The Montseny brook newt (Calotriton arnoldi) is a critically endangered amphibian species which inhabits a small 20 km2 holm oak and beech forest area in NE Spain. Calotriton arnoldi strictly lives in running waters and might be highly vulnerable to hydrological perturbations expected to occur under climate and vegetation cover changes. Knowledge about the potential response of the species habitat to environmental changes can help assessing the actions needed for its conservation. Based on knowledge of the species and supported by observations, we proposed daily low and high streamflow event thresholds for the viability of C. arnoldi. We used the rainfall–runoff model PERSiST to simulate changes in the frequency and duration of these events, which were predicted under two climate and four vegetation cover scenarios for near‐future (2031–2050) and far‐future (2081–2100) periods in a reference catchment. All future scenarios projected a significant decrease in annual streamflow (from 21% to as much as 67%) with respect to the reference period. The frequency and length of low streamflow events will dramatically increase. In contrast, the risk of catastrophic drift linked to high streamflow events was predicted to decrease. The potential change in vegetation toward an expansion of holm oak forests will be more important than climate changes in determining threshold low flow conditions. We thus demonstrated that consideration of potential changes in vegetation and not only changes in climate variables is essential in simulating future streamflows. This study shows that future low streamflow conditions will pose a severe threat for the survival of C. arnoldi and may help taking management actions, including limiting the expansion of holm oak forest, for ameliorating the species habitat and help its conservation.  相似文献

Mitochondrial DNA (mtDNA) sequence variation was examined in the three species belonging to the newt genus Euproctus: E. asper, E. montanus, and E. platycephalus, and in three other species belonging to the same family: Triturus carnifex, T. vulgaris and Pleurodeles waltl. The Euproctus species inhabit mountain streams in the Pyrenean region, Corsica, and Sardinia, respectively. This vicariant distribution is believed to be a result of the disjunction and rotation of the Sardinia-Corsica microplate from the Pyrenean region and suggested dates for each cladogenetic event are available. A total of 915 bp from 12S and 16S ribosomal rRNA genes were compared for each taxon. These are the first mt-rDNA sequence data for salamanders. Sequences were used to reconstruct phylogenetic trees, investigate evolutionary rates for these genes, calibrate them with absolute time since divergence, and compare rates with published ones. Using P. waltl as the outgroup, all phylogenetic methods used (parsimony, maximum likelihood, and Neighbor Joining) produced trees with identical topologies and similar bootstrap values associated with each node. These sequence data cannot unambiguously resolve the splitting events leading to the main radiation of the genus Triturus and the origin of the genus Euproctus. These events may well have occurred very close in time, consistent with other sorts of data. Although it is unlikely strict linearity holds for all kinds of substitutions, relative rate tests of the molecular clock hypothesis could not reject clock-like behavior of sequence changes along Euproctus lineages. Estimates of absolute rates of base changes are 0.35% per Myr since divergence for all substitutions and 0.14% per Myr for transversions; these estimates are similar to other vertebrate estimates. A comparison with distance measures from allozyme studies agrees quite well with regard to relative divergences of the three Euproctus species.  相似文献

Two BamHI families of repeated sequences were characterized from the genome of the Italian smooth newt, Triturus vulgaris meridionalis (Amphibia, Urodela). The first family, which is divided into subfamilies, consists of tandemly arranged arrays whose basic repeat is around 398 bp long; these arrays are dispersed throughout the entire chromosome sets of the various species of Triturus tested. Moreover the family is widely conserved among Salamandridae, being detected by genomic DNA blotting of Notophthalmus viridescens, Taricha granulosa, Salamandrina terdigitata and Euproctus platycephalus. The second BamHI family is represented by a cloned sequence of 419 bp, which is dispersed in the chromosome set of several species of Triturus. The sequence is also conserved in S. terdigitata and in E. platycephalus but is not detectable in N. viridescens or T. granulosa. Historical factors (colonization scenarios, demographic oscillations) and contemporary processes (population connectivity, current population size) largely contribute to shaping species’ present‐day genetic diversity and structure. In this study, we use a combination of mitochondrial and nuclear DNA markers to understand the role of Quaternary climatic oscillations and present‐day gene flow dynamics in determining the genetic diversity and structure of the newt Calotriton asper (Al. Dugès, 1852), endemic to the Pyrenees. Mitochondrial DNA did not show a clear phylogeographic pattern and presented low levels of variation. In contrast, microsatellites revealed five major genetic lineages with admixture patterns at their boundaries. Approximate Bayesian computation analyses and linear models indicated that the five lineages likely underwent separate evolutionary histories and can be tracked back to distinct glacial refugia. Lineage differentiation started around the Last Glacial Maximum at three focal areas (western, central and eastern Pyrenees) and extended through the end of the Last Glacial Period in the central Pyrenees, where it led to the formation of two more lineages. Our data revealed no evidence of recent dispersal between lineages, whereas borders likely represent zones of secondary contact following expansion from multiple refugia. Finally, we did not find genetic evidence of sex‐biased dispersal. This work highlights the importance of integrating past evolutionary processes and present‐day gene flow and dispersal dynamics, together with multilocus approaches, to gain insights into what shaped the current genetic attributes of amphibians living in montane habitats.  相似文献

