The Michaelis-Menten parameters, JM and Km of the initial 1-min fluxes of uptake of l-phenylalanine and of α-aminoisobutyric acid were determined for extracellular concentrations of Na+ ranging from 0.5 to 110 mequiv/l for Ehrlich ascites tumor cells. The maximal initial flux, JM, decreased with decrease in extracellular Na+ for both α-aminoisobutyric acid and phenylalanine but the Km for α-aminoisobutyric acid increased markedly as the Na+ concentration fell whereas the Km for phenylalanine decreased. Cycloleucine behaved like phenylalanine.The data provides strong evidence that the Na+-independent flux of phenylalanine is an exchange diffusion flux that can be varied by changing the intracellular level of amino acids such as phenylalanine. For phenylalanine, cyclolcucine, and methionine this exchange diffusion flux appears to be additive with the Na+-dependent initial flux. α-Aminoisobutyric acid also has an exchange diffusion that is Na+-independent but it has a high Km and is not additive with the Na+-dependent flux.  相似文献

Uptake of methionine, α-aminoisobutyric acid, and α-(methyl-amino)-isobutyric acid has been shown to occur by at least two transport systems, one sensitive and the other insensitive to the Na+ concentration. For α-aminoisobutyric acid and its N-methyl derivative, the Na+-insensitive uptake is not concentrative and its rate increases almost linearly with concentration within the range examined. In contrast, the Na+-insensitive uptake of methionine is concentrative and subject to inhibition by such amino acids as phenylalanine, leucine, and valine, although not in a manner to indicate that the uptake is mediated by a single agency. This component is not produced by a residual operation of the Na+-requiring transport system, handicapped by the absence of Na+ or by its having combined with α-aminoisobutyric acid. The increase in the rate of methionine uptake is linear with concentration only above about 16 mM methionine. The Na+-sensitive uptakes of methionine, α-aminoisobutyric, and α-(methylamino)-isobutyric acid appear to occur by the same population of transport-mediating sites. Both Km and V max of the Na+-sensitive uptake of these three amino acids change with changes in the concentration of Na+, an effect which is shown to have a theoretical basis. A similarity in the values of Vmax for ten amino acids entering principally by the Na+-sensitive agency indicates that differences in their Km values probably measure differences in their affinities for that transport-mediating system.  相似文献

Summary A model with a carrier having sites for both amino acid and Na+ can account for AIB (-aminoisobutyric acid) transport kinetics observed in membrane vesicles from SV3T3 (simian virus 40-tranformed Balb/c3T3 cells) and 3T3 (the parent cell line). The main feature of this cotransport model is that Na+ binding to carrier decreases the effectiveK m for AIB transport, Na+ transport kinetics observed in both vesicle systems can be described by passive (possibly facilitated) diffusion. The lag of Na+ transport across the membrane compared to that for AIB, coupled to the Na+-dependent decrease in theK m for AIB, accounts for the overshoot in intravesicular AIB observed for SV3T3 in the presence of an initial Na+ gradient. Extra-vesicular Na+ maintains a derease in theK m for AIB influx before intra-vesicular Na+ has accumulated to balance it with a comparable decrease in theK m for AIB efflux. 3T3 vesicles display little overshoot, and this finding can be explained mostly by a lower carrier affinity for Na+.  相似文献

