以较耐盐花生品种‘花育25’、‘鲁花12’和盐敏感品种‘海花1’、‘花育20’为材料,采用盆栽试验,设置0、1.0、2.0、3.0 g/kg土壤NaCl胁迫浓度梯度,测定其净光合速率、表观量子效率、气孔导度等光合特性,以及抗氧化酶活性和渗透调节物质含量等指标,明确NaCl胁迫条件下不同耐盐性花生品种光合和生理生化特性的适应特征。结果表明:(1)NaCl胁迫明显抑制各品种花生叶片光合作用,净光合速率随盐胁迫浓度的升高呈明显降低的趋势。(2)各品种花生叶片净光合速率均先随光照强度的增强而升高,当光强达到一定数值时趋于平稳;光补偿点和光饱点因品种和盐胁迫浓度差异较大,较高的盐胁迫浓度使叶片光补偿点升高,盐敏感品种的光饱和点降低。(3)盐胁迫条件下,各品种叶片表观量子效率和最大净光合速率均随盐胁迫强度的增加呈显著降低趋势,盐敏感品种利用弱光的能力在低盐胁迫下强于耐盐品种,其最大净光合速率在较高盐胁迫浓度(3.0 g/kg)下明显低于耐盐品种,但两类品种的叶片表观量子效率降幅相近(78.65%~88.00%)。(4)在NaCl胁迫下,耐盐品种叶片自由水含量显著高于盐敏感品种;在2.0~3.0 g/kg NaCl胁迫下,耐盐品种叶片SOD、CAT、POD活性和MDA含量的升降幅度均低于盐敏感品种;耐盐品种在NaCl浓度低于2.0 g/kg时的抗氧化能力明显高于盐敏感品种。研究发现,盐胁迫下花生品种抗盐耐逆的主要生理响应特征是提高光补偿点和最大净光合速率,增强叶片持水能力和物质代谢能力,以及提升抗氧化和渗透调节能力。  相似文献

Water deficit is the major yield‐limiting factor of crop plants. The exposure of plants to this abiotic stress can result in oxidative damage due to the overproduction of reactive oxygen species. The aim of this work was to study the antioxidant‐stress response of drought‐tolerant (SP83‐2847 and SP83‐5073) and drought‐sensitive (SP90‐3414 and SP90‐1638) sugarcane varieties to water‐deficit stress, which was imposed by withholding irrigation for 3, 10 and 20 days. The drought‐sensitive varieties exhibited the lowest leaf relative water content and highest lipid peroxidation, hydrogen peroxide (H2O2) and proline contents during the progression of the drought‐stress condition. The antioxidant enzymes superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (GPOX) and glutathione reductase (GR) activities changed according to variety and stress intensity. SP83‐2847 exhibited higher CAT and APX activities than the other varieties in the early stage of drought, while the activities of GPOX and GR were the highest in the other varieties at the end of the drought‐stress period. A Cu/Zn SOD isoenzyme was absent at the end of drought period from the SP90‐3414‐sensitive variety. The results indicate that lipid peroxidation and early accumulation of proline may be good biochemical markers of drought sensitivity in sugarcane.  相似文献

Aims

A causal relationship between salinity and oxidative stress tolerance and a suitability of using root antioxidant activity as a biochemical marker for salinity tolerance in barley was investigated.

Methods

Net ion fluxes were measured from the mature zone of excised roots of two barley varieties contrasting in their salinity tolerance using non-invasive MIFE technique in response to acute and prolonged salinity treatment. These changes were correlated with activity of major antioxidant enzymes; ascorbate peroxidase, catalase, and superoxide dismutase.

Results

It was found that genotypic difference in salinity tolerance was largely independent of root integrity, and observed not only for short-term but also long-term NaCl exposures. Higher K+ retention ability (and, hence, salinity tolerance) positively correlated with oxidative stress tolerance. At the same time, antioxidant activities were constitutively higher in a sensitive but not tolerant variety, and no correlation was found between SOD activity and salinity tolerance index during large-scale screening.

