The purpose of this study was to examine changes in fatty acyl chain composition of major cardiac phospholipids in relation to down-regulation of -adrenoceptors during various forms of stress or chronic adrenergic stimulation. Analysis of the fatty acid profile of phosphatidylcholine (PC) and phosphatidylethanolamine (PE) in sarcolemma or cardiac muscle membranes showed partial replacement of 18:2n-6 by 20:4n-6 in PC and replacement of both 18:2n-6 and 20:4n-6 by 22:6n-3 in PE during daily administration of epinephrine or norepinephrine for 7 or 15 days, respectively These changes in membrane PC and PE coincided with down-regulation or the decrease in Bmax of -adrenoceptors during adrenergic stimulation. Cardiac membrane response to other forms of stress or chronic adrenergic stimulation such as neonatal stress, restriction stress or restricted food intake was expressed in the same way, that is replacement of 18:2n-6 by 20:4n-6 in PC and replacement of 18:2n-6 and 20:4n-6 by 22:6n-3 in PE.Conclusion: Adaptation to stress includes a decrease in the density of binding sites or down-regulation of -adrenoceptors in sarcolemma synchronized with specific alterations in the fatty acyl chain composition within the membrane bilayer. The changes in the lipid milieu of the membrane may facilitate conformational changes in the transmembrane segment of the receptor forming the ligand binding sites of the -adrenoceptor.  相似文献

Plasma membrane lipid rafts have been considered as a sort of "chamber", where several subcellular activities, including CD95/Fas-mediated pro-apoptotic signaling, can take place. Recently, we demonstrated that, after CD95/Fas triggering, raft-like microdomains could be detected in mitochondrial membranes. The mitochondrion appears as a dynamic and subcompartmentalized organelle in which microdomains might act as controllers of apoptosis-associated fission that results in the release of apoptogenic factors. Here, we hypothesize that some "small" mitochondria, possibly derived from their fission process, can reach the nuclear envelope and strictly interact with this. Mitochondria could act as a signaling "device" contributing to molecular trafficking of molecules, including raft-like components, during apoptosis.  相似文献

This paper reports on a multidisciplinary meeting held to discuss ethical issues in medical research in the developing world. Many studies, including clinical trials, are conducted in developing countries with a high burden of disease. Conditions under which this research is conducted vary because of differences in culture, public health, political, legal and social contexts specific to these countries. Research practices, including standards of care for participants, may vary as a result. It is therefore not surprising that ethical issues emerge. This meeting sought to identify and discuss these issues from the perspectives of the many actors in such research, including community representatives, with a view to finding ethical and pragmatic solutions to these issues. Dialogue between these actors was also promoted, with a view to identifying the need to develop such dialogue in future.
Drawing from the experiences of the speakers, the colloquium attempted to outline some answers to several key questions characterising the field today. Experiences related to epidemiologic research, vaccine trials, drug trials, diagnostic tests and to some fundamental ethical issues in health research. Speakers were from different countries, disciplines and professions. The meeting provided a forum for consultation and debate between different ethics actors. Both encouraging findings and challenges emerged.  相似文献

The cabbage butterfly, Pieris melete is multivoltine with a pupal summer and winter diapause. Summer and winter diapause are induced principally by relatively long and short daylengths, respectively. The intermediate to relatively short daylengths of autumn permitted some pupae to develop without diapause in the field. A short daylength had a stronger diapause inducing effect than a relatively long one under higher temperatures. The principal sensitive phase for photoperiodic response occurred before the late 3rd larval instar. The critical daylength for wild autumnal populations was between 12h 30min and 12h 40min at an average temperature of 20.5 degrees C. A night interruption by 2h of light averted diapause most effectively when it was placed 10 to 12h after lights-off. High temperatures and long days during summer inhibited the incidence of diapause, suggesting that the occurrence of summer diapause is due to the specific climatic conditions occurring in April and early May, rather than to the high temperatures in summer. This indicates that the butterfly has a cryptic ability to reproduce in summer. High temperatures delayed diapause development, whereas low temperatures enhanced it, indicating that the optimum temperature of diapause development is lower. The diapause regulating mechanisms thus ensure that the species synchronises its development and reproduction with the growth seasons of the host plants and provide the species with a high degree of flexibility in its life cycle.  相似文献

