs. 78%). The solute marker (MRT 22-26 h) was excreted faster than the particle markers; there was no difference in the MRT of 10 and 20 mm particles (MRT 28-32 h), but 2mm particles were retained longer (MRT 39-40 h). Because the solute marker was not selectively retained, and wet-sieving of gut contents of slaughtered animals did not indicate smaller particles in the caeca, the long MRT of small particles is interpreted as intermittent excretion from the gizzard, potentially due to entrapment in small grit. The marker excretion pattern also showed intermittent peaks for all markers in five of the animals, which indicates non-continuous outflow from the gizzard. When adding our data to literature data on avian herbivores, a dichotomy is evident, with ostrich and hoatzin (Opisthocomus hoazin) displaying long MRTs, high digestibilities, and gut capacities similar to mammalian herbivores, and other avian herbivores such as grouse, geese or emus with shorter MRTs, lower fibre digestibilities and lower gut capacities. In the available data for all avian herbivores where food intake and MRTs were measured, this dichotomy and food intake level, but not body mass, was related to MRT, adding to the evidence that body mass itself may not be sole major determinant of digestive physiology. The most striking difference between mammalian and avian herbivores from the literature is the fundamentally lower methane production measured in the very few studies in birds including ostriches, which appears to be at the level of reptiles, in spite of general food intake levels of a magnitude as in mammals. Further studies in ostriches and other avian herbivores are required to understand the differences in digestive mechanisms between avian and mammalian herbivores.  相似文献

Production of inhibitory substances by strains of the Streptococcus mutans group is well documented, but the nature of the substances implied is often unknown. Of nine laboratory strains known to produce inhibitory substances, the optimal conditions for producing inhibition zones on solid media were found to vary between strains but good production was generally obtained on all-purpose media with Tween 80 at 37 degrees C after 2-4 days of aerobic incubation. Streptococcus sanguis Ny101 was found to be more sensitive than Streptococcus rattus LG-1 to all inhibitory substances produced by the S. mutans strains tested. While all strains showed some inhibition, only six showed inhibition after neutralization; arginine incorporated in agar at 0.75% completely eliminated all inhibition zones. However 1% arginine in the overlays did not affect the production of inhibition zones by strains of S. mutans C67-1, Ny257, Ny266, and T8. These strains were shown to elaborate (in a reproducible fashion) inhibitory substances which were not organic acids. Inhibitory activity was never obtained in liquid preparations, except for strains Ny257 and T8 where it was found to be very unstable.  相似文献

Lantibiotics such as gallidermin are lanthionine-containing polypeptide antibiotics produced by gram-positive bacteria that might become relevant for the treatment of various infectious diseases. So far, self-toxicity has prevented the isolation of efficient overproducing strains, thus hampering their thorough investigation and preventing their exploitation in fields other than the food area. We wanted to investigate the effect of lantibiotic precursor peptides on the producing strains in order to evaluate novel strategies for the overproduction of these promising peptides. In this study, gallidermin was chosen as a representative example of the type A lantibiotics. A Staphylococcus gallinarum Tü3928 mutant, whose gene for the extracellular pregallidermin protease GdmP was replaced by a kanamycin-resistance gene, was constructed. Mass spectrometry (MS) analysis indicated that this mutant produced fully posttranslationally modified gallidermin precursors with truncated versions of the leader peptide, but not the entire leader as predicted from the gdmA sequence. In filter-on-plate assays, these truncated pregallidermins showed no toxicity against Staphylococcus gallinarum Tü3928 up to a concentration of 8 g/liter (corresponding to approximately 2.35 mM), while gallidermin produced clear inhibitory zones at concentrations as low as 0.25 g/liter (0.12 mM). We showed that the lack of toxicity is due entirely to the presence of the truncated leader, since MS as well as bioassay analysis showed that the peptides resulting from tryptic cleavage of pregallidermins and gallidermin produced by S. gallinarum Tü3928 had identical masses and approximately the same specific activity. This demonstrates that even a shortened leader sequence is sufficient to prevent the toxicity of mature gallidermin. In nonoptimized fermentations, the gdmP mutant produced pregallidermin to a 50%-higher molar titer, suggesting that the absence of self-toxicity has a beneficial effect on gallidermin production and giving a first confirmation of the suitability of the overproduction strategy.  相似文献