Conclusion

Although salinity tolerance in barley correlates with its oxidative stress tolerance, higher antioxidant activity at one particular time does not correlate with salinity tolerance and, as such, cannot be used as a biochemical marker in barley screening programs.  相似文献

Bermudagrass (Cynodon dactylon) is a widely used warm-season turfgrass and one of the most drought tolerant species. Dissecting the natural variation in drought tolerance and physiological responses will bring us powerful basis and novel insight for plant breeding. In the present study, we evaluated the natural variation of drought tolerance among nine bermudagrass varieties by measuring physiological responses after drought stress treatment through withholding water. Three groups differing in drought tolerance were identified, including two tolerant, five moderately tolerant and two susceptible varieties. Under drought stress condition, drought sensitive variety (Yukon) showed relative higher water loss, more severe cell membrane damage (EL), and more accumulation of hydrogen peroxide (H2O2) and malondialdehyde (MDA), while drought tolerant variety (Tifgreen) exhibited significantly higher antioxidant enzymes activities. Further results indicated that drought induced cell injury in different varieties (Yukon, SR9554 and Tifgreen) exhibited liner correlation with leaf water content (LWC), H2O2 content, MDA content and antioxidant enzyme activities. Additionally, Tifgreen plants had significantly higher levels of osmolytes (proline level and soluble sugars) when compared with Yukon and SR9554 under drought stress condition. Taken together, our results indicated that natural variation of drought stress tolerance in bermudagrass varieties might be largely related to the induced changes of water status, osmolyte accumulation and antioxidant defense system.  相似文献

In the phytotron experiment, the effect of elevated atmospheric CO2 (EC, 750 μmol mol?1) on the drought tolerance was studied in two winter varieties (Mv Mambo, tolerant; Mv Regiment, moderately tolerant) and in one spring variety of wheat (Lona, sensitive to drought). Changes in net photosynthetic rate (P N), stomatal conductance, transpiration, wateruse efficiency, effective quantum yield of photosystem II, and activities of glutathione reductase (GR), glutathione-Stransferase (GST), guaiacol peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX) were monitored during water withdrawal. Drought caused a faster decline of P N at EC, leading to the lower assimilation rates under severe drought compared with ambient CO2 (NC). In the sensitive variety, P N remained high for a longer period at EC. The growth at EC resulted in a more relaxed activation level of the antioxidant enzyme system in all three varieties, with very low activities of GR, GST, APX, and POD. The similar, low values were due to decreases in the varieties which had higher ambient values. A parallel increase of CAT was, however, recorded in two varieties. As the decline in P N was faster at EC under drought but there was no change in the rate of electron transport compared to NC values, a higher level of oxidative stress was induced. This triggered a more pronounced, general response in the antioxidant enzyme system at EC, leading to very high activities of APX, CAT, and GST in all three varieties. The results indicated that EC had generally favourable effects on the development and stress tolerance of plants, although bigger foliage made the plants more prone to the water loss. The relaxation of the defence mechanisms increased potentially the risk of damage due to the higher level of oxidative stress at EC under severe drought compared with NC.  相似文献

The effects of salt stress were studied on the accumulation and metabolism of proline and its correlation with Na+ and K+ content in shoots and callus tissue of four potato cultivars, viz., Agria, Kennebec (relatively salt tolerant), Diamant and Ajax (relatively salt sensitive). Na+ and proline contents increased in all cultivars under salt stress. However, K+ and protein contents decreased in response to NaCl treatments. The activities of enzymes involved in proline metabolism, Δ1-pyrroline-5-carboxylate synthetase (P5CS) and proline dehydrogenase (ProDH) increased and decreased, respectively, in response to elevated NaCl concentrations. The changes of P5CS and ProDH activities in more salt sensitive cultivars (Diamant, Ajax) were more than those in the tolerant ones. Then the stimulation of synthesis in combination with a partially increase of protein proteolysis, a decrease in proline utilization and inhibition of oxidation resulted in high proline contents in seedlings and calli under salt stress. In callus tissue, reduced growth and cell size may be partially responsible for high proline accumulation in response to high NaCl levels. However, although the basic proline contents in the seedlings of more salt tolerant cultivars were higher than the sensitive ones, a clear relationship was not generally observed between accumulation of proline and salt tolerance in potato.  相似文献

