Onion (Allium cepa L.) plants were examined to determine the photosynthetic role of CO2 that accumulates within their leaf cavities. Leaf cavity CO2 concentrations ranged from 2250 L L–1 near the leaf base to below atmospheric (<350 L L–1) near the leaf tip at midday. There was a daily fluctuation in the leaf cavity CO2 concentrations with minimum values near midday and maximum values at night. Conductance to CO2 from the leaf cavity ranged from 24 to 202 mol m–2 s–1 and was even lower for membranes of bulb scales. The capacity for onion leaves to recycle leaf cavity CO2 was poor, only 0.2 to 2.2% of leaf photosynthesis based either on measured CO2 concentrations and conductance values or as measured directly by 14CO2 labeling experiments. The photosynthetic responses to CO2 and O2 were measured to determine whether onion leaves exhibited a typical C3-type response. A linear increase in CO2 uptake was observed in intact leaves up to 315 L L–1 of external CO2 and, at this external CO2 concentration, uptake was inhibited 35.4±0.9% by 210 mL L–1 O2 compared to 20 mL L–1 O2. Scanning electron micrographs of the leaf cavity wall revealed degenerated tissue covered by a membrane. CO2 exchange components of a temperate semi-desert sand grassland ecosystem in Hungary were measured 21 times in 2000–2001 using a closed IRGA system. Stand CO2 uptake and release, soil respiration rate (R s), and micrometeorological values were determined with two types of closed system chambers to investigate the daily courses of gas exchange. The maximum CO2 uptake and release were –3.240 and 1.903 mol m–2 s–1, respectively, indicating a relatively low carbon sequestration potential. The maximum and the minimum R s were 1.470 and 0.226 mol(CO2) m–2 s–1, respectively. Water shortage was probably more effective in decreasing photosynthetic rates than R s, indicating water supply as the primary driving variable for the sink-source relations in this ecosystem type.  相似文献

The steady-state and dynamic photosynthetic response of two poplar species (Populus tremuloides and P. fremontii) to variations in photon flux density (PFD) were observed with a field portable gas exchange system. These poplars were shown to be very shade intolerant with high light saturation (800 to 1300 mol photons m–2 s–1) and light compensation (70 to 100 mol m–2 s–1) points. Understory poplar leaves showed no physiological acclimation to understory light environments. These plants become photosynthetically induced quickly (10 min). Activation of Rubisco was the primary limitation for induction, with stomatal opening playing only a minor role. Leaves maintained high stomatal conductances and stomata were unresponsive to variations in PFD. Leaves were very efficient at utilizing rapidly fluctuating light environments similar to those naturally occurring in canopies. Post-illumination CO2 fixation contributed proportionally more to the carbon gain of leaves during short frequent lightflecks than longer less frequent ones. The benefits of a more dynamic understory light environment for the carbon economy of these species are discussed.  相似文献

Strawberry (Fragaria ananassaDuch. cv. Fengxiang) plantlets were cultured under two in vitroenvironments for rooting, and then acclimatized under two ex vitroirradiance conditions. At the end of rooting stage plant height, fresh weight and specific leaf area of T1-plants grown under high sucrose concentration (3 sucrose), low photosynthetic photon flux density (30 mol m–2 s–1) and normal CO2 concentration (350–400 l l–1) were significantly higher than those of T2-plantlets grown under low sucrose concentration (0.5), high photosynthetic photon flux density (90 mol m–2 s–1) and elevated CO2 concentration (700–800 l l–1). But T2-plantlets had higher net photosynthetic rate (Pn), effective photochemical quantum yield of PSII (PSII), effective photosynthetic electron transport rate (ETR), photochemical quenching (qP) and ratio of chlorophyll fluorescence yield decrease (Rfd). After transfer, higher irradiance obviously promoted the growth of plantlets and was beneficial for the development of photosynthetic functions during acclimatization. T2-plantlets had higher fresh weight, leaf area, PSII and ETR under higher ex vitroirradiance condition.  相似文献

