Rat dams and offspring were exposed to 1.5-g, 1.75-g or 2.0-g hypergravity (hg) from Gestational day [G] 11 until Postnatal day [P] 10. To ascertain the role of maternal factors in reduced postnatal body weights of offspring developed in hg, the dams' lactational hormones were measured. Oxytocin (OT), the major hormone responsible for milk ejection, was reduced in hg dams whereas prolactin (Prl), involved in milk production, was unchanged. Video analyses of nursing behavior revealed that hg dams spent more time nursing relative to 1-g controls. We hypothesized impaired milk transfer from dam to pup, however pup body weight gains following a discrete suckling episode were comparable across conditions. Changes in lactational hormones and nursing behavior by dams exposed to hg do not account for reduced body masses of their offspring. 相似文献

A rapid radioimmunoassay for estrone (total unconjugated and sulfated) was developed to determine plasma estrone (E1) concentrations in inseminated gilts that conceived and those that had not. Thirty-one 160 day-old prepuberal gilts were induced to ovulate with gonadotropins and were artificially inseminated 10 hr before the expected time of ovulation (Day 1 - day of insemination). Unconjugated E1 and E1SO4 were extracted from 20 to 500 microl of plasma twice with 5 ml of tetrahydrafuran:ethyl acetate (1:1). Aliquots of a standard E1SO4 preparation were dissolved in 500 microl of distilled water and extracted at the same time as the plasma samples. The dried extracts were solvolyzed for 1 hr at 50 degrees C in 0.6 ml of glacial acetic acid:ethyl acetate (1:1), and the dried residue was redissolved in 0.2 ml of distilled water and extracted once with 2 ml of diethyl ether. Twenty of 31 gilts were pregnant at Days 29 to 31 of the induced cycle. Plasma E1 in pregnant gilts increased from 85 pg/ml on Day 18 to 702, 1879 and 2793 pg/ml, respectively, on Days 22, 25 and 29 to 31. Three of the non-pregnant gilts had plasma progesterone secretion maintained until Day 22; they also had a transitory increase in plasma E1 on Day 22 (215 pg/ml). Some blastocysts may have been present to exert a temporary luteotropic effect, but not enough blastocysts to completely overcome the luteolytic effect of the uterus. Quantification of plasma E1SO4 could be used as a pregnancy test in the pig. 相似文献

Peripubertal gilts (n = 25) were treated with corn oil (CO) or ovarian steroids, one month following an ovariectomy. The first day of treatment was assigned as the first day of the experiment. The gilts received: Group (Gr) I (n = 4)--CO (2 mL x day(-1) from 1st to 12th day), Gr II (n = 4) and Gr III (n = 4)--progesterone (P4; 10 to 100 mg x day(-1) from 1st to 12th day), Gr IV (n = 5)--estradiol benzoate (EB; 400 microg x day(-1) from 1st to 3rd day), Gr V (n = 4) and Gr VI (n = 4)--EB + P4 (EB 400 microg x day(-1) from 1st to 3rd day, 20 microg x day(-1) at 6th and 9th day, 50 microg at 12th day plus P4 10 to 100 mg from 4th to 15th day). All gilts were injected with oxytocin (OT; 20 IU; i.v.) on the following days of the experiment: 13th (Gr I and Gr II), 15th (Gr III and Gr IV), 16th (Gr V) and 18th (Gr VI). Concentrations of the PGF2alpha metabolite--PGFM were determined in blood samples, collected from 30 min before to 120 min after OT injection. Baseline PGFM concentrations (30 min before OT) differed among treatment groups and were the highest in Gr V and Gr VI (P < 0.01 vs. other groups). The magnitude of the PGFM response to OT increased only in four of the five gilts of Gr IV and in three of the four gilts of Gr VI, and it was higher (P = 0.009) in Gr VI than in Gr IV. In the remaining groups, PGFM concentrations did not increase above the baseline in response to OT. The day after OT injection, oxytocin receptors (OTR) were found in the uterine tissues of all animals studied. The lowest OTR concentrations were in Gr I--75.5 +/- 11.2 fmol x mg protein(-1) and the highest in Gr IV--712.9 +/- 86.7 fmol x mg protein(-1); (P < 0.05 vs. other groups). The values of K of OTR differed among groups (P < 0.001) and ranged from 1.62 +/- 0.44 nM in Gr I to 12. 08 +/- 1.9 nM in Gr VI. A positive correlation (r = 0.54; P < 0.01) between plasma E2 and uterine OTR concentrations was observed. In conclusion, E2 and P4 are involved in both PGF2 synthesis/secretion and OTR formation, however, full PGF response to OT does not develop before puberty. The neuropeptide oxytocin (OXT) is a key factor in the initiation and regulation of sociosexual behavior. The present study analyzes the effects of cohabitation and social challenge on plasma OXT concentration rates in guinea pig pairs in relation to male sociosexual behavior. The cohabitation phase lasted 3 days. On day 4, the pair was socially challenged by introducing an unfamiliar male. Displayed male sexual behavior varied significantly during cohabitation, with peaks on day 1. Sociopositive behavior, i.e., side-by-side contact, was increased on days 3 and 4. Cohabitation per se led to elevated plasma OXT concentrations only in males. In contrast, both sexes reacted with increased plasma OXT concentrations to the social challenge (day 4). At that time, male OXT was significantly correlated with sexual behavior and female OXT with sociosexual behavior received from the partner. Additionally, pairs were synchronized in their OXT release during days 3 and 4. We conclude that cohabitation causes sexually dimorphic plasma OXT concentration patterns in guinea pigs. Secondly, the conformity of OXT release in both sexes may represent an endocrine marker for long-term cohabitation, which is reflected behaviorally by increased spatial proximity. 相似文献

