Susceptibility to acephate, methomyl, and permethrin was determined with laboratory bioassays of field-collected adults from 15 populations of the B biotype of sweetpotato whitefly,Bemisia tabaci (Gennadius) (Homoptera:Aleyrodidae), from Hawaii. Comparisons at the LC50 showed up to 24-fold resistance to acephate, 18-fold resistance to methomyl, and 4-fold resistance to permethrin. Analysis of variance showed significant intra-island variation in susceptibility to each insecticide, but no significant variation among islands. Insecticide use varied from 4 to 103 insecticide sprays per site per season. Acephate and methomyl were used more often than permethrin. The frequency of application and LC50 for each insecticide were positively correlated across sites. These results suggest that local variation in insecticide use was a primary cause of variation in susceptibility. If local insecticide use is a key determinant of resistance, as our results suggest, growers can retard resistance development locally by reducing their own insecticide use.  相似文献

The purpose of this study was to evaluate the cytochrome P450-dependent monooxygenase activities in methidathion resistant and susceptible strains of Amblyseius womersleyi Schicha. Artificial laboratory selections for resistance and susceptibility to methidathion were performed in an organophosphate resistant strain of A. womersleyi (Kanaya strain). Selections for susceptibility were also performed in a susceptible strain of this predaceous mite (Ishigaki Strain). After the selection process, the LC50 of methidathion for the selected strains of A. womersleyi were 816 mg/l (Kanaya R), 4.61 mg/l (Kanaya S) and 1.59 mg/l (Ishigaki S). The monooxygenase activities were determined biochemically by the O-deethylation of 7-ethoxycoumarin (7-EC). The monooxygenase activity in adult females of Kanaya R strain (51.1 pmol/30 min/mg protein) was 3.60- and 5.42-fold higher than the activity observed for Kanaya S and Ishigaki S strains, respectively. Significant correlation between monooxygenase activity and LC50 (mg/l) of methidathion was observed analyzing 16 populations of A. womersleyi with different susceptibilities to methidathion. Monooxygenase activity was also evaluated in different life stages (egg, larva, protonymph, deutonymph and adult) of A. womersleyi. The lowest activity was observed for the larval stage, which presented the highest susceptibility to methidathion. Protonymph, deutonymph and adult presented the highest monooxygenase activities. These stages were the most tolerant to methidathion. Monooxygenase activities of the Kanaya R strain were higher than of the Kanaya S strain in all developmental stages. The present study can be helpful for the implementation of a program involving release of insecticide-resistant populations of A. womersleyi in the field. The monooxygenase activity determination is easier and quicker than the estimation of LC50, requiring fewer mites.  相似文献

The toxicity of dimethoate 30EC and methidathion 40EC was tested against four strains ofPhytoseiulus persimilis obtained from commercial greenhouse rose crops. A single strain ofP. persimilis demonstrated tolerance to the field rate of 1.0 ml/l dimethoate used to controlHeliothrips haemorrhoidalis as indicated by the LC50 of 1.5 ml/l ai and a significantly different response at the LC90 level (P<0.05) to the three other strains tested. None of the strains was different in level of response to methidathion (P>0.05) and were unable to tolerate the recommended field rate of methidathion. Selection to dimethoate by routine field spraying has produced improved tolerance inP. persimilis. The dimethoate-tolerant strain was not different in developmental period and mean number of eggs produced per female when compared with the Sydney strain (P>0.05).  相似文献

Nine Washington populations ofTyphlodromus (=Metaseiulus) occidentalis Nesbitt collected from commercial apple orchards were surveyed for resistance to ten pesticides. A susceptible population collected from wild blackberry which had no history of pesticide exposure was used in estimating resistance development. All populations from apple orchards were tolerant to high concentrations of azinphosmethyl, diazinon, endosulfan and propargite. Resistance was also apparent to these materials, especially for azinphosmethyl which produced over 100-fold resistance levels for several populations. Moderate to low levels of tolerance and resistance development were seen to field concentrations of cyhexatin, formetanate and carbaryl. Fenvalerate, methomyl and methidathion were all very toxic at field concentrations and little evidence of resistance development was apparent. The use of fenvalerate, methomyl or methidathion at a time whenT. occidentalis is active would likely abrogate favorable biological control of pestiferous orchard mites.Scientific Paper No. 7502  相似文献