Conservation Reserve Program (CRP) fields may provide good habitat for nesting and brood-rearing ring-necked pheasants (Phasianus colchicus) during early stages of succession. But, the success of hens in early successional CRP, relative to late successional CRP and other grassland habitats, has yet to be evaluated. The reproductive period is especially critical for populations of pheasants, and CRP's benefits to hens and chicks may decrease as fields age because of loss of vegetative diversity, decrease in vegetation density, and accumulation of residual litter. During 2005–2006, we evaluated spatial and temporal variation in nest and brood survival for radio-marked hen pheasants in areas of northeastern Nebraska where portions of CRP fields had been recently disced and interseeded (DICRP) with legumes. Nests in DICRP tended to have a higher daily survival rate (0.984; 95% CI: 0.957–0.994) than nests in grasslands (including CRP) that were unmanaged (0.951; 95% CI: 0.941–0.972). The probability of 23-day nest success was 0.696 (95% CI: 0.631–0.762) for DICRP and 0.314 (95% CI: 0.240–0.389) for unmanaged grasslands. Daily brood survival rates varied by habitat type, brood age, and date of hatch. The probability of a brood surviving to day 21 was 0.710 (95% CI: 0.610–0.856). Brood survival rates increased with time spent in DICRP and as the brood aged. Survival decreased as broods spent more time in cropland and peaked seasonally with broods that hatched on 15 June. Brood survival probability, to 21 days, would be reduced to 0.36 (95% CI: 0.100–0.701) if broods in our sample had not used DICRP. We combined nest and brood survival in a productivity model that suggested 2,000 hens, in a landscape with no DICRP, would produce 1,826 chicks, whereas the same hens in a landscape of 100% DICRP would produce 5,398 chicks. Production of first-year roosters more than doubled when hens nested in DICRP. Without DICRP, population growth rates of pheasant populations usually declined; with DICRP, populations stabilized with at annual survival rates of 0.3 or greater. The positive response of nest and brood survival to discing and interseeding CRP provides further evidence that CRP fields must be managed to optimize wildlife benefits. © 2012 The Wildlife Society.  相似文献

In Collembola, pheromones appear to be present in the faecal pellets. Pheromone release after cessation of faeces production points to the digestive tract as a possible site of biosynthesis.During the pre-moulting periods Collembola do not react to pheromones, possibly due to their low activity at that time, whereas the production of the pheromones continues.Starvation periods of up to 14 days diminish pheromone release but do not cause complete cessation. Production per animal seems to decrease at increasing densities.The effect of pheromones on the reproductive efficiency of Collembola is discussed in the context of their physiological and behavioural ecology.  相似文献

Lantibiotics, a group of lanthionine-containing peptides, display their antibiotic activity by combining different killing mechanisms within one molecule. The prototype lantibiotic nisin was shown to possess both inhibition of peptidoglycan synthesis and pore formation in bacterial membranes by interacting with lipid II. Gallidermin, which shares the lipid II binding motif with nisin but has a shorter molecular length, differed from nisin in pore formation in several strains of bacteria. To simulate the mode of action, we applied cyclic voltammetry and quartz crystal microbalance to correlate pore formation with lipid II binding kinetics of gallidermin in model membranes. The inability of gallidermin to form pores in DOPC (1,2-dioleoyl-sn-glycero-3-phosphocholine) (C18/1) and DPoPC (1,2-dipalmitoleoyl-sn-glycero-3-phosphocholine) (C16/1) membranes was related to the membrane thickness. For a better simulation of bacterial membrane characteristics, two different phospholipids with branched fatty acids were incorporated into the DPoPC matrix. Phospholipids with methyl branches in the middle of the fatty acid chains favored a lipid II–independent DPoPC permeabilization by gallidermin, while long-branched phospholipids in which the branch is placed near the hydrophilic region induced an identical lipid II–dependent pore formation of gallidermin and nisin. Obviously, the branched lipids altered lipid packing and reduced the membrane thickness. Therefore, the duality of gallidermin activity (pore formation and inhibition of the cell wall synthesis) seems to be balanced by the bacterial membrane composition.  相似文献

The aim of the study was to determine susceptibility of 587 strains of S. aureus and 85 strains of coagulase-negative staphylococci isolated from outpatients in Poznań to co-trimoxazole, amoxycillin/clavulanic acid, erythromycin, gentamycin, doxycycline, ampicillin, oxacillin, cephradine, clindamycin and neomycin. Also methicillin-resistant strains were determined as well as strains ability to produce beta-lactamases. Susceptibility testing and examination of methicillin-resistant strains were performed by the disc diffusion techniques according to recommendation of NCCLS. Methicillin-resistant strains were additionally examined to their sensitivity to vankomycin and teicoplanin. beta-lactamase production was detected using nitrocefin impregnated discs and iodometric method. Amoxacillin/clavulanic acid, gentamycin, co-trimoxazole, cephradin, oxacillin and clindamycin occurred to be very active against both, S. aureus and coagulase-negative staphylococci. 84.7% to 100% of examined strains were sensitive to these drugs. Doxycyclin, erythromycin and ampicillin were less effective. Nine strains (1.5%) of 587 strains of S. aureus as well as 7 strains (8.7%) of coagulase-negative staphylococci were methicillin-resistant. All of methicillin-resistant strains were sensitive to vancomycin and teicoplanin. More than 75% of S. aureus and close to 50% of coagulase-negative staphylococci were able to produce beta-lactamases.  相似文献