To determine if administration of the anaesthetic cocktail, telazol-ketamine-xylazine (TKX) and pentothal (PEN) decreases serum concentrations of luteinizing hormone (LH) in pigs, the following experiment was performed. On day 1, eight gilts and six barrows of similar weight (75 kg) were anaesthetized with TKX (1 mL/22.5 kg body weight [BW] intramuscularly) and indwelling jugular catheters were inserted. On days 2, 6 and 8 blood samples were taken every 20 min, for 4 h before pigs were administered saline (day 2) or anaesthetized with TKX (day 6) and PEN (8.9 mg/kg BW intravenously, day 8). Blood samples were taken every 20 min for 4 h following administration of saline and anaesthetics. Mean serum concentrations of LH and cortisol did not differ (P > 0.05) within barrows or gilts from before administration of saline (day 2) to following saline administration. Mean serum concentrations of LH and cortisol were not different (P > 0.05) within barrows and gilts before administration of TKX (pre-TKX) or PEN compared with day 2 samples. Following administration of TKX (post-TKX), mean serum concentrations of LH decreased (P < 0.05) and remained decreased for 140 min, while mean serum concentrations of cortisol increased (P < 0.05) post-TKX and remained elevated for 140 min. In gilts, mean serum concentrations of LH did not differ (P > 0.05) from pre- to post-TKX. However, mean serum concentrations of cortisol increased (P < 0.05) post-TKX in gilts and remained elevated for 240 min. Following administration of PEN, mean serum concentrations of LH and cortisol within barrows and gilts were not different (P > 0.05) from concentrations before administration of PEN. Administration of TKX to barrows and gilts increased serum concentrations of cortisol, but transient decreases in serum concentrations of LH were observed only in barrows, indicating gonadal status and/or sex may influence the ability of TKX to alter circulating concentrations of LH in pigs. 相似文献

The hormone oxytocin (OT) is released both centrally and peripherally during and after mating. Although research in humans suggests a central role in sexuality, the most reliable findings to date involve peripheral activation. This review will discuss these results and will particularly focus on understanding the most recent findings from fMRI data and the effects of exogenous peripheral OT administration. We will then consider hypotheses of the roles played by central and systemic OT release as well as their control and modulation in the female, summarizing recent findings from animal research. The mating behaviour of 28 gilts was studied. The gilts were reared under two different social conditions known to affect both their puberty attainment and reproductive parameters during early pregnancy. The different social conditions were applied from an average age of 137 days onwards. Ten gilts were housed individually, having neither tactile nor visual contact with other pigs. The remaining gilts ( n=18) were housed pairwise, having additional contact with gilts in adjacent pens and daily boar contact from 180 days of age onwards. At third oestrus, the gilts were artificially inseminated and subsequently introduced to one of three vasectomized boars for a period of 20 min. The gilts were slaughtered 10±1 days after insemination. The mating behaviour varied considerably between individual gilts, partly because of differences in mating behaviour between the two groups of gilts. More (P<0.05) individually housed gilts showed a standing response latency upon introduction of the boar. During this latency period, the individually housed gilts initiated contact with the boar. Once the standing response was elicited, mating behaviour was similar in gilts of both social groups. One individually housed gilt did not show a standing response and consequently was not mated. The mating behaviour of the boars did not differ for the gilts of the two social conditions. It was concluded that the social conditions of gilts during rearing affected their introductory sexual behaviour. The relationship with reproductive performance during early pregnancy is discussed. 相似文献

