The different invasive and noninvasive diagnostic tests for Helicobacter pylori have been applied mainly in emerging countries. Molecular methods have been developed, especially a test for detection of H. pylori and its clarithromycin resistance directly from stools. The long-term effects of eradication on histologic lesions have been studied in a meta-analysis and the prognostic value of post-treatment in gastric mucosa-associated lymphoid tissue lymphoma has been assessed. An operating link for gastritis assessment (the OLGA staging) has also been published. Attempts to simplify the urea breath test protocol have been made, and new stool antigen tests have been proposed and compared to those previously available.  相似文献

Gas chromatography mass spectrometry has been used for over a decade for the determination of the amino acid sequences of fragment peptides derived from larger parent molecules. The majority of these fragments have from four to seven residues and several different methods of derivatization have been devised. Few reports have been published in which similar techniques have been used for the quantification of such peptides, but there is a growing list of small peptides which have been shown to have biological activity in their own right. This report is concerned with the development of a gas chromatographic mass spectrometric assay for the two eosinophil chemotactic peptides, Val-Gly-Ser-Glu and Ala-Gly-Ser-Glu, which appear to have a role to play in the course of the inflammatory process in skin disorders.  相似文献

1. Platelets have been isolated from plasma and their surface glycoconjugates radioactively-labelled using galactose oxidase and NaB3H4. 2. Conditions have been defined for optimal labelling of glycoproteins and a membrane fraction enriched in plasma membrane has been prepared and characterized by sodium dodecyl sulphate/polyacrylamide-gel electrophoresis. 3. Desialylated glycoproteins that act as receptors to peanut agglutinin and lentil lectin have been purified from a detergent extract of plasma membrane. 4. Two glycosylated polypeptides that are able to bind to the surfaces of platelets have been identified and some characteristics of the binding have been investigated.  相似文献

The presence or absence of a chemoreceptive capacity in marine mammals has drawn relatively little attention from the research community outside the Soviet Union. Toothed whales are typically labelled anosmic (lacking a sense of smell) since they do not have the peripheral olfactory structures typically associated with terrestrial mammals. Baleen whales are known to possess reduced olfactory tracts; their olfactory bulbs also may be reduced or absent. Although the neural structures that mediate taste in terrestrial mammals have been reported to be present in both groups of whales, cetaceans have been considered to have a poor sense of taste because typical mammalian taste receptors have been thought to be absent. Soviet researchers, however, recently have reported that gustatory receptors are present on some cetacean tongues and that the tongue of Tursiops truncatus appear to be well innervated. These workers also have been conducting investigations which seem to be aimed at describing a specialized gustatory capability in cetaceans. No experimental work has been reported by Western scientists. Little work has been done by either Western or Soviet researchers with regard to chemoreception among the other orders of marine mammals (Pinnipedia, Carnivora and Sirenia). Pinnipedia are typically labelled microsmatic (having a poor sense of smell); research has been restricted to histological examination of the nasal pathways, and neural anatomy. Sea otters are credited with a keen sense of smell, but no quantitative work has been reported. The chemosensory abilities of Sirenia remain unknown. The tongues of non-cetacean marine mammals have been histologically examined and found to resemble those of terrestrial mammals. No other investigations of gustation in non-cetacean marine mammals have been reported.  相似文献

Since its invention in the mid 1980s atomic force microscopy has revolutionised the way in which surfaces can be imaged. Close to atomic resolution has been achieved for some materials and numerous images of molecules on surfaces have been recorded. Atomic force microscopy has also been of benefit to biology where protein molecules on surfaces have been studied and even whole cells have been investigated. Here we report a study of red blood cells which have been imaged in a physiological medium. At high resolution, the underlying cytoskeleton of the blood cell has been resolved and flaws in the cytoskeleton structure may be observed. Comparison of the normal 'doughnut' shaped cells with swollen cells has been undertaken. Differences in both the global properties of the cells and in the local features in cytoskeleton structure have been observed.  相似文献

In this, a preliminary report, the advantages and disadvantages of upper lobe lobectomy and concomitant thoracoplasty in tuberculosis have been summarized. There has been no undue increase in the operative hazard, there have been no complications, and the immediate results have been entirely satisfactory.Twenty-six patients were subjected to a combined operation (17 upper lobectomy-thoracoplasty and nine pneumonectomy-thoracoplasty) with one postoperative death. Indications for primary lobectomy have been discussed. The technique of resection and concomitant thoracoplasty has been briefly described.  相似文献

