In Brants' whistling rat, Parotomys brantsii, single animals inhabit unusually complex burrow systems containing several nest chambers. We investigated burrow use and examined the effect of ectoparasites on choice of alternative nest chambers by radiotelemetrically monitoring the nocturnal sleeping locations of individual whistling rats, before and after treating them to remove ectoparasites. Prior to treatment, animals used several different nest chambers within a single burrow, moving from one chamber to another every 1.6 d on average, and most individuals also slept in more than one burrow. Anti‐parasite treatment reduced the rate at which animals switched from one nest chamber to another within a burrow. Screening of a separate sample of animals for ectoparasites revealed that they were infested with fleas, mainly Xenopsylla eridos. We suggest that by switching periodically from one nest chamber to another, whistling rats reduce the rate at which ectoparasites, especially fleas, accumulate.  相似文献

Examination of the topographical anatomy of the stomach complex and intestinal tract of an adult male and a newborn female pigmy hippopotamus (Choeropsis liberiensis) shows that the stomach consists of four chambers, the first three constituting the proventriculus and the fourth the glandular stomach. The proventriculus is made up of a visceral and parietal blind sac opening into a connecting chamber, which in turn opens into the glandular compartment. The walls of the proventricular chambers are covered with villi and non-glandular mucous membrane. The significant difference between the animals is that whereas the connecting chamber of the adult stomach lies transversely deep in the cranial part of the abdomen and connects with the glandular chamber on the right side, that of the newborn lies almost vertically and connects ventrally with the glandular compartment situated on the floor of the abdomen. A groove which in the adult runs more or less horizontally from the cardia through the visceral sac and connecting chambers is aligned almost vertically in the newborn. In the adult the connecting chamber is the largest compartment, but in the neonate the visceral blind sac and the glandular compartment are proportionally larger. Functional aspects of these anatomical differences are discussed in relation to suckling behavior. The intestine consists of a continous tube. There is no caecum; instead the junction between small and large intestine is distinguished by an increase in diameter, a change in epithelial lining, and anastomoses of the jejunal and colic arteries. Large amounts of fat are present in the omentum and mesentery of the neonate.  相似文献

The growth of granulopoietic progenitor cells (CFU-C) in diffusion chambers during culture of peripheral blood leukocytes from 10 normal subjects has been studied. At various times after initiation of diffusion chamber culture, cells harvested from the chambers were transferred to agar culture for measurement of CFU-C concentration. Under these conditions colonies could be grown successfully in agar culture provided pronase, necessary for the chamber harvesting procedure, was first removed by careful washing. A marked increase in the number of CFU-C, up to 25-fold the initial value, was observed in 8 out of 10 subjects. Here the growth pattern was similar, independent of the initial CFU-C values, with an immediate rise to a maximum between 6 and 13 days of culture followed by a decrease. In the other two subjects the growth of CFU-C throughout the diffusion chamber culture period was very poor. The growth of CFU-C from a given individual's blood was shown to be reproducible in repeated studies in 2 subjects, one of whom showed a proliferative and the other a non-proliferative pattern. Evidence suggests that the increase in CFU-C in diffusion chambers is the result of both self-renewal of these cells and influx from a more primitive compartment, although the present data do not allow an estimate of the relative magnitude of each.  相似文献

Temperature and food availability are two important factors which affect fish growth and therefore are expected to influence habitat choice in fish. In this study, shoals of 16 juvenile roach, Rutilus rutilus , were given a choice between two chambers that differed in temperature by 1·5°C or 3°C whereas food availability was the same in both chambers (ratio 1 : 1) or higher in the colder one (ratio 4 : 1). The number of fish in each chamber was recorded for 10 min each during a pre-feeding, feeding and post-feeding period. Roach generally preferred the warmer over the colder chamber during the pre-feeding periods. Temperature had a significant effect on the distribution of fish during all three time periods whereas food availability was a significant factor only during the feeding period. The important role of temperature was emphasized further by the fact that a relatively small difference in the temperature gradient of 1·5°C had a stronger effect on fish distribution than a four times higher feeding rate during the feeding period. The implications for growth rates of such short-term decision-making of roach are discussed.  相似文献