Exchange of CO2 and O2 and chlorophyll fluorescence were measured in the presence of 360 1 · 1–1 CO2 in nitrogen in Helianthus annuss L. leaves which had been preconditioned in the dark or at a photon flux density (PFD) of 24 mol · m–2 · s–1 either in 21 or 0% O2. An initial light-dependent O2 outburst of 6 mol · m–2 was measured after aerobic dark incubation. It was attributed to the reduction of electron carriers, predominantly plastoquinone. The maximum initial rate of O2 evolution at PFD 8000 mol · m–2 · s–1 was 170 mol · m–2 · s–2 or about four times the steady CO2-and light-saturated rate of photosynthesis. Fluorescence measurements showed that the rate was still acceptor-limited. Fast O2 evolution ceased after electron carriers were reduced in the dark-adapted leaf, but continued for a short time at the lower rate of 62 mol · m–2 · s–1 in the light-adapted leaf. The data are interpreted to show that enzymes involved in 3-phosphoglycerate reduction are dark-inhibited, but were fully active in low light. In a dark-adapted leaf, respiratory CO2 evolution continued under nitrogen; it was partially inhibited by illumination. Prolonged exposure of a leaf to anaerobic conditions caused reducing equivalents to accumulate. This was shown by a slowly increasing chlorophyll fluorescence yield which indicated the reduction of the PSII acceptor QA in the dark. When the leaf was illuminated, no O2 evolution was detected from short light pulses, although transient O2 production was appreciable during longer light pulses. Leaf photosynthetic rates were measured on field-grown soybeans during the 1980 season. Comparisons were made between different cultivars and isolines representative of maturity groups I–IV. Mature, fully expanded leaves at different nodes on the plant were measured in high light to determine which had the highest potential photosynthetic rates at any one time. Successive leaves during the growing season had maximum rates which increased from about 22 mol CO2 m-2 s-1 on 25 June to a peak of 30–44 mol CO2 m-2 s-1 in early August.The persistency and eventual decline in the maximum rate was associated with the maturity group and related dates of flowering, pod fill and onset of senescence. Early maturing cultivars (groups I and II) had higher peak rates (38–44 mol CO2 m-2 s-1) than later maturing cultivars (30–35 mol CO2 m-2 s-1, groups III and IV). However, the photosynthetic rates of early maturing cultivars declined rapidly after attaining their peak, whereas the leaves of later maturing cultivars maintained their photosynthetic activity for much longer.  相似文献

This study reports survival and physiological responses of micropropagated Ceratonia siliqua L. cvs. Galhosa and Mulata plants during ex vitro acclimatization under ambient (AC; 330 mol mol–1) or elevated (EC; 810 mol mol–1) CO2 concentration and a photosynthetic photon flux density of 125 mol m–2 s–1. CO2 enrichment during acclimatization did not improve survival rate that was around 80 % for both treatments. Eight weeks after ex vitro transplantation, photosynthetic capacity and apparent quantum yield in acclimatized leaves were higher in comparison with those in in vitro-grown leaves, without any significant difference between CO2 treatments. Chlorophyll content increased after acclimatization. However, EC led to a decrease in the total amount of chlorophyll in new leaves of both cultivars, compared to those grown at AC. Soluble sugars and starch contents were not markedly affected by growth EC, although starch had significantly increased after transfer to ex vitro conditions. EC induced an increase in the stem elongation and in the effective life of leaves, and a decrease in the number of new leaves.  相似文献

Chlorophyll (Chl) a and Chl b contents, rate of CO2 gas exchange, quenching coefficients of chlorophyll fluorescence, and endogenous phytohormones have been studied in primary leaves of barley seedlings cultivated under blue (BL) or red (RL) light. Photon flux densities (PFD) were between 0.3 and 12 mol m-2 s-1. Plants grown at PFD of 0.3 mol m-2 s-1 demonstrated in BL tenfold and in RL threefold decreased Chl content compared to plants grown at 12 mol m-2 s-1. Chl a/b ratio increased from 2.3–2.5 to 4.4–4.5 in BL, not in RL, following the decrease in PFD at plant cultivation from 12 to 0.3 mol m-2 s-1. Plants cultivated at weak BL demonstrated severalfold decreased rate of photosynthetic CO2 uptake, whereas decrease in PFD of RL from 12 to 0.3 mol m-2 s-1 caused only 20% de cline in the rate of photosynthesis. Decrease in PFD during a plant cultivation reduced the maximum quantum yield of photosynthesis in BL, not in RL leaves. Light response curves of non-photochemical and photochemical quenching of chlorophyll fluorescence calculated on the basis of absorbed quanta were not affected by PFD of RL during plant cultivation. On the contrary, both non-photochemical quenching and accumulation of QA -, reduced primary acceptor of Photosystem II, occurred at lower amounts of absorbed quanta in leaves of BL plants grown at 0.3 than at 12 mol m-2 s-1. Two photoregulatory reactions were suggested to exert the light control of the development of photosynthetic apparatus in the range of low PFDs. The photoregulatory reaction saturating by very low PFDs of RL was supposed to be mediated by phytochrome. Phytochrome was proposed to enhance (as related to other pigment-protein complexes of thylakoids) the accu mulation of chlorophyll- b-binding light-harvesting complex of Photosystem II (LHC II). It acts independently of the pigment mediating the second photoregulatory reaction, as evidenced by the results of experiments with plant growth under mixed blue plus red light. The contents of cytokinins and indole-3-acetic acid in a leaf were not significantly affected by either light quality and PFD thus indicating those phytohormones not to be involved into photoregulatory processes.  相似文献

