Twenty-four yearling Boer × Spanish wethers (7/8 Boer; initial body weight (BW) of 34.1 ± 1.02 kg) were used to determine effects on methane (CH4) emission of dietary levels of a condensed tannin (CT)-containing forage, Kobe lespedeza (Lespedeza striata; K), and a forage very low in CT, sorghum-sudangrass (Sorghum bicolor; G). Treatments were dietary K levels (dry matter (DM) basis) of 1.00, 0.67, 0.33, and 0 (100, 67, 33, and 0 K, respectively). Forages were harvested daily and fed at approximately 1.3 times maintenance metabolizable energy requirement. The experiment lasted 21 days, with most measures on the last 8 days. The CT concentration was 0.3 and 151 g/kg DM in G and K, respectively. DM intake was similar among treatments (i.e., 682, 675, 654, and 648 g/day; S.E. = 30.0) and gross energy (GE) digestibility increased linearly (P<0.05) with decreasing K (0.472, 0.522, 0.606, and 0.666 for 100, 67, 33, and 0 K, respectively). CH4 emission changed quadratically (P<0.05) with decreasing K (10.9, 13.8, 17.6, and 26.2 l/day; 32, 42, 57, and 88 kJ/MJ GE; 69, 81, 94, and 133 kJ/MJ digestible energy for 100, 67, 33, and 0 K, respectively). In vitro CH4 emission by incubation of ruminal fluid for 3 weeks with a medium for methanogenic bacteria and other conditions promoting activity by methanogens also was affected quadratically (P<0.05) by K level (7.0, 8.1, 9.2, and 16.1 ml for 100, 67, 33, and 0 K, respectively). The total bacterial count of ruminal samples was similar among K levels, but the number of total protozoa increased linearly (P<0.05) as K declined (8.3, 11.8, 15.6, and 27.1 × 105 ml−1 for 100, 67, 33, and 0 K, respectively). The CT-containing forage K decreased CH4 emission by goats regardless of its feeding level, although the effect per unit of K increased with decreasing K. Forage type (i.e., legume versus grass) may have contributed to the effect of K on CH4 emission, but most of the change appeared attributable to CT, which appeared to directly impact activity of methanogenic bacteria, although alterations of protozoal activity could have been involved. These findings suggest that relatively low dietary levels of CT could be employed to lessen CH4 emission without a marked detrimental effect on other conditions such as total tract protein digestion.  相似文献

The beneficial effects of forages containing condensed tannins (CTs) on ruminants are well documented, but the chemical features of CT that yield benefits have not been defined. Some evaluations of limited numbers of highly purified compounds have resulted in positive correlations between CT molecular weight (MW) and biological activity, while others have failed to show a correlation. The objectives of this study were to determine if MW of CT could predict biological activity relative to protein precipitability. MW of condensed tannin, protein-precipitable phenolics (PPP), and the amount of protein bound (PB) were determined for nine species of warm-season perennial legumes. There was no correlation between PPP or PB and MW (R2 0.11 and R2 0.02, respectively). However, CT concentration did correlate with PPP and PB (R2 0.81 and R2 0.69, respectively). It was concluded that CT MW does not explain the variation in protein precipitation by CT from the forage legumes surveyed.  相似文献

The percentage of tannins in leaves, bark, wood, and immature fruits of several species of Acacia and related mimosoid legumes from the southwestern U.S. and Mexico, along with a few from Costa Rica and Argentina, was determined by a modified hide powder procedure and by precipitation with casein. The relative percentages of hydrolyzable and condensed tannins were determined by the iodate and the vanillin-HCl methods, respectively. Gallotannins of selected samples were also determined by the rhodanine method. Although the amount of total tannins was similar for the first two methods, values for condensed tannins by the vanillin-HCl method were frequently two to four times greater than the total tannin values.  相似文献

Protein in white clover (Trifolium repens L.) is poorly utilised by ruminants because of its extensive degradation to ammonia in the rumen. However, white clover produces condensed tannins (CT) in its flowers, which can reduce rumen proteolysis. Effects of increasing proportions of clover dry matter (DM) as flowers (and therefore floral CT) on soluble protein, ammonia and volatile fatty acid (VFA) concentrations were determined with in vitro incubations. Minced mixtures of 0, 250, 500, 750 and 1000 g/kg of DM as white clover flower (F) with the remainder as white clover leaf, were incubated in vitro and sampled after 0, 2, 4, 8, 12 and 24 h. Treatments contained 0, 13, 26, 39 and 52 g CT/kg DM, respectively. A further treatment with 500 g/kg DM as flower and 500 g/kg DM as leaf had polyethylene glycol added to remove effects of CT. Increasing the proportion of white clover as flowers from 0 to 1000 g/kg DM reduced net conversion of plant N to ammonia N from 290 to 120 mM/M at least partly due to reduced solubility of the protein. Treatments with 750 g/kg DM or more as clover flowers reduced ammonia concentrations to levels likely to limit microbial growth. Total VFA production was not affected by flower content, although the proportion of acetate to propionate increased. The contribution of CT to treatment effects was small compared to effects attributed to difference in chemical composition between flowers and leaves.  相似文献

