Seedlings of the rootstocks Pineapple sweet orange (SwO), Carrizo citrange (CC), and sour orange (SO) were grown in low phosphorus (P) sandy soil and either inoculated with the vesicular-arbuscular mycorrhizal (VAM) fungus,Glomus intraradices, or were non-mycorrhizal (NM) and fertilized with P. VAM and NM seedings of similar shoot size and adequate P-status were selected for study of salinity and flooding stress. One-third of each of the VAM and NM plants were given 150 mM NaCl for a period of 24 days. One-third of the plants were placed into plastic bags and flooded for 21 days while the remaining third were non-stressed controls. In general, neither stress treatment affected mycorrhizal colonization. Salinity stress reduced the hydraulic conductivity of roots, leaf water potential, stomatal conductance and net assimilation of CO2 (ACO2) of mycorrhizal and non-mycorrhizal seedlings to a similar extent. VAM plants of CC and SO accumulated more Cl in leaves than NM plants. Cl was higher in non-mycorrhizal roots of SwO and CC than in mycorrhizal roots. Flooding the root zone for 3 weeks did not produce visible symptoms in the shoot but did influence plant water relations and reduce ACO2 of all 3 rootstocks. VAM and NM plants of each rootstock were affected similarly by flooding. Comparable reduction in nitrogen and P content of both mycorrhizal and non-mycorrhizal plants suggested that flooding stress was primarily affecting root rather than hyphal nutrient uptake. Kinetics of net phosphate (Pi) uptake was measured on intact ectomycorrhizal and non‐mycorrhizal Pinus sylvestris seedlings using a semihydroponic cultivation method. The depletion of Pi in a nutrient solution was assessed over a 160–0.2 μM Pi gradient. Growth of the pine seedlings was P limited and measurements were performed 7 and 9 weeks after inoculation. Three ectomycorrhizal fungi were studied: Paxillus involutus, Suillus bovinus and Thelephoraterrestris. Pi uptake was extremely fast in plants colonised by P. involutus. The Pi concentration dropped below 0.2 μM within 4–5 h. In plants colonised with S. bovinus this occurred in 5–6 h and in plants associated with T. terrestris 8 h were needed to run through the whole concentration range. Non‐mycorrhizal plants of similar size and nutrient status decreased Pi to a concentration between 1 and 2 μM in 18 h. Data were curve fitted to a two‐phase Michaelis‐Menten equation. The apparent kinetic constants, Km and Vmax, for the high affinity Pi uptake system of the pine roots could be estimated accurately. Vmax of this system was up to 7 times higher in pines associated with P. involutus than in non‐mycorrhizal seedlings. The intact extraradical mycelium greatly increased the absorption surface area of the roots (Vmax). Non‐mycorrhizal plants had a Km between 7.8 and 16.4 μM Pi. Plants mycorrhizal with P. involutus had Km values between 2.4 and 7.2, plants colonised with S. bovinus had a Km between 5.1 and 12.3, and seedlings associated with T. terrestris had a Km from 4.6 to 10.1 μM Pi. All 3 ectomycorrhizal fungi had a strong impact on the Pi absorption capacity of the pine seedlings. The results also demonstrated that there is substantial heterogeneity in kinetic parameters among the different mycorrhizal root systems.  相似文献

The influence of phosphorus (P) and nitrogen (N) supply on biomass, leaf area, photon saturated photosynthetic rate (Pmax), quantum yield efficiency (), intercellular CO2 concentration (Ci), and carboxylation efficiency (CE) was investigated in Vicia faba. The influence of P on N accumulation, biomass, and leaf area production was also investigated. An increase in P supply was consistently associated with an increase in N accumulation and N productivity in terms of biomass and leaf area production. Furthermore, P increased the photosynthetic N use efficiency (NUE) in terms of Pmax and . An increase in P supply was also associated with an increase in CE and a decrease in Ci. Under variable daily meteorological conditions specific leaf nitrogen content (NL), specific leaf phosphorus content (PL), specific leaf area (L), root mass fraction (Rf), Pmax, and remained constant for a given N and P supply. A monotonic decline in the steady-state value of Rf occurred with increasing N supply. L increased with increasing N supply or with increasing NL. We tested also the hypothesis that P supply positively affects both N demand and photosynthetic NUE by influencing the upper limit of the asymptotic values for Pmax and CE, and the lower limit for Ci in response to increasing N.This revised version was published online in March 2005 with corrections to the page numbers.  相似文献

