The effect of gramicidin C added to the medium at various periods of cultivation in concentrations of 20, 40 and 100 gamma/ml on sporulation of P+-variant of Bac. brevis var. GB was studied. The most effective increase in the sporulation rate and percentage of the cells germinating into the spores was observed on addition of the antibiotic to the medium in amounts of 20 and 40 gamma/ml in 13 hours of the culture development. The amount of gramicidin C during sporulation decreased and partially passed into the spores which did not differ after germination from those of P+-variant grown on the synthetic medium with glucose and without preliminary addition of the antibiotic. Addition of gramicidin C in an amount of 100 gamma/ml at the end of the lag phase, i.e. 4 hours after the culture inoculation suppressed sporulation and had no effect on growth of the cells of its own producing organism.  相似文献

Gramicidin S is known to prolong the outgrowth stage of spore germination in the producing culture. Bacillus brevis strain Nagano and its gramicidin S-negative mutant, BI-7, were compared with respect to cell-surface hydrophobicity and germination of their spores. Parental spores were hydrophobic as determined by adhesion to hexadecane, whereas mutant spores showed no affinity to hexadecane. Addition of gramicidin S to mutant spores resulted in a high cell surface hydrophobicity and a delay in germination outgrowth. The hydrophobicity of parental spores was retained throughout most of the germination period. Hydrophobicity was lost as outgrowing spores entered into the stage of vegetative growth. The data indicate that gramicidin S is responsible for the hydrophobicity of B. brevis spores. It is suggested that in making spores hydrophobic, the antibiotic plays a role in concentrating the spores at interfaces where there is a higher probability of finding nutrients for germination and growth.Abbreviation GS Gramicidin S  相似文献

Rates of commitment to germinate and germination of Bacillus subtilis spores with mixtures of low concentrations of germinants acting on different germinant receptors (GRs) were much higher than the sums of the rates of commitment and germination with individual germinants alone. This synergism with mixtures of nutrient germinants was not seen with spores lacking GRs responsible for recognizing one or several components of the germinant mixtures and was not eliminated by either a gerD mutation or overexpression of one of the GRs involved in this synergism. This synergism was also not seen between the germinant L-valine, which acts via a GR, and the germinant dodecylamine, which does not act via any GR. These results indicate that spores not only integrate but can also amplify signals from multiple germinants and multiple GRs in determining rates of commitment and overall spore germination. This amplification can be explained by a simple mechanism in which a single signal integrator triggers germination above an accumulation threshold. Direct cooperative action between GRs may further add to the synergism seen in germination triggered by multiple GRs. Further experiments and modeling are required to determine the relative contributions of these different mechanisms.  相似文献

Stereoisomeric alanylalanine (Ala-Ala) derivatives were examined for their effects on germination of Bacillus thiaminolyticus spores. L-Ala-L-Ala and L-Ala-glycine were effective in inducing germination, and their activities were completely inhibited by D-Ala. L-Ala-D-Ala and glycine-D-Ala competitively prevented L-Ala-induced germination. Sarcosine- or beta-Ala-containing L-alanyldipeptides and eight kinds of alanyltripeptides did not show any detectable effect on germinability or any inhibitory effect. No detectable amounts of Ala were found in germination exudates when alanylpeptides were incubated with spores. The ability of these peptides to induce or inhibit germination depends on their steric conformation and a certain distance between the primary amino group and the free carboxyl groups. Involvement of L-Ala dehydrogenase in the initiation of germination is unlikely because L-Ala-L-Ala was not a substrate and L-Ala-D-Ala was not an effective inhibitor of enzyme activity.  相似文献

The phenolic compounds extracted from olives with ethyl acetate inhibited germination and outgrowth of Bacillus cereus T spores. Purified oleuropein, a well-characterized component of olive extract, inhibited these processes also. The addition of oleuropein and olive extracts 3 or 5 min after germination began, immediately decreased the rate of change of phase bright to phase dark spores and delayed significantly outgrowth.  相似文献

The review deals with recent results and problems of gene expression during germination of Bacillus spores. Three problems were selected: 1. The activation of metabolism as a prerequisite for the synthesis of nucleic acids and proteins. 2. The activation of nucleic acid and protein synthesis during germination. 3. The gene expression programme of germinating spores. Using the highly sensitive two-dimensional polyacrylamide gel analysis three major classes of proteins were distinguished, depending on the time of onset and duration of their syntheses: a) proteins made throughout germination (main class), b) proteins whose synthesis started only after a lag phase and then continued throughout germination, and c) proteins which are synthesized only during the early phases of germination. The programme of protein synthesis is an indicator for the control of gene expression during germination. The regulation of expression of these major gene groups during spore outgrowth is discussed.  相似文献

We have confirmed the finding of Murray et al. [Lett Appl Microbiol 1: 63–65, 1985] that most of theBacillus brevis spores undergoing the gramicidin S-delayed outgrowth stage of germination are killed by gramicidin S, the antibiotic produced during sporulation. We found, however, that 1% of the population resists this suicidal event even when high concentrations of gramicidin S are added and outgrowth is further delayed. It is obviously this small fraction of the population which, at the end of the long outgrowth stage, develops into vegetative cells. Previous work indicates that this minor population is not genetically resistant to gramicidin S. We conclude that the long delay in germination outgrowth is brought about by two effects of gramicidin S: (1) killing; and (2) decreasing the rate of one or more of the cellular metabolic activities necessary for outgrowth.  相似文献

The content of dipicolinic acid (DPA) was assayed in the spores of Bacillus anthracoides 96 during various stages of its growth. The content of DPA was ca. 10.7 per cent of the dry biomass weight in the seven-day-old culture containing 96 to 99 per cent of the spores in a "starvation" medium. The morphology of the culture was modified, and the content of DPA in the spores fell to 3.6 per cent half an hour after the inoculation into the medium favourable for the growth (MPA). During the following one to four hours of the germination, the refraction index of the spores and the content of DPA in them decreased (the content of DPA to 2 per cent).  相似文献

Triggering of germination in Bacillus megaterium QM B1551 spores with D-glucose was studied. First, the interaction of glucose with spores for less than 1 min resulted in triggering almost 90% of the spores after the glucose was removed by dilution. Therefore only a brief time is needed for glucose to trigger germination, and then the continuous presence of glucose is not necessary. Detectable uptake of glucose began 2 to 3 min after absorbance loss started, and a non-metabolizable glucose analog, methyl-alpha-D-glucopyranoside, triggered germination in the absence of detectable uptake. Several inhibitors that reduced or eliminated glucose uptake did not block triggering of germination. Therefore, glucose uptake may be a relatively late event and not a prerequisite for triggering of germination.  相似文献

Germinating spores of Bacillus brevis are sensitive to inhibition by gramicidin S prior to emergence whereas once emergence is underway inhibition is lost and newly emerged vegetative cells are not affected by the antibiotic. Under conditions of overcrowding the concentration of antibiotic released is sufficient to render the germinated spores non-viable although dormant spores still retain their viability. Considering these points and the manner in which spore populations germinate we outline a strategy of germination for survival of Bacillus brevis .  相似文献
