Subepithelial blood vessels of the rat lingual papillae and their spatial relations to the connective tissue papillae and surface structures were demonstrated by light and scanning electron microscopy. In the rat, four types of papillae were distinguished on the dorsal surface of the tongue, i.e. the filiform, fungiform, foliate and circumvallate papillae. Vascular beds of various appearance were found in all four types of lingual papillae: a simple or twisted capillary loop in the filiform papilla; a basket- or petal-like network in the fungiform papilla; a ring-like network in the foliate papilla, and a conglomerated network surrounded by double heart-shaped capillary networks in the circumvallate papilla. These characteristic vascular beds corresponded to the shape of the connective tissue papillae and surface structures. The vascular bed beneath the gustatory epithelium in the fungiform, foliate and circumvallate papilla consisted of fine capillary networks next to the taste buds.  相似文献

Study by transmission and scanning electron microscopy of the morphogenesis of three types of lingual papillae in the albino rat (Rattus rattus).—Acta Zoologica (Stockholm) 91 : 267–278 Tongues were removed from albino rat foetus on days 12 (E12) and 16 (E16) of gestation and from newborns (P0) and from juvenile rats on days 7 (P7), 14 (P14) and 21 (P21) postnatally for investigation by light, scanning, and transmission electron microscopy. Significant changes appeared during the morphogenesis of the papillae. At E12, two rows of rudiments of fungiform papillae were extended bilaterally on the anterior half of the tongue. At E16, the rudiments of fungiform papillae were regularly arranged in a lattice‐like pattern. A rudiment of circumvallate papillae could be recognized. No rudiment of filiform papillae was visible. No evidence of keratinization was recognizable. At P0, rudiments of filiform papillae were visible but had a more rounded appearance, with keratinization. The fungiform and circumvallate papillae were large and their outlines were somewhat irregular as that found in the adult rat. At P7, the filiform papillae were large and slender. The fungiform papillae became large and the shape of circumvallate papillae was almost similar to that observed in the adult. At P14 and P21, the shape and structure of the three types of papillae were irregular as those found in the adult. In conclusion, the rudiments of the fungiform and circumvallate papillae were visible earlier than those of the filiform papillae. The morphogenesis of filiform papillae advanced in a parallel manner with the keratinization of the lingual epithelium, in the period from just before birth to a few weeks after birth.  相似文献

In mammals, taste buds develop in different regions of the oral cavity. Small epithelial protrusions form fungiform papillae on the ectoderm-derived dorsum of the tongue and contain one or few taste buds, while taste buds in the soft palate develop without distinct papilla structures. In contrast, the endoderm-derived circumvallate and foliate papillae located at the back of the tongue contain a large number of taste buds. These taste buds cluster in deep epithelial trenches, which are generated by intercalating a period of epithelial growth between initial placode formation and conversion of epithelial cells into sensory cells. How epithelial trench formation is genetically regulated during development is largely unknown. Here we show that Pax9 acts upstream of Pax1 and Sox9 in the expanding taste progenitor field of the mouse circumvallate papilla. While a reduced number of taste buds develop in a growth-retarded circumvallate papilla of Pax1 mutant mice, its development arrests completely in Pax9-deficient mice. In addition, the Pax9 mutant circumvallate papilla trenches lack expression of K8 and Prox1 in the taste bud progenitor cells, and gradually differentiate into an epidermal-like epithelium. We also demonstrate that taste placodes of the soft palate develop through a Pax9-dependent induction. Unexpectedly, Pax9 is dispensable for patterning, morphogenesis and maintenance of taste buds that develop in ectoderm-derived fungiform papillae. Collectively, our data reveal an endoderm-specific developmental program for the formation of taste buds and their associated papilla structures. In this pathway, Pax9 is essential to generate a pool of taste bud progenitors and to maintain their competence towards prosensory cell fate induction.  相似文献

