Addition of the l-proline analogue l-azetidine 2-carboxylic acid to growing cultures of Saccharomyces cerevisiae var. ellipsoideus promoted fast deactivation of the general aminoacid permease, measured as l-valine uptake, without an immediate decrease in the growth rate. Cells preincubated with the analogue for 3 h were unable to restore either growth ability or general aminoacid permease activity in analogue-free medium. Eadie-Hofstee plots of l-valine uptake in the presence of the analogue are consistent with a strong reduction in the number of active molecules of the general amino-acid permease located in the plasma membrane. Three new water soluble titanocene–aminoacid complexes have been synthesized via the reaction of Cp2TiCl2 and two equivalents of aminoacid (L) in methanol, affording [Cp2TiL2]Cl2, L=L-cysteine (2), D-penicillamine (3) and L-methionine (4). These complexes have been characterized by 1H, IR and UV-Vis spectroscopies, elemental analysis and cyclic voltammetry. Kinetic studies of ligand hydrolysis have been monitored at low pH using UV-Vis and 1H NMR spectroscopies to assess their stability in aqueous solution. At low pH, aminoacid ligands are lost one order of magnitude faster than cyclopentadienyl. However, at physiological pH, in Tris buffer solution, the complexes decompose rapidly to form an insoluble titanium compound. The affinity of these complexes to apo-transferrin was also investigated to elucidate how the ancillary aminoacid ligands affect the titanium intake by apo-transferrin.Electronic Supplementary Material Supplementary material is available for this article at  相似文献

Three isolates ofA. tenuis isolated from the diseased leaves ofMangifera indica l. Musa paradisiaca l. andPsidium guajava l. were investigated. They were grown on different sources of nitrogen viz., potassium nitrate, sodium nitrate, calcium nitrate, ammonium nitrate, sodium nitrite, ammonium sulphate, ammonium chloride, glycine, DL-valine, L-glutamic acid, urea, thiourea, L-asparagine and peptone. They were also grown on the medium lacking nitrogen. A wide variation was observed in the growth and reproduction of the different isolates. The growth of all of them was good on potassium nitrate, calcium nitrate, glycine, DL-valine, L-glutamic acid, L-asparagine and peptone but the sporulation was satisfactory on calcium nitrate only. Sodium nitrite supported moderate growth of banana leaf isolate whereas there was no growth of the other two isolates. None of the organisms could grow on the medium lacking nitrogen as well as on thiourea. The results obtained with the isolates under study have been compared with those of earlier investigators and it has been clearly established that the different isolates ofA. tenuis could show marked differences in their nitrogen requirements.  相似文献

The interaction of free IAA and its amino acid conjugates on growth and development of cultured tomato hypocotyl tissue (Lycopersicon esculentum Mill. cv. Marglobe) was studied. In a nutrient medium containing 10 mol/L of benzyladenine, free IAA stimulated shoot and root development with little callus proliferation. In contrast, all IAA-amino acid conjugates tested supported mostly callus growth. Simultaneous application of free IAA and its conjugates resulted in the expression of mixed morphogenetic responses (i.e., both vigorous callus growth and organogenesis resulted). Free D- and L-alanine contents were comparable in the body wall and introvert cum retractor muscles of Phascolosoma arcuatum. In contrast, the content of free D-alanine in the internal organs was twice that of free L-alanine. Since alanine aminotrans-ferase from P. arcuatum was L-alanine specific, D-alanine appeared to be synthesized from L-alanine through the action of alanine racemase. Alanine racemase activity was higher in the D-alanine-forming direction in the three body parts of P. arcuatum. In addition, the ratio of DL/LD racemase activity in the internal organs was the lowest among the body parts studied. These results indicate that free D-alanine might be of lesser importance than the free D-isomer to the internal organs as compared to the body wall and introvert cum retractor muscles. Indeed, L-alanine inhibited pyruvate kinase from the body wall and introvert cum retractor muscles but had no effect on the pyruvate kinase from the internal organs. Furthermore, the activity of alanopine dehydrogenase present in the internal organs was significantly lower than those of the body wall and introvert cum retractor muscles. L-Alanine was an essential substrate for alanopine formation in the body wall and introvert cum retractor muscles during hypoxia since alanopine dehydrogenases from these body parts were L-alanine specific. When P. arcuatum was confronted with hypo-osmotic stress, the free D-alanine/total free alanine ratio in the internal organs increased approximately from 0.6 to 0.8 as the total free alanine content decreased. In comparison, those ratios in the body wall and introvert cum retractor muscles remained relatively constant. The effect of a number of compounds structurally related to glutamic acid and other nitrogenous compounds on the composition of three forms of glutamine synthetase (GS) inRhizobium phaseoli has been examined in detail. Amino acids like glutamic acid, glutamine, and a fixed source of nitrogen like ammonium chloride did not alter the relative glutamine synthetase composition.l-Methioninedl-sulfoximine (MSX), a glutamate analogue, significantly repressed the synthesis of GSIII to a greater extent.,N-oxalyl,-diaminopropionic acid (ODAP), another glutamate analogue, selectively stimulated the synthesis of GSII, and the effect of ODAP on GSII synthesis was greatly enhanced in the presence of ethylenediamine or ammonium chloride. Ethylenediamine itself caused a predominant synthesis of GSIII.-Cyanoalanine-grownR. phaseoli did not synthesize GSI. The synthesis of the three different glutamine synthetases can thus be differentially modulated.  相似文献