This work was devoted to the study of the structure-affinity relationships in neutral amino acid transport by intestinal brush border of marine fish (Dicentrarchus labrax). The effects of the length of the side chain on kinetics of glycine, alanine, methionine and amino isobutyric acid were investigated. In the presence of K+ two components were characterized: one is saturable by increased substrate concentrations, whereas the other can be described by simple diffusion mechanism. Simple diffusion, a passive, non-saturable, Na+-independent route, contributes largely to the transport of methionine and to a much lesser extend to alanine, glycine or alphaaminoisobutyric acid uptakes. If a branched chain is present, as in the case of amino isobutyric acid, diffusion is low. A Na+-independent, saturable system has been fully characterized for methionine, but not for branched amino acids such as amino isobutyric acid. In the presence of Na+ saturable components were shown. Two distinct Na+-dependent pathways have been characterized for glycine uptake, with low and high affinities. For alanine and methionine only one Na+-dependent high affinity system exists with the same half-saturation concentration and the same maximum uptake at saturable concentrations. Glycine high affinity system has the same half-saturation concentration as methionine or alanine uptake, whereas maximum uptake is lower. The substitution of the hydrogen by a methyl group results in a severe decrease of uptake (aminoisobutyric acid). Mutual inhibition experiments indicate that the same carriers could be responsible for methionine and alanine uptakes and probably glycine Na+-dependent uptake. The influence of Na+ concentrations (100-1 mol·l-1) on amino acid uptake was examined. Glycine, alanine, methionine and amino isobutyric acid transport can be described by a hyperbolic function, with a saturation uptake which is highly increased for methionine. However, the half-saturation concentration does not seem to be strongly affected by the amino acid structure. The effect of Na+ concentration (25 and 100 mmol·l-1) on the kinetics of methionine uptake have been also examined. The uptake of l-phenylalanine into brush border microvilli vesicles and basolateral plasma membrane vesicles isolated from rat kidney cortex by differential centrifugation and free flow electrophoresis was investigated using filtration techniques.Brush border microvilli but not basolateral plasma membrane vesicles take up l-phenylalanine by an Na+-dependent, saturable transport system. The apparent affinity of the transport system for l-phenylalanine is 6.1 mM at 100 mM Na+ and for Na+ 13 mM at 1 mM l-phenylalanine. Reduction of the Na+ concentration reduces the apparent affinity of the transport system for l-phenylalanine but does not alter the maximum velocity.In the presence of an electrochemical potential difference for Na+ across the membrane (ηNa0 >ηNa1) the brush border microvilli accumulate transiently l-phenylalanine over the concentration in the incubation medium (overshoot phenomenon). This overshoot and the initial rate of uptake are markedly increased when the intravesicular space is rendered electrically more negative by membrane diffusion potentials induced by the use of highly permeant anions, of valinomycin in the presence of an outwardly directed K+ gradient and of carbonyl cyanide p-trifluoromethoxyphenylhydrazone in the presence of an outward-directed proton gradient.These results indicate that the entry of l-phenylalanine across the brush border membrane into the proximal tubular epithelial cells involves cotransport with Na+ and is dependent on the concentration difference of the amino acid, on the concentration difference of Na+ and on the electrical potential difference. The exit of l-phenylalanine across the basolateral plasma membranes is Na+-independent and probably involves facilitated diffusion.  相似文献

The activation of Ca2+-dependent K+ channel by propranolol or by ascorbate-phenazine methosulphate stimulates Na+-dependent transport of α-aminoisobutyric acid. This stimulation arises from a membrane hyperpolarization due to the specific increase of membrane K+ conductance. The characteristics of α-aminoisobutyric acid translocation were examined in membrane vesicles from obligately alkalophilic Bacillus alcalophilus and its non-alkalophilic mutant derivative, KM23. Vesicles from both strains exhibited α-aminoisobutyric acid uptake upon energization with ascorbate and N,N,N′,N′-tetramethyl-p-phenylenediamine. The presence of Na+ caused a pronounced reduction in the Km for α-aminoisobutyric acid in wild-type but not KM23 vesicles; the maximum velocity (V) was unaffected in vesicles from both strains. Passive efflux and exchange of α-aminoisobutyric acid from wild-type vesicles were Na+-dependent and occurred at comparable rates (with efflux slightly faster than exchange). This latter observation suggests that the return of the unloaded carrier to the inner surface is not rate-limiting for efflux. The rates of α-aminoisobutyric acid efflux and exchange were also comparable in KM23 vesicles, but were Na+-independent. Furthermore, in vesicles from the two strains, both efflux and exchange were inhibited by generation of a transmembrane electrochemical gradient of protons, outside positive. This suggests that the ternary complex between solute, carrier, and coupling ion bears a positive charge in both strains even though the coupling ion is changed. Evidence from experiments with an alkalophilic strain that was deficient in l-methionine transport indicated that the porters, i.e., the solute-translocating elements, used by non-alkalophilic mutants are not genetically distinct from those used by the alkalophilic parent; that is, the change in coupling ion cannot be explained by the expression of a completely new set of Na+-independent, H+-coupled porters upon mutation of B. alcalophilus to non-alkalophily.  相似文献