The dorsal surface of the mammalian tongue is covered with four kinds of papillae, fungiform, circumvallate, foliate and filiform papillae. With the exception of the filiform papillae, these types of papillae contain taste buds and are known as the gustatory papillae. The gustatory papillae are distributed over the tongue surface in a distinct spatial pattern. The circumvallate and foliate papillae are positioned in the central and lateral regions respectively and the fungiform papillae are distributed on the anterior part of the tongue in a stereotyped array. The patterned distribution and developmental processes of the fungiform papillae indicate some similarity between the fungiform papillae and the other epithelial appendages, including the teeth, feathers and hair. This is because 1) prior to the morphological changes, the signaling molecules are expressed in the fungiform papillae forming area with a stereotyped pattern; 2) the morphogenesis of the fungiform papillae showed specific structures in early development, such as epithelial thickening and mesenchymal condensation and 3) the fungiform papillae develop through reciprocal interactions between the epithelium and mesenchymal tissue. These results led us to examine whether or not the early organogenesis of the fungiform papillae is a good model system for understanding both the spacing pattern and the epithelial-mesenchymal interaction during embryogenesis.  相似文献

Summary Taste buds (TB) in the foliate, circumvallate and fungiform papillae of the rabbit tongue were examined with lectin histochemistry by means of light (LM) and electron (EM) microscopy. Biotin- and gold-labeled lectins were used for the detection of carbohydrate residues in TB cells and subcutaneous salivary glands. At the LM level, the lectins of soybean (SBA) and peanut (PNA) react with material of the foliate and circumvallate taste pores only after pretreatment of the section with neuraminidase. This indicates that the terminal trisaccharide sequences are as follows: Sialic acid-Gal-GalNAc in O-glycosylated glycoproteins or Sialic acid-Gal-GlcNAc in N-glycosylated glycoproteins. In fungiform taste buds the lectins of Dolichos biflorus (DBA) and Helix pomatia (HPA), also specific to GalNAc residues, are reactive without preincubation with neuraminidase. Wheat germ agglutinin (WGA), specific to GlcNAc, reacts with TBs of all papillae; and the lectin from Ulex europaeus (UEA I), specific to fucose, binds to individual TB cells. The presence of sialic acid may protect mucus or other glycoproteins in TB cells and inside the taste pore from premature enzymatic degradation. In a post-embedding EM procedure on LR-White-embedded tissue sections, only gold-labeled HPA was found to bind especially on membrane surfaces of the microvilli which protrude into the taste pore; however HPA did not bind to the electron-dense mucus inside the taste pore. The mucus situated in the trough and at the top of the adjacent epithelial cells also is strongly HPA-positive, but is of different origin and composition than that found in the taste pore. These results demonstrate distinct carbohydrate histochemical differences between fungiform and circumvallate/foliate taste buds. The different configuration of galactosyl residues and the occurrence of mannose in circumvallate and foliate TBs leads to the suggestion that the lectin reactivities of TBs are not only due to the presence of mucins, but also to N-linked glycoproteins, possibly with a hormone-like, paraneuronal function. A possible relationship to v. Ebner glands in these papillae is discussed.  相似文献

Immunoreactivity to neuron-specific enolase (NSE), a specific neuronal marker, and calcitonin gene-related peptide (CGRP) was localized in lingual taste papillae in the pigs. Sequential staining for NSE and CGRP by an elution technique allowed the identification of neuronal subpopulations. NSE-staining revealed a large neuronal network within the subepithelial layer of all taste papillae. NSE-positive fibers then penetrated the epithelium as isolated fibers, primarily in the foliate and circumvallate papillae, or as brush-shaped units formed by a multitude of fibers, especially in the fungiform papillae and in the apical epithelium of the circumvallate papilla. Taste buds of any type of taste papillae were found to express a dense subgemmal/intragemmal NSE-positive neuronal network. CGRP-positive nerve fibers were numerous in the subepithelial layer of all three types of taste papillae. In the foliate and circumvallate papillae, these fibers penetrated the epithelium to form extragemmal and intragemmal fibers, while in the fungiforms, they concentrated almost exclusively in the taste buds as intragemmal nerve fibers. Intragemmal NSE- and CGRP-positive fiber populations were not readily distinguishable by typical neural swellings as previously observed in the rat. The NSE-positive neuronal extragemmal brushes never expressed any CGRP-like immunoreactivity. Even more surprising, fungiform taste buds, whether richly innervated by or devoid of NSE-positive intragemmal fibers, always harboured numerous intragemmal CGRP-positive fibers. Consequently, NSE is not a general neuronal marker in porcine taste papillae. Our observations also suggest that subgemmal/intragemmal NSE-positive fibers are actively involved in synaptogenesis within taste buds. NSE-positive taste bud cells were found in all three types of taste papillae. CGRP-positive taste bud cells were never observed.  相似文献