Phytostabilization strategies may be suitable to reduce the dispersion of uranium (U) and the overall environmental risks of U-contaminated soils. The role of Glomus intraradices, an arbuscular mycorrhizal (AM) fungus, in such phytostabilization of U was investigated with a compartmented plant cultivation system facilitating the specific measurement of U uptake by roots, AM roots and extraradical hyphae of AM fungi and the measurement of U partitioning between root and shoot. A soil-filled plastic pot constituted the main root compartment (CA) which contained a plastic vial filled with U-contaminated soil amended with 0, 50 or 200 mg KH2PO4−P kg–1soil (CB). The vial was sealed by coarse or fine nylon mesh, permitting the penetration of both roots and hyphae or of just hyphae. Medicago truncatula plants grown in CA were inoculated with G. intraradices or remained uninoculated. Dry weight of shoots and roots in CA was significantly increased by G. intraradices, but was unaffected by mesh size or by P application in CB. The P amendments decreased root colonization in CB, and increased P content and dry weight of those roots. Glomus intraradices increased root U concentration and content in CA, but decreased shoot U concentrations. Root U concentrations and contents were significantly higher when only hyphae could access U inside CB than when roots could also directly access this U pool. The proportion of plant U content partitioned to shoots was decreased by root exclusion from CB and by mycorrhizas (M) in the order: no M, roots in CB > no M, no roots in CB > M, roots in CB > M, no roots in CB. Such mycorrhiza-induced retention of U in plant roots may contribute to the phytostabilization of U contaminated environments.  相似文献

Response of net photosynthetic rate (P N), stomatal conductance (g s), intercellular CO2 concentration (c i), and photosynthetic efficiency (Fv/Fm) of photosystem 2 (PS2) was assessed in Eucalyptus cladocalyx grown for long duration at 800 (C800) or 380 (C380) μmol mol-1 CO2 concentration under sufficient water supply or under water stress. The well-watered plants at C800 showed a 2.2 fold enhancement of P N without any change in g s. Under both C800 and C380, water stress decreased P N and g s significantly without any substantial reduction of c i, suggesting that both stomatal and non-stomatal factors regulated P N. Vesicular-arbuscular mycorrhizae may increase resistance of plants to drought by a number of mechanisms, such as increased root hydraulic conductivity, stomatal regulation, hyphal water uptake and osmotic adjustment. However, a substantial contribution of vesicular-arbuscular mycorrhizal (VAM) hyphae to water uptake has not been demonstrated unequivocally. The objective of this investigation was to examine the contribution of hyphae from two VAM fungi to water uptake and transport by the host plant. Lettuce (Lactuca sativa L.) plants were grown in a container divided by a screen into two compartments. One was occupied by roots, the other only by VAM hyphae, which the screen permitted to pass. Roots were colonized by the VAM fungi Glomus deserticola or Glomus fasciculatum, or were left uninoculated but P-supplemented. Water was supplied to the hyphal compartment at a distance of 10 cm from the screen (root). CO2 exchange rate, water-use efficiency, transpiration, stomatal conductance and photosynthetic phosphorus-use efficiency of VAM or P-amended control plants were evaluated at three levels of water application in the hyphal compartment. Results indicate that much of the water was taken up by the hyphae in VAM plants. VAM plants, which had access to the hyphal compartment, had higher water and nutrient contents. G. deserticola functioned efficiently under water limitation and mycelium from G. fasciculatum-colonized plants was very sensitive to water in the medium. This discrepancy in VAM behaviour reflects the various abilities of each fungus according to soil water levels. Different abilities of specific mycelia were also expressed in terms of nutritional and leaf gas-exchange parameters. G. fasciculatum caused a significant increase in net photosynthesis and rate of water use efficiency compared to G. deserticola and P-fertilized plants. In contrast, the G. deserticola treatment was the most efficient affecting N, P and K nutrition, leaf conductance and transpiration. Since no differences in the intra- and extra-radical hyphal extension of the two endophytes were found, the results demonstrate that mycorrhizal hyphae can take up water and that there are considerable variations in both the behaviour of these two VAM fungi and in the mechanisms involved in their effects on plant water relations.  相似文献

