Exposure of Solanum nigrum L. seeds to high temperatures duringimbibition affected their leakage pattern: (1) The rate of leakageof total electrolytes was markedly increased with elevationof temperature. The increase was highest during the first 3h of imbibition but with a reduced rate thereafter. (2) Leakageof Na+ was almost complete after 6 h of imbibition at both temperatures,but much more Na+ leaked out at 50?C than at 25?C. (3) A markedincrease in leakage of K+ occurred after 24 h of exposure to50?C so that after 96 h three times more K+ leaked out at 50?Cthan at 25?C. (4) After 6 h of imbibition Ca11 and Mg++ continuedto leak out at 25?C and at 50?C at a similar rate. (5) Imbibitionat an elevated temperature induced a marked increase in theleakage of both nucleic acids and proteins. (6) Malate dehydrogenasewas not detected in the leachate at 25?C, but was found after48 h at 50?C. It is assumed that this enzyme was of cytoplasmicorigin, indicating heat damage to membranes. A comparison of mitochondria isolated from 2 and 24 C grown winter wheat (Triticum aestivum L.) and winter rye (Secale cereale L.) seedlings revealed no correlation between changes in swelling and contraction characteristics and extent of cold hardiness. The swelling response changed markedly due to growth at low temperature, but the change was similar for the four cultivars examined. The swelling response was also observed to change rapidly during aging of isolated mitochondria, either at 2 or 24 C. Spontaneously swollen mitochondria, isolated from 24 C grown seedlings, contracted abruptly upon addition of certain oxidizable substrates, but this response was lost when seedlings were transferred from 24 to 2 C. Studies on the effect of various substrates and respiratory inhibitors on the swelling and contraction responses indicate that inhibitors which reduce or stop electron flow through the electron transport chain also inhibit substrate induced mitochondrial contraction.  相似文献

A linear relationship between constant temperatures in the sub-optimaltemperature range and germination rate is shown in both Quercusrobur L and Castanea sativa Mill germinated under nominal darkconditions. The mean base temperature was interpolated for Qrobur as 0 8 ? or 2-4 ?, depending on seed lot provenance, andfor C. sativa as 1 -4? The optimum temperature for germinationin Q. robur was about 20? compared with around 28 ? in C. sativaOver the sub-optimal temperature range the distribution of thermaltimes was log-normal for each population studied their spreadvarying both between Q robur seed lots and between species However,in C. sativa germinated close to the mean base temperature,the distribution in thermal times was reduced Thermal timesto germination were decreased in Q. robur and C sativa by approximately0 3 and 0-5 log-units, respectively, when the pericarp was removed,i.e in the seeds, but the sensitivity of the response remainedrelatively unaltered In both species the germination rate was the same when nominaldark or safe green light conditions were employed during thegermination test. However, at 21 ? Q robur exhibited the highirradiance reaction (HIR) at photon doses above 30mmol m–2d–1. HIR first affected the germination rate by an inhibitionof radicle extension The sensitivity of the response to thermaltime was reduced as photon dose increased. This photo-inhibitionwas exacerbated at supra-optimal temperatures. Period lengths at different temperatures and phase responsecurves at a high temperature (35°C) of circadian conidiationrhythms were examined in 13 temperature-sensitive (un) strainsof Neurospora crassa. Two strains, un-16 and un-18, had longerperiod lengths than the wild-type strain even at permissivetemperatures. Period lengths of six strains, un-4, un-11, un-16,un-18, un-19 and un-22, changed differently from that of thewild-type strain at restrictive temperatures. However, the shapeof phase response curves for high temperature (35°C) for3 h was almost the same for all un strains and the wild-typestrain. We isolated 97 temperature-sensitive mutants with periodlengths from 19.2 to 24.8 h and determined the dependence ontemperature of the period length of the conidiation rhythm foreach mutant. Infection threads were observed abundantly in the root hairsof Lotus corniculatus L., but very rarely in L. hispidus, Desf.,in response to infection by Rhizobium strains 3001 and 3002.Numbers of infections differed between species and strains andwere also affected by temperature. In L. corniculatus all thenodules originated from infection threads, but in L. hispidusmost nodules appeared to originate by direct bacterial penetrationthrough the