The function of the esophagus is mechanical. To understand the function, it is necessary to know how the stress and strain in the esophagus can be computed, and how to determine the stress-strain relationship of the wall materials. The present article is devoted to the issue of determining the incremental elastic moduli in the layers of the esophagus under homeostatic conditions. The esophagus is treated as a two-layered structure consisting of an inner collagen-rich submucosa layer and an outer muscle layer. We adopt a theory based on small perturbation experiments at homeostatic conditions for determination of incremental moduli in circumferential, axial, and cross directions in the two layers. The experiments are inflation, axial stretching, circumferential bending, and axial bending. The analysis takes advantage of knowing the esophageal zero-stress state (an open sector with an opening angle of 59.4 +/- 13.2 deg). The neutral axis was located 27% +/- 1.9%away from the mucosal surface. It is demonstrated that under homeostatic conditions, the incremental moduli are layer and direction dependent. The incremental modulus is the highest in the axial direction. Furthermore, the axial moduli for the two layers are similar, whereas in the circumferential direction, the incremental modulus is a factor of 6 higher in the mucosa-submucosa layer compared to the muscle layer. Hence, the esophagus has to be treated as a composite, anisotropic body. With this additional information, we can then look forward to a vision of truly understanding the mechanical events of the esophagus.  相似文献

The passive mechanical properties of blood vessel mainly stem from the interaction of collagen and elastin fibers, but vessel constriction is attributed to smooth muscle cell (SMC) contraction. Although the passive properties of coronary arteries have been well characterized, the active biaxial stress-strain relationship is not known. Here, we carry out biaxial (inflation and axial extension) mechanical tests in right coronary arteries that provide the active coronary stress-strain relationship in circumferential and axial directions. Based on the measurements, a biaxial active strain energy function is proposed to quantify the constitutive stress-strain relationship in the physiological range of loading. The strain energy is expressed as a Gauss error function in the physiological pressure range. In K(+)-induced vasoconstriction, the mean ± SE values of outer diameters at transmural pressure of 80 mmHg were 3.41 ± 0.17 and 3.28 ± 0.24 mm at axial stretch ratios of 1.3 and 1.5, respectively, which were significantly smaller than those in Ca(2+)-free-induced vasodilated state (i.e., 4.01 ± 0.16 and 3.75 ± 0.20 mm, respectively). The mean ± SE values of the inner and outer diameters in no-load state and the opening angles in zero-stress state were 1.69 ± 0.04 mm and 2.25 ± 0.08 mm and 126 ± 22°, respectively. The active stresses have a maximal value at the passive pressure of 80-100 mmHg and at the active pressure of 140-160 mmHg. Moreover, a mechanical analysis shows a significant reduction of mean stress and strain (averaged through the vessel wall). These findings have important implications for understanding SMC mechanics.  相似文献

Experiments were performed to study biaxial anisotropy in relaxed canine carotid arteries subjected to a wide range of longitudinal lengths and transmural pressures. In order to encompass the range of vessel lengths which occurs in situ, 120 carotid arteries were studied at 20%, 40%, 60% and 80% extension relative to retracted vessel length. These studies were performed in systematically randomized order. At each length, the vessels were pressurized in steps up to 200 mmHg transmural pressure, or until the traction force fell to zero. Results showed that longitudinal extension markedly increased the longitudinal elastic modulus, but had only a slight effect on the circumferential modulus. At physiological pressures, the vessels were nearly isotropic at about 70% longitudinal extension, but were anisotropic at shorter lengths. Estimates of the anisotropic circumferential modulus by a number of simplified methods revealed that use of a 'pressure-elastic modulus' (Ep) underestimated the anisotropic modulus by 80%, but was extremely consistent. Therefore when suitably corrected, Ep could be used to reliably estimate the anisotropic modulus of carotid arteries over a wide range of pressures and vessel lengths.  相似文献

