酶促反应速度是酶促反应动力学中的一个基本概念。一定条件下 ,酶催化反应的活力大小要由该反应条件下酶促反应速度来反映和确定 :酶促反应速度越小 ,酶活力越小 ;反之 ,酶促反应速度越大 ,酶活力也越大。在研究底物浓度、酶浓度、温度、pH、激活剂、抑制剂等因素对酶活力的影响时 ,都必须测定酶促反应的速度。为了排除实验中存在的一些干扰 ,一个酶的酶促反应的速度要在酶促反应进行的初期测定 ,即测定所谓的酶促反应的初速度。在教学过程中 ,学生不易弄清楚酶促反应速度与酶促反应的初速度之间的关系。我们发现 :当一般地问起酶反应速度…  相似文献

超声对胃蛋白酶,胰蛋白酶,过氧化氢酶作用的研究   总被引:3,自引:0,他引:3  
以胃蛋白酶,胰蛋白酶,过氧化氢酶溶液在超声处理下的酶活变化为指标研究超声对蛋白质作用的机理和影响因素。结果表明超声对蛋白质的破坏程度随着功率的升高或处理时间的延长而增加;三种酶在超声作用下酶活变化形式和程度各不同;浓度是影响超声对酶作用效果的一个重要因素,可通过调整酶溶液浓度来减少酶所受到的破坏程度。自由基清除剂甘露醇和非离子表面活性剂吐温-80可以对酶活在超声作用下起到一定的保护作用,说明自由基和超声空穴是超声破坏酶结构的重要机理,研究结果同时表明对于不同的酶,超声的破坏作用可能有不同的发挥主导作用机理。  相似文献

 兎肌肌酸激酶被LDS变性后,底物能够诱导变性酶使其活力和构象得到部分恢复。变性程度不同的酶,构象和活力的恢复程度也不同:低浓度LDS变性酶,恢复程度较高;反之亦然。活力的恢复与构象的恢复两者呈对应关系。底物修复作用的pH以8.2为好。底物修复作用受其它蛋白质(例如BSA)存在的影响。等速电泳结果表明,BSA能竞争性结合LDS-酶复合物的LDS,使酶成为游离酶。变性酶先与BSA保温再加底物所得的活力恢复,大约是变性酶与含BSA的底物保温所得活力的10倍。这一结果似表明LDS变性酶仍能结合底物;被结合的底物还能使变性酶的构象发生变化。  相似文献

以酶促聚合为代表的绿色高分子合成途径,以其反应条件温和、产物多分散性低、无金属催化剂残留、高度立体和区位选择性等优势,成为医用高分子材料合成领域中的研究热点。目前,氧化还原酶、水解酶、转移酶均成功应用于聚合反应,其中脂肪酶催化的缩聚反应及开环聚合反应研究最为广泛,同时,以可逆加成-断裂链转移聚合和原子转移自由基聚合为代表的酶促可逆失活自由基聚合得到了快速发展。针对酶促聚合中单体及合成产物结构与性能单一、应用范围有限等缺陷,基于酶促聚合与原子转移自由基聚合、开环易位聚合等反应的偶联,制备了多种不同结构与性能的聚合物材料,推动了上述材料在药物与基因递送领域中的应用。本文综述了脂肪酶催化聚合、酶促可逆失活自由基聚合、酶促化学偶联催化等方面的研究进展,并探讨了目前研究的局限性和未来研究方向。  相似文献

从埋麻土壤中分离到放线菌298株,经初筛和复筛得到产酶活性较高的一株放线菌5-71。最适产酶条件是:果胶0.5%,葡萄糖0.5%,蛋白胨0.5%,酵母粉0.1%,K2HPO4 0.1%,KH2PO4 0.1%,NaCl0.1%,MgSO4·7H2O0.05%,pH8.0。25℃培养3天达产酶高峰。通气量对产酶影响不大。酶促反应最适条件是:pH9.6,45℃,底物果胶浓度0.75%,作用时间90mi  相似文献

氨基酰化酶在阴离子去污剂十二烷基硫酸溶液中的失活与去折叠的研究结果表明,在低浓度的LDS溶液中变性时,以荧光和紫外差吸收方法监测的酶分子构象尚未发生明显变化,而酶的活力已经大部分或几乎全部丧失。当LDS浓度达1.6mmol/L时,此时酶分子的构象变化才达到最大程度。在实验使用的LDS的浓度范围内,用远紫外CD光谱监测的二级结构没有发生明显的变化。  相似文献