Summary In order to document the natural CO2 environment of the moss Hylocomium splendens, and ascertain whether or not the moss was adapted to this, and its interactions with other microenvironmental factors, two studies were carried out. Firstly, the seasonal variations of CO2 concentration, photosynthetically active radiation (PAR), tissue water content and temperature were measured in the natural microenvironment of H. splendens in a subarctic forest during the summer period (July–September). Secondly, the photosynthetic responses of the species to controlled CO2 concentrations, PAR, temperature, and hydration were measured in the laboratory. CO2 concentrations around the upper parts of the plant, when PAR was above the compensation point (30 mol m–2 s–1), were mostly between 400 and 450 ppm. They occasionally increased up to 1143 ppm for short periods. PAR flux densities below saturating light levels for photosynthesis (100 mol m–2 s–1), occurred during 65% (July), 76% (August) and 96% (September) of the hours of the summer period. The temperature optimum of photosynthesis was 20° C: this temperature coincided with PAR above the compensation point during 5%, 6% and 0% of the time in July, August and September, respectively. Optimal hydration of tissues was infrequent. Hence PAR, temperature and water limit CO2 uptake for most of the growing season. Our data suggest that the higher than normal ambient CO2 concentration in the immediate environment of the plant counteracts some of the limitations in PAR supply that it experiences in its habitat. This species already experiences concentrations of atmospheric CO2 predicted to occur over the next 50 years.  相似文献

The responses of photosynthesis, Rubisco activity, Rubisco protein, leaf carbohydrates and total soluble protein to three carbon dioxide treatments were studied in winter wheat [Triticum aestivum (L.)] and barley [Hordeum vulgare (L.)]. Barley and wheat plants were grown in small field plots during 1995 and 1996 in clear, acrylic chambers (1.2–2.4 m2) and were provided with continuous carbon dioxide fertilization at concentrations of 350, 525 and 700 mol mol–1. Photosynthetic rates of barley penultimate leaves and wheat flag leaves measured at growth carbon dioxide concentrations decreased with leaf age in all three CO2 treatments during 1995 and 1996. Photosynthetic acclimation to elevated CO2 was observed on seven of eight measurement dates for barley and ten of eleven measurement dates for wheat over both years. Initial Rubisco activity, total soluble protein and Rubisco protein in barley penultimate leaves and wheat flag leaves also decreased with leaf age. Total Rubisco activity was not used because of enzyme degradation. There was a significant CO2 treatment effect on initial Rubisco activity, total soluble protein and Rubisco protein for wheat in 1995 and 1996 and for barley in 1995. Responses of barley penultimate leaf Rubisco activity and leaf protein concentrations to elevated carbon dioxide were nonsignificant in 1996. A significant CO2 treatment effect also was detected when means of Rubisco activity, soluble protein and Rubisco protein for wheat flag leaves were combined over harvests and years. These three flag leaf parameters were not significantly different in the 350 and 525 mol mol–1 CO2 treatments but were decreased during growth in 700 mol mol–1 CO2 relative to the other two CO2 treatments. Ratios of photosynthesis at 700 and 350 mol mol–1 were compared to ratios of Rubisco activity at 700 and 350 mol mol–1 using wheat flag leaf data from 1995 and 1996. Regression analysis of these data were linear [y = 0.586 + 1.103t x (r2 = 0.432)] and were significant at P 0.05. This result indicated that photosynthetic acclimation was positively correlated with changes of initial Rubisco activity in wheat flag leaves in response to CO2 enrichment. Effects of elevated CO2 on wheat leaf proteins during 1995 and 1996 and on barley during 1995 were consistent with an acceleration of senescence.  相似文献