The development of microsatellite loci has become more efficient using next‐generation sequencing (NGS) approaches, and many studies imply that the amount of applicable loci is large. However, few studies have sought to quantify the number of loci that are retained for use out of the thousands of sequence reads initially obtained. We analyzed the success rate of microsatellite loci development for three amphibian species using a 454 NGS approach on tetra‐nucleotide motif‐enriched species‐specific libraries. The number of sequence reads obtained differed strongly between species and ranged from 19,562 for Triturus cristatus to 55,626 for Lissotriton helveticus, with 52,075 reads obtained for Calotriton asper. PHOBOS was used to identify sequences with tetra‐nucleotide repeat motifs with a minimum repeat number of ten and high quality primer binding sites. Of 107 sequences for T. cristatus, 316 for C. asper and 319 for L. helveticus, we tested the amplification success, polymorphism, and degree of heterozygosity for 41 primer combinations each for C. asper and T. cristatus, and 22 for L. helveticus. We found 11 polymorphic loci for T. cristatus, 20 loci for C. asper, and 15 loci for L. helveticus. Extrapolated, the number of potentially amplifiable loci (PALs) resulted in estimated species‐specific success rates of 0.15% (T. cristatus), 0.30% (C. asper), and 0.39% (L. helveticus). Compared with representative Illumina NGS approaches, our applied 454‐sequencing approach on specifically enriched sublibraries proved to be quite competitive in terms of success rates and number of finally applicable loci.  相似文献

According to theories on cave adaptation, cave organisms are expected to develop a lower metabolic rate compared to surface organisms as an adaptation to food scarcity in the subterranean environments. To test this hypothesis, we compared the oxygen consumption rates of the surface and subterranean populations of a surface‐dwelling species, the newt Calotriton asper, occasionally found in caves. In this study, we designed a new experimental setup in which animals with free movement were monitored for several days in a respirometer. First, we measured the metabolic rates of individuals from the surface and subterranean populations, both maintained for eight years in captivity in a natural cave. We then tested individuals from these populations immediately after they were caught and one year later while being maintained in the cave. We found that the surface individuals that acclimated to the cave significantly reduced their oxygen consumption, whereas individuals from the subterranean population maintained in the cave under a light/dark cycle did not significantly modify their metabolic rates. Second, we compared these metabolic rates to those of an obligate subterranean salamander (Proteus anguinus), a surface aquatic Urodel (Ambystoma mexicanum), and a fish species (Gobio occitaniae) as references for surface organisms from different phyla. As predicted, we found differences between the subterranean and surface species, and the metabolic rates of surface and subterranean C. asper populations were between those of the obligate subterranean and surface species. These results suggest that the plasticity of the metabolism observed in surface C. asper was neither directly due to food availability in our experiments nor the light/dark conditions, but due to static temperatures. Moreover, we suggest that this adjustment of the metabolic level at a temperature close to the thermal optimum may further allow individual species to cope with the food limitations of the subterranean environment.  相似文献

Islands are hotspots of biodiversity, with a disproportionately high fraction of endemic lineages, often of ancient origin. Nevertheless, intra‐island phylogeographies are surprisingly scarce, leading to a scanty knowledge about the microevolutionary processes induced on island populations by Plio‐Pleistocene climatic oscillations, and the manner in which these processes contributed to shape their current genetic diversity. We investigated the phylogeography, historical demography and species distribution models of the Corsican endemic newt Euproctus montanus (north‐western Mediterranean). As for many island endemics, the continuous distribution of E. montanus throughout its range has hitherto been considered as evidence for a single large population, a belief that also guided the species' categorization for conservation purposes. Instead, we found a geographic mosaic of ancient evolutionary lineages, with five main clades of likely Pliocene origin (2.6–5.8 My), all but one restricted to northern Corsica. Moreover, the copresence between main lineages in the same population was limited to a single case. As also suggested by growing literature on intra‐island phylogeographic variation, it seems that the extensive use of simplifying assumption on the population structure and historical demography of island populations—both in theoretical and applicative studies—should be carefully reconsidered, a claim that is well exemplified by the case presented here.  相似文献

