The hemoglobin from a European Bison (Bison bonasus) was analysed and the complete primary structures of the alpha I-, alpha II-and beta-chains have been determined. The alpha I- and alpha II-chains differ only at position alpha 19 (Asp----Gly). The beta-chains are homogeneous. The sequences are compared with the globin chains of Bison bison and bovine and the polymorphism of the alpha-chains is discussed. On the basis of the primary structure it may be concluded that the hemoglobin of Bison bonasus belongs to the group of hemoglobins with intrinsically low oxygen affinity.  相似文献

The structurally reinforced jaws of the cownose ray, Rhinoptera bonasus testify to this species' durophagous diet of mollusks, but seem ill-suited to the behaviors necessary for excavating such prey. This study explores this discordance by investigating the prey excavation and capture kinematics of R. bonasus. Based on the basal suction feeding mechanism in this group of fishes, we hypothesized a hydraulic method of excavation. As expected, prey capture kinematics of R. bonasus show marked differences relative to other elasmobranchs, relating to prey excavation and use of the cephalic lobes (modified anterior pectoral fin extensions unique to derived myliobatiform rays). Prey are excavated by repeated opening and closing of the jaws to fluidize surrounding sand. The food item is then enclosed laterally by the depressed cephalic lobes, which transport it toward the mouth for ingestion by inertial suction. Unlike in most sharks, upper jaw protrusion and mandibular depression are simultaneous. During food capture, the ray's spiracle, mouth, and gill slit movements are timed such that water enters only the mouth (e.g., the spiracle closes prior to prey capture and reopens immediately following). Indigestible parts are then hydraulically winnowed from edible prey portions, by mouth movements similar to those used in excavation, and ejected through the mouth. The unique sensory/manipulatory capabilities of the cephalic lobes, as well as the cownose ray's hydraulic excavation/winnowing behaviors and suction feeding, make this species an effective benthic predator, despite its epibenthic lifestyle.  相似文献

The aim of this study was to identify the molecular genetic origin underlying the I variant of αs1-casein and to develop a DNA-based test for this polymorphism as a tool for genetic analyses independent of milk sample testing. All coding exons and flanking regions of the α s 1 -casein gene were sequenced in DNA samples from cattle of known α s 1 -casein genotypes ( BI , CI , II , CC ), determined by isoelectric focusing of milk samples. A nucleotide substitution (A>T) in exon 11 (g.19836A>T) leads to the exchange of Glu with Asp at amino acid position 84 of the mature protein (p.Glu84Asp) and perfectly co-segregated with the presence of the αs1-casein I variant in the milk of the analysed animals. Genotyping of a total of 680 DNA samples from 31 Bos taurus and Bos indicus cattle breeds and from Bos grunniens , Bison bison and Bison bonasus by restriction fragment length polymorphism analysis revealed the occurrence of Asp at position 84 at low frequencies in Bos taurus and Bos indicus breeds and established its origin from the α s1-casein C variant (p.Glu192Gly). Ten different intragenic haplotypes in the gene region from intron 8 to intron 12 were observed by sequencing, of which two occurred in Bison bison and one in Bison bonasus only. Using available casein gene complex information, an association of Asp at position 84 to β-casein A 2 and κ-casein B was shown in the Bos indicus breed Banyo Gudali . Taken together, we can postulate that the α s1-casein variant I is caused by a non-synonymous nucleotide substitution in exon 11 of the gene and that it originated within Bos indicus and spread to Bos taurus subsequently.  相似文献

Since 1980 severe chronic balanoposthitis has been observed in free-living European bison (Bison bonasus) in the Bia?owieza Primeval Forest (Poland). Sera of 50 bison with balanoposthitis and 48 clinically healthy male and 49 female bison were investigated for antibodies against Mycoplasma bovis and M. bovigenitalium by western blot analysis. Prevalence of antibodies against M. bovigenitalium was significantly higher in bison with balanoposthitis than in unaffected male bison. Mycoplasma bovigenitalium may play a role in the pathogenesis of balanoposthitis in European bison.  相似文献