Condensed tannins in plants are found free and attached to protein and fibre but it is not known whether these fractions influence rumen degradation and microbial colonisation. This study explored the rumen degradation of tropical tannin-rich plants and the relationship between the disappearance of free and bound condensed tannin fractions and microbial communities colonising plant particles using in situ and in vitro experiments. Leaves from Calliandra calothyrsus, Gliricidia sepium, and Leucaena leucocephala, pods from Acacia nilotica and the leaves of two agricultural by-products: Manihot esculenta and Musa spp. were incubated in situ in the rumen of three dairy cows to determine their degradability for up to 96 h. Tannin disappearance was determined at 24 h of incubation, and adherent microbial communities were examined at 3 and 12 h of incubation using a metataxonomic approach. An in vitro approach was also used to assess the effects of these plants on rumen fermentation parameters. All plants contained more than 100 g/kg of condensed tannins with a large proportion (32–61%) bound to proteins. Calliandra calothyrsus had the highest concentration of condensed tannins at 361 g/kg, whereas Acacia nilotica was particularly rich in hydrolysable tannins (350 g/kg). Free condensed tannins from all plants completely disappeared after 24-h incubation in the rumen. Disappearance of protein-bound condensed tannins was variable with values ranging from 93% for Gliricidia sepium to 21% for Acacia nilotica. In contrast, fibre-bound condensed tannin disappearance averaged ~ 82% and did not vary between plants. Disappearance of bound fractions of condensed tannins was not associated with the degradability of plant fractions. The presence of tannins interfered with the microbial colonisation of plants. Each plant had distinct bacterial and archaeal communities after 3 and 12 h of incubation in the rumen and distinct protozoal communities at 3 h. Adherent communities in tannin-rich plants had a lower relative abundance of fibrolytic microbes, notably Fibrobacter spp. whereas, archaea diversity was reduced in high-tannin-containing Calliandra calothyrsus and Acacia nilotica at 12 h of incubation. Concurrently, in vitro methane production was lower for Calliandra calothyrsus, Acacia nilotica and Leucaena leucocephala although for the latter total volatile fatty acids production was not affected and was similar to control. Here, we show that the total amount of hydrolysable and condensed tannins contained in a plant govern the interaction with rumen microbes affecting degradability and fermentation. The effect of protein- and fibre-bound condensed tannins on degradability is less important.  相似文献

Transformants of Arabidopsis thaliana can be generated without using tissue culture techniques by cutting primary and secondary inflorescence shoots at their bases and inoculating the wound sites with Agrobacterium tumefaciens suspensions. After three successive inoculations, treated plants are grown to maturity, harvested and the progeny screened for transformants on a selective medium. We have investigated the reproducibility and the overall efficiency of this simple in planta transformation procedure. In addition, we determined the T-DNA copy number and inheritance in the transformants and examined whether transformed progeny recovered from the same Agrobacterium-treated plant represent one or several independent transformation events. Our results indicate that in planta transformation is very reproducible and yields stably transformed seeds in 7–8 weeks. Since it does not employ tissue culture, the in planta procedure may be particularly valuable for transformation of A. thaliana ecotypes and mutants recalcitrant to in vitro regeneration. The transformation frequency was variable and was not affected by lower growth temperature, shorter photoperiod or transformation vector. The majority of treated plants gave rise to only one transformant, but up to nine siblings were obtained from a single parental plant. Molecular analysis suggested that some of the siblings originated from a single transformed cell, while others were descended from multiple, independently transformed germ-line cells. More than 90% of the transformed progeny exhibited Mendelian segregation patterns of NPTII and GUS reporter genes. Of those, 60% contained one functional insert, 16% had two T-DNA inserts and 15% segregated for T-DNA inserts at more than two unlinked loci. The remaining transformants displayed non-Mendelian segregation ratios with a very high proportion of sensitive plants among the progeny. The small numbers of transformants recovered from individual T1 plants and the fact that none of the T2 progeny were homozygous for a specific T-DNA insert suggest that transformation occurs late in floral development.National Research Council of Canada Publication No. 38003  相似文献

