This study examined whether or not the reproductive response of female sheep to photoperiod varies with seasonal reproductive state. The specific objective was to test the hypothesis that the reproductive response to a long-day pattern of melatonin varies with the reproductive state of the ewe. The response examined was the synchronization of reproductive neuroendocrine induction (rise in serum luteinizing hormone, or LH) following nocturnal infusion of melatonin into pinealectomized ewes for 35 consecutive nights. This infusion restored a pattern of circulating melatonin similar to that in pineal-intact ewes maintained in a long photoperiod (LD 16:8). The ewes had been pinealectomized and without melatonin replacement for 16-25 months prior to the study. They were in differing reproductive states at the start of the infusion, as their endogenous reproductive rhythm had become desynchronized among individuals and with respect to time of year. Noninfused pinealectomized ewes served as controls. Regardless of the reproductive state at the start of the 35-day infusion of the long-day pattern of melatonin, all treated ewes exhibited the same reproductive neuroendocrine response after the infusion was ended. This consisted of a synchronized rise in LH some 6-8 weeks after the infusion was terminated, the maintenance of a high level of serum LH for some 15 weeks, and a subsequent precipitous fall in LH to a very low level. These results provide evidence that a long-day pattern of melatonin can synchronize reproductive neuroendocrine induction in the ewe, regardless of reproductive condition, and thus do not support the hypothesis that this response differs with seasonal reproductive state.  相似文献

Two experiments were conducted to examine the effects of treating seasonally anoestrous ewes with melatonin before ram introduction on reproductive response, and on LH secretion in anoestrous ewes induced to ovulate by rams.In Experiment 1, a total of 667 ewes from three flocks involving Merino (Flock 1, N = 149), Merino entrefino (Flock 2, N = 325) and Rasa Aragonesa (Flock 3, N = 203) breeds were used. Within each flock, ewes isolated from rams since the previous lambing were assigned at random to receive melatonin implants of Regulin (75, 175 and 105 in Merino, Merino entrefino and Rasa Aragonesa flocks, respectively) or to serve as untreated controls (74 in Merino, 150 in Merino entrefino and 98 in Rasa Aragonesa flocks). Fertile rams were introduced into all flocks 5 weeks after implantation in March (Flocks 1 and 2) or April (Flock 3), and remained with the ewes for a 50 day mating period. Percentage of ewes with luteal activity at ram introduction did not differ between melatonin treated and control ewes in any flock. There were no significant differences in either the mean interval from ram introduction to lambing or the distribution of lambing. Implantation with melatonin resulted in an improvement of prolificacy in all three flocks, although this only reached statistical significance in the Merino flock (1.15 vs. 1.03 in treated and control ewes, respectively, P < 0.05). Fertility was increased significantly (P < 0.05) in the Merino entrefino flock (64.5% in treated vs. 51.3% in control ewes).In Experiment 2, two trials were undertaken utilizing a total of 63 ewes. Trial 1 involved 24 mature Manchega ewes and Trial 2 involved 39 Merino ewe lambs. Half of the animals in each trial received a Regulin implant on 28 February (Trial 1) or 12 March (Trial 2) and the remaining half acted as controls. Rams were introduced 5 weeks after implantation and remained with the ewes for a 25 day period. In both trials, anoestrous ewes at ram introduction were bled at 20 min intervals for 3 h before and 5 h after ram introduction and then at 3 h intervals over the next 24 h for assessment of plasma concentrations of LH. Secretion of LH before or following introduction of rams was not affected by melatonin. Both treated and control anoestrous ewes in each trial responded to introduction of rams with an increase in the frequency of the LH pulses (P < 0.05), but no significant changes were detected in pulse amplitude or mean levels of LH. A preovulatory surge of LH was detected between 8 and 26 h after ram introduction, but neither mean interval from ram introduction to the peak of LH surge, nor the magnitude of the LH peak, was influenced by melatonin treatment.Results from this study show that: (1) melatonin implants administered during early seasonal anoestrus have the potential to improve reproductive performance in Spanish breeds of sheep, but the response is conditioned by breed, management system and environmental factors; (2) melatonin did not modify the secretion of LH in anoestrous ewes induced to ovulate by the ram effect under our experimental conditions.  相似文献

