The influence of immunoregulating humoral thymus factor T-activin (fraction AFT-6) on the activity of xenobiotic metabolism enzymes (EMX) and cellular-dependent cytotoxicity was investigated in germ-free guinea-pigs. Germ-free and conventional animals were injected 5 micrograms of T-activin intraperitoneally, on 3 successive days. The control animals were injected a physiologic saline. A day after the last injection the animals were killed, and EMX and K cell activity was measured. It was found that EMX activity in germ-free animals was decreased. T-activin stimulated cytochrome P-450, NADP-cytochrome-C-reductase, glutathione-S-transferase, benzopyrene hydroxylase and epoxyhydrase activity. In conventional animals the activity of these enzymes remained unchanged under the influence of T-activin. K-cell activity in germ-free animals was decreased, as compared to conventional guinea-pigs. Under the influence of T-activin the parameter increased and stood at about 70% of normal values.  相似文献

Evidence of the impact of air temperature and pressure on cardiovascular morbidity is still quite limited and controversial, and even less is known about the potential influence of geomagnetic activity. The objective of this study was to assess impacts of air temperature, barometric pressure and geomagnetic activity on hospitalizations with myocardial infarctions and brain strokes. We studied 2,833 myocardial infarctions and 1,096 brain strokes registered in two Moscow hospitals between 1992 and 2005. Daily event rates were linked with meteorological and geomagnetic conditions, using generalized linear model with controls for day of the week, seasonal and long-term trends. The number of myocardial infarctions decreased with temperature, displayed a U-shaped relationship with pressure and variations in pressure, and increased with geomagnetic activity. The number of strokes increased with temperature, daily temperature range and geomagnetic activity. Detrimental effects on strokes of low pressure and falling pressure were observed. Relative risks of infarctions and strokes during geomagnetic storms were 1.29 (95 % CI 1.19–1.40) and 1.25 (1.10–1.42), respectively. The number of strokes doubled during cold spells. The influence of barometric pressure on hospitalizations was relatively greater than the influence of geomagnetic activity, and the influence of temperature was greater than the influence of pressure. Brain strokes were more sensitive to inclement weather than myocardial infarctions. This paper provides quantitative estimates of the expected increases in hospital admissions on the worst days and can help to develop preventive health plans for cardiovascular diseases.  相似文献

The influence of phospholipids on the activity of the soluble phosphatidate phosphohydrolase from rat liver was studied. Phosphatidylethanolamine stimulated the enzyme activity whereas phosphatidylglycerol, phosphatidylserine, and phosphatidylinositol were inhibitory. At a phospholipid concentration of 0.7 mg/ml, phosphatidylglycerol inhibited phosphatidate phosphohydrolase activity by 75%, while the enzyme activity was stimulated twofold in the presence of phosphatidylethanolamine. Both lysophosphatidylglycerol and lysophosphatidylethanolamine inhibited phosphatidate phosphohydrolase activity as did octylglucoside, sodium cholate, and Tween 20. The finding that phospholipids influence hepatic phosphatidate phosphohydrolase activity indicates that changes in the lipid environment may modulate the enzyme activity.  相似文献

AIMS: To determine the influence of alternansucrase-derived oligosaccharides (AOS) and other carbohydrates on alpha-galactosidase and alpha-glucosidase activity in Bifidobacterium adolescentis. METHODS AND RESULTS: Activities for alpha-galactosidase and alpha-glucosidase were determined from cell extracts of B. adolescentis grown on 18 test carbohydrates including AOS. alpha-galactosidase activity was enhanced on a variety of alpha-linked or beta-linked carbohydrates regardless of a galactoside or glucoside. alpha-glucosidase, however, was enhanced only on alpha-linked carbohydrates. AOS significantly enhanced enzyme activity compared with most of the carbohydrates tested. Most of the AOS showed significant increases in activity for both enzymes over that displayed by their corresponding acceptor carbohydrates. CONCLUSIONS: alpha-galactosidase may serve as a biomarker for microbial metabolic activity within the large intestine for potential prebiotics composed of alpha-linked or beta-linked oligosaccharides whereas alpha-glucosidase activity may be restricted to assessing the influence of only alpha-linked carbohydrates. The AOS synthesis process provided a value-added component to carbohydrates by increasing metabolic activity (via alpha-galactosidase and alpha-glucosidase) over certain acceptor carbohydrates. SIGNIFICANCE AND IMPACT OF THE STUDY: Fundamental knowledge of enzyme activity in Bifidobacterium may aid in the design of more effective prebiotics and may also help identify enzyme indicators of metabolic activity when assessing influence within the intestine.  相似文献

