Biology cannot accommodate all aspects of culture. Aspects of culture that a biological approach can take into account can be covered by the biological categories of phenotype and environment. There is no need to treat culture as a separate category. Attempts to elaborate biological explanations of cultural variation will meet with success only if biologists expand theories of development, and integrate them in evolutionary biology. The alternative — elaborating the idea of so-called cultural inheritance — makes little sense from a biological point of view.  相似文献

The properties of the -2-l-fucosyltransferases in submaxillary gland preparations from blood group ABH secrefors and non-secretors were compared. The level of activity in the non-secretor gland homogenates amounted to about 5% only of that found in the secretor gland preparations. The enzymes from the two sources differed in solubility properties, charge and affinities for donor and acceptor substrates. The enzyme from secretor glands showed a preference for acceptors with Type 1 [d-galactosyl(1–3)-N-acetyl-d-glucosamine] structures whereas the enzyme from non-secretor glands had a preference for Type 2 [d-galactosyl(1–4)-N-acetyl-d-glucosamine] structures.These results demonstrate that expression of the secretor gene (Se) is associated with a molecular form of the -2-l-fucosyltransferase that is different from the species present in the same tissue when theSe gene is not expressed.  相似文献

A glasshouse experiment was conducted to study the effect of Ni on the growth and nutrients concentration in wheat (Triticum aestivum Cv. WH 291) in the presence and absence of applied N as urea. Responses to N application were observed up to 120 g N g–1 soil. No response to Ni was observed in the dry matter yield of wheat tops (leaves + stem) in the absence of applied N while in the presence of applied N, significant yield increases were obtained at 12.5g Ni g–1 soil. Nickel was not toxic to wheat up to 50g Ni g–1 soil in the presence of 120g N g–1 soil. Nitrogen and Ni concentration in wheat tops and roots increased with increasing levels of applied N and Ni, respectively. Applied Ni had an antagonistic effect on N concentration. Similarly, N reduced the Ni concentration in the wheat tissues. Positive growth responses to Ni were associated with 22 and 15g Ni g–1 in wheat tops, in the presence of applied N at 60 and 120g N g–1 soil, while Ni toxicity was associated with 63, 92.5 and 112.5g Ni g–1 in wheat tops, in the absence and presence of applied N at 60 and 120g N g–1 soil, respectively.  相似文献

Summary The growth of two strains of the petitenegative yeast Kluyveromyces lactis is inhibited by Tetracycline in different ways under the same culture conditions. Tetracycline resistant mutants of one strain have been isolated which can tolerate doses as high as 3000 g/ml of antibiotic.The segregation pattern of this character obtained by random spore analysis of the ascospores derived from the cross of the two strains strongly suggests that the resistance to tetracycline is under mitochondrial control.  相似文献

Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. Within the temporal pattern of primary differentiation the capacity of chlorophyll — a biosynthesis in the cotyledons ofSinapis alba L. seedlings is controlled by phytochrome (in continuous light) or by releasing the circadian rhythm either with lightdark cycles or by a lightdark transition. The sensor pigment for this process is phytochrome. Unravelling the factors that contribute to the formation and the stability of -sheet structure in peptides is a subject of great current interest. A -hairpin, the smallest -sheet motif, consists of two antiparallel hydrogen-bonded -strands linked by a loop region. We have performed a statistical analysis on protein -hairpins showing that the most abundant types of -hairpins, 2:2, 3:5 and 4:4, have characteristic patterns of 13C and 13C conformational shifts, as expected on the basis of their and angles. This fact strongly supports the potential value of 13C and 13C conformational shifts as a means to identify -hairpin motifs in peptides. Their usefulness was confirmed by analysing the patterns of 13C and 13C conformational shifts in 13 short peptides, 10–15 residues long, that adopt -hairpin structures in aqueous solution. Furthermore, we have investigated their potential as a method to quantify -hairpin populations in peptides.  相似文献