The pineal gland, through its nocturnal melatonin secretion, mediates the effects of inhibitory (long) and stimulatory (short) photoperiod on reproduction in female sheep. Earlier studies revealed that duration of the nighttime melatonin rise is important in determining the inhibitory effect of day length on reproduction in the ewe. The present study tested whether the duration is also important in mediating the inductive response to short days. Pinealectomized ewes, housed under long days, received a short-day melatonin infusion (16-h duration) for 90 days. Reproductive status was monitored from the response to estradiol negative feedback of luteinizing hormone (LH) secretion. This short-day melatonin pattern led to unambiguous reproductive induction, despite the exposure to inhibitory long days. The increase in serum LH was comparable, in terms of latency and magnitude, to that in pinealectomized controls receiving the same short-day melatonin pattern under short days, and in pineal-intact controls transferred from long to short days. Since the reproductive status conformed to the length of time that melatonin was elevated each day rather than to photoperiod, these results support the conclusion that duration of the nighttime melatonin rise mediates the reproductive response of the ewe to an inductive photoperiod. In all, the melatonin rhythm is considered an integral component of the physiologic mechanism measuring day length; through duration of its nocturnal secretion, melatonin encodes both inhibitory and stimulatory photoperiods.  相似文献

The present study tested the hypothesis that the nocturnal melatonin rhythm in the fetal sheep results from transfer across the placenta of melatonin from maternal circulation. Pregnant ewes were exposed to an artificial reverse photoperiod at about 100 days gestation (n = 6; lights on 10 h, 2200-0800 h PST). This treatment tested for entrainment in the ewe and its fetus of the 24-h pattern of melatonin production from the pineal gland. Other ewes were pinealectomized at 55 days post-breeding (n = 6), and similarly treated. Catheters were implanted and blood samples were collected between 117 and 142 days gestation at two 48-h periods, about every 0.5-4 h, to assess the pattern of melatonin in maternal and fetal circulations. In pineal-intact ewes and their fetuses, melatonin rhythms conformed to the reverse photoperiod, i.e. plasma melatonin concentrations were relatively low during the light period and significantly increased for the duration of darkness. In contrast, maternal pinealectomy abolished the melatonin rhythms in both the ewe and fetus; melatonin concentrations remained at or below the limits of detection. Pineal-intact sheep gave birth about 139 +/- 2 days (mean +/- SE, n = 4) at 1915 +/- 0.7 h and pinealectomized ewes (n = 5 of 6) lambed at 149 +/- 2 days at 0424 +/- 0.5 h. Finally, in lambs (n = 3) born to pinealectomized ewes, typical melatonin rhythms were present within the first week of life. The findings indicate that the maternal pineal gland is responsible for the 24-h pattern of melatonin in the ewe and its fetus during the last trimester of pregnancy.  相似文献

Although many species display endogenous circannual rhythms of biological activity that are synchronized by day length, the specific photoperiodic requirements for synchronizing such rhythms are not established for any species. We tested the hypothesis that the circannual reproductive rhythm of sheep can be synchronized by exposure to just one or two discrete blocks of photoperiodic information each year. Ewes were pinealectomized to prevent their ability to transduce photoperiodic information into altered reproductive neuroendocrine activity. During the 53/4 yr following pinealectomy, specific photoperiodic signals were restored for discrete periods of time via replacement of 24-h patterns of melatonin, the pineal hormone that transmits photic information to the reproductive neuroendocrine axis. The ewes were kept in a 12-mo photoycycle that alternated between short (8L:16D) and long (16L:8D) days every 6 mo and that was 6 mo out of phase with the geophysical year. Pineal-intact control ewes exhibited synchronous annual reproductive cycles. Noninfused pinealectomized control ewes did not exhibit synchronous cycles. Pinealectomized ewes infused with alternating 70-day blocks of short- and long-day patterns of melatonin every 6 mo for the first 21/2 yr of the experiment exhibited synchronous annual reproductive cycles that were 6 mo out of phase with those of ewes maintained outdoors. This synchrony persisted when the frequency of the melatonin treatment was reduced to just one 70-day block of a long-day pattern of melatonin each 365 days. Cycle period was 368 +/- 3 days; standard deviation of the date of onset of reproductive induction averaged only 3 days. Our study provides the first direct evidence that a single block of photoperiodic information a year can synchronize a circannual rhythm.  相似文献