It has been proposed that short-term activation of the hypothalamo-pituitary adrenal axis, with a consequent increase in the secretion of cortisol, may disrupt the endocrine events prior to ovulation and thereby impair reproduction in females. We investigated this concept in gilts in which oestrus was detected by introduction to boars, where intense physical contact is possible, or by applying pressure to the back of gilts (back-pressure test) during fence-line exposure to boars, where intense physical contact is prohibited. We expected that there would be a greater release of cortisol and that reproduction would be inhibited in gilts introduced to boars compared to gilts in which the back-pressure test was used. As expected, introduction of gilts to boars resulted in a significant transient increase in plasma concentrations of cortisol while there was no significant effect of using the back-pressure test on plasma cortisol. Nevertheless, introduction of gilts to boars did not impair reproduction and there was no effect of method of detecting oestrus on duration of oestrus, sexual receptivity, fertility or fecundity. The length of the oestrous cycle was decreased and ovulation rate increased in gilts that were introduced to boars compared to gilts that underwent the back-pressure test, indicating that introduction of gilts to boars may have stimulated these aspects of reproduction. These stimulatory effects may have been due to an increased exposure of gilts to sexual behaviour and stimuli from boars when introduced to boars and/or to stimulatory effects of the hypothalamo-pituitary adrenal axis on some aspects of reproduction. 相似文献

Fourteen crossbred beef bulls were assigned at random to receive one of four sexual stimuli treatments. Treatments consisted of: (1) controls (four bulls), no visual or physical contact with any cows; (2) false mount (two bulls), allowed to mount an estrual cow; (3) mated (four bulls), allowed to mount an estrual cow with intromission and ejaculation; (4) electroejaculated (four bulls), no exposure to cows. Serum from blood samples taken at 15-min intervals from -15 min to 2 hr from sexual stimuli were radioimmunoassayed for luteinizing hormone (LH). Four bulls had elevated levels at the pretreatment bleeding, but none of the stimuli induced or were associated with LH releases. Basal levels of LH were consistent within bulls but varied considerably among bulls. Conclusion is that stimuli associated with mating do not cause a release of LH. 相似文献

In plasma of newborn piglets 12.6 +/- 2.9 mug cortisol/100 ml was estimated radioimmunologically. During the first 24 hours of life the concentration decreases to 1/3 of the starting level. After application of 5 IU ACTH/kg body weight, there was in newborn as in 5-day-old piglets an increase in the concentration of cortisol, while an influence on the level of glucose in plasma could not be detected in 0-day-old animals. 相似文献

Twenty-four gilts detected in estrus were fed a barley-soybean meal diet which was essentially free of vitamin A and β-carotene. At the second estrus, gilts were randomly assigned to be fed daily the same basal control diet (CON), CON + 8200 IU vitamin A in the diet (VIT), CON + 4100 IU vitamin A + 8.2 mg β-carotene in the diet (CAR), or VIT + 16.4 mg β-carotene injected intramuscularly every other day (INJ). All gilts were sham-bred at the third estrus (day 0). On day 15, uterine secretions were flushed and uterine protein fractions were quantified after sequential chromatographic separations on Sephadex G-200 and G-50. Acid phosphatase activity in uterine flushings was measured also. Results showed nonsignificant treatment differences in the number of corpora lutea, ovarian weight, length of the estrous cycle and concentrations of plasma progesterone on days 0, 3, 6, 9, 12 and 15. The amount of total secretory protein was not different among treatment groups. However, CON had the lowest quantity of serum-like proteins and uterine-specific proteins and in the total and specific acid phosphatase activities. Group INJ tended to have the highest amount of each uterine protein fraction examined. Thus, deficiencies in vitamin A and β-carotene decreased certain uterine protein components while injected β-carotene tended to have an opposite effect. Such differences in uterine proteins may influence embryonic survival in pigs. 相似文献