epidermis, and infected root hairs were very rarelyseen. Both species of Lotus were tolerant to cold temperatures,the minimum temperature for nodulation being 10 ?C. The optimumtemperature for nodulation of L. corniculatus was 20 ?C with3001 and between 27 and 30 ?C with 3002, a few nodules beingformed with both strains at 35 ?C. L. hispidus formed more nodulesthan L. corniculatus and the optimum temperature for both thestrains was between 25 and 27 ?C. No infection threads were seen in root hairs or nodules of Stylosanthesguyanensis (Aubl.) S. W. and S. humilis H.B.K. infected withRhizobium strain CB1552, and all the nodules were formed inthe axils of lateral roots. Optimum temperature for nodulationin S. guyanensis and S. humilis was around 27 ?C; nodulationwas completely inhibited at 15 ?C and very few nodules wereformed at 35 ?C. Both in Lotus and Stylosanthes the transfer of plants from suboptimalto optimal and supraoptimal temperatures increased nodulation.Delayed inoculation and excision of root tips increased nodulation.  相似文献

For a single seed population of each of four species of grainlegume positive linear relationships were shown between temperatureand rate of germination for different fractions (G) of eachpopulation, from a base temperature, Tb(G), at which germinationrate is zero, to an optimum temperature, To(G) at which germinationrate is maximal. At constant temperatures warmer than To(G)there were negative relations (probably linear) between temperatureand rate of germination to the maximum temperature for germination,Tm(G), Within each population Tb(G) did not differ, but it didvary between species, viz.0.0?C, 0.25?C, 4.and 8.5?C for chickpea(Cicer arietinum L.), lentil (Lens culinaris Medic.), soyabean(Glycine max [ Merr.) and cowpea (Vigna unguiculata [L.] Walp.),respectively. In contrast, To(G) varied both within each populationand also between the four species: 80% of seeds in each populationhad To(G) values within the range 31.8?C to 33.8 ?C, 24.0?Cto 24.4?C, 34.0?C to 34.5?C and 33.2?C to >40?C, respectively.Values of Tm(G) were much more vanable: the 80% population rangewas 48 .0?C to 60.8?C for chickpea, 31.8?C to 34.4?C for lentiland 46.8?C to 55.2?C for soyabean; reliable estimates couldnot be made for cowpea, but the results suggest higher and morevariable values of Tm(G) than in the other three species. Atsub-optimal temperatures the distribution of thermal time forthe different fractions of each population was normal, exceptfor lentil where it was log-normal. A single equation is proposedto describe the influence of sub-optimal temperatures on ratesof germination for whole seed populations. At supra-optimaltemperatures, variation in thermal time for the different fractionsof each population was only slight. The implications of thesefindings for the adaptation of grain legume crops to differentenvironments, and for the screening of germplasm, are discussed. Leaf temperatures (T1) of the parasitic plant Striga hermonthicaare substantially below those of the air (Ta), [TaT1]reaching 7 ?C at Ta = 40 ?C. This results from high rates oftranspiration and the consequent evaporative cooling of theleaf. Application of an antitranspirant, which mechanicallyimpedes foliar loss of water vapour, reduced transpiration andstomatal conductance by 40% and 57%, respectively, and reduced[TaT1] to 2 ?C at Ta = 40 ?C. The temperature sensitivityof photosynthesis in the host-parasite association differed,the optima (Topt) being 37.2 and 40.1 ?C for S. hermonthicaand sorghum, respectively. Once Topt had been exceeded in S.hermonthica net photosynthesis declined rapidly, reaching thelethal limit (Tmax) at 42.6 ?C. S. hermonthica is particularlysensitive to high temperatures and antitranspirant-induced overheatingleads to blackening and shrivelling of the leaf after as littleas 4 h at Ta = 40 ?C. Application of an antitranspirant underfield conditions in the Sudan at Ta = 40 ?C resulted in 28%and 67% reductions in transpiration and stomatal conductance,together with a 5 ?C increase in T1, and subsequent leaf death.In addition to these short-term physiological responses, antitranspirantspraying of the arasite increased the grain and straw yieldof the crop by factors of 3.4 and 2.6, respectively. Antitranspirantsmay have potential use as a method of controlling Striga inthe field. Carbon dioxide and water vapour exchanges for single attachedleaves of the temperate C4 grass Spartina townsendii were measuredunder controlled environment conditions in an open gas-exchangesystem. The responses of net photosynthesis, stomatal resistance,and residual resistance to leaf temperature and photon fluxdensity are