An improved filter paper disk bioassay for determining activities of plant growth regulators was developed and evaluated. Indole-3-acetic acid (IAA), (±)-abscisic acid (ABA), and 6-furfurylamino-purine (kinetin) were dissolved in 95% ethanol at fixed dilutions. Specific concentrations of each growth regulator were then evenly dropped onto individual 6-cm paper disks and the solvent evaporated. The activities of the above three water-insoluble plant growth regulators in both solution and the disk assay were compared using the excised cucumber (Cucumis sativus L.) cotyledon root formation bioassay (IAA), wheat (Triticum aestivum L.) coleoptile straight growth bioassay (ABA), and cucumber cotyledon expansion bioassay (kinetin), and similar results were obtained at each concentration. The possible principle of this method has been studied using the cucumber cotyledon expansion bioassay. The results suggested that the disks, as carriers, had highly dispersed kinetin molecules on them and greatly accelerated the dissolution and diffusion of kinetin from disks to water.  相似文献

The role of auxins in induction of roots byAgrobacterium rhizogenes was studied in carrot root disks. Transformed roots were produced on root disks by inoculation withA. rhizogenes, A4. Measurement of indole-3-acetic acid (IAA) by gas chromatography-mass spectrometry (GC-MS) indicated that there was a significant increase in the concentration of IAA in transformed callus and induced roots compared with initial IAA concentrations in carrot disks. Indole-3-butyric acid (IBA) was found to occur naturally in carrot roots. The presence of IBA, a potent root inducer, must be taken into account when assessing the role of auxin during transformation and induction of roots byA. rhizogenes.  相似文献

The addition of 1.5 mM putrescine to the culture medium facilitated the conversion of protocorm-like bodies (PLBs) of Dendrobium officinale to shoots. The administration of 1.0 mM of the polyamine inhibitor, α-DL-difluoromethylarginine, decreased the conversion of PLBs to shoots. Compared to the control, the PLBs treated with 1.5 mM putrescine had higher levels of total intracellular free polyamines. The enhanced conversion of PLBs to shoots following the addition of putrescine was accompanied by an increase in the ratio of total cytokinins (CTKs) to indole-3-acetic acid (IAA). Analysis of enzyme activities indicated that the increased endogenous level of total CTKs driven by putrescine was associated with the inhibition of CTK decomposition by CTK oxidase, while the decreased endogenous level of IAA was associated with the promotion of IAA decomposition by IAA oxidase. In addition, putrescine increased the ratio of reduced glutathione to oxidized glutathione in the conversion process of PLBs to shoots.  相似文献

Epicotyl dormancy was broken in cultured peony (Paeonia lactiflora Pall.) embryos after topical application of agarose gels containing gibberellic acid, with optimum growth at 1.5 mM gibberellic acid. Addition of 100 M abscisic acid to the medium resulted in complete inhibition of gibberellic acid-stimulated promotion of dormant epicotyls. Epicotyl dormancy was also broken in embryos by culture on media containing 1 or 10 M benzylaminopurine. A highly significant increase in leaf number occurred when embryos were both cultured on medium containing benzylaminopurine and treated topically with gibberellic acid. Anatomical and morphological studies indicated that the increase in shoot growth was due to the development and growth of 1) buds formed at the cotyledonary node, 2) axillary buds, and 3) adventitious meristems originating from subepidermal parenchymatous tissue.Abbreviations ABA abscisic acid - BA N6-benzylaminopurine - DMSO dimethyl sulfoxide - GA3 gibberellic acid - LS Linsmaier and Skoog  相似文献