Engorged female Rhipicephalus sanguineus sensu lato (Ixodida: Ixodidae) were collected from dogs in the state of Yucatán, Mexico. Fourteen tick populations were collected from dogs at seven veterinary clinics, four residential homes and three cattle farms. The larval immersion test was used in the progeny of collected adult females to test susceptibility to amitraz and cypermethrin. Dose–mortality regressions, 50% lethal concentrations (LC50), confidence intervals and slope were estimated by probit analysis. For amitraz, 12 tick populations (85.7%) were classified as resistant and low inter‐population variation in the phenotypic level of resistance was evident [resistance ratios (RRs) at LC50: 1.0–13.0]. For cypermethrin, 12 tick populations (85.7%) were classified as resistant and substantial inter‐population variation in the phenotypic level of resistance was evident (RRs at LC50: 1.0–104.0). Thus, amitraz resistance in R. sanguineus s.l. is common, but generally occurs at low levels; however, alarmingly high levels of cypermethrin resistance are present in R. sanguineus s.l. populations in dogs in Yucatán, Mexico. The intensive use of both acaricides to control ectoparasites on dogs is likely to lead to more serious resistance problems that may cause high levels of control failure in the future.  相似文献

Two strains of Drosophila melanogaster represent the extremes in resistance and sensitivity to the lethal effects of CdCl2. The strain containing the mutations vermilion and brown (v; bw) and the strain Austin had LC50's of 3.3 and 1.3mm CdCl2, respectively. The three major chromosomes from these two strains were assorted genetically into the six possible combinations. The measured LC50's for CdCl2 for these six genotypes fell into two groups according to the X chromosome; those containing the X chromosome from v; bw had LC50's 0.5–1.0mm greater than those in which the X chromosome was from Austin. Since the parent strains differed by 2mm, we suggest that the X chromosome is a major, but not the sole, site of genes to produce resistance to CdCl2. When 109Cd was in the diet the uptake by v; bw and Austin over 2 days was the same. After 4 days of uptake, the Austin strain excreted the 109Cd five times faster than v; bw but the six genotypes did not differ appreciably in excretion rate from one another and resembled the sensitive parent Austin more than the resistant one. The seasonal changes of insecticide resistance and stability in hymenopteran Cotesia plutellae, collected from Jianxin, Fuzhou-City, and Shangjie, Minhou-County, Fujian, China, were assessed by using a dry residual film method. The resistance to two insecticides in the field populations of C. plutellae was not stable under insecticide-free conditions in the insectarium. Compared with susceptible F11 progeny of C. plutellae in the insectarium, the resistance ratios (RR) in F0 parents were 18.4 for fenvalerate and 11.4 for cypermethrin based on LC50 at 9 hours, and 32.8 for fenvalerate and 28.5 for cypermethrin based on LC50 at 24 hours when the parasitoids were left in contact with the insecticides for 1 hour and mortalities were recorded at 9 and 24 hours, respectively. However, the RR in a field population of C. plutellae were 9.2 for fenvalerate and 12.7 for cypermethrin, if the parasitoids were left in contact with the insecticides for 24 hours. The resistances to the two pyrethroids in other field populations collected from Jianxin and Shangjie from November 2000 and July 2004 were also determined. Significant seasonal variations of resistance to the two insecticides in the field populations of C. plutellae were found. The RR were 3.0-18.4 for fenvalerate and 4.8-20.6 for cypermethrin in Jianxin populations from November 2000 to April 2002 based on LC50 at 9 h, and 2.3-13.6 for fenvalerate and 3.6-16.0 for cypermethrin in Shangjie populations from May 2002 to July 2004 based on LC50 at 24 hours. The resistance levels were high in spring and autumn and decreased sharply in summer. In addition, significant recovery from the knocked-down caused by the insecticides was found in the F0 and field populations of C. plutellae which were resistant to fenvalerate and cypermethrin if the parasitoids were left in contact with the pyrethroids for 1 hour. In Argentina, studies on Varroa destructor resistance to coumaphos are still unknown. At present, high infestation levels of V. destructor are being detected in colonies of Apis mellifera after treatment with this acaricide. The aim of the present study was to determine the LC50 of coumaphos in V. destructor from four apiaries with high mite density after treatment with coumaphos. The LC50’s were 112, 319, 127 and 133 μg/Petri dish for mites from the four apiaries. Significant LC50 differences were detected between resistant and susceptible mites. LC50 increased 197–559-fold when compared to the corresponding baseline, suggesting the development of resistance. These results are the first report of resistance to coumaphos in V. destructor in Argentina.  相似文献