Turtlegrass, Thalassia testudinum, produces high concentrations of proanthocyanidins (condensed tannins) which we hypothesized are induced by grazing, as a component of a general wound response. To test this we quantified condensed tannins in a variety of turtlegrass tissues following simulated fish grazing, grazing by the urchin Lytechinus variegatus, and treatment with the natural plant wound hormone jasmonic acid. We observed that simulated fish grazing triggered rapid induction of condensed tannins by an average of 10 mg tannin g−1 dry mass (DM) after 5 days. Condensed tannin accumulations were correlated with a reduction of blade extension rates. Further, we observed that constitutive tannin levels in developing first-rank leaves were strongly correlated with the width of second-rank leaves on each shoot, with an increase of 7.7 mg tannin g−1 tissue dry mass per millimeter blade width on average. We propose that wider source leaves provide additional resources for phenolic biosynthesis. There was no induction of tannins in leaves, meristematic or sheath tissues in response to grazing by the urchin L. variegatus, the presence of potential waterborne cues from nearby grazed plants, or to treatment with 5 mM jasmonic acid. However, urchin grazing did induce tannin production in root/rhizome tissues, where they accumulated to levels 3–4 times higher than in blades (up to 350 mg tannin g−1 tissue dry mass). These results confirm the potential for rapid wound-induced condensed tannin accumulations in T. testudinum. The link between blade widths and the tannin content of new leaves indicates that leaf morphology may be a useful bioindicator for predicting herbivore and disease-resistance in the field.  相似文献

随着测序技术和体外培养技术的进步,越来越多的未知甲烷菌被发现。近年来,第七个甲烷菌目"Methanomassiliicoccales"(简称Mmc)被发现并建立。这一目甲烷菌在进化关系上与热原体古菌相近,但又存在较远距离,独立成簇。Mmc分布广泛,遍布哺乳动物和昆虫的消化道以及稻田、湿地等环境,但不同来源菌株表现出生境偏好性。Mmc缺少将CO2还原为甲基辅酶M的完整代谢途径,导致它们严格利用氢气还原甲基底物生成甲烷。全面深入地了解Mmc在反刍动物瘤胃中发挥的功能,将有助于新型高效反刍动物甲烷减排策略的提出。因此,本文主要综述了Mmc菌株的分离培养、生理生化和基因组特性及其在瘤胃甲烷生成中的作用。  相似文献

带叶兜兰种子原地共生萌发及有效菌根真菌的分离与鉴定   总被引:1,自引:0,他引:1  
为获得带叶兜兰(Paphiopedilum hirsutissimum)种子萌发的共生真菌,采用原地共生萌发技术获得了2株自然萌发的小幼苗,并分离和筛选出了有效的种子萌发共生菌——瘤菌根菌(Epulorhiza sp.)。为验证分离菌株对带叶兜兰种子萌发的有效性,将Phs34号菌株与带叶兜兰种子在灭菌后的原生境基质上进行室内共生萌发试验,结果表明,经过6周的培养,对照组没有观察到种子的萌发;接菌的种子胚明显膨大,突破种皮,形成原球茎,平均萌发率为(58.35±3.41)%。这表明分离得到的瘤菌根菌能促进带叶兜兰的种子萌发。  相似文献

Non-adapted plants growing in high altitude such as bracken fern Pteridium arachnoideum are exposed to environmental extremes that may induce a chemical adaptive response. Here we show that there is a non-uniform distribution of low (LMP) and high molecular weight (HMP) phenolics in the frond parts of P. arachnoideum growing at high elevation. LMP–HMP levels were measured in sun-exposed (E) and self shaded (SS) pinnae between 2100 and 3190 m in the tropical Andes, during dry and rainy seasons. While there was no difference in E vs. SS contents of LMP at 2100 m, E accumulated greater LMP–HMP concentrations relative to SS as altitude increased. This difference was increased during the dry season. Linear correlations between the position of each pinnae relative to the ground level and LMP–HMP occurred along a 2570–3190 m transect. Water restriction in the dry season also caused increase of LMP and HMP. We conclude that excess UV-B radiation and water availability are important modelers of the non-adapted plant acclimation response to stress in tropical high mountain habitats.  相似文献