described. The light and temperature responses ofnet photosynthesis in S. townsendii are compared to informationon these responses in both temperate C3 grasses and sub-tropicalC4 grasses. Adaptation of photosynthesis in this C4 speciesto a cool temperate climate is indicated both by the light andtemperature responses of net photo-synthesis. Unlike the C4grasses examined previously, significant rates of net photosynthesiscan be detected at leaf temperatures below 10?C. Rates of netphotosynthesis equal or exceed those reported for temperateC3 grasses at all of the temperature (5–40?C) and photonflax density (13–2500µmol m–2 s–1) conditionsexamined. Maximum rates of net photosynthesis in S. townsendiiare almost double those reported for C3 herbage grasses. Unliketemperate C3 grasses, the major limitation to net photosynthesisat low leaf temperatures (10?C and below) is the stomatal resistance,showing that the low residual resistance characteristic of C4species is maintained in S. townsendii even at low leaf temperatures.  相似文献

Germination characteristics of groundnut (Arachis hypogaeaL.) and pearl millet (Pennisetum typhoides S. & H.).—J.exp. Bot. 39: 1121–1128. The germination at constant temperature of several genotypesof groundnut and pearl millet was investigated between 0?C and50?C on a thermal gradient plate. Large differences in bothgermination rate and percentage germination were observed inboth species. Base temperatures vary from 8–11.5?C and 8–13.5?Cin groundnut and millet, respectively and optimum temperaturesfrom about 29–36.5?C in both. Maximum temperatures forgermination ranged from 41–47?C. When Lemna gibba G3 was in contact with 10–5 M Ach oreserine, floral response to chilling changed diurnally under[24(0)], as was the case for control cultures exposed to [16(8)].On the other hand, with a rise in temperature, the min-[24(0)]decreased discontinuously in 24-hr units in Ach cultures subjectedto [24(0)], as did the min-[16(8)] in control cultures. Thusin the presence of Ach or eserine, this long-day plant tookthe continuous light regime, i.e. [24(0)], for another typeof long-day regime consisting of a long light period and a shortdark period, e.g. [16(8)]. In Ach cultures, the lower limitof the min-[24(0)] (72 hr) was attained at 22.5?C and remainedunaltered at the higher temperatures examined. The min-[16(8)]for control cultures, however, known to reach its lower limit(48 hr) at 26?C. Sodium lauryl sulphate (10–5 M) and ouabain(10–6 M) also caused a similar diurnal change in temperaturesensitivity of 24(0)] cultures. We surmised that exogenous Ach or inserted dark period modifiesthe relative rates chemical and physical component reactionsinvolved in the floral evocation processes, resulting in therhythmical floral response to chilling. The effects of root age, temperature, and soil water statuson root hydraulic conductivity (LP) were investigated for twocactus species, Ferocactus acanthodes and Opuntia ficus-indica.The volumetric flux density of water was measured for excisedroot segments, either using negative hydrostatic pressures appliedto the proximal end or using reverse flow of water from theroot to the soil. For both species, LP at 20 ?C increased withroot age, average values reaching a maximum of 3.9 ? 10–7m s–1 MPa–1 for F. acanthodes and 5.2 ? 10–7m s–1 MPa–1 for O.ficus-indica at 11 to 17 weeksof age; LP subsequently declined with increasing root age forboth species. LP was maximal at a temperature of about 10 ?Cfor the youngest roots (1–3 weeks), this optimum shiftingto 40 ?C for 8-week-old roots of both species. For older roots(up to 1.5-years-old), LP increased with temperature from 0?C to 50 ?C, with a Q10 of 1.3 between 20 ?C and 30 ?C. At asoil water potential (soil) of –0.016 MPa, root LP wasindependent of the direction of water flow for both species.Depending on root age, LP declined 45- to 500-fold for F. acanthodesand 90- to 800-fold for O.ficus-indica as soil was reduced from–0.016 to –1.06 MPa, consistent with a rectifier-likebehaviour with respect to water movement between soil and roots.Incorporation of such responses into water uptake models shouldlead to a better understanding of root function. Bien venu) was grown for49 d in flowing nutrient solution at pH 6?0 with root temperaturedecrementally reduced from 20?C to 5?C; and then exposed todifferent root temperatures (3, 5, 7, 9, 11, 13,17 or 25?C)held constant for 14 d. The air temperature was 20/15?C day/nightand nitrogen was supplied automatically to maintain 10 mmolm–3 NH4NO3 in solution. Total dry matter production wasexponential with time and similar at all root temperatures givinga specific growth rate of 0?0784 g g–1 