We describe gross and histopathology of cysts found in the epididymis of the European bison (Bison bonasus). The material was collected from 107, 3 mo to 20 yr old, free-ranging, culled males from the Bia?owieza Primeval Forest (Poland). Epididymal cysts were observed in 65 (61%) of bison of varying ages. They were primarily in the head of epididymis and less often in the corpus or tail. The cysts contained opalescent fluid devoid of spermatozoa. They were lined by epithelium resembling that of efferent ducts and appeared to be congenital abnormalities.  相似文献

Between July 2003 and November 2004, 21 cownose rays Rhinoptera bonasus were tagged and tracked within Pine Island Sound estuary, Florida, using passive acoustic telemetry. Residence time of individuals ranged between 1 and 102 days. No relationship was detected between ray activity and tidal stage or time of day. Minimum convex polygons (MCP) and kernel utilization distributions (KUD) were calculated over several time frames to demonstrate the extent of an animal's home range and core areas of use. Total MCPs ranged between 0·81 and 71·78 km2 (mean = 22·01 km2), with daily MCPs as large as 25·8 km2. Total 95% KUDs ranged between 0·18 and 62·44 km2 (mean = 22·63 km2), while total 50% KUDs were smaller, ranging from 0·09 to 9·68 km2 (mean = 3·33 km2). Both MCP and KUD areas exhibited a positive relationship with residence time and R. bonasus size. As mobile, pelagic swimmers capable of traversing large distances, these data show that cownose rays travel extensively throughout this estuary, yet may remain within very small areas for extended periods.  相似文献

The comparative analysis of genetic differentiations between three species of Bovinae--Bos taurus, Bison bonasus, Bison bison with the use of different types of molecular-genetic markers--genetical-biochemical (35 loci) and DNA markers (RAPD-PCR, ISSR-PCR) was carried out. It was shown, that the evaluation of interspecies genetic interrelations was connected more with the determined molecular-genetic markers (loci), included in analysis, than with the marker's belonging to certain type (protein polymorphism, variability of DNA repeat distributions).  相似文献

From 1980 to 1983, blood was taken from 60 selected European bison (Bison bonasus) in Poland. Serum samples were tested for the presence of antibodies against Brucella abortus, 14 serovars of Leptospira interrogans, Chlamydia spp., Coxiella burnetti; foot and and mouth disease virus, bovine leukemia virus and bovine herpes virus-1. In addition, an attempt was made to isolate bovine herpes virus-1 from the prepuce of selected bulls. Serological tests suggested chlamydial infection in 28 bison, subclinical Q-fever of a 2-yr-old heifer, subclinical bovine leukemia virus infection in a 12-yr-old bull and bovine herpes virus-1 infection in five bulls and three cows. Attempts to isolate bovine herpes virus-1 were not successful. These results suggest the possibility of cross transmission of several of these bovine pathogens between free-ranging bison and domestic cattle in Poland.  相似文献

Genome fingerprinting with a hypervariable minisatellite sequence of phage M13 DNA was used to study the genetic variation in individual species of the genera Bos and Bison (subfamily Bovinae) and in their interspecific and intergeneric hybrids. DNA fingerprints were obtained for domestic cow Bos taurus primigenius, vatussy Bos taurus macroceros, banteng Bos javanicus, gaur Bos gaurus, wisent Bison bonasus, bison Bison bison, and for the interspecific and intergeneric hybrids. Compared with the original species, most hybrids showed a greater variation in number and size of hybridization fragments. An association was revealed between the number of hybridization fragments and blood composition of interspecific hybrids resulting from unique crossing of domestic cow and banteng. Pairwise similarity coefficients were calculated to construct a dendrogram of genetic similarity, which reflected the relationships between the parental species and hybrids varying in blood composition. The applicability of the method for identifying interspecific and intergeneric hybrids and for studying the consequences of distant hybridization in the subfamily Bovinae is discussed.  相似文献