The pineal controls the reproductive response of ewes to both stimulatory (short) and inhibitory (long) day lengths. Melatonin, a pineal hormone whose nocturnal secretion is entrained by photoperiod, mediates the effect of stimulatory photoperiod. We now report that melatonin also mediates the effect of inhibitory day length, monitored as response to estradiol negative feedback on luteinizing hormone (LH) secretion. Ovariectomized, estradiol-implanted ewes were pinealectomized and intravenously infused with melatonin to restore the nightly melatonin rise. Following transfer from short to long days, and a concurrent switch from short- to long-day melatonin patterns, LH dropped precipitously in pinealectomized ewes, matching the photoinhibitory response of pineal intact controls. LH dropped similarly in pinealectomized ewes when long-day melatonin was infused under short days. Pinealectomized ewes transferred from long to short days displayed a marked LH rise, provided melatonin was also switched to the short-day pattern. LH remained suppressed if long-day melatonin was infused following transfer to short days. These data indicate the nighttime melatonin rise mediates reproductive responses to inhibitory, as well as stimulatory photoperiods; they further suggest the duration of this rise controls suppression of LH under long days. Rather than being strictly pro- or antigonadal, the pineal participates in measuring day length.  相似文献

An endogenous circannual rhythm drives the seasonal reproductive cycle of a broad spectrum of species. This rhythm is synchronized to the seasons (i.e., entrained) by photoperiod, which acts by regulating the circadian pattern of melatonin secretion from the pineal gland. Prior work has revealed that melatonin patterns secreted in spring/summer entrain the circannual rhythm of reproductive neuroendocrine activity in sheep, whereas secretions in winter do not. The goal of this study was to determine if inability of the winter-melatonin pattern to entrain the rhythm is due to the specific melatonin pattern secreted in winter or to the stage of the circannual rhythm at that time of year. Either a summer- or a winter-melatonin pattern was infused for 70 days into pinealectomized ewes, centered around the summer solstice, when an effective stimulus readily entrains the rhythm. The ewes were ovariectomized and treated with constant-release estradiol implants, and circannual cycles of reproductive neuroendocrine activity were monitored by serum LH concentrations. Only the summer-melatonin pattern entrained the circannual reproductive rhythm. The inability of the winter pattern to do so indicates that the mere presence of a circadian melatonin pattern, in itself, is insufficient for entrainment. Rather, the characteristics of the melatonin pattern, in particular a pattern that mimics the photoperiodic signals of summer, determines entrainment of the circannual rhythm of reproductive neuroendocrine activity in the ewe.  相似文献

Fifteen pinealectomized and 15 unoperated ewes were exposed to constant light for 3 weeks before and 10 weeks after lambing. Fourteen pinealectomized and 15 unoperated ewes were allowed to lamb outdoors. Five ewe lambs born in constant light to the 2 groups of dams were pinealectomized at 10 weeks of age. Ewes and lambs were then returned to the field. Puberty (determined by weekly progesterone analysis) was significantly delayed (P less than 0.05) in the pinealectomized ewe lambs. Median pubertal age in pineal-intact ewe lambs was 37 weeks compared to 49 weeks in pinealectomized lambs. Constant light during the first 10 weeks of life had no effect upon puberty onset nor did the pineal status of the dam. Control lambs entered seasonal anoestrus at the time pinealectomized ewe lambs were entering puberty. Pinealectomized lambs entered anoestrus at the same time as control lambs were beginning their second breeding season. These results confirm a key role of pineal-mediated hormonal signals in the control of puberty in the sheep.  相似文献