Lac+ recombinant of Shigella flexneri 2a carrying R-factors of the standard set have been constructed. The R+ transconjugants obtained have been compared with the original strain with respect to their susceptibility to a set of bacteriophages recommended for phagetyping of Shigella strains and to a set of colicins. It have been found that as a result of acquiring some R-factors the susceptibility to bacteriophages changes; presence of certain R-factors causes susceptibility, that of other R-factors--resistance. Similar changes have been observed in the case of susceptibility to colicins. Propable mechanisms of the changes of susceptibility to these factors are discussed.  相似文献

Optical-fiber bundles have been employed as a versatile substrate for the fabrication of high-density microwell arrays. In this minireview, we discuss the application of optical-fiber-bundle arrays for a variety of biological problems. For genomics studies and microbial pathogen detection, individual beads have been functionalized with DNA probes and then loaded into the microwells. In addition, beads differentially responsive to vapors have been employed in an artificial olfaction system. Microwell arrays have also been loaded with living cells to monitor their individual response to biologically active compounds over long periods. Finally, the microwells have been sealed to enclose single enzyme molecules that can be used to measure individual molecule catalytic activity.  相似文献

For 30 days Wistar rats have been kept under conditions of hypokinesia. Using stereological methods, the volume and surface density of muscle fibers, microcirculatory bed and connective tissue components have been defined, and volumetric and surface--volume ratios of parenchymal and stromal structures have been calculated. Intracellular stereological parameters of the cardiac myocytes have been studied. Three-dimensional parameters in the parenchymatous-stromal interrelations of the atrophying myocardium accompanied with a deep rearrangement of the intracellular architectonics have been determined, as well as certain destructive changes of the cardiac myocyte ultrastructures. Such a treatment as a decreased functional loading is demonstrated to be the cause of cardiomyocytic lesions.  相似文献

The solution conformations of thyrotrophin-releasing hormone (TRH), a biologically-active tripeptide, have been predicted computationally using a method which represents solution effects as a dielectric continuum surrounding the molecule throughout energy minimisation; this is described as the reaction field contribution. The predicted conformations have been compared with previous experimental and theoretical studies. Favoured conformations of TRH under other simulated conditions have also been examined, including the behaviour of the cis isomer, and have been related to possible interactions with intermediates and with specific receptors. The relevance of these predictions to the design of biologically-active analogues of TRH has been analysed.  相似文献

The yeast Saccharomyces cerevisiae produces a peptide pheromone, termed the alpha-factor, as a prelude to sexual conjugation. Haploid MAT alpha-cells, but not haploid MAT a-cells or MAT a/alpha-diploids, produce this tridecapeptide of the structure: Trp-His-Trp-Leu-Gln-Leu-Lys-Pro-Gly-Gln-Pro-Met-Tyr. Structural analogues of the alpha-factor have been prepared with alterations in many of the residues, derivatized peptides have been synthesized, and truncated and elongated peptides have been studied. These peptides have been analyzed for their biological activities by various assays. Mutants of S. cerevisiae have been isolated that do not respond to alpha-factor or are supersensitive to the pheromone and its analogues. The mating system of S. cerevisiae provides a powerful model in which genetics, biochemistry, and molecular biology can be used to unravel the mysteries of peptide hormone structure and function.  相似文献

Humoral and cellular mechanisms of defense have been described for cephalopods, a relatively advanced group of mollusks. Typical of other mollusks, cephalopod agglutinins are the most documented component of humoral immunity. Lectins, which have agglutinating properties, have been described and characterized from octopuses. Agglutinins from cephalopod hemolymph have also been shown to agglutinate a variety of vertebrate red blood cells, as well as potential bacterial pathogens. Hemocytes are the primary component of cellular immunity. Although the hemocyte role in phagocytosis has been extensively studied in other mollusks, the mechanisms of phagocytosis have not been described extensively for cephalopods. Cephalopod hemocytes have phagocytic capabilities and may function in encapsulation and neutralization of foreign substances; however, the effects of environmental factors and the full extent of phagocytic capabilities of cephalopod hemocytes have not been reported. Hemocytes from cephalopods have a role in wound healing and inflammation which have been reported in detail by several investigators.  相似文献