For a right coronary artery, three-dimensional stress and strain distributions at a physiological intraluminal pressure and an axial extension ratio were computed on the basis of a two-layer elastic model. To validate the model, curves of external radius versus pressure and of axial force versus pressure were computed for three axial extension ratios. To analyze mechanical properties, stress-free configurations of media and adventitia, and the constitutive law of each layer in literature, were used. The present study showed that the peak circumferential stress and the peak axial stress appear in the media at the boundary between the media and adventitia. This result is due to the opening angle of the media being larger than π (rad) and the larger value of a material constant of the strain energy function for the media than for the adventitia. The circumferential stress and strain were discontinuous at the boundary. On the other hand, the radial stress was continuous at the boundary because of the boundary condition for stress. The circumferential stress and axial stress in the adventitia were almost uniformly distributed, and smaller than in the media. The residual stress and strain were also computed. The circumferential residual stress and strain were almost linearly distributed in each layer, although discontinuity appeared at the boundary between the two layers.  相似文献

The importance of mechanical stresses and strains has become well recognized in vascular physiology and pathology. To compute the stress and strain on the various components of the vessel wall, we must know the constitutive equations for the different layers of the vessel wall. The objective of the present study is to determine the constitutive equation of the coronary artery treated as a two-layer composite: intima-media and adventitial layers. Twelve hearts were obtained from a local slaughterhouse, and the right coronary artery and left anterior descending artery were dissected free from the myocardium. The vessel wall was initially mechanically tested biaxially (inflation and axial extension) as a whole (intact wall) and subsequently as intima-media or adventitial layer. A Fung-type exponential strain energy function was used to curve fit the experimental data for the intact wall and individual layers for the right coronary artery and left anterior descending artery. Two methods were used for the determination of material constants, including the Marquardt-Levenberg nonlinear least squares method and the genetic algorithm method. Our results show that there were no statistically significant differences in the material constants obtained from the two methods and that either set of elastic constants results in good fit of the data. Furthermore, at an in vivo value of axial stretch ratio, we find that the stiffness is as follows: intima-media > intact > adventitia. These results underscore the composite nature of coronary arteries with different material properties in each layer. The present results are necessary for analysis of coronary artery mechanics and to provide a fundamental understanding of vessel physiology.  相似文献

The normal coronary artery consists of two mechanically distinct layers: intima-media and adventitia. The objective of this study is to establish a two-layer three-dimensional (3-D) stress-strain relation of porcine coronary arteries. Experimental measurements were made by a series of biaxial tests (inflation and axial extension) of intact coronary arteries and, subsequently, their corresponding intima-media or adventitia layer. The Fung-type exponential strain energy function was used to describe the 3-D strain-stress relation for each layer and the intact wall. A genetic algorithm was used to determine the material constants in the Fung-type constitutive equation by curve fitting the experimental data. Because one layer must be sacrificed before the other layer can be tested, the material property of the missing layer was computed from the material constants of the intact vessel and the tested layer. A total of 20 porcine hearts were used: one group of 10 hearts for the left anterior descending artery and another group of 10 hearts for the right coronary artery. Each group was further divided into two subgroups of five specimens tested for the intact wall and the intima-media layer and for the intact wall and the adventitia layer. Our results show statistically significant differences in the material properties of the two layers. The mathematical model was validated by experimental stress-strain data for individual layers. The validated 3-D constitutive model will serve as a foundation for formulation of layer-specific boundary value problems in coronary physiology and cardiology.  相似文献

A novel approach is developed for mathematically modeling the variability observed in experimentally determined elastic moduli of longitudinally oriented fibrous tissues such as ligaments and tendons. The elastic modulus of these tissues is modeled with a rule of mixtures (ROM) where each parameter (fibril and matrix moduli and fibril volume fraction) is assumed to be an independent random variable. A joint density function formed from the independent densities results in a probabilistic ROM (pROM). This pROM is used to generate a distribution of moduli which agrees well with moduli determined from tests of rabbit medial collateral ligaments (Woo and Ohland, 1994, Unpublished experimental data as gift). Minimizing the error between the pROM and experimental distributions resulted in an integrated error of 9% for a constrained set of independent distribution parameters derived from the literature. This pROM thus incorporates microstructural observations (fibril and matrix moduli and fibril volume fraction) to partially explain the experimentally observed variability in a macroscopic property (tissue modulus).  相似文献

