Cryptomonas erosa Skuja, a planktonic alga, was grown in batch culture at different combinations of light intensity and temperature, under nutrient saturation. Growth was maximal (1.2 divisions · day?1) at 23.5 C and 0.043 ly · min?1, declining sharply with temperature (0.025 divisions-day?1 at 1 C). With decreasing temperature, the cells showed both light saturation and inhibition at much reduced light intensities. At the same time the compensation light intensity for growth declined towards a minimum of slightly above 0.4 × 10?4 ly · min?1 (~1 ft-c) at 1 C or <0.1 ly · day?1 (PAR). Cell division was more adversely affected by low temperature than carbon uptake, and the resulting excess production of photosynthate was both stored and excreted. Extreme storage of carbohydrates resulted in cell volumes and carbon content ca. 22 and 30 × greater, respectively, than the maxima observed for cells incubated in the dark, whereas, at growth inhibitory light levels, as much as 57% of the total assimilated carbon was excreted. A marked increase in cell pigment was observed at the lowest light levels (<10?3 ly · min?1), at high temperature. The growth response of C. erosa in culture provides insight into the abundance and distribution of cryptomonads and other small algal flagellates in nature.  相似文献

The relative importance of respiration and organic carbon release to the efficiency of carbon specific growth of Skeletonema costatum (Grev.) Clave was evaluated over a light range from 1500–15 μE · m?2· s?1. Net growth efficiency ranged from 0.45–0.69 with a maximum at 130 μE · m?2· s?1. Respiration was 93% or more of the variations in growth efficiency. Organic carbon release ranged from 0–7% of gross production and increased with light intensity. Carbon specific particulate production was a hyperbolic function of incident light intensity and was related exponentially to particulate carbon production per unit chlorophyll a. Full sunlight conditions, 1500 μE · m?2· s?1, did not induce photoinhibition of gross production. Variations in the efficiency of growth of S. costatum were minimized over a wide range of light intensities mainly because of variations in cellular pigments which permitted the efficient utilization of available light energy, and a reduction in the losses of carbon which increases the growth rate, possibly as a consequence of the recycling of respired carbon within the cell.  相似文献

The effects of pulsed ammonium additions on the ammonium-limited marine diatoms, Chaetoceros gracile Schutt and Skeletonema costatum (Grev.) Cleve were studied. Two culture systems were maintained for each species. One culture was grown in a chemostat which provided a homogeneous distribution of the limiting nutrient. In the other continuous culture, ammonium was added once daily giving rise to a patchy distribution. Ammonium patchiness increased both the V'max (from 4.1 ± 0.3 to 11.8 ± 0.9 · h?1) and the Vi (from 0.19 ± 0.02 to 0.36 ± 0 0.04 · h?1) for ammonium in S. costatum and the V'max (from 0.91 ± 0.07 to 2.9 ± 0 0.2 · h?1) and the Vi (from 0.16 ± 0.01 to 0.31 ± 0.05 · h?1) for ammonium in Chaetoceros gracile. The once-per-day addition of ammonium also induced a diel periodicity in photosynthetic rate and fluorescence although these cultures were growing under continuous light. The relative amplitude of the periodicity was greater for Skeletonema than for Chaetoceros. These observations are considered with regard to the hypothesis that limiting nutrient patchiness may alter the growth kinetics of marine phytoplankton.  相似文献

Production of domoic acid (DA), a neurotoxin, by the diatom Pseudo-nitzschia multiseries (previously Nitzschia pungens f. multiseries) Hasle and its cellular chemical composition were studied in phosphate-limited chemostat continuous cultures and in subsequent batch cultures. Under steady-state chemostat conditions, DA production increased from 0.01 to 0.26 pg DA · cell?1· d?1 as the growth rate decreased. When the nutrient supply was discontinued (to produce a batch culture), DA production was enhanced by a factor of ca. 3. DA production was temporarily suspended upon addition of phosphate to the batch cultures but resumed 1 d later at a higher rate coincident with the decline of phosphate uptake. In both steady-state continuous culture and batch culture, more DA was produced when alkaline phosphatase activity (APA) was high. The association of high DA production with high levels of APA and high cellular N:P ratios strongly suggests that phosphate limitation enhances DA production. Also, DA production was high when other primary metabolism (e.g. uptake of carbon, nitrogen, phosphorus and silicon, and cell division) was low, but chlorophyll a and adenosine triphosphate were generally high. This suggests that the synthesis of DA requires a substantial amount of biogenic energy.  相似文献

