The experimentally measured oxygen consumption rate by the cladoceran,Ceriodaphnia dubia, showed a linear increase between 5 and 20°C.Oxygen consumption rates of C. dubia were estimated in situfrom respiratory electron transport system (ETS) activity inLake Rotongaio during summer stratification and winter mixing.Oxygen consumption was 0.002 µl O2 animal–1 h–1in the hypolimnion and 0.076 µl O2 animal–1 h–1in the epilimnion during stratification. Implications of respiredoxygen for metabolic carbon requirements are discussed.  相似文献

In order to clarify the metabolic adaptation of respiratorypathways in plants to limited levels of Pi, the effects of long-termstarvation of Pi on the activities of various enzymes relatedto respiratory metabolism were examined in suspension-culturedCatharanthus roseus cells. When the activities were expressedas units per g fresh weight, only those of phosphoenolpyruvate-hydrolyzing(PEP-hydrolyzing) enzyme (which may possibly be equivalent tothe acid phosphatase activity derived from vacuoles) and PEPcarboxylase were higher in the Pi-starved cells than in controlcells. Activities of other enzymes in the Pi-starved cells werelower than or similar to those of the control cells. Time-coursestudies indicated that PEP-hydrolyzing activity was inducibleby starvation of Pi. However, in contrast to the results reportedby Duff et al. [(1989a) Plant Physiol. 90: 1275.], fluctuationsin the activity of PP1:fructose-6-phosphate 1-phosphotransferaseduring starvation of Pi were similar to those in levels of phosphofructokinaseand 6-phosphogluconate dehydrogenase. These data suggest thatthe concept of the phosphate starvation-inducible ‘bypasses’,which are engineered via the coarse control (i.e., induction)of specified enzymes and were proposed initially by Duff etal. in Brassica nigra cells, is not directly applicable to Catharanthusroseus cells in suspension. Tracer experiments using [U-14C]glutamineindicated that a significant proportion of respiratory substratescould be supplied from the enlarged pool of amino acids duringstarvation of Pi. Cycloheximide (2·0 µg ml–1) inhibits theincorporation of [14C]phenylalanine and [14C]adenine into insolublecompounds in Ankistrodesmus braunii. 6-Methylpurine (1·0mM) inhibits only the incorporation of [14C]adenine and it isconcluded that it inhibits RNA synthesis. When ammonium-growncells of Ankistrodesmus or Chlorella are nitrogen-starved orwhen ammonium-grown cells of Dunalitlla are resuspended in nitratemedium, the appearance of nitrate reductase in these organismsis not inhibited by 6-methylpurine. The appearance of nitratereductase activity in Ankistrodesmus or Chlorella is inhibitedby 6-methylpurine when ammonium-grown organisms are preincubatedwith this substance for 1-2 h before nitrogen starvation. Itis concluded that cells growing with ammonium and lacking nitratereductase activity nevertheless contain preformed mRNA for nitratereductase synthesis.  相似文献

The metabolic transformation of glycolate to glycine occurringin photosynthesizing cells of Chromatium was investigated bythe radioisotopic technique and by amino acid analysis. By analyzingthe distribution of radiocarbon upon feeding [1-14C] glycolate,[2-14C] glyoxylate and [1-14C] glycine to bacterial cells, itwas demonstrated that glycolate is converted to glycinc viaglyoxylate, and both glycolate and glycine are excreted extracellularly.Although the formation of serine was barely detected by theabove two techniques in both N2 and O2 atmospheres, it was foundthat 14CO2 is evolved quite markedly from both [1-14C] glycolateand [1-14C] glycine fed to the Chromatium cells. Analyticalresults of transient changes in amino acid compositions underatmospheric changes of N2O2 and by the addition of exogenousglycolate in N2 confirm the notion that glycolate is convertedto glycine. Acidic amino acids (glutamic acid and aspartic acid)appear to take part in glycine formation as amino donors. Theformation of glycine from glycolate in a N2 atmosphere suggeststhat an unknown glycolate dehydrogenation reaction may operatein the overall process. 