The cation dye acridine orange (AO) was shown to inhibit ATPase activity of myosin, DTNB-myosin and heavy meromyosin and not to influence that of EDTA-S-1 at low ionic force. The inhibiting effect is concerned with the presence of KCl in solution. The allosteric influence of AO on myosin ATPase activity is discussed and dependence of this effect on charge distribution on the surface of the protein molecule is considered.  相似文献

Effects of angiotensin-converting enzyme (ACE) inhibitors, enalaprilat and imidaprilat, on bradykinin (BK) metabolizing enzymes, aminopeptidase P (APP), neutral endopeptidase (NEP) and carboxypeptidase N (CPN), were examined. APP activity in the mouse lung was inhibited by enalaprilat in a concentration-dependent manner while imidaprilat did not influence the enzyme activity. The inhibitory effects of these ACE inhibitors on the NEP activity in the mouse lung and the CPN activity in the mouse serum were negligible. These data suggested that the influence of enalaprilat on the APP activity and subsequent BK metabolism are different from those of imidaprilat.  相似文献

The influence of KCl and CaCl2 on ATPase activity of ventricular myosin of the mouse, rat, rabbit and cow, the temperature dependence of ATPase and the effect of pCMB treatment and tryptic digestion on ATPase activity of these myosins were studied. 2. Ca2+ - and K+ -ATPase activities of myosins were inversely related to body size of the animal species; when K+ -ATPase activities were measured in the absence of EDTA, the body size/ATPase dependence was only slightly apparent. 3. The influence of temperature, the effect of pCMB and the influence of tryptic digestion on Ca2+ - ATPase activity distinguished the compared myosins. 4. There was a marked alteration of the effect of myosin treatment with pCMB or trypsin on K+ -ATPase activity of these myosins and in this case differences in K+ -ATPase activities were less pronounced.  相似文献

The dependence of the effects of serotonin and cholecystokinin on the activity of peptidases and glycosidases in the intestine of the carp Cyprinus carpio from the content of zinc and copper ions in different photoperiods (6: 18 and 0: 24 h) was studied. It was shown that the activity of enzymes in the intestine of fish under the influence of both hormones in conditions of light deprivation significantly increases, especially under the action of copper. The mechanisms of the influence of these factors on the activity of peptidases and glycosidases of fish intestines are discussed.  相似文献

In order to study the influence of amino group on antioxidant activity of oligosaccharides, an amino disaccharide, 6,6′-diamino-6,6′-dideoxytrehalose (DAMDT) was successfully prepared in this paper, and its antioxidant activities against DPPH, superoxide, hydrogen peroxide, and hydroxyl radicals, and reducing power were evaluated, respectively. The results indicated that DAMDT had better antioxidant activity than trehalose at any tested concentration. The influence of amino group on antioxidant activity of disaccharides is positive based on the results in this paper, and amination should be an effective method to improve the bioactivity of saccharides.  相似文献

The influences of hydrocortisone and thyroxine on the developmental changes of arginase activity in intestine, kidney, and brain of suckling rats were studied. A single injection of hydrocortisone (50 mg/kg) into rats aged 9 days evoked premature increase of jejunal arginase activity due to precocious formation of arginase A4. Arginase A4 can be detected about 48 hr after hydrocortisone injection, whereas in intact rats the enzyme appears in the intestinal mucosa on the 19th-21st days of postnatal life. After hydrocortisone administration to rats aged 6 days, a similar pattern of arginase activity in jejunum was observed. Under the same conditions, the influence of hydrocortisone on kidney arginase was weaker. The hormone did not have any influence on the activity of brain arginase. Daily injection of thyroxine (2 mg/kg) to 6-day-old rats (for 6 consecutive days) caused a precocious increase of the arginase activity in intestine. Under the same conditions, only a slight increase of the arginase activity was observed in kidney, whereas in brain the activity was unaffected.  相似文献