Clones of Skeletonema costatum (Grev.) Cl. isolated from Narragansett Bay, R.I., during different seasons were grouped according to their electrophoretic banding patterns. The growth rates, pg chlorophyll · cell?1, carbon uptake · cell?1· h?1, and carbon uptake · pg chl?1· h?1 were measured at 20°C, in a 14:10 h L:D cycle at 180 μE · m?2· s?1. Statistically significant sources of variation were found among groups of clones in growth rate, pg chl · cell?1, and carbon uptake · pg chl?1· h?1. It was concluded that there is a significant relationship between the physiological characteristics of clones isolated from populations in different seasons and patterns of genetic variation inferred from the electrophoretic studies. However, genetic diversity detected by banding patterns tends to underestimate the total genetic diversity in natural populations. The groups of clones most common in summer bloom populations had significantly higher growth rates, lower values of pg chl · cell?1, and higher rates of carbon uptake · pg chl?1· h?1 at 20°C than did the group of clones most common in winter bloom populations. However, differences among groups in these parameters at 20°C alone cannot account for the seasonal cycling of genetically variable populations of Skeletonema in Narragansett Bay. The range of growth rates among clones of this species is 0.1–5.0 divisions · d?1 under a single set of temperature and light conditions. Chlorophyll concentrations range from 0.2–1.7 pg chl · cell?1 and carbon uptake · pg chl?1· h?1 varies by a factor of 7 among clones. The range of physiological variation in this species means that it is difficult to use laboratory studies of single clones to analyze the responses of natural populations of Skeletonema.  相似文献

The underwater light field in blackwater environments is strongly skewed toward the red end of the electromagnetic spectrum due to blue light absorption by colored dissolved organic matter (CDOM). Exposure of phytoplankton to full spectrum irradiance occurs only when cells are mixed up to the surface. We studied the potential effects of mixing‐induced changes in spectral irradiance on photoacclimation, primary productivity and growth in cultures of the cryptophyte Rhodomonas salina and the diatom Skeletonema costatum. We found that these taxa have very different photoacclimation strategies. While S. costatum showed classical complementary chromatic adaption, R. salina showed inverse chromatic adaptation, a strategy previously unknown in the cryptophytes. Transfer of R. salina to periodic full spectrum light (PFSL) significantly enhanced growth rate (μ) by 1.8 times and primary productivity from 0.88 to 1.35 mg C · (mg Chl?1) · h?1. Overall, R. salina was less dependent on PFSL than was S. costatum, showing higher μ and net primary productivity rates. In the high‐CDOM simulation, carbon metabolism of the diatom was impaired, leading to suppression of growth rate, short‐term 14C uptake and net primary production. Upon transfer to PFSL, μ of the diatom increased by up to 3‐fold and carbon fixation from 2.4 to 6.0 mg C · (mg Chl?1) · h?1. Thus, a lack of PFSL differentially impairs primarily CO2‐fixation and/or carbon metabolism, which, in turn, may determine which phytoplankton dominate the community in blackwater habitats and may therefore influence the structure and function of these ecosystems.  相似文献

Uptake and assimilation of nitrogen and phosphorus were studied in Olisthodiscus luteus Carter. A diel periodicity in nitrate reductase activity was observed in log and stationary phase cultures; there was a 10-fold difference in magnitude between maximum and minimum rates, but other cellular features such as chlorophyll a, carbon, nitrogen, C:N ratio (atoms) · cell?1 were less variable. Ks values (~2 μM) for uptake of nitrate-N and ammonium-N were observed. Phosphorus assimilated · cell?1· day?1 varied with declining external phosphorus concentrations; growth rates <0.5 divisions · day?1 were common at <0.5 μM PO4-P. Phosphate uptake rates (Ks= 1.0–1.98 μM) varied with culture age and showed multiphasic kinetic features. Alkaline phosphatase activity was not detected. Comparisons of the nutrient dynamics of O. luteus to other phytoplankton species and the ecological implications as related to the phytoplankton community of Narragansett Bay (Rhode Island) are discussed.  相似文献