1 This is paper XXXVII in the series ‘Structure and Functionof Chloroplast Proteins’. Carbonic anhydrase (CA) activity in wild type cells of Chlamydomonasreinhardtii was low when cells were cultured under 2% CO3 inthe light. When the gas phase was changed to air, CA activityincresaed as much as 20 fold over the next 24 hours. In contrast,CA activity did not change markedly in cells of the mutantspet 20-8 (PS II-negative), lip 10-2 (photophosphorylation-negative),and F60 (phosphoribulokinase-negative), when they were subjectedto the same induction regimen. DCMU (10–5 M) and cydoheximide(3 µg/ml) severely inhibited the induction in wild typecells. Seedlings from Euphorbia canariensis and Euphorbia lambii weregrown in the dark at 25 °C. Protein and triglyceride contentas well as levels of sugars and amino acids in the endospermwere determined during endosperm depletion. In the endospermof Euphorbia canariensis, relatively low levels of amino acids(up to 1 µmol.endosperm–1) were found of which glutamine/glutamateaccounted for 40% at the stage of radicle emergence. High levelsof amino acids (up to 4 µmol.endosperm–1) comparedwith sugars (up to 2 µmol sucrose.endosperm–1) weredetected in the endosperm of Euphorbia lambii. Arginine wasthe main component (28 µmol%) of the amino acids in thistissue. In both species amino acid composition changed graduallyduring endosperm depletion. Cotyledons retained their ability to absorb a variety of watersoluble substrates after removal of the endosperm. 14C from[U-14C]sucrose was effectively incorporated into the triterpenesof the laticifers and to a lesser extent into the sterols ofthe seedling. The highest incorporation values were found inyoung seedlings about 2 d after the emergence of the radicle.Seedlings of this age also showed high incorporation rates of14C from labelled alanine, serine, threonine, valine, leucineand isoleucine into both triterpenols and sterols, but no generalconclusions about metabolic channelling in lipid synthesis couldbe made. When Phaseolus mungo seeds were allowed to germinate in 5–20mM quinate solution in the dark, a marked increase in the endogenousshikimic acid level occurred in their tissues. The acid levelrose distincdy on the 2nd day of germination and reached a maximumon the 4th day. The quinate-14C fed to germinating seeds waspredominantly converted to shikimic acid, and little radioactivitywas found in 3-dehydroshikimic acid. When quinate-14C was suppliedsimultaneously with relatively high concentrations of 3-dehydroquinicacid or 3-dehydroshikimic acid, its conversionto shikimic acidwas restrained, but hardly any radioactivity was trapped ineither of the dehydro compounds. 3-Dehydroquinic acid-14C or3-dehydroshikimic acid-14C fed to the seeds was metabolizedpreferentially to shikimic acid. The experimental results arediscussed with respect to the metabolic relationship betweenquinic acid and odier alicyclic acids in the aromatic biosynthesisof P. mungo seedlings. The parasitoid nanoflagellate (PNF) Pirsonia diadema is hostspecific for the marine centric diatom Coscinodiscus spp. Experimentsshowed that flagellates significantly prefer C. wailesii overC.granii as host species (interspecific selectivity). This preferencewas independent of light conditions (dark, irradiance of 10and 70 µmol m–2 s–1) and temperature (10 and15C). Among unicellular host diatoms, the infection behaviourwas selective for individual cells: already infected C.graniicells were more attractive for further flagellate attachmentthan non-infected cells (intraspecific selectivity). Both predicted (incubator) and measured (in situ) 14C-assimilationrates were studied from February to November 1981 at three stationsin Boknafjorden, a deep silled fjord of western Norway. Sampleswere taken from different light depths within the euphotic zone.A high degree of