Salinity is one of the most severe abiotic stresses for crop production. The present study investigates the salinity-induced modulation in growth indicators, morphology and movement of stomata, photosynthetic pigments, activity of carbonic anhydrase as well as nitrate reductase, and antioxidant systems in two varieties of chickpea (Pusa-BG5023, and Pusa-BGD72). On 20th day of sowing, plants were treated with varying levels of NaCl (0, 50, 100, 150 and 200 mM) followed by sampling on 45 days of sowing. Recorded observations on both the varieties reveal that salt stress leads to a significant decline in growth, dry biomass, leaf area, photosynthetic pigments, protein content, stomatal behavior, cell viability, activity of nitrate reductase and carbonic anhydrase with the rise in the concentration of salt. However, quantitatively these changes were less in Pusa-BG5023 as compared to Pusa-BGD72. Furthermore, salinity-induced oxidative stress enhanced malondialdehyde content, superoxide radicals, foliar proline content, and the enzymatic activities of superoxide dismutase, catalase, and peroxidase. The variety Pusa-BGD72 was found more sensitive than Pusa-BG5023 to salt stress. Out of different graded concentrations (50, 100, 150 and 200 mM) of sodium chloride, 50 mM was least toxic, and 200 mM was most damaging. The differential behavior of these two varieties measured in terms of stomatal behavior, cell viability, photosynthetic pigments, and antioxidant defense system can be used as prospective indicators for selection of chickpea plants for salt tolerance and sensitivity.  相似文献

The growth of the wild-type and three salt tolerant mutants of barnyard grass ( Echinochloa crusgalli L.) under salt stress was investigated in relation to oxidative stress and activities of the antioxidant enzymes superoxide dismutase (SOD: EC 1.15.1.1), catalase (CAT: EC 1.11.1.6), phenol peroxidase (POD: EC 1.11.1.7), glutathione reductase (GR: EC 1.8.1.7) and ascorbate peroxidase (APX: EC 1.11.1.1). The three mutants ( fows B17, B19 and B21) grew significantly better than the wild-type under salt stress (200 m M NaCl) but some salt sensitive individuals were still detectable in the populations of the mutants though in smaller numbers compared with the wild-type. The salt sensitive plants had slower growth rates, higher rates of lipid peroxidation and higher levels of reactive oxygen species (ROS) in their leaves compared with the more tolerant plants from the same genotype. These sensitivity responses were maximized when the plants were grown under high light intensity suggesting that the chloroplast could be a main source of ROS under salt stress. However, the salt sensitivity did not correlate with reduced K +/Na + ratios or enhanced Na + uptake indicating that the sensitivity responses may be mainly because of accumulation of ROS rather than ion toxicity. SOD activities did not correlate to salt tolerance. Salt stress resulted in up to 10-fold increase in CAT activity in the sensitive plants but lower activities were found in the tolerant ones. In contrast, the activities of POD, APX and GR were down regulated in the sensitive plants compared with the tolerant ones. A correlation between plant growth, accumulation of ROS and differential modulation of antioxidant enzymes is discussed. We conclude that loss of activities of POD, APX and GR causes loss of fine regulation of ROS levels and hence the plants experience oxidative stress although they have high CAT activities.  相似文献