Inoubation of thin slioes ofTaraxacum root segments in 3-indoleaoetic acid solutions enhanced the level of total proteins, RNA, DNA and a number of enzymes reaching an optimum at 0.01 mg 1-1. It has been shown that the auxin promotes the synthesis of DNA, again with an optimum promotion at 0.01 mg 1-1 IAA. The effect of UV-C (220–280 nm) on the accumulation of phytoalexin and resistance to Botrytis cinerea was studied in cold-stored carrots. Carrots were surface-wounded, treated with a range of UV doses and stored at 1 °C for 25 days in lots of 22 roots. The level of the phytoalexin, 6-methoxymellein, in each lot was then assayed in the peel of eight roots. Twelve of the remaining roots were subsequently inoculated with mycelial plugs to evaluate their level of disease resistance. The elicitation of 6-methoxymellcin by UV increased significantly the resistance of the roots to B. cinerea. The effect of UV in freshly harvested carrots was curvilinear, showing an optimum between 0.44 and 0.88 Merg/cm2. However, only a linear relationship was observed with aged (stored for 4 months at 1 °C) carrots for the same doses, suggesting a modification in the response to UV with age. Wounding was necessary for carrots kept at 1 °C to respond to UV treatment. Neither UV nor wounding alone caused any elicitation at this temperature. Since unwounded roots could respond to UV at 20 °C, it is hypothesized that the level of physiological activity of the roots determines their response to UV. An increase in the physiological activity by higher temperatures or wounding would allow the elicitation process to take place. Since UV irradiation can increase the level of disease resistance in treated tissues, this treatment has potential as an alternative method for the control of post-harvest diseases m carrots.  相似文献

The reducing agents, potassium ferrocyanide, β-mercaptoethylamine, cysteine, reduced DPN, ferrous sulfate, methyl viologen and ascorbic acid caused the expansion in darkness of disks of primary leaf tissue cut from dark-grown bean plants. The reducing agents interacted synergistically with low irradiances of red radiation and additively with high irradiance of red light. Exposure of disks treated with reducing agents to far red light repressed disk expansion and the decay in sensitivity to far red radiation showed the same time relations as sequential exposure to red and far red radiation.

The oxidizing agents, 1,4-naphthoquinone, ferric sulfate, hydrogen peroxide, t-butyl hydroperoxide, cystine, and potassium ferricyanide repressed the expansion of leaf disks initiated by exposure to red radiation. The oxidizing agents interacted synergistically with low irradiances of far red light and additively with irradiances of far red light.

The release (=the measured loss) of amino acids was studied in Commelina benghalensis leaf disks. The release is assumed to be the result of influx and efflux, therefore, both movements were investigated.The uptake of 14C-labeled valine exhibited a biphasic isotherm. The uptake was pH-dependent, especially at low substrate concentrations (pH optimum 4.8). Signals for amino acid/proton co-transport were observed: stimulation of the uptake by fusicoccin (FC), inhibition by diethylstilbestrol (DES) or by high K+ concentrations. In the light, the ATP level of the disks was maintained during the uptake period (2 h), in darkness the ATP content decreased from 87 to 24 nmol g–1 fr. wt. However, light-promoted uptake, which is explained in the proton pump concept by an intensified proton extrusion as the result of high ATP production, was lacking.The release of amino acids was increased by washing with p-chloromercuriphenyl sulphonic acid (PCMBS), nystatin, 3(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU), or KCN. The release (Q10 about 1.5) was independent of the external pH and was linearly related to the intracellular amino acid concentration. Light enhanced the rate of release to the same extent at all intracellular concentrations. The present results suggest that the release is balanced by a, at least partially, proton-driven influx and a diffusional ligh-promoted efflux. The effects of salt stress and abscisic acid (ABA) on the expression of betaine aldehyde dehydrogenase (BADH) were determined in sorghum (Sorghum bicolor L.) plants. BADH mRNA expression was induced by salinity, and the timing coincided with the observed glycinebetaine (betaine) accumulation. The leaf water potential in the leaves of the sorghum plants was significantly affected by salinity. In response to salinity, betaine, ABA, Na and Cl accumulations increased 6-, 16-, 90-, and 3-fold, respectively. In the leaf disks from unsalinized plants incubated on NaCl, or ABA solution, the BADH mRNA level was lower than in the ABA-treated disks. Exogenous application of the ABA biosynthetic inhibitor fluridone to the NaCl-treated disks reduced the ABA accumulation and BADH mRNA levels compared with NaCl-treated leaves. The results indicate that the salt-induced accumulation of betaine and BADH mRNA coincides with the presence of ABA.  相似文献