conformity was found between the two approaches.Daily values of carbon assimilation integrated over the euphoticzone varied between 0.05 and 1.4 g C m–2. Yearly primaryproduction varied between stations from 82 to 112 g C m–2(120–148 g C m–2 when based on average light conditions).The light-saturation curve parameters B and PBmax ranged from0.0056 to 0.0537 mg C mg Chla–1 h–1 µE–1m2 and from 0.7 to 8.5 mg C mg Chla–1 h–2 (in situassimilation numbers ranged from 0.9 to 9.3 mg C mg Chla–1h–1) respectively, which compare well with those publishedfrom the northwestern side of the Atlantic. The overall importanceof light in controlling photosynthesis throughout the year wasrevealed by the light utilization index , estimated to be 0.43mg C mg Chla–1 E–1 m2. The maximum quantum yieldwas encountered on August 17, with 0.089 mol CE–1. Chla/Cratios above and below 0.010 were found to be typical for shade-and light-adapted cells respectively. Assimilation numbers andgrowth rates were linearly related only when considering light-adaptedcells. Consistent with the findings of this study, the applicabilityof IK, B and PBmax as indicators of light-shade adaptation propertiesshould be questioned. Maximum growth rates were encounteredduring an autumn bloom of the dinoflagellate Gyrodinium aureolum(1.0 doublings day–1), while 0.7–0.8 doublings day–1were found for a winter bloom (water temperature of 2°C)of the diatom Skeletonema costatum. No unambiguous temperatureeffect on assimilation number and growth of phytoplankton couldbe recognized in Boknafjorden. A tendency towards increasedassimilation numbers coinciding with increased water columnstability was revealed. The highest PBmax values were oftenencountered at almost undetectable nutrient concentrations.At least during summer this could be attributed to recyclingof nutrients by macro- and/or microzooplankton, responsiblefor a greater part of the primary production now being grazeddown. This study supports the convention that the depth of theeuphotic zone may extend considerably below the 1% light depth.  相似文献

1) A partially disintegrated cell preparation of Cyanidium caldarium(i.e. the 100G fraction; cells from which the outer parts ofthe cell wall had been removed) was obtained by differentialcentrifugation of a cell suspension treated with a French press.2) The 100G fraction cells had completely lost the outer partsof their cell walls, but retained their subcellular structureunchanged and had Hill activity that was equally as high asthat of the intact cells. 3) The Hill activity of the 100G fractionshowed an optimum at pH 7.0 and at 35°C. This activity waslost under acid and high temperature conditions (e.g. pH 3,50°C) under which intact cells showed high activity. 4)The Hill activity of the 100G fraction was lost by heat treatment(55°C, 10min), acid-treatment (pH 3.0, 10 min) or pre-illumination(3xl05lux, 30 min) though intact cells were not inactivatedby these treatments. However, no remarkable difference in sensitivitytowards inhibitors was found between the 100G fraction and intactcells. 5) We thus concluded that the cell wall plays an importantrole in the acido- and thermo-phily of Cyanidium cells. The effect of heat stress on processes related to carbohydratepartitioning was investigated in young bell pepper (Capsicumannum L. cv. Maor) plants in relation to abscission of theirreproductive organs at different stages of development. None of the reproductive organs abscised after 5 d in a normalday/night temperature regime (25/18°C). With a temperatureregime of 35°C day, 25°C night, abscission occurredin only a small portion of the flower buds and none of the flowersand fruitlets. However, when temperatures in the day and nightwere reversed (25/35°C, day/night) all the buds and someof the flowers abscised during that time period. The young fruitat the first node did not abscise under any temperature regime.The abscission rate of the flower buds was reduced under heatstress if the