Changes in nuclear DNA content and cell size of adaxial andabaxial epidermal pavement cells were investigated using brightlight-induced leaf expansion of Phaseolus vulgaris plants. Inprimary leaves of bean plants grown under high (sunlight) ormoderate (ML; photon flux density, 163 µmol m–2s–1) light, most adaxial epidermal pavement cells hada nucleus with the 4C amount of DNA, whereas most abaxial pavementcells had a 2C nucleus. In contrast, plants grown under lowintensity white light (LL; 15 µmol m–2 s–1)for 13 d, when cell proliferation of epidermal pavement cellshad already finished, had a 2C nuclear DNA content in most adaxialpavement cells. When these LL-grown plants were transferredto ML, the increase in irradiance raised the frequency of 4Cnuclei in adaxial but not in abaxial pavement cells within 4d. On the other hand, the size of abaxial pavement cells increasedby 53% within 4 d of transfer to ML and remained unchanged thereafter,whereas adaxial pavement cells continuously enlarged for 12d. This suggests that the increase in adaxial cell size after4 d is supported by the nuclear DNA doubling. The differentresponses between adaxial and abaxial epidermal cells were notinduced by the different light intensity at both surfaces. Itwas shown that adaxial epidermal cells have a different propertythan abaxial ones. Using a laboratory-constructed system that can measure the gas exchange rates of two leaf surfaces separately, the light responses of the adaxial and abaxial stomata in intact leaves of sunflower ( Helianthus annuus L.) were investigated, keeping the intercellular CO2 concentration ( C i) at 300  µ L L−1. When evenly illuminating both sides of the leaf, the stomatal conductance ( g s) of the abaxial surface was higher than that of the adaxial surface at any light intensity. When each surface of the leaf was illuminated separately, both the adaxial and abaxial stomata were more sensitive to the light transmitted through the leaf (self-transmitted light) than to direct illumination. Relationships between the whole leaf photosynthetic rate ( A n) and the g s for each side highlighted a strong dependence of stomatal opening on mesophyll photosynthesis. Light transmitted through another leaf was more effective than the direct white light for the abaxial stomata, but not for the adaxial stomata. Moreover, green monochromatic light induced an opening of the abaxial stomata, but not of the adaxial stomata. As the proportion of blue light in the transmitted light is less than that in the white light, there may be some uncharacterized light responses, which are responsible for the opening of the abaxial stomata by the transmitted, green light.  相似文献

A comparison of photosynthesis-irradiance response curves (PEresponse curves) obtained through fast repetition rate (FRR)fluorometry and radiocarbon (14C) tracer method was made inthe chlorophyte, Dunaliella tertiolecta, grown under differentirradiance conditions. In FRR-based PE response curveexperiments, actinic light provided by white light-emittingdiodes (LEDs) was increased gradually from 0 to 1500 µmolquanta m–2 s–1 and the rate of photosyntheticelectron transport was determined at each light level. Short-termexperiments (20 min) of 14C-based PE response curvewere carried out with an improved photosynthetron, which containswhite LEDs as the light source. Irrespective of growth irradiance,the ratios of FRR to 14C-based initial slopes were almost uniform.The ratios of FRR- to 14C-based maximum rates were 25–36%higher than those of FRR- to 14C-based initial slopes. The relationshipbetween electron transport and carbon assimilation was non-linearwith increasing discrepancy towards high actinic light. Thisnon-linear relationship between FRR- and 14C-based estimatesis primarily due to the effect of physiological processes stimulatedat high levels of light, such as cyclic electron flow and theMehler reaction. The results of this study indicate that theFRR fluorometry can be used as a good indicator of photosyntheticrates from low to middle light levels, but becomes increasinglyquestionable as the maximum photosynthetic rate is approached.The degree to which this relationship is further affected bynutrient-status warrants investigation.  相似文献

