A multivariate morphometric study of theCardamine pratensis group is presented, based on 84 population samples collected from the Carpathian and Pannonian area in the Czech Republic, Slovakia, Poland, Ukraine, Hungary, and Romania. Among the multivariate methods, principal component analysis, cluster analysis, and classificatory and canonical discriminant analysis were used. The analysis of chromosome numbers from all populations studied showed wide variation. The morphometric study showed that not all groups of populations characterised by their chromosome numbers and geographical criteria are morphologically, and thus taxonomically, distinguishable. Besides the morphologically well characterised speciesCardamine dentata andC. rivularis, the following species were recognised in the area studied:C. matthioli, C. majovskii andC. pratensis. Within the last species, besides the typical populations, two diploid types are provisionally recognised: type ucranica and type rivularis auct..  相似文献

The Rubus species R. parviflorus, R. spectabilis and R. strigosus interfere with conifer seedling establishment on forest regeneration sites in Canada and the United States. As a first step towards microbial metabolite-based control, callus and cell suspension cultures of the Rubus species were developed as a bioassay system to detect phytotoxic compounds that may have relevance in a vegetation control context. Rapidly growing friable callus and suspension cultures were obtained from leaf disks of the three weedy Rubus species using similar culture media conditions (modified Murashige and Skoog) but required different plant growth regulators (R. parviflorus, 4.5 M 2,4-D; R. spectabilis, 26.9 M NAA/0.5 M zeatin; and R. strigosus, 12.4 M picloram). Cell growth and health attributes including callus circumference, degree of browning and suspension culture cell viability as measured by the TTC vital stain assay were developed and were rapid and convenient to use. We have established Rubus tissue culture systems that will make it possible for large scale screening of phytotoxic metabolites.  相似文献

Summary In this paper, the results of the preceding electrophysiological study of sodium-alanine cotransport in pancreatic acinar cells are compared with kinetic models. Two different types of transport mechanisms are considered. In the simultaneous mechanism the cotransporterC forms a ternary complexNCS with Na+ and the substrateS; coupled transport of Na+ andS involves a conformational transition between statesNCS andNCS with inward- and outward-facing binding sites. In the consecutive (or ping-pong) mechanism, formation of a ternary complex is not required; coupled transport occurs by an alternating sequence of association-dissociation steps and conformational transitions. It is shown that the experimentally observed alanine- and sodium-concentration dependence of transport rates is consistent with the predictions of the simultaneous model, but incompatible with the consecutive mechanism. Assuming that the association-dissociation reactions are not rate-limiting, a number of kinetic parameters of the simultaneous model can be estimated from the experimental results. The equilibrium dissociation constants of Na+ and alanine at the extracellular side are determined to beK N <-64mm andK S <-18mm. Furthermore, the ratioK N /K N S of the dissociation constants of Na+ from the binary (NC) and the ternary complex (NCS) at the extracellular side is estimated to be <-6. This indicates that the binding sequence of Na+ andS to the transporter is not ordered. The current-voltage behavior of the transporter is analyzed in terms of charge translocations associated with the single-reaction steps. The observed voltage-dependence of the half-saturation concentration of sodium is consistent with the assumption that a Na+ ion that migrates from the extracellular medium to the binding site has to traverse part of the transmembrane voltage.  相似文献

Summary A new cellulase gene was cloned and expressed inEscherichia coli from a thermophilic anaerobe, strain NA10. A 7.4 kbEcoRI fragment of NA10 DNA encoded the cellulase which hydrolyzed carboxymethyl cellulose, lichenan, andp-nitrophenyl--d-cellobioside, but could not digest laminarin andp-nitrophenyl--d-glucoside. The cloned enzyme could digest cellooligosaccharides and release cellobiose as a main product from cellotetraose but could not digest cellobiose. It was distinct from the endoglucanase which was cloned by us previously from NA10 strain in terms ofp-nitrophenyl--d-cellobioside degradation activity and the location of restriction enzyme sites. The enzyme produced byE. coli transformant was extremely heat-stable and the optimum temperature for the enzymatic reaction was 80°C. Fifty three percent of the cloned enzyme was detected in the periplasm and the remaining activity existed in the cellular fraction in theE. coli transformant.  相似文献