We studied the life-history of the Sierra Madre sparrow (Xenospiza baileyi) in a subalpine grassland-agriculture mosaic south of Mexico City. From March to early September 1999 we captured and color-banded 53 adults with mist-nets and mapped the breeding territories of 21 males. We again mapped territories in the same spot in April and May 2000. The number of breeding territories was found to be the same in the two consecutive years and interannual survivorship was found to be relatively high. Breeding territories were restricted to the bunchgrass-covered areas. We used the density of territories and the amount of remaining habitat to estimate a total population size of 5,380–6,150 adults for this species. Using this and other recent data, we recommend raising the Sierra Madre sparrow to the status of critically endangered using BirdLife International criteria.  相似文献

The zooplankton community structure, including copepods, euphausiids, chaetognaths, and decapod larvae, was monitored during six circadian cycles using Bongo net (500 microns mesh net) samples from Bahía Magdalena, on the southwest coast of Baja California, México. Samples were obtained during three oceanographic surveys (March, July, and December 1996) to describe the changes in the zooplankton community structure throughout the main mouth of Bahía Magdalena. The zooplankton community structure showed strong changes with a close relation to environmental conditions. During March, a well-mixed water column with low temperature and salinity indicated an influence of the California Current water and local upwelling processes. During July, temperature increased and a wide salinity range was recorded. The stratification of the water column was intense during summer, enhancing the thermocline. The highest temperatures and salinity were recorded in December, related to the presence of the Costa Rica Coastal Current (CRCC). The thermocline deepened as water temperature increased. A typical temperate community structure with low specific richness dominated by Calanus pacificus, Nyctiphanes simplex, and Acartia clausi and high zooplankton biomass (average 9.3 and 5.5 ml 1000 m-3 respectively) during March and July shifted to a more complex tropical community structure with a low zooplankton biomass in December (average 0.37 ml 1000 m-3). The mouth of Bahía Magdalena has a vigorous exchange of water caused by tidal currents. The zooplankton community structure was not significantly different between the central part of Bahía Magdalena and the continental shelf outside the bay for all months. The results suggest a more dynamic inside-outside interaction of zooplankton assemblages than first thought.  相似文献

The conditions of eutrophication are described in three lagoon systems differing by their structure, their catchment area and their connection with the sea: the Bassin d'Arcachon on the Atlantic coast, SW France, the semi-artificial fish ponds of the Bassin d'Arcachon, and the Étang du Prévost on the Mediterranean. The Bassin d'Arcachon is a shallow semi-enclosed bay, strongly influenced by climatic factors and tidal currents. The Bassin receives significant inflow of freshwater and the waters are only partially renewed. The greatest part of the primary production is due to the seagrass Zostera noltii. Although the ecosystem remains on the whole in steady state, some evidence of potential eutrophication are visible. For instance, the flux of nitrogen into the Bassin d'Arcachon has increased by more than 50% during the last 25 years. The most significant change among primary producers is the massive development since 1988 of the green alga Monostroma obscurum. The fish reservoirs of the Bassin d'Arcachon are man-made enclosures designed for extensive aquaculture and where the water renewal is only possible during certain periods of time. Thus, because of the shallowness and the confined nature of these fish ponds, acute eutrophication is sometimes observed in summer. The Étang du Prevost is extremely eutrophic due to agricultural and urban run-off. Red waters occur periodically during the warm summer months as a consequence of ecological events beginning in the early spring with a bloom of green macroalgae (Ulva sp.). In summer, the algal biomass is degraded by aerobic heterotrophic bacteria; the oxygen demand encompasses the oxygen production, leading to the predominence of anaerobic processes and dystrophic crisis. From the comparison of the selected sites, three stages of eutrophication are recognized according to the conceptual model of Nienhuis (1992) describing the relation between the relative dominance of primary producers connected to the availability of nutrients. Such macroscopic observations should, now, be explained by the study of microbiological processes including meiofauna, protozoa, bacteria and all the components of the microbial loop.  相似文献