Oscillatory and pulsatile flows of Newtonian fluids in straight elastic tubes are simulated numerically with the aid of Ling and Atabek's "local flow" assumption for the nonlinear convective acceleration terms. For the first time, a theoretical assessment of the local flow assumption is presented, and the range of validity of the assumption is estimated by comparison with perturbation solutions of the complete flow problem. Subsequent simulations with the local flow model indicate that the flow field and associated wall shear stress are extremely sensitive to the phase angle between oscillatory pressure and flow waves (impedance phase angle). This phase angle, which is a measure of the wave reflection present in the system, is known to be altered by arterial disease (e.g., hypertension) and vasoactive drugs. Thus, the paper elucidates a mechanism by which subtle changes in systemic hemodynamics (i.e., phase angles) can markedly influence local wall shear stress values.  相似文献

The model of the lung as an elastic continuum undergoing small distortions from a uniformly inflated state has been used to describe many lung deformation problems. Lung stress-strain material properties needed for this model are described by two elastic moduli: the bulk modulus, which describes a uniform inflation, and the shear modulus, which describes an isovolume deformation. In this study we measured the bulk modulus and shear modulus of human lungs obtained at autopsy at several fixed transpulmonary pressures (Ptp). The bulk modulus was obtained from small pressure-volume perturbations on different points of the deflation pressure-volume curve. The shear modulus was obtained from indentation tests on the lung surface. The results indicated that, at a constant Ptp, both bulk and shear moduli increased with age, and the increase was greater at higher Ptp values. The micromechanical basis for these changes remains to be elucidated.  相似文献

The development of tolerance and dependence has traditionally been considered an integral aspect of the drug addiction process, and opiate dependence has been studied extensively as a model system in this regard. However, recent emphasis on the positive reinforcing properties of drugs has led to the suggestion that tolerance, dependence, and withdrawal may be of secondary or even negligible importance in motivating compulsive drug use. The current article argues for an integrated view of addiction in the form of a homeostatic neuroadaptation model which emphasizes the motivational significance of both the positive affective state produced by opiates and the negative affective state characteristic of drug withdrawal. The model is supported by evidence at both the behavioral and neural systems levels of analysis. Understanding the important distinction between somatic and affective components of opiate withdrawal is key to recognizing the factors which contribute to the motivational significance of opiate dependence and withdrawal. In addition, the critical role of conditioning processes in the maintenance of compulsive drug use and relapse after periods of abstention is discussed. Finally, it is argued that both the positive reinforcement produced by acute administration of a drug and the negative affective state produced by withdrawal are common to multiple classes of abused drugs, suggesting that an understanding of homeostatic neuroadaptation within motivational systems provides a key to the etiology, treatment and prevention of drug addiction. We develop equilibrium fluctuation formulae for the isothermal elastic moduli of discrete biomembrane models at different scales. We account for the coupling of large stretching and bending strains of triangulated network models endowed with harmonic and dihedral angle potentials, on the basis of the discrete-continuum approach presented in Schmidt and Fraternali (J Mech Phys Solids 60:172-180, 2012). We test the proposed equilibrium fluctuation formulae with reference to a coarse-grained molecular dynamics model of the red blood cell (RBC) membrane (Marcelli et al. in Biophys J 89:2473-2480, 2005; Hale et al. in Soft Matter 5:3603-3606, 2009), employing a local maximum-entropy regularization of the fluctuating configurations (Fraternali et al. in J Comput Phys 231:528-540, 2012). We obtain information about membrane stiffening/softening due to stretching, curvature, and microscopic undulations of the RBC model. We detect local dependence of the elastic moduli over the RBC membrane, establishing comparisons between the present theory and different approaches available in the literature.  相似文献