In the Bialowieza Primeval Forest (Poland) a chronic disease of the external genital organs has been observed in free-living male European bison (Bison bonasus) since 1980. Investigations on this disease started in the late 1980s. The most striking findings are necrotic and ulcerative lesions of the prepuce and penis of bison aged from 6 mo to >10 yr. Histologic examination of tissue samples from the prepuce of six bison (9-mo- to 8-yr-old), and from the penis of two bison (3- and 8-yr-old), were characteristic of necrobacillosis. Masses of slender, Gram-negative, rod-like or filamentous bacteria occurred in necrotic tissue. At the periphery of necrotic tissue filamentous bacteria were often arranged in large clusters and strands that advanced towards healthy tissue. Immunolabeling and electron microscopy also suggest that these organisms are Fusobacterium sp.  相似文献

The aim of this study was to assess whether animal tuberculosis (TB) is transmitted between free-living European bison (Bison bonasus caucasicus), wild boars (Sus scrofa), and protected carnivores such as grey wolves (Canis lupus), brown bears (Ursus arctos), and Eurasian lynx (Lynx lynx) in the Bieszczady Mountains in Southern Poland. Results of animal studies suggest that TB transmission from bison or wild boars to grey wolves is possible. These are the first described cases where Mycobacterium caprae was detected in samples collected from grey wolves.  相似文献

Five European bison (Bison bonasus) from three European zoos were shipped to the Bukovské Vrchy Hills (Slovakia) in June 2004 and kept together in an acclimatization enclosure. The European bison were released into the wild in December 2004. At that time, papillomas were found at the medial canthus of the left eye of a 12-yr-old female bison. Cutaneous papillomatosis was confirmed histologically. Negative stain transmission electron microscopic examination revealed papillomavirus in the papillomas, and papillomavirus DNA also was detected using the polymerase chain reaction with FAP59 and FAP64 primers. The amplified 413 bp DNA sequence was identical to that of BAPV2 bovine papillomavirus. This paper is the first report of papillomatosis in European bison.  相似文献

European bison (Bison bonasus) populations from both the Polish (PL) and the Belarusian (BY) sides of the Bia?owie?a Forest represent the Lowland genetic line (LB line) – progeny of the Lowland bison (Bison bonasus bonasus) that inhabited western, central, and south‐eastern Europe in historical times. During the species recovery, one of the founders was a descendant of the extinct Caucasian bison (Bison bonasus caucasicus) and its descendants formed the other genetic line – Lowland–Caucasian (LC). There have been justified suspicions that LB European bison in the former Soviet Union had undergone cross‐mating with the LC line. We performed a comparative genetic analyses on European bison from the BY and PL parts of the Bia?owie?a Forest, the LC line and extinct Caucasian bison, based on a set of 19 microsatellite markers and 1512 bovine single nucleotide polymorphism (SNP) markers, polymorphic in at least one of the studied populations. Although genetic variability (mean allele number and expected heterozygosity) for both populations were similar, the FST jack‐knifing and principal component analyses PCA revealed highly significant differences between PL and BY bison from the Bia?owie?a Forest. Examining DNA of the extinct Caucasian bison revealed that at least part of the genetic variants found in the BY, but not the PL, population were of Caucasian origin. The results indicate that the contemporary population of European bison from the BY part of the Bia?owie?a Forest should not be regarded as a LB line. The results also suggest that the actual global population size of the LB line European bison is only a half of its official status. We conducted virologic investigations on postmortem specimens from 261 free-living European bison (Bison bonasus) from the Bialowieza Primeval Forest, Poland collected between 1990 and 2000. Fifty-four of 94 males had balanoposthitis; none of the 167 female bison examined had reproductive tract lesions. Peripheral blood, swabs, and various tissues were analyzed for bovine viruses as well as for viral DNA by bovine herpesvirus 1 (BoHV-1) and bovine herpesvirus 4 (BoHV-4) specific polymerase chain reaction (PCR) technique. An infectious bovine rhinotracheitis like BoHV-1 strain was isolated from the spleen of a female bison calf and additionally was detected by nested PCR from splenic tissue. None of the bison had significant antibody titers against BoHV-1, bovine herpesvirus 2, BoHV-4, caprine herpesvirus 1, cervid herpesvirus 1, or bovine viral diarrhea (BVD) virus-1. However, low antibody titers in two animals indicate that this European bison population has been exposed to BVD virus or BVD-like viruses and BoHV-2.  相似文献