developing fruit at the first node had been removed. High temperature during either the light or dark periods reducedthe export of [14C]sucrose from the source leaf (fed for 48h with [14C]sucrose). Both heat stress and fruit presence reduced the relative amountof [14C]sucrose which was exported to the flower buds, flowersand roots. Likewise, these treatments reduced the concentrationof reducing sugars in the reproductive organs. Concomitantly,the heat stress and fruit presence on the first node reducedthe activity of soluble acid invertase in the flower buds andthe roots, but not in young leaves. Overall, the results show that heat stress causes alternationin sucrose distribution in the plant, but may also have specificeffects on metabolic activities related to sucrose import andutilization in flower buds and flowers which in turn may enhancetheir abscission. Most of the 14C added as glucose to carbohydrate-starved cellsof Chlorella Vulgaris can be recovered as alcohol-soluble compoundsor as polysaccharide. Only 5–I6 per cent., depending onthe position of 14C in the glucose supplied, is released ascarbon dioxide. Similar results were obtained with Chlorellapyrenoidosa and Ankistrodesmus. The labelled alcohol-solublecompounds in Chlorella vulgaris include amino-acids, particularlyglutamic acid, aspartic acid, and alanine, and, when glucose-I-14Cis metabolized, the amount of 14C recovered in these amino-acidsis about the same as that recovered as carbon dioxide. Degradationof the glucose incorporated into polysaccharide shown that theC1 and C6 atoms of glucose rapidly interchange when in the cells.The bearing of these results on attempts to estimate the relativeimportance of different pathways of glucose breakdown is discussed.  相似文献

A Cyt P450 (P450C4H) possessing trans-cinnamate 4-hydroxylase(C4H) activity was purified to apparent homogeneity from microsomesof etiolated mung bean seedlings. Upon SDS-polyacrylamide gelelectrophoresis, the purified preparation gave a single proteinband with a molecular mass of 58-kDa. Its specific P450 contentwas 12.6 nmol (mg protein)–1. Using NADPH as electrondonor, purified P450C4H aerobically converted trans-cinnamicacid to p-coumaric acid with a specific activity of 68 nmolmin–1 nmol–1 P450 in a reconstituted system containingNADPH-Cyt P450 reductase purified from the seedlings or rabbitliver microsomes, dilauroyl phosphatidylcholine, and cholate.This specific activity is by far the highest for reconstitutedC4H systems so far reported and provides direct evidence thatC4H activity is actually associated with a P450 protein. Inthe oxidized state P450C4H showed a typical low-spin type absorptionspectrum with a Soret peak at 419 nm. A partial spectral shiftto the high spin state was observed when trans-cinnamic acidwas added to oxidized P450C4H. By spectral titration, the dissociationconstant of the cinnamic acid-P450C4H complex was determinedto be 2.8 µM. The activity of plasma membrane H+-ATPase was measured withmembrane fragments of guard-cell protoplasts isolated from Viciafaba L. ATP hydrolytic activity was slightly inhibited by oligomycinand ammonium molybdate, and markedly inhibited by NO3and vanadate. In the presence of oligomycin, ammonium molybdateand NO3, the ATP-hydrolyzing activity was strongly inhibitedby vanadate. It was also inhibited by diethylstilbestrol (DES),p-chloromercuribenzoic acid (PCMB) and Ca2+, but slightly stimulatedby carbonyl cyanide m-chlorophenylhydrazone (CCCP). The acitivityhad higher specificity for ATP as a substrate than other phosphoricesters such as ADP, AMP, GTP and p-nitrophenylphosphate; theKm was 0.5 mM for ATP. The activity required Mg2+ but was notaffected by K+, and it was maximal around pH 6.8. When guard-cellprotoplasts were used instead of membrane fragments, the ATPaseactivity reached up to 800µmol Pi.