Where its populations are isolated in higher mountain regions, Abies mariesii is one of the more important conifers of Japan's alpine forest zone. In this study we tried to clarify the genetic variation of chloroplast DNA (cpDNA) in A. mariesii. Cones and fresh needles were collected from seven mountain regions. Total DNAs were extracted from individual seedlings, and these were digested by 15 restriction endonucleases. Southern hybridization was then done using cpDNA clones of Cryptomeria japonica and tobacco as probes. CpDNA variation was detected with enzyme-probe combinations: HindIII+pCS10 probe, HindIII+pCS7, and BglII+pCS7 in preliminary screening. These variations were considered to be caused by the same insertion, deletion or inversion. All populations surveyed for the combination HindIII+pCS10 resulted in only two frequency variations in each population. This indicates a gradual cline along latitude and longitude.  相似文献

Allelic frequencies at the polymorphic enzyme loci IDH-1, IDH-2, LAP-1, and 6-PGDH-1 were determined in population samples ofAbies alba from 5 different areas in Austria. The results reveal clear genetic differentiation between the eastern and the western provenances. Gradual frequency clines are observed with all four loci. The amount of variation, measured as average heterozygosity and genotypic diversity, is significantly higher in the west. The multilocus genotypes proved to be useful markers for the identification of clones. Functional differences with respect to substrate specifity were found for the two allozymes coded by the coexisting alleles of the IDH-2 locus.  相似文献

The growth pattern was analysed forAbies mariesii Mast. saplings in the subalpine climax forests of the northern Yatsugatake Mountains, Central Japan. The lateral branches were distinguished from the trunk not only with respect to the branching habits and needle arrangement but also with respect to the distribution of dry matter to needles, the slenderness of shoot-stem and the longevity of needles on them. Based on these differences, the contribution of the trunk and lateral branches to the matter production function of saplings is discussed. The lateral branches showed plastic changes in their growth characters in order to expand and maintain an assimilation system effectively, under conditions of suppression. In addition, the trunk growth was inhibited compared with the lateral branch growth and the apical dominance among lateral branches was weakened in the suppressed saplings. It was concluded that these growth habits resulted in the observed difference in the crown shape of saplings in the different habitats. Arguments are presented regarding the adaptive meanings of such a growth pattern for the shade tolerance and survival of the saplings and for the regeneration of the forest.  相似文献

Seedling and tree-establishment microhabitats of Tsuga diversifolia and Abies mariesii were examined on the herb- and dwarf bamboo-dominated forest floor on Mount Hayachine, a mountain under intermediate snow conditions in northern Honshu, Japan. The four microsite types were fallen logs, buttresses, rocks and ground. The ground substratum was further divided into four subtypes by dominant undergrowth species: Lycopodium, Pteridophyllum, Carex and dwarf bamboo. The establishment of T. diversifolia seedlings on the ground was scarce, and depended mostly on non-ground microsites (i.e. fallen logs, buttresses and rocks). The seedling establishment of A. mariesii was not dependent on specific substrata, although on the ground, establishment sites were limited to the Lycopodium subtype situated on convex sites. Among the microhabitats for seedling establishment, larger trees of A. mariesii rarely occurred on higher portions of the non-ground microsites. In contrast, T. diversifolia could grow up to the height of canopy-layer trees in such microsites. Thus, non-ground microsites seem to be unsuitable for tree establishment in A. mariesii, and are probably useful microhabitats for regenerating T. diversifolia to avoid competition with A. mariesii. We also compared seedling-establishment microhabitats for the two conifers between Mount Hayachine and two other mountain regions under different undergrowth conditions (moss-dominated and dense dwarf bamboo-dominated). Our findings suggested that seedling recruitment in non-ground microsites was primarily determined by undergrowth conditions; T. diversifolia preferred such microsites where the moss-covered area was low, and A. mariesii preferred where dwarf bamboo-covered area was high.  相似文献

Summary Variation at a polymorphic Aspartate aminotransferase locus was assayed in a sample of 298 accessions from the ICARDA germplasm collection of the domesticated lentil (Lens culinaris). Two alleles Aat-1 F and Aat-1 S were detected with global frequencies of 0.51 and 0.49, respectively. Fifty-nine percent of accessions were polymorphic for both alleles. The frequency of outcrossing was estimated from the observed heterozygosity to be about 1%. This is higher than direct estimates of outcrossing and implicates selection in favour of heterozygous gene combinations. Significant variation in allele frequency and in the occurrence of polymorphic accessions was observed between countries or geographic areas. Significant associations were observed between the allozymes and agronomic characters. In particular high frequency of Aat-1 F appeared to be associated with late flowering and maturity and low yield.  相似文献