An investigation was undertaken to study the neuro-histochemistry of tonue of Gallus domesticus (white Leghorn) by cholinesterase technique under maintained pH of 5.2, temperature 40 degrees C and incubation period 20 h, and Ungewitter's silver impregnation method. Fungiform (8--16 in number), filiform (5--16 in number), circumvallate and foliate (numerous) were recorded profusely innervated in the various regions of the tongue. Neuro-muscular spindles related either with the non-myelinated, myelinated nerve fibres or with the ganglia via pre- or postganglionic nerve fibre showed cholinesterase (AChE) positive activity in the form of dark brown patches. Neural network over the glands, formed the basket envelope likestructures. Glands showed positive cholinesterase (AChE) reaction on the periphery. Dot-like nerve endings were connected with the neural terminal network in the foliate papillae region and motor end plate like endings on blood vessel, were occasionally recorded. Parasympathetic ganglia of various shape and sizes, arranged in chain-like fashion were observed in the connective tissue space between the fungiform and circumvallate papillae and foliate papillae region. The pre- and postganglionic fibres of these ganglia were related with the anteriorly and posteriorly located papillae via the neural terminal network. Cholinesterase (AChE) activity was marked either in the centre, of the fungiform or on the periphery of the filiform, circumvallate and foliate papillae.  相似文献

The role of amiloride-sensitive Na+ channels (ASSCs) in the transduction of salty taste stimuli in rat fungiform taste buds has been well established. Evidence for the involvement of ASSCs in salt transduction in circumvallate and foliate taste buds is, at best, contradictory. In an attempt to resolve this apparent controversy, we have begun to look for functional ASSCs in taste buds isolated from fungiform, foliate, and circumvallate papillae of male Sprague-Dawley rats. By use of a combination of whole-cell and nystatin-perforated patch-clamp recording, cells within the taste bud that exhibited voltage-dependent currents, reflective of taste receptor cells (TRCs), were subsequently tested for amiloride sensitivity. TRCs were held at - 70 mV, and steady-state current and input resistance were monitored during superfusion of Na(+)-free saline and salines containing amiloride (0.1 microM to 1 mM). Greater than 90% of all TRCs from each of the papillae responded to Na+ replacement with a decrease in current and an increase in input resistance, reflective of a reduction in electrogenic Na+ movement into the cell. ASSCs were found in two thirds of fungiform and in one third of foliate TRCs, whereas none of the circumvallate TRCs was amiloride sensitive. These findings indicate that the mechanism for Na+ influx differs among taste bud types. All amiloride-sensitive currents had apparent inhibition constants in the submicromolar range. These results agree with afferent nerve recordings and raise the possibility that the extensive labeling of the ASSC protein and mRNA in the circumvallate papillae may reflect a pool of nonfunctional channels or a pool of channels that lacks sensitivity to amiloride.  相似文献