用胃毒触杀法确定8种常用杀虫剂对韭菜迟眼蕈蚊Bradysiaodoriphaga 3龄幼虫的LC20和LC50剂量后,以其LC20分别处理3龄幼虫,将存活幼虫正常饲养,测定该剂量药剂对韭菜迟眼蕈蚊化蛹率、蛹重、羽化率,成虫存活率、雌雄比、单雌产卵量及卵孵化率等指标的影响。结果表明:毒死蜱等4种药剂的影响较显著,其影响程度由高到低依次为毒死蜱>辛硫磷>阿维菌素溴虫腈;而另4种药剂丙硫克百威、灭多威、丁硫克百威和吡虫啉则影响不显著。此外也测定了用后4种药剂LC50剂量处理的存活试虫的上述生物学指标,结果表明:除化蛹率和羽化率较对照降低且差异显著外,其他生物学指标与对照处理无明显变化。  相似文献

The successful use of chemical rotations to manage insecticide resistance requires reversion between alternate chemical applications. We tested a tebufenpyrad resistant population of Tetranychus urticae Koch after some 55 months laboratory culture without pesticide selection and found LC50 level resistance had dropped from 63.29- to 2.41-fold. However, the population was still heterogeneous with LC99 level resistance at 38.03-fold. It is likely that a lack of reversion contributed directly to the initial tebufenpyrad control failure. Susceptibility of tobacco blue mould (Peronospora hyoscyami) to metalaxyl   总被引:1,自引:0,他引:1  
The lethal dose (LC50) of metalaxyl for a wild isolate of Peronospora hyoscyami f. sp. tabacina was determined using a bioassay. Infection levels were recorded using 5-wk-old tobacco seedlings that had been treated with foliar sprays of metalaxyl (Ridomil®) 24 h prior to inoculation. Plants were maintained in a controlled environment cabinet. LC50 estimates determined by probit analysis of the data from three independent experiments ranged between 0–46 and 0–58 μg/ml. Pooling the results for all three experiments, probit analysis gave a slope of 8·733 (S.E. 0·805) with an LC50 of 0·51 μg/ml (95% fiducial limits 0·45, 0·57). This determination will be useful as a guide to future monitoring of P. hyoscyami for resistance to metalaxyl.  相似文献

In this study, the knockdown and mortality effects of imidacloprid and methomyl were investigated. The residual surface applications were carried out to determine the knockdown effects (KDt50 and KDt95) and mortality (LD50 and LD95) induced by each insecticide. For mortality comparisons, the susceptible house fly (Musca domestica L., Diptera: Muscidae) of a WHO population and three natural field‐collected M. domestica populations from Turkey were used. In conclusion, it was found that the resistance to imidacloprid and methomyl was significantly higher in the field populations when compared to the susceptible population from WHO. The results showed that applicators and pest management decision‐makers should control and conduct an integrated pest management strategy by including biological agents to prevent the development of high levels of resistance in the field populations.  相似文献

Persea mite, Oligonychus perseae Tuttle, Baker, and Abatiello, susceptibility to abamectin and milbemectin was evaluated in 2003 to determine baseline susceptibility levels in avocado groves in San Diego and Ventura Counties (California, USA) where more than 70% of the state’s avocado production is concentrated. Milbemectin has yet to be used in avocado production in California and abamectin has been available for use since 1999. Baseline susceptibility ratios (in relation to the most susceptible population) of five persea mite field strains to milbemectin varied 2.1- to 2.8-fold at the LC50 and LC90, respectively. The susceptibility of seven field strains to abamectin varied slightly more (2.1- to 3.5-fold) with one strain subjected to seven sprays over the past 4 years showing slight but significant separation of LC50 and LC90’s from the most susceptible strain, which is suggestive of the early stages of resistance to this product. Based on these data, baseline susceptibility levels are proposed that might be used to monitor for future persea mite resistance to these chemicals as their use in California avocado production continues.  相似文献

