Summary Cross-compatibility of species in section Arachis Krap. et Greg. nom. nud., and chromosome pairing and pollen fertility in their interspecific F1 hybrids were studied to further understand the phylogenetic relationships among these species. Except those with A. batizocoi Krap. et Greg. nom. nud., hybrids between diploid species have near normal bivalent frequency (9.1–9.8) and moderate to high pollen fertility (60–91%). Hybrids between A. batizocoi and other species have low bivalent frequency (5.2–6.9) and very low pollen fertility (3–7%). These results confirm the earlier separation of these species into two groups based on karyomorphology and Mahalanobis D2 calculated on arm ratios. These studies also provide a picture of relative affinities between A. batizocoi, the lone member of one cluster, and the other species, and among the rest of the species. They also indicate that the basic chromosome complement in the two groups of species is the same. Chromosome pairing in triploid hybrids, (A. hypogaea L. X diploid wild species), suggests that A. batizocoi is the closest diploid relative of A. hypogaea. It is closer to A. hypogaea subspecies fastigiata Waldron than to A. hypogaea subspecies hypogaea Krap. et. Rig. Other diploid species of the section Arachis are equidistant from A. hypogaea, and have the same genome which has strong homology to one of the genomes of A. hypogaea. Based on the present results, the two tetraploid species, A. monticola Krap. et Rig. and A. hypogaea can be recognised as two forms of the same species. Breeding implications have been discussed in the light of chromosome behaviour observed in hybrids of A. hypogaea X diploid species, and on the presumptions that A. hypogaea has an AABB genomic constitution, and that among the diploid species, the B genome is present in A. batizocoi while the A genome is common to the other diploid species of section Arachis.Submitted as Journal Article No. 328 by the International Crops Research Institute for the Semi-Arid Tropics (ICRISAT)  相似文献

The present work provides new data on the banding pattern of two cyprinid fish species Phoxinus phoxinus and Eupallasella perenurus from Poland. C-banding, silver-staining (Ag), and fluorescent staining with chromomycin A3 techniques were used to describe the karyotypes. Both of the species karyotypes of 2n=50 were characterised by one pair of acrocentric chromosomes, the largest in the set, and by two pairs of NOR-bearing chromosomes. In the chromosome set of Ph. phoxinus Ag-stained NORs were located on telomeres of two metacentric and two submetacentric chromosomes, but in most metaphases only one of the two homologous was observed. The karyotype of E. perenurus was characterised by Ag-NOR regions at a telomeric position on the shorter arm of two submetacentric chromosome pairs. In most metaphases only three NOR-bearing chromosomes were observed. In both investigated species the location of the A3 positive signals corresponded with the location of Ag-stained NORs and these sites were associated with heterochromatin shown as C-bands. The results of cytogenetical studies on other related, mainly the North American phoxinins, species are compared and discussed.  相似文献

Summary The seed proteins of Chinese Spring wheat stocks which possess single chromosomes from other plant species related to wheat have been separated by gel electrophoresis in the presence of sodium dodecyl sulphate. Marker protein bands have been detected for both arms of barley chromosome 5, chromosome E (= 1R) and B (= 2R) of rye, chromosomes A,B (= 1Cu) and C (= 5Cu) of Aegilops umbellulata and chromosomes I and III of Agropyron elongatum. These studies, and previous findings, indicate that chromosome 5 of barley, chromosome 1R of rye, chromosome I of Ag. elongatum and possibly chromosome 1Cu of Ae. umbellulata are similar to chromosomes 1A, 1B and 1D in hexaploid wheat in that they carry genes controlling prolamins on their short arms and genes controlling high-molecular-weight (apparent molecular weight greater than 86,000) seed protein species on their long arms. These findings support the idea that all these chromosomes are derived from a common ancestral chromosome and that they have maintained their integrity since their derivation from that ancestral chromosome.  相似文献

Summary Meiotic chromosome pairing and Giemsa C-banding analyses in crosses of several European blue-grained wheat strains with Chinese Spring double ditelosomic and other aneuploid lines showed that Triticum aestivum Blaukorn strains Berlin, Probstdorf, Tschermak, and Weihenstephan are chromosome substitutions, in which the complete wheat chromosome 4A pair is replaced, whereas the strains Brünn and Moskau are 4B substitutions. The alien chromosome pair in all of these strains is an A genome chromosome (4A) from diploid Triticum monococcum or T. boeoticum not present in common tetraploid and hexaploid cultivated wheats. The Blaukorn strain Weihenstephan W 70a86 possesses, in addition to a rye chromosome pair 5R compensating for the loss of part of chromosome 5D, a 4A/5DL translocation replacing chromosome pair 4B of wheat.  相似文献