Allozyme variation in viviparid snails from the genus Mekongia in Thailand were examined across the different species, subspecies and geographical locations (river drainage systems). Using horizontal starch gel electrophoresis, 11 presumed allozyme loci (eight polymorphic) from eight enzyme systems were screened. Heterozygosity was moderately low (Hexp = 0.000–0.109, mean = 0.037). One population of Mekongia pongensis (Nong Khai) was monomorphic at all 11 examined loci in contrast to the other two populations, suggesting bottleneck within this population. Populations were more differentiated in the Mekongia sphaericula complex (FST = 0.587) than in either the Mekongia swainsoni complex (FST = 0.161) or M. pongensis species (FST = 0.073). Mekongia sphaericula sphaericula and Mekongia sphaericula extensa exhibited fixed allele differences at two loci, a high genetic distance (D = 0.265–0.300) and a potential polyphyletic relationship, suggesting two distinct lineages (species).  相似文献

In the subalpine areas of the snowy regions of Japan (the Japan Sea side), there are some mountains with no or very small stands ofAbies mariesii, although this species dominates the subalpine coniferous forests of the region. In order to discuss the cause and process of this phenomenon, present horizontal and vertical ranges, as well as physiographic conditions, of theA. mariesii forest were examined in detail on the mountains in the Tohoku District. Sites in the subalpine zone were classified into two types: ‘azonal sites’ which should be excluded from the habitat ofA. mariesii because of their edaphic or small-scale climatic properties, and ‘zonal sites’. Mountains with vast less-inclined zonal sites generally had well developed stands ofA. mariesii forest. On the mountains with only small, solitary stands ofA. mariesii, the distribution was limited to flats or slightly inclined slopes at relatively low altitudes. These less-inclined zonal sites were regarded as an important habitat for theA. mariesii forest in the Hypsithermal period and the extent of these sites controls the extent of the stands in that period and the success of the subsequent range expansion of the forest.  相似文献

Patterns of allozyme variation have been investigated in 101 clones representing most of the morphological variation (and taxa) found in the amphi-apomictic genus Pilosella Hill. in the Nordic countries. Levels of variation were very high, both within narrowly defined taxa and within the whole data-set, but the correlation between a morphology-based taxonomy and patterns of allozyme variation was very weak. Almost all enzyme bands were found in more than one taxon and the differences between broadly defined species in terms of the frequency of bands were generally small. Cluster analyses did not perfectly recover any taxon but there were some tendencies for samples of some broadly defined taxa (e.g. P. officinarum s. l., and P. cymosa ssp. cymosa) to cluster together. It is concluded that there is considerable gene-flow among virtually all Nordic morpho-types of Pilosella and this is considered an argument for applying a very broad species concept in this genus.Financial support for the present study has been recieved from Stiftelsen Harald E. Allozyme variation was examined in three diploid taxaChionographis japonica var.japonica, var.kurokamiana, andC. koidzumiana and three tetraploid taxaC. japonica var.kurohimensis, ssp.hisauchiana, and ssp.minoensis. Results show thatC. japonica var.kurokamiana is genetically closer toC. koidzumiana than to var.japonica. In the tetraploid taxa, fixed heterozygosities were found at several loci, and this supports the hypothesis that these taxa are allotetraploids. Furthermore, the tetraploid taxa have many unique alleles not found in the diploid taxa. This suggests that sufficient time has passed since the origin of tetraploids for new mutations to have been fixed.  相似文献

Clinal variation in life histories can be genetically based, resulting from selection imposed by different environments, or it may be due to the differential expression of phenotypically plastic traits. We examined the cline in voltinism in the egg-diapausing cricket Allonemobius socius, with populations spanning the switch from a univoltine to a bivoltine phenology. A common garden experiment was employed, using environments that mimicked photoperiod and temperature conditions found in the field. There were only small differences in development time among populations, and the difference in phenology observed in the field is likely due to clinal variation in the length of the growing season. We found large genetically-based differences in the reaction norm for egg diapause that were further magnified by environmental cues. The synergism of genetic and environmental effects was an example of cogradient selection. In the zone of transition between phenologies, voltinism appeared to be a conditional strategy, rather than a genetic polymorphism. First-generation females from this area can lay either direct-developing or diapause eggs depending on the likelihood that a second generation will have sufficient time to develop. For this species, the cline in voltinism is the result of a combination of environmental effects on development, and genetic and environmental influences on egg diapause propensity.  相似文献