In order to verify whether harmful algal blooms increased since 1979, a long-term project for the study of harmful algal blooms in Mazatlán Bay began 25 years ago (1979-2003). Observations were done daily and results indicate five trend types: (I) years without discoloration related to El Ni?o events; (II) years with days of sudden occurrence of discoloration, related to the release of accumulated energy after El Ni?o events; (III) years with similar number of discoloration events (days); (IV) years with a progressive decrease of the occurrence (days) of discoloration events; (V) years with gradual increase of discoloration events. The annual mean average of discoloration is of 25.3 days. Ninety-six discoloration events were recorded during year 2000, and represent the highest number of events in the last 25 years. Discoloration events are more common in March (174 days), harmful and toxic blooms included. This indicates continuity in the occurrence of the highest content of microalgae, and therefore, it is concluded that they play an important role for a better understanding of the environmental deterioration undergone by the coastal zone due to an increase of harmful and toxic microalgae blooms.  相似文献

The aim of this work is to study heat waves (HWs) in Mexicali, Mexico, because numerous deaths have been reported in this city, caused by heatstroke. This research acquires relevancy because several studies have projected that the health impacts of HWs could increase under various climate change scenarios, especially in countries with low adaptive capacity, as is our case. This paper has three objectives: first, to analyze the observed change in the summer (1 June to 15 September) daily maximum temperature during the period from 1951 to 2006; secondly, to characterize the annual and monthly evolution of frequency, duration and intensity of HWs; and finally, to generate scenarios of heat days (HDs) by means of a statistical downscaling model, in combination with a global climate model (HadCM3), for the 2020s, 2050s, and 2080s. The results show summer maximum temperatures featured warming and cooling periods from 1951 until the mid-1980s and, later, a rising tendency, which prevailed until 2006. The duration and intensity of HWs have increased for all summer months, which is an indicator of the severity of the problem; in fact, there are 2.3 times more HWs now than in the decade of the 1970s. The most appropriate distribution for modeling the occurrence of HDs was the Weibull, with the maximum temperature as co-variable. For the 2020s, 2050s, and 2080s, HDs under a medium-high emissions scenario (A2) could increase relative to 1961–1990, by 2.1, 3.6, and 5.1 times, respectively, whereas under a medium-low emissions scenario (B2), HDs could increase by 2.4, 3.4, and 4.0, for the same projections of time.  相似文献

Summary During fictive swimming in the isolated spinal cord of the lamprey (Ichthyomyzon unicuspis andPetromyzon marinus) the membrane potentials of motoneurons (MNs), lateral interneurons (L INs), and CC interneurons (CC INs) oscillate between a depolarised and a relatively hyperpolarised phase. After intracellular Cl injections (usually combined with a DC hyperpolarisation) IPSP's became depolarising, and in cells which were phasically inhibited, phases of relative hyperpolarisation became phases of relative depolarisation. The peak depolarisation and/or spike burst mid point in MNs after Cl injection occurred at a phase of 0.65 ± 0.12 (mean ±S.D.) in the cycle, with zero being the start of the ipsilateral ventral root burst. In CC INs the peak depolarisation and/or spike burst mid point after Cl occurred significantly earlier, at a phase of 0.40 ± 0.18. L INs were also phasically inhibited with peak depolarisation and/or spike burst mid point after Cl injection at an intermediate phase of 0.52 ± 0.21. Abstract

The etiologies of idiopathic sudden sensorineural hearing loss (SSNHL) and Ménière's disease remain unclear. Recently, accumulating evidence has demonstrated that free radicals are related to the pathology of inner ear disease. Because genetic factors may contribute partly to the etiologies of SSNHL and Ménière's disease, we investigated the association between genetic polymorphisms located in genes related to the free-radical process and susceptibility to SSNHL and Ménière's disease. We compared 83 patients affected by SSNHL and 83 patients affected by Ménière's disease with 2048 adults (for SSNHL) and 1946 adults (for Ménière's disease) who participated in the National Institute for Longevity Sciences, Longitudinal Study of Aging. Multiple logistic regression was used to calculate odds ratios (ORs) for SSNHL and Ménière's disease in individuals with polymorphisms in the genes: methionine synthase (MTR; rs1805087); methionine-synthase reductase (MTRR; rs1801394); nitric oxide synthase 3 (NOS3; rs1799983); caveolin 1 (Cav1; rs3840634); melatonin receptor 1B (MTNR1B; rs1387153); NAD(P)H oxidase p22(phox) subunit (NADH/NADPHp22phox; rs4673); and mitochondria 5178 (MT5178; rs28357984). The NOS3 polymorphism was significantly associated with a risk of SSNHL; in addition, the OR for the NOS3 polymorphism and SSNHL risk was 2.108 (CI, 1.343–3.309) with adjustment for age and sex. The Cav1 polymorphism was significantly associated with a risk of Ménière's disease; moreover, the OR for the Cav1 polymorphism and Ménière's disease risk was 1.849 (CI, 1.033–3.310) with adjustment for age and sex. In conclusion, the NOS3 and Cav1 polymorphisms were significantly associated with the risk of SSNHL and Ménière's disease, respectively.  相似文献

