Effect of long term soil salinity (control-S0 and three levels S1 to S3) was studied in two maize (Zea mays L.) genotypes, PEHM 3 (comparatively tolerant) and Navjot (susceptible) at vegetative and anthesis stages during summer-rainy season. Salinity stress decreased relative water content (RWC), chlorophyll (Chl) and carotenoid (Car) contents, membrane stability index (MSI), potassium and calcium contents, and increased the contents of superoxide radical (O2 ·−), hydrogen peroxide (H2O2), thiobarbituric acid reactive substances (TBARS), proline, glycinebetaine, total soluble sugars, and sodium, and Na+/K+ and Na+/Ca2+ ratios in both the genotypes. Contents of zinc, copper, manganese and iron increased up to S2. Though under S0 PEHM 3 had higher content of all the metals, Navjot recorded higher content of Zn at all salinity levels and contents of all metal ions at S2 and S3. Activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), catalase (CAT) and glutathione reductase (GR) increased upto S2 in both the genotypes, and upto S3 in PEHM 3 at the two stages. Salinity induced decrease in RWC, Chl, Car, MSI, K+ and Ca2+ was significantly greater in Navjot, which also recorded higher Na+ content and Na+/K+ and Na+/Ca2+ ratios than PEHM-3. PEHM-3 recorded higher contents of proline, glycine-betaine, total soluble sugars, K+, Ca2+, activity of SOD, APX, CAT, GR, and comparatively lower O2 ·−, H2O2 and TBARS contents compared to Navjot.  相似文献

The objective of this study was to examine the role of antioxidant enzymes in waterlogging tolerance of pigeonpea (Cajanus cajan L. Halls) genotypes ICP 301 (tolerant) and Pusa 207 (susceptible). Waterlogging resulted in visible yellowing and senescence of leaves, decrease in leaf area, dry matter, relative water content and chlorophyll content in leaves, and membrane stability index in roots and leaves. The decline in all parameters was greater in Pusa 207 than ICP 301. Oxidative stress in the form of superoxide radical, hydrogen peroxide and thiobarbituric acid reactive substances (TBARS) contents initially decreased, however at 4 and 6 d of waterlogging it increased over control plants, probably due to activation of DPI-sensitive NADPH-oxidase. Antioxidant enzymes such as superoxide dismutase, ascorbate peroxidase, glutathione reductase and catalase also increased under waterlogging. The comparatively greater antioxidant enzyme activities resulting in less oxidative stress in ICP 301 could be one of the factor determining its higher tolerance to flooding as compared to Pusa 207. This study is the first to conclusively prove that waterlogging induced increase in ROS is via NADPH oxidase.  相似文献

Abscisic acid (ABA) and salicylic acid (SA) were sprayed on leaves of wheat genotypes C 306 and Hira at 25 and 40 d after sowing under moderate water stress (−0.8 MPa) imposed by adding PEG-6000 in nutrient solution. ABA and SA increased the activities of superoxide dismutase, ascorbate peroxidase, glutathione reductase, and catalase in comparison to unsprayed control plants. Both ABA and SA treatments decreased the contents of hydrogen peroxide and thiobarbituric acid reactive substances, a measure of lipid peroxidation, compared to unsprayed plants. The beneficial effect of increase in antioxidant enzymes activity and decrease in oxidative stress was reflected in increase in chlorophyll and carotenoid contents, relative water content, membrane stability index, leaf area and total biomass over control plants. The lower concentrations of ABA (0.5 mM) and SA (1.0 mM) were generally more effective than higher concentrations.  相似文献