(mg Chl)–1.h–1in the presence of lysolecithin. Water exchange, temperature tolerance and oxygen consumptionof the snail, Trigonephrus sp., from the southern Namib desertof Namibia were examined and related to activity. At 25°Cand 15% R.H. mean water loss and food and water uptake were5.95 mg. day–1 and 630 mg.day–1, respectively. Bodytemperature tracked sand temperature. Snails tolerated sandtemperatures as high as 45°C. Mean ± S.D. oxygenconsumption rates were 32.0 ± 2.94 µlO2.g totalbody mass–1.h–1 at 15°C, when the snails wereactive, and 11.27 µlO2.g total body mass–1.h–1at 25°C, when the snails were inactive. These values are2-6 times lower than those recorded for the similarly sizedmesic snail, Helix aspersa. Activity experiments indicated thatlow ambient temperatures and high humidities were favoured bythe snails. This, together with the burying behaviour of thesesnails during high temperatures, suggests that they limit stressby restricting activity to physiologically-favourable periods,even though more-extreme conditions may be tolerated. The light-catalysed isomerization of (+)-abscisic acid (ABA)to its trans isomer during isolation from leaves was monitoredby the addition of (±)-[2-14C]ABA to the extraction medium.(+)Trans-abscisic acid (t-ABA) was found to occur naturallyin rose (Rosa arvensis) leaves at 20µg/kg fresh weight;(+)-ABA was present at 594µg/kg. (±)-[2-14D]Trans-abscisicacid was not isomerized enzymically to ABA in tomato shoots. (±)-Abscisic acid was converted by tomato shoots to awater-soluble neutral product, ‘Metabolite B’, whichwas identified as abscisyl-ß-D-glucopyranoside. When(±)-[2-14C]trans-abscisic acid in an equimolar mixturewith (±)-[2-14C}ABA was fed to tomato shoots it was convertedto its glucose ester 10 times faster than was ABA. Trans-abscisyl-ß-D-glucopyrano8ide only was formedfrom (±)-[2-14C]t-ABA in experiments lasting up to 30h. Glucosyl abscisate was formed slowly from ABA and the freeacid fraction contained an excess of the unnatural (–).ABAas did the ABA released from abscisyl-ß-D-glucopyranosideby alkaline hydrolysis. The (+).ABA appeared to be the solesource of the acidic ‘Metabolite C" previously noted. The concentrations of endogenous (+)-, (+)-[2-14C]-, and (–)-[2-14C]ABAremaining as free acid, and also in the hydrolysate of abscisyl-ß-D-glucopyranoside,were measured by the ORD, UV absorption, and scintillation spectrometryof highly purified extracts of ABA from tomato shoots whichhad been supplied with racemic [2-l4C]ABA.  相似文献

5-Keto-D-[1-14C]gluconic acid, the most effective precursorof L(+)tartaric acid among all labeled compounds which haveever been tested in grapes, was found to be a good precursorof L(+)tartaric acid in a species of Pelargonium. The synthesisof labeled L(+)tartaric acid from D-[1-14C]glucose in Pelargoniumwas remarkably depressed when a 0.5% solution of D-gluconateor 5-keto-D-gluconate was administered continuously to leavestogether with D-[1-14C]glucose. Our results provide strong evidence that D-[1-14C]glucose ismetabolized in Pelargonium to give labeled L(+)tartaric acidvia (probably D-gluconic acid and) 5-keto-D-gluconic acid withoutpassing through L-ascorbic acid. Labeled L-idonic acid was found in young leaves of Pelargoniumwhich had been labeled with L-[U-14C]ascorbic acid. The synthesisof the labeled L-idonic acid increased when a 0.1% solutionof L-threonate was administered continuously to leaves togetherwith L-[U-14C]ascorbic acid. Specifically labeled compounds, recognized as the members ofthe synthetic pathway for L(+)tartaric acid from L-ascorbicacid via L-idonic acid in grapes, were administered to youngleaves of Pelargonium. Each compound (2-keto-L-[U-14C]idonicacid, L-[U-14C]idonic acid, 5-keto-D-[1-14C]gluconic acid and5-keto-D-[6-14C]gluconic acid) was partly metabolized, as ingrapes. The metabolic pathway starting from L-ascorbic acidto L(+)tartaric acid via L-idonic acid, however, did not actuallycontribute to the synthesis of L(+)tartaric acid in Pelargoniumprobably because the activity of each metabolic step was muchlower than that observed in grapes.