Karyotype analyses were conducted onCunninghamia konishii, Cunninghamia lanceolata, andTaiwania cryptomerioides, all members ofTaxodiaceae. The somatic chromosome number was found to be 2n = 2x = 22 in all species which concurrs with previous reports. The karyotypes are generally asymmetrical with the smaller chromosomes being more submedian than the larger ones. Chromosomes with unusual or specific structures, thought to be associated with the nucleolar organizing region, were found in each species.Cunninghamia species have a marker chromosome pair with an unusually long secondary constriction.Taiwania has an unusually long kinetochore region present in a submedian chromosome pair.  相似文献

The interaction of 3,5-bis-O-(,,,-tetrafluoropyrid--yl)thymidine with various nucleophilic reagents was studied to evaluate the possibility of molecular design of new types of nucleic acid analogues using S NAr reactions. The reactions with morpholine and sodium azide led to the introduction of one and two nucleophilic residues into each of the polyfluorinated pyridine rings. The nucleophilic polycondensation with bifunctional reagents ethylenediamine and hexamethylenediamine depended on the nature of nucleophile and reaction conditions and resulted in the formation of supramolecules containing about five or more than 20 pyrimidine bases.  相似文献

Constitutive heterochromatin of a karyotypically conserved species of harvest mouse was compared to that of three karyotypically derived species of harvest mice by examining banding patterns produced on metaphase chromosomes with three restriction endonucleases (EcoRI, MboI and PstI). Banding patterns produced by two of these restriction endonucleases (EcoRI and MboI) were compared to published G- and C-banded karyotypes and in situ hybridization of a satellite DNA repeat for these taxa. The third restriction endonuclease (PstI) did not produce a detectable pattern of digestion. For the most part, patterns produced by EcoRI and MboI can be related to C-banded chromosomes and in situ hybridization of satellite DNA sequences. Moreover, digestion with EcoRI reveals bands not apparent with these other techniques, suggesting that restriction endonuclease digestion of metaphase chromosomes may provide additional insight into the structure and organization of metaphase chromosomes. The patterns produced by restriction endonuclease digestion are compatible with the chromosomal evolution of these taxa, documenting that in the highly derived taxa not only are the chromosomes rearranged but the abundance of certain sequences is highly variable. However, technical variation and difficulty in producing consistent results even on a single slide with some restriction endonucleases documents the problems associated with this method.  相似文献

Summary The nature of genome change during polyploid evolution was studied by analysing selected species within the tribe Triticeae. The levels of genome changes examined included structural alterations (translocations, inversions), heterochromatinization, and nucleotide sequence change in the rDNA regions. These analyses provided data for evaluating models of genome evolution in polyploids in the genus Triticum, postulated on the basis of chromosome pairing at metaphase I in interspecies hybrids.The significance of structural chromosome alterations with respect to reduced MI chromosome pairing in interspecific hybrids was assayed by determining the incidence of heterozygosity for translocations and paracentric inversions in the A and B genomes of T. timopheevii ssp. araraticum (referred to as T. araraticum) represented by two lines, 1760 and 2541, and T. aestivum cv. Chinese Spring. Line 1760 differed from Chinese Spring by translocations in chromosomes 1A, 3A, 4A, 6A, 7A, 3B, 4B, 7B and possibly 2B. Line 2541 differed from Chinese Spring by translocations in chromosomes 3A, 6A, 6B and possibly 2B. Line 1760 also differed from Chinese Spring by paracentric inversions in arms 1AL and 4AL whereas line 2541 differed by inversions in 1BL and 4AL (not all chromosomes arms were assayed). The incidence of structural changes in the A and B genomes did not coincide with the more extensive differentiation of the B genomes relative to the A genomes as reflected by chromosome pairing studies.To assay changing degrees of heterochromatinization among species of the genus Triticum, all the diploid and polyploid species were C-banded. No general agreement was observed between the amount of heterochromatin and the ability of the respective chromosomes to pair with chromosomes of the ancestral species. Marked changes in the amount of heterochromatin were found to have occurred during the evolution of some of the polyploids.The analysis of the rDNA region provided evidence for rapid fixation of new repeated sequences at two levels, namely, among the 130 bp repeated sequences of the spacer and at the level of the repeated arrays of the 9 kb rDNA units. These occurred both within a given rDNA region and between rDNA regions on nonhomologous chromosomes. The levels of change in the rDNA regions provided good precedent for expecting extensive nucleotide sequence changes associated with differentiation of Triticum genomes and these processes are argued to be the principal cause of genome differentiation as revealed by chromosome pairing studies.  相似文献