Feeding behavior and habitat use of the king angelfish, Holacanthus passer, was studied in the southern Sea of Cortés, México. H. passer fed on benthic communities (algae and sessile invertebrates) and in the water column (mainly feces from the damselfish Chromis atrilobata). Although there were not significant differences in feeding rate between sexes, coprophagy was more common in males, while grazing was more common in females. Spatial distribution of size classes followed a pattern of decreasing size with increasing depth. Feeding rate was significantly different among habitats: small females had a higher feeding rate on the bottom, big females and small males had similar feeding rates from the bottom to 3m above the bottom, and big males had higher feeding rates from 5m above the bottom to the surface. Habitat was clearly partitioned, and there was significant habitat overlap only between big females and small males. The abundance of H. passer was partly explained (34% of the total variance) by the abundance of the damselfish C. atrilobata. There was a clear trophic association between C. atrilobata schools and H. passer feeding damselfish feces in the water column. The sex ratio male:female of H. passer populations was >1 at several sites, an unusual pattern for a protogynous fish. The sex ratio on the H. passer water column stock was also biased towards males at most sites. Although there is a positive relationship between C. atrilobata abundance and H. passer, there are factors other than damselfish abundance which cause this dominance of males.  相似文献

The class II major histocompatibility complex antigens are cell-surface heterodimers consisting of an a and a chain. Cosmid cloning has shown that the three families of clas II antigens, DR, DQ, and DP, are encoded within the HLA-D region of chromosome 6 as a series of discrete gene clusters. The DP cluster contains two pairs of a and genes, one of which encodes the biochemically-defined DP antigen. In order to assess whether the other two genes, SXa and SX, are also expressed, potential coding regions have been subcloned and sequenced. The SX3 gene is shown to contain region closely homologous to all six exons of DP. A 1 bp deletion in the 2 exon, also observed for the SX4 allele, causes a translation frameshift, suggesting that SX is a pseudogene. However, all the other exons, as well as their splice sites and the putative promoter region, appear to be intact.  相似文献

The properties of the -2-l-fucosyltransferases in submaxillary gland preparations from blood group ABH secrefors and non-secretors were compared. The level of activity in the non-secretor gland homogenates amounted to about 5% only of that found in the secretor gland preparations. The enzymes from the two sources differed in solubility properties, charge and affinities for donor and acceptor substrates. The enzyme from secretor glands showed a preference for acceptors with Type 1 [d-galactosyl(1–3)-N-acetyl-d-glucosamine] structures whereas the enzyme from non-secretor glands had a preference for Type 2 [d-galactosyl(1–4)-N-acetyl-d-glucosamine] structures.These results demonstrate that expression of the secretor gene (Se) is associated with a molecular form of the -2-l-fucosyltransferase that is different from the species present in the same tissue when theSe gene is not expressed.  相似文献

We have studied the effect of such milk proteins as caseins, lactalbumin, and lactoglobulin, on proliferation and immunoglobulin production of human-human hybridoma HB4C5 cells. It was found that -, -, and -caseins stimulated both proliferation and IgM product ion of human-human hybridoma HB4C5 cells, while the activities of -lactalbumin and -lactoglobulin were negligible. To localize the active sites of these caseins, effect of protease treatments on the activities were examined. When caseins were digested with trypsin, casein digests stimulated proliferation of the hybridoma, but not their IgM production. When -casein was digested with chymosin and fractionated top--casein and glycomacropeptide, both fragments stimulated proliferation of the cells, but onlyp--casein fragment stimulated IgM production. These results indicate that -casein has at least two proliferation stimulating sites and an IgM production stimulating site in thep--casein region.  相似文献

In the present report, we used serological, cellular, and restriction fragment length polymorphism (RFLP) to investigate the DR1 haplotype in the Israeli population. We describe an Israeli homozygous typing cell (HTC), HLA-DwLVA, which defines a new lymphocyte-activating determinant associated with Bw65, DR1 and distinct from Dwl. The parents of this donor, non-Ashkenazi Algerian Jews, are first cousins and share HLA-Cw8, Bw65, BfS, DR1, DQw1, DPw4. No specificity could be assigned to HLA-DwLVA using the 91 Ninth Workshop HTCs. Two families and forty unrelated DR1 individuals were studied with DwLVA and a panel of DR1/Dw1 HTCs. HLA-DwLVA showed segregation as a single determinant within families. This new specificity was present in 24 out of 40 (60%) unrelated DR1 individuals, indicating that in the Israeli population DwLVA is the main lymphocyte-defined determinant associated with the serologically defined DRI specificity, in contrast to non-Jewish Caucasoids where DR1 is significantly associated with Dw1. The vast majority of DwLVA-positive carriers were also Bw65 carriers, indicating that Bw65, DR1, DwLVA may represent a typical allele combination in the Israeli population. The RFLP analysis established the correlation of certain RFLPs with Dw1 and DwLVA. In addition, we describe a cluster of RFLPs that may correspond to a new Dw subtype associated with DR1, for which no serological and cellular reagents have been described so far.  相似文献