【目的】开发一种稳定可控的从反刍动物瘤胃中分离、培养真核微生物尖尾内毛虫的技术方法,为原生动物瘤胃纤毛虫内毛虫的种质资源储备和生理功能研究提供足够的实验材料。【方法】首先采用瘤胃插管法从武汉地区奶牛瘤胃中采集瘤胃液,并通过微孔滤网过滤法逐级分离富集瘤胃纤毛虫尖尾内毛虫,然后用本研究改良的SP培养基在厌氧培养瓶中对所分离富集的尖尾内毛虫进行体外培养,经过纯化培养和单种培养获得瘤胃纤毛虫尖尾内毛虫武汉分离株系的单一种培养体系。其次,结合形态观察和18S rRNA基因测序进行物种鉴定及系统发育分析。最后,采用对半转移培养法计算单种培养的尖尾内毛虫武汉分离株系的世代时间。【结果】从奶牛的瘤胃液中分离、培养得到一个尖尾内毛虫武汉分离株系的体外单种培养体系。本研究所用改良SP培养基由改良SP盐溶液、无原虫瘤胃液上清、半胱氨酸盐酸盐、抗生素、淀粉和草粉等组合而成,虫体在此培养基中由起始接种密度320个/ml经16 d培养可实现最高培养密度37 000个/ml的单种培养,而且可进行稳定生长传代增殖。基于形态观察、18S rRNA基因的同源搜索和系统发育分析表明,本研究所培养的虫体为尖尾内毛虫武汉分离株系,命名为Entodinium caudatum strain WH。尖尾内毛虫武汉分离株系的世代时间为19.0 h。【结论】本研究成功建立反刍动物瘤胃内主要真核微生物瘤胃纤毛虫的单种体外培养方法,实现尖尾内毛虫武汉分离株系的实验室稳定可控培养,为深入开展瘤胃纤毛虫种质资源的探索和尖尾内毛虫的功能研究提供足够的实验材料。  相似文献

Summary Linear, extrachromosomal DNA's of the filamentous fungus Ascobolus immersus are localized within the mitochondria. These linear plasmids have no homology to the high molecular weight mtDNA (hmw mtDNA). For analysis of plasmid replication an in organello DNA synthesis system was developed, in which radionucleotides were incorporated into intact mitochondria. Plasmid DNA is labelled preferentially in this system. From replication analysis of a specific plasmid there is evidence of a virus-like protein-primed replication. Sequence analysis of this plasmid reveals that a viral DNA polymerase is encoded. Thus, these genetic elements presumably are viral remnants rather than true plasmids.  相似文献

Interleukin-21 (IL-21) is a class I cytokine with antitumor properties due to enhanced proliferation and effector function of CD8+ T cells and natural killer (NK) cells. Here we have explored the magnitude and time-course of cytostatics-induced lymphopenia in mice and investigated whether treatment with cytostatics influences the antitumor effect of IL-21 in mouse tumor models. We show that pegylated liposomal doxorubicin (PLD), irinotecan and oxaliplatin induced transient lymphopenia, whereas 5-fluorouracil (5-FU) transiently increased lymphocyte counts. B cells were more sensitive than T cells towards irinotecan and oxaliplatin. Additive antitumor effects were observed after combining IL-21 with PLD, oxaliplatin and to less extent 5-FU but not irinotecan, and larger effect was observed when IL-21 administration was postponed relative to chemotherapy, suggesting that these agents may transiently impair immune function. However, the chemotherapies did not significantly alter the levels of circulating regulatory T cells and only marginally affected the ability of CD8+ T cells to respond to IL-21 measured as increased granzyme B mRNA. Our results show that IL-21 therapy can be successfully combined with agents from different chemotherapeutic drug classes, i.e. topoisomerase II inhibitors (PLD), anti-metabolites (5-FU) and platinum analogs (oxaliplatin) provided that IL-21 therapy is delayed relative to chemotherapy.  相似文献