Tachykinins are present in the pituitary gland and in brain areas involved in the control of the secretion of pituitary hormones. Tachykinins have been demonstrated to stimulate prolactin release acting directly on the anterior pituitary gland. These peptides have also been revealed to be able to act at the hypothalamic level, interacting with neurotransmitters and neuropeptides that have the potential to affect prolactin secretion. Tachykinins seem to act by stimulating or inhibiting the release of the factors that affect prolactin secretion. Among them, tachykinins have been demonstrated to stimulate oxytocin and vasopressin release, which in turn results in prolactin release. Tachykinins also potentiated the response to vasoactive intestinal peptide (VIP) and reinforced the action of glutamate, which in turn result in prolactin release. They have also been shown to interact with serotonin, a neurotransmitter involved in the control of prolactin secretion. In addition, tachykinins have been shown to inhibit GABA release, a neurotransmitter with prolactin-release inhibiting effect. This inhibition may result in an increased prolactin secretion by removal of the GABA inhibition. On the other hand, tachykinins have also been shown to stimulate dopamine release by the hypothalamus, an action that results in an inhibition of prolactin release. Dopamine is a well known inhibitor of prolactin secretion. In conclusion, although tachykinins have been shown to have a predominantly stimulatory effect on prolactin secretion, especially at the pituitary level, under some circumstances they may also exert an inhibitory influence on prolactin release, by stimulating dopamine release at the hypothalamic level.  相似文献

The drug Bitter Aloes has been used for several thousands of years because of its purgative properties and later several species of the plant have been cultivated for horticultural interest. In addition, many more local therapeutic properties have been recorded. Research has been largely directed towards the physiologically active components during the 50s and 60s. Extensions to this work in the last decade have been made in Germany, Japan and South Africa. Compounds have been characterized from the dried exudate prepared for drug purposes and from a few species. A number of anthraquinones have been recognized both in the free state and as glycosides. Other compounds with phenolic groups include chromones and a phenyl-pyranone. Chromatographic methods have been developed for recognizing some of these compounds and estimating the levels of those of pharmacological interest. Distribution of some of the compounds among a number of species is described showing that among some groups it has taxonomic significance. The biology of secretion and exudation from Aloë leaves remains largely unknown although fragmentary evidence points to interesting and potentially useful activity in some of the components.  相似文献

The breeder and the chemostat have been used to measure mutation rates for two mutations under a variety of steady state growth conditions. These rates have been found to be higher in complex medium than in minimal (F) medium. The effects of changes in nutritional conditions on these high rates have been described. In addition, the mutation rates at short generation times, in complex medium, have been shown to decrease with increasing generation time.  相似文献

Two rat neurophysins have been purified by salt precipitation, molecular sieving and ion-exchange chromatography. The proteins, performic-acid oxidized or reduced-alkylated, have been split either by trypsin or by staphylococcal proteinase and fragments have been separated by peptide mapping. Amino acid sequences of tryptic peptides have been determined either directly or after cleaving the large fragments by subtilisin, chymotrypsin, elastase or staphylococcal proteinase and characterizing the subfragments. Tryptic peptides have been ordered through the fragments given by staphylococcal proteinase. The N-terminal sequences of both proteins have also been established by automated degradation.The two usual types of mammalian neurophysins have been identified. One neurophysin belongs to the MSEL-neurophysin family and shows 11 substitutions and a 2-residue C-terminal truncation when compared with bovine MSEL-neurophysin. The other belongs to the VLDV-neurophysin family and shows 8 substitutions when compared with bovine VLDV-neurophysin. There are 23 differences between the MSEL- and VLDV-neurophysins of the rat.  相似文献

Connection between the putamen, the brain cortex and the hypothalamus, as well as the role of the former in different aspects of purposive behaviour have been studied in a complex morpho-physiological investigation. In 12 cats, after developing a symmetrical active-defensive conditioned reflex, unilateral electrolysis of the putamen has been performed and the number of conditioned-reflexive reactions have been counted before and after coagulation. The brains have been treated after Nauta--Gygax, Fink--Heimer with additional staining after Kawamura--Niimi. Monosynaptic connections of the putamen with frontal, precentral, postcentral, orbital, parietal cortical areas have been revealed; direct pathways from the putamen to the infundibulum of the grey tuber, to the posterior and lateral hypothalamic nucleus have been demonstrated; participation of the putamen in the formation of active-defensive conditioned reflexes has been stated, as well as in emotional behaviour with a preferable use either the right or the left foreleg.  相似文献

Probiotics have many potential therapeutic uses, but have not been universally accepted because of a lack of understanding of their action. Lactic acid bacteria (LAB) have been modified by traditional and genetic engineering methods to produce new varieties. Modern techniques of molecular biology have facilitated the identification of probiotic LAB strains, but only a few LAB have been modified by recombinant-DNA technology because of consumer resistance to their introduction to markets, especially in Europe.  相似文献