利用水平板淀粉凝胶电泳技术检验了板栗8个居群在6个同工酶位点(Per-2、Got-2、Est-2、Gpi-2、Idh-1、Idh-2 )上的遗传变异(多态位点百分率为97.9%,平均等位基因数为2.54,平均预期杂合值为0.452 ) 。在同一水平上,其遗传多样性水平显著高于欧洲栗。总的基因多样性中,89.2%发生在居群内,10.8%发生于居群间,各居群之间的遗传距离为0.036~0.394,有些居群分化较大,尤其在个别位点上更为明显。初步研究表明,其丰富的遗传变异及其高水平的分化度可能与其生境的多样性、风媒异交等因素造成的长距离的基因漂变有关。  相似文献

Allozyme variation at eleven loci encoding seven enzyme systems were examined in 20 populations of diploid (genome AA, 2n = 16)Scilla scilloides in China. In comparison with the average species of seed plants studied, populations of this species display a high amount of genetic variation (A = 2.0, P = 58.6%, Ho = 0.172, and He = 0.185). Allozyme variation pattern revealed predominant outcrossing within populations and considerable differentiation (FST = 0.314) among populations as well as between the subtropic and temperate regions. The wide distribution, long existence and outcrossing are presumably the main factors responsible for the high genetic diversity within populations. But the gravity dispersal of seeds and pollination by small insects set limits to the increase of genetic variation within populations and promote differentiation between populations and regions. In addition, allozyme variation does not distinguishS. scilloides var.albo-viridis and suggests that subtropic populations may be considered as a genetic entity.  相似文献

InVicia benghalensis two biotypes differing in biochemical and cytological features were identified in previous studies. It was hypothesized that these biotypes could also vary in reproductive behaviour and be genetically isolated. To better understand this issue, 25 distinct samples were studied using allozymes. Nine enzyme systems, accounting for 15 putative loci, were scored. The data obtained were analysed using population diversity parameters; Nei's genetic distance was calculated and used to construct a dendrogram. Principal component analysis was conducted to verify the dendrogram and to determine which alleles had stronger influence on the grouping of populations. The analyses demonstrated that the two previously identified biotypes are genetically different: Nei's distance between the two groups is larger than the distance within each group. Three-dimensional plotting of the first three principal components corroborates this division. Genetic diversity parameters confirm that the two biotypes also differ in outcrossing rates. These results denote the existence of two highly divergent genepools inV. benghalensis and suggest that a speciation process may be acting. The consequence of the present findings on the management of gene bank collections is analysed.  相似文献

We examined the allozymic variation of Zostera japonica, a seagrass in brackish-water zones, for plant samples collected from the Ohashi River (four localities including nine sampling sites within 7.5km) and Lake Nakaumi (one location), to which it is connected, in Japan to study plant propagation. Among five enzymes detected, phosphoglucoisomerase (PGI) showed three alleles (b, c, d) and five genotypes (bb, cc, dd, bd, cd), but the other four enzymes were monomorphic in 185 plant samples. PGI genotype variation was found not only in large meadows but also in small (<1 m) vegetation patches, indicating genet coexistence within a small spatial scale. The observed genotype frequencies in large meadows did not differ significantly from Hardy—Weinberg equilibrium. However, no plant sample included genotype bc. These results suggest that new genet recruitment by mating among genets has occurred but that recruitment may not be very frequent. The allele frequencies differed significantly, not only between distant (1.4 km) meadows but also within short distances (20–40 m) within a meadow. Plants carrying allele c were collected from the upper but not from the lower part of the Ohashi River. These results suggest successful dispersal by water currents is restricted.  相似文献