Salivary carbonic anhydrase VI (CA VI) appears to contribute to taste function by protecting taste receptor cells (TRCs) from apoptosis. The serous von Ebner's glands locating in the posterior tongue deliver their saliva into the bottom of the trenches surrounding the TRC-rich circumvallate and foliate papillae. Because these glands deliver their saliva directly into the immediate vicinity of TRCs, we investigated whether CA VI is secreted by the von Ebner's glands, using immunochemical techniques. The immunohistochemical results showed that CA VI is present in the serous acinar cells, ductal cells, and ductal content of von Ebner's glands and in the demilune and ductal cells plus ductal content of rat lingual mucous glands. More importantly, CA VI was also detected in taste buds and in the taste pores. Western blotting of saliva collected from the orifices of human von Ebner's glands and CAs purified from rat von Ebner's glands confirmed that CA VI is expressed in these glands and secreted to the bottom of the trenches surrounding the circumvallate and foliate papillae. These findings are consistent with the hypothesis that locally secreted CA VI is implicated in the paracrine modulation of taste function and TRC apoptosis. During development, axons of the chorda tympani nerve navigate to fungiform papillae where they penetrate the lingual epithelium, forming a neural bud. It is not known whether or not all chorda tympani axons initially innervate fungiform papillae correctly or if mistakes are made. Using a novel approach, we quantified the accuracy with which gustatory fibers successfully innervate fungiform papillae. Immediately following initial targeting (E14.5), innervation was found to be incredibly accurate: specifically, 94% of the fungiform papillae on the tongue are innervated. A mean of five papillae per tongue were uninnervated at E14.5, and the lingual tongue surface was innervated in 17 places that lack fungiform papillae. To determine if these initial errors in papillae innervation were later refined, innervation accuracy was quantified at E16.5 and E18.5. By E16.5 only two papillae per tongue remained uninnervated. Innervation to inappropriate regions was also removed, but not until later, between E16.5 and E18.5 of development. Therefore, even though gustatory fibers initially innervate fungiform papillae accurately, some errors in targeting do occur that are then refined during later embryonic periods. It is likely that trophic interactions between gustatory neurons and developing taste epithelium allow appropriate connections to be maintained and inappropriate ones to be eliminated.  相似文献

Two experiments were conducted to investigate the psychophysicalresponse characteristics of single circumvallate papillae. InExperiment 1, 12 circumvallate papillae in four subjects werechemically stimulated to assess identification of taste qualities.Single circumvallate papillae were found to mediate multipletaste qualities, and the taste profiles obtained from differentpapillae were similar within the same subject. Moreover, sucrose,quinine monohydrochloride and citric acid elicited unitary andcharacteristic quality responding in these papillae from allsubjects, whereas NaCl elicited predominantly sour and/or bitterresponses from three of the four subjects. Experiment 2 directly compared responses obtained from singlecircumvallate papillae with those obtained from fungiform regionsof the tongue. Data for 10 subjects showed significantly greatersour responses to citric acid and NaCl in circumvallate papillaeand significantly greater salty responses to these compoundson the anterior tongue. In addition, the taste profiles forcitric acid and NaCl were distinct for circumvallate papillae,while those from the anterior tongue were similar. These datasuggest that the bitterness and sweetness of quinine and sugar,respectively, can be identified on the basis of sensory informationarising from either circumvallate or fungiform regions, butthat differentiation of the tastes of salts and acids may dependon a comparison of the input from both regions and/or additionalinformation arising from foliate regions.  相似文献

The dual innervation of the foliate papillae of the rat by thechorda tympani and the glossopharyngeal nerves was determinedin this electrophysiological study. The magnitude of the chordatympani response to the chemical stimulation of the foliatepapillae was small and variable in different animals. Summatedresponses of both the chorda tympani and the glossopharyngealnerves to the stimulation of the foliate papillae showed similarpatterns such as long latencies, slowly rising responses afteronset of stimulation and slow declines by water ringing. Therelative responsiveness to 5 different chemicals applied tothe foliate papillae were almost the same for the chorda tympaniand the glossopharyngeal nerves. The staining and demonstration of neural elements in relation with other associated structures was most satisfactory at pH 5.2, incubation period 20 h, and temperature 40degreesC. Ganglia of various shape and size were observed on or near the fungiform, filiform and foliate papillae. In the region of the fungiform papillae, a chain of ganglia (parasympathetic) was observed. The nerve cells of these ganglia showed cholinesterase (ChE) activity. The neural networks of the foliate papillae were connected with the neural terminal network and occasionally to the ganglia. The innervation of blood vessels (artery and vein) was profuse. At times ganglia were noticed on the periphery of the artery. Neuromuscular spindle-like structures were noticed occasionally. Dot-like free nerve endings were also observed. The ChE activity was very marked in the muscle fibres lying in the region of the circumvallate papillae but was less marked in the muscle fibres lying in the region of the filiform and fungiform papillae.  相似文献