Salinity could damage cellular membranes through overproduction of reactive oxygen species (ROS), while antioxidant capacities play a vital role in protecting plants from salinity caused oxidative damages. The objective of this study was to investigate the toxic effect of salt on the antioxidant enzyme activities, isoforms and gene expressions in perennial ryegrass (Lolium perenne L.). Salt-tolerant ‘Quickstart II’ and salt-sensitive ‘DP1′ were subjected to 0 and 250 mM NaCl for 12 d. Salt stress increased the content of lipid peroxidation (MDA), electrolyte leakage (EL) and hydrogen peroxide (H2O2), to a greater extent in salt-sensitive genotype. Salt-stressed plant leaves exhibited a greater activity of superoxide dismutase (SOD, EC 1.15.1.1), peroxidase (POD, EC 1.11.1.7), ascorbate peroxidase (APX, EC 1.11.1.11) at 4 d after treatment (DAT), but a lower level of enzyme activity at 8 and 12 d, when compared to the control. Catalase (CAT, EC 1.11.1.6) activity was greater at 4 DAT and thereafter decreased in salt tolerant genotype relative to the control, whereas lower than the control during whole experiment period for salt-sensitive genotype. There were different patterns of five isoforms of SOD, POD and two isoforms of APX between two genotypes. Antioxidant gene expression was positively related to isoenzymatic and total enzymatic activities during 12-d salt-treated leaves of two genotypes, with a relatively higher level in salt-tolerant genotype. Thus, salt tolerance could be related to the constitutive/induced antioxidant gene, leading to more efficient enzyme stimulation and protection in perennial ryegrass.  相似文献

Wheat (Triticum aestivum L.) genotypes K-65 (salt tolerant) and HD 2329 (salt sensitive) were grown in pots under natural conditions and irrigated with NaCl solutions of electrical conductivity (ECe) 4.0, 6.0, and 8.0 dS m−1. Control plants were irrigated without saline water. Observations were made on the top most fully expanded leaf at tillering, anthesis, and grain filling stages. The net photosynthetic rate (P N), stomatal conductance (g s), and transpiration rate (E) were reduced with the addition of NaCl. The reduction was higher in HD 2329 than in K-65. Salinity enhanced leaf to air temperature gradient (ΔT) in both the genotypes. NaCl increased the activities of superoxide dismutase (SOD) and peroxidase (POX); the percent increment was higher in K-65. The sodium and potassium contents were higher in the roots and leaves of K-65 over HD 2329. Thus at cellular level K-65 has imparted salt tolerance by manipulating P N, E, g s, and K accumulation in leaves along with overproduction of antioxidative enzyme activities (SOD and POX).  相似文献

Effects of long-term sodium chloride salinity (100 and 200 mM NaCl; ECe = 6.85 and 12.3 dS m–1) were studied in tolerant (Kharchia 65, KRL 19) and susceptible (HD 2009, HD 2687) wheat genotypes. NaCl decreased relative water content (RWC), chlorophyll content (Chl), membrane stability index (MSI) and ascorbic acid (AA) content, and increased the contents of hydrogen peroxide, thiobarbituric acid reactive substances (TBARS), and activities of superoxide dismutase (SOD), ascorbate peroxidase (APOX) and glutathione reductase (GR). Kharchia 65 showed lowest decline in RWC, Chl, MSI and AA content, lowest increase in H2O2 and TBARS contents and higher increase in SOD and its isozymes, APOX and GR, while HD2687 showed the highest decrease in AA content, highest increase in H2O2 and TBARS contents and smallest increase in activities of antioxidant enzymes. KRL 19 and HD 2009 showed intermediate response both in terms of oxidative stress and antioxidant activity.  相似文献

The response of two maize (Zea mays L.) genotypes, named GR (salt-tolerant) and SK (salt-sensitive), to salt stress (150 mM NaCl) was investigated under controlled environmental growth conditions. Genotype SK experienced more oxidative damage than the GR genotype when subjected to salt stress, which corresponded to higher O2 ? production rate and H2O2 content in the SK genotype than the GR genotype. Induction of caspase-like activity in response to salt stress was stronger in the SK genotype than in the GR genotype. On the other hand, induction of antioxidant enzyme activity to scavenge O2 ? and H2O2 in response to salt stress was weaker in the SK genotype than in the GR genotype. Consequently, the higher level of oxidative damage in the SK genotype in response to salt stress was manifested as more extensive cell death and biomass reduction in the SK genotype than it was in the GR genotype. Our results suggest that a direct relationship exists between salt stress-induced oxidative damage and cell death-inducing caspase-like activity, with tolerance to the salt stress being controlled by the efficiency of the plant antioxidant enzymes in limiting salt stress-induced oxidative damage and thus limiting cell death-inducing caspase-like activity.  相似文献