The influence of the freeze-thawing rates on ATP synthetase (ATPase) complex of intact liver mitochondria was investigated. It was shown that the increase in latent ATPase activity and decrease in ATP synthetase activity resulted from an influence on the inner mitochondrial membrane. An increase in freeze-thawing rates led to the preservation of ATP synthetase activity and ATP hydrolysis reduction. Kinetic parameter changes of the ATP synthetase reaction resulted from an insignificant nonspecific increase in the inner mitochondrial membrane permeability and changes in its electrochemical potential level.  相似文献

The influence of supernatants and cell extracts of vaginal lactic acid bacilli on the growth, catalase, antilysozyme and anticomplementary activity of S. epidermidis and E. coli was studied. Changes observed in the persistence characteristics of these microorganisms were found to develop in different directions: in S. epidermidis a decrease in antilysozyme activity was mainly observed, while in E. coli the suppression of anticomplementary activity was registered. The modifying influence of lactic acid flora, revealed in this investigation, is regarded as one of the mechanisms facilitating the formation of microbiocenosis and ensuring colonization resistance.  相似文献

The influence of lipid peroxidation (LPO) inductor H2O2 on spontaneous contractility and electrical activity of the right atrium was studied. LPO induction caused positive inotropic catecholamine-like effect, followed by brady-arrhythmia with the action of potential prolongation, blockade of electrical activity and atrial arrest. Electrical pacing during atrial arrest caused regular contractions of the same force as before electrical activity blockade. The results suggest that the arrest of spontaneous atrial activity under the influence of LPO inductor is due to the impairment of sinus node automaticity.  相似文献

An influence of somatotropin, prolactin and insulin on destructive processes in bovine granulosa cells from small antral follicles following atresia in vivo was studied in vitro. As compared to control, the addition of the studied hormones to serum-free suspension system was shown to result in increase in number of cells without signs of chromosome degeneration after 24 and 48 hrs of incubation. The revealed inhibitory action of somatotropin, prolactin and insulin on chromatin degeneration in granulosa cells was not due to the hormonal influence on proliferative activity of the cells. The stimulatory action of insulin on the viability and estrogen-secretory activity of granulosa cells cultured for 1 day was also found. At the same time, somatotropin and prolactin did not affect the estradiol and progesterone production by the cells. The data obtained suggest that the inhibitory action of somatotropin and prolactin on destructive processes in cultured granulosa cells is not related to the hormonal regulation of the steroidogenic activity of the cells, whereas the similar action of insulin may be partially due to its stimulatory influence on the estradiol secretion.  相似文献

Model compounds of modified hydrophobicity (Eta), hydrophobic moment (mu) and angle subtended by charged residues (Phi) were synthesized to define the general roles of structural motifs of cationic helical peptides for membrane activity and selectivity. The peptide sets were based on a highly hydrophobic, non-selective KLA model peptide with high antimicrobial and hemolytic activity. Variation of the investigated parameters was found to be a suitable method for modifying peptide selectivity towards either neutral or highly negatively charged lipid bilayers. Eta and mu influenced selectivity preferentially via modification of activity on 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphatidylcholine (POPC) bilayers, while the size of the polar/hydrophobic angle affected the activity against 1-palmitoyl-2-oleoylphosphatidyl-DL-glycerol (POPG). The influence of the parameters on the activity determining step was modest in both lipid systems and the activity profiles were the result of the parameters' influence on the second less pronounced permeabilization step. Thus, the activity towards POPC vesicles was determined by the high permeabilizing efficiency, however, changes in the structural parameters preferentially influenced the relatively moderate affinity. In contrast, intensive peptide accumulation via electrostatic interactions was sufficient for the destabilization of highly negatively charged POPG lipid membranes, but changes in the activity profile, as revealed by the modification of Phi, seem to be preferentially caused by variation of the low permeabilizing efficiency. The parameters proved very effective also in modifying antimicrobial and hemolytic activity. However, their influence on cell selectivity was limited. A threshold value of hydrophobicity seems to exist which restricted the activity modifying potential of mu and Phi on both lipid bilayers and cell membranes.  相似文献