脂肪酶具有非水催化作用,但其非水催化活性和稳定性需进一步提高,这是非水酶学的瓶颈问题之一。理想的策略是模拟脂肪酶的界面活化机制,以大分子代替水,优化、稳定化和有效分散酶蛋白,阻止其在有机相中变性。因此,选用多羟基、比表面积大、惰性、且与酶蛋白能兼容的大分子--脱脂棉纤维,作为固定化载体,以1∶0.9的质量比,通过物理吸附,将假单胞菌脂肪酶(Pseudomonas cepacia lipase)固定在脱脂棉纤维上。在催化己醇与乙酸乙烯酯的转酯反应中,反应1 h,脱脂棉固定化脂肪酶转化底物的能力是酶粉的3.7倍。在每次6 h共6次的循环催化中,固定化酶和酶粉转化底物的能力分别平均每次降低约0.3%和2.4%。表明脱脂棉固定化脂肪酶的非水活性,尤其是稳定性明显提高。这为通过固定化有效提高脂肪酶的非水催化作用,以满足工业应用的需要,提供了一种有效的途径和重要参考。  相似文献

许多脂肪酶在有机体系中表现出催化作用,可用于绿色有机合成. 但其催化活性和稳定性明显低于水/油(有机相)界面上的表现. 为了提高脂肪酶在有机反应体系中的活性和稳定性,依据脂肪酶的界面活化机制,以水为酶蛋白构象优化剂、羧甲基纤维素为赋形剂,通过物理吸附的方式,将典型的假单胞菌脂肪酶(Pseudomonas cepacia lipase)固定在锥形瓶的内壁上,形成简易的生物反应器. 为方便检测器壁固定化酶促反应动力学,选择特征吸收为640 nm的生化指示剂2,6-二氯靛酚为反应底物,乙酸乙烯酯为酰化试剂,丙酮为溶剂. 光谱检测表明,催化反应0.5 h后,器壁固定化脂肪酶转化底物的能力是脂肪酶粉的6倍. 在每次催化5 h共10次的循环催化中,器壁固定化脂肪酶的催化活性平均每次仅降低3.2%,而酶粉降低11.8%. 结果表明,该器壁固定化脂肪酶的活性和稳定性相对于酶粉明显提高,这将为通过固定化有效提高脂肪酶的非水催化作用提供重要的参考.  相似文献

Salt hydrates very frequently are utilized as in situ water activity buffers in reaction mixtures of enzymes in nonaqueous media. In addition to buffering water activity, there is evidence that salt hydrates also often affect initial rates in other ways. This has been generally overlooked or thought to be related to water transfer effects. Here we show that salt hydrates can have important acid-base effects on enzymes in nonaqueous media. We performed transesterification reactions in n-hexane and in supercritical ethane catalyzed by cross-linked crystals of subtilisin, differing in the method used to set a(W), and confirmed that the presence of salt hydrate pairs significantly affected the catalytic performance of the enzyme. However, in the presence of a solid-state acid-base buffer, salt hydrates had no effect on enzymatic activity. Direct evidence for the acid-base effects of salt hydrates was obtained by testing their effect on the protonation state of an organo-soluble H(+)/Na(+) indicator. The four salt hydrate pairs tested affected the indicator to very different extents. By promoting the exchange of H(+) for Na(+), salt hydrates will tend to affect the ionization state of acidic residues in the protein and, hence, enzymatic activity. In fact, salt hydrates were able to affect the pH memory of subtilisin lyophilized from different aqueous pHs, bringing about up to 20-fold enhancements and up to 5-fold decreases in catalytic activity. The possibility of such acid-base effects need to be considered in all experiments using salt hydrates to control water activity.  相似文献

探明二斑叶螨取食对棉花叶片内应激防御性酶活性的影响。在棉苗5-6叶期接种不同数量(10-40头)的二斑叶螨雌成虫,取食1-4 d后采摘不同处理的叶片,利用分光光度计测定过氧化氢酶(CAT)、过氧化物酶(POD)、多酚氧化酶(PPO)、脂氧合酶(LOX)和苯丙氨酸解氨酶(PAL)活性。结果表明二斑叶螨取食后,棉花叶片CAT活性显著升高,且随诱导时间的延长其活性变化呈先上升后下降的趋势,为害3 d时,10、20、H(每株接棉蚜跟叶螨各40头)头/叶的CAT活性达到最高峰,显著高于对照(P0.05);POD、PAL活性也随诱导时间的延长和螨口数量的增加呈上升趋势,为害4 d时达到最高峰;而PPO、LOX活性随着时间的增加呈现上升后下降趋势,且为害2 d时达到最高峰,仍显著高于对照(P0.05)。棉叶应激防御酶活性随着二斑叶螨为害时间与螨口密度的增加而增加,表明二斑叶螨的危害胁迫能诱导寄主植物产生一系列应激生化反应,不同的酶活性表现有所不同。  相似文献