Bean (Phaseolus vulgaris L. cv. Dwarf) roots were inoculated with Rhizobium phaseoli and colonized by the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus fasciculatum Gerd. and Trappe or left uncolonized as controls. The symbiotic associations were grown in an inert substrate using 0, 25, 50, 100, or 200 milligrams hydroxyapatite (HAP) (Ca10[PO4]6[OH]2) per pot as a P amendment. Plant and nodule dry weights and nodule activity increased for both VAM and control plants with increasing P availability, but values for VAM plants were significantly lower in all parameters than for controls. Inhibition of growth and of N2 fixation in VAM plants was greatest at the lowest and highest P regimes. It was smallest at 50 milligrams HAP, where available P at harvest (7 weeks after planting) was 5 micrograms P per gram substrate. At this level of P availability, the association apparently benefited from increased P uptake by the fungal endophyte. Percent P values for shoots, roots, and nodules did not differ significantly (p > 0.05) between VAM and control plants. The extent of colonization, fungal biomass, and the fungus/association dry weight ratio increased several fold as HAP was increased from 0 to 200 milligrams. It is concluded that intersymbiont competition for P and photosynthate was the primary cause for the inhibition of growth, nodulation, and nodule activity in VAM plants. Impaired N2 fixation resulted in N stress which contributed to inhibition of host plant growth at all levels of P availability.  相似文献

The role of mycorrhizal fungi in acquisition of mineral nutrients by host plants is examined for three groups of mycorrhizas. These are; the ectomycorrhizas (ECM), the ericoid mycorrhizas (EM), and the vesicular-arbuscular mycorrhizas (VAM). Mycorrhizal infection may affect the mineral nutrition of the host plant directly by enhancing plant growth through nutrient acquisition by the fungus, or indirectly by modifying transpiration rates and the composition of rhizosphere microflora. A capacity for the external hyphae to take up and deliver nutrients to the plant has been demonstrated for the following nutrients and mycorrhizas; P (VAM, EM, ECM), NH4 + (VAM, EM, ECM), NO3 - (ECM), K (VAM, ECM), Ca (VAM, EM), SO4 2- (VAM), Cu (VAM), Zn (VAM) and Fe (EM). In experimental chambers, the external hyphae of VAM can deliver up to 80% of plant P, 25% of plant N, 10% of plant K, 25% of plant Zn and 60% of plant Cu. Knowledge of the role of mycorrhiza in the uptake of nutrients other than P and N is limited because definitive studies are few, especially for the ECM. Although further quantification is required, it is feasible that the external hyphae may provide a significant delivery system for N, K, Cu and Zn in addition to P in many soils. Proposals that ECM and VAM fungi contribute substantially to the Mg, B and Fe nutrition of the host plant have not been substantiated. ECM and EM fungi produce ectoenzymes which provide host plants with the potential to access organic N and P forms that are normally unavailable to VAM fungi or to non mycorrhizal roots. The relative contribution of these nutrient sources requires quantification in the field. Further basic research, including the quantification of nutrient uptake and transport by fungal hyphae in soil and regulation at the fungal-plant interface, is essential to support the selection and utilization of mycorrhizal fungi on a commercial scale.  相似文献

On the first day after foliar application, chitosan pentamer (CH5) and chitin pentamer (CHIT5) decreased net photosynthetic rate (P N) of soybean and maize, however, on subsequent days there was an increase in P N in some treatments. CH5 caused an increase in maize P N on day 3 at 10–5 and 10–7 M; the increases were 18 and 10 % over the control plants. This increase was correlated with increases in stomatal conductance (g s) and transpiration rate (E), while the intercellular CO2 concentration (C i) was not different from the control plants. P N of soybean plants did not differ from the control plants except for treatment CH5 (10–7 M) which caused an 8 % increase on day 2, along with increased g s, E, and C i. On days 5 and 6 the CHIT5 treatment caused a 6–8 % increase in P N of maize, which was accompanied by increases in g s, E, and C i. However, there was no such increase for soybean plants treated with CHIT5. In general, foliar application of high molecular mass chitin (CHH) resulted in decreased P N, particularly for 0.010 % treated plants, both in maize and soybean. Foliar applications of chitosan and chitin oligomers did not affect (p > 0.05) maize or soybean height, root length, leaf area, shoot or root or total dry mass.  相似文献