Aims: Kluyveromyces lactis was cultured in cheese whey permeate on both batch and continuous mode to investigate the effect of time course and growth rate on β‐galactosidase activity, lactose consumption, ethanol production and protein profiles of the cells. Methods and Results: Cheese whey was the substrate to grow K. lactis as a batch or continuous culture. In order to precise the specific growth rate for maximum β‐galactosidase activity a continuous culture was performed at five dilution (growth) rates ranging from 0·06, 0·09, 0·12, 0·18 to 0·24 h?1. The kinetics of lactose consumption and ethanol production were also evaluated. On both batch and continuous culture a respirofermentative metabolism was detected. The growth stage for maximum β‐gal activity was found to be at the transition between late exponential and entrance of stationary growth phase of batch cultures. Fractionating that transition stage in several growth rates at continuous culture a maximum β‐galactosidase activity at 0·24 h?1 was observed. Following that stage β‐gal activity undergoes a decline which does not correlate to the density of its corresponding protein band on the gel prepared from the same samples. Conclusion: The maximum β‐galactosidase activity per unit of cell mass was found to be 341·18 mmol ONP min?1 g?1 at a dilution rate of 0·24 h?1. Significance and Impact of the Study: The physiology of K. lactis growing in cheese whey permeate can proven useful to optimize the conversion of that substrate in biomass rich in β‐gal or in ethanol fuel. In addition to increasing the native enzyme the conditions established here can be set to increase yields of recombinant protein production based on the LAC4 promoter in K. lactis host.  相似文献

Two axenic, in vitro liquid suspension cultures were established for Agardhiella subulata (C. Agardh) Kraft et Wynne, and their growth characteristics were compared. This study illustrated how reliable routes for the development of suspension cultures of macrophytic red algae of terete thallus morphology can be achieved for biotechnology applications. Undifferentiated filament clumps of 2–8 mm diameter were established by induction of callus-like tissue from thallus explants, and lightly branched microplantlets of 2–10 mm length were established by regeneration of filament clumps. The filament clumps were susceptible to regeneration. Adventitious shoot formation was reliably induced from 40% to 70% of the filament clumps by gentle mixing at 100 rev min?1 on an orbital shaker. The specific growth rate of the microplantlets was higher than the filament clumps in nonagitated well plate culture (4%–6% per day for microplantlets vs. 2%–3% per day for filament clumps) at 24° C and 8–36 μmol photons·m?2·s?1 irradiance (10:14 h LD cycle) when grown on ASP12 artificial seawater medium at pH 8.6–8.9 with 20%–25% per day medium replacement. Oxygen evolution rate vs. irradiance measurements showed that relative to the filament clumps, microplantlets had a higher maximum specific oxygen evolution rate (Po,max= 0.181 ± 0.035 vs. 0.130 ± 0.023 mmol O2·g?1 dry cell mass·h?1), but comparable respiration rate (Qo= 0.040 ± 0.013 vs. 0.033 ± 0.017 mmol O2·g?1 dry cell mass·h?1), compensation point (Ic= 3.8 ± 2.4 vs. 5.7 ± 1.2 μmol photons·m?2·s?1), and light intensity at 63.2% of saturation (Ik= 17.5 ± 3.9 vs. 14.9 ± 2.6 μmol photons·m?2·s?1). The microplantlet culture was more suitable for suspension culture development than the filament clump culture because it was morphologically stable and exhibited higher growth rates.  相似文献