A study was made of the effects of solar ultraviolet‐B radiation (UV‐B) on the growth of the dominant plant species of a shrub‐dominated ecosystem in Tierra del Fuego. This part of southern Argentina can be under the direct influence of the Antarctic ‘ozone hole’ during the austral spring and lingering ozone‐depleted air during the summer. The plant community is dominated by an evergreen shrub (Chiliotrichum diffusum) with an herbaceous layer of Gunnera magellanica and Blechnum penna‐marina in the interspaces between the shrubs. Inspections of ozone trends indicate that the springtime and summertime ozone column over Tierra del Fuego has decreased by 10–13% from 1978/9 to 1998/9. In a set of well‐replicated field plots, solar UV‐B was reduced to approximately 15–20% of the ambient UV‐B using plastic films. Polyester films were used to attenuate UV‐B radiation and UV‐transparent films (~90% UV‐B transmission) were used as control. Treatments were imposed during the growing season beginning in 1996 and continued for three complete growing seasons. Stem elongation of the shrub C. diffusum was not affected by UV‐B attenuation in any of the three seasons studied. However, frond length of B. penna‐marina under attenuated UV‐B was significantly greater than that under near‐ambient UV‐B in all three seasons. Attenuation of solar UV‐B also promoted the expansion of G. magellanica leaves in two of the growing seasons. Differences between treatments in leaf or frond length in B. penna‐marina and G. magellanica did not exceed 12%. Another significant effect of UV‐B attenuation was a promotion of insect herbivory in G. magellanica, with a 25–75% increase in the leaf area consumed. Changes in plant phenology or relative species cover were not detected within the time frame of this study. The results suggest that the increase in UV‐B radiation associated with the erosion of the ozone layer might be affecting the functioning of this ecosystem to some degree, particularly by inhibiting the growth of some plant species and by altering plant–insect interactions.  相似文献

The effects of the auxin transport inhibitors 2,3,5-triiodobenzoic acid (TIBA), 9-hydroxyfluorene-9-carboxylic acid (HFCA), and 1-N-naphthylphthalamic acid (NPA) on gibberellins (GAs) in the garden pea (Pisum sativum L.) were studied. Application of these compounds to elongating internodes of intact wild type plants reduced markedly the endogenous level of the bioactive gibberellin A1 (GA1) below the application site. Indole-3-acetic acid (IAA) levels were also reduced, as was internode elongation. The auxin transport inhibitors did not affect the level of endogenous GA1 above the application site markedly, nor that of GA1 precursors above or below it. When plants were treated with [13C,3H]GA20, TIBA reduced dramatically the level of [13C,3H]GA1 recovered below the TIBA application site. The internodes treated with auxin transport inhibitors appeared to be still in the phase where endogenous GA1 affects elongation, as indicated by the strong response to applied GA1 by internodes of a GA1-deficient line at the same stage of expansion. On the basis of the present results it is suggested that caution be exercised when attributing the developmental effects of auxin transport inhibitors to changes in IAA level alone. Many herbivorous insects induce preferences for host plants. Recent work in Manduca sexta indicates that induced preferences are mediated by a “tuning” of the peripheral taste system to chemicals within host plant foliage. We tested this hypothesis by rearing caterpillars on artificial diet or potato foliage, and then examining olfactory- and taste-mediated responses to potato foliage extract. First, we confirmed earlier reports that consumption of potato foliage tunes the peripheral taste system by reducing responsiveness to glucose and increasing responsiveness to foliage extract. Second, we offered caterpillars a choice between disks treated with foliage extract (experimental) or solvent alone (control). The foliage-reared caterpillars approached and consumed the experimental disks disproportionately, whereas the diet-reared caterpillars approached and consumed both disks indiscriminately. This indicated that induced preferences involve olfaction and taste. Third, we ran choice tests with foliage-reared caterpillars deprived of either olfactory or gustatory input. Caterpillars lacking olfactory input approached both disks indiscriminately, but fed selectively on experimental disks. In contrast, caterpillars lacking gustatory input approached experimental disks selectively, but fed indiscriminately on both types of disk. We conclude that even though olfaction helps caterpillars locate potato foliage, it is the “tuned” gustatory response that ultimately mediates the induced preference.  相似文献

