A new method was developed to synthesize a cortexolone-substituted affinity matrix, based on the fast, mild and quantitative reaction between alpha-ketomesylates and thiols. The resulting cortexolone-Sepharose absorbed easily the cytosolic chick thymus glucocorticoid receptor. Owing to the relatively fast dissociation of the glucocorticoid receptor-cortexolone complex, glucocorticoid receptor could be eluted with cortexolone as well as with triamcinolone acetonide from the affinity gel with similarly good yields. We obtained 75-150-fold purification factors (yield: 20-30%) using this column procedure. The partially purified glucocorticoid receptor was obtained in non-activated form. It had a Stokes radius of 6.2 +/- 0.1 nm. It could be activated to DNA-cellulose binding form by heat or 0.3 M KCl. KCl treatment activated 30-50% of the partially purified glucocorticoid receptor. Heat activation, however, was rather poor. Cortexolone-complexed, partially purified glucocorticoid receptor dissociated easily, and partially purified free glucocorticoid receptor, capable of steroid binding, could be obtained. Binding properties of the partially purified glucocorticoid receptor were then analyzed using different steroids. Dissociation rate constants were similar to those of the cytosolic glucocorticoid complexes. Association rate constants were consistently smaller than in the case of cytosolic glucocorticoid receptor, but the relative order of rates for different steroids was basically the same for glucocorticoid receptor in the two studied systems.  相似文献

We report on the discovery of two new lead series for the development of glucocorticoid receptor agonists. Firstly, the discovery of tetrahydronaphthalenes led to metabolically stable and dissociated compounds. Their binding mode to the glucocorticoid receptor could be elucidated through an X-ray structure. Closer inspection into the reaction path and analyses of side products revealed a new amino alcohol series also addressing the glucocorticoid receptor and demonstrating strong anti-inflammatory activity in vitro.  相似文献

Glucocorticoid resistance due to mutations in the gene for the glucocorticoid receptor has been suggested to be more common than is thought at present, owing to the relative mildness of its symptoms and the difficulty of its diagnosis. To investigate the prevalence of mutations in the glucocorticoid receptor gene responsible for relative insensitivity to glucocorticoids, we carried out polymerase chain reaction/single-strand conformation analysis of the glucocorticoid receptor gene in a group of 20, otherwise healthy, persons with a reduced response in a dexamethasone suppression test and in 20 controls. We did not find mutations or polymorphisms associated with a reduced sensitivity to glucocorticoids. However, we identified five novel polymorphisms in the gene for the human glucocorticoid receptor, which may be useful in analyzing whether loss of (part of) the glucocorticoid receptor gene plays a role in glucocorticoid-resistant malignancies. Although relative resistance to glucocorticoids seems to be rather frequent in otherwise healthy persons, it is not usually associated with mutations or polymorphisms in the glucocorticoid receptor gene. Glucocorticoids are widely used in the treatment of inflammatory and other diseases. However, high-dose or chronic administration often triggers troublesome side effects such as metabolic syndrome and osteoporosis. We recently described that one glucocorticoid receptor gene produces eight translational glucocorticoid receptor isoforms that have distinct gene-regulatory abilities. We show here that specific, but not all, glucocorticoid receptor isoforms induced apoptosis in human osteosarcoma U-2 OS bone cells. Whole human genome microarray analysis revealed that the majority of the glucocorticoid target genes were selectively regulated by specific glucocorticoid receptor isoforms. Real-time PCR experiments confirmed that proapoptotic enzymes necessary for cell death, granzyme A and caspase-6, were induced by specific glucocorticoid receptor isoforms. Chromatin immunoprecipitation assays further suggested that glucocorticoid receptor isoform-dependent induction of proapoptotic genes was likely due to selective coregulator recruitment and chromatin modification. Interestingly, the capabilities to transrepress proinflammatory genes were similar among glucocorticoid receptor isoforms. Together, these findings provide new evidence that translational glucocorticoid receptor isoforms can elicit distinct glucocorticoid responses and may be useful for the development of safe glucocorticoids with reduced side effects.  相似文献