Tendons of the jaw adductor muscles of a hard prey crushing stingray exhibit similar adaptations to compressive and shear loads as those seen in mammalian tendons. Ventral intermandibular tendon from the cownose ray, Rhinoptera bonasus, has a prominent fibrocartilaginous pad that lies between a fibrous region of the tendon and the mineralized tissue of the jaw. Histologically the pad is similar to the fibrocartilaginous meniscus of mammals, and these tissues also share some biochemical traits. Proteoglycan (PG) content in the fibrocartilaginous pad is nearly four times higher than in the linearly arrayed tendinous tissue. The predominant PGs appear to be an aggrecan-like molecule and a decorin-like molecule. The decorin-like molecule is quite small when compared to mammalian decorin (20-80 kDa vs. 100-200 kDa). This study is the first to document adaptations to compressive/shear loading in tendon from a cartilaginous fish, and the similarities to the mammalian condition argue for the early evolution of this reactive ability of tendinous tissue.  相似文献

对欧拉型藏绵羊生长激素受体(GHR)基因5′侧翼区(包括P1启动子和外显子1A)进行了T-A克隆和序列测定(GenBank accession No. EF116490), 在分析其序列结构特征的基础上与GenBank中摩弗伦羊、山羊、普通牛、欧洲野牛进行了比较基因组学和系统进化研究, 结果表明: (1)欧拉型藏绵羊GHR基因启动子P1区存在C/EBP、C/EBPb、SP1、Cap、USF、HFH-2、HNF-3b、Oct-1等多个潜在的转录因子结合位点, 可能与GHR基因的转录调控和起始以及特异表达有关。在该非编码序列中, 重复序列所占比率为2.55%, 不存在SINEs、LINEs、LTR类反转录元件和DNA转座子元件, 而发现存在一(TG)11微卫星位点; (2)在启动子P1区, 藏绵羊与摩弗伦羊、山羊、普通牛、欧洲野牛各物种间同源性大小分别为99.7%、94.2%、85.9%、86.5%; 而在外显子1A区段, 藏绵羊与摩弗伦羊、山羊、普通牛、欧洲野牛各物种间同源性大小分别为 99.0%、97.0%、92.7%、94.6%。物种间欧拉型藏绵羊与摩弗伦羊同源性最高, 而欧拉型藏绵羊与普通牛最低。(3)邻接法(即NJ法)构建的分子系统进化树聚类结果表明, 欧拉型藏绵羊与摩弗伦羊先聚为一类, 再与山羊聚类形成一个大分支, 而普通牛和欧洲野牛先聚类形成另一大分支, 两大分支最后再聚为一起, 其聚类结果与线粒体DNA和动物学分类的研究结果一致。  相似文献

Analyses of hematological parameters were carried out on eight axis deer (Cervus axis), 12 fallow deer (Cervus dama), 16 red deer (Cervus elaphus hippelaphus), three sambar (Cervus unicolor), nine Père David deer (Elaphurus davidianus), 20 European bison (Bison bonasus), seven nilgai (Boselaphus tragocamelus), eight mouflon (Ovis musimon), four white-bearded gnu (Connochaetes taurinus) and six barbary sheep (Ammotragus lervia). The following parameters were determined: packed cell volume, hemoglobin concentration, red blood cell number, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, leukocyte number and differential leukocyte count. Some degree of interspecific variability was found in the artiodactyls for these hematological parameters. An inverse relationship between mean corpuscular volume and red blood cell number is described, and specific values for these parameters are given for the four subfamilies.  相似文献