Differences in environmental conditions such as those between lakes and streams can produce phenotypic variation and ultimately promote evolutionary diversification. Some species of newts and salamanders can occupy these habitats and express alternative phenotypes: metamorphs that lose gills at metamorphosis and paedomorphs that retain them at the adult stage. Whereas this process is facultative in some species, it is obligatory in others, thus suggesting that isolation and environmental pressures may have canalized developmental pathways. In this study, we focused our research on the Pyrenean brook newt, Calotriton asper, which is present in both lakes and streams, but whose fully aquatic paedomorphic individuals are only present in lakes. We aimed to determine the genetic structure and differentiation of two paedomorphic populations, including their surrounding stream and lake metamorphic populations, to test whether populations of paedomorphs can constitute evolutionary significant units. Although gene flow was identified between lakes and nearby stream populations, there was a low percentage of dispersers, and the paedomorphic populations were genetically differentiated from the populations of metamorphs. It is likely that the studied lakes have offered peculiar conditions that have allowed the development of a paedomorphic phenotype. These populations and phenotypes therefore constitute good models to understand local adaptations. As each of these populations of paedomorphs can be considered evolutionary significant units that cannot be replaced by other nearby populations in case of a population crash, conservation actions should be focused directly on them.  相似文献

Batrachochytrium dendrobatidis (Bd), the causative agent of the amphibian disease chytridiomycosis, is an important factor in the global decline of amphibians. Within Europe, animals that exhibit clinical signs of the disease have only been reported in Spain despite the pathogen’s wide, but patchy, distribution on the continent. Recently, another occurrence of chytridiomycosis was reported in Euproctus platycephalus, the Sardinian brook newt, on the Mediterranean island of Sardinia, but without any evidence of fatal disease. We report further evidence of the emergence of Bd on Sardinia and the first evidence of lethal chytridiomycosis outside of Spain. Unusual mortalities of the Tyrrhenian painted frog (Discoglossus sardus) were found at three sites in the Limbara mountains of northern Sardinia. Molecular and histological screens of corpses, frogs, and tadpoles from these sites revealed infection with Bd. Infection and mortality occurred at locations that are unusual in terms of the published habitat requirements of the pathogen. Given the endemicity, the IUCN Red List status of the amphibian species on Sardinia, and the occurrence of infection and mortality caused by chytridiomycosis, there is serious reason for concern for the impact that disease emergence may have on the conservation of the amphibians of the island.  相似文献

Summary The somatic chromosomes of T. striatus lysteri, E. minimus operarius, E. speciosus frater, E. quadrivittaius hopiensis and E. umbrinus montanus have been analyzed utilizing a method employing pretreatment with colchicine and hypotonic citrate followed by staining and squashing.Each animal had a modal diploid chromosome number of 38.A preliminary chromosome classification in chipmunks, based on chromosome size and centromere position, has been proposed. Utilizing this classification, karyograms of T. striatus lysteri, E. minimus operarius, E. quadrivittatus hopiensis and E. umbrinus montanus were analyzed. Karyograms of E. umbrinus montanus and E. quadrivittatus hopiensis were similar and differed from those of E. minimus operarius and from those of T. striatus lysteri. A comparison of terrestrial small mammal coenoses belonging to nine different biotopes in the tips of the Iberian and Italian peninsulas was carried out using the pitfall trapping method. The influence of both habitat type and peninsular effect on composition of small mammal coenoses was analysed. In Southern Italy, 203 specimens belonging to seven species were trapped: Suncus etruscus (Savi, 1822), Crocidura suaveolens (Pallas, 1811), C. leucodon (Hermann, 1780), Microtus savii (de Sélys-Longchamps, 1838), Apodemus sylvaticus (L., 1758), A. flavicollis (Melchior, 1834) and Mus musculus domesticus Schwarz & Schwarz, 1943. In Southern Spain 428 specimens belonging to five species were trapped: Suncus etruscus, Crocidura russula (Hermann, 1780), Microtus duodecimcostatus (de Sélys-Longchamps, 1839), Apodemus sylvaticus and Mus spretus Lataste, 1883. The relative density of small mammals occurring in the nine Spanish sampling stations was twice that recorded in the Italian stations; however the number of species recorded in the different biotopes show similar mean values, ranging from three to five in Andalusia and from three to six in Calabria. Apodemus sylvaticus was the dominant species in the Calabrian stations, whereas Crocidura russula prevailed in Andalusia. The biotic diversity values are very similar in the Calabrian and Andalusian biotopes. By contrast, the Insectivora/Rodentia ratio was always higher in Andalusia. The more xerophytic biotopes showed greater similarities between the communities in Southern Spain and Southern Italy, while the cooler biotopes differed between these two peninsulas.  相似文献