Adjustment in leaf area : mass ratio called leaf area ratio (LAR) is one of the strategies to optimize photon harvesting. LAR was recorded for 10 genotypes of Hevea brasiliensis under high irradiance and low temperature and the genotypes were categorized into two groups, i.e. high LAR and low LAR types. Simultaneously, the growth during summer as well as winter periods, photosynthetic characteristics, and in-vitro oxidative damage were studied. Low LAR (19.86±0.52 m2 kg–1) types, recorded an average of 18.0 % chlorophyll (Chl) degradation under high irradiance and 7.1 % Chl degradation under low temperature. These genotypes maintained significantly higher net photosynthetic rate (P N) of 10.4 mol(CO2) m–2 s–1 during winter season. On the contrary, the high LAR (24.33±0.27 m2 kg–1) types recorded significantly lower P N of 4 mol(CO2) m–2 s–1 and greater Chl degradation of 37.7 and 13.9 % under high irradiance and low temperature stress, respectively. Thus LAR may be one of the physiological traits, which are possibly involved in plant acclimation process under both stresses studied.  相似文献

Structural, functional, and biochemical characteristics of the photosynthetic apparatus of a nemoral herbaceous perennial plant Ajuga reptansL. inhabiting the middle taiga subzone were investigated. Plant leaves were characterized by a high content of green (3.1 mg/dm2) and yellow (0.64 mg/dm2) pigments and contained moderate-sized chloroplasts with grana consisting of ten thylakoids or more. The maximum rate of photosynthesis in summergreen leaves (5–8 mg CO2/(dm2h)) was observed at 14–16°C under a saturating photosynthetically active radiation of 50 W/m2. At 6–7°C, the rate of CO2assimilation was reduced to 60–80% of the maximum one. The temperature optimum of photosynthesis was not constant and shifted by 2–6°C depending on the changes in the ambient temperature. Wintergreen leaves were capable of photosynthesis in late autumn after heavy freezes and in early spring after a long winter. The accumulation of soluble carbohydrates and free amino acids in leaves helps to maintain the functional activity of the photosynthetic apparatus.  相似文献

Dry weight and Relative Growth Rate of Lemna gibba were significantly increased by CO2 enrichment up to 6000 l CO2 l–1. This high CO2 optimum for growth is probably due to the presence of nonfunctional stomata. The response to high CO2 was less or absent following four days growth in 2% O2. The Leaf Area Ratio decreased in response to CO2 enrichment as a result of an increase in dry weight per frond. Photosynthetic rate was increased by CO2 enrichment up to 1500 l CO2 l–1 during measurement, showing only small increases with further CO2 enrichment up to 5000 l CO2 l–1 at a photon flux density of 210 mol m–2 s–1 and small decreases at 2000 mol m–1 s–1. The actual rate of photosynthesis of those plants cultivated at high CO2 levels, however, was less than the air grown plants. The response of photosynthesis to O2 indicated that the enhancement of growth and photosynthesis by CO2 enrichment was a result of decreased photorespiration. Plants cultivated in low O2 produced abnormal morphological features and after a short time showed a reduction in growth.  相似文献

Photosynthetic-induction response and light-fleck utilization were investigated for the current-year seedlings of Quercus serrata, a deciduous tree found in temperate regions of Japan. The tree seedlings were grown under three light regimes: a constant low photosynthetic photon flux density (PFD) regime of 50 mol m–2 s–1, a constant high PFD regime of 500 mol m–2 s–1, and a lightfleck regime with alternated low (lasting 5 s) and high (lasting 35 s) PFD. The photosynthetic-induction response following a sudden increase of PFD from 50 to 500 mol m–2 s–1 exhibited two phases: an initial fast increase complete within 3–5 s, and a second slow increase lasting for 15–20 min. Induction times required to reach 50% and 90% of steady-state assimilation rates were significantly shorter in leaves from the constant low PFD than those from the high PFD regime. During the first 60–100 s, the ratio of observed integrated CO2 uptake to that predicted by assuming that a steady-state assimilation would be achieved instantaneously after the light increase was significantly higher for leaves from the low PFD regime than from the high PFD regime. Lightfleck utilization was examined for various durations of PFD of 500 mol m–2 s–1 on a background PFD of 50 mol m–2 s–1. Lightfleck utilization efficiency was significantly higher in low PFD leaves than in the high PFD leaves for 5-s and 10-s lightflecks, but showed no difference among different light regimes for 100-s lightflecks. The contribution of post-illumination CO2 fixation to total carbon gain decreased markedly with increasing lightfleck durations, but exhibited no significant difference among growth regimes. Photosynthetic performances of induction response and lightfleck utilization in leaves from the lightfleck regime were more similar to those in leaves from the low PFD regime. It may be the total daily PFD rather than PFD dynamics in light regimes that affects the characteristics of transient photosynthesis in Q. serrata seedlings.  相似文献