Several studies have suggested that phytoplankton play a role in the iodine cycle. Using a short-term incubation technique for determining the uptake of iodate by phytoplankton, cultures of Thalassiosira oceanica Hasle, Skeletonema costatum (Greville) Cleve, Emiliania huxleyi (Lohmann) Hay and Mohler, and Dunaliella tertiolecta Butcher were found to be capable of assimilating iodate at rates ranging from 0.003 to 0.24 nmol IO3?·μg chlorophyll a?1·h?1. The kinetics for the uptake of iodate can be modeled, and the similarity between the model and experimental results suggests that there is a steady state between iodate uptake and release of dissolved iodine from the cells, presumably in the form of iodide. Two experiments were conducted in the Sand Shoal Inlet of the Cobb Bay estuary (37°15′N, 75°50′W). The uptake of iodate was 0.26 and 0.08 nmol IO3?·μg chlorophyll a?1·h?1 during high and low tide, respectively. Using field estimates based on measured levels of iodate in the estuary, we estimate that phytoplankton can take up as much as 3% of the ambient pool of iodate on a daily basis and the entire pool in about 1 month. Thus, phytoplankton can be a significant component of the global iodine cycle by mediating changes in the speciation of iodine in the marine environment.  相似文献

The uptake kinetics of ammonium and phosphate by Gracilaria tikvahiae McLachlan were studied under field conditions. Seaweeds, pulse fed once a week for 6 h over a 4-week period, had maximum uptake rates of 19 μmol·g fwt?1·h?1 for ammonium and 0.28 μmol·g fwt?1·h?1 for phosphate. For both nutrients there was a positive linear correlation between uptake rate (v) and concentration (S) over the entire range of concentration tested. In a nutrient depletion experiment, the phosphate uptake curve determined over a wide range of concentrations consisted of two stages of saturation at low concentrations, and a linear phase at high concentrations. Ash free dry weight, chlorophyll a, phycoerythrin, and protein content were higher in pulse fed plants than in control plants receiving no nutrient additions, while the reverse held true for carbohydrate contents and the C/N ratios. The C/N ratio inversely correlated with ammonium and phosphate uptake rate as well as protein and phycoerythrin content, and positively with carbohydrate content.  相似文献

The effect of external glucose (51 mM) and acetate (13 mM) on growth and photosynthetic capacity of Ulva lactuca L. was tested in laboratory cultures over 41 days in the dark and in dim light (0.9 μmol photons·m?2·s?1) at 7–8° C. Glucose and acetate had a significant positive effect on growth rate, chlorophyll content, and quantum yield for discs grown in the dark and in dim light. The carbon gain from heterotrophic uptake was low and only allowed U. lactuca to maintain a specific uptake was low and only allowed U. lactuca to maintain a specific growth rate of 0.005 day?1 compared to 0.06–0.1 day?1 at higher light intensities. However, plants with added organic substrate maintained a normal chlorophyll content and were able to photosynthesize whereas control plants lost pigmentation and photosynthetic capability after 41 days in both dim light and darkness, probably because of disorganization of the photosynthetic apparatus. This suggest that the ecological significance of heterotrophic uptake is to allow U. lactuca to survive during prolonged low light conditions with an intact photosynthetic apparatus.  相似文献

Growth, blade shape and blade thickness of young gametophytes of Porphyra abbottae Krishnamurthy cultured from conchospores were determined at various combinations of temperature (8, 10, 12° C), photon flux density (17.5, 70, 140 μmol·m-?2·S?1), nutrient concentration (5, 25, 50, 100% f medium) and water motion (0, 50, 100, 150 rpm). Growth (as surface area) was light-saturated at 70 μmol· m?2· S?1, light-inhabited at 140 μmol·m?2· S?1, and nutrient-saturated an 25% f medium. Temperature had no significant effect on growth. Water motion and nutrients had an interactive effect on growth, with water motion having the greatest effect at the lowest nutrient concentrations. Water motion enhanced growth even at saturating nutrient concentrations. Blade length / width ratio was greater in low light (2.5) than in saturating light (1.9); with increasing water motion the ratio increased from 1.2 to 2.4. Blade thickness (53-88 μm) was greatest at the highest nutrient concentrations and at the lowest water motion levels. Temperature and light did not have a consistent effect on blade thickness.  相似文献