Net photosynthetic rate (P N), stomatal conductance (g S), transpiration rate (E), intercellular CO2 concentration (C i), leaf water potential (w), leaf area, chlorophyll (Chl) content, and the activities of photosynthetic carbon reduction cycle (PCR) enzymes in two mulberry (Morus alba L.) cultivars (drought tolerant Anantha and drought sensitive M-5) were studied during water stress and recovery. During water stress, P N, g S, and E declined whereas C i increased. P N, g S, and E were less affected in Anantha than in M-5, which indicates tolerance nature of Anantha over M-5. Activities of ribulose-5-phosphate kinase, NAD- and NADP-glyceraldehyde-3-phosphate dehydrogenases, and 3-phosphoglycerate kinase decreased with increasing stress in both the cultivars. The enzyme activities less affected in tolerant (Anantha) than in sensitive cultivar (M-5) were restored after re-watering to almost initial values in both the cultivars. Re-watering of the plants led to an almost complete recovery of P N, E, and g S, indicating that a short-term stress brings about reversible effect in these two cultivars of mulberry.  相似文献

Plants associated with arbuscular mycorrhizal fungi (AMF) acquire phosphorus via roots and extraradical hyphae. How soil P level affects P accumulation within hyphae and how P in hyphae influences the accumulation of metal minerals remains little explored. A bi-compartmented in vitro cultivation system separating a root compartment (RC), containing a Ri T-DNA transformed carrot root associated to the AMF Rhizophagus irregularis DAOM 197198, from a hyphal compartment (HC), containing only the extraradical hyphae, was used. The HC contained a liquid growth medium (i.e., the modified Strullu-Romand medium containing P in the form of KH2PO4) without (0 μM) or adjusted to 35, 100, and 700 μM of KH2PO4. The accumulation of P and metal minerals (Ca, Mg, K, Na, Fe, Cu, Mn) within extraradical hyphae and AMF-colonized roots, and the expression of the phosphate transporter gene GintPT were assessed. The expression of GintPT in the extraradical hyphae did not differ in absence of KH2PO4 or in presence of 35 and 100 μM KH2PO4 in the HC but was markedly reduced in presence of 700 μM KH2PO4. Hyphal P concentration was significantly lowest in absence of KH2PO4, intermediate at 35 and 100 μM KH2PO4 and significantly highest in presence of 700 μM KH2PO4 in the HC. The concentrations of K, Mg, and Na were positively associated with the concentration of P in the extraradical hyphae developing in the HC. Similarly, P concentration in extraradical hyphae in the HC was related to P concentration in the growth medium and influenced the concentration of K, Mg, and Na. The accumulation of the metal mineral K, Mg, and Na in the extraradical hyphae developing in the HC was possibly related to their function in neutralizing the negative charges of PolyP accumulated in the hyphae.  相似文献

In bean (Phaseolus vulgaris L.) seedlings well supplied with water, rates of transpiration (E) and CO2 assimilation (P N) of the primary leaves were measured under blue (BR) or red (RR) irradiance of 150 µmol(photon) m–2 s–1. The leaf conductance to H2O vapour transfer (g H2O), as well as the intercellular concentrations of H2O vapour (e i) and of CO2 (C i) were calculated. Under BR, g H2O was significantly greater, but P N was lower, and E similar as compared with corresponding values found under RR. The increase of stomata aperture under BR was evident although C i was higher and e i was lower than under RR. Results agree with the suggestion that BR directly activates guard cell metabolism and in well watered plants determines mainly the stomata aperture.  相似文献