A method is described for the extraction of microsomal ouabain-sensitive (Na+ + K+)-activated ATPase from separated frog skin epithelium. The method yields a microsomal fraction containing (Na+ + K+)-stimulated activity in the range of 30–40 nmol · mg−1 · min−1 at 26 °C. This portion, which is also ouabain sensitive, is about half of the total activity in media containing Mg2+, Na+ and K+. These preparations also contain Mg2+-dependent or Ca2+-dependent activities which are not additive and which are not significantly affected by ouabain, Na+, K+ or Li+.The activations of the ouabain-sensitive ATPase activity by Mg2+, Na+, and K+ are similar to those described in other tissues. It is found that Li+ does not substitute for Na+ as an activator but in high concentrations does produce partial activation in the presence of Na+ with no K+. These results are pertinent to the reported observations of ouabain-sensitive Li+ flux across frog skin. It is concluded that this flux is not apparently due to a direct activating effect of Li+ on the sodium pump.  相似文献

A procedure has been developed for the separation of intrinsic proteins of plasma membranes from the electric organ of Torpedo marmorata. (Na+ + K+)-ATPase, nicotinic acetylcholine receptor and acetylcholinesterase remained active after solubilization with the nonionic detergent dodecyl octaethylene glycol monoether (C12E8). These components could be separated by ion exchange chromatography on DEAE-Sephadex A-25. Fractions enriched in ouabain-sensitive K+-phosphatase or (Na+ + K+)-ATPase activity showed two bands in sodium dodecyl sulphate polyacrylamide gel electrophoresis corresponding to the α- and β-subunits. The (Na+ + K+)-ATPase was shown to have immunological determinants in common with a 93 kDa polypeptide which copurified with the nicotinic acetylcholine receptor, also after solubilization in Triton X-100 and chromatography on Naja naja siamensis α-toxin-Sepharose columns. The data suggest that the α-subunit of (Na+ + K+)-ATPase associates with the acetylcholine receptor in the membranes of the electric organ.  相似文献

The pH-dependent fluorescence quenching of acridine orange was used to study the Na+- and K+-dependent H+ fluxes in tonoplast vesicles isolated from storage tissue of red beet and sugar beet (Beta vulgaris L.). The Na+-dependent H+ flux across the tonoplast membrane could be resolved into two components: (a) a membrane potential-mediated flux through conductive pathways; and (b) an electroneutral flux which showed Michaelis-Menten kinetics relationship to Na+ concentration and was competitively inhibited by amiloride (Ki = 0.1 millimolar). The potential-dependent component of H+ flux showed an approximately linear dependence on Na+ concentration. In contrast, the K+-dependent H+ flux apparently consisted of a single component which showed an approximately linear dependence on K+ concentration, and was insensitive to amiloride. Based on the Na+- and K+-dependent H+ fluxes, the passive permeability of the vesicle preparation to Na+ was about half of that to K+.

The apparent Km for Na+ of the electroneutral Na+/H+ exchange varied by more than 3-fold (7.5-26.5 millimolar) when the internal and external pH values were changed in parallel. The results suggest a simple kinetic model for the operation of the Na+/H+ antiport which can account for the estimated in vivo accumulation ratio for Na+ into the vacuole.

Gallbladder Na+ absorption is linked to gallstone formation in prairie dogs. Na+/H+ exchange (NHE) is one of the major Na+ absorptive pathways in gallbladder. In this study, we measured gallbladder Na+/H+ exchange and characterized the NHE isoforms expressed in prairie dogs. Na+/H+ exchange activity was assessed by measuring amiloride-inhibitable transepithelial Na+ flux and apical 22Na+ uptake using dimethylamiloride (DMA). HOE-694 was used to determine NHE2 and NHE3 contributions. Basal J Na ms was higher than J Na sm with J Na net absorption. Mucosal DMA inhibited transepithelial Na+ flux in a dose-dependent fashion, causing J Na ms equal to J Na sm and blocking J Na net absorption at 100 μm. Basal 22Na+ uptake rate was 10.9 ± 1.0 μmol · cm−2· hr−1 which was inhibited by ∼43% by mucosal DMA and ∼30% by mucosal HOE-694 at 100 μm. RT-PCR and Northern blot analysis demonstrated expression of mRNAs encoding NHE1, NHE2 and NHE3 in the gallbladder. Expression of NHE1, NHE2 and NHE3 polypeptides was confirmed using isoform-specific anti-NHE antibodies. These data suggest that Na+/H+ exchange accounts for a substantial fraction of gallbladder apical Na+ entry and most of net Na+ absorption in prairie dogs. The NHE2 and NHE3 isoforms, but not NHE1, are involved in gallbladder apical Na+ uptake and transepithelial Na+ absorption. Received: 9 February 2001/Revised: 11 April 2001  相似文献