Continuous cultures of Merismopedia tenuissima Lemmerman, limited by phosphorus, nitrogen, sulfur, or carbon, were compared to non limited batch cultures by two methods. The cellular content of photosynthetic pigments (chlorophyll and phycocyanin) was found to decrease in all nutrient limited cultures, except for the carbon limited culture. The ratio of carbohydrate to protein was 4- to 7-fold higher in P, N or S limited cultures than in non-limited or C limited cultures. The macromolecular products of photosynthesis were determined in samples to which NaH14CO3 was added. Relative incorporation into protein decreased in P or N limited cultures, increased accumulation of low molecular weight compounds was found in S and P limited cultures, and little change was noted in C limited cultures as compared to non-limited cultures. Although relative incorporation into protein was significantly greater at 20μEin·m?2·s?1 light intensity than at 180 μEin·m?2.s?1 in non-limited cultures, this effect was abolished in all nutrient limited cultures. These results suggest that measurement of the cellular carbohydrate to protein ratio and the products of photosynthesis would be useful in the analysis of algal population dynamics in nature.  相似文献

Emiliania huxleyi (strain L) expressed an exceptional P assimilation capability. Under P limitation, the minimum cell P content was 2.6 fmol P·cell?1, and cell N remained constant at all growth rates at 100 fmol N·cell?1. Both, calcification of cells and the induction of the phosphate uptake system were inversely correlated with growth rate. The highest (cellular P based) maximum phosphate uptake rate (VmaxP) was 1400 times (i.e. 8.9 h?1) higher than the actual uptake rate. The affinity of the P‐uptake system (dV/dS) was 19.8 L·μmol?1·h?1 at μ = 0.14 d?1. This is the highest value ever reported for a phytoplankton species. Vmax and dV/dS for phosphate uptake were 48% and 15% lower in the dark than in the light at the lowest growth rates. The half‐saturation constant for growth was 1.1 nM. The coefficient for luxury phosphate uptake (Qmaxt/Qmin) was 31. Under P limitation, E. huxleyi expressed two different types of alkaline phosphatase (APase) enzyme kinetics. One type was synthesized constitutively and possessed a Vmax and half‐saturation constant of 43 fmol MFP·cell?1·h?1 and 1.9 μM, respectively. The other, inducible type of APase expressed its highest activity at the lowest growth rates, with a Vmax and half‐saturation constant of 190 fmol MFP·cell?1·h?1 and 12.2 μM, respectively. Both APase systems were located in a lipid membrane close to the cell wall. Under N‐limiting growth conditions, the minimum N quotum was 43 fmol N·cell?1. The highest value for the cell N‐specific maximum nitrate uptake rate (VmaxN) was 0.075 h?1; for the affinity of nitrate uptake, 0.37 L·μmol?1·h?1. The uptake rate of nitrate in the dark was 70% lower than in the light. N‐limited cells were smaller than P‐limited cells and contained 50% less organic and inorganic carbon. In comparison with other algae, E. huxleyi is a poor competitor for nitrate under N limitation. As a consequence of its high affinity for inorganic phosphate, and the presence of two different types of APase in terms of kinetics, E. huxleyi is expected to perform well in P‐controlled ecosystems.  相似文献

The photoprotective response in the dinoflagellate Glenodinium foliaceum F. Stein exposed to ultraviolet‐A (UVA) radiation (320–400 nm; 1.7 W · m2) and the effect of nitrate and phosphate availability on that response have been studied. Parameters measured over a 14 d growth period in control (PAR) and experimental (PAR + UVA) cultures included cellular mycosporine‐like amino acids (MAAs), chls, carotenoids, and culture growth rates. Although there were no significant effects of UVA on growth rate, there was significant induction of MAA compounds (28 ± 2 pg · cell?1) and a reduction in chl a (9.6 ± 0.1 pg · cell?1) and fucoxanthin (4.4 ± 0.1 pg · cell?1) compared to the control cultures (3 ± 1 pg · cell?1, 13.3 ± 3.2 pg · cell?1, and 7.4 ± 0.3 pg · cell?1, respectively). In a second investigation, MAA concentrations in UVA‐exposed cultures were lower when nitrate was limited (P < 0.05) but were higher when phosphate was limiting. Nitrate limitation led to significant decreases (P < 0.05) in cellular concentration of chls (chl c1, chl c2, and chl a), but other pigments were not affected. Phosphate availability had no effect on final pigment concentrations. Results suggest that nutrient availability significantly affects cellular accumulation of photoprotective compounds in G. foliaceum exposed to UVA.  相似文献

