Acquisition of desiccation tolerance in soybeans   总被引:10,自引:0,他引:10  
The entry into a desiccation-tolerant state is a major developmental component of seed maturation. Development of desiccation tolerance of embryonic axes of soybean [Glycine max (L.) Merrill cv. Chippewa 64] was studied by measuring changes in electrolyte leakage. germination and relative growth rate after axes were rapidly air-dried to various water contents. Axes acquired the full capacity for germination at 34 days after flowering (DAF). and reached physiological maturity (maximum dry weight) at 48 DAF. When dried to water content h = 0. 08 (g water g−1 dry weight). few axes germinated before 42 DAF. but more than 90% germinated after 48 DAF. However, electrolyte leakage of rehydrated axes showed a linear decline from 30 to 55 DAF. For developing axes there was a critical water content or desiccation threshold. which could be estimated by using the electrolyte leakage method. The threshold of desiccation tolerance decreased gradually from h = 1. 10 to 0. 18 as axes matured from 28 to 55 DAF. The development of desiccation tolerance continued after physiological maturity at 48 DAF. We conclude that the acquisition of desiccation tolerance of soybean axes is a gradual event, rather than an abrupt transition.  相似文献

The sensitivity of a Polish provenance of dormant Corylus avellana seeds to extreme desiccation and cryopreservation in liquid nitrogen (LN, ?196°C) was investigated. No sensitivity to desiccation was observed as seeds readily germinated and exhibited seedling emergence, even when the critical water content (WC) of seeds (nuts devoid of pericarp) was reduced to 0.027 g H2O/g dry mass, g g?1 by drying. Results of germination and seedling emergence tests indicated that seeds tolerated cooling in LN when desiccated to a WC in the range of 0.05–0.10 and 0.08–0.10 g g?1, respectively. The results of this study demonstrate the feasibility of long‐term cryopreservation of European hazelnut seeds. As the seeds of this species have been classified in several different categories (orthodox, suborthodox and recalcitrant), based on their response to desiccation and low temperature, the assignment of the seeds of hazelnut to a specific category is provided and discussed.  相似文献

Some cultured plant cells are able to acquire tolerance to various stresses when they are cultured under suitably controlled conditions. Induction of a high level of desiccation tolerance in suspension-cultured cells of the liverwort Marchantia polymorpha was examined for studying the mechanisms of desiccation tolerance and vitrification at the cellular level. Desiccation tolerance level of cells was very low and the survival rate was less than 10% after exposure to drying below 0.1 g H2O g−1 dry weight (DW). Preculture treatment in 0.5 M sucrose medium was the most effective method for inducing a high level of desiccation tolerance in cells and the survival rate was 87% even after being desiccated to below 0.1 g H2O g−1 DW. Preculture treatment caused alteration of cell structures and accumulation of a large amount of sucrose and newly synthesized proteins in cells. Abundant sucrose and preculture-induced proteins were necessary for full development of desiccation tolerance in the cells. When water content decreased to below 0.1 g H2O g−1 DW, desiccation-tolerant cells that had been precultured were vitrified above 0°C and maintained stable viability. We have succeeded in the induction of desiccation tolerance that allows formation of intracellular glass with cell viability at ambient temperatures by controlling culture conditions, and our results suggest that suspension-cultured cells of M. polymorpha are useful for studying cellular mechanisms for the development of desiccation tolerance and the stabilization of vitrified cells.  相似文献

The relationships between water content of desiccated embryonic axes (using different methods of desiccation), the availability of water determined by differential scanning calorimetry (DSC) analysis and recovery percentage after liquid nitrogen (LN) exposure of Fortunella polyandra embryonic axes were investigated. The objectives were to understand thermal properties of desiccated embryonic axes during cryopreservation and to determine the critical moisture contents for successful cryopreservation of the embryonic axes. Excised embryonic axes were desiccated under laminar air flow (0, 10, 15, 30 and 45 min), over silica gel (0, 5, 15, 30 and 60 min), and ultra-rapidly (0, 5, 10, 20 and 25 min). Desiccation under laminar air flow resulted in an optimal water content of 0.150 gH2O g?1 dw and a survival of 50 % after cryopreservation, while the unfrozen water content (WCu) was 0.126 gH2O g?1 dw. After drying over silica gel, the optimal water content was 0.190 gH2O g?1 dw, where the survival was 40 % after cryopreservation and the WCu was determined as 0.177 gH2O g?1 dw. Using the flash-drying method, the optimal water content was found to be 0.145 gH2O g?1 dw, the survival was 50 % after cryopreservation and the WCu was 0.133 gH2O g?1 dw. Embryonic axes of F. polyandra showed low-to-moderate tolerance to desiccation. The results of the freezing transitions for all the desiccation times and methods showed that the onset temperature and the peak of the mean enthalpy decreased in size with decreasing water content. DSC elucidated the critical moisture contents and the cooling and melt enthalpies for successful cryopreservation of F. polyandra embryonic axes.  相似文献