Summary The humoral leukocyte adherence inhibition (H-LAI) assay has recently been found to measure an antitumor immunefactor. In this assay, trypsinized leukocytes from control persons are used as indicator cells and 0,25% serum from the patient is added to the assay system together with the relevant tumor antigen.In the present work, evidence is presented that the H-LAI response is mediated through in vitro-formed immune complexes. Different antibody-antigen pairs (anti-albumin — albumin; anti-2microglobulin — 2microglobulin; anti-carcinoembryonic antigen — carcinoembryonic antigen; anti-transferrin — transferrin) have been added to the assay mixture. A significant H-LAI response was observed when immune complexes were formed. On the other hand, when unrelated antibody — antigen pairs were added, no response was found. The specificity was demonstrated in experiments where two different antibodies were added simultaneously and the response tested both against the two corresponding antigens and against unrelated antigens.Since the same trypsinized indicator cells can be used for different immune complexes, it is likely that the response is mediated through common receptors on the cell surface with affinity for immune complexes, i.e., Fc-receptors. Presumably, the H-LAI test gives response to immune complexes in general and is as such not specific. The specificity is achieved through the addition of specific antigen and the subsequent in vitro formation of immune complexes.  相似文献

Summary Several agents known to interact with the (Na++K+)-pump were tested for their effects on the components of steady-state K+ flux in ascites cells.86Rb+ was used as a tracer for K+, and influx was differentiated into a ouabain-inhibitable pump component, a Cl-dependent and furosemide-sensitive exchange component, and a residual leak flux. All agents tested (ouabain, quercetin, oligomycin, phosphate) affected both the pump flux and the Cl-linked flux. These findings suggest a linkage between the activity of the Na/K ATPase and the Cl-dependent K+ exchange flux. In the discussion we point out that the mechanism of this linkage could be direct; e.g., Cl-dependent exchange may represent a mode of operation of the Na/K ATPase. However, data from this and other systems tend to suggest an indirect linkage between the Na+ pump and a KCl symporter, perhaps via a change in the level of intracellular ATP.  相似文献

Summary Chromosome 5 was examined by fluoresence microscopy in 13 individuals whose karyotype was 5p-. Great variation was observed in length (30–80% of the total short arm length), in type (translocation, interstitial and terminal deletions), and in localization of the deleted segment. There was no evidence of correlation between length, type or localization of deletion and triosephosphate isomerase activity. Triosephosphate isomerase activity of the 13 cases of karyotype 5p- was 2,74 moles/min/mg hemoglobin (95% confidence limits 2.49–3.32). Identical values were obtained in 16 normal individuals; 2.44 moles/min/mg hemoglobin (95% confidence limits 2.16–2.72). The present investigation does not confirm the postulated localization of the gene for triosephosphate isomerase on the short arm of human chromosome 5.
Zusammenfassung 13 Patienten mit einem 5p—-Karyotyp wurden durch Fluorescenzuntersuchungen identifiziert und das Ausmaß der Deletion bestimmt. Triosephosphatisomeraseaktivitäten dieser 13 Fälle waren 2,74 mol/min/mg Hämoglobin, 95%-Vertrauensgrenzen 2,49–3,32.Identische Werte wurden bei 16 Normalpersonen gefunden (2,44 mol/min/mg Hämoglobin, 95%-Vertrauensgrenzen 2,16–2,72).Diese Befunde lassen sich dahingehend interpretieren, daß keine Assoziation zwischen Triosephosphatisomerase und Chromosom 5 besteht.

This paper defines a four-tiered, hierarchical system for classifying subsistence production types in Nepal and uses the system to describe the subsistence communities of central Nepal. The system first divides Nepal into a grid of homogenous parts or cells, second, it divides, the cells into ridge-valley slope sequences, third, defines production types for the ridge-valley slope sequences, and, finally, distinguishes local variants of the production types. After describing these tiers, I apply these categories to the northern portions of the central and western development regions where I have done research. I then define how representatives of Nepal as a whole are the production types defined for the central and western development regions. Finally, the paper locates some completed studies within its framework.  相似文献

Summary Osmium tetroxide and OTAN histochemical reactions have been reappraised following recent publications in this journal by Ellbder and Lojda. Provided that the standard OTAN reaction is used on frozen sections of conventional thickness, unsaturated hydrophilic polar lipids are stained in an orange or red shade while unsaturated hydrophobic non-polar lipids are stained brown-black or black. We have confirmed the anomaly — pointed out by Elleder and Lojda — that the polar lipid reaction of the atherosclerotic plaque is essentially extinguised by acetone. Solubility and blockading methods provide circumstantial evidence that Elleder and Lojda's assumed OsO4-protein reactions are due to the lipid moiety of a lipoprotein complex.  相似文献