The effect of simultaneous expression of genes encoding three antioxidant enzymes, copper zinc superoxide dismutase (CuZnSOD, EC 1.15.1.1), ascorbate peroxidase (APX, EC 1.11.1.11), and dehydroascorbate (DHA) reductase (DHAR, EC 1.8.5.1), in the chloroplasts of tobacco plants was investigated under oxidative stress conditions. In previous studies, transgenic tobacco plants expressing both CuZnSOD and APX in chloroplast (CA plants), or DHAR in chloroplast showed enhanced tolerance to oxidative stresses, such as paraquat and salt. In this study, in order to develop transgenic plants that were more resistant to oxidative stress, we introduced the gene encoding DHAR into CA transgenic plants. Mature leaves of transgenic plants expressing all three antioxidant genes (CAD plants) had approximately 1.6–2.1 times higher DHAR activity, and higher ratios of reduced ascorbate (AsA) to DHA, and oxidized glutathione (GSSG) to reduced glutathione (GSH) compared to CA plants. CAD plants were more resistant to paraquat-induced stress, exhibiting only 18.1% reduction in membrane damage relative to CA plants. In addition, seedlings of CAD plants had enhanced tolerance to NaCI (100 mM) compared to CA plants. These results indicate that the simultaneous expression of multiple antioxidant enzymes, such as CuZnSOD, APX, and DHAR, in chloroplasts is more effective than single or double expression for developing transgenic plants with enhanced tolerance to multiple environmental stresses.  相似文献

Role of oxidative stress and antioxidant enzymes in Crohn's disease   总被引:1,自引:0,他引:1  
There is increasing interest in oxidative stress being a potential aetiological factor and/or a triggering factor in Crohn's disease, rather than a concomitant occurrence during the pathogenesis of the disease. Recent research has shown that the immune mononuclear cells of Crohn's disease patients are induced to produce hydrogen peroxide (H2O2). Similarly, the regulation of antioxidant enzymes during disease in these cells has been unravelled, showing that SOD (superoxide dismutase) activity and GPx (glutathione peroxidase) activity is increased during active disease and returns to normal in remission phases. However, catalase remains constantly inhibited which supports the idea that catalase is not a redox-sensitive enzyme, but a regulator of cellular processes. ROS (reactive oxygen species) can be produced under the stimulus of different cytokines such as TNFα (tumour necrosis factor α). It has been shown in different experimental models that they are also able to regulate apoptosis and other cellular processes. The status of oxidative stress elements in Crohn's disease and their possible implications in regulating cellular processes are reviewed in the present paper.  相似文献

Spermine and putrescine enhance oxidative stress tolerance in maize leaves   总被引:3,自引:0,他引:3  
The protective effects of spermine (SPM) and putrescine (PUT) against paraquat (PQ), a herbicide in agriculture and oxidative stress inducer, were investigated in the leaves of maize. Maize leaves were pretreated to SPM and PUT at concentrations of 0.2 and 1 mM and treated with PQ afterwards. Pretreatment with 1 mM of SPM and PUT significantly prevented the losses in chlorophyll and carotenoid levels induced by PQ. Ascorbic acid content in the leaves pretreated with both polyamines was found to be higher than those of the leaves pretreated with water. Also, pretreatment with SPM and PUT was determined to have some effects on the activities of superoxide dismutase (SOD) and peroxidase (POD). 1 mM of SPM increased SOD activity, but PUT has no significant effect on SOD activity. On the other hand, POD activity was recorded to increase slightly in response to both concentrations of SPM and 1 mM of PUT. The results showed that such polyamine pretreated plants may become more tolerant to oxidative stress due to increases in the antioxidative enzymes and antioxidants.  相似文献

Changes of carotenoids and anthocyanins content, lipid peroxidation, and activity of antioxidant enzymes were studied in wild type and tocopherol-deficient lines vte1 and vte4 of Arabidopsis thaliana subjected to 200 mM NaCI during 24 h. The salt stress enhanced the intensity of lipid peroxidation to different extent in all three plant lines. Salt stress resulted in an increase of carotenoid content and activity of catalase, ascorbate peroxidase, guaiacol peroxidase and glutathione reductase in wild type and tocopherol-deficient vte1 mutant. However, the increase in anthocyanins concentration was observed in vte1 mutants only. In vte4 mutant, which contain gamma-tocopherol instead of alpha-tocopherol, the response to salt stress occurred via coordinative action of superoxide dismutase and enzymes of ascorbate-glutathione cycle, in particular, ascorbate peroxidase, glutathione reductase, dehydroascorbate reductase, and glutathione-S-transferase. It can be concluded, that salt stress was accompanied by oxidative stress in three studied lines, however different mechanisms involved in adaptation of wild type and tocopherol-deficient lines to salt stress.  相似文献