The single rooted leaf of soybean (Glycine max L. Merr.) wasused to study source-sink relationships in photosynthesis. Whenthe leaves were kept under a regime of 10 h light (410–480µmol photons m–2, 400–700 nm)–14 h dark,they did not expand, the increase in leaf dry weight almoststopped, and photosynthetic activity remained at a high andconstant level for 8 d while the dry weight of the roots increasedat a constant rate throughout the period. Thus, under this conditionthe leaf and the root system served as the only source and sinkorgans, respectively. When leaves grown for 7 d under this conditionwere placed under continuous light to alter the source/sinkbalance in photosynthate, the root dry weight increased at aconstant rate equal to that found under the 10 h light–14h dark condition. The leaf dry weight markedly increased andby day 5 of continuous light had increased 1.6-fold, mainlyas a consequence of accumulation of starch and sucrose, whichwere not translocated for root growth. The continuous lightcaused an abrupt decrease in the photosynthetic activity (40%of initial value by day 5). However, the activity recoveredalmost completely after a 32-h transfer to darkness. Significantnegative correlations existed between photosynthetic activityand the sucrose and starch contents in the rooted leaves placedunder continuous light. When the plants were treated with variouslight conditions, there was no significant difference (p<0.01)among the regression line slopes for photosynthetic activityon the sucrose content, but there was some deviation among thosefor the photosynthetic activity on the starch content. Thisresult suggests that sucrose accumulated in the leaf has a moredirect influence on photosynthetic activity when the source/sinkbalance was altered. Gossypium hirsutum L. (upland cotton) and G. barbadense L. (Pimacotton) are two of the most important fibre producing cottonspecies in cultivation. When grown side-by-side in the field,G.hirsutum has higher photosynthetic and transpiration rates (Luet al., 1997. Australian Journal of Plant Physiology24: 693–700).The present study was undertaken to determine if the differencesin physiology can be explained by leaf and canopy morphologyand anatomy. Scanning electron microscopy was used to comparethe leaf anatomy of field-grown upland (‘Delta’and ‘Pine Land 50’) and Pima (‘S6’)cotton. Compared to G. hirsutum, mature leaves of G. barbadenseare larger and thinner, with a thinner palisade layer. G. barbadenseleaves show significant cupping or curling which allows fora more even absorption of insolation over the course of theday and much more light penetration into the canopy. AlthoughG. barbadense leaves have a 70–78% higher stomatal densityon both the abaxial and the adaxial surfaces, its stomates areonly one third the size of those of G. hirsutum. This resultsin G. barbadense having only about 60% of the stomatal surfacearea per leaf surface area compared to G. hirsutum. These resultsare indicative of the anatomical and physiological differencesthat may limit the yield potential of G. barbadense in certaingrowing environments. Rice is believed to show photosynthetic symmetry between adaxial and abaxial leaf sides. To verify this, we re‐examined dorsoventral asymmetry in photosynthesis, chlorophyll fluorescence and anatomical traits in flag leaves of two Oryza sativa cultivars that differ in nitrogen (N) response and in leaf angle: ‘Akenohoshi’, a cultivar that can adapt to low‐N (LN), with low leaf angle (more erect leaves), and ‘Shirobeniya’, a cultivar that is unable to adapt to LN, with higher leaf angle. Plants were grown under standard‐N (SN) and LN conditions. LN leaves of both cultivars became more erect than SN, but LN Akenohoshi still had more erect ones than Shirobeniya. Contrary to results of previous studies, leaves of both cultivars showed an asymmetry in photosynthetic rate between adaxial and abaxial sides (higher on the adaxial side) under SN. SN leaves of both cultivars showed lower susceptibility to photoinhibition on the adaxial side than on the abaxial side. However, leaves of Akenohoshi showed less asymmetry in these traits under LN than under SN, whereas leaves of Shirobeniya had similar degrees of asymmetry in these traits under both SN and LN. Both cultivars also showed dorsoventral asymmetry in anatomical traits of mesophyll tissue regardless of N level, but the degree of asymmetry was lower in LN Akenohoshi. These data reveal that rice leaves exhibit dorsoventral asymmetry in photosynthetic and anatomical features, and that the degree of asymmetry varies with cultivar and N level. It is suggested that lower leaf angles (particularly in Akenohoshi) in the presence of LN represent a light acclimation to prevent photoinhibition.  相似文献