Since its invasion of North America in 2000, the soybean aphid, Aphis glycines Matsumura (Homoptera: Aphididae) has notably changed the arthropod community of the soybean agroecosystem. The objectives of this study were to characterize the foliar predatory fauna associated with A. glycines in commercial soybean fields in Québec, Canada, and to evaluate the suitability of A. glycines as prey for three coccinellid species: Harmonia axyridis Pallas, Propylea quatuordecimpunctata L. and Coleomegilla maculata lengi Timberlake (Coleoptera: Coccinellidae). Field surveys showed that several predators responded rapidly to expanding and increasing populations of the soybean aphid. Coccinellidae were the most important aphidophagous predators observed in 2002 (58.6%) and 2003 (44.8%), with mainly four native and naturalized species co-occurring with the soybean aphid throughout the growing season. Measurement of fitness parameters under laboratory conditions (survival, development time, longevity, fecundity) indicated that A. glycines is an excellent prey for the development and reproduction of all three of the coccinellid species studied. The intrinsic rate of natural increase (rm) was highest for H. axyridis (0.238 d−1), intermediate for P. quatuordecimpunctata (0.215 d−1) and lowest for C. maculata (0.134 d−1).  相似文献

Climate refugia, or places where habitats are expected to remain relatively buffered from regional climate extremes, provide an important focus for science and conservation planning. Within high-priority, multi-jurisdictional landscapes like the Madrean sky islands of the United States and México, efforts to identify and manage climate refugia are hindered by the lack of high-quality and consistent transboundary datasets. To fill these data gaps, we assembled a bi-national field dataset (n?=?1416) for five pine species (Pinus spp.) and used generalized boosted regression to model pine habitats in relation to topographic variability as a basis for identifying potential microrefugia at local scales in the context of current species’ distribution patterns. We developed additional models to quantify climatic refugial attributes using coarse scale bioclimatic variables and finer scale seasonal remote sensing indices. Terrain metrics including ruggedness, slope position, and aspect defined microrefugia for pines within elevation ranges preferred by each species. Response to bioclimatic variables indicated that small shifts in climate were important to some species (e.g., P. chihuahuana, P. strobiformis), but others exhibited a broader tolerance (e.g., P. arizonica). Response to seasonal climate was particularly important in modeling microrefugia for species with open canopy structure and where regular fires occur (e.g., P. engelmannii and P. chihuahuana). Hotspots of microrefugia differed among species and were either limited to northern islands or occurred across central or southern latitudes. Mapping and validation of refugia and their ecological functions are necessary steps in developing regional conservation strategies that cross jurisdictional boundaries. Mutations at codons 526 and 531 in the rpoB gene and at 315 in the katG gene are considered diagnostic markers for resistance to rifampin and isoniazid in tuberculosis. The aim of this study was to design and evaluate three TaqMan probes for the identification of these mutations in 138 respiratory samples positive for acid-fast bacilli, and 32 clinical isolates from a region with considerable levels of drug resistance. The specificities of the probes for the diagnosis of resistance to both drugs were 100%; however, the sensitivities were calculated to be 50% for isoniazid and 56% for rifampin. DNA sequencing of rpoB and katG; and the spoligotyping assay of the clinical isolates, confirmed the diversity of the mutations and the presence of 11 spoligotypes with a shared international type and eight unique spoligotypes. Analysis of the respiratory samples identified 22 (16%) as drug-resistant and 4 (3%) as multidrug-resistant tuberculosis. The diagnostic value of the TaqMan probes was compromised by the diversity of mutations found in the clinical isolates. This highlights the need for better understanding of the molecular mechanisms responsible for drug resistance prior to the use of molecular probes, especially in regions with significant levels of drug-resistant tuberculosis.  相似文献