Experiments were conducted to examine whether the refractoriness of the Suffolk ewe to the reproductive effects of day length is associated with a deficit in the generation of the circadian rhythm of melatonin secretion or in the postpineal processing of this photoperiodic message. Using serum luteinizing hormone (LH) concentrations in ovariectomized ewes bearing constant-release estradiol implants as a marker of reproductive induction, ewes with intact pineal glands were found to become unresponsive to fixed artificial photoperiods that initially had been either inductive (short days) or inhibitory (long days). The loss of the photoperiodic response was not associated with notable changes in the 24-h secretory pattern of melatonin, which remained characteristically low throughout the day and rose at night. In pinealectomized ewes, nightly infusion of a stimulatory pattern of melatonin (simulating that seen on short days) initially provoked reproductive induction; this response then lessened over much the same time course that pineal intact ewes became refractory to short days. These results support the hypothesis that photorefractoriness reflects a deficit in the postpineal processing of the photoperiodic message. Further, in view of recent evidence that photorefractoriness normally leads to both onset and cessation of the breeding season in Suffolk ewes maintained outdoors, these findings suggest that the loss of response to the melatonin signal contributes to at least one of these reproductive transitions, the cessation of the breeding season, under natural environmental conditions.  相似文献

The beneficial effects of nutrition on reproduction in sheep have been described, particularly on ovulation rate. However, the relationships between nutrition and reproductive seasonality are not well known. This review will deal with the effects of body fat or food intake on sexual and hypothalamic/pituitary activity in sheep, mainly focused on Mediterranean genotypes. Although only severe malnutrition can significantly extend the length of the seasonal anestrous period, the level of fat reserves can play a significant role on reproductive seasonality delaying the onset of seasonal anoestrus, particularly on the Mediterranean environment. The effect of overfeeding on LH secretion has also been reported, specially at short term. Several experimental approaches have elucidated that both high body fat and food intake are able to modify the sensitivity of the hypothalamus to oestradiol negative feedback during seasonal anoestrus, with those effects being associated to a reduced amount of NPY mRNA and to an increase of plasma insulin, glucose and leptin concentrations, particularly in the late scenario. However, the highest receptivity to nutritional stimulation in terms of increasing LH occurs when ewes are subjected to a photoperiodic state of early anoestrus or late breeding season rather than under a photoperiod characteristic of the end of anoestrus or the beginning of the breeding season.  相似文献

The role of endogenous opioids and nutrition on the inhibition of luteinizing hormone (LH) secretion during the postpartum period was investigated in a Spanish breed of sheep lambing in the mid-late breeding season. Two groups of adult Rasa Aragonesa ewes housed in individual pens and lambing on 30 December were fed during the suckling period to provide maintenance requirements and the production of 1.1 (M; n=8) or 0.55 (L; n=8) kg of milk per day. On days 10, 20 and 30 after lambing, the effect of a treatment with the opiate receptor antagonist naloxone (1 mg/kg at four hourly intervals) on LH secretion was assessed in half of the ewes of each group, the remaining females receiving four saline injections. After weaning, animals were fed to provide requirements for maintenance of liveweight. Blood samples were collected twice a week from day 20 postpartum until the end of March, and assayed for progesterone and prolactin. Although underfed ewes showed significantly lower mean plasma concentrations during the control period on day 20 postpartum, nutrition did not seem to modify LH secretion before naloxone or saline injections. Moreover, no differences between nutritional groups in the response to naloxone injections on pattern of LH secretion were found. In fact, naloxone treatment induced an increase of mean LH concentrations on days 10, 20 and 30 postpartum (at least, P<0.05), of LH pulse frequency on days 20 and 30 (P<0.05), and of LH pulse amplitude on days 10 and 20 (P<0.05). Underfed ewes during the postpartum period showed a slower decline in plasma prolactin levels, with significant differences on days 29, 36 and 39 after lambing (P<0.05). Only 3 M ewes ovulated before the onset of the seasonal anoestrus period. It is concluded that endogenous opioids are involved in the inhibition of LH secretion during the early suckling period of a reduced seasonality breed of sheep without any influence of nutrition on the response to naloxone treatment; however, ewes underfed before weaning failed to reactivate their cyclicity prior to the onset of the seasonal anoestrus.  相似文献