The effect of sodium chloride and triadimefon (TDM) on the chlorophyll (Chl) content, net photosynthetic rates (PN), rate of transpiration (E), and intercellular CO2 concentration (Ci) in Raphanus sativus was studied. The effect of NaCl salinity was partially ameliorated by TDM which caused increase in Chl content, PN, and Ci. TDM also increased root dry matter production, decreased E, and increased the water use efficiency. Tolerance to phosphorus stress was studied in Capsicum annuum L. Chile ancho cv. San Luis and bell pepper cv. Jupiter plants. Plants were fertilized weekly with Long-Ashton nutrient solution (LANS) modified to supply 0, 11, 22, 44, 66, or 88 g(P) m-3 (P0, P11, P22, P44, P66, P88). Phosphorus stress occurred in both cultivars at P0 and P11, with reduced plant growth and development. At P0, the lowest percentage of total biomass was directed toward reproductive growth. The root/shoot ratio was greatest at P0, reflecting greater dry matter partitioning to the root system. Growth of ‘San Luis’ was more sensitive to phosphorus stress than ‘Jupiter’. A greater percentage of total biomass was directed towards reproductive growth in ‘Jupiter’ than ‘San Luis’. Increasing P nutrition elevated leaf tissue P in both cultivars with highest leaf tissue P at P88. There were no differences in tissue P between P0 and P11 ‘San Luis’ plants, whereas P0 ‘Jupiter’ plants had the lowest tissue P. Low P-plants generally had the highest tissue N and lowest S, Mn, and B. In both cultivars, gas exchange was lowest at P0, as indicated by reduced stomatal conductance (gs) and net photosynthetic rate (PN). Internal CO2 concentration and leaf-to-air vapor pressure difference (VPD) were generally highest with P-stressed plants. Phosphorus use efficiency, as indicated by PN per unit of leaf tissue P concentration (PN/P), was highest at P11. Generally, no P treatments exceeded the gas exchange levels obtained by P44 (full strength LANS) plants. Both PN and gs declined during reproductive growth in ‘San Luis’, which fruits more rapidly than ‘Jupiter’, whereas no reduction in gas exchange occurred with ‘Jupiter’. Changes in net photosynthetic rate (PN), stomatal conductance (gs), intercellular CO2 concentrations (Ci), transpiration rate (E) and water use efficiency (WUE) were measured in Plantago major L. plants grown under sufficient soil water supply or under soil water stress conditions. The plants had high PN in a wide range of soil water potential and temperature regimes. Soil water had little effect on PN under ambient CO2 concentrations, which was explained by a high carboxylation rate, but increased the dark respiration rate. Carboxylation activity at low Ci depended on RuBP regeneration, whereas at high Ci it depended on the phosphate regeneration rate. The gs and E values were low in plants under stress as compared to the controls that resulted in an increase of WUE. The results obtained show that Plantago major plants have different ways of adaptation to soil water deficit conditions.  相似文献

Seventeen clones of C4 grass Miscanthus spp. collected from different climatic regions and elevations of Taiwan were transplanted in pots. 15–16 months after collection the plants received 0, 1, and 2 g of nitrogen fertiliser (N0, N1, and N2, respectively) per pot. All the measurements were done 10–12 d after N application. The relationships between net photosynthetic rate (P N) and photon flux density (PFD) showed a saturated curve, with PFD saturation at about 1 000 µmol m–2 s–1. The ranges of PFD saturated P N (P sat) for all the tested clones with N0, N1, and N2 were 8–16, 11–18, and 12–21 µmol m–2 s–1, respectively. The clones from southern Taiwan, a tropical region, showed the highest P sat, followed by the clones from northern Taiwan, a subtropical region, while those from mountainous area showed the lowest P sat. The clones collected from southern Taiwan showed the highest frequency of stomata on the adaxial surface, and those collected from the high mountainous area showed the lowest frequency. Also the adaxial surface of leaves from the higher mountainous area had more wax deposited than the leaves from the lowland. Thus the low P sat in mountain clones is limited by both stomatal and non-stomatal factors. Further, the lower leaf conductance and different epidermal characteristics of mountain clones might prevent excessive loss of heat through transpiration and provide production against ultraviolet-B radiation.  相似文献