A strain improvement program was developed to increase extracellular phytase (E.C. 3.1.3.8.) production by Aspergillus niger (syn. A. ficuum) NRRL 3135. Ultraviolet (UV) radiation was used as the mutagen and resistance to 50 g/ml of hygromycin B as the selection method. Mutant 2DE, the product of two UV treatments, had phytase (PhytA) activity at pH 5.0 in the extracellular filtrate that was 3.3-fold higher than the wild-type activity. The activity of the non-specific acid phosphatase with a pH optimum of 6.0 (Pase) was one-fifth the activity of the wild type and the non-specific acid phosphatase with a pH optimum of 2.5 (PhytB) was not significantly different from the wild type. The mutant and wild-type PhytA, PhytB and Pase responsed similarly in inhibition studies. However, the wild-type enzymes were inhibited more by 1 mm sodium fluoride and 1 mm phosphomycin. PhytA production by the mutant was repressed 60% by inorganic phosphate concentrations of 0.006% (wt/vol) or above. The mutant had an extracellular protein concentration 3.2-fold higher than the wild type, which correlated with its higher phytase activity at pH 5.0, but not with phytase activity at pH 2.5 and acid phosphatase activities. The isolate may be a phytase catalytic mutant, as well as, on overproducer of phytase. UV‐induced synthesis/accumulation of photoprotective pigments and antioxidant activity were investigated in the hot‐spring cyanobacterium Leptolyngbya cf. fragilis. The results indicated that UV radiation may induce biosynthesis of carotenoids, allophycocyanin, phycoerythrin, and scytonemin while phycocyanin degrades in response to longtime UV radiation. Moreover, pigment composition of L. cf. fragilis was significantly altered with increasing UV radiation times, probably due to destruction and resynthesis of accessory pigments as an adaptation strategy to UV stress. The in vitro antioxidant analysis of different extracts of UV treated cyanobacteria exhibited concentration‐dependent antioxidant activity. Ethyl acetate extract of 72 h UV treatment showed maximum total antioxidant activity (IC50 = 71.73 ± 5.3 μg mL?1) followed by ethyl acetate control (non‐UV irradiated) extract (IC50 = 109.43 ± 2.76 μg mL?1). This is the first report for the UV‐induced synthesis of photoprotective pigments and their antioxidant activity in L. cf. fragilis.  相似文献

The aim of this study was to investigate the protective effect of ferulic acid at different doses (50 mg kg?1 alternative day and 50 mg kg?1 daily) on the streptozotocin (STZ)‐induced post‐diabetes rat testicular damage. Diabetes was induced by a single intraperitoneal injection of STZ (50 mg/kg). Rats treated with ferulic acid were given once a day orally for 10 weeks, starting 3 days after STZ injection. Testis tissue and blood samples were collected for investigating biochemical analysis, antioxidant status, sperm parameters, and histopathological, immunohistochemical and apoptotic studies. Treatment with ferulic acid to diabetic rats significantly improved the body weight, testis weight, serum insulin level, serum testosterone level and sperm parameters (viability, motility and count). Histopathological study also revealed that ferulic acid‐treated diabetic rats showed an improved histological appearance. Our data indicated that significant reduction in the activity of apoptosis by using terminal deoxyuridine triphosphate nick end‐labelling and reduced expression of transforming growth factor‐β1 and interleukin‐1β in the testis tissue of ferulic acid‐treated diabetic rats. Conversely, it was also revealed that ferulic acid‐treated diabetic rats markedly enhanced the serine/threonine protein kinase protein expression in the testis tissue. Our result suggests that ferulic acid inhibits testicular damage in diabetic rats by declining oxidative stress. Copyright © 2013 John Wiley & Sons, Ltd.  相似文献

The response of pericarp disks from ripening tomato (Lycopersicon esculentum Mill. cv. Traveler‘76) to CaCl2, additions was studied to determine the effect of Ca2+ on ethylene and CO2 production. Application of 5 mM CaCl2 resulted in a 2, 20, 33, 39, and 50% increase in ethylene production in disks obtained from preclimacteric minimum, climacteric rise, climacteric peak, one, and two days postclimacteric fruit, respectively. CaCl2 concentrations of 10 and 50 mM gave no additional stimulation of ethylene production; CO2 production at 5 mM CaCl2 was not different from controls, but is increased at 10 and 50mM CaCl2. CaCl2 also increased ethylene production in disks treated with 1-aminocyclopropane-1-carboxylic acid (ACC) or aminoethoxy-vinylglycine. Chloride salts of K+, Na+, Mg2+, Sr2+ and La3+ did not stimulate ethylene production. SrCl2 stimulated ethylene production to a lesser degree than CaCl2. Disks from potato (Solanum tuberosum L. cv. Katahdin) tubers produced greater quantities of ethylene and ACC when 5 mM CaCl2 was included in the incubation medium (K. B. Evensen, 1983. Physiol. Plant. 60:125–128). Ca2+-treated disks had more than three times as much ACC synthase activity as control disks after 18 to 24 h incubation, when ethylene and ACC were maximal. The apparent Km for S-adenosylmethionine was 13 μM at 29°C, pH 8.0 in extracts from both Ca2+-treated and control disks. Inclusion of 1 to 50 mM CaCl2 in the assay medium did not significantly affect enzyme activity. ACC synthase extracted from control and Ca2+-treated disks had a pH optimum of 8.5 and an apparent molecular weight of 72 kdalton, estimated by gel filtration. It is likely that the presence of Ca2+ in the buffer allows greater synthesis of ACC synthase as part of the wound-healing response in potato, while in tomato the predominant effect is on membrane stabilization.  相似文献