Oligomeric procyanidins were isolated from the leaves and flowers of hawthorn (Crataegus laevigata). A trimer, epicatechin-(4β→8)-epicatechin-(4β→6)-epicatechin, and a pentamer consisting of (−)-epicatechin units linked through C-4β/C-8 bonds have been isolated from hawthorn for the first time, in addition to known procyanidins including dimers B-2, B-4 and B-5, trimers C-1 and epicatechin-(4β→6)-epicatechin-(4β→8)-epicatechin, and tetramer D-1. Human cystic echinococcosis is a highly endemic zoonotic disease in the province of Neuquén, Patagonia Argentina, although a hydatid control programme has been carried out since 1970. Human infection due to Echinococcus canadensis (G6 genotype) is frequent in Neuquén. However, the reservoir for this species remains undetermined in a region where camels are absent. We investigated the fertility, viability and molecular epidemiology of hydatid cysts obtained from local goats, pigs and sheep in order to identify the possible reservoirs of E. canadensis (G6). We also analyzed isolates from infected dogs. A total of 67 isolates were identified by the DNA sequencing of the mitochondrial cytochrome c oxidase subunit 1 gene. Cysts from sheep (n = 16), goats (n = 23) and pigs (n = 18) and adult worms from 10 infected dogs were analyzed. The fertility of the hydatid cysts was 78.6%; 90.4% and 94.4% for sheep, goats and pigs, respectively. We detected E. canadensis (G6) in 21 of 23 goat samples and in 1 dog isolate, E. canadensis (G7) in all the pig isolates, E. granulosus sensu stricto (G3) in 1 sheep and the G1 genotype in 15 sheep, 2 goats and 9 dog samples. The G1 haplotypes included the common sheep strain sequence and 2 microvariants of this sequence. E. granulosus sensu stricto (G3) is described for the first time in South America. We conclude that goats act as reservoir for E. canadensis (G6) in Neuquén, and that control strategies may have to be adapted to local molecular epidemiology to improve the control of parasite transmission.  相似文献

The effect of vasoactive intestinal peptide (VIP) on anterior pituitary hormone release was examined in a variety of in vitro preparations. Synthetic VIP was capable of stimulating increased prolactin (PRL) release from male rat hemipituitaries in doses as low as 10−9 M only when the enzyme inhibitor bacitracin was present in the incubation medium. Natural porcine VIP was similarly capable of stimulating PRL release, but only at higher doses (10−6 M). Additionally, synthetic VIP was capable of stimulating PRL release from dispersed anterior pituitary cells harvested from adult male and lactating female rats and from an enriched population of lactotrophs obtained by unit gravity sedimentation of similar dispersed cells from infantile female rats. No effect of VIP on luteinizing hormone, growth hormone or thyroid stimulating hormone release was seen. These findings taken in concert with the presence of VIP in the hypothalamus, pituitary and hypophyseal portal plasma of the rat suggest a physiological role for VIP in the control of PRL secretion.  相似文献

为研究巴西橡胶树(Hevea brasiliensis)中HbSUT3和HbSUT5基因的功能,采用地高辛标记的RNA探针与橡胶树嫩茎和中脉两种组织切片分别进行RNA原位杂交,对这2种SUT基因在组织中的表达区域与表达特点进行了分析。结果表明,在橡胶树嫩茎中,两个SUT基因主要在树皮的韧皮部和皮层细胞中表达;在中脉中,两个SUT基因在除木质部导管系统外的其它部位均有表达;HbSUT3基因在嫩茎和中脉中的表达量相近,而HbSUT5基因在嫩茎中的表达量远高于中脉。这些揭示HbSUT3和HbSUT5基因可能广泛参与韧皮部装载、蔗糖运输与库细胞供给等活动,同时两个SUT基因也存在功能分化。  相似文献

Until recently the contact loads acting in the glenohumeral joint have been calculated using musculoskeletal models or measured in vitro. Now, contact forces and moments are measured in vivo using telemeterized shoulder implants. Mean total contact forces from four patients during eight activities of daily living are reported here.Lifting a coffee pot (1.5 kg) with straight arm caused an average force of 105.0%BW (%body weight) (range: 90–124.6%BW), while setting down the coffee pot in the same position led to higher forces of 122.9%BW on the average (105.3–153.4%BW). The highest joint contact forces were measured when the straight arm was abducted or elevated by 90° or more, with a weight in the hand. Lifting up 2 kg from a board up to head height caused a contact force of 98.3%BW (93–103.6%BW); again, setting it down on the board led to higher forces of 131.5%BW (118.8–144.1%BW). In contrast to previously calculated high loads, the contact force during passive holding of a 10 kg weight laterally was only 12.3%BW (9.2–17.9%BW), but when lifting it up to belt height it increased to 91.5%BW (87–95%BW).The moments transferred inside the joint at our patients varied much more than did the forces both inter and intra-individually.Our data suggest that patients with shoulder problems or during the first post-operative weeks after shoulder fractures or joint replacements should avoid certain activities encountered during daily living e.g. lifting or holding a weight with an outstretched arm. Some energy-related optimization criteria used in the literature for analytical musculoskeletal shoulder models must now be reconsidered.  相似文献