Summary The DNA polymerase III holoenzyme is a complex, multisubunit enzyme that is responsible for the synthesis of most of the Escherichia coli chromosome. Through studies of the structure, function and regulation of this enzyme over the past decade, considerable progress has been made in the understanding of the features of a true replicative complex. The holoenzyme contains at least seven different subunits. Three of these, , and , compose the catalytic core. Apparently is the catalytic subunit and the product of the dnaE gene. Epsilon, encoded by dnaQ (mutD), is responsible for the proofreading 35 activity of the polymerase. The function of the B subunit remains to be established. The auxiliary subunits, , and , encoded by dnaN, dnaZ and dnaX, respectively, are required for the functioning of the polymerase on natural chromosomes. All of the proteins participate in increasing the processivity of the polymerase and in the ATP-dependent formation of an initiation complex. Tau causes the polymerase to dimerize, perhaps forming a structure that can coordinate leading and lagging strand synthesis at the replication fork. This dimeric complex may be asymmetric with properties consistent with the distinct requirements for leading and lagging strand synthesis.  相似文献

Summary Somatic chromosomes of two cultivais of Cajanus cajan, eight species of Atylosia (A. albicans, A. cajanifolia, A. lineata, A. platycarpa, A. scarabaeoides, A. serica, A. trinervia and A. volubilis), and of Rhynchosia rothii were analysed. All species had 2n=22. Eight of the 10 species studied had two pairs of satchromosomes while A. scarabaeoides and A. sericea had only one sat-chromosome pair. Based on relative chromosome length (L%), arm ratio (pa-value) and presence or absence of secondary constriction, a karyotype formula for each species was formulated. Based on these parameters the chromosome pairs could also be assigned to groups ranging from 8 to 10 in different species. Except for the asymmetrical karyotype of A. albicans, the other species had rather moderately symmetrical karyotypes.  相似文献

Whether the two tetraploid wheat species, the well known Triticum turgidum L. (macaroni wheat, AABB genomes) and the obscure T. timopheevii Zhuk. (AtAtGG), have monophyletic or diphyletic origin from the same or different diploid species presents an interesting evolutionary problem. Moreover, T. timopheevii and its wild form T. araraticum are an important genetic resource for macaroni and bread-wheat improvement. To study these objectives, the substitution and genetic compensation abilities of individual T. timopheevii chromosomes for missing chromosomes of T. aestivum Chinese Spring (AABBDD) were analyzed. Chinese Spring aneuploids (nullisomic-tetrasomics) were crossed with a T. timopheevii x Aegilops tauschii amphiploid to isolate T. timopheevii chromosomes in a monosomic condition. The F1 hybrids were backcrossed one to four times to Chinese Spring aneuploids without selection for the T. timopheevii chromosome of interest. While spontaneous substitutions involving all At- and G-genome chromosomes were identified, the targeted T. timopheevii chromosome was not always recovered. Lines with spontaneous substitutions from T. timopheevii were chosen for further backcrossing. Six T. timopheevii chromosome substitutions were isolated: 6At (6A), 2G (2B), 3G (3B), 4G (4B), 5G (5B) and 6G (6B). The substitution lines had normal morphology and fertility. The 6At of T. timopheevii was involved in a translocation with chromosome 1G, resulting in the transfer of the group-1 gliadin locus to 6At. Chromosome 2G substituted for 2B at a frequency higher than expected and may carry putative homoeoalleles of gametocidal genes present on group-2 chromosomes of several alien species. Our data indicate a common origin for tetraploid wheat species, but from separate hybridization events because of the presence of a different spectrum of intergenomic translocations.  相似文献