Desiccation tolerance is initiated in wheat (Triticum aestivum L.) embryos in planta at 22 to 24 d after anthesis, at the time that the embryo water content has decreased from about 73% fresh weight (2.7 g water/g dry weight) to about 65% fresh weight (1.8 g water/g dry weight). To determine if desiccation tolerance is fully induced by the loss of a relatively small amount of water, detached wheat grains were treated to reduce the embryo water content by just a small amount to approximately 69% (2.2 g water/g dry weight). After 24 h of such incipient water loss, subsequently excised embryos were able to withstand severe desiccation, whereas those embryos that had not previously lost water could not. Therefore, a relatively small decrease in water content for only 24 h acts as the signal for the development of desiccation tolerance. Embryos that were induced into tolerance by a 24-h water loss had no detectable raffinose. The oligosaccharide accumulated at later times even in embryos of detached grains that had not become desiccation tolerant, although tolerant embryos (i.e. those that previously had lost some water) contained larger amounts of the carbohydrate. It is concluded that desiccation tolerance and the occurrence of raffinose are not correlated. Immunodetected dehydrins accumulated in embryos in planta as desiccation tolerance developed. Detachment of grains induced the appearance of dehydrins at an earlier age, even in embryos that had not been made desiccation tolerant by incipient drying. It is concluded that a small reduction in water content induces desiccation tolerance by initiating changes in which dehydrins might participate but not by their interaction with raffinose.  相似文献

Germination of Archontophoenix alexandrae seeds and embryos were studied under gradient water content treatments throughout the seed development phases of maturation in 2005 to investigate seed desiccation tolerance and storage characteristics. During the maturation process, seed water content decreased gradually from55 DAF (days after flowering) to 70 DAF, and seeds reached the maximum dry-weight at 90 DAF. Seed germinability appeared after 60 DAF. Seeds germinated with a temperature range from15℃- 40℃ under alternating photoperiod (14 h light, 10 h dark, 12μmol m- 2s - 1 ), while the best germination percentage was obtained between 30℃- 35℃. A maximum germination capacity reached at 70 DAF. However, seed germination was greatly inhibited by light. Desiccation tolerance of seeds and embryos increasedgradually from 55 DAF to 90 DAF and reached the maximum at 90 DAF with a semilethal water content of 0.18 g/g ( seed) and 0.3 g/g ( embryo) respectively. Rapid dehydration maintained higher seed germination percentage than thatof slow dehydration when drying to the same water content. Seeds with without water content treatments failed to germinate after 1 month storage under - 18℃, whereas appropriate desiccation treatment prolonged seed longevity under 4℃, 10℃ and 15℃ storage temperatures. It revealed obviously the recalcitrant characteristics of Archontophoenix alexandrae seeds torage behaviour which are tolerant toward neither deep desiccation nor low temperatures.  相似文献

Freezing and melting transitions of cellular water in embryonic axes and cotyledonary tissues of recalcitrantQuercus rubra(red oak) seeds were compared under slow and rapid cooling conditions. The relevance of desiccation sensitivity (critical water content) and state/phase transition behaviors to cryopreservation was examined. Under a slow to intermediate cooling condition (≤10°C min−1), unfrozen water content in the tissues decreased to less than the critical water content, resulting in a dehydration damage. Under a rapid cooling condition (>100°C min−1) using liquid nitrogen (LN2), freeze-induced dehydration damage could be avoided if the initial water content was >0.50 g g−1dry wt. However, at water content >0.50 g g−1dry wt, the vitrified cellular matrix was highly unstable upon warming at 10°C min−1. These results offered a theoretical explanation on the difficulty for successful cryopreservation of recalcitrant red oak embryonic axes. A complete state/phase transition diagram for red oak axes was constructed, and a vitrification-based cryopreservation protocol that employed predehydration and rapid cooling was examined. State/phase transition behaviors of cellular water are important parameters for cryopreservation; however, vitrification alone was not sufficient for seed tissues to survive the cryopreservation condition.  相似文献