This study was designed to assess effects of exogenous oxytocin (OT) on uterine activity, and to compare three different sexual stimuli in their effects on OT release, uterine activity and receptive behavior in estrous sows. Uterine activity was recorded nonsurgically, by transcervical insertion of an open-end catheter into the caudal part of the uterine lumen. After recording spontaneous uterine activity, exogenous OT was administered (Experiment 1), or one of the following stimuli was applied to the sow (Experiment 2): tactile stimuli, i.e. manual stimulation of the sow's back and flanks, tactile stimulation in combination with boar pheromone spray (5alpha-androstenon), or tactile stimuli in the presence of a boar. Both exogenous OT and endogenously released OT increased uterine activity. The effect depended on the uterine activity before treatment, with the effect being greater in those sows with lower uterine activity before treatment. In Experiment 2, boar presence was the only stimulus that elicited a clear, surge-like release of OT, and also clearly increased uterine activity. Release of OT was not necessary for induction of receptive behavior: tactile stimulation alone and in combination with pheromone spray elicited a standing response in one third of the sows, but had no effect on OT release. 相似文献

In order to investigate the significance of oxytocin in pregnancy and labor, oxytocin concentrations in plasma and cerebrospinal fluid (CSF) were determined using the specific radioimmunoassay. Plasma and CSF samples were obtained from 23 pregnant women (11 pre labor, 12 in labor), 15 nonpregnant women and 4 men at spinal puncture for anesthesia. In males and nongravidas, CSF levels of oxytocin were significantly higher than plasma levels. Plasma levels in pregnant patients pre or in labor were significantly higher than those in nongravidas. No significant difference between CSF levels in prelabor gravidas (mean +/- SE, 9.7 +/- 1.5 mu u/ml) and nongravidas (10.1 +/- 1.2 mu u/ml) was found. However, CSF levels in gravidas in labor (18.6 +/- 2.3 micromicrons/ml were significantly higher than the levels in prelabor gravidas. These results strongly suggest that oxytocin levels in human plasma and CSF are controlled by different mechanisms and that the increased oxytocin could have some specific central actions. 相似文献

The effect of adrenocorticotropic hormone (ACTH) administration on plasma cortisol concentrations was determined in pregnant gilts and their fetuses. In a first experiment, 100 IU ACTH (Synacthen Depot) was administered intramuscularly to the gilts every second day from Days 49 to 75 of gestation. ACTH injections were carried out at 08:00 h and, thereafter, 10 blood samples were taken within the following 8h via jugular catheters. Blood samples were analysed for plasma cortisol concentrations, and results were compared with values from animals which were treated with physiological saline and untreated animals (blood sampling only). The values for plasma cortisol concentrations increased until 3h after ACTH applications to a mean maximum level of 276.5+/-17.2 nmol/l in the whole 4-week stimulation period. Plasma cortisol levels did not return to pre-treatment values within the 8 h post-injection. No differences in cortisol levels were found between the physiological saline and untreated control, and no habituation of the adrenocortical response to ACTH was found during the 4-week stimulation period. In a second experiment, 100 IU ACTH were administered to pregnant gilts at gestation Day 65. After 3 h, fetuses were recovered under general anaesthesia and blood samples were taken from the umbilical vein, artery, and, after decapitation, from periphery. Application of ACTH to the sows significantly increased their plasma cortisol concentrations (P<0.001), and also increased plasma cortisol concentrations in peripheral blood samples from the fetuses (P=0.09) and in the umbilical vein (P<0.001) and artery (P<0.01), respectively. Plasma ACTH concentrations did not differ in fetuses from ACTH-treated or control sows. The results show that in gilts the adrenocortical response to an exogenous application of Synacthen Depot is consistent over time during mid-gestation. Furthermore, cortisol but not ACTH levels were increased in fetuses from ACTH-treated sows, indicating that maternal cortisol can cross the placenta during mid-gestation. The stimulation of maternal cortisol release through exogenous ACTH with subsequent elevation of fetal cortisol levels is, therefore, a useful approach for studying effects of elevated maternal glucocorticoids in prenatal stress studies in pigs. 相似文献

The increase in serum cortisol concentrations following naloxone administration to female pigs was abolished by hypophysial stalk-transection, even though CRH and ACTH stimulated cortisol release in these animals.