In Evernia prunastri, photosynthetic gas exchange was saturated with yellow radiation (SOX) at 400 mol m–2s–1, and then red (R), far-red (FR), or blue (B) radiations at irradiance of 15 mol m–2s–1 were added. Because of photosynthesis saturation, any stimulation or decay in CO2 assimilation by any radiation quality could be attributed to the involvement of a non-photosynthetic photoreceptor. Thus CO2 assimilation, effective quantum yield, and photochemical quenching were enhanced when R was included, and decreased with FR. Blue radiation completely abolished CO2 fixation. Hence different spectral radiation qualities may activate non-photosynthetic photoreceptors such as phytochrome and blue photoreceptors, which are involved in regulating the photosynthetic activity in E. prunastri.  相似文献

The responses to irradiance of photosynthetic CO2 assimilation and photosystem 2 (PS2) electron transport were simultaneously studied by gas exchange and chlorophyll (Chl) fluorescence measurement in two-year-old apple tree leaves (Malus pumila Mill. cv. Tengmu No.1/Malus hupehensis Rehd). Net photosynthetic rate (P N) was saturated at photosynthetic photon flux density (PPFD) 600-1 100 (mol m-2 s-1, while the PS2 non-cyclic electron transport (P-rate) showed a maximum at PPFD 800 mol m-2 s-1. With PPFD increasing, either leaf potential photosynthetic CO2 assimilation activity (Fd/Fs) and PS2 maximal photochemical activity (Fv/Fm) decreased or the ratio of the inactive PS2 reaction centres (RC) [(Fi – Fo)/(Fm – Fo)] and the slow relaxing non-photochemical Chl fluorescence quenching (qs) increased from PPFD 1 200 mol m-2 s-1, but cyclic electron transport around photosystem 1 (RFp), irradiance induced PS2 RC closure [(Fs – Fo)/Fm – Fo)], and the fast and medium relaxing non-photochemical Chl fluorescence quenching (qf and qm) increased remarkably from PPFD 900 (mol m-2 s-1. Hence leaf photosynthesis of young apple leaves saturated at PPFD 800 mol m-2 s-1 and photoinhibition occurred above PPFD 900 mol m-2 s-1. During the photoinhibition at different irradiances, young apple tree leaves could dissipate excess photons mainly by energy quenching and state transition mechanisms at PPFD 900-1 100 mol m-2 s-1, but photosynthetic apparatus damage was unavoidable from PPFD 1 200 mol m-2 s-1. We propose that Chl fluorescence parameter P-rate is superior to the gas exchange parameter P N and the Chl fluorescence parameter Fv/Fm as a definition of saturation irradiance and photoinhibition of plant leaves.  相似文献

Net photosynthetic rates and mesophyll conductances at 25 °C at light saturation and air levels of carbon dioxide and oxygen were measured on recently fully expanded leaflets of second trifoliolate leaves of soybeans (Glycine max cv. Kent). Plants were grown outdoors in pots at Beltsville, Maryland with 14 planting times from May through August, 1983. Air temperature and humidity, and photosynthetically active radiation (PAR) were measured for the expansion periods of the second trifoliolate leaves. Rates of net photosynthesis ranged from 24 to 33 mol m–2 s–1, and mesophyll conductances from 0.24 to 0.35 cm s–1 for the different planting dates. Mean 24-h air temperatures ranged from 20.6 to 29.0 °C, and mean daily PAR ranged from 29.4 to 58.4 mol m–2 d–1 for the leaf expansion periods. There was a positive relationship between photosynthetic characteristics and PAR during leaf expansion, and a negative relationship between photosynthetic characteristics and leaf expansion rates, with 96% of the variation in photosynthetic characteristics accounted for by these two variables. Leaf expansion rates were highly correlated with air temperature.  相似文献