Despite recent advances in identifying the causative agents of disease in corals and understanding the impact of epizootics on reef communities, little is known regarding the interactions among diseases, corals, and their dinoflagellate endosymbionts (Symbiodinium spp.). Since the genotypes of both corals and their resident Symbiodinium contribute to colony-level phenotypes, such as thermotolerance, symbiont genotypes might also contribute to the resistance or susceptibility of coral colonies to disease. To explore this, Symbiodinium were identified using the internal transcribed spacer-2 region of ribosomal DNA from diseased and healthy tissues within individual coral colonies infected with black band disease (BB), dark spot syndrome (DSS), white plague disease (WP), or yellow blotch disease (YB) in the Florida Keys (USA) and the US Virgin Islands. Most of the diseased colonies sampled contained B1, B5a, or C1 (depending on host species), while apparently healthy colonies of the same coral species frequently hosted these types and/or additional symbiont diversity. No potentially “parasitic” Symbiodinium types, uniquely associated with diseased coral tissue, were detected. Within most individual colonies, the same dominant Symbiodinium type was detected in diseased and visually healthy tissues. These data indicate that specific Symbiodinium types are not correlated with the infected tissues of diseased colonies and that DSS and WP onset do not trigger symbiont shuffling within infected tissues. However, few diseased colonies contained clade D symbionts suggesting a negative correlation between hosting Symbiodinium clade D and disease incidence in scleractinian corals. Understanding the influence of Symbiodinium diversity on colony phenotypes may play a critical role in predicting disease resistance and susceptibility in scleractinian corals.  相似文献

This process is known as racemization and is most likely to occur in long-lived low-turnover tissues such as normal cartilage. During an investigation of the disease profile of Withania somnifera, it was observed that leaf spot is the most prevalent disease. Repeated isolations from infected leaf tissues and pathogenicity tests showed the association of fungal pathogen identified as Alternaria alternata (Fr.) Keissler. Scanning electron microscopy showed various histological changes in the leaf tissues of infected plants. A decrease in total content of reducing sugars (20%) and chlorophyll (26.5%) was observed in diseased leaves whereas an increase was noticed in proline (25%), free amino acids (3%) and proteins (74.3%). High performance thin layer chromatography (HPTLC) analysis of secondary metabolites viz. withanolides, withaferin-A and total alkaloids of the diseased leaves vis-à-vis control revealed reduction in withaferin-A and withanolides contents by 15.4% and 76.3% respectively, in contrast to an increase in total alkaloids by 49.3%, information hitherto unreported in W. somnifera.  相似文献

The location of enzyme activity in gangrene‐diseased tubers was determined using the nitrocellulose blotting method. The activity of aminopeptidase and esterase was located in tissues adjacent to dry rot caused by Phoma exigua var. foveata and in other apparently healthy tissues. The activity of glucuronidase, succinic and glucose‐6‐phosphate dehydrogenases (G‐6‐PDH), however, was confined to tissues adjacent to the rotted tissue. The pathogen produces very active α‐ and β‐glycosidases, so their highest activity occurred in rotten tissue that was filled with fungal mycelium. Results suggest that all these enzymes are involved in alteration of cell metabolism and the destruction of diseased tuber tissue.  相似文献

Abstract

In southern Italy, Spartium junceum (Spanish broom) is severely affected by a phytoplasmal disease, Spartium witches’‐broom (SpaWB). The volatile fractions extracted from flowers of healthy and diseased plants, examined by gas chromatography and gas chromatography–mass spectrometry, appeared to be quantitatively and qualitatively different. In both the healthy and the diseased plants, the main components were n‐alkanes, which occurred at a rate of 55.2% and 38.8%, respectively. The level of aliphatic acids was considerably lower in flowers of the diseased plants than in those of the healthy plants (4.5% vs. 18.7%). Sesquiterpenes were detected only in the diseased plants. It is possible that the changes in the composition of secondary metabolites of diseased plants can be related to plant defense responses.