Tolerance to low temperature and paraquat-mediated oxidative stress was investigated in two Zea mays genotypes, VA36 and A619, grown at 25/22 C and 16/14 C for 50 d after germination. VA36, the tolerant genotype, showed an enhanced resistance to paraquat as compared to A619, the sensitive genotype, when grown at low temperature. In VA36, superoxide dismutase and ascorbate peroxidase activities increased during growth at both 25/22 °C or 16/14 °C. In A619, superoxide dismutase activity was similar in plants grown at both 16/14 °C or 25/22 °C. Ascorbate peroxidase activity was always significantly lower in plants grown at low temperature than in plants grown at 25/22 °C. The total ascorbate peroxidase activity was correlated with the cytosolic ascorbate peroxidase protein content in all but A619 plants grown at low temperature for 25 d. The isozyme pattern of SOD showed a higher abundance of MnSOD in VA36 than in A619 and of FeSOD in A619 compared to VA36. Growth at low temperature enhanced resistance to paraquat infiltration more in VA36 than in A619. SOD and APX activities were generally higher and more stable with the increase of paraquat concentration in VA36 than in A619. Damage indicated by Fv/Fm and ion leakage after paraquat infiltration were generally higher in plants grown at 25/22 °C than at 16/14 °C and higher in A619 than in VA36. However, no causal link is proved between the extent of damage and the increase of SOD and APX activities alone. It is suggested that tolerance to oxidative stress requires an integrated enhancement of the antioxidant system.  相似文献

Nimbolide is a bioactive compound found in Azadirachta indica. This work was devised to investigate the potential effects of nimbolide on intracellular lipid deposition and its associated redox modulation in primary hepatocytes (Heps). Lipid accumulation was induced in Heps by supplementing 1 mM oleic acid for 24 h which was marked by significant accumulation of lipids. The results demonstrated that nimbolide can decrease intracellular cholesterol, free fatty acids and triglycerides. Nimbolide may also improve hepatocytes function through its antioxidant effects by inhibiting oxidative DNA damage and lipid peroxidation by curtailing the reactive oxygen species levels. Further it also restore the mitochondrial potential, improving the endogenous antioxidant levels such as GSH and antioxidant enzyme activities. Nimbolide increased (P?<?0.05) liver X receptor-α (LXRα), peroxisome proliferator-activated receptor-γ (PPARγ) and sterol regulatory element-binding protein-1c (SREBP1c) gene expression in Heps. The biological significance of nimbolide may involve hypolipidemic effect, lipid peroxidation inhibition, DNA damage inhibition, ROS inhibition, restoring mitochondrial function, increases in GSH and SOD & CAT activities, and direct regulation of LXRα, PPARγ and SREBP1c gene expression. Nimbolide may be used as effective lipid lowering compound and lipid deposition-induced Heps changes.  相似文献

Silicon is known to improve resistance against salinity stress in maize crop. This study was conducted to evaluate the influence of silicon application on growth and salt resistance in maize. Seeds of two maize genotypes (salt-sensitive ‘EV 1089’ and salt-tolerant ‘Syngenta 8441’) were grown in pots containing 0 and 2 mM Si with and without 50 mM NaCl. After detailed investigation of ion concentrations in different maize organs, both genotypes were further selected in hydroponic experiment on basis of their contrasting response to salinity stress. In the second experiment, pre-germinated seedlings were transplanted into nutrient solution with 0 and 60 mM NaCl with and without 2 mM Si. Both genotypes differed significantly in their response to salinity. Silicon addition alleviated both osmotic and oxidative stress in maize crop by improving the performance of defensive machinery under salinity stress. Silicon application also improved the water-use efficiency in both tested genotypes under both normal and salinity stress conditions. In conclusion, this study implies that the silicon-treated maize plants had better chance to survive under salinity conditions and their photosynthetic and biochemical apparatus was working far better than that of silicon-non-treated plants.  相似文献