Circulating patterns of luteinizing hormone (LH) and prolactin (PRL) were monitored for 5 yr in ewes maintained either outdoors in natural conditions or indoors in a fixed, short photoperiod (8L:16D). The ewes were ovariectomized and each was treated with a Silastic implant containing estradiol to provide a fixed negative feedback signal to the reproductive neuroendocrine axis. Serum concentrations of LH and PRL were subjected to a statistical algorithm developed for the purpose of detecting hormone cycles. In ewes maintained outdoors, serum concentrations of both hormones underwent high amplitude cycles with a period no different from 365 days. Among ewes maintained in the fixed photoperiod, unambiguous cycles of LH and PRL persisted through the 5 yr of exposure to short days. Period of these cycles differed from 365 days. Further, the LH cycles became desynchronized among ewes housed together and desynchronized with respect to the LH cycles in ewes kept outdoors. These findings document the existence of an endogenous circannual rhythm of reproductive neuroendocrine function in ewes.  相似文献

The reproductive performance (fertility, prolificacy and fecundity of totally confined sheep in a controlled breeding program at the Animal Research Centre (ARC) was analyzed to evaluate the effect of pregnant mares' serum gonadotrophin (PMSG) on ewes synchronized for estrus with fluorogestone acetate (FGA). PMSG did not significantly enhance reproductive performance. Fertility for ewes receiving 500 IU PMSG was 57% and for ewes not receiving PMSG it was 47%. Corresponding values for prolificacy were 2.4 and 2.2, and for fecundity, 144 and 103%. The reproductive performance was not affected by any of the other factors (flock, strain, lighting regime, ram age or reproductive status of the ewe) indigenous to the controlled breeding program for sheep housed in total confinement.  相似文献

Exposure of anoestrous ewes to rams induces an increase in LH secretion, eventually leading to ovulation. This technique therefore is an effective, low-cost and hormone-free way of mating sheep outside the breeding season. However, the use of this technique is limited by the variability of the ewes’ responses. In this study, our objective was to understand more completely the origins of this variability and to determine the relative roles of breed, the point in time during anoestrus and the depth of anoestrus on the response to the ‘ram effect’. In the first experiment, the pattern of anoestrus on the basis of the concentration of progesterone determined weekly, was determined in four breeds including two less seasonal (Mérinos d'Arles and Romane), one highly seasonal (Mouton Vendéen) and one intermediate (Île-de-France) breeds. Anoestrus was longer and deeper in Mouton Vendéen and Île-de-France than in Romane or Mérinos d'Arles. In the second experiment, we used the same four breeds and tested their hypophyseal response to a challenge with a single dose of 75 ng gonadotrophin-releasing hormone (GnRH) in early, mid and late anoestrus, and then we examined their endocrine and ovarian responses to the ‘ram effect’. Most (97%) ewes responded to GnRH and most (93%) showed a short-term increase in LH pulsatility following the ‘ram effect’. The responses in both cases were higher in females that went on to ovulate, suggesting that the magnitude of the hypophyseal response to a GnRH challenge could be a predictor of the response to the ‘ram effect’. As previously observed, the best ovarian response was in Mérinos d'Arles at the end of anoestrus. However, there was no relationship between the proportion of females in the flock showing spontaneous ovulation and the response to the ‘ram effect’ of anoestrous ewes from the same flock.  相似文献