A heavy metal responsive gene PvSR3 (GenBank accession number U54703) encoding an acid dehydrin was isolated from a mercuric chloride-treated bean (Phaseolus vulgaris L.) leaf cDNA library by differential screening using cDNAs derived from treated and untreated plants. The PvSR3 cDNA is 981-bp long and has a 606-bp open-reading frame with a 202-residue-deduced amino acid sequence. The PvSR3 sequence contains two conserved repeats of the characteristic lysine-rich K segment (EKKGIMDKIKEKLPG) preceded by an 8-serine residue stretch, whereas the Y segment (DEYGNP) conserved motif is absent. The deduced protein has a calculated molecular weight of 23 kDa and an isoelectric point of 5.2. Sequence similarity and comparative analysis showed that PvSR3 shares 70 and 73% similarity with the dehydrin of poplar and pepper, respectively. Southern hybridizations indicated that PvSR3 was a low copy-number gene. Northern blot analysis revealed that PvSR3 mRNA was weakly detected in seedling leaves. However, the gene expression was strongly stimulated by heavy metals, such as mercury, cadmium, arsenic, and coppper, whereas virus infection and salt had little effect on it. In contrast, PvSR3 was not responsive to drought or abscisic acid (ABA), and was downregulated by UV radiation. Furthermore, PvSR3 was upregulated by the exogenous signaling molecules, including salicylic acid (SA) and hydrogen peroxide (H2O2). It is suggested that PvSR3 is extremely related to heavy metal stress, and might play an important role in metal detoxification and resistance to the damage caused by heavy metals.  相似文献

The effects of kinetin (Kn), gibberellic acid (G), and light(L) on cell expansion and division in disks from 6-day-old etiolatedprimary leaves of dwarf bean are described. Cell number wasdetermined by direct counting after disks were digested in apectinase/EDTA mixture to separate the cells. Kn increased leafexpansion wholly by increasing cell size. G also increased cellsize; it increased cell division in the dark but not in thelight. Light also increased cell size and cell division; iteliminated the effect of G on cell division but enhanced theeffect of G on cell expansion.  相似文献

The effects of enhanced UV‐B radiation on the oogenesis and morpho‐anatomical characteristics of the European solitary red mason bee Osmia bicornis L. (Hymenoptera: Megachilidae) were tested under laboratory conditions. Cocooned females in the pupal stage were exposed directly to different doses (0, 9.24, 12.32, and 24.64 kJ/m2/d) of artificial UV‐B. Our experiments revealed that enhanced UV‐B radiation can reduce body mass and fat body content, cause deformities and increase mortality. Following UV exposure at all 3 different doses, the body mass of bees was all significantly reduced compared to the control, with the highest UV dose causing the largest reduction. Similarly, following UV‐B radiation, in treated groups the fat body index decreased and the fat body index was the lowest in the group receiving the highest dose of UV radiation. Mortality and morphological deformities, between untreated and exposed females varied considerably and increased with the dose of UV‐B radiation. Morphological deformities were mainly manifested in the wings and mouthparts, and occurred more frequently with an increased dose of UV. Cell death was quantified by the Terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay (DNA fragmentation) during early stages of oogenesis of O. bicornis females. The bees, after UV‐B exposure exhibited more germarium cells with fragmented DNA. The TUNEL test indicated that in germarium, low doses of UV‐B poorly induced the cell death during early development. However, exposure to moderate UV‐B dose increased programmed cell death. In females treated with the highest dose of UV‐B the vast majority of germarium cells were TUNEL‐positive.  相似文献