This study aimed to monitor the present and future developments of the resistance of Tetranychus urticae Koch to fenpyroximate and pyridaben, using the relationship of the LC50 and slope of the concentration-mortality line in a probit model, for the provision of reliable resistance management tactics. Tetranychus urticae populations were collected from 16 commercial greenhouses, where various crops were cultivated, as well as from 10 apple orchards throughout Korea. The resistance to fenpyroximate and pyridaben of each population was estimated by calculating the median lethal concentration (LC50), resistance ratio (RR) and slope of the concentration-mortality regression. Most of the greenhouse populations exhibited moderate levels of resistance, whereas the apple orchard populations showed only low levels, indicating that T. urticae populations in greenhouses were more strongly selected than those in apple orchards. Four population groups were established based on either the habitats (greenhouse and apple orchard) or acaricides (fenpyroximate and pyridaben). To test the hypothesis, “the slope is greatest at low and high levels of resistance,” the slopes were regressed as a function of the LC50, and fitted to a polynomial regression. The polynomial regression model explained this relationship well for the four population groups (p < 0.05), indicating that the development of resistance toward fenpyroximate or pyridaben was consistent with the gradient. A laboratory selection study agreed with the results from both acaricide field populations. These results suggest that the gradient was a good indicator of the susceptibility of T. urticae to genetic variations, which was related to the LC50. The application of these findings is also discussed in relation to the resistance management of T. urticae.  相似文献

The mealybug Phenacoccus gossypiphilous (Stanley) played havoc with the cotton crop in Pakistan during 2005. To control this pest, insecticides of different groups were evaluated in both the laboratory and in field conditions. In the laboratory, bifenthrin, profenofos and chlorpyrifos proved to be the best insecticides for mealybug control, based on their susceptibility with the leaf dip method for their LC50. In field conditions, the recommended application rates of methomyl, profenofos and chlorpyrifos provided the best control: the lethal time studies proved their efficiency for better and timely control of this sporadic pest. The present study has shown that the insecticides tested, in particular profenofos, chlorpyrifos, methomyl and bifenthrin, provide satisfactory control of the cotton mealybug. The control of the insect pest complex throughout the cotton crop predominantly depends on wise and justified use of these chemicals, and necessitates development of an integrated pest management strategy.  相似文献

Tetranychus urticae Koch (Acari: Tetranychidae), twospotted spider mite, is a major secondary pest of strawberry and can cause significant yield loss. Tetranychus urticae is typically controlled using miticides, which has led to rapid resistance development. In South Carolina (USA), extension agents and growers have reported field failures of miticides (inadequate pest suppression), but resistance has not been quantitatively determined. In 2018, we determined the level of miticide resistance of six T. urticae populations found on strawberry across South Carolina. We examined efficacy of all miticides registered for use on US strawberry by conducting an initial diagnostic bioassay at 20% of the maximum labeled field rate. Any population?×?active ingredient combination resulting in?<?55% mortality was identified as ‘potentially resistant’ and concentration–response bioassays were then conducted to calculate LC50 values for an individual population. These values were compared with those of a known-susceptible laboratory population to calculate resistance ratios (RR). Our results indicate that examined South Carolina populations of T. urticae from strawberry were highly resistant to bifenthrin (RR?=?100–60,000) and there was reduced susceptibility to fenbutatin-oxide (RR?=?25–123). The 'Sardinia' population had decreased abamectin susceptibility (RR?=?25). No resistance to hexythiazox, etoxazole, acequinocyl, bifenazate, fenpyroximate, spiromesifen, or cyflumetofen was found. Based on available data, it appears that miticide resistance is not a likely cause of field failures and issues related to application error and coverage should be investigated. Overall, this work supports the need to reduce the use of broad-spectrum pesticides and older products, in favor of newer miticide chemistries due to resistance issues.