The effects of environmental variables, particularly irradiance, on the sinking rates of phytoplankton were investigated using cultures of Chaetoceros gracilis Schütt and C. flexuosum Mangin in laboratory experiments; these data were compared with results from assemblages in the open ocean and marginal ice zone of the Greenland Sea. In culture experiments both the irradiance under which the diatom was grown and culture growth rate were positively correlated with sinking rates. Sinking rates (ψ) in the Greenland Sea were smallest when determined from chlorophyll (mean ψchl= 0.14 m · d?1) and biogenic silica (ψsi= 0.14 m · d?1) and greatest when determined from particulate carbon (ψc= 0.55 m · d?1) and nitrogen (ψN= 0.64 m · d?1). Field measurements indicated that variations in sinking may be associated with changes in irradiance and nitrate concentrations. Because these factors do not directly affect water density, they must be inducing physiological changes in the cell which affect buoyancy. Although a direct response to a single environmental variable was not always evident, sinking rates were positively correlated with growth rates in the marginal ice zone, further indicating a connection to physiological processes. Estimats of carbon flux at stations with vertically mixed euphotic zones indicated that approximately 30% of the daily primary production sank from the euphotic zone in the form of small particulates. Calculated carbon flux tended to increase with primary productivity.  相似文献

Microalgae are a promising feedstock for biofuel production. Lipid content in microalgae could be enhanced under nutrient depletion. This work investigated the effect of the nutrient on lipid accumulation in Ankistrodesmus sp. culture. Batch cultures were carried out using fresh BG11 medium, and after the harvest, the medium was reused for the next culture; this method was repeated two times. The maximum lipid productivity of 29.75 mg L?1 day?1 was obtained from the culture with the second reuse medium. In continuous cultures, Ankistrodesmus sp. was cultured in both fresh and modified BG11 mediums. The modified BG11 medium was adjusted to resemble the content of the first reuse medium. As a comparison between batch and continuous cultures, it was proven that the productivity in the continuous culture was better than in the batch, where the achievable maximum biomass and lipid were 188.30 and 38.32 mg L?1 day?1. The maximum lipid content of 34.22% was obtained from the continuous culture at a dilution rate of 0.08 day?1, whereas the maximum saturated and unsaturated fatty acid productivities of 79.96 and 104.54 mg L?1 day?1 were obtained at a dilution rate of 0.16 day?1.  相似文献

Cellular nutrient concentrations and nutrient uptake rates of Cladophora glomerata (L.) Kuetzing were determined during summer and fall in 1989–1990 at a site on the upper Clark Fork of the Columbia River, Montana. Both physiological tests indicated that Cladophora growth is likely to be limited by nitrogen during late summer-early fall. Maximum uptake rates of ammonia-N and nitrate-N were 5935–6991 and 507–984 μg · g DW?1· h?1, respectively, during July–October when dissolved inorganic nitrogen (DIN) concentrations in the river were less than 10 μg · L?1. During November-December, when DIN was 72–376 μg · L?1, maximum ammonia-N uptake was 1137–1633 μg · g DW?1· h?1 and maximum nitrate-N uptake was 0–196 μg · g DW?1· h?1. Cellular nitrogen during summer–early fall was 0.78–1.80% of Cladophora dry weight, frequently at or below 1.1%, a level suggested as a critical minimum N concentration for maximum growth. In contrast, cellular P was 0.18–0.36% of dry weight, 3–6 times the suggested critical P concentration of 0.06%. Molar ratios of cellular N:P (< 16:1) and DIN: SRP (< 4:1) during late summer-early fall also indicated potential N limitation. Cellular N and P from Cladophora collected from a second site influenced by a municipal wastewater discharge in 1990 displayed similar seasonal trends. At both sites, seasonal fluctuations in DIN were closely tracked by changes in cellular N, Cellular P, however, increased through the growing season despite declining levels of SRP in the river.  相似文献