Summary The effect of extracellular and intracellular Na+ (Na o + , Na i + ) on ouabain-resistant, furosemide-sensitive (FS) Rb+ transport was studied in human erythrocytes under varying experimental conditions. The results obtained are consistent with the view that a (1 Na++1 K++2 Cl) cotransport system operates in two different modes: modei) promoting bidirectional 11 (Na+–K+) cotransport, and modeii) a Na o + -independent 11 K o + /K i + exchange requiring Na i + which, however, is not extruded. The activities of the two modes of operation vary strictly in parallel to each other among erythrocytes of different donors and in cell fractions of individual donors separated according to density. Rb+ uptake through Rb o + /K i + exchange contributes about 25% to total Rb+ uptake in 145mm NaCl media containing 5mm RbCl at normal Na i + (pH 7.4). Na+–K+ cotransport into the cells occurs largely additive to K+/K+ exchange. Inward Na+–Rb+ cotransport exhibits a substrate inhibition at high Rb o + . With increasing pH, the maximum rate of cotransport is accelerated at the expense of K+/K+ exchange (apparent pK close to pH 7.4). The apparentK m Rb o + of Na+–K+ cotransport is low (2mm) and almost independent of pH, and high for K+/K+ exchange (10 to 15mm), the affinity increasing with pH. The two modes are discussed in terms of a partial reaction scheme of (1 Na++1 K++2 Cl) cotransport with ordered binding and debinding, exhibiting a glide symmetry (first on outside = first off inside) as proposed by McManus for duck erythrocytes (McManus, T.J., 1987,Fed. Proc., in press). N-ethylmaleimide (NEM) chemically induces a Cl-dependent K+ transport pathway that is independent of both Na o + and Na i + . This pathway differs in many properties from the basal, Na o + -independent K+/K+ exchange active in untreated human erythrocytes at normal cell volume.     The uptake of 3H-labeled choline by a suspension of isolated type II epithelial cells from rat lung has been studied in a Ringer medium. Uptake was linear for 4 min at both 0.1 μm and 5.0 μm medium choline; at 5 μm, only 10% of the label was recovered in a lipid fraction. Further experiments were conducted at the low concentration (0.1 μm), permitting characterization of the properties of high-affinity systems. Three fractions of choline uptake were detected: (i) a sodium-dependent system that was totally inhibited by hemicholinium-3 (HC-3); (ii) a sodium-independent uptake, when Na+ was replaced by Li+, K+ or Mg2+, inhibited by HC-3; (iii) a residual portion persisting in the absence of Na+ and unaffected by HC-3. Choline uptake was sigmoidally related to the medium Na+ concentration. Kinetic properties of the uptake of 0.1 μm 3H-choline in the presence and absence of medium Na+ were examined in two ways. (a) Inhibition by increasing concentrations of unlabeled choline (0.5–100 μm) was consistent with the presence of two Michaelis-Menten-type systems in the presence of Na+; a Na+-dependent portion (a mean of 0.52 of the total) had a K m for choline of 1.5 μm while K m in the absence of Na+ (Li+ substituting) was 18.6 μm. (b) Inhibition by HC-3 (0.3–300 μm) gave Ki values of 1.7 μm and 5.0 μm HC-3 for the Na+-dependent and -independent fractions. The apparent K m of the Na+-dependent uptake is lower than that reported previously for lung-derived cells and is in the range of the K m values reported for high-affinity, Na+-dependent choline uptake by neuronal cells.     Earlier studies by our laboratory have suggested a relationship between an amiloride-sensitive Na+−H+ exchange process and the physical state of the lipids of rat colonic brush-border membrane vesicles. To further assess this possible relationship, a series of experiments were performed to examine the effect of dexamethasone administration (100 μg/100 g body wt. per day) subcutaneously for 4 days on Na+−H+ exchange, lipid composition and lipid fluidity of rat distal colonic brush-border membrane vesicles. The results of these studies demonstrate that dexamethasone treatment significantly: (1) increased the Vmax of the Na+−H+ exchange without altering the Km for sodium of this exchange process, utilizing the fluorescent pH-sensitive dye, acridine orange. 22Na flux experiments also demonstrated an increase in amiloride-sensitive proton-stimulated sodium influx across dexamethasone-treated brush-border membrane vesicles; (2) increased the lipid fluidity of treated-membrane vesicles compared to their control counterparts, as assessed by steady-state fluorescence polarization techniques using three different lipid-soluble fluorophores; and (3) increased the phospholipid content of treated-membrane vesicles thereby, decreasing the cholesterol/phospholipid molar ratio of treated compared to control preparations. This data, therefore, demonstrates that dexamethasone administration can modulate amiloride-sensitive Na+−H+ exchange in rat colonic distal brush-border membrane vesicles. Moreover, it adds support to the contention that a direct relationship exists between Na+−H+ exchange activity and the physical state of the lipids of rat colonic apical plasma membranes.  相似文献