The effects of two phosphorus (P) sources (triple superphosphate and Ghafsa phosphate rock), applied at rates equivalent to 44kg ha-1 and 22 kg ha-1, on vesicular-arbuscular mycorrhizal (VAM) fungal infection in roots, dry matter yield and nutrient content of maize grown in an oxisol and an alfisol, were investigated in a growth cabinet. The application of 44 kg P ha -1 resulted in root infection by VAM fungi not was significantly different (P<-0.01) from when no P was applied. Root infection was significantly greater when P was applied as triple superphosphate at the rate of 22 kg ha-1 the higher rate. Phosphate rock treatments at both rates of application resulted in significantly greater root infection than in controls with no P or when triple superphosphate was applied at 44 kg ha-1. Plant P uptake increased in all soils with the different P treatments compared with the control. No direct effects of the treatments on the aluminium and zinc contents of maize plants were observed. In the gleyic alfisol, reduced Mn uptake as a result of increased infection of plants with the superphosphate treatments was observed. Higher Mn was also found in plants with the higher rate of superphosphate treatment than with the phosphate rock treatments in the haplustox, although infection rates in plants with the latter treatments were higher. With the exception of plants with the phosphate rock treatment applied at 22kg ha-1, dry matter yields of plants with all P sources were significantly greater than the controls.  相似文献

At the grain-filling stage, net photosynthetic rate (P N), stomatal conductance (g s), and ribulose-1,5-bisphosphate carboxylation efficiency (CE) were correlated in order to find the determinant of photosynthetic capacity in rice leaves. For a flag leaf, P N in leaf middle region was higher than in its upper region, and leaf basal region had the lowest P N value. The differences in g s and CE were similar. P N, g s, and CE gradually declined from upper to basal leaves, showing a leaf position gradient. The correlation coefficient between P N and CE was much higher than that between P N and g s in both cases, and P N was negatively correlated with intercellular CO2 concentration (C i). Hence the carboxylation activity or activated amount of ribulose-1,5-bisphosphate carboxylase/oxygenase rather than gs was the determinant of the photosynthetic capacity in rice leaves. In addition, in flag leaves of different tillers P N was positively correlated with g s, but negatively correlated with C i. Wheat plants grown in controlled growth chambers were exposed to drought stress (DS) and high temperature (HT) singly and in combination (DS+HT). The effects of these two stresses on net photosynthetic rate (P N), stomatal conductance (g s), intercellular CO2 concentration (C i), quantum efficiency of photosystem 2 (ΦPS2), variable to maximum chlorophyll (Chl) fluorescence (Fv/Fm), photochemical (qp) and non-photochemical (NPQ) Chl fluorescence, and yield were investigated. Grain yield was decreased by 21 % due to DS, while it was increased by 26 % due to HT. P N, g s, C i, and Chl fluorescence were dramatically reduced to DS, HT, and their interaction, except NPQ which showed an increase due to HT.  相似文献

Five-year-old plants of two olive cultivars (Frantoio and Moraiolo) grown in large pots were exposed for 7 to 8 months to ambient (AC) or elevated (EC) CO2 concentration in a free-air CO2 enrichment (FACE) facility. Exposure to EC enhanced net photosynthetic rate (P N) and decreased stomatal conductance, leading to greater instantaneous transpiration efficiency. Stomata density also decreased under EC, while the ratio of intercellular (C i) to atmospheric CO2 concentration and chlorophyll content did not differ, except for the cv. Moraiolo after seven months of exposure to EC. Analysis of the relationship between photosynthesis and C i indicated no significant change in carboxylation efficiency of ribulose-1,5-bisphosphate carboxylase/oxygenase after five months of exposure to EC. Based on estimates derived from the P N-C i relationship, there were no apparent treatment differences in daytime respiration, CO2 compensation concentration, CO2-saturated photosynthetic rate, or photosynthetic rate at the mean C i, but there was a reduction in stomata limitation to P N at EC. Thus 5-year-old olive trees did not exhibit down regulation of leaf-level photosynthesis in their response to EC, though some indication of adjustment was evident for the cv. Frantoio with respect to the cv. Moraiolo.  相似文献