d–1. Partitioningof dry matter was influenced by root temperature; shoot: rootratios increased during treatment at 17?C and 25?C but decreasedafter 5 d at 3?C and 5?C. The ratio of shoot specific growthrate: root specific growth rate increased with the ratio ofwater soluble carbohydrates (shoot: root). Concentrations ofwater soluble carbohydrates in shoot and root were inverselyrelated to root temperature; at 3, 5 and 7?C they increasedin stem + petioles throughout treatment, coinciding with a decreasein the weight of tissue water per unit dry matter. These resultssuggest that the accumulation of soluble carbohydrates at lowtemperature is the result of metabolic imbalance and of osmoticadjustment to water stress. Plants were grown for 49 d in flowing nutrientsolution at pH 6?0 with root temperature decrementally reducedfrom 20?C to 5?C; and then exposed to different root temperatures(3, 5, 7, 9, 11, 13, 17 or 25?C) held constant for 14 d. Theair temperature was 20/15?C day/night and nitrogen was suppliedautomatically to maintain 10 mmol m–3 NH4NO3 in solution.Total uptake of nitrogen over 14 d increased threefold between3–13?C but was constant above 13?C. Net uptake of NH4+exceeded that of NO3 at all temperatures except 17?C,and represented 47–65% of the total uptake of nitrogen.Unit absorption rates of NH4+ and of 1?5–2?7 for NO3suggested that NO3 absorption was more sensitive thanNH4+ absorption to temperature. Rates of absorption were relativelystable at 3?C and 5?C compared with those at 17?C and 25?C whichincreased sharply after 10 d. Tissue concentration of N in theshoot, expressed on a fresh weight basis, was independent ofroot temperature throughout, but doubled between 3–25?Cwhen expressed on a dry weight basis. The apparent proportionof net uptake of NO3 that was assimilated was inverselyrelated to root temperature. The influence of storage temperature on the onset of rapid ethyleneproduction was investigated for fruits of Conference pear (Pyruscommunis L.) and five cultivars of apple (Malus domestica Borkh.).The time taken from harvest to rapid ethylene production wasshorter and more uniform at 3 ?C than at 18–20 ?C forConference pears and Golden Delicious apples. Increases in internalethylene concentration, 1-amino cyclopropane-1-carboxylic acidconcentration and ethylene production were simultaneous in GoldenDelicious apples at 3 ?C. When Golden Delicious apples wereheld at 3 ?C for 48 h and then kept at 20 ?C the mean time ofonset of ethylene production was similar to that for applesheld continuously at 20 ?C. However, two periods of 48 h at3 ?C caused earlier ethylene production. Conversely, ethyleneproduction at 3 ?C was delayed by transfer to 20 ?C for twoperiods of 48 h. Cox's Orange Pippin and other apple cultivarstended to show more synchronous ethylene production at 3 ?Cthan at higher temperatures but the mean time of onset was eitherunaffected by temperature or slighdy delayed at lower temperature.Acceleration of the onset of ethylene production by low temperaturewas never observed in Cox's Orange Pippin apples harvested atweekly intervals from 10 August to 17 September. Use of the Weibull Function to Calculate Cardinal Temperatures in Faba Bean   总被引:1,自引:0,他引:1  
The onset of germination of faba bean seeds at constant temperaturewas progressively delayed as that temperature diverged froman optimum of 25.5 ?C. At temperatures below 10 ?C, or above28 ?C, the maximum germination percentage fell to below 90%.There was no germination at 39 ?C. Positive and negative linearrelationships were established between the constant temperaturesand the rates of progress of germination to different percentiles,at sub-optimal and supra-optimal temperatures, respectively.Like germination rates, base temperature (Tb) declined from3.71 to –0.83 ?C as the percentile value increased from10% to 80%. Caution was urged in extrapolating beyond the experimentaldata set. Differences in the ceiling temperature (Tc) with percentilecould not be discerned. Cumulative germination progress curves at each temperature weremodelled by the Weibull, logistic, and cumulative normal distributionfunctions. Cardinal temperatures (Tb and Tc) calculated fromthese data reasonably approximated the actual data. The Weibullfunction demonstrated a good approximation at all percentilelevels, while the logistic and cumulative normal distributionfunctions, as a result of their inherent symmetry, deviatedat the extreme percentiles. It was concluded that the Weibullfunction not only accurately modelled cumulative germinationbut could also be used in the calculation of cardinal temperatures. The germination of pearl millet (Pennisetum typhoides