    Ion and acid–base balance were examined in the freshwater-adapted mummichog (Fundulus heteroclitus) using a series of treatments designed to perturb the coupling mechanisms. Unidirectional Cl uptake (JClin) was extremely low whereas JNain was substantial (three- to sixfold higher); comparable differences occurred in unidirectional efflux rates (JClout, JNaout). JClin was refractory to all treatments, suggesting that Cl/base exchange was unimportant or absent. Indeed, no base excretion or modulation of ion fluxes occurred for acid–base balance for up to 8 h after NaHCO3 loading (injections of 1000 or 3000 nequiv.·g−1). Acute environmental low pH (4.5) and amiloride (10−4 M) treatments caused concurrent inhibition of JNain and net H+ excretion (JH+net), indicating the presence of Na+/H+ exchange. JNain was elevated and JH+net restored during recovery from both treatments, but this exchange did not appear to be dynamically adjusted for acid–base homeostasis. High external ammonia exposure (1 mmol·l−1) initially blocked ammonia excretion (JAmmnet) but had no effect on JNain, whereas high pH (9.4) reduced both JAmmnet and JNain. Inhibition of JNain by the low pH and amiloride treatments had no effect on JAmmnet. These results indicate that ammonia excretion is entirely diffusive and independent of both Na+uptake and the protons that are transported via the Na+/H+ coupling. In addition, ureagenesis served as a compensatory mechanism during high external ammonia exposure, as a marked elevation in urea excretion partially replaced the inhibited JAmmnet. In all treatments, changes in the Na+–Cl net flux differential were consistent with changes in JH+net measured by traditional water titration techniques, indicating that the former can be used as an estimate of the acid–base status of the fish. Overall, the results demonstrate that the freshwater-adapted F. heteroclitus does not conform to the ion/acid–base relationships described in the standard model based on commonly studied species such as trout, goldfish, and catfish.  相似文献

    A Na+-specific and Na+-stimulated active α-aminoisobutyric acid transport system was reconstituted from plasma membranes isolated from mouse fibroblast BALB/c 3T3 cells transformed by simian virus 40. The plasma membranes were treated with dimethylmaleic anhydride and then extracted with 2% cholate. The cholate-solubilized supernatant proteins were combined with exogenous phospholipids and eluted through a Sephadex G-50 column. This yielded reconstituted vesicles which in the presence of Na+ could actively transport α-aminoisobutyric acid as shown by the transient accumulation above the equilibrium level (overshoot). The overshoot was not obtained with other monovalent cations such as K+, Li+, and choline+. The electrochemical effect of the lipophilic anion, SCN?, led to greater α-aminoisobutyric acid uptake as compared to that observed with Cl? or SO42?. The Na+-stimulated transport of a-aminoisobutyric acid was a saturable process with an apparent Km of 2 mm. Studies of the inhibition of α-aminoisobutyric acid transport by other amino acids showed that methylaminoisobutyric acid [specifically transported by A system (alanine preferring)]had a pronounced inhibitory effect on a-aminoisobutyric acid uptake in contrast to the slight inhibitory effect produced by phenylalanine [primarily transported by L system (leucine preferring)]. The results show that the reconstituted vesicles, prepared from partially purified membrane proteins and exogenous phospholipids, regained the same important transport properties of native membrane vesicles, i.e., Na+-specific and Na+-stimulated concentrative α-aminoisobutyric acid uptake.  相似文献