A leaf photosynthesis model was constructed based upon the notionthat the leaf photosynthesis is a summation of photosynthesisof each chloroplast under in situ micro-environmental conditions.Intra-leaf light environment was calculated using the valuesof transmittance and reflectance of leaf tissues reported previously.Simulations of light response curves of whole leaf photosynthesiswere carried out for the model leaves with different patternsof gradients in light environment and/or in photosynthetic activities.The results indicate that the higher absorption coefficientof chlorophyll in spongy tissue than in palisade tissue andintra-leaf vertical gradient in photosynthetic activity of thechloroplasts as reported for real dorsiventral leaves are bothadvantageous to the productivity of the leaf because these propertiesraise the efficiency of the light utilization. The dependence of photosynthetic capacity on imported and locally-assimilatedsupplies of carbon during leaf development under different irradianceswas investigated in Glycine max. The potential export of carbonto the developing, mainstem trifoliate leaf (source-potential)was restricted non-destructively by shading all lower, sourceleaves (source-shading), while local photosynthesis was modifiedconcurrently by exposing the young leaf to different light levelsduring development. When source-shading was applied below the2nd mainstem trifoliate leaf at the bud stage of development,photosynthetic capacity was unaffected in leaves which had developedunder moderate and low irradiances (500 and 250 µmol PARm –2 s–1 respectively), but was reduced significantlyin leaves developed under a high irradiance (900 µmolPAR m –2 s–1). If source-shading was applied beneaththe 2nd leaf at unfolding, the reduction of photosynthetic capacityunder the high irradiance was relatively minor. The photosyntheticcapacity attained by the 2nd leaf during development under differentirradiances was influenced by the previous light environmentof the whole plant. In contrast to the 2nd leaf, the photosyntheticcapacities of the 1st and 4th mainstem leaves were relativelyunaffected by source-shading, even under the highest light regime.While photosynthetic capacity showed a widespread insensitivityto the light level of the lower region of the canopy, source-shadingreduced final leaf size irrespective of node position or localirradiance during leaf development. These effects were not relatedto differences in daily photosynthesis by the expanding leaf,and are discussed in terms of the source/sink balance of thedeveloping leaf. Three-week acclimation of winter oilseed rape (Brassica napusL. var. oleifera L.) plants in the cold (2 °C) resultedin a modified pattern of leaf cell enlargement, indicated bythe increased thickness of young leaf blades and modified dimensionsof mesophyll cells, as compared with non-acclimated tissuesgrown at 20/15 °C (day/night). The thickness of leaf cellwalls also increased markedly during cold acclimation but itdecreased in response to a transient freezing event (5 °Cfor 18 h followed by 6 or 24 h at 2 °C, in the dark). Cellwalls of the upper (adaxial) epidermis were most affected. Theirultrastructure was modified by cold and freezing treatmentsin different ways, as revealed by electron microscopy. Possiblereasons for the cold- and freezing-induced modifications inthe leaf and cell wall morphology and their role in plant acclimationto low temperature conditions are discussed. The azimuth of vertical leaves of Silphium terebinthinaceum profoundly influenced total daily irradiance as well as the proportion of direct versus diffuse light incident on the adaxial and abaxial leaf surface. These differences caused structural and physiological adjustments in leaves that affected photosynthetic performance. Leaves with the adaxial surface facing East received equal daily integrated irradiance on each surface, and these leaves had similar photosynthetic rates when irradiated on either the adaxial or abaxial surface. The adaxial surface of East-facing leaves was also the only surface to receive more direct than diffuse irradiance and this was the only leaf side which had a clearly defined columnar palisade layer. A potential cost of constructing East-facing leaves with symmetrical photosynthetic capcity was a 25% higher specific leaf mass and increased