Field drought studies were performed in order to assess oxidative stress, proteolytic activity and yield loss under natural stress conditions. Flag leaves of two drought-tolerant (Yantar and Zlatitsa) and two drought-sensitive (Miziya and Dobrudjanka) winter wheat varieties were analyzed. Stress intensity was assessed by relative electrolyte leakage and proline accumulation. Senescence progression was followed by loss of chlorophyll and protein. Lipid peroxidation, H2O2 content, activities of superoxide dismutase (SOD), catalase (CAT), and non-specific peroxidase (GPX) isoforms, as well as proteolytic activities were analyzed from heading throughout grain filling. Weakening of membrane integrity and oxidative damage to lipids were more pronounced in the sensitive varieties under field drought. The activities of Fe- and Cu/Zn SOD isoforms decreased in the controls, but remained high in drought-treated plants. The activities of MnSOD isoforms and CAT were enhanced towards grain filling, especially in the sensitive varieties under drought. GPX activities were rised under drought but progressively diminished. Accelerated senescence under field drought was linked to higher proteolytic activity with variety specific differences in the protease response, but without a clear correlation to drought resistance or sensitivity. Field drought led to higher oxidative stress more pronounced for drought sensitive varieties, especially during the grain filling period.  相似文献

The investigation was carried out to study the effect of halopriming on NaCl and polyethylene glycol-6000 (PEG-6000) induced stress tolerance potential of three Vigna radiata (L.) Wilczek varieties, with varied abiotic stress tolerance potential. Halopriming is a seed priming technique in which the seeds were soaked in various salt solutions (in this study NaCl was used). The results of the study indicated that the application of stresses (both NaCl and PEG) induced retardation of growth attributes (measured in terms of shoot length, fresh weight, dry weight) and decrease in physiological attributes like total chlorophyll content, metabolites, photosynthetic and mitochondrial activity of the seedlings in all three V. radiata (L.) varieties. However, halopriming of the seeds could reduce the extent of decrease in these biological attributes. NaCl and PEG stress also caused increase in MDA content (a product of membrane lipid peroxidation) in all the varieties studied and this increase was significantly minimized under halopriming. From the present investigation it was evident that among the green gram varieties studied, Pusa Vishal, a NaCl tolerant variety showed enhanced tolerance to NaCl and PEG induced stress, when the seeds were subjected to halopriming followed by Pusa Ratna (stress sensitive variety). Pusa 9531 (drought tolerant variety) also showed positive halopriming effects but it was less significant when compared to other two varieties. It could be concluded that halopriming improved the drought and salinity stress tolerance potential of all varieties and it was significantly higher in the Pusa Vishal as compared to Pusa 9531 and Pusa Ratna.  相似文献

Drought is one of the main factors affecting the productivity of agricultural crops, and plants respond to such stress by activating various physiological and biochemical mechanisms against dehydration. The present study investigated two varieties of sugarcane (Saccharum spp.) with contrasting responses to drought (RB867515, more tolerant; and RB855536, less tolerant) and subjected them to progressive drought conditions (2, 4, 6 and 8 days) followed by rehydration. Drought caused a decrease in water potential (ψw) and osmotic potential (ψos) in the leaves, which recovered to normal levels after rehydration only up to the fourth day of drought. Water stress changed the carbon metabolism of leaves by reducing starch and sucrose contents and increasing glucose and fructose contents in both varieties. Water deficit caused a significant reduction in the maximum quantum efficiency of photosystem II (Fv/Fm) and effective quantum yield (ΦPSII) in both varieties; however, RB867515 recovered faster after rehydration. Under water stress, the more tolerant variety RB867515 exhibited increased activity of the antioxidant enzymes catalase, ascorbate peroxidase and superoxide dismutase compared with the RB855536 variety. The results suggest that RB867515 is more tolerant to drought conditions because of a more efficient antioxidant system, which results in reduced photosynthesis photoinhibition during water stress, thus revealing itself as a potential physiological marker for drought tolerance studies.  相似文献