Arachis batizocoi Krap. & Greg. is a suggested B genome donor to the cultivated peanut,A. hypogaea L. Until recently, only one accession of this species was available in U.S.A. germplasm collections for analyses and species variability had not been documented. The objective of this study was to determine the intraspecific variability ofA. batizocoi to better understand phylogenetic relationships in sect.Arachis. Five accessions of the species were used for morphological and cytological studies and then F1 intraspecific hybrids analyzed. Some variation was observed among accessions—for example, differences in seed size, plant height and branch length. The somatic chromosomes of accessions 9484, 30079, and 30082 were nearly identical, whereas, the karyotypes of accessions 30081 and 30097 have several distinct differences. For example, 30081 had significantly more asymmetrical chromosomes 2 and 6 and more median chromosomes 7 and 10, and 30097 had significantly more asymmetrical chromosomes 3 and 10 and more median chromosomes 1 and 5 than accessions 9484, 30079, and 30082. All F1 hybrids among accessions were highly fertile. Meiotic observations indicated that hybrids among accessions 9484, 30079, or 30082 had mostly bivalents. However, quadrivalents were observed when either 30081 or 30097 was crossed with the above three accessions and 30081 × 30097 had quadrivalents, hexavalents and octavalents. The presence of translocations is the most likely cause of multivalent formation inA. batizocoi hybrids. C-banding polymorphism was analyzed in 14 accessions of Triticum searsii from Israel, and a generalized idiogram of the species was established. One accession was homozygous for whole arm translocations T1SsS·4SsS and T1SsL·4SsL. C-banding analysis was also used to identify 7 T. aestivum cv Chinese Spring-T. searsii disomic chromosome addition lines, 14 ditelosomic chromosome addition lines, 21 disomic whole chromosome, and 31 ditelosomic chromosome substitution lines. The identity of these lines was further confirmed by meiotic pairing analysis. Sporophytic and gametophytic compensation tests were used to determine the homoeologous relationships of the T. searsii chromosomes. The results show that the T. searsii chromosomes do not compensate well for their wheat homoeologues. The C-banding patterns of T. searsii chromosomes are distinct from those of other S-genome species and from the B-genome chromosomes of wheat, indicating that T. searsii is not a direct B-genome donor species of T. turgidum and T. aestivum.Contribution No. 95-72-J from the Kansas Agricultural Experiment Station, Kansas State University, Manhattan, Kansas, USA  相似文献

Summary Restriction fragment length polymorphisms (RFLP) were assessed among accessions within six peanut species of the Arachis section: tetraploid cultivated species, A. hypogaea; tetraploid wild species, A. monticola; and four diploid wild species, A. batizocoi,A. cardenasii, A. duranensis and A. glandulifera. While the two tetraploid species did not show polymorphism with 16 PstI-generated random genomic probes, two of seven seed cDNA probes detected polymorphisms. The RFLP variation detected by two seed cDNA probes appeared to be related to structural changes occurring within tetraploid species. The botanical var. fastigiata (Valencia market type) of A. hypogaea subspecies fastigiata was shown to be the most variable. Arachis monticola was found to be more closely related to A. hypogaea subspecies hypogaea than to subspecies fastigiata. Diploid species A. cardenasii, A. duranensis, and A. glandulifera showed considerable intraspecific genetic diversity, but A. batizocoi showed little polymorphism. We report here the in vivo production of type 2 fucosylated-N-acetyllactosamine oligosaccharides in Escherichia coli. Lacto-N-neofucopentaose Gal1-4GlcNAc1-3Gal1-4(Fuc1-3)Glc, lacto-N-neodifucohexaose Gal1-4(Fuc1-3)Glc-NAc1-3Gal1-4(Fuc1-3)Glc, and lacto-N-neodifucooctaose Gal1-4GlcNAc1-3Gal1-4(Fuc1-3)GlcNAc1-3Gal1-4(Fuc1-3)Glc were produced from lactose added in the culture medium. Two of them carry the Lewis X human antigen. High cell density cultivation allowed obtaining several grams of fucosylated oligosaccharides per liter of culture. The fucosylation reaction was catalyzed by an -1,3 fucosyltransferase of Helicobacter pylori overexpressed in E. coli with the genes lgtAB of N. meningitidis. The strain was genetically engineered in order to provide GDP-fucose to the system, by genomic inactivation of gene wcaJ involved in colanic acid synthesis and overexpression of RcsA, positive regulator of the colanic acid operon.To prevent fucosylation at the glucosyl residue, lactulose Gal1-4Fru was assayed in replacement of lactose. Lactulose-derived oligosaccharides carrying fucose were synthesized and characterized. Fucosylation of the fructosyl residue was observed, indicating a poor acceptor specificity of the fucosyltransferase of H. pylori.  相似文献