To evaluate the feasibility of long-term cryopreservation of Polish provenances of silver birch (Betula pendula), the sensitivity of conditionally dormant seeds to extreme desiccation and/or the ultra-low temperature of liquid nitrogen (LN; ?196°C) was evaluated. The critical water content (WC) of desiccated seeds and the high-moisture freezing limit of seeds desiccated or moistened to various WCs and frozen for 24 h or for 2 years in LN was also determined. Germination tests revealed no critical WC for seeds [to 0.02 g H2O g?1 dry mass (g g?1)]. Seeds tolerated freezing in LN within specific safe range of WC 0.02–0.23 g g?1 (nuts). Seeds desiccated to the safe WC and stored in LN for 2 years had similar or higher germination as seeds stored at ?3°C for 2 years, depending on provenance. Therefore, long-term cryopreservation of B. pendula seeds in gene banks is feasible.  相似文献

Summary Cotyledonary somatic embryos ofLarix × leptoeuropaea that developed after various maturation times on media containing abscisic acid showed different frequencies of conversion into plants. Drying of these somatic embryos under high relative humidity (RH) before germination improved plantlet recovery and eliminated differences in the performance of somatic embryos matured for different times. However, dehydration of somatic embryos under 98% RH to a water content below that of zygotic embryos excised from mature seeds (0.97 and 1.36 g H2O/g dry weight, respectively) showed a strong positive correlation between longer maturation time and desiccation tolerance. Drying somatic embryos at 4° C under 59% RH for 1 wk resulted in desiccation to a water content of 0.30 g H2O/g dry weight, which was the closest to the hydration state of zygotic embryos in dried, stored seeds (0.20 g H2O/g dry weight). Under this condition, only somatic embryos matured for 5 wk germinated and produced plantlets at a relatively high frequency (73 and 41%, respectively).  相似文献

Water regulation caused by enzymes, such as carbonic anhydrase (CA), changes the water status, making it difficult to diagnose water deficit using leaf water potential (ψL) or stomatal conductance (gs). Therefore, new methods for timely and accurately determining plant water status should be established. In this study, CA activity, ψL, leaf tensity (Td), photosynthetic characteristics and plant growth of Brassica napus L. seedlings under drought and subsequent rewatering were analysed. Results indicated that Td could reflect the plant water status better than ψL or gs and played an important role in the photosynthesis of B. napus. B. napus exhibited good restorability at the 40?g?L?1 polyethylene glycol level. The rewatering strategy for B. napus was excellent at 40?g?L?1 (?0.15?MPa) →20?g?L?1 (?0.11?MPa). Td could be used for the rapid determination of water requirement information in B. napus during winter drought period.  相似文献

对离体玉米胚脱水耐性的变化以及不同脱水速率对其脱水耐性的影响进行了研究。授粉后16d的玉米胚能耐轻微脱水,含水量从1.45降低到0.28gH2Og-1DW时胚的萌发率为100%,但含水量低于0.1gH2Og-1DW时胚死亡。胚的脱水耐性随着发育逐渐加强,表现为电解质渗漏速率逐渐降低,萌发率和幼苗干重逐渐增加。授粉后20d胚内超氧化物歧化酶(SOD)和抗坏血酸过氧化物酶(APX)活性较高,过氧化氢酶(CAT)活性较低;授粉后24d,这些酶的活性与授粉后20d的正好相反。脂质过氧化产物丙二醛(MDA)在种子发育过程中呈下降趋势。不同脱水速率明显地影响胚的脱水耐性:在慢速脱水到含水量0.1~0.18gH2Og-1DW时,胚的萌发率和幼苗干重比快速脱水高,电解质渗漏速率比快速脱水低;在快速脱水条件下胚中的SOD、APX活性和MDA含量也比慢速脱水高;CAT活性的变化不明显。  相似文献