5S rRNA intergenic spacers were amplified from two elite sugarcane (Saccharumhybrids) cultivars and their related taxa by polymerase chain reaction (PCR) with 5S rDNA consensus primers. Resulting PCR products were uniform in length from each accession but exhibited some degree of length variation among the sugarcane accessions and related taxa. These PCR products did not always cross hybridize in Southern blot hybridization experiments. These PCR products were cloned into a commercial plasmid vector PCR™ 2.1 and sequenced. Direct sequencing of cloned PCR products revealed spacer length of 231–237 bp for S. officinarum, 233–237 for sugarcane cultivars, 228–238 bp for S. spontaneum, 239–252 bp for S. giganteum, 385–410 bp for Erianthusspp., 226–230 bp for Miscanthus sinensisZebra, 206–207 bp for M. sinensisIMP 3057, 207–209 bp for Sorghum bicolor, and 247–249 bp for Zea mays. Nucleotide sequence polymorphism were found at both the segment and single nucleotide level. A consensus sequence for each taxon was obtained by Align X. Multiple sequences were aligned and phylogenetic trees constructed using Align X, CLUSTAL and DNAMAN programs. In general, accessions of the following taxa tended to group together to form distinct clusters: S. giganteum, Erianthusspp., M. sinensis, S. bicolor, and Z. mays. However, the two S. officinarumclones and two sugarcane cultivars did not form distinct clusters but interrelated within the S. spontaneumcluster. The disclosure of these 5S rRNA intergenic spacer sequences will facilitate marker-assisted breeding in sugarcane. We report the presence of pigmented skin tumors in three populations of the endangered amphibian Montseny brook newt, Calotriton arnoldi, one of the European amphibian species with the smallest distribution range (40 km(2) in the Montseny Natural Park, Catalonia, Spain). Examination of one of the tumors by light microscopy was consistent with chromatophoroma and was most suggestive of a melanophoroma. Tumors were not found in juveniles. In adults, only two of three populations were affected. The proportions of males and females affected were not significantly different, but there was a positive correlation between body size and presence of tumors in both sexes. The etiology of chromatophoromas remains unknown but, in our study, they do not appear to have been caused by water quality or Ultraviolet B.  相似文献

Length–weight relationships (LWRs) are presented for five species, namely: Garra nasuta (McClelland, 1838); Garra gotyla gotyla (Gray, 1830); Devario aequipinnatus (McClelland, 1839); Barilius bendelisis (Hamilton, 1807) and Pseudolaguvia shawi (Hora, 1921) collected from the Basistha River, Guwahati, Assam, India. The organization of 5S ribosomal RNA genes (rDNA) was examined for threeJapanese Laminaria species, L. japonica, L.religiosa and L. ochotensis. The linkage of 5SrDNA with the 18S-5.8S-25S rDNAs unit known in the brown algaScytosiphon lomentaria could not be detected inLaminaria. Instead, the tandem repeats of 5S rDNA were notassociated with the 18S-5.8S-25S rDNAs unit. The nucleotide sequence of 5S rDNAwas completely identical among these three species and its length was 118bp. However, a difference of nucleotide arrangement was detectedinthe spacer region of the tandemly repeated 5S rDNAs. Several nucleotideinsertion / deletions and substitutions were confirmed between differentindividuals of L. japonica, which were collected from notonly disjunct localities, but also the same locality. The lengths of the spacerregion of L. japonica, L. religiosaand L. ochotensis were 247–252 bp, 232bp and 252 bp, respectively.  相似文献