The levels of plasma LH and FSH were measured in serial blood samples taken at 15-min intervals for 6 h from ewes that had remained fertile after grazing oestrogenic pasture (clover-fertile ewes), from ewes that were permanently affected by clover disease (clover-infertile ewes) and from normal ewes. Two flocks of ewes from different locations were studied. In flock 1, tonic LH secretion (total area under the curve of LH concentration versus time, 1 area unit = 1 ng ml-1 x 1 h) was significantly (P < 0.05) greater in clover-infertile ewes (10.4 area units) during anoestrus than in ewes that had remained fertile after prolonged grazing of oestrogenic clover (5.4 area units). Tonic LH and FSH secretions during the bleeding season and FSH secretion during anoestrus were not significantly different. In flock 2, LH levels during the breeding season were significantly (P < 0.05) elevated in clover-infertile ewes (10.9 area units) compared to normal ewes (5.4 area units) that had never grazed oestrogenic clover. LH secretion in clover-infertile ewes (7.8 area units) was intermediate to that found in infertile and control ewes. Concentrations of FSH, progesterone and ovarian vein oestradiol-17 beta (E2) during the breeding season were similar in the three groups. In another experiment, the positive feedback release of LH following administration of E2 (12.5, 25 or 50 micrograms per ewe) was measured in anoestrous ewes of flock 2. Significantly (P < 0.01) more clover-infertile ewes demonstrated a positive feedback effect than control ewes when given 12.5 micrograms E2 but not when given higher doses. The elevation of LH secretion in permanently affected clover-infertile ewes is inconsistent with the hypothesis that the hypothalamo-pituitary axis of these ewes is less responsive to the negative feedback effect of oestrogen. Furthermore, the patency of the positive feedback loop is consistent with the ability to ovulate.  相似文献

Reproductive responses to photoperiod were directly compared in mature ewes and in their spring-born twin female lambs. All females were ovariectomized and treated with oestradiol implants before transfer into artificial photoperiod; serum LH concentrations and pulsatile LH patterns provided an index of neuroendocrine reproductive activity. Mothers were transferred from natural photoperiod to artificial long days (16 h light:8 h dark) at the summer solstice so that no decrease in photoperiod would be experienced. These ewes began reproductive activity synchronously at the expected time in the autumn. One of each pair of twin lambs was treated exactly as the mothers; to determine the normal timing of puberty the remaining twin was maintained in a photoperiod simulating the natural decrease in daylength. In all 6 control lambs experiencing the simulated natural photoperiod, reproductive activity occurred synchronously at 28 +/- 1 weeks of age (2 October +/- 7 days). However, in their twin sisters which did not experience a decrease in photoperiod, only 2 of 6 lambs had begun reproductive activity by the end of the experiment at 52 weeks of age (March), and these were both delayed relative to their twin control lambs exposed to decreasing daylength. Therefore, a decrease in photoperiod is necessary for the normal timing of puberty in the spring-born, female sheep, whereas seasonally anoestrous, mature sheep can enter the breeding season at a normal time in the absence of decreasing photoperiod. We suggest that the requirement for a decreasing photoperiod by the spring-born lamb reflects its limited photoperiodic history as compared to the adult.  相似文献