Lipozyme IM20 from Novo Nordisk (Denmark) was examined after various treatments. Conditions were chosen to reflect those that would be considered in the design of an industrial process. A two-level factorial design was employed to assess the effects of pressurization/depressurization cycles, rate of depressurization and exposure length. A significant three-factor interaction was observed. Lowest residual activity was observed for runs in which the depressurization rate was 86–89 bar min–1. Incubation for 12 h also yielded low residual activity but only when exposing the immobilized enzyme to one cycle. The highest residual activity was obtained for immobilized enzymes repeatedly exposed for periods of 12 h (5 times) with a depressurization rate of 4.3 to 4.45 bar min–1. This effect may be due to the extraction of an inhibiting compound. Tuning process parameters can lead to a seven-fold change in residual activity.  相似文献

We discuss the hydrolysis of cellulose using a pure cellulase: endo-1,4-β-D-glucanase (EG) from the fungus, Aspergillus niger, in buffer, the pure ionic liquid (IL), tris-(2-hydroxyethyl)-methylammonium methylsulfate (HEMA), and various mixtures of the two at different temperatures. Steady-state fluorescence and absorbance studies were performed to monitor the stability and activity of EG using cellulose azure as the substrate. EG attains its highest activity at 45°C in buffer and denatures at ~55°C. On the other hand, HEMA imparts substantial stability to the enzyme, permitting the activity to peak at 75°C. The relative roles of temperature, viscosity, pH, polarity, and the constituent ions of the ILs on the hydrolysis reaction are examined. It is demonstrated that pretreatment of cellulose with ILs such as BMIM Cl, MIM Cl, and HEMA results in more rapid conversion to glucose than hydrolysis with cellulose that is not pretreated. The percent conversion to glucose from pretreated cellulose is increased when the temperature is increased from 45 to 60°C. Two different ILs are used to increase the efficiency of cellulose conversion to glucose. Cellulose is pretreated with BMIM Cl. Subsequent hydrolysis of the pretreated cellulose in 10-20% solutions of HEMA in buffer provides higher yields of glucose at 60°C. Finally, to our knowledge, this is the first study dealing with a pure endoglucanase from commercial A. niger. This enzyme not only shows higher tolerance to ILs, such as HEMA, but also has enhanced thermostability in the presence of the IL.  相似文献

Understanding the characteristics that define temperature-adapted enzymes has been a major goal of extremophile enzymology in recent decades. In the present study, we explore these characteristics by comparing psychrophilic, mesophilic, and thermophilic enzymes. Through a meta-analysis of existing data, we show that psychrophilic enzymes exhibit a significantly larger gap (Tg) between their optimum and melting temperatures compared with mesophilic and thermophilic enzymes. These results suggest that Tg may be a useful indicator as to whether an enzyme is psychrophilic or not and that models of psychrophilic enzyme catalysis need to account for this gap. Additionally, by using predictive protein stability software, HoTMuSiC and PoPMuSiC, we show that the deleterious nature of amino acid substitutions to protein stability increases from psychrophiles to thermophiles. How this ultimately affects the mutational tolerance and evolutionary rate of temperature adapted organisms is currently unknown.  相似文献

Salt-activation of nonhydrolase enzymes for use in organic solvents   总被引:1,自引:0,他引:1  
Enzymatic reactions are important for the synthesis of chiral molecules. One factor limiting synthetic applications of enzymes is the poor aqueous solubility of numerous substrates. To overcome this limitation, enzymes can be used directly in organic solvents; however, in nonaqueous media enzymes usually exhibit only a fraction of their aqueous-level activity. Salt-activation, a technique previously demonstrated to substantially increase the transesterification activity of hydrolytic enzymes in organic solvents, was applied to horse liver alcohol dehydrogenase, soybean peroxidase, galactose oxidase, and xanthine oxidase, which are oxidoreductase and oxygenase enzymes. Assays of the lyophilized enzyme preparations demonstrated that the presence of salt protected enzymes from irreversible inactivation. In organic solvents, there were significant increases in activity for the salt-activated enzymes compared to nonsalt-activated controls for every enzyme tested. The increased enzymatic activity in organic solvents was shown to result from a combination of protection against inactivation during the freeze-drying process and other as-yet undetermined factors.  相似文献

在5 L发酵罐进行甘油脉冲流加发酵,分析了不同pH值对克雷伯氏肺炎杆菌发酵特性的影响,pH 6.5为菌体最佳生长条件,克雷伯氏肺炎杆菌合成1,3-丙二醇的产量最高。在1,3-丙二醇合成速率较大的对数中前期,进行甘油脉冲流加发酵,提高甘油浓度促进甘油脱水酶、1,3-丙二醇氧化还原酶和甘油脱氢酶活性。不同pH值的脉冲试验表明,甘油脱水酶,2,3-丁二醇脱氢酶比酶活随着pH值的升高而升高,1,3-丙二醇氧化还原酶,乳酸脱氢酶比酶活在pH6.5最高,因此偏酸性的发酵条件和对数期维持一定的甘油浓度能够促进1,3-丙二醇的合成。  相似文献