Between April 2008 and March 2009, we analyzed the pattern of coccidian oocysts present in the feces of the European bison (Bison bonasus L., 1758) and found 4 species (Eimeria bovis , E. canadensis, E. ellipsoidalis, E. zuernii) previously reported from this host and 3 species (Eimeria alabamensis, E. cylindrica, E. pellita) that are new host and locality records. All the species occurred in bison females, and only 4 occurred in males; E. bovis was the most prevalent in both sexes. The overall prevalence of Eimeria spp. invasion reached 34.7% in cows and 13.9% in bulls. The highest prevalence was noted in early spring, with a peak in April, and the lowest in late autumn and winter. The oocyst count per gram of feces (OPG) varied from 50 to 1,350; no symptoms of clinical coccidiosis were observed. We found a significant influence of winter aggregations of bison on shedding of coccidian oocysts. The prevalence and OPG values were higher in bison congregating in large numbers around winter-feeding sites in comparison to other sites. We suggest that the coming together of cows during the growing season impacts the gender-related differences in prevalence and the number of coccidian species involved. This observation probably results from an increased production of oocysts by sub-clinically infected individuals in high-density bison populations.  相似文献

To describe genetic variability and population diversity in domesticated populations of American bison (Bison bison), aurochs (Bison bonasus), and gray Ukrainian cattle (Bos taurus) different variants of DNA fingerprinting technique (utilizing the M13 phage DNA, (TTAGGG)4 synthetic oligonucleotide, and three arbitrary primers as hybridization probes) were used. Several parameters characterizing polymorphism and genetic diversity levels in each population (species) were evaluated on the basis of the profiles obtained. Dendrograms reflecting similarities between individual animals were constructed. Genetic variability of minisatellite and telomeric markers observed in the gray Ukrainian cattle flock was higher than that in aurochs and bisons. Comparison of the intrapopulation similarity (S) and gene diversity (H) indices along with the analysis of clusters in the dendrograms showed that the relatedness between the aurochs individuals was much higher than between the individual animals in the bison and gray Ukrainian cattle flocks. Furthermore, the gray Ukrainian cattle flock was represented by more distant relatives than the bison flock. It is suggested that reduced genetic variability and the appearance of deviant genotype observed in the two bison lines under selection, resulted from close inbreeding and the founder effect. The diagnostic value and efficacy of utilization of different molecular markers for estimation of genetic diversity and relatedness in domesticated animal populations is discussed.  相似文献

A free-living population of European bison Bison bonasus in the Białowieża Primeval Forest originated from only seven founder animals after a severe bottleneck that occurred at the beginning of the 20th century. Consequently, the contemporary population of the species is characterized by low genetic diversity. We studied a total of 195 individuals (127 males and 68 females). A 1429 bp fragment of mitochondrial DNA (mtDNA) including the D-loop region was analyzed in 87 individuals and revealed only three distinct haplotypes. Nucleotide ( π ) and haplotype ( H d) diversity values were estimated for the European bison and were compared with π and H d estimated from three individuals of American bison Bison bison . Very low diversity values were found in the European bison in comparison with the diversity values found in the American bison. The low mtDNA variability in the European bison is in concordance with theoretical expectations for a species that has undergone a severe and recent bottleneck. A management strategy for the preservation of the rare and very rare haplotypes present in the Białowieża population of the European bison is discussed. Furthermore, all 195 individuals were investigated for heteroplasmy involving these three haplotypes, in order to detect a possible association between heteroplasmy and the incidence of males affected by posthitis , a disease that affects the male reproductive organs, leading to necrotic lesions. Heteroplasmy was found in 15 females, in 17 males affected by posthitis and in 11 non-affected males, and no significant association was found.  相似文献