S. &H.) seeds was investigated at constant temperatures between12 ?C and 47 ?C on a thermal gradient plate. The rate of germination increased linearly with temperaturefrom a base Tb to a sharply defined optimum To beyond whichthe rate decreased linearly with temperature, reaching zeroat Tm. The linearity of the response both above and below Toallowed time and temperature to be combined in a thermal timeat which a specified fraction of the seeds germinated. The seismonastic plant Biophytum sensitivum was stimulated electricallyto produce reproducible action potentials. It was found thatnegative voltages only induced a response and that stimulationcould also be achieved by electrostatic means by depositingnegative charge on the leaf surface. A study of the effectsof temperature on the propagation rates of action potentialsin different aged leaves showed that for leaves of all agesthe fastest propagation occurred at 24 ?C. As the temperaturewas decreased or increased from 24 ?C a reduction in the propagationrate occurred. No propagation was found at, or below, 14 ?Cand the propagation rate decreased with increasing leaf age.Both the threshold voltage level and the charge required toproduce action potentials in young leaves was lowest at 24 ?Cand had to be increased at either lower or higher temperaturesto cause leaf closure. The duration of the electrical stimulusrequired to induce action potentials was found to decrease asthe voltage was increased. Mechanisms which could account forthe effects of temperature on action potentials are discussedin relation to the chill-sensitivity of the plant. The diversion of electron flow to the alternate pathway in mitochondriaof chillsensitive callus of Cornus stolonifera was studied withreference to the immediate response of the cells to chilling.Temperatures below 15?C diverted the bulk of respiration tothe alternate path, proportionally as the temperature decreasedto 0?C. The alternate path, however, existed in cells in a suppressedform at temperatures above 15?C. In chill-resistant callus ofSambucus Sieboldiana, no alteration was noted in electron apportionmentbetween the cytochrome path and the alternate path. Thus, thereseems to be a marked difference in the mode of respiration betweensensitive and resistant plant cells at low temperatures. Therespiratory control ratio also markedly declined at temperaturesbelow 15?C in chill-sensitive callus, but a higher respiratorycontrol ratio was observed below 10?C in chill-resistant callus.From these results, an alteration in the regulatory system forelectron apportionment between the two paths is probably themost immediate response of chill-sensitive cells to low temperatures. 1Contribution No. 2154 from the Institute of Low TemperatureScience. Growth parameters of the diatom Astenonella formosa Hass, andits fungal parasite Rhizophydium planktoniacum Canter emend,were measured at five temperatures and six light intensitieswith a 15?9 h light:dark cycle, using laboratory cultures ofboth organisms. With the parameter values obtained, thresholdhost densities were calculated in order to estimate the effectsof light and temperature on survival and epidemic developmentof the parasite The uninfected host reached light-saturatedgrowth rates between 0.917 day1 at 21?C and 0 285 day1at 2?C. Under light limitation the optimum growth temperaturefor Asterionella decreased because of a reduced growth efficiencyGrowth inhibition at high irradiances was only observed at 2?CThe parasite reached the highest zoospore production at 2?Cand saturating irradiances: 30 2 zoospores per sporangium. Thisvalue was consistently reduced by lower irradiances and highertemperatures to only 2.2 zoospores at the opposite light andtemperature extremes Low light conditions depressed also theinfectivity of the zoospores At very low irradiances, they becamecompletely uninfective The light dependence of zoospore productionand infectivity was restricted to light intensities that limitedthe growth rate of the host. The development time of the sporangiaand the mfecti ve lifetime of the zoospores were not affectedby light but only by temperature, and ranged from 19.0 and 121 days respectively at 2?C to 1.9 and 2 1 days at 21?C- Theseeffects result in optimal conditions for the development ofa Rhizophydium epidemic at 11?C and a moderate light limitationof Astenonella At temperature above 7?C, the possibilities forepidemic development are only slightly affected by light andtemperature, except for very low irradiance levels, when thezoospores of the parasite become uninfective. However, below5?C the development of an epidemic is only possible at limitinglight levels.