Summary Two antisera, Y-10 and Y-18 were raised in rabbits against synthetic human -endorphin conjugated to bovine serum albumin and keyhole limpet haemocyanin respectively. Antiserum Y-10 has been shown by radioimmunoassay to be highly specific for human -endorphin with minimal or no cross-reactivity against other pituitary peptides whilst antiserum Y-18 crossreacted on an equimolar basis against -endorphin and -lipotropin. When used in the immunohistochemical procedure, both antisera specifically stained the corticotrophs in human anterior pituitary tissue. A similar effect was observed when antiserum Y-18 was applied to rat anterior pituitary tissue in the immunohistochemical procedure. Y-10 antiserum, on the other hand, stained not only rat corticotrophs but also somatotrophs. The somatotrophin staining could not be attributed to the enkephalins reported to be present in these cells.The non-specific -endorphin antiserum Y-18 was used to stain anterior pituitaries from dehydrated and adrenalectomized rats as well as rats of the Brattleboro strain. In tissues from the three experimental animals, cells that stained positively for -endorphin did not give a positive immunoreaction for ACTH and vice versa in some other sections. It is concluded that under the physiological conditions, formalin fixation of the tissue causes the proopiocortin molecule to be trapped in a conformation such that either ACTH or -endorphin-like determinants are available for reacting with the appropriate antiserum.This work was financed by the Medical Research Council of New Zealand, NIH Research Program Project Grant HD-12303 and by U.S.P.H.S. Summary Plant water relations and shoot growth rate of shrubs resprouting after fire or unburnt were measured in a semi-arid poplar box (Eucalyptus populnea) shrub woodland of eastern Australia. In vegetation unburnt for about 60 years, the dawn xylem water potential (x) of the dominant shrub species was about-1.0 MPa when the soil was wet and-8.0 MPa when the soil was very dry. At any one time, the dominant shrub species,Eremophila mitchellii, E. sturtii, Geijera parviflora andCassia nemophila, were similar in x butAcacia aneura andDodonaea viscosa were consistently higher in x than this group when the soil was moist and lower when the soil was dry. The dominant tree species,Eucalyptus populnea andE. intertexta, appeared to have access to additional water beneath the hardpan which is located 60–80 cm below the surface. When shrubs were under extreme water stress (x of-8 MPa), the trees had a x of-3 to-3.6 MPa. Following a fire, both x and leaf stomatal conductance (g s) of resprouting shrubs were higher for about 5 years than comparable-aged unburnt vegetation, with relative differences in x increasing with drought stress. Elongation rate of resprouts was positively linked to prefire shrub height in 3 of 4 species. However, shrubs resprouting after high intensity fires had substantially higher rates of shoot elongation than after low intensity fires which were in turn higher than for foliar expansion of unburnt shrubs. It is concluded that the growth rate of resprouting shrubs is primarily determined by physiological/ morphological factors associated with plant size but is also assisted by greater availability of water and possibly nutrients for a period after fire.  相似文献