The role of plant antioxidant systems in stress tolerance was studied in leaves of three contrasting wheat genotypes. Drought imposed at two different stages after anthesis resulted in an increase in H2O2 accumulation and lipid peroxidation and decrease in ascorbic acid content. Antioxidant enzymes like superoxide dismutase, ascorbate peroxidase and catalase significantly increased under water stress. Drought tolerant genotype C 306 which had highest ascorbate peroxidase and catalase activity and ascorbic acid content also showed lowest H2O2 accumulation and lipid peroxidation (malondialdehyde content) under water stress in comparison to susceptible genotype HD 2329 which showed lowest antioxidant enzyme activity and ascorbic acid content and highest H2O2 content and lipid peroxidation. HD 2285 which is tolerant to high temperature during grain filling period showed intermediate behaviour. Superoxide dismutase activity, however, did not show significant differences among the genotypes under irrigated as well as water stress condition. It seems that H2O2 scavenging systems as represented by ascorbate peroxidase and catalase are more important in imparting tolerance against drought induced oxidative stress than superoxide dismutase alone. Role of Ca2+ in Drought Stress Signaling in Wheat Seedlings   总被引:1,自引:0,他引:1  
Plants use complex signal transduction pathways to perceive and react to various biotic and/or abiotic stresses. As a consequence of this signaling, plants can modify their metabolism to adapt themselves to new conditions. One such change is the accumulation of proline in response to drought and salinity stresses. We have studied drought and salinity induced proline accumulation and the roles of Ca2+ (10 mM) and indoleacetic acid (IAA, 0.3 mM) in this response. Subjecting seedlings to both drought (6% polyethylene glycol, PEG) and salinity (150 mM NaCl) stress resulted in a dramatic increase in proline accumulation (7-fold higher than control level). However, the application of Ca2+ along with these stress factors had different effects. Unlike the salinity stress, Ca2+ prevented the drought induced proline accumulation indicating that these stress factors use distinct signaling pathways to induce similar responses. Experiments with IAA and EGTA (10 mM) supported this interpretation and suggested that Ca2+ and auxin participate in signaling mechanisms of drought-induced proline accumulation. Drought and salt stress-induced proline accumulation was compared on salt resistant (cv. Gerek 79) and salt sensitive (cv. Bezostaya) wheat varieties. Although proline level of the first was twofold lower than that of the second in control, relative proline accumulation was dramatically higher in the case of the salt resistant wheat variety under stress conditions.  相似文献

Seedlings of selected six genotypes of maize (Zea mays L.) differing in their drought sensitivity (LM5 and Parkash drought-tolerant and PMH2, JH3459, Paras and LM14 as drought-sensitive) were exposed to 72 h drought stress at two leaf stage. Alterations in their antioxidant pools combined with activities of enzymes involved in defense against oxidative stress were investigated in leaves. Activities of some reactive oxygen species (ROS)-scavenging enzymes, catalase (CAT) and ascorbate peroxidase (APX) were enhanced in tolerant genotypes in response to drought stress. Superoxide dismutase (SOD) activity was significantly decreased in sensitive genotypes, but remained unchanged in tolerant genotypes under stress. Peroxidase (POX) activity was significantly induced in tolerant, as well as sensitive genotypes. Imposition of stress led to increase in H2O2 and malondialdehyde (MDA, a marker for lipid peroxidation) content in sensitive genotypes, while in tolerant genotypes no change was observed. Significant increase in glutathione content was observed in sensitive genotypes. Ascorbic acid pool was induced in both tolerant and sensitive genotypes, but induction was more pronounced in tolerant genotypes. Significant activation of antioxidative defence mechanisms correlated with drought-induced oxidative stress tolerance was the characteristic of the drought tolerant genotypes. These studies provide a mechanism for drought tolerance in maize seedlings.  相似文献