Backward pairings of a distinctive chamber as a conditioned stimulus and wheel running as an unconditioned stimulus (i.e., running-then-chamber) can produce a conditioned place preference in rats. The present study explored whether a forward conditioning procedure with these stimuli (i.e., chamber-then-running) would yield place preference or aversion. Confinement of a rat in one of two distinctive chambers was followed by a 20- or 60-min running opportunity, but confinement in the other was not. After four repetitions of this treatment (i.e., differential conditioning), a choice preference test was given in which the rat had free access to both chambers. This choice test showed that the rats given 60-min running opportunities spent less time in the running-paired chamber than in the unpaired chamber. Namely, a 60-min running opportunity after confinement in a distinctive chamber caused conditioned aversion to that chamber after four paired trials. This result was discussed with regard to the opponent-process theory of motivation.  相似文献

Infection of subcutaneusly implanted chambers in guinea pigs conferred immunity against homologous infection of other chambers in the same animals. However, attempts to immunize guinea pigs by subcutaneous injection of filtered fluid from infected chambers, or with small doses of formalin-killed, chamber gonococci were not successful. Thus, neither organisms grown in vivo nor their extracellular products appeared to be exceptionally immunogenic. In immunizing tests with different isolates of gonococci adapted to growth in guinea-pig chambers, cross-immunity to chamber infection with low challenge doses was detected only between two of six isolates. The killing of gonococci in chambers of immunized animals, which occurred only after homologous challenge or with the heterologous strain showing cross-immunity, was not due primarily to humoral factors in the chamber fluid but probably to an enhanced effectiveness of phagocytosis. The serum of immunized animals was bactericidal for homologous strains and for the strain showing cross-immunity but not for strains showing no cross-immunity. Hence, serum bactericidal activity might be a useful indicator for investigating the specificity of immunity produced by different gonococcal strains.  相似文献

Since consistent data on endogenous circadian rhythms of Mongolian gerbils are not available, the main aim of our study was to identify suitable conditions to receive stable and reproducible free-running rhythms of activity under different light intensities. Another objective was to determine the role of social cues as an exogenous zeitgeber in the absence of a light-dark (LD) cycle. We performed two long-term sets of experiments with adult male gerbils kept in climatic chambers under various photoperiods of at least 30 days each. In all cases, the time of lights on in the chambers differed from the daily starting hour of work in the animal house. Always, two animals per chamber were kept separately in cages with a running wheel while their activity was monitored continuously. During the first set, only three of eight animals developed intra- and interindividual variable free-running rhythms. The activity patterns seemed to be influenced by human activities outside, indicating high sensitivity to external factors. Subsequently, we damped the chambers and the room and restricted access to the room. In the following noise-reduced set, all gerbils developed comparable free-running rhythms of activity. We determined the mean of the free-running period tau, the activity-rest relationship alpha/theta and the amount of running wheel activity per day: tau = 23.7h +/- 0.08h under low light (5 lux) and 25.5h +/- 0.19h under high light intensities (450 lux); alpha/theta = 0.53 +/- 0.08 under 5 lux and 0.34 +/- 0.04 under 450 lux. The amount of daily activity was 12 times as high under 5 lux as under 450 lux. There was no indication that the two animals in one chamber socially synchronized each other. In conclusion, the pronounced rhythm changes in accordance with Aschoff's theory support the view that gerbils are mainly nocturnal animals.  相似文献

A variety of bone chambers are used in orthopedic research to study bone and tissue ingrowth in small and large animals. If different bone chambers are placed in one species, differences in bone ingrowth are observed. For instance, bone ingrowth in the bone conduction chamber (BCC) is high, but is low or absent in the repeated sampling bone chamber (RSBC). This difference may be explained by the design and fixation of these chambers. It is known that stress shielding and micromovement can influence bone formation. The objective of the study reported here was to determine whether stress shielding or soft tissue movement affected bone ingrowth in the BCC in the goat. Two types of caps were made, with fixation similar to that of the fixation plate of the RSBC. By placing the caps over the BCCs and fixating the caps directly to the tibial bone, the effect of stress shielding was studied. One cap was in direct contact with the bone chamber underneath, the other cap did not touch the chamber. This difference was used to observe whether movement of the soft tissue on top of the chamber and cap would affect bone ingrowth. Each limb received one control chamber without a cap and a chamber with a cap, either with or without contacting the BCC, yielding four implants per goat. After 12 weeks, bone and total tissue ingrowths were measured. Bone ingrowth was seen in 38 of 40 chambers. Total tissue and bone ingrowths were comparable between control chambers and BCCs with a cap, irrespective of type. Neither stress shielding, nor lack of movement of soft tissue affected bone ingrowth. Other factors in the design of the chambers were responsible for the difference in bone ingrowth between the BCC and the RSBC.  相似文献