The objective of this study was to investigate Cd phytoremediation ability of Indian mustard, Brassica juncea. The study was conducted with 25, 50, 100, 200 and 400 mg Kg?1 CdCl2 in laboratory for 21 days and Cd concentrations in the root, shoot and leaf tissues were estimated by atomic absorption spectroscopy. The plant showed high Cd tolerance of up to 400 mg Kg?1 but there was a general trend of decline in the root and shoot length, tissue biomass, leaf chlorophyll and carotenoid contents. The tolerance index (TI) of plants were calculated taking both root and shoot lengths as variables. The maximum tolerance (TI shoot = 87.4 % and TI root = 89.6 %) to Cd toxicity was observed at 25 mg Kg?1, which progressively decreased with increase in dose. The highest shoot (10791 μg g?1 dry wt) and root (9602 μg g?1 dry wt) Cd accumulation was achieved at 200 mg kg?1 Cd treatment and the maximum leaf Cd accumulation was 10071.6 μg g?1 dry wt achieved at 100 mg Kg?1 Cd, after 21 days of treatment. The enrichment coefficient and root to shoot translocation factor were calculated, which, pointed towards the suitability of Indian mustard for removing Cd from soil.  相似文献

The moss Physcomitrella patens , a model system for basal land plants, tolerates several abiotic stresses, including dehydration. We previously reported that Physcomitrella patens survives equilibrium dehydration to ?13 MPa in a closed system at 91% RH. Tolerance of desiccation to water potentials below ?100 MPa was only achieved by pretreatment with exogenous abscisic acid (ABA). We report here that gametophores, but not protonemata, can survive desiccation below ?100 MPa after a gradual drying regime in an open system, without exogenous ABA. In contrast, faster equilibrium drying at 90% RH for 3–5 days did not induce desiccation tolerance in either tissue. Endogenous ABA accumulated in protonemata and gametophores under both drying regimes, so did not correlate directly with desiccation tolerance. Gametophores of a Ppabi3a/b/c triple knock out transgenic line also survived the gradual dehydration regime, despite impaired ABA signaling. Our results suggest that the initial drying rate, and not the amount of endogenous ABA, may be critical in the acquisition of desiccation tolerance. Results from this work will provide insight into ongoing studies to uncover the role of ABA in the dehydration response and the underlying mechanisms of desiccation tolerance in this bryophyte.  相似文献

The moss Physcomitrella patens is becoming the model of choice for functional genomic studies at the cellular level. Studies report that Physcomitrella survives moderate osmotic and salt stress, and that desiccation tolerance can be induced by exogenous ABA. Our goal was to quantify the extent of dehydration tolerance in wild type moss and to examine the nature of cellular damage caused by desiccation. We exposed Physcomitrella to humidities that generate water potentials from −4 (97% RH) to −273 MPa (13% RH) and monitored water loss until equilibrium. Water contents were measured on a dry matter basis to determine the extent of dehydration because fresh weights (FW) were found to be variable and, therefore, unreliable. We measured electrolyte leakage from rehydrating moss, assessed overall regrowth, and imaged cells to evaluate their response to drying and rehydration. Physcomitrella did not routinely survive water potentials <−13 MPa. Upon rehydration, moss dried to water contents >0.4 g g dm−1 maintained levels of leakage similar to those of hydrated controls. Moss dried to lower water contents leaked extensively, suggesting that plasma membranes were damaged. Moss protonemal cells were shrunken and their walls twisted, even at −13 MPa. Moss cells rehydrated after drying to −273 MPa failed to re-expand completely, again indicating membrane damage. ABA treatment elicited tolerance of desiccation to at least −273 MPa and limited membrane damage. Results of this work will form the basis for ongoing studies on the functional genomics of desiccation tolerance at the cellular level.  相似文献

In a comparison of six cowpea cultivars, we determined the variation in abscisic acid (ABA) production as an ‘early warning signal’ produced in response to drought stress. By imposing drought only to the upper 20 cm rooting zone, we compared the rates of ABA synthesis relative to (i) total root mass and (ii) inherent variation per unit root mass. We were able to relate the intensity of the stress response to these two factors, and determine which is quantitatively more important as the primary signal indicating responsiveness to drought stress. Plants were grown in 1.2 m long columns and a soil drying treatment imposed in such a way that that upper roots were in dry soil and deep roots in soil at field capacity. Relative water contents (RWC) of stressed plants were similar and not significantly different from those of well watered controls. However, roots accumulated ABA in the dehydrated zone, where root water content ranged from 10–12 g g?1 DW. The soil moisture contents and root ‐water contents in the dry zone were similar for each of the different varieties. However, the ABA contents were significantly different in drought‐stressed (upper) roots and ranged from 7.82 nmol g?1 DW in cv. APC 689 to 16.02 nmol g?1 DW in cv. APC 370, such that for varieties with similar overall root weights (e.g. APC 580 and APC 540) the different ABA contents were related to the capacity for ABA synthesis. The relationship between stomatal conductance and total root ABA was assessed, with a negative relation (r= 0.90, n= 24, P= 0.05) suggesting that the intrinsic capacity of cowpea varieties for ABA synthesis could play an important role in regulating stomatal conductance in a drying soil and provide useful selection criteria for tolerance to drought stress.  相似文献