Summary The influence of salivation on the location of gustatory alkaline phosphatase has been examined. In untreated rats, taste buds at the ends of fungiform papillae showed almost no activity. However, if salivation was suppressed for 12 hours in fasted animals, alkaline phosphatase activity could be clearly demonstrated in association with these taste buds. The results indicated that alkaline phosphatase may be removed from its site of secretion by saliva and that the enzyme is secreted from fungiform as well as circumvallate and foliate papillae.  相似文献

The morphogenesis of fungiform papillae occurs in a stereotyped pattern on the dorsal surface of the mammalian tongue via epithelial–mesenchymal interactions. These interactions are thought to be achieved via intercellular communication. Gap junctions can be observed in many developing tissues and have been suggested to participate in a variety of functions, including the regulation of cell proliferation, differentiation, and apoptosis. Here, we demonstrate that the expression of Connexin 43 (Cx43), a gap junction protein, is correlated significantly with the development of fungiform papillae, which exhibit a pattern formation and morphogenesis similar to the development of other epithelial appendages. Antisense-oligodeoxynucleotide (AS-ODN) against Cx43 was used to assess the developmental functions of Cx43. The expression patterns of the signaling molecules were disrupted by Cx43 inhibition. Interestingly, the expression patterns of Shh, a key molecule in the determination of the spacing patterns of fungiform papillae, were disturbed after treatment with Cx43 AS-ODN. We have also attempted to determine the functions of Bmp-2 by applying NOGGIN protein to tongue cultures. Our results indicate that upstream regulation via Cx43 controls the Shh and Bmp-2 pathways for the morphogenesis and pattern formation of fungiform papillae.This work was supported by grant no. Localization of type II collagen in the lingual mucosa of rats during the morphogenesis of circumvallate papillae. —Acta Zoologica (Stockholm) 92 : 67–74. Immunoreactivity specific for type II collagen was recognized first in the mesenchymal connective tissue just beneath the circumvallate papilla placode in fetuses on E13. At this stage, most of the lingual epithelium was pseudostratified epithelium composed of one or two layers of cuboidal cells. However, the epithelium of the circumvallate papilla placode was composed of several layers of cuboidal cells. Immunoreactivity specific for type II collagen was detected mainly on the lamina propria just beneath the lingual epithelium of the rudiment of the circumvallate papilla in fetuses on E15 and on E17, and slight immunostaining was detected on the lamina propria around the rudiment. In fetuses on E19, immunoreactivity specific for type II collagen was widely and densely distributed on the connective tissue around the developing circumvallate papillae and on the connective tissue that surrounded the lingual muscle. Immunoreactivity specific for type II collagen was sparsely distributed on the lamina propria of central bulge. After birth, morphogenesis of the circumvallate papillae advanced gradually with the increase in size of the tongue. Immunoreactivity specific for type II collagen was distinctively distributed in the lamina propria around circumvallate papilla, in the central bulge, and in the connective tissue that surrounded the lingual muscle.  相似文献

The localization of adenylate cyclase activity in the fungiform,foliate and circumvallate papillae of rats, rabbits, cats anddogs was determined histochemically using an incubation mediumwith a high pH. Light-microscopic study showed that adenylatecyclase activity is localized not only at the apex of tastebuds but also in other tissues, such as the von Ebner's glandsand the blood vessels or capillaries. The adenylate cyclaseactivity at the apex of taste buds was detectable in all thetaste papillae of rats, rabbits, cats and dogs except for thefungiform papillae of rabbits, though the amount of reactionproduct varied in different papillae. Electron-microscopic studyshowed that the number and density, as well as the size, ofsmall round-shaped electron-dense granules caused by the precipitationof lead with imidodiphosphate at the apex of taste buds arelow in the circumvallate papillae of cats compared with thosein the foliate papillae of rabbits. This may explain the resultthat the amount of reaction product varied in different papillae.  相似文献