This study documents that ethnomedical beliefs and practices play an important role in primary care in a southern community. Thirty-three of 73 patients from a rural Appalachian area coming to a university primary care internal medicine practice presented 54 ethnomedical complaints such as high blood (24.1%), Weak 'n dizzy (22.2%), nerves (16.7%), sugar (5.6%) and fallin' out (3.7%). Thirty-three patients had both biomedical and ethnomedical complaints, 40 patients had biomedical complaints without ethnomedical complaints and no patients presented with ethnomedical complaints alone. Over two-thirds of all patients consulted non-medical personnel for their complaints, mostly family and friends, and 70 percent self-treated prior to clinic consultation. Patients presenting with ethnomedical complaints when compared with those presenting with biomedical complaints sought advice of non-physicians significantly more often (p < 0.02); no statistical difference, however, was found in their self-treatment practices. Ninety-two of 130 biomedical complaints were recorded by the patient's physician but none of the 54 ethnomedical complaints were formally recorded (p < 0.001). The high incidence of ethnomedical complaints in this population and the failure of physicians to recognize these complaints demand that primary care medicine residents be taught improved history-taking skills and the essentials of ethnomedical illnesses if they are to provide culturally-sensitive patient care. Summary In situ hybridization has been performed in sections through ovaries ofAcipenser ruthenus andAcipenser güldenstädti in order to detect the rDNA sequences. Hybridization resulted in specific labelling of the caps of extrachromosomal DNA present in pachytene oocyte nuclei and of the chromatin granules distributed beneath the nuclear envelope in early diplotene nuclei. In the same sections, the nuclei of all ovarian cells in both species (oogonia, leptotene, and zygotene stage oocytes, follicular cells, connective tissue cells) showed a very low, but similar labelling.Amplification of genes for rRNA thus occurs at the pachytene stage in early oogenesis ofAcipenseridae. Seed dispersal by the Clark's nutcracker (Nucifraga columbiana Wilson) may markedly influence the growth form and genetic population structure of limber pine (Pinus flexilis James). The nutcracker buries clusters of seeds in subterranean caches; germination of clustered seeds often results in a growth form characterized by two or more genetically distinct trees with fused or contiguous trunks (tree clusters). The occurrence of a morphologically similar form, the multi-trunk tree (a single genet branched near the base), as well as the typical single-trunked tree, complicates the study of limber pine populations. We examined growth form distribution and genetic relationships in tree clusters in limber pine populations at four elevations (from 2585 m to 3460 m) in the Colorado Front Range. At three study areas, relative occurrence of limber pine growth forms, as well as that of associated pines, was examined by a point-centered quarter survey. From the four study areas, we collected foliage from each trunk from a total of 74 clumps (combined tree clusters and multi-trunk trees) in order to differentiate the two growth forms using starch gel protein electrophoresis. Tree clumps were significantly more common in limber pine than in ponderosa or lodgepole pine (P<0.010). Although single-trunk limber pine was the most common growth form, except at the highest elevation, both multi-trunk trees and tree clusters were present in each stand. Tree clusters were estimated to comprise about 20% of the tree sites in each limber pine stand; the estimated proportion of multi-trunk trees varied by site from 5% to 77%. Trees in clusters were related, on average, as half to full siblings (mean r=0.43), but were unrelated to trees in other clusters (mean r=0.01). Electrophoretic analysis suggests possible genetic differentiation in limber pine that may be the result of different selection pressures on the growth forms.  相似文献

Summary The plasmid pHCP2 carries the lamB gene of E. coli. This plasmid was transferred to 4 strains of Erwinia using transformation or conjugal mobilisation. All 4 plasmid-containing strains became sensitive to infection. After infection with ::Tn5 and the higher-hopper Tn10 derivatives, a variety of auxotrophic and enzyme-deficient mutants were isolated. Summary Nonparenchymal liver cells from untreated C3HeB/FeJ mice, when incubated in medium containing-10% fetal bovine serum or portal serum, produced significant amounts of interferon alpha/beta (IFN/). In contrast, other cell populations (spleen, mononuclear blood cells and peritoneal cells) from C3HeB/FeJ mice or nonparenchymal liver cells from other strains of mice (C3H/HeJ, germ-free C3H/HeN and C57Bl/6J) produced little or no detectable IFN in fetal bovine serum under the same culture conditions. The cells in the nonparenchymal liver cell population responsible for IFN/ production were adherent, phagocytic, silica-sensitive, carbonyl-iron-sensitive, and Thy1.2, presumably Kupffer cells or resident liver macrophages. IFN/ production by cultured Kupffer cells was not observed if medium containing fetal bovine serum or portal serum was treated with polymyxin B or if Kupffer cells were cultured in serum-free medium. This suggested that small amounts of endotoxin in fetal bovine or portal serum stimulated Kupffer cells to produce IFN/. Possibly, Kupffer cells are in a different state of activation/maturation than peritoneal and splenic macrophages since the sensitivity of resident Kupffer cells from C3HeB/FeJ mice to the stimulatory effects of endotoxin. Summary The statocysts inProtodrilus ciliatus, P. oculifer, P. haurakiensis andP. helgolandicus are situated in the prostomium anterior to the palps and have been investigated by electron microscopy. The sensory organs were reconstructed from serial sections, volumes were calculated from areas of consecutive section profiles, and additional data on surface area of distal receptor elements have been determined. In spite of variations in size (diameter 8–20 m) their structure is nearly identical. The organs consist of one cup-shaped supportive cell, one large bi- or multiciliated sensory cell and two small uni- or biciliated sensory cells forming an extracellular cavity. This cavity is completely filled with microvillus-like or paracrystalline structures and there are no signs of statoliths composed of extracellular material. The most striking feature is the occurrence of paracrystals made up of undulating ciliary membranes extending from the large sensory cell and occupying 75–90% of the cavity inP. ciliatus, P. oculifer andP. haurakiensis. The remaining space is filled with microvilli or dendritic processes of the sensory cells. InP. helgolandicus the ciliary paracrystals are almost completely replaced by microvillus-like branches of cilia of the corresponding sensory cell. Paracrystals fill less than 10% of the cavity and are formed of flattened membranes. These sensory organs enclose large surface areas of membranes (15,000–38,000 m2). The surface areas of the paracrystals composed of undulating membranes is almost identical to that of densely arranged arrays of microvilli (about 25 m2 per m3). These sensory organs are so different from all known statocysts that it is likely that they have another function. Their greatest structural correspondence is to light-receptive organs, especially in the structure and arrangement of microvilli. The role the paracrystals play is discussed: they might bear photopigments or simply represent a lens — a transparent, refractile and crystalline structure. These sensory organs are completely different from pigmented ocelli and phaosomes occurring in some protodrilids and represent a type of sensory organ thus far undescribed in polychaetes.  相似文献