Caribbean corals, including sea fans (Gorgonia spp.), are being affected by severe and apparently new diseases. In the case of sea fans, the pathogen is reported to be the fungus Aspergillus sydowii, and the disease is named aspergillosis. In order to understand coral diseases and pathogens, knowledge of the microbes associated with healthy corals is also necessary. In this study the fungal community of healthy Gorgonia ventalina colonies was contrasted with that of diseased colonies. In addition, the fungal community of healthy and diseased tissue within colonies with aspergillosis was contrasted. Fungi were isolated from healthy and diseased fans from 15 reefs around Puerto Rico, and identified by sequencing the nuclear ribosomal ITS region and by morphology. Thirty fungal species belonging to 15 genera were isolated from 203 G. ventalina colonies. Penicillum and Aspergillus were the most common genera isolated from both healthy and diseased fans. However, the fungal community of healthy fans was distinct and more diverse than that of diseased ones. Within diseased fans, fungal communities from diseased tissues were distinct and more diverse than from healthy tissue. The reduction of fungi in diseased colonies may occur prior to infection due to environmental changes affecting the host, or after infection due to increase in dominance of the pathogen, or because of host responses to infection. Data also indicate that the fungal community of an entire sea fan colony is affected even when only a small portion of the colony suffers from aspergillosis. An unexpected result was that A. sydowii was found in healthy sea fans but never in diseased ones. This result suggests that A. sydowii is not the pathogen causing aspergillosis in the studied colonies, and suggests several fungi common to healthy and diseased colonies as opportunistic pathogens. Given that it is not clear that Aspergillus is the sole pathogen, calling this disease aspergillosis is an oversimplification at best. Communicated by Biology Editor Dr Michael Lesser  相似文献

Interactions between the mycorrhizal fungus Glomus monosporum and the root rot pathogen Thielaviopsis basicola and their effects on tobacco plants were investigated over a 4 week period. Mycorrhizal tobacco plants, obtained by preinoculation with G. monosporum, showed a better tolerance to T. basicola than non-mycorrhizal seedlings. Root and leaf dry weights of mycorrhizal plants were greater than those of controls. Mycorrhizal plants inoculated with T. basicola showed higher root and leaf dry weights than non-mycorrhizal infected plants, but lower values than mycorrhizal plants which were not infected. No appreciable differences in free aminoacid composition were observed among the different treatments with two exceptions: proline content was higher in infected and mycorrhizal infected plants compared to control and mycorrhizal plants; arginine content was higher in infected and mycorrhizal infected plants compared to control and mycorrhizal plants; arginine content was higher in mycorrhizal plants than in all the other treatments. The mechanisms by which (VAM) fungi can reduce disease incidence and pathogen development are discussed.  相似文献