Censuses are widely used to monitor populations. One of the most interesting modern techniques is distance sampling, which depends on some crucial assumptions, including the random distribution of animals with respect to transect lines, which are often asphalt and gravel roads. We tested the assumption of a random distribution of animals with respect to roads for females of ring-necked pheasants (Phasianus colchicus). Roads can be used to census pheasants using distance sampling, without significant bias because pheasants are randomly distributed with respect to roads. Our method is easy to apply to other field conditions where radio tracking data are available and can be used to test the assumption in other studies and/or with different species.  相似文献

A collection of 59 ureolytic and lactic acid-producing ruminal staphylococci and enterococci, isolates from domestic and wild ruminants, were tested for sensitivity or resistance to lantibiotic nisin. All strains tested were sensitive with zones of inhibition, around wells containing 250 μg nisin, from 6 to 26 mm. 74.6% of isolates had zones of inhibition more than 10 mm and 11.1% more than 20 mm. Nisin was more effective against enterococci than staphylococci. Sensitivity of ruminal isolates to nisin may be used to control bacterial growth during the colonization of the rumen or to study the role of antibacterial activity in microbial interactions. Results obtained can be also used in experiments on gnotobiotic animals.  相似文献

Stimulated rabbit peritoneal polymorphonuclear leukocyte (PMN) preparations simultaneously produce prostaglandin-like material and mediators that induce metabolic alterations in experimental animals characteristic of the host's responses to inflammation. The alterations observed in rats include responses by: proteins, carbohydrates, hormones, trace metals, and total blood neutrophils. This study demonstrates a possible relationship between prostaglandins and PMN-derived substances that mediate plasma zinc depression, hepatic amino acid uptake, and increased numbers of blood neutrophils. Production of these mediators by stimulated-PMN preparations was prevented by 23 muM indomethacin or 93 muM aspirin. Conversely, morphine (2 mM or less) had no detrimental effect on production of these mediators, although, it consistently stimulated production of a substance stimulating total blood neutrophia. In addition, 2 muM prostaglandin E and F stimulated production of substances mediating hepatic amino acid uptake plasma zinc depression, respectively. At this concentration, neither prostaglandin significantly altered production of substances mediating increased numbers of total blood neutrophils. A partial-nitrogen atmosphere, dibutyryl cyclic analogs of AMP and GMP, or homogenization of the PMN had no effect on mediator production. The inhibitory effect of indomethacin and aspirin also was observed with PMN-homogenastes. These experimental observations suggest that prostaglandin synthesis has a function in production of mediators by stimulated-PMN preparations.  相似文献

Predation on brown hares and pheasants in relation to their production was examined in an open field area in southern Sweden. Biomass and numbers of hares and pheasants eaten by the predators were calculated from data on food habits, food requirements, and numbers of the mammalian and avian predators present (11 species). At the same time estimates of numbers and production of hares and pheasants were obtained. At least 40% of the estimated annual production of hares and almost 60% of subadult-adult pheasants in autumn were consumed by the predators. Estimated numbers of hares and pheasants taken by the predators greatly exceeded shooting and road mortality. Foxes and cats were the predominant predators on hares (about 90% of total hare consumption). Foxes accounted for about two-thirds of predation on pheasants; cats and goshawks were of secondary importance. Pheasant nests were preyed upon by hooded crows and also by badgers. SUMMARY.
  • 1 This study documents the life history of the xylophagous elmid beetle, Lara avara, and estimates its contribution to wood degradation in Oregon streams. The life cycle was found to be 4 to 6 or more years long, with all but 2–3 months of that spent in the larval stage.
  • 2 Larvae grow through seven instars, taking about 1 year for instars 1–3, and from 3 to 5 or more years for instars 4–7.
  • 3 Last-instar larvae leave the water to pupate. Adults live approximately 3 weeks and occur from May to August. The eggs are deposited on submerged wood.
  • 4 Larvae probably obtain their nutrition by absorbing substances liberated into decaying wood by microbial activity. They do not produce their own cellulase, nor do they have a symbiotic gut flora similar to that of xylophagous cranefly (Tipulidae) larvae.
  • 5 Faecal production by L. avara larvae averaged 13% dry body wt d-1. This yields an estimate of faecal production of 1.6 g m-2 y-1 in Oregon Coast Range streams (about 0.3% y-1 of wood standing crop).