Since atherosclerotic lesions tend to be localized at bends and branching points, knowledge of wall shear rate patterns in models of these geometries may help elucidate the mechanism of atherogenesis. This study uses the photochromic method of flow visualization to determine both the mean and amplitude of the wall shear rate waveform in straight and curved elastic arterial models to demonstrate the effects of curvature, elasticity, and the phase angle between the flow and pressure waveforms (impedance phase angle). Under sinusoidal flow conditions characteristic of large arteries, the mean shear rate at the inner wall of the curved tube is reduced 40–56% from its steady flow value, depending on the phase angle. Wall shear rate amplitudes in the curved tube are significantly reduced by wall motion (36–55% of the Womersley amplitude for a straight rigid tube). The shear rate amplitude at the outer wall decreases 30% as the phase angle is reduced from −20° to −66°, while the shear rate amplitude at the inner wall increases 45%. As a result, the oscillatory nature of flow at the outer wall decreases with decreasing negative phase angle, but flow at the inner wall becomes much more oscillatory. At large negative phase angles, characteristic of hypertension or vasoactive agents, the shear rate at the inner wall has a small mean and cycles through positive and negative values; the shear rate at the outer wall remains positive throughout the flow cycle. Thus, the impedance phase angle could affect atherogenesis along the inner wall if temporal and directional changes in wall shear rate play a role.  相似文献

In vitro experiments were conducted to measure the oscillatory flow pressure gradient along an elastic tube in order to assess the recent nonlinear theory of Wang and Tarbell. According to this theory, in an elastic tube with oscillatory flow, the mean (time-averaged) pressure gradient cannot be calculated using Poiseuille's law. The effect of wall motion creates a nonlinear convective acceleration, and an induced mean pressure gradient is required to balance the convective acceleration. The induced mean pressure gradient depends on the diameter variation over a cycle, the pulsatility and unsteadiness of the flow, and the phase difference between the pressure wave form and the flow wave form. The amplitude of the pressure gradient also depends on these parameters and may deviate significantly from Womersley's rigid tube theory. A flow loop was constructed to produce oscillatory flow in an elastic tube. Flow wave forms were measured with an ultrasonic flow probe, and ultrasonic diameter crystals were used to measure wall movement. A special device for pressure drop measurement was constructed using Millar catheter tip transducers to obtain both forward and backward pressure drops that were then averaged. This averaging method eliminated the static error of the pressure transducers. The pressure-flow phase angle was varied by clamping a distal elastic section at various locations. Pressure gradients were obtained for a range of phase angles between −55 ° and +49 °. The mean and amplitude of the measured pressure gradient were compared to theoretical values. Both positive and negative induced mean pressure gradients were measured over the range of phase angles. The measured pressure gradient amplitudes were always lower than predicted by Womersley's rigid tube theory. The experimental means and amplitudes are in good agreement with the elastic tube theoretical values. Thus, the experiments verify the theory of Wang and Tarbell.  相似文献

The anatomical variation of orthotropic elastic moduli of the cancellous bone from three human proximal tibiae was investigated using an ultrasonic technique. With this technique, it was possible to measure three orthogonal elastic moduli and three shear moduli from cubic specimens of cancellous bone as small as 8 mm per side. Correlation with mechanical tensile testing has shown this technique to offer a precise measure of cancellous modulus (Eten = 0.94Eult + 144.6 MPa, r2 = 0.96, n = 34). The cancellous bone of the proximal tibia was found to be very inhomogeneous, with the axial modulus ranging between 340 and 3350 MPa. A course map is presented, showing measured Young's moduli as a function of anatomical position. The anisotropy of the cancellous bone, determined by the relative differences between the three orthogonal moduli, was shown to be relatively constant over the entire range of cancellous densities tested. The relationship between the axial elastic modulus and the apparent density was found to be approximately linear, as reported by others for proximal tibial cancellous bone.  相似文献