The study presents the impact of drought stress on five finger millet varieties (PR202, VL146, VL315, PES400 and VR708), representing contrasting areas of Indian sub-continent. Drought stress induced increase in the activity of superoxide dismutase, ascorbate peroxidase and glutathione reductase was higher in PR202 and VL315, while the activity was lower in the varieties PES400 and VR708. Ascorbate peroxidase : superoxide dismutase ratio, which is a crucial factor in alleviating drought stress, was higher in varieties PR202 and VL315, whilst the varieties PES400 and VR708 exhibited a lower ratio under stress. The variety PES400 recorded maximum stress induced damage, as indicated by higher accumulation of malondialdehyde and hydrogen peroxide; whereas the variety PR202 recorded least stress induced cytotoxic damage. The results clearly indicate that better drought tolerance of the variety PR202 is positively related to the capacity of its antioxidant system to scavenge reactive oxygen species, resulting in a reduced incidence of oxidative damage. Ascorbate peroxidase : superoxide dismutase ratio is found to be a critical factor governing the stress tolerance potential of different varieties. Therefore, varieties PR202 and VL315 were found to be tolerant while PES400 was susceptible to drought stress.  相似文献

Salt tolerance of sorghum varieties in terms of fresh weight, ion accumulations, proline content and peroxidase activity was analyzed in this study. Three sorghum varieties, Payam, Kimia, and Jambo, differing in salt tolerance, were grown in a greenhouse-hydroponic culture with a complete nutrition solution to which 0, 50, 100, 150 and 200 mM NaCl was added. Plant roots and leaves were harvested at 15 and 30 days after treatment and subjected to analysis. Clear decline in K+ and Ca2+ concentrations and increase in Na+ and proline contents were observed in the root and leaf tissues at each NaCl concentration in all varieties during the NaCl treatment. The Ca2+ concentration in leaves was higher than in roots, and had the following order in the tested cultivars: Jambo, Kimia, and Payam. Total peroxidase activity increased under salinity stress and it was proportional with the salt concentration. Payam had the largest decrease (46.95%) in fresh weight caused by NaCl, while Jambo had the lowest decrease, 28.63%. Linear regression analysis revealed significant relationships between the estimated factors and fresh weight. The profiles of isoperoxidases were modified under stress conditions. Two isoforms, A1 and A2, were detected in all three varieties with different intensities. Under NaCl stress, isoperoxidases were strongly expressed and a third isoform, A3, was specifically found in variety Jambo suggesting that A3 is implicated in salt adaptation of this variety.  相似文献

Summary This study provides first-hand information on the salinity and copper-induced oxidative damage and its protection in Anabaena doliolum by the antioxidant defence system. Oxidative damage measured in terms of lipid peroxidation, electrolyte leakage and H2O2 production was induced by different concentrations of NaCl and Cu2+. A greater electrolyte leakage by NaCl than Cu2+ supported the hypothesis of salinity being more injurious than copper. To explore the survival strategies of A. doliolum under NaCl and Cu stress, enzymatic antioxidant activities e.g. superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR) and nonenzymatic antioxidant contents such as glutathione reduced (GSH), ascorbate, α-tocopherol, and carotenoid were measured. A general induction in SOD and APX activities as well as ascorbate and α-tocopherol contents was found under NaCl and Cu2+ stress. In contrast to this, an appreciable decline in GR activity, GSH pool and carotenoid content under Cu2+ and an increase under NaCl stress were observed. CAT activity was completely inhibited at high doses of NaCl but stimulated following Cu2+ treatment. The above results suggest the involvement of APX and CAT in the scavenging of H2O2 under Cu2+ stress. In contrast to this, only APX was involved in H2O2 scavenging under salt stress. Our postulate of Cu2+-mediated antagonism of salt stress can be explained by a conceivable reversion of Na+-induced disturbance of cellular homeostasis by redox active Cu2+.  相似文献