Zusammenfassung In der vorliegenden Arbeit werden Beobachtungen ausgewertet, die in den letzten Jahren in einer Seggenrohrsängerpopulation der Mark Brandenburg gesammelt wurden. Brutbiotop ist eine Großseggenwiese. und haben getrennte Reviere, wobei sich -Reviere, -Reviere und auch - und -Reviere zum Teil sehr erheblich überlagern. Keinem konnte ein bestimmtes zugeordnet werden, sie leben also in Keinehe. versuchen, aus der Nestnähe zu vertreiben. Aggressive Handlungen zwischen benachbarten konnten nicht festgestellt werden. Ihre Singflüge unterscheiden sich von denen der Schilfrohrsänger. 28 Eier maßen im Durchschnitt 17,76×13,14 mm. Nur die brüten und Füttern. Die durchschnittliche Fütterungsfrequenz liegt bei 19,7 Fütterungen pro Stunde, die tägliche Aktivitätszeit der ist im Juli etwa 1 Stunde kürzer als im Juni. 13–14 Tage nach dem Schlüpfen verlassen die Jungen das Nest. In der Nestlingsnahrung ist bei Julibruten der hohe Heuschreckenanteil auffällig. Nachdem die Jungen selbständig sind, streunen sie im Brutbiotop umher. Zweitbruten sind sehr wahrscheinlich, konnten aber noch nicht nachgewiesen werden.
Summary Observations made during the last years on a population of the Aquatic Warbler (Acrocephalus paludicola) in Brandenburg, Germany, gave the following results:In a sedge meadow, withCarex elata the dominant species, males and females ofAcrocephalus paludicola held separate territories, those of males as well of females sometimes considerably overlapping territories held by other individuals of the same or the opposite sex. In no case a certain male could be considered as having formed a firm pair bond with a certain female: on the contrary, the relations between the sexes were very loose, and probably the birds are even promiscuous. Females try to chase away males from the neighbourhood of their nests. Aggressiveness between neighbouring males has not been observed. The display flights of the males are different from those of the Sedge Warbler(A. schoenobaenus). 28 eggs measure on the average 17,76×13,14 mm. Females alone incubate and feed the young, at an average rate of 19,7 times per hour. The young leave the nest 13–14 days after hatching. Young reared in July are fed a great amount of grasshoppers. Second broods probably occur, but could not be proved.

Mortality factors were determined in over 20000 pupal skins of Brassolis sophorae and Opsiphanes invirae, collected from 55 sites in the urban area of Campinas, São Paulo, southeast Brazil. Almost 50% of B. sophorae pupae and 30% of O. invirae pupae were killed by various identifiable factors, which may have an appreciable influence in determining adult population size. Mortality was related to type of attachment site, distance from the center of the city, area occupied by buildings and houses, vegetation cover, and other urban variables. Exposed pupation sites such as outside walls had a lower frequency of successful eclosions than darker or more sheltered sites, such as offset skirting boards, garages, or mailboxes, which act as partial refuges against parasitoids. Mortality levels were higher in regions of the city with high vegetation cover. Vegetation cover and distance from the city center were the best predictors of parasitism level of B. sophorae by the chalcidid wasp Spilochalcis morleyi. The viability of S. morleyi (data from parasitized B. sophorae) did not differ significantly among different regions of the city. These data do not support an exclusive model of escape from parasites (or other natural enemies) for explaining the abundance of urban butterflies.This paper was presented at the VI INTECOL, Manchester, UK, in September 1994  相似文献