Past studies of the effects of varying levels of photosynthetic photon flux density (PPFD) on the morphology and physiology of the epiphytic Crassulacean acid metabolism (CAM) plant Tillandsia usneoides L. (Bromeliaceae) have resulted in two important findings: (1) CAM, measured as integrated nocturnal CO2 uptake or as nocturnal increases in tissue acidity, saturates at relatively low PPFD, and (2) this plant does not acclimate to different PPFD levels, these findings require substantiation using photosynthetic responses immediately attributable to different PPFD levels, e.g., O2 evolution, as opposed to the delayed, nocturnal responses (CO2 uptake and acid accumulation). In the present study, instantaneous responses of O2 evolution to PPFD level were measured using plants grown eight weeks at three PPFD (20–45, 200–350, and 750–800 mol m-2s-1) in a growth chamber, and using shoots taken from the exposed upper portions (maximum PPFD of 800 mol m-2s-1) and shaded lower portions (maximum PPFD of 140 mol m-2s-1) of plants grown ten years in a greenhouse. In addition, nocturnal increases in acidity were measured in the growth chamber plants. Regardless of the PPFD levels during growth, O2 evolution rates saturated around 500 mol m-2s-1. Furthermore, nocturnal increases in tissue acidity saturated at much lower PPFD. We investigated seasonal patterns of photosynthetic responses to CO2 concentrations in Spartina alterniflora Loisel, an aerenchymous halophyte grass, from a salt marsh of the Bay of Fundy (NB, Canada), and from plants grown from rhizome in controlled-environment chambers. From late May to August, CO2 compensation concentrations () of field-grown leaves varied between 2.5–10.7 cm3(CO2) m–3, with a mean of 5.4 cm3(CO2) m–3. From September onwards field leaves showed CO2 compensation concentrations from 6.6–21.1 cm3(CO2) m–3, with a mean of 13.1 cm3 m–3 well into the C3–C4 intermediate range. The seasonal variability in did not result from changing respiration, but rather from a sigmoidal response of net photosynthetic rate (P N) to applied CO2 concentration, found in all tested leaves but which became more pronounced late in the season. One explanation for the sigmoidal response of P N to external CO2 concentration could be internal delivery of CO2 from roots and rhizomes to bundle sheath cells via the aerenchyma, but the sigmoidal responses in S. alterniflora persisted out to the tips of leaves, while the aerenchyma extend only to mid-leaf. The sigmoidicity persisted when CO2 response curves were measured from low to high CO2, or from high to low CO2, and even when prolonged acclimation times were used at each CO2 concentration.  相似文献

Transgenic tobacco (Nicotiana tabacum L. cv. W38) with an antisense gene directed against the mRNA of the ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) small subunit was used to determine the kinetic properties of Rubisco in vivo. The leaves of these plants contained only 34% as much Rubisco as those of the wild type, but other photosynthetic components were not significantly affected. Consequently, the rate of CO2 assimilation by the antisense plants was limited by Rubisco activity over a wide range of CO2 partial pressures. Unlike in the wild-type leaves, where the rate of regeneration of ribulose bisphosphate limited CO2 assimilation at intercellular partial pressures above 400 ubar, photosynthesis in the leaves of the antisense plants responded hyperbolically to CO2, allowing the kinetic parameters of Rubisco in vivo to be inferred. We calculated a maximal catalytic turnover rate, kcat, of 3.5+0.2 mol CO2·(mol sites)–1·s–1 at 25° C in vivo. By comparison, we measured a value of 2.9 mol CO2·(mol sites)–1·–1 in vitro with leaf extracts. To estimate the Michaelis-Menten constants for CO2 and O2, the rate of CO2 assimilation was measured at 25° C at different intercellular partial pressures of CO2 and O2. These measurements were combined with carbon-isotope analysis (13C/12C) of CO2 in the air passing over the leaf to estimate the conductance for transfer of CO2 from the substomatal cavities to the sites of carboxylation (0.3 mol·m–2·s–1·bar–1) and thus the partial pressure of CO2 at the sites of carboxylation. The calculated Michaelis-Menten constants for CO2 and O2 were 259 ±57 bar (8.6±1.9M) and 179 mbar (226 M), respectively, and the effective Michaelis-Menten constant for CO2 in 200 mbar O2 was 549 bar (18.3 M). From measurements of the photocompensation point (* = 38.6 ubar) we estimated Rubisco's relative specificity for CO2, as opposed to O2 to be 97.5 in vivo.