Karyotype studies on 20 taxa of gymnosperms endemic to New Zealand show a wide diversity of chromosome number and form. Fluorochrome banding with DAPI and CMA reveals a depauperate pattern of bands with CMA and no reliable banding with DAPI. Characteristically one pair of chromosomes shows a prominent CMA band, which may or may not be associated with a secondary constriction. A band size polymorphism was observed in all plants ofDacrycarpus dacrydioides, irrespective of the sex of the plant. Measurements of genome size by flow cytometry show a range of values from 12.3 pg to 40.0 pg DNA per 2C nucleus. Intraspecific variation in genome size was observed inManoao colensoi.  相似文献

The karyotypes ofElymus dentatus from Kashmir andE. glaucescens from Tierra del Fuego, both carrying genomesS andH, were investigated by C- and N-banding. Both taxa had 2n = 4x = 28. The karyotype ofE. dentatus was symmetrical with large chromosomes. It had 18 metacentric, four submetacentric and six satellited chromosomes. The karyotype ofE. glaucescens resembled that ofE. dentatus, but a satellited chromosome pair was replaced by a morphologically similar, non-satellited pair. The C-banding patterns of both species had from one to five conspicuous and a few inconspicuous bands per chromosome. N-banding differentiated the chromosomes of the constituent genomes by producing bands in theH genome only. TheS genomes of both species were similar with five metacentric and two satellited chromosomes having most conspicuous C-bands at telomeric and distal positions. They resembled theS genome of the genusPseudoroegneria. TheH genomes had four similar metacentric and two submetacentric chromosomes. The seventhH genome chromosome ofE. dentatus was satellited, that ofE. glaucescens nonsatellited, but otherwise morphologically similar. The C-bands were distributed at no preferential positions. TheH genome ofE. dentatus resembles theH genomes of some diploidHordeum taxa.  相似文献

Summary Isoelectric focusing of extracts from the endosperm of mature grains of hexaploid wheat and related species was used to study the genetic control of Iodine binding factor (IBF). Ten IBF bands were present in Chinese Spring (CS) and analysis of the nullisomictetrasomic and ditelosomic lines of CS showed nine of them to be controlled by genes on the long arms of the homoeologous group 5 chromosomes. Five alleles were detected at Ibf-A1 locus, four at Ibf-B1 and four at Ibf-D1 among a sample of 46 wheat genotypes. Homoeoloci were found on chromosome 5R of Secale cereale, 5E of Agropyron elongatum, 5U of Aegilops umbellulata, 5Agi of Agropyron intermedium, 5S1 and 4S1 of Aegilops sharonensis and 4H of Hordeum vulgare.  相似文献

Summary Despite an average difference of about 50% in DNA amount, homoeologous chromosomes pair effectively at first metaphase in the diploid interspecific hybrid between Lolium temulentum and Lolium perenne. However, in the presence of accessory B chromosomes and diploidising genes pairing at metaphase I is severely reduced. Reconstruction of serial electron micrographs through pollen mother cell nuclei show that synaptonemal complexes are formed at pachytene between not only homoeologous but also non-homologous chromosome segments resulting in multivalent formation. These associations are largely ineffective in terms of chiasma formation and degenerate at late pachytene. It is highly probable that the pairing determinants exercise their control on chromosome pairing largely by prohibiting the siting of crossovers in homoeologously paired chromosome segments.  相似文献

The geographical distribution of the grass genusHelictotrichon in the Mediterranean Region is analysed and mapped for 33 taxa. Based on only a single life form (perennial herb), a variety of edaphically, climatically and altitudinally differently adapted species complexes has evolved in the area. Most of these complexes show west-east disjunctions and contain geographically, sometimes even edaphically vicarious taxa with complementary distribution. A transition from mesomorphic to xeromorphic habit occurred independently in different species groups and led to the establishment of the modern Mediterranean taxa which are in part highly polyploid derivatives of more mesophilic diploids. The significance of polyploidy, patterns of parapatric and sympatric distribution, biogeographical borders, and centres of species diversity are discussed in context with the history of the Mediterranean vegetation. New combinations are:Helictotrichon setaceum subsp.petzense, H. pratense subsp.lusitanicum, H. praetutianum.  相似文献