Murine lymphoma cell lines such as WEHI-7 exhibit a cytolytic response to both cAMP and glucocorticoids. We have exploited this behavior to ask if cyclic AMP-dependent protein kinase plays a role in regulating glucocorticoid receptor function. We have found that cAMP-resistant cell lines containing a defective cAMP-dependent protein kinase activity give rise to spontaneous steroid-resistant variants at a high frequency (approximately 10(-7)) relative to wild type cells (less than 10(-10)). Unlike previous results with wild type cells, nearly complete loss of glucocorticoid receptor function was observed in a single selection using unmutagenized cAMPr derivatives of WEHI-7. Thus, the initial selection of the cAMPr phenotype serves as a permissive step toward the acquisition of glucocorticoid resistance in WEHI-7. In addition, cAMP was found to increase the levels of steroid binding in these cell lines, and the dose response was dependent upon the phenotype of the cyclic AMP-dependent protein kinase. The results demonstrate an important role for cAMP in regulating glucocorticoid receptor activity and strongly suggest that this novel two-step selection scheme leads to the isolation of new forms of glucocorticoid resistance.  相似文献

Mutation of a small number of amino acids in the DNA-binding domain of the estrogen receptor to the corresponding sequence of the glucocorticoid receptor switches the specificity of the receptor in transactivation assays (Mader, S., Kumar, V., de Verneuil, H., and Chambon, P. (1989) Nature 338, 271-274). We have made the corresponding reciprocal mutations in the context of the glucocorticoid receptor DNA-binding domain and studied the binding of wild type and mutant purified proteins to palindromic glucocorticoid and estrogen response elements as well as to elements of intermediate sequence, using gel mobility shift assays. We show here that a protein with two altered amino acids binds glucocorticoid and estrogen response elements with a low but equal affinity, whereas a protein with an additional changed residue has a high affinity for estrogen response elements but still retains a considerable affinity for glucocorticoid response elements. Using binding sites of intermediate sequence we have further characterized the interaction with DNA. The in vitro DNA binding results are confirmed by in vivo transactivation assays in yeast. Finally we suggest a testable model for amino acid/base pair interactions involved in recognition by the glucocorticoid receptor DNA-binding domain of its target sequence.  相似文献

Tissue-specific regulation of glucocorticoid receptor mRNA by dexamethasone   总被引:13,自引:0,他引:13  
The effect of glucocorticoids on tissue-specific regulation of glucocorticoid receptor mRNA was studied in intact and adrenalectomized rats. Glucocorticoid receptor mRNA was examined by Northern blot hybridization and quantitated by slot blot hybridization using a glucocorticoid cRNA probe. Glucocorticoid receptor mRNA was greatest in the lung with the relative levels in other tissues as follows: spleen, 70%; brain, 55%; liver, 50%; kidney, 43%; heart, 35%; adrenal, 13%; and testis only 8%. A tissue-specific difference in glucocorticoid receptor mRNA accumulation was found after adrenalectomy. There was little change in glucocorticoid receptor mRNA levels in liver and lung, but the brain and kidney demonstrated a 40 and 80% increase in mRNA, respectively. In contrast, dexamethasone treatment resulted in a consistent decrease of 40-60% in the accumulation of glucocorticoid receptor mRNA in all tissues studied. These results provide in vivo evidence for the autoregulation of the glucocorticoid receptor by its homologous ligand and demonstrate the existence of tissue-specific regulation of the glucocorticoid receptor mRNA levels in states of glucocorticoid excess and depletion.  相似文献

Glucocorticoids deplete the AtT-20 mouse pituitary tumor cell of its glucocorticoid receptors. Once placed into steroid-free medium, however, these 'receptor-deplete' cells gradually regain their ability to bind glucocorticoids displaceably. The goal of this paper is to determine whether replete binding-species is indeed a glucocorticoid receptor, and whether any precursor or modified forms are seen during repletion. Once depleted, cells take nearly 3-4 days to replete their full complement of cytosolic glucocorticoid-binding species. Scatchard analysis revealed only one binding site, of which the affinity for dexamethasone was identical to that of the native receptor. The steroid-binding specificity of the 'replete' binding species was exactly the same as that of the original receptor. The molecular size and surface charge density of the glucocorticoid-binding species obtained throughout the process of repletion were identical to those of the original receptor. Finally, studies in which replete cells were exposed to glucocorticoid agonists showed that the cell was able to decrease its production of ACTH, indicating that the regenerated binding species was able to function as a biologically active glucocorticoid receptor. We conclude that the replete species is a glucocorticoid receptor identical to the original, and that probably no precursor or modified forms of the repleting receptor exist.  相似文献
