The properties of sodium fusidate micelles were determined by a spectral shift technique, surface tension measurements, and ultracentrifugal analysis. The critical micellar concentrations, mean molecular areas, and apparent aggregation numbers were estimated as a function of the concentration of counterion (0.001-1.0 m Na(+)) at 20 degrees C. The critical micellar concentrations were studied over a temperature range of 10 degrees C to 40 degrees C at one counterion concentration (0.001 m Na(+)), and from these data the standard thermo-dynamic functions of micellization were calculated. The ability of sodium fusidate solutions to solubilize the insoluble swelling amphiphiles, lecithin and monoolein, was investigated, and the results were compared with the solubilizing properties of sodium taurocholate. The critical micellar concentrations of sodium fusidate approximated those of sodium taurocholate. The values fell in the range of 1.44-4.56 mm, varying with the technique used, counterion concentration, and temperature. The percentage of counterions bound to fusidate micelles in water, calculated from the log critical micellar concentration-log Na(+) curve, was estimated to be negligible, which compares with sodium taurocholate micelles. The critical micellar concentration of sodium fusidate exhibited a minimum at 20 degrees C, a phenomenon observed with other ionic detergents and with bile salts. Micelle formation in sodium fusidate solutions was shown to be primarily entropy-driven at 10 degrees and 20 degrees C, whereas at 30 degrees and 40 degrees C the enthalpy factor predominated. From the surface tension measurements the molecular areas of sodium fusidate and sodium taurocholate were calculated. The mean molecular area of fusidate was 101 A(2), whereas sodium taurocholate possessed a molecular area of 88 A(2). It was demonstrated that the sodium fusidate molecule, like a bile salt molecule, lies with its longitudinal axis horizontal at an air-water interface. The apparent aggregation number of sodium fusidate micelles increased from 5 to 16 as the concentration of counterion increased from 0.01 to 0.60 m Na(+). These values are slightly larger than the corresponding aggregation numbers of sodium taurocholate micelles.  相似文献

The complexation of two steroid antibiotics of the fusidane family, sodium fusidate and potassium helvolate, by beta-CD and gamma-CD has been studied by using 1D and 2D-NMR techniques. Both guests form 1:1 complexes with gamma-CD and 1:2 (guest:cyclodextrin) complexes with beta-CD. Thus, both antibiotics behave as monotopic and ditopic guests when they are complexed by gamma-CD and beta-CD, respectively. Both steroids enter into the cavity of the gamma-CD by the side chain, reaching the central region of the steroid (rings C and D), whereas the A and B (partially) rings remain outside. For beta-CD complexes, ROESY spectra show a remarkable absence of interactions of the protons of the C and D rings, whereas clear interactions corresponding to the side chain, and A and B rings are observed. The obtained equilibrium constants (see previous paper) are discussed in terms of the structures proposed for the complexes. NMR spectra of sodium fusidate are revised, and a full assignment of the 1H and 13C NMR spectra is presented for potassium helvolate.  相似文献

The use of a slowly released oral preparation of sodium chloride is described. It was given to patients and athletes to treat or prevent acute and chronic sodium chloride deficiency. Gastrointestinal side effects were not encountered after the ingestion of up to 500 mEq in one day or 200 mEq in 10 minutes.  相似文献

Methods are described for extracting the rate constants, governing the transitions between states, for a generalized, linear three discrete state model for the Na conductance,g Na. It is shown that the problem cannot be solved given only the time course ofg Na during some test potential step with fixed initial conditions. However, if the effects of any two durations of some conditioning potential step on the peakg Na during the test step are taken into account, and the physically required assumption that the steady state is an equilibrium state is made, then the rate constants for both conditioning and test potentials can be solved for simultaneously. These methods were applied to thev 5 model of Goldman (5) in which theg Na(t) was described in terms of a generalized linear second order variable. After extensive computations involving some 20,000 combinations of experimentally determined input values, it was not found possible to extract a set of rate constants that were all real and positive as is required. It was concluded that if the transitions between states are first order and there are no subunit interactions, then the Na gating unit displays more than three states.  相似文献

The review is concerned with three Na(+)-dependent biotin-containing decarboxylases, which catalyse the substitution of CO(2) by H(+) with retention of configuration (DeltaG degrees '=-30 kJ/mol): oxaloacetate decarboxylase from enterobacteria, methylmalonyl-CoA decarboxylase from Veillonella parvula and Propiogenium modestum, and glutaconyl-CoA decarboxylase from Acidaminococcus fermentans. The enzymes represent complexes of four functional domains or subunits, a carboxytransferase, a mobile alanine- and proline-rich biotin carrier, a 9-11 membrane-spanning helix-containing Na(+)-dependent carboxybiotin decarboxylase and a membrane anchor. In the first catalytic step the carboxyl group of the substrate is converted to a kinetically activated carboxylate in N-carboxybiotin. After swing-over to the decarboxylase, an electrochemical Na(+) gradient is generated; the free energy of the decarboxylation is used to translocate 1-2 Na(+) from the inside to the outside, whereas the proton comes from the outside. At high [Na(+)], however, the decarboxylases appear to catalyse a mere Na(+)/Na(+) exchange. This finding has implications for the life of P. modestum in sea water, which relies on the synthesis of ATP via Delta(mu)Na(+) generated by decarboxylation. In many sequenced genomes from Bacteria and Archaea homologues of the carboxybiotin decarboxylase from A. fermentans with up to 80% sequence identity have been detected.  相似文献