leaf thickness in comparison to asymmetrical South-facing leaves. The adaxial surface of South-facing leaves received approximately three times more daily integrated irradiance than the abaxial surface. When measured at saturating CO2 and irradiance, these leaves had 42% higher photosynthetic rates when irradiated on the adaxial surface than when irradiated on the abaxial surface. However, there was no difference in photosynthesis for these leaves when irradiated on either surface when measurements were made at ambient CO2. Stomatal distribution (mean adaxial/abaxial stomatal density = 0.61) was unaffected by leaf orientation. Thus, the potential for high photosynthetic rates of adaxial palisade cells in South-facing leaves at ambient CO2 concentrations may have been constrained by stomatal limitations to gas exchange. The distribution of soluble protein and chlorophyll within leaves suggests that palisade and spongy mesophyll cells acclimated to their local light environment. The protein/chlorophyll ratio was high in the palisade layers and decreased in the spongy mesophyll cells, presumably corresponding to the attentuation of light as it penetrates leaves. Unlike some species, the chlorophyll a/b ratio and the degree of thylakoid stacking was uniform throughout the thickness of the leaf. It appears that sun-shade acclimation among cell layers of Silphium terebinthinaceum leaves is accomplished without adjustment to the chlorophyll a/b ratio or to thylakoid membrane structure.  相似文献

The tetraammonium salt of the K+ binding fluorescent dye benzofuranisophthalate (PBFI) was used to investigate the influence ofpotassium nutrition (0.1–2.1 mol m–3) on apoplasticK+ inVicia faba leaves by means of ratio imaging. As a referencethe infiltration-centrifugation method was used. Both methodsreflected the influence of K+ supply on apoplastic K+ concentration.The abaxial leaf side revealed significantly higher K+ concentrations(20-25 mol m–3) than the adaxial side (5–8 mol m–3).Application of CCCP led to an immediate increase in apoplasticK+ demonstrating the reliability of the PBFI method. Equipment is described which delivers air with concentrationsof CO2 and water vapour closely controlled in the ranges 0 to2500 ppm and 5 to 15 mb respectively, at flow rates of up to10 1 min-1, to each four leaf chambers. The leaf temperatureis controlled to ±0.5 °C and, with a light intensityof 0.3 cal cm-2 min-1 visible radiation (0.4 to 0.7 µm)leaf temperature can be maintained at 17.5 °C.The apparatusused to measure the concentration differences between the watervapour and CO2 entering and leaving the leaf chamber (used tocalculate transpiration, photosynthetic, and respiration rates)is described in detail.Results of tests, which show the necessityfor mounting a fan within the leaf chamber, are reported.Typicallight- and CO2-response curves are given for kale leaves (Brassicaoleracca var. acephala) and an attempt is made to quantify theerrors in the measurement of photosynthesis and transpiration.  相似文献

Diverse Responses of Maple Saplings to Forest Light Regimes   总被引:7,自引:1,他引:6  
Seedlings of 11 species of forest maples (AcerL.) were grownoutdoors from budburst to senescence under three light regimes:‘gap centre under clear skies’ (approx. 20% opensky irradiance; red:far-red ratio=1.12); ‘gap centre undercloudy skies’ (1.5%, ratio=1.03); and ‘gap edge’(2.5%, ratio=0.6). Seedlings grown under the gap centre (clearsky) regime had significantly greater height growth, greaterspecific leaf mass, higher root:shoot ratio, greater investmentin roots, higher leaf nitrogen concentrations, greater chlorophylla:bratio,lower photosynthetic rates under dim light, higher maximum photosyntheticrate, higher stomatal conductance, and lower leaf internal CO2concentrationscompared with those grown in either gap edge or gap centre (cloudy)regimes. Responses to the gap edgevs.gap centre (cloudy) treatmentsdiffer little, suggesting that shade acclimation in forest mapleseedlings is mainly a response to light intensity rather thanspectral quality. The ubiquitous and, except for leaf internalCO2concentration, highly significant interspecific variationin traits was broad-ranging and continuous. Garlic cultivars ‘Bangladesh Local’ and ‘Fructidor’were grown under field conditions in the south-east of Englandand subjected to different shading treatments. The effects ofshading on final leaf size were related to cell numbers anddimensions. An