Caco-2 cells grown in bicameral chambers are a model system to study intestinal iron absorption. Caco-2 cells exhibit constitutive transport of iron from the apical (luminal) chamber to the basal (serosal) chamber that is enhanced by apo-transferrin in the basal chamber, with the apo-transferrin undergoing endocytosis to the apical portion of the cell. With the addition of iron to the apical surface, divalent metal transporter 1 (DMT1) on the brush-border membrane (BBM) undergoes endocytosis. These findings suggest that in Caco-2 cells DMT1 and apo-transferrin may cooperate in iron transport through transcytosis. To prove this hypothesis, we determined by confocal microscopy that, after addition of iron to the apical chamber, DMT1 from the BBM and Texas red apo-transferrin from the basal chamber colocalized in a perinuclear compartment. Colocalization was also observed by isolating endosomes from Caco-2 cells after ingestion of ultra-small paramagnetic particles from either the basal or apical chamber. The isolated endosomes contained both transferrin and DMT1 independent of the chamber from which the paramagnetic particles were endocytosed. These findings suggest that iron transport across intestinal epithelia may be mediated by transcytosis.  相似文献

An intraperitoneal chamber implant system has been used to investigate the phenotype of Staphylococcus aureus growing in the rat and the effect of the antibiotic flucloxacillin on bacterial growth in vivo. Titanium chambers were implanted in the peritoneum: a period of 3-4 days equilibration allowed diffusion of host proteins into the chamber fluid prior to inoculation with bacteria. S. aureus inoculated into the chamber fluid, grew rapidly over a 72 h period, reaching counts of > 10(9) per ml. Organisms harvested from chambers were analysed by SDS-PAGE and showed significant differences in polypeptide profiles from the same strain grown in nutrient broth in vitro. Analysis of whole cell extracts by Western-blotting revealed that protein A expression was repressed in S. aureus grown in vivo. Following subcutaneous administration, flucloxacillin levels in serum peaked earlier and were higher than those detected in chamber fluid. The inhibitory effect of the antibiotic on the growth of S. aureus in chambers in treated animals could be monitored easily by sequential sampling of the chamber fluid. These results indicate the potential of the chamber implant model for investigation of microbial phenotype in vivo and development of alternative methods for assessment of antimicrobial efficacy in vivo.  相似文献

A new dynamic dialysis method has been developed for studying protein-ligand binding phenomena. The method depends on analysis of the elution pattern of ligand in a single dialyzing process where the ligand concentration in the sample compartment changes greatly with time. The dialyzer is composed of a long, narrow chamber (the sample compartment) between two sheets of semipermeable membrane and two outside chambers (the sink compartment) connected as a single path. Eluting buffer flows in the sink compartment to exchange the ligand with the solution in the sample compartment. Therefore, the ligand concentration gradient in the sink compartment is in the longitudinal direction. The mathematical expressions to analyze the experimental data were derived from a modified theory of chromatography. Examination of the binding of sulfanilamide to bovine serum albumin using this method shows that these equations are valid for use in studying protein-ligand binding.  相似文献

Polyclonal antibody production in subcutaneous chambers was compared to traditional antibody production methods in rabbits. The chamber, a sterilized plastic wiffle golf ball that had been surgically implanted in the subcutis of the thoracic region, was immunized via a percutaneous injection of antigen into the core of the ball through one of the perforations in the chamber wall. Rabbits bearing chambers were immunized on the same schedule and with the same concentrations of antigens as were provided the adjuvant injected rabbits. Fluid volumes of 12 to 22 ml could be removed from each chamber at weekly intervals. Chamber antibody to specific microbial antigens was equal to or better than serum antibody produced to the same antigens with Freund's or acrylamide adjuvants. The comfort of the rabbit, the ease in chamber immunization, and the recovery of high titer antibody in large volumes make the subcutaneous chamber an attractive method for polyclonal antibody production.  相似文献