The chemoautotrophic symbiont-bearing clam Lucinoma aequizonata contains very high levels of free d-alanine in all tissues. The possible sources for this amino acid and its involvement in the clams' metabolism were investigated. Very low levels of d-alanine (generally below 1 mol·l-1) were measured in the sediment porewaters from the habitat of the clams. Experiments with 14C-labeled tracers demonstrate an active metabolism of d-alanine in the clams rather than a role as inert waste product. d-alanine is metabolized at about 0.12 mol·g fw-1·h-1. Label from aspartate, but not glucose and CO2, is incorporated into d-alanine. Incubation with labeled d-alanine did not result in formation of radioactive l-alanine. Tests for alanine racemase (EC 5.1.1.1) and d-amino acid oxidase (EC 1.4.3.3.) did not show activity in either gill, i.e. symbiont and host, or foot tissue. d-Alanine amino transferase (EC 2.6.1.b.) was demonstrated in gill and foot tissues. The control of malate metabolism and stimulation of 1-sinapolyglucose: L-malate sinapoyltransferase (SMT) activity in radish (Raphanus sativus L. var. sativus) cotyledons has been studied. The light-induced and nitrate-dependent activity of SMT catalyzes the formation of O-sinapoly-L-malate via 1-O-sinapoyl--D-glucose. When dark-grown radish seedlings, cultivated in quartz sand with nutrient solution containing NO 3 - as the sole N source, were treated with light, SMT activity increased concomitantly with free malate in the cotyledons. This light effect was suppressed in seedlings grown in a culture medium which contained in addition to NO 3 - also NH 4 + . However, treatment with methionine sulfoximine neutralized this ammonium effect, resulting again in both rapid accumulation of malate and rapid increase in SMT activity. When seedlings grown on NO 3 - nitrogen were subsequently supplied with NH 4 + nitrogen, the accumulated level of L-malate rapidly dropped and the SMT increase ceased. The enzyme activity decreased later on, reaching the low activity level of plants which were grown permanently on NO 3 - /NH 4 + -nitrogen. An external supply (vacuum infiltration) of malate to excised cotyledons and intact seedings, grown on NO 3 - /NH 4 + -nitrogen medium, specifically promoted a dose-dependent increase in the activity of SMT. Summary l-proline uptake via the intestinal brush-borderIMINO carrier was tested for inhibition by 41 compounds which included sugars, N-methylated, -,-, - and -amino and imino acids, and heterocyclic analogs of pyrrolidine, piperidine and pyridine. Based on competitive inhibitor constants (apparentK/'s) we find that theIMINO carrier binding site interacts with molecules which possess a well-defined set of structural prerequisites. The ideal inhibitor must 1) be a heterocyclic nitrogen ring, 2) have a hydrophobic region, 3) be thel-stereoisomer of 4) an electronegative carbonyl group which is 5) separated by a one-carbon atom spacer from 6) an electropositive tetrahedral imino nitrogen with two H atoms. Finally, 7) the inhibitor conformation determined by dynamic ring puckering must position all these features within a critical domain. Summary The soil fungi from an agricultural field in Allahabad where sugarcane is being grown for many years, have been isolated from various depths during different seasons and were identified. The inter-relations of chemical composition of soil and distribution of fungi is also being shown here.The techniques for the isolation and the study of the fungus flora was that ofGoddard modified bySaksena &Mehrotra. Soil samples were examined from 1–6 depths in three seasons of the year and were mechanically and chemically analysed.For the isolation, soil dilution plate method, a modification ofMenzies' method, direct method ofWaksman, Rossi Cholodny Burried slide technique, and screened immersion plate method, were followed Fifty five different species of microfungi belonging to Phycomycetes, Ascomycetes and Fungi — Imperfecti were isolated and identified. The moisture contents, hydrogen-ion concentrations, carbon, nitrogen, phosphorus, calcium, magnesium and iron, etc., of the soil samples from 1–6 inches depths, were also studied.Out of these 12 species covering 9 genera belonged to the Phycomycetes, nine genera of Ascomycetes and nine genera of Fungi Imperfecti were recorded.P. multicolor Grigorieva-Monoilova &Poradielova, andP. roqueforti Thom.,Gliocladium vermoesoni (Biourge)Thom. andMasoniella grisea (Smith)Smith were recorded for the first time from Indian Soil. The tissue distributions of sinapic acid esters (1-sinapoylglucose, sinapolyl-l-malate, 6,3-disinapoylsucrose), kaempferol glycosides, free malic acid and of the enzyme involved in the synthesis of sinapoyl-l-malate, 1-sinapoylglucose: l-malate sinapoyltransferase (SMT), have been investigated in cotyledons of Raphanus sativus L. seedlings. The kaempferol glycosides were mainly localized in the upper epidermis. The sinapoyl esters were found in all tissues, but differed markedly in their concentrations. While disinapoylsucrose was localized predominantly in the mesophyll, most sinapoylmalate was found in the epidermal layers, as was most SMT activity. Ultraviolet microscopy and microfluorospectrophotometry of isolated epidermal peels indicated that the epidermal sinapoyl esters were restricted to guard cells, guard mother cells and adjacent epidermal cells. Upon excitation by UV light (365 nm) these exhibited strong blue fluorescence with an emission maximum at about 480 nm. Our results indicate a highly tissue-and cell-specific secondary metabolism in Raphanus cotyledons and indicate that the biosynthesis of sinapoylmalate is intimately related to the malic-acid metabolism of the guard cells.Abbreviations HPLC high-performance liquid chromatography - SMT 1-sinapoylglucose: l-malate sinapoyltransferase  相似文献

A 2 × 2 factorial arrangement of treatments in randomized design was conducted to investigate the effect of different selenomethionine (SM) sources and levels on the productive performance of breeder hens and the Se distribution in the inclusion of eggs and serum and tissues of breeder hens and its offspring. A total of 480 Ling-Nan-Huang breeder hens, 48 weeks of age, were allocated to four treatments, each of which included three replicates of 40 hens. Pretreatment period was 2 weeks, and the experiment lasted 8 weeks. Two SM forms of dl-SM and l-SM were supplemented at 0.15 or 0.30 mg Se/kg into the basal diet. Results showed that the Se level of 0.15 mg/kg supplemented in the diet, compared to 0.30 mg/kg, significantly elevated the percentage of egg production (p < 0.05), hatchability (p < 0.01), and birthrate (p < 0.01), whereas the Se level of 0.30 mg/kg led to a higher Se content in egg contents, serum, and all tissues (p < 0.01). In addition, the form of dl-SM showed a significant increase in Se content of egg inclusion (p < 0.01), serum (p < 0.01), and all tissues (p < 0.01, except breeder hens’ pancreas and its offspring’s liver and breast muscle). The birthrate and yolk Se content were markedly influenced by the interaction between Se source and Se level (p < 0.01). The above results suggested that dl-SM, compared to l-SM, had a similar equal effect on the performance of breeder hens, but dl-SM was superior to l-SM with respect to selenium distribution in egg inclusion, serum, and tissues.  相似文献