This study was conducted to investigate the ecotoxicological effects of exposure to copper oxide nanoparticles (CuO NPs) on the gill of the swan mussel Anodonta cygnea using several approaches including qualitative and quantitative histopathology, ultra-morphology (scanning electron microscopy [SEM]) and measures of clearance rate (CR) and bioaccumulation of CuO NPs. Histological alterations in mussels exposed to 0.25 (T1), 2.5 (T2) and 25.0?µg L?1 (T3) CuO NPs for 12 days include changes in the length and form of gill lamellae, changes in inter-lamellar spaces, epithelial hyperplasia, atrophy and tissue rupture. Ultra-morphological changes following CuO NP exposure included epithelial hyperplasia and hypertrophy, epithelial lifting, tissue rupture (water channel fusion) and extensive necrosis of the gill surfaces. IGill (gill damage severity) index values for both histopathological and ultra-morphological data were significantly (P?0.05) higher in T3. The CR of mussels was significantly (P??1 g?1 dry weight]) in comparison to controls (CR?=?108?±?47.14 [L min?1 g?1 dry weight]). CuO NPs accumulated in exposed mussels at all exposure concentrations until day 4, but there was no further change in accumulation levels by the end of the exposure period. The accumulated content of CuO NPs was significantly (P??1 exposure concentration. Based on these results, significant accumulation of CuO NPs in the gills of swan mussel could affect histological and ultra-structural characteristics of this organ and consequently have deleterious impacts on its filtration activity.  相似文献

Wheat seedlings obtained after 2 or 3 days of seed germination in darkness at 20°C (i.e. with a 0.5–0.7 cm long coleoptile) were still viable after drying in darkness in ambient conditions which reduced the shoot moisture content to about 0.30 g H2O g?1 dry mass (DM). Coleoptile and primary leaf growth resumed upon rehydration, but primary roots died and new roots regenerated. In the present work we have investigated whether desiccation tolerance of the shoot (coleoptile and primary leaf combined) was related to some reversible cellular or metabolic changes induced by dehydration. Non‐dehydrated shoots were high in moisture content (4.0–5.0 g H2O g?1 DM) and exhibited an active metabolism as indicated by a high energy charge (EC = 0.85) and cells with well developed mitochondria, endoplasmic reticulum, polysomes and Golgi bodies. Dehydration induced changes in cell membrane properties since it reduced in vivo capacity of the shoot to convert 1‐aminocyclopropane 1‐carboxylic acid (ACC) to ethylene (i.e. ACC oxidase activity). This effect was already observed at 4–5 h of dehydration, namely when shoot moisture content dropped down below about 3.0 g H2O g?1 DM, and ACC‐dependent ethylene production became almost nil when shoot moisture content reached 1.0 g H2O g?1 DM. Dehydration also resulted in decreases in ATP and non‐adenylic triphosphate nucleotide (NTP) contents down to 1–2% of their initial values, and in EC value to 0.20. Concomitant with water loss, sucrose content of the shoot increased and was maximal (about 330 mg g?1 DM, namely three‐fold that of non‐dehydrated organs) after 2 days of drying. Upon rehydration, shoots regained their original moisture content within 3 days, during which they progressively recovered apparent normal metabolism. Reversal of extensive dehydration‐associated cell wall folding occurred between 2 and 3 days of rehydration, when the ultrastructure of coleoptile and primary leaf cells also provided evidence of intensive autophagic activity, indicative of the removal of damaged cell components. Concomitantly, apparently undamaged organelles and endomembranes persisted in the cytoplasm. Restoration of 60–70% of ACC oxidase activity and 80–90% of EC value occurred within 48 and 18 h, respectively. However, the values of the ATP/ADP and NTP/ATP ratios remained lower than in control non‐dehydrated shoots, indicating that not all metabolic deterioration induced by dehydration was completely repaired. Differences in relationships between shoot moisture content and ACC‐oxidase activity or energy metabolism during dehydration and upon rehydration, and cell ultrastruture analyses suggest that desiccation tolerance of wheat seedling shoot is related to mechanisms involved in the maintenance of cell structure during water loss and the cell capacity to repair the dehydration damage.  相似文献