Under photoperiod regulated conditions, some Pelibuey ewes appear to express estrous cycles continuously throughout the year. Ovulatory activity of Suffolk ewes has not being determined at 19 degrees N. The aim of the present study was to describe the annual reproductive cycle of Pelibuey and Suffolk ewes under natural conditions at 19 degrees N. Ten adult ewes of each breed were maintained under natural environmental conditions, on a constant plane of nutrition and isolated from rams. The ovulatory activity of both breeds was monitored over 2 years (2003 and 2004) by quantifying progesterone concentrations in blood samples taken twice a week. For both breeds, the proportion of ovulating ewes per month and year was analyzed by chi(2). The duration of anestrus was compared between breeds and years by analysis of variance. Correlation of ambient temperature and weight changes with the ovulatory activity was evaluated by regression procedure. Ovulatory activity was different (P<0.05) between breeds from February to July. Pelibuey sheep (60%, 6 of 10) ovulated continuously during the entire study; one animal was eliminated in the second year, due to ovarian follicular cysts and three ewes had anestrous periods from 65+/-46 to 70+/-36 days. Suffolk ewes displayed reproductive seasonality similar to that observed in sheep of the same breed in latitudes greater than 35 degrees . Within breed, reproductive behavior of sheep was similar between years, with the only difference (P<0.05) observed in February for Pelibuey and in July for Suffolk relative to other months. Ambient temperature was not correlated with ovulatory activity. The body weight of Pelibuey ewes was constant during the entire study and was not correlated with ovulatory activity. There was, however, an increase on the body weight of Suffolk ewes, but this variation was not related to ovulatory activity. These findings lead to the conclusion that at 19 degrees N, a high proportion of Pelibuey ewes is capable of ovulating throughout the year, whereas Suffolk ewes undergo seasonal anestrous periods similar to those observed in individuals of the same breed at latitudes greater than 35 degrees . Identification of continuous ovulatory Pelibuey ewes may be the first step for developing a selection program against seasonality to increase lambing frequency.  相似文献

Oestrous cycles were induced in seasonally anoestrous ewes by introducing rams into the flock and giving to the ewes one intramuscular injection of 20 mg progesterone. At the second ovulation the onset of oestrus and the preovulatory surge of luteinizing hormone (LH) were recorded. It was found that the LH surge began in significantly more ewes during the night (79%) than during the day (21%). The onset of oestrus tended to follow a similar pattern. This temporal pattern was not related to the time of ram introduction, but may be the result of daily rhythms in ovarian activity. The objective of this study was to characterize follicular development, onset of oestrus and preovulatory LH surge, and in vivo embryo yields of sheep superovulated after treatment with a single dose of 1.5mg of GnRH antagonist (GnRHa). At first FSH dose, ewes treated with GnRH antagonist (n=12) showed a higher number of gonadotrophin-responsive follicles, 2-3mm, than control ewes (n=9, 13.5+/-3.8 versus 5.3+/-0.3, P<0.05). Administration of FSH increased the number of >or=4mm follicles at sponge removal in both groups (19.3+/-3.8, P<0.0005 for treated ewes and 12.7+/-5.4, P<0.01 for controls). Thereafter, a 25% of the GnRHa-treated sheep did not show oestrous behaviour whilst none control sheep failed (P=0.06). The preovulatory LH surge was detected in an 88.9% of control ewes and 66.7% of GnRHa-treated sheep. A 77.8% of control females showed ovulation with a mean of 9.6+/-0.9 CL and 3.3+/-0.7 viable embryos, while ewes treated with GnRHa and showing an LH surge exhibited a bimodal distribution of response; 50% showed no ovulatory response and 50% superovulated with a mean of 12.2+/-1.1 CL and 7.3+/-1.1 viable embryos. In conclusion, a single dose of GnRHa enhances the number of gonadotrophin-dependent follicles able to grow to preovulatory sizes in response to an FSH supply. However, LH secretion may be altered in some females, which can affect the preovulatory LH surge and/or can weak the terminal maturation of ovulatory follicles.  相似文献

Follicles of various sizes at the surface of the ovary were ablated by electrocautery at the time of cloprostenol-induced luteolysis in ewes and the interval from cloprostenol treatment to the onset of the LH surge determined as an index of the time from luteolysis to ovulation. When follicles 2-4 mm or greater than 4 mm diameter remained in the ovaries, the interval from cloprostenol treatment to the onset of the LH surge was similar to that in sham-operated (control) ewes (55-60 h), whereas when the only follicles remaining were less than 2 mm, the interval was extended by 24 h (P less than 0.05). This study demonstrates that follicles capable of ovulating can be selected from those greater than or equal to 2 mm diameter at luteolysis, emphasizing the flexibility of the sheep ovary in its final selection of the ovulatory follicle.  相似文献