    We measured Na+/K+ ATPase activity in homogenates of gill tissue prepared from field caught, winter and summer acclimatized yellow perch, Perca flavescens. Water temperatures were 2–4°C in winter and 19–22°C in summer. Na+/K+ ATPase activity was measured at 8, 17, 25, and 37°C. Vmax values for winter fish increased from 0.48±0.07 μmol P mg−1 protein h−1 at 8°C to 7.21±0.79 μmol P mg−1 protein h−1 at 37°C. In summer fish it ranged from 0.46±0.08 (8°C) to 3.86±0.50 (37°C) μmol P mg−1 protein h−1. The Km for ATP and for Na+ at 8°C was ≈1.6 and 10 mM, respectively and did not vary significantly with assay temperature in homogenates from summer fish. The activation energy for Na+/K+ ATPase from summer fish was 10 309 (μmol P mg−1 h−1) K−1. In winter fish, the Km for ATP and Na+ increased from 0.59±0.08 mM and 9.56±1.18 mM at 8°C to 1.49±0.11 and 17.88±2.64 mM at 17°C. The Km values for ATP and Na did not vary from 17 to 37°C. A single activation energy could not be calculated for Na/K ATPase from winter fish. The observed differences in enzyme activities and affinities could be due to seasonal changes in membrane lipids, differences in the amount of enzyme, or changes in isozyme expression.  相似文献

    Summary Intracellular pH (pHi) regulation was studied in crayfish neurons with pH-, and Na+-sensitive microelectrodes. It was confirmed to involve both a HCO 3 -dependent and a HCO 3 -independent mechanism. The latter was identified as the amiloride-sensitive Na+/H+ exchange described in vertebrate cells. Its dependence on extracellular pH (pHe) and Na+ concentration ([Na+]e) was studied in CO2-free external solutions at 20°C. The steady state pHi and the rate constant (k) of the exponential pHi recovery following an acid load were determined. At pHe=7.5 and [Na+]e=200 mM, the average steady state pHi was 7.09±0.12 (as compared to 7.30±0.10 in the presence of 5 mM bicarbonate). The dependence of the rate constant of recovery on [Na+]e could be described by Michaelis-Menten kinetics; at pHe=7.5 the apparentK m andK max were 39 mM and 1.4 mmol·l–1·min–1, respectively. Decreasing pHe reduced the rate of recovery, the variations ofk with pHe conforming to a simple titration curve with an apparent pK of 7.05±0.21. These kinetic properties of the Na+/H+ exchange in crayfish neurons are similar to those described in vertebrate cells.Preliminary results were presented at the First International Congress of Comparative Physiology and Biochemistry (Liège, Belgium, 1984)  相似文献

    Summary When the mulletMugil capito is transferred to medium lacking Ca++ (either Ca++-free seawater or distilled water) the passive permeability of the gill to Na+ and Cl is increased and the activating effect of external K+ on the Na+ and Cl effluxes in hyposaline media is inhibited. The permeability of the gill increases progressively in proportion to the time of Ca++ deprivation; it declines when Ca++ is added again to the external medium. The active mechanisms for ion excretion are not reversible. At external Ca++ concentrations from 0.1 to 10 mM the Na+ permeability is constant but the activation of Na+ efflux by K+ shows a maximum at a Ca++ concentration of about 1 mM. For activation of Cl efflux external bicarbonate must be present, in addition to Ca++, suggesting the existence of a Cl/HCO 3 exchange. The mechanism by which Ca++ controls the passive branchial permeability is thus probably different from that involved in K+ activation of ion excretion. The Ca++ effect on the K+ sensitive ionic excretory mechanisms seems to be related to intracellular Ca++ movements. Thus, on the one hand, substances such as Ruthenium Red and La+++ which both inhibit Ca++ exchange, in media containing Ca++ and HCO 3 also inhibit K+ activation of Na+ and Cl effluxes; on the other hand, the ionophore A 23187, a stimulator of Ca++ exchange, when added to these media, activates the Na+ and Cl effluxes; its maximal effect on the Na+ flux occurs at 2 mM Ca++.Abbreviations ASW-Ca artificial seawater minus calcium - DW deionised water - DWCa deionised water with 1 mM Ca++ added - DWCaHCO 3 DW with calcium plus bicarbonate - DWHCO 3 DW with 1 mM sodium bicarbonate added - FW freshwater (tap water) - FWK freshwater with K+ added - P.