The present research focused on enhancing the production of wedelolactone through cell suspension culture (CSC) in Eclipta alba (L.) Hassk. With an aim of attaining a sustainable CSC, various plant growth regulators, elicitors and agitation speed were examined. Nodal segments of in vitro propagated plantlets induced the maximum percentage (93.47?±?0.61%) of callus inoculated on Murashige and Skoog (MS) medium fortified with picloram (2 mg L?1). The growth kinetics of CSC exhibited a sigmoid pattern with a lag phase (0–6 days), a log phase (6–18 days), a stationary phase (18–24 days) and then death phase thereafter. The highest biomass accumulation in CSC with 7.09?±?0.06 g 50 mL?1 fresh weight, 1.52?±?0.02 g 50 mL?1 dry cell weight, 1.34?±?0.01?×?106 cell mL?1 total cell count and 57.00?±?0.58% packed cell volume was obtained in the liquid MS medium supplemented with 1.5 mg L?1 picloram plus 0.5 mg L?1 kinetin at 120 rpm. High performance thin layer chromatography confirmed that yeast extract (biotic elicitor) at 150 mg L?1 accumulated more CSC biomass with 1.22-fold increase in wedelolactone (288.97?±?1.94 µg g?1 dry weight) content in comparison to the non-elicited CSC (237.78?±?0.04 µg g?1 dry weight) after 120 h of incubation. Contrastingly, methyl jasmonate (abiotic elicitor) did not alter the biomass but increased the wedelolactone content (259.32?±?1.06 µg g?1 dry weight) to an extent of 1.09-fold at 100 µM. Complete plantlet regeneration from CSC was possible on MS medium containing N6-benzyladenine (0.75 mg L?1) and abscisic acid (0.5 mg L?1). Thus, the establishment of protocol for CSC constitutes the bases for future biotechnological improvement studies in this crop.  相似文献

Abstract

This study reports the combined use of a rhamnolipid type biosurfactant (BS) along with phytoremediation and bioaugmentation (BA) for bioremediation of hydrocarbon-contaminated soils. Bacterial isolates obtained from hydrocarbon contaminated soil were screened for rhamnolipid production and isolate BS18, identified as Shewanella seohaensis, was selected for bioremediation experiments. Growth of BS18 in mineral salt medium (MSM) with diesel oil as the carbon source showed a maximum biomass of 8.2?g L?1, rhamnolipid production of 2.2?mg g?1 cell dry weight, surface tension reduction of 28.6?mN/m and emulsification potential (EI24%) of 65.6. Characterization of rhamnolipid based on Fourier transmittance infrared (FTIR) analysis confirmed the presence of OH, CH2/CH3, C=O, and COO stretching vibrations, respectively, which are distinctive features of rhamnolipid type BSs. In bioremediation experiments, the lowest hydrocarbon concentration of 2.1?mg g?1 of soil for non-sterilized soil and 4.3?mg g?1 of soil for sterilized soil was recorded in the combined application of rhamnolipid, phytoremediation, and BA. Production of lipid from oleaginous yeast using starch as a carbon source is not a common practice; therefore, the purpose of this investigation was to explore the capability of starch assimilating microbes to produce oil, which was determined in terms of biomass weight, productivity, and lipid yield. Saccharomyces pastorianus, Rhodotorula mucilaginosa, Rhodotorula glutinis, and fungal isolate Ganoderma wiiroense were screened for the key parameters. The optimization was also performed by one-factor-at-a-time approach. Considering the specific yield of lipid and cell dry weight yield, R. glutinis and R. mucilaginosa showed superiority over other strains. G. wiiroense, a new isolate, would also be a promising strain for starch waste utilization in terms of extracellular and intracellular specific yield of lipids. Extracellular specific yield of lipid was highest in R. glutinis culture (0.025?g?g?1 of biomass) followed by R. mucilaginosa (0.022?g?g?1 of biomass) and G. wiiroense (0.020?g?g?1 of biomass). Intracellular lipid was again highest in R. glutinis (0.048?g?g?1 of biomass). The most prominent fatty acid methyl esters among the lipid as detected by GC-MS were saturated lipids mainly octadecanoic acid, tetradecanoate, and hexadecanoate. Extracellular lipid produced on starch substrate waste would be a cost-effective alternative for energy-intensive extraction process in biodiesel industry.  相似文献