The present study was aimed at investigating the effects of different concentrations of sodium alginate (NaAlg) (0.075 and 0.15% (w/v)) on the production of secondary metabolites (SMs) and antioxidant activity of seven safflower genotypes under in vitro salinity stress. The results showed that total phenolic content (TPC), total flavonoids (TFD), total flavonols (TFL), anthocyanin (Ant), total antioxidant capacity (TAC), phenylalanine ammonia-lyase (PAL), catalase (CAT) activity, and lipid peroxidation significantly increased under salinity stress consisting of the concentration of 1.5% (w/v) of NaCl, but callus growth traits decreased. The highest amount of TPC, Ant, and callus growth traits was observed under the elicitation of the sample with the concentration of 0.075(%) NaAlg under salinity stress, but the highest amount for TFD, TFL, CAT, PAL, and TAC was observed under elicitation of the sample with the concentration of 0.15% of NaAlg under salinity stress. This indicated the superiority of NaAlg for elicitation to increase SMs in safflower under salinity stress. Overall, the results showed that genotypes of Mex.22-191 and GE62918 could be processed to produce SMs by eliciting NaCl in safflower as an important medicinal plant at cellular level. A positive and significant correlation between CAT and TPC was observed and indicates that phenolic compounds are the major contributors to the antioxidant potential in safflower. This new elicitor introduced new ways to select and exploit the best NaAlg concentration to develop SMs that are of tremendous importance in terms of commercial purposes along with medical features in safflower at cellular level.

The role of plant antioxidant system in water stress tolerance was studied in three contrasting wheat genotypes. Water stress imposed at different stages after anthesis resulted in a general increase in lipid peroxidation (LPO) and decrease in membrane stability index (MSI), and contents of chlorophylls (Chl) and carotenoids (Car). Antioxidant enzymes like glutathione reductase and ascorbate peroxidase significantly increased under water stress. Genotype C 306, which had highest glutathione reductase and ascorbate peroxidase activity, also showed lowest LPO and highest MSI, and Chl and Car contents under water stress in comparison to susceptible genotype HD 2329, which showed lowest antioxidant enzyme activity as well as MSI, Chl and Car contents and highest LPO. HD 2285 which is tolerant to high temperature during grain filling period showed intermediate behaviour. Thus, the relative tolerance of a genotype to water stress as reflected by its comparatively lower LPO and higher MSI, Chl and Car contents is closely associated with its antioxidant enzyme system. The effects of single or combined stress of aluminum (Al) and chromium (Cr) on plant growth, root dehydrogenase, oxidative stress and antioxidative enzymes were studied using two barley genotypes differing in Al tolerance in a hydroponic experiment. Al or Cr stress decreased plant growth, lowered root dehydrogenase activity and caused oxidative damage, as characterized by increased MDA and H2O2 contents. Under Al or Cr stress, the activities of antioxidative enzymes, including superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX), glutathione reductase (GR) and catalase (CAT), were dramatically increased in plant tissues. Gebeina, an Al-tolerant genotype, had less oxidative damage than Shang 70-119, an Al-sensitive genotype. The extent of oxidative damage induced by Cr varied with the pH of the culture solution, with lower pH values (4.0) being more severe than higher pH values (6.5). The combination of Cr and Al caused a further decrease in plant growth, a decrease in root dehydrogenase activity and an increase in MDA and H2O2 contents as well as the activities of antioxidative enzymes. There was also a marked difference between the two barley genotypes in the extent of increased antioxidative enzyme activity under the Cr and Al stresses.  相似文献

In the present study, we measured the markers of oxidative stress as well as activity of antioxidative enzymes and content of α-tocopherol in the acclimated and non-acclimated cucumber (Cucumis sativus L.) cell suspension cultures subjected to 150 and 200 mM NaCl. The content of carbonyl groups and lipid peroxidation were lower in the acclimated cultures than in the non-acclimated ones as well as their increases after NaCl treatments. Both NaCl concentrations enhanced activity of glutathione peroxidase in the examined cultures whereas activity of glutathione-S-transferase rose only in the acclimated ones. The increase in content of α-tocopherol induced by NaCl was more pronounced in the acclimated cultures. NaCl caused high decline in cell vigour in the non-acclimated cultures up to 80–90 % at the end of the experiment. The presented data suggest that the acclimated cultures coped with the salt stress better than the non-acclimated ones.  相似文献