increase in light intensity reduced leaf lengthand size of epidermal cells. In the cv. Fructidor, cell lengthwas reduced, but in the cv. Bangladesh Local, both length andwidth of cells were reduced. The stomatal index decreased withincreasing light intensity in both cultivars. Leaf thicknessincreased with light intensity, resulting in corresponding gainsin leaf d. wt per unit area. Data from cv. Fructidor growing in full sunlight indicated thatthe epidermal tissue adjacent to the abaxial leaf surface hada greater number of cells than that next to the adaxial surface,but no significant differences were found in the correspondingcell depths. The size of epidermal cells in both epidermisesdecreased from the base to tip of the leaf due to reductionin cell length and width. Leaf age effects on the leaf conductance to water vapour diffusionof the adaxial and abaxial leaf surfaces were measured in themorning and in the afternoon on 17 different plantain and banana(Musa spp.) genotypes. The irradiance levels increased three-foldwhile leaf to air vapour pressure deficit levels increased two-to four-fold from morning to afternoon during the sampling periodin a field site located in the humid forest-moist savanna transitionzone of Nigeria. Conductance values were reduced in older, andsenescing leaves relative to the young and mature leaves bothin the morning and in the afternoon. Conductances were higherfor the abaxial leaf surfaces than the adaxial surface and higherin the afternoon than in the morning, with some genotypic differences.Lower values of leaf conductance to water vapour in the afternoonunder a short dry spell was sufficiently variable (P 0·05)among the test genotypes to indicate potential adaptation totransient dry conditions. Differential and relative leaf conductanceadjustments were noted among genotypes experiencing a shortdry spell versus non-limiting soil moisture conditions. Significantgenotypic differences were observed for leaf conductance amongthe 17 genotypes during the afternoon on the lower leaf surfaceof younger leaves. In wave-regenerating Abies forests in central Japan, light availabilityon the forest floor of a mature-tree stand increases graduallyover 4 to 5 years during the senescence of trees. The understoreyevergreen perennialPteridophyllum racemosum Sieb. et Zucc. (Papaveraceae)is commonly found in such forests. To determine the acclimationcharacteristics of this species to gradual changes in lightavailability, leaf photosynthetic traits and biomass allocationpatterns were determined along a transect running from fullyshaded understorey to a canopy gap. Along the transect, thephoton flux density on the forest floor relative to full sun(RPFD) varied from 2.3–36.7%. The light-saturated netphotosynthetic rate of field-grown plants increased as the lightenvironment increased from 2.3–14.1% RPFD. However, aslight conditions increased from moderate (14.1%) to high (36.7%),the light-saturated net photosynthetic rate decreased to a levelcomparable to that exhibited by low-light grown plants. On theother hand, root mass per unit leaf area (RMA) increased withincreasing light conditions. A higher RMA contributes to a higherwater uptake capacity per unit leaf area which in turn supportshigher rates of photosynthesis and transpiration in high light.We conclude that in wave-regenerating forests P. racemosum cansurvive in high-light environments by avoiding a severe waterdeficit and heat load by increasing RMA. We measured the light response curve of photosynthetic oxygen evolution by illuminating a leaf disc in an air-tight windowed chamber. Oxygen production was measured by monitoring the quenching of luminescence of an organometallic ruthenium compound. A photodiode based chlorophyll a fluorometer was used to measure the luminescence intensity. Oxygen evolution measurements with a traditional oxygen electrode gave the same numerical values at different light intensities when the same leaf disk was tested. The quality of the measurement signal of the new method was found to be similar to that obtained with the oxygen electrode method. The new luminescence based system is more stable against electrical disturbances than an oxygen electrode, its response to oxygen pressure changes is very rapid, and the new method allows the same basic equipment to be used for chlorophyll fluorescence and oxygen measurements.  相似文献

Photosynthesis by developing embryos of oilseed rape (Brassica napus L.)   总被引:1,自引:0,他引:1  
