The presence of adenylate cyclase (ATP pyrophosphate-lyase (cyclizing) EC 4.6.1.1) activity was demonstrated in human erythrocyte ghosts and was found to be around 3 pmol adenosine ′,5′-monophosphatase (cyclic AMP) · 2 h?1 · mg?1 protein. This enzymatic activity is strongly stimulated by NaF and 5′-guanylimidodiphosphate, is slightly stimulated by epinephrine, norephrine, soproterenol, and prostaglandin E, and is inhibited by calcium. The hormone stimulation is not potentiated by 5′-guanylylimidodiphosphate.  相似文献

This study has examined the effect of MDA, an end product of lipid peroxidation, on the viscosity of AA and SS RBC suspensions. MDA accumulation in RBC was accomplished in vitro in human RBC by treating them with exogenous standard MDA. MDA accumulation assessed by the thiobarbituric acid reactivity of in vitro MDA-treated RBC was comparable to that of the RBC in vivo in hemolytic anemias. There was a significant increase in the viscosity of both AA and SS RBC suspensions after in vitro treatment with MDA. The increase in viscosity or RBC was significantly positively correlated with the extent of MDA accumulation.  相似文献

Exogenous sialic acid at 3 mM and higher concentrations inhibits the basal adenylate cyclase activity and the activity stimulated by thyrotropin (TSH) and fluoride in the human thyroid membrane fraction; 30 mM-sialic acid acts as an inhibitor of TSH binding. The decrease of these activities at high sialic acid concentrations might be ascribed to changes in membrane conformation caused by acidic character of this sugar.  相似文献

The activity of the adenylate cyclase catalytic subunit is higher in Harvey and Kirsten Murine Sarcoma Viruses-infected thyroid epithelial cells than in uninfected control cells either in the presence of Mg2+ alone or following stimulation by Mn2+ or forskolin. The higher activity is associated with an increased cAMP cellular content. The Gpp(NH)p and F- anion are more effective positive modulators in the control than in the virus infected cells: these results exclude therefore that the ras p21 proteins can act as the G-protein alpha-subunit and suggest that they negatively interfere with the G-protein modulation of the adenylate cyclase system.  相似文献

Effect of prostacyclin (PGI2) on adenylate cyclase activity in human thyroid membranes was examined. PGI2 caused a dose- and time-dependent production of cyclic AMP (cAMP) with high potency. When GTP was added in concentrations up to 100 uM, the activation of adenylate cyclase by PGI2 was increased. In the assay medium containing 3 mM ATP, 10 uM GTP and nucleotide regenerating system, the replacement of Mg2+ by increasing concentrations of Mn2+ caused a progressive loss of PGI2 as well as TSH-stimulated adenylate cyclase activities, while high concentrations of Mg2+ (12 or 18 mM) slightly suppressed the activity stimulated by either PGI2 or TSH. Both agents had an additive effect on the stimulation of adenylate cyclase activity in the presence of either 6 mM Mg2+ or 6 mM Mn2+. Gamma-globulin fraction containing non-stimulatory TSH receptor antibody which was prepared from a patient with chronic thyroiditis, suppressed only TSH- but not PGI2-stimulation of the adenylate cyclase activity. These results suggest that PGI2 can stimulate the adenylate cyclase activity in human thyroid tissue, and that PGI2-stimulation may be mediated by the different system from TSH-dependent one.  相似文献

The effects of three different ammonium ion concentrations (0.143; 1.43; 14.3 mmol .1(-1)) on adenylate cyclase activities in liver, muscle, fat and brain tissue in vitro were studied in the rat. At physiological levels, ammonium ions reduced adenylate cyclase activity in liver and fat by about 30%, did not affect enzyme activity in muscle tissue but increased adenylate cyclase activity in brain by 40%. Similar effects were observed at higher ammonium ion concentrations though there were no statistically significant correlations between ammonium ion concentration and the degree of change of enzyme activity.  相似文献

Thyroid hormones effectively inhibited partially purified cyclic AMP phosphodiesterase from the anterior and posterior lobes of bovine pituitary gland competitively with the substrate, but thyronine, diiodotyrosine, monoiodotyrosine, thyrotropin, thyrotropin releasing hormone, dexamethasone, and luteinizing hormone releasing hormone did not inhibit this enzyme activity.K i values were found to be 1.0 and 2.5 M for thyroxine, and 8.0 and 13.5 M for triiodothyronine in the anterior and posterior lobes, respectively.  相似文献