It has been an enduring belief that increasing aridity combined with Pliocene‐Pleistocene glacial cycles resulted in the formation of distinct arid zone and temperate zone faunas within Australia. We assembled a molecular phylogeny for the Egernia whitii species group, an endemic group of skinks that comprises representatives from arid and temperate Australia, in order to test several biogeographical hypotheses regarding the origin of the Australian arid zone fauna. Sequence data were obtained from ten of the 11 species within the species group, plus three other Egernia species and an outgroup (Eulamprus heatwolei). We targeted portions of the ND4 (696 base pairs) and 16S rRNA (500 bp) mitochondrial genes and the β‐Fibrinogen 7th Intron nuclear gene (648 bp). The edited alignment comprised 1844 characters, of which 551 (30%) were variable and 382 (69%) were parsimony informative. We analysed the data using maximum likelihood and Bayesian techniques and produced a single optimal tree. Our phylogeny strongly supports two major clades within the species group, corresponding to temperate‐adapted rock‐dwelling species and arid‐adapted obligate burrowing species. However, the phylogenetic affinities of E. pulchra were not resolved. Our topology indicates that the New South Wales population of E. margaretae is actually E. whitii and reveals that E. margaretae margaretae and E. m. personata are distinct species. There also appears to be a major phylogeographical break within E. whitii occurring in eastern Victoria. Although our data supported several previously proposed phylogenetic relationships, Shimodaira–Hasegawa tests soundly rejected several suggested affinities between certain species. The arid zone members of the E. whitii species group had been suggested to have originated as a result of multiple periods of colonization during the Pleistocene glaciation cycles. However, our genetic data suggest a single origin (presumably from a semiarid E. multiscutata‐like ancestor) for the arid zone members of the group prior to the Plio‐Pleistocene, probably during the late Miocene to early Pliocene. Our topology displays substantial sequence divergence between species with short internodes and long terminal branches, indicating rapid adaptive radiations within the arid and temperate zones. The presence of temperate‐adapted species within more mesic refugia of the arid zone suggests that the necessary adaptations to aridity for colonizing the dry interior of the continent have not evolved since the initial period of adaptive radiation, despite the long evolutionary history of the species group. © 2004 The Linnean Society of London, Biological Journal of the Linnean Society, 2004, 83 , 157–173.  相似文献

The phylogenetic relationships and species identification of pufferfishes of the genus Takifugu were examined by use of randomly amplified polymorphic DNA (RAPD) and sequencing of the amplified partial mitochondrial 16S ribosomal RNA genes. Amplifications with 200 ten-base primers under predetermined optimal reaction conditions yielded 1962 reproducible amplified fragments ranging from 200 to 3000 bp. Genetic distances between 5 species of Takifugu and Lagocephalus spadiceus as the outgroup were calculated from the presence or absence of the amplified fragments. Approximately 572 bp of the 16S ribosomal RNA gene was amplified, using universal primers, and used to determine the genetic distance values. Topological phylogenic trees for the 5 species of Takifugu and outgroup were generated from neighbor-joining analysis based on the data set of RAPD analysis and sequences of mitochondrial 16S rDNA. The genetic distance between Takifugu rubripes and Takifugu pseudommus was almost the same as that between individuals within each species, but much smaller than that between T. rubripes, T. pseudommus, and the other species. The molecular data gathered from both analysis of mitochondria and nuclear DNA strongly indicated that T. rubripes and T. pseudommus should be regarded as the same species. A fragment of approximately 900 bp was amplified from the genome of all 26 T. pseudommus individuals examined and 4 individuals of intermediate varieties between T. rubripes and T. pseudommus. Of the 32 T. rubripes individuals, only 3 had the amplified fragment. These results suggest that this fragment may be useful in distinguishing between T. rubripes and T. pseudommus. Summary DNA sequence analysis and the localization of the 5 and 3 termini by S1 mapping have shown that the mitochondrial (mt) small subunit rRNA coding region fromPodospora anserina is 1980 bp in length. The analogous coding region for mt rRNA is 1962 bp in maize, 1686 bp inSaccharomyces cerevisiae, and 956 bp in mammals, whereas its counterpart inEscherichia coli is 1542 bp. TheP. anserina mt 16S-like rRNA is 400 bases longer than that fromE. coli, but can be folded into a similar secondary structure. The additional bases appear to be clustered at specific locations, including extensions at the 5 and 3 termini. Comparison with secondary structure diagrams of 16S-like RNAs from several organisms allowed us to specify highly conserved and variable regions of this gene. Phylogenetic tree construction indicated that this gene is grouped with other mitochondrial genes, but most closely, as expected, with the fungal mitochondrial genes.  相似文献