An enrichment culture method was applied to the isolation of a bacterial strain responsible for biodegradation of methidathion residues, from a methidathion-treated orchard. The strain (SPL-2) was identified as Serratia sp. according to its physiological characteristics and 16S rRNA gene phylogenetic analysis. Serratia sp. was able to grow in a poor medium consisting of mineral salts and using methidathion as the sole carbon source at a concentrations of 50–150 mg/L. The effects of multifactors on degradation of methidathion in pure cultures by Serratia sp. were investigated using an orthogonal experimental design L9 (34). On the basis of range analysis and ANOVA results, the most significant factors were temperature and inoculum size. The optimal conditions for methidathion biodegradation in pure cultures were a temperature in 30 °C, an inoculum size of 10 %, pH?=?7 and an aeration rate of 200 rpm. Two different concentrations of strain SPL-2 fermenting liquids (OD600?=?0.2 and OD600?=?0.4) were prepared and applied to remove methidathion residues from agricultural products, and this process can be described by a first order rate model. In contrast to controls, the DT50 of methidathion was shortened by 35.7 %, 8.2 % and by 62.3 %, 57.5 % on OD600?=?0.2 and OD600?=?0.4 treated haricot beans and peaches, respectively. These results suggest that the isolated bacterial strain may have potential for use in bioremediation of methidathion-contaminated crops.  相似文献

The present study was designed to investigate the neuropathological effect of the two carbamate pesticides: methomyl and methiocarb on the neurons of the buccal ganglia in the land snail Eobania vermiculata using topical application and baiting technique. Their in vivo effects on acetylcholinesterase (AChE, EC 3.1.1.7) activity were also investigated. Sublethal dose and concentration (1/4 LD50 and 1/4 LC50) of both pesticides were used, and the experiment lasted for 14 days. Histopathological and ultrastuctural alterations in the buccal ganglia were more obvious after the baiting technique treatment than after the topical application method, and methomyl was found to be more toxic than methiocarb. These alterations included shrinkage of the perikarya of neurons, increased cytoplasmic basophilia, and extreme indentation of the plasma membrane. In addition, the nuclei appeared karyolitic, eccentric, and highly shrunken with an irregular nuclear envelope. The most outstanding symptom observed after topical application of methiocarb was a highly vacuolated cytoplasm with a peripheral increase in electron density associated with dense accumulations of free ribosomes. On the other hand, an increased number of lysosomes and autophagosomes were observed after topical application of methomyl. Mitochondrial damage, increased number of lipid droplets, and myelin figures were frequently observed in ganglia treated with either methomyl or methiocarb. Moreover, it was noticed that both compounds induced reductions in AChE activity. However, methomyl exhibited more potency in reducing AChE activity than methiocarb.  相似文献

Imidacloprid and thiamethoxam are neonicotinoids that have been tested in several Orius species, including Orius laevigatus (Fieber) (Hemiptera: Anthocoridae), but not the variability in their effect among Orius populations of a single species. In this study, the variation in susceptibility to imidacloprid and thiamethoxam in 30 Mediterranean wild populations and four commercial populations of O. laevigatus was investigated in the laboratory using a standard dip bioassay method. Lethal concentration values (LC50) and the mortality of adults at the maximum field rate (MFR) were calculated. The range of LC50 of thiamethoxam was from 0.7 to 5.9 mg l?1, an 8.4‐fold variability, obtaining mortality at MFR (100 mg l?1) of >89.1% in all populations. The baseline obtained a value of 2.1 mg l?1, which is very low compared to the MFR. For imidacloprid, the LC50 varied from 7.7 to 94.7 mg l?1 (12.3‐fold variability). Mortalities at the MFR (150 mg l?1) were 57.7–99.2%, that is, more variable than for thiamethoxam. The LC50 value of the baseline was 48.7 mg l?1, also low compared to the MFR. This variation was exploited to select two populations resistant to thiamethoxam and imidacloprid, respectively. Artificial selection for on average 40 cycles significantly increased the resistance to thiamethoxam (LC50 = 149.1 mg l?1) and imidacloprid (LC50 = 309.9 mg l?1). Mortalities at the MFR in the thiamethoxam‐ and imidacloprid‐resistant populations were 44.5 and 36.9%, respectively. These results demonstrate that resistance can be enhanced in biocontrol agents by artificial selection under laboratory conditions, starting with populations showing no or very low tolerance. Our neonicotinoid‐resistant populations might enhance the wider adoption of biological control by allowing punctual or hotspot applications of neonicotinoids to control several main and secondary pests.  相似文献