Electrophoretic surveys have demonstrated that populations of the sea anemone Metridium senile along the northeast coast of the United States are polymorphic at four enzyme loci. Phosphoglucose isomerase (PGI) has two alleles in most populations, phosphoglucomutase (PGM) has three alleles, and two leucine aminopeptidase loci have two common alleles each. Phosphoglucose isomerase displays clinal variation and an apparent association with environmental temperature. Phosphoglucomutase shows clinal variation north of Cape Cod for two of the three alleles, while the two leucine aminopeptidase loci are not clinal. All loci show a great deal of variation in populations on Cape Cod, but there is no apparent systematic pattern to this variation. Allozyme variation was examined inCarex sect.Phyllostachys (Cyperaceae) to provide insight into phylogenetic relationships hypothesized in an earlier study and to determine the degree of genetic differentiation within and between taxa. Genetic identity values are concordant with the morphological differences found between species. The lowest values are found between species with the greatest morphological dissimilarity. Conversely, the highest values are associated with species pairs distinguished by relatively few morphological differences. Conspecific populations possess high genetic identities, although interpopulation differentiation has characterized the evolutionary history of some species. Geographic patterning is also evident within species, with geographically proximate populations often having the highest identity values. Phylogenetic trees produced using different cladistic methods were poorly supported and varied in their depiction of relationships among species. One cladogram produced using presence/absence allelic data is more or less congruent with a topology recovered from an earlier analysis utilizing molecular and morphological data. The wide- and narrow-scaled clades are maintained as are the sister species pairsC. backii/C. saximontana, C. basiantha/C. superata, andC. jamesii/C. juniperorum. Contrary to the finding of our previous study, however,C. willdenowii is aligned withC. jamesii/C. juniperorum.  相似文献

Allozyme variation at 11 loci (with 37 alleles) was studied electrophoretically in seven outbreeding, closely related diploid and tetraploid taxa, seven from sect.Leptogalium and two from sect.Leiogalium. Whereas the sections are clearly distinct by several different alleles, aggregates, species and subspecies differ only in the frequency or presence/absence of common alleles. The resulting dendrogram suggests phylogenetic relationships and is supported by other multidisciplinary evidence. Tetraploids have originated independently in several groups, and there is evidence for tetrasomic inheritance and thus for autopolyploidy in spite of normal meiotic bivalent pairing and partly suspected hybrid origin. Tetraploids differ from related diploids only little in number of alleles and expected heterozygosity within populations, but clearly exhibit higher numbers of genotypes. This often corresponds to their greater morphological variability, increased adaptive flexibility, and better colonizing capacity compared to related diploids.  相似文献

Drosophila antonietae is an endemic South American cactophilic species found in relictual xerophytic vegetation, mostly associated with Cereus hildmaniannus cactus. Low differentiation among populations of this species has been detected using several markers. In this work, we performed an allozyme genetic variability analysis of 11 natural populations of D. antonietae and included a discussion about the possible influences of several evolutionary processes that might be acting to maintain the pattern observed. The genetic variability of 14 isoenzyme loci was analysed and showed a high genetic diversity (average observed heterozygosity = 0.319) and a moderate genetic differentiation among populations ( F statistics = 0.0723). A correlation between genetic and geographical and ecological distances was detected among pairs of populations and the regional equilibrium analysis was thus applied. This analysis resulted in Nm (number of migrants) of approximately 3.21, indicating that moderate levels of both gene flow and genetic drift occur in this species, with gene flow overlapping genetic drift. However, considering ecological features of drosophilids, we propose a hypothesis to explain the moderate differentiation encountered as a result of three different processes, or a combination of them: (1) gene flow; (2) a short period of differentiation, i.e. maintenance of ancestral polymorphism; and (3) action of natural selection. Moreover, if gene flow is present, the high genetic diversity compared with other cactophilic and non-cactophilic species could be due to differential selection in different populations followed by gene exchange among them. These factors are discussed in the light of D. antonietae 's historical and evolutionary association with the host cactus.  相似文献

New Zealand populations of Gracilaria chilensis are uniform in anatomical reproductive characteristics but vary morphologically and have been found to separate into two distinct groups with respect to agar methylation level, namely low (24–30%) and high (43–47%). To investigate the genetic variation within New Zealand populations of this species, 14 isozyme loci detected by starch-gel electrophoresis were examined in 17 wild populations from a wide range of localities, and in cultures derived from these populations. Five of these loci were polymorphic, but the genetic variation within populations was low: of the 17 populations examined, 15 were fixed at all loci (heterozygosity 0.000) and in the remaining two populations the observed heterozygosity was still low (0.004 and 0.011). The genetic distances between the populations ranged from 0.00 to 0.43. UPGMA cluster analysis separated the populations into two groups, a northern group and a group found throughout the country. Although these two groups do not correlate with the two groups based on agar methylation level at every locality, the correlation is sufficiently striking to merit further investigation.  相似文献