From April 1978 to April 1980 the exploratory behaviour of more than 1000 wild wood mice was studied in a modified open field test situation. The influence of sex, sexual condition and age on the exploratory activity was investigated. Sufficient data being available, the exploratory activity of both sexually active and sexually inactive adult males and adult females, of both sexually active and sexually inactive subadult males and subadult females and of both sexually inactive juvenile males and females was compared by means of a one way analysis of variance for each two-month period. In other analyses the influence of age of adult males and adult females on the exploratory behaviour was further examined. No differences could be found in the exploratory activity displayed by males or females and by sexually active or sexually inactive animals. Also a clear influence of age was absent. The interindividual variation was very large.  相似文献

Activity of endogenous cytokinins and gibberellins was determined in the shoots and roots of oak seedlings following the submersion of roots in water acidified with sulphuric add to pH 3.0, 2.5, 2.0, 1.7 and 5.6 (control) for 6 and 72 h. After 6 h the cytokinins increased in the shoots and in roots at pH 2.5 and 2.0, respectively. None of the tested pH values decreased the cytokinin activity. The acidification of solutions exhibited no remarkable influence on gibberellin activity. Only slight stimulation of the gibberellin activity was exhibited at pH 2.0. After 72 h a slight decrease in the activity of cytokinins was observed in the shoots at pH 2.5, remarkable decrease was revealed at pH 2.0 and 1.7. Nevertheless, in the roots stimulation of cytokinin activity appeared up to pH 2.0 (the highest at pH 2.5), and significant decrease was shown at pH 1.7. The activity of gibberellins in shoots showed similar dependence of changes as the activity of cytokinins in roots. Acidification stress exerted no influence on the gibberellin. activity in roots.  相似文献

It was found that haemolytic activity of Fushimi strain of Sendai virus multiplied in allantoic cavity of chicken embryos is independent on its haemagglutinating titer and also on allantoic fluid urea and uric acid content. It was shown in experiments with embryonated eggs that these two compounds have no also influence on haemolytic activity induction in Sendai virus. Moreover, the results of an experiment in which allantoic fluid was replaced by Eagle's liquid suggest that most probably the other components present in allantoic fluid do not also influence the appearance of haemolytic activity of this virus.  相似文献

The binding of actin to myosin containing phosphorylated and dephosphorylated light chains (LC2) was investigated by studying the influence of actin on Mg2+- and K+-stimulated ATPase of phosphorylated and dephosphorylated myosin and by comparing the influence of PPi on actomyosin formed from pure actin and phosphorylated or dephosphorylated myosin. The concentration of actin producing inhibition of one half of myosin K+-ATPase activity was 4.1 micro M and 7.7 micro M for phosphorylated and dephosphorylated myosin, respectively. Actomyosin formed from dephosphorylated myosin dissociated at lower PPi concentration than did that from the phosphorylated form. The extrapolated values of Km obtained from studies of the influence of actin on Mg2+-ATPase activity of dephosphorylated myosin were about twice as high as for the phosphorylated form. Thus, the affinity of phosphorylated myosin for actin was significantly higher under conditions studied.  相似文献

For assessing monoamine oxidase (MAO-A and -B) activities in the hypothalamus, ovaries and uterus, mature female rats were exposed to either continuous light or dark over 6 weeks. Confirming previous studies, continuous light induced constant estrus in all animals. The majority of animals kept under continuous dark during the six weeks remained mostly in diestrus with estrus appearing sporadically. The endocrinological function of the ovaries was disturbed by continuous light, which resulted in the development of polycystic ovaries, their morphological appearance being not significantly affected by continuous dark. Hypothalamic MAO-A activity was markedly reduced under the influence of both light (p < 0.05) and dark (p < 0.01). The activity of hypothalamic MAO-B was reduced only under the influence of dark (p < 0.01). In the ovaries, no significant differences were detected in either MAO-A or -B activity as a result of these environmental manipulations. In the uterus of rats exposed to continuous light which developed polycystic ovaries, MAO-A activity was significantly lower (p < 0.05) in comparison to either intact controls or rats exposed to continuous dark. These results demonstrate that chronic changes in photoperiodicity may considerably influence MAO-A activity and to a lesser extent MAO-B activity dependent on the tissue studied.  相似文献