The effect of the modification of synaptosomal membrane glycoproteins on the activity of adenylate cyclase was studied. It was found that the binding of concanavalin A to unmodified guinea pig cerebral cortex synaptosomal membrane did not change adenylate cyclase activity. Concanavalin A binding to synaptosomal membrane of hypoxic brain cortex resulted in no decrease of enzyme activity. The level of protein-bound sialic acid in these synaptosomal fractions was 20% lower than in the control. Treatment of synaptosomal membranes with neuraminidase resulted in a decrease of sialic acid content by about 70%, but it had no significant effect on adenylate cyclase activity. The modification with concanvalin A of sugar end groups exposed by neuraminidase treatment resulted in significant decrease of both basal and fluoride-stimulated adenylate cyclase activity. These results seem to indicate that some component of the adenylate cyclase complex of brain synaptosomal membranes is closely interacting with a carbohydrate-containing macromolecule on the cell surface.This work was supported by, the Polish Academy of Sciences within the project 10.4.  相似文献

The characteristics of the iodide-induced inhibition of cyclic AMP accumulation in dog thyroid slices have been previously described [Van Sande, J., Cochaux, P. and Dumont, J. E. (1985) Mol. Cell. Endocrinol. 40, 181-192]. In the present study we investigated the characteristics of the iodide-induced inhibition of adenylate cyclase activity in dog and horse thyroid. The inhibition of cyclic AMP accumulation by iodide in stimulated horse thyroid slices was similar to that observed in dog thyroid slices. The inhibition was observed in slices stimulated by thyroid-stimulating hormone, cholera toxin and forskolin. Increasing the concentration of the stimulators did not overcome the iodide-induced inhibition. Adenylate cyclase activity, assayed in crude homogenates or in plasma-membrane-containing particulates (100,000 x g pellets), was lower in homogenates or in particulates prepared from iodide-treated slices than from control slices. This inhibition was observed on the cyclase activity stimulated by forskolin, fluoride or guanosine 5'-[beta, gamma-imino]triphosphate, but also on the basal activity. It was relieved when the homogenate was prepared from slices incubated with iodide and methimazole. Similar results were obtained with dog thyroid. The inhibition persisted when the particulate fraction was washed three times during 1 h at 100,000 x g, in the presence of bovine serum albumin or increasing concentration of KCl. It was similar whatever the duration of the cyclase assay, in a large range of protein concentration. These results indicate that a stable modification of adenylate cyclase activity, closely related to the plasma membrane, was induced when slices were incubated with iodide. Iodide inhibition did not modify the affinity of adenylate cyclase for its substrate (MgATP), but induced a decrease of the maximal velocity of the enzyme. The percentage inhibition was slightly decreased when Mg2+ concentration increased, and markedly decreased when Mn2+ concentration increased. A detectable adenylate cyclase activity was demonstrated when intact slices were incubated in the presence of [alpha-32P]ATP, probably because of the presence of broken cells produced during the slicing. Iodide had no direct effect on this cyclase system, which confirms that iodide needs the integrity of the cell to induce the inhibition and suggests that the inhibition is not transmitted between cells.  相似文献

The effect of divalent cations on bovine sperm adenylate cyclase activity was studied. Mn2+, Co2+, Cd2+, Zn2+, Mg2+ and Ca2+ were found to satisfy the divalent cation requirement for catalysis of the bovine sperm adenylate cyclase. These divalent cations in excess of the amount necessary for the formation of the metal-ATP substrate complex were found to stimulate the enzyme activity to various degrees. The magnitude of stimulation at saturating concentrations of the divalent cations was strikingly greater with M2+ than with either Ca2+, Mg2+, Zn2+, Cd2+ or Co2+. The apparent Km was lowest for Zm2+ (0.1 - 0.2 mM) than for any of the other divalent cations tested (1.2 - 2.3 mM). The enzyme stimulation by Mn2+ was decreased by the simultaneous addition of Co2+, Cd2+, Ni2+ and particularly Zn2+ and Cu2+. The antagonism between Mn2+ and Cu2+ or Zn2+ appeared to have both competitive and non-competitive features. The inhibitory effect of Cu2+ on Mn2+-stimulated adenylate cyclase activity was prevented by 2,3-dimercaptopropanol, but not by dithiothreitol, L-ergothioneine, EDTA, EGTA or D-penicillamine. Ca2+ at concentrations of 1-5 mM was found to act synergistically with Mg2+, Zn2+, Co2+ and Mn2+ in stimulating sperm adenylate cyclase activity. The Ca2+ augmentation of the stimulatory effect of Zn2+, Co2+, Mg2+ and Mn2+ appeared to be specific.  相似文献

Melittin, a surface-active, 26-amino acid polypeptide from bee venom, has been reported to alter a variety of membrane properties including stability, permeability, and fluidity, the latter having been shown to be altered in a biphasic manner. Melittin induced a biphasic alteration of rat heart microsomal adenylate cyclase activity, stimulating it at low concentration (<30 μg/ml) and inhibiting it at higher concentrations (100 μg/ml or higher). Melittin potentiated sodium fluoride and 5′-guanylylimidodiphosphate activation of adenylate cyclase below 40 μg/ml but it inhibited at high concentrations, except in the presence of high concentrations of 5′-guanylylimidodiphosphate (10?4m). Basal and fluoride-activated adenylate cyclase exhibited no significant change in the Km for ATP in the presence of melittin at <40 μg/ml, but the V was elevated. Potentiation by melittin of adenylate cyclase was observed at all fluoride, 5′-guanylylimidodiphosphate, and magnesium concentrations tested. The observed effects of melittin on rat heart adenylate cyclase are consistent with it acting by altering the properties of membrane lipids with which the enzyme is associated.  相似文献