Microtubules play a number of important mechanical roles in almost all cell types in nearly all major phylogenetic trees. We have used a molecular mechanics approach to perform tensile tests on individual tubulin monomers and determined values for the axial and circumferential moduli for all currently known complete sequences. The axial elastic moduli, in vacuo, were found to be 1.25 GPa and 1.34 GPa for α- and β-bovine tubulin monomers. In the circumferential direction, these moduli were 378 MPa for α- and 460 MPa for β-structures. Using bovine tubulin as a template, 269 homologous tubulin structures were also subjected to simulated tensile loads yielding an average axial elastic modulus of 1.10 ± 0.14 GPa for α-tubulin structures and 1.39 ± 0.68 GPa for β-tubulin. Circumferentially the α- and β-moduli were 936 ± 216 MPa and 658 ± 134 MPa, respectively. Our primary finding is that that the axial elastic modulus of tubulin diminishes as the length of the monomer increases. However, in the circumferential direction, no correlation exists. These predicted anisotropies and scale dependencies may assist in interpreting the macroscale behavior of microtubules during mitosis or cell growth. Additionally, an intergenomic approach to investigating the mechanical properties of proteins may provide a way to elucidate the evolutionary mechanical constraints imposed by nature upon individual subcellular components.  相似文献

Thin layers of gels with mechanical properties mimicking animal tissues are widely used to study the rigidity sensing of adherent animal cells and to measure forces applied by cells to their substrate with traction force microscopy. The gels are usually based on polyacrylamide and their elastic modulus is measured with an atomic force microscope (AFM). Here we present a simple microfluidic device that generates high shear stresses in a laminar flow above a gel-coated substrate and apply the device to gels with elastic moduli in a range from 0.4 to 300 kPa that are all prepared by mixing two components of a transparent commercial silicone Sylgard 184. The elastic modulus is measured by tracking beads on the gel surface under a wide-field fluorescence microscope without any other specialized equipment. The measurements have small and simple to estimate errors and their results are confirmed by conventional tensile tests. A master curve is obtained relating the mixing ratios of the two components of Sylgard 184 with the resulting elastic moduli of the gels. The rigidity of the silicone gels is less susceptible to effects from drying, swelling, and aging than polyacrylamide gels and can be easily coated with fluorescent tracer particles and with molecules promoting cellular adhesion. This work can lead to broader use of silicone gels in the cell biology laboratory and to improved repeatability and accuracy of cell traction force microscopy and rigidity sensing experiments.  相似文献

The sheep is considered to be a suitable model for cardiovascular surgery because of its ease of handling, size, and vascular anatomy which bears close resemblance to the human. Several difficulties, however, have limited the use of the sheep for such a purpose-mainly the high infection rate resulting from median sternotomy incision and its susceptibility to intractable ventricular fibrillation (VF) with the slightest manipulation of the heart, and even the risk of short periods of myocardial ischaemia. We have used the sheep model extensively to perform coronary artery bypass surgery and were successful in overcoming these difficulties. Fifty-seven adult female sheep were used to test a new anastomotic device for the creation of a sutureless connection between venous and arterial grafts and the coronary arteries. The study required full access to the heart and great vessels and mobilization of one of the internal mammary arteries. Changing to the left lateral thoracotomy (LLT) approach solved the initial fatal problems of postoperative infected median sternotomy incisions. Aggressive prophylactic treatment with anti-arrhythmic drugs, maintenance of normothermia and myocardial preconditioning rendered the heart much less vulnerable to manipulations and ischaemia. These measures have reduced the mortality rate from 45% to 0% (P <0.0001). With specific operative techniques and pharmaceutical interventions, the sheep can be effectively and safely used as a model for coronary artery surgery.  相似文献