The effects of meat-curing agents on germination and outgrowth of putrefactive anaerobe 3679h (PA 3679h) spores were studied in microcultures. Nitrite concentrations up to 0.06% at pH 6.0 or between 0.8 and 1% at pH 7.0 allowed emergence and elongation of vegetative cells but blocked cell division. The newly emerged cells then lysed. With more than 0.06% nitrite at pH 6.0 or more than 0.8 to 1% at pH 7.0, the spores lost refractility and swelled, but vegetative cells did not emerge. Even as much as 4% nitrite failed to prevent germination (complete loss of refractility) and swelling of the spores. Sodium chloride concentrations above 6% prevented complete germination (i.e., the spores retained a refractile core). In the presence of 3 to 6% sodium chloride, most of the spores germinated and produced vegetative cells, but cell division was often blocked. Sodium nitrate had no apparent effect on germination and outgrowth at concentrations up to 2%.  相似文献

Bean plants were grown under constant levels of sodium chlorideand sodium sulphate salinity, and under changing levels of sodiumsulphate salinity. Although growth was suppressed similarly by the two types ofsalinity when expressed on an osmotic basis, other parametersshowed different responses according to salinity type. Chloride-salinatedplants have thicker leaves with higher water content than thesulphate and control plants. The relative water content of thesulphate plants was somewhat lower. Transpiration rates weresuppressed more by chloride salinity. Osmotic adjustment seemsto be faster under chloride salinity and was of a differentnature. Chloride accumulated to much higher levels than sulphate.Increase in potassium ion concentration and decrease in calciumion concentration was more pronounced under sulphate salinity,but the total cation concentration in the sap was similar forall treatments. As a result, the inorganic ions' electricalbalance was more negative under chloride salinity. Leaf expansion in the changing-level treatment reflected rapidlythe variation m substrate salinity. However, total yield wassuppressed by the changing treatments to a similar extent asby constant salinity. According to other parameters the changingregime may be considered as a shorter period of exposure tosalinity.  相似文献

Unidirectional Na fluxes from frog''s striated muscle were measured in the presence of 0 to 5 mM sodium azide. With azide concentrations of 2 and 5 mM the Na efflux was markedly stimulated; the Na efflux with 5 mM azide was about 300 per cent greater than normal. A similar increase was present when all but the 5.0 mM sodium added with azide was replaced by choline. 10-5 M strophanthidin abolished the azide effect on Na24 efflux. Concentrations of azide of 1.0 mM or less had no effect on Na efflux. The Na influx, on the other hand, was only increased by 41 per cent in the presence of 5 mM NaN3. From these findings it is concluded that the active transport of Na is stimulated by the higher concentrations of azide. The hypothesis is advanced that the active transport of Na is controlled by the transmembrane potential and that the stimulation of Na efflux is produced as a consequence of the membrane depolarization caused by the azide.  相似文献

Enterococcus hirae grows in a broad pH range from 5 to 11. An E. hirae mutant 7683 lacking the activities of two sodium pumps, Na+-ATPase and Na+/H+ antiporter, does not grow in high Na+ medium at pH above 7.5. We found that 7683 grew normally in high Na+ medium at pH 5.5. Although an energy-dependent sodium extrusion at pH 5.5 was missing, the intracellular levels of Na+ and K+ were normal in this mutant. The Na+ influx rates of 7683 and two other strains at pH 5.5 were much slower than those at pH 7.5. These results suggest that Na+ elimination of this bacterium at acid pH is achieved by a decrease in Na+ entry and a normal K+ uptake.  相似文献

Voltage-gated ion channels are well known for their functional roles in excitable tissues. Excitable tissues rely on voltage-gated ion channels and their auxiliary subunits to achieve concerted electrical activity in living cells. Auxiliary subunits are also known to provide functional diversity towards the transport and biogenesis properties of the principal subunits. Recent interests in pharmacological properties of these auxiliary subunits have prompted significant amounts of efforts in understanding their physiological roles. Some auxiliary subunits can potentially serve as drug targets for novel analgesics. Three families of sodium channel auxiliary subunits are described here: beta1 and beta3, beta2 and beta4, and temperature-induced paralytic E (TipE). While sodium channel beta-subunits are encoded in many animal genomes, TipE has only been found exclusively in insects. In this review, we present phylogenetic analyses, discuss potential evolutionary origins and functional data available for each of these subunits. For each family, we also correlate the functional specificity with the history of evolution for the individual auxiliary subunits.  相似文献