1-adrenaline, ACTH and glucagon activate the adenylate cyclase of rat adipocytes by decreasing its S0.5(Mg2+) (concentration yielding 0.5 Vmax) from its basal value of 11.5 to 1.2, 0.3 and 1.8 mM and by increasing its Ki(ATP4?) from 0.03 to 0.25; 0.62 and 0.16 mM respectively. The kinetic properties of the enzyme are regulated by its state of saturation with ATP4? or Mg2+; its saturation with ATP4? and citrate3? suppressed its basal and hormone-dependent activities. The hormone-dependent decrease in Km and increase in Vmax of the enzyme occur when shifting from suboptimal low concentrations of hormone and Mg2+ to optimal conditions, i.e., high concentration of hormone and low concentration of Mg2+. The increase in the state of saturation of the enzyme with Mg2+ decreases the hormone-dependent effects on Vmax and results in identical values of Km (0.14 mM) for its basal and 1-adrenaline dependent activities. CaCl2 saturation curves at 5 mM ATP with either 5, 10 or 20 mM MgCl2 show that the substitution of 5 mM MgCl2 by 10 mM and 20 mM MgCl2 increased the Ki(Ca2+) of the enzyme from 0.19 to 0.49 and 0.94 mM but decreased its Ki(CaATP) from 0.42 to 0.19 and 0.14 mM respectively. Only when the concentration of MgCl2 exceeded that of ATP did 1-adrenaline and ACTH activate the enzyme by increasing its Ki(Ca2+), although only ACTH increased its Ki(CaATP). An increase in energy charge would decrease the intracellular concentrations of Mg2+ and Ca2+ because ATP4? has stronger binding constants for Mg2+ and Ca2+ than ADP3? and AMP2?. Hence, the reported properties of the enzyme suggests that changes in energy charge may allow for metabolic feedback control of the hormonal responsiveness of the Mg2+, Ca2+, ATP4? -sensitive adenylate cyclase.  相似文献

Adenylate cyclase (EC 4.6.1.1) was studied in membrane preparations of reticulocyte-rich blood obtained from phenylhydrazine-treated rabbits and compared to that of untreated animals.Basal and fluoride-stimulated activities were decreased 2- and 4-fold, respectively, during the process of maturation.Catalytic parameters such as time course, protein, ATP, Mg2+ concentration curves and Km have been determined and were found to be similar in the reticulocyte and the erythrocyte.Adenylate cyclase was sensitive to GTP, 5′-guanylyl imidodiphosphate, prostaglandin E1 and prostaglandin E2. Activation by prostaglandin E1 was higher than that produced by prostaglandin E2. Only additive effect was found when 5′-guanylyl imidodiphosphate or GTP was added to hormone-stimulated activity. The sensitivity of the enzyme to these effectors was decreased over the transition reticulocyte-erythrocyte.In either cell the enzyme was not activated by catecholamines (epinephrine, norepinephrine, isoproterenol).  相似文献

TSH (thyrotropin)-stimulated human thyroid adenylate cyclase has a biphasic response to Ca2+, being activated by submicromolar Ca2+ (optimum 22nM), with inhibition at higher concentrations. Calmodulin antagonists caused an inhibition of TSH-stimulated adenylate cyclase in a dose-dependent manner. Inhibition of TSH-and TSIg-(thyroid-stimulating immunoglobulins)-stimulated activity was more marked than that of basal, NaF- or forskolin-stimulated activity. This inhibition was not due to a decreased binding of TSH to its receptor. Addition of pure calmodulin to particulate preparations of human non-toxic goitre which had not been calmodulin-depleted had no effect on adenylate cyclase activity. EGTA was ineffective in removing calmodulin from particulate preparations, but treatment with the tervalent metal ion La3+ resulted in a loss of up to 98% of calmodulin activity from these preparations. Addition of La3+ directly to the adenylate cyclase assay resulted in a partial inhibition of TSH- and NaF-stimulated activity, with 50% inhibition produced by 5.1 microM and 4.0 microM-La3+ respectively. Particulate preparations with La3+ showed a decrease of TSH- and NaF-stimulated adenylate cyclase activity (approx. 40-60%). In La3+-treated preparations there was a decrease in sensitivity of TSH-stimulated adenylate cyclase to Ca2+ over a wide range of Ca2+ concentrations, but most markedly in the region of the optimal stimulatory Ca2+ concentration. In particulate preparations from which endogenous calmodulin had been removed by La3+ treatment, the addition of pure calmodulin caused an increase (73 +/- 22%; mean +/- S.E.M., n = 8) in TSH-stimulated thyroid adenylate cyclase activity.