The benefits of mycorrhizas for host plants are well known for a large number of species. However, experimental evaluations of the hyphal contribution to the total water uptake and the assessment of the bulk flow velocity in the hyphae are so far contradictory. Barley (Hordeum vulgaris L. Scarlet) with the inoculum Glomus intraradices was grown in a split plant-hyphal chamber with a 5 mm air gap. During the preparation of the chambers with a loamy-silt soil, water content sensors were inserted in each of the plant and the hyphal compartments. These sensors allow non-destructive measurements with high resolution. In total, 8 drying periods with a length of several days were applied with repeated watering following each drying period. A clear decline in water content in the hyphal compartment during each drying period supports the ability of hyphae to transfer water into the plant compartment. The difference between the decline in the hyphal compartment with and without arbuscular mycorrhyzal fungi is significant at the p?<?0.000001 level. The direct and indirect hyphal contribution to the total water uptake was estimated to be about 20%. The application of capacitance sensors for water content determination with a special geometry adapted to the plant-hyphal chambers allows the evaluation of the hyphal water flow with high accuracy.  相似文献

A compartmental model of toad bladder sodium content has been developed, whereby it is possible to measure the four unidirectional fluxes across the opposite faces of the transport compartment, as well as the amount of sodium in the compartment. 24Na is added to the mucosal medium of a short-circuited bladder mounted between halves of a chamber in which the fluid is stirred by rotating impellers. After a steady state is reached, nonradioactive medium is flushed through both sides of the chamber, collected, and counted. The data from each chamber are fitted to sums of exponentials and interpreted in terms of conventional compartmental analysis. Three exponentials are required, with half-times of 0.2, 2.2, and 14.0 min. It is shown that the first of these represents chamber washout, the second the transport pool, and the third a tissue compartment which is not involved in active sodium transport and which does not communicate with the transport pool. The second compartment contains 10.5 µEq of sodium per 100 mg dry weight, an amount equal to approximately 30% of total tissue sodium. The results also indicate, as expected from electrophysiological data, that the mucosal-facing side of the transport compartment is over 10 times as permeable to sodium as the serosal, or pump, side.  相似文献

This paper proposes a new double-chamber model (DCM) of ion channels. The model ion channel consists of a series of three pores alternating with two chambers. The chambers are net negatively charged. The chamber's electric charge originates from dissociated amino acid side chains and is pH dependent. The chamber's net negative charge is compensated by cations present inside the chamber and in a diffuse electric layer outside the chamber. The pore's permeability is constant independent of time. One pore of the sodium channel and one of the potassium channel is a voltage-sensing pore. Due to the channel's structure, ions flow through the pores and chambers in a time-dependent manner. The model reproduces experimental voltage clamp and action potential data. The current flowing through a single sodium channel is less then one femtoampere. The DCM is considerably simpler then the Hodgkin and Huxley model (HHM) used to describe the electrophysiological properties of an axon. Unlike the HHM, the DCM can explain refractoriness, anode break excitation, accommodation and the effect of pH and temperature on the channels without additional parameters. In the DCM, the axon membrane shows repetitive activity depending on the channel density, sodium to potassium channel ratio and external potassium concentration. In the DCM, the action potential starts from 'hot spot areas' of higher channel densities and a higher sodium to potassium channel ratio, and then propagates through the whole axon.  相似文献

The arterial system is assumed to consist of two elastic chambers connected by a conducting channel. It is assumed that a current of fluid enters one chamber, whereas the other chamber is drained by a pipe with a certain peripheral resistance. The continuity of the fluid is described by a differential equation for each chamber. The inertia resistance of the conducting channel is taken into consideration. It is shown that the system may possess a resonance frequency. The latter, if it exists, as well as the damping coefficients are expressed in terms of the elastic moduli of the chambers, the conductivity of the channel, and the peripheral resistance. It is shown that with plausible values of the latter variables the resonance frequency as determined theoretically has the right order of magnitude as found experimentally.  相似文献

Inhalation exposure studies, in which test subjects are fully or partially immersed in an atmosphere containing a compound of interest, are usually carried out using one of two possible exposure systems: large whole-body chambers or systems that expose only the animal's nose or head. Whole-body chambers may require large quantities of test compound, which can pose a problem if the chemical is expensive or available in limited quantities. Nose- or head-only systems can help conserve test compound but may cause stress or injury to animals. To address these concerns, the authors developed an exposure system consisting of small single-animal whole-body chambers. They exposed 80 mice and 80 rats to five test compounds at various concentrations. Though the system was labor-intensive for animal care technicians, it effectively exposed animals to precise chemical doses without causing adverse effects, using less test compound than would have been required in a conventional whole-body chamber.  相似文献