The aim of this study was to assess the photosynthetic potentialof developing seeds of oilseed rape (Brassica napus L.) andto compare photosynthetic properties of embryo plastids withthose of leaf chloroplasts from the same species. Measurementsof CO2-dependent O2 evolution show that developing seeds ofB. napus are photosynthetically active in vitro. Essentially,all of the photosynthetic activity of the developing seed isaccounted for by the embryo. The rate of photosynthesis by developingembryos increased until the onset of desiccation, after whichit declined, so that by maturity embryos were no longer photosyntheticallyactive. Photosynthetic activity was positively correlated withchlorophyll content throughout development. Comparison of thephotosynthetic characteristics of leaf and embryo chloroplastsrevealed that rates of uncoupled electron transport were 2.5-foldgreater in those from the embryo. Light-saturated rates of CO2-dependentO2 evolution, per unit chlorophyll, and CO2 saturation pointswere similar for chloroplasts from both tissues. However, light-saturationpoints and chlorophyll a/b ratios were lower for embryo thanfor leaf choroplasts. Embryos and embryo chloroplasts also containedconsiderably less ribulose 1,5-bisphosphate carboxylase/oxygenaseprotein per unit total protein, than leaves. Although excisedembryos were capable of high rates of CO2-dependent O2 evolution(90–100 mol mg–1 chlorophyll h–1) under asaturating photosynthetic photon flux density (PPFD), low transmittanceof light through the silique wall (30%), together with the highPPFD required to achieve light compensation points in developingseeds (500 mol m–2 s–1), suggests that photosynthesisin vivo is unlikely to make a net contribution to carbon economyunder normal environmental conditions. Acclimation of fully developed leaves of Bischofia javanicaBlume to shadelight was examined. Seedlings were grown undersimulated daylight (1000 µmol m–2 s–1), thentransferred to a simulated shadelight (40 µmol m–2s–1). When a high-light leaf was transferred to low light, large negativenet photosynthetic rates (Pm) were recorded. This decrease wasrapid, but within 7 d the rate increased and became equal tothe low-light control leaf. These changes in photosynthesisdid not follow the pattern of changes in stomatal conductance(gs). Transfer to the low light resulted in a dramatic decreasein leaf weight per unit area (Lw), and most of the decreasesin Lw occurred within 3 d of transfer when the Pm of the transferredleaf was well below that of the low-light control leaf. There was a significant decrease in chlorophyll a in the transferredleaf without an appreciable change in chlorophyll b resultingin a large decrease in the chlorophyll a to chlorophyll b ratio.Leaf chlorophylls per unit area were higher in the transferredleaf than the low-light control leaf. The evaluation of the factors controlling the penetration of4-amino-3,5,6-trichloropicounic acid (picloram), containing14C in the carboxyl group, into leaves of Phaseolus vulgarisis complicated by the ability of the leaf tissue to decarboxylatethe compound rapidly. The degradation system becomes fully activatedonly after some hours following treatment with picloram whilethe capacity for decarboxylation declines with the age of theleaf. When allowance for decarboxylation is made, the pattern of penetrationfor picloram resembles in particular that of 2,2-dichloropropionicacid (dalapon). At the abaxial surface penetration continuesover 24 h at a fairly constant rate but in light, which enhancespenetration, a ‘surge’occurs between the secondand sixth hour. Raising the external concentration promotespenetration and in light entry is directly proportional to concentrationup to at least 2.5 x 10–l M. The response to varying lightintensity resembles that of dalapon rather than 2,4-D. Evenlow intensities promote penetration; the effect is maximal atabout 10 000 lx at the abaxial surface but continues at leastup to 20 000 lx at the adaxial surface. Penetration is retarded at 4°C but the decarboxylation systemremains functional. In light, but not in darkness, penetrationis strikingly reduced by 3-(3,4-dichlorophenyl)-l,l-dimethylurea(DCMU). Raising the external pH from 4.2 to 7.2 retards entry. It is concluded that the factors which determine the penetrationof picloram into Phaseolus leaves also control the entry ofa wide range of organic compounds and inorganic ions.  相似文献