Cytogenetical studies of the F1 hybrid between the commercially cultivated tuber crop, cassava (Manihot esculenta Crantz.) and the closely related wild speciesManihot glaziovii Muell. (Ceara rubber) used as donor specles for Cassava mosaic discase and drought-resistant genes and back crosses (to cassava parent) were made. The contrasting parental characters showed partial to total dominance in the F1 hybrid, while the back cross plants were similar to cassava in most of their characters. Eleven of the twelve backeross plants exhibited resistance to Cassava mosaic under field conditions. Karyological similarities and differences as resolved on the basis of a comparative study of the karyotypes of the cassava parent and coara rubber were corroborated by the study of chromosomal pairing in the F1 at pachytene. Major chromosomal differentiation in the two species involved three chromosomes of their haploid complement which were represented by three heteromorphic bivalent associations in F1 each consisting of a probably basic chromosomal type and a derived type. Pachytene analyses of three back cross plants provided direct proof for random transmission of marker chromosomes of both the parents through male gametes of the F1 hybrid. An increase in the chiasma frequency in the back cross plants over the F1 hybrid at metaphase I stage was also observed. Pollen fertility of the backeross plants showed considerable variation.  相似文献

Chloroplast DNA restriction fragment length polymorphisms have been used to reconstruct the maternal phylogeny of all the known taxa in the small neotropical legume genusLeucaena. Three major plastome clades were recognized, but these did not conform with relationships between the taxa proposed on other characters from morphology, cytology or hybridization. The maternal parentage of tetraploids within the genus has been proposed. Evidence for introgression was found between diploidL. diversifolia and tetraploidL. diversifolia. The implications of these results for the origin of the cultivated taxa are discussed.  相似文献

The -anomer of glucose relative to the -anomer was more rapidly metabolized into lactate by rat erythrocytes at 37° C (/ ratio = ca. 1.3): the amounts of - and -D-glucose metabolized into lactate during 3 min were 0.21 and 0.27 mol/gHb, respectively. Also, the transport of -D-glucose into erythrocytes was more rapid than that of -D-glucose: the amounts of - and -D-glucose transported into erythrocytes during 3 min were approximately 3.5 and 5.0 mol/gHb, respectively. Glucose phosphorylation by rat erythrocyte hexokinase (i.e., a possible rate-limiting step in glycolysis) occurred at higher velocities with the -anomer than with the a-anomer (/ ratio = 1.28). The Km value of hexokinase for either anomer of glucose was 53 M. The glucose concentrations in erythrocytes incubated with - and -D-glucose reached about 1 mM in 1 min, indicating that hexokinase is almost completely saturated with glucose within less than 1 min. The results suggest that glucose phosphorylation and glucose transport are major and minor determinants, respectively, for the anomeric preference of glucose utilization in rat erythrocytes.  相似文献

The deciduous wood flora of southern Europe is characterized as a special nemoral-Submediterranean element. The distribution patterns of some representatives (total ranges and local areas in Roumania) are described and explained by recent climatic conditions. The northern limits of Submediterranean and Submediterranean-middle European deciduous forest taxa exhibit a continuous gradation, but only a few species with a wide ecological amplitude extend into the temperate zone.  相似文献

Picoplankton (plankton 3 m) biomass was determined by flow cytometry in three European estuarine systems (Krka Estuary in Croatia, Rhône Delta in France, and Lena Delta and Laptev Sea in Russia). The size of natural phytoplankton groups was obtained by a calibration curve, with different picoplankton's strains (from 1.6 to 3.4 m), measured by a Coulter counter (size) and a flow cytometer (light-scattering). Two natural groups of picoplankton were identified by flow cytometry in the three systems: Synechococcus sp and picoeukaryotes. Picoplankton cells abundance ranged between: 2800 and 42000, 5000 and 37000, 1000 and 50000 cells ml–1 in the Krka estuary, in the Rhône delta and in the Lena-Laptev system, respectively. In the Krka estuary, picoplankton biomass ranges between 11 and 68 gC l–1. It can make up as much as 88% of the total photosynthetic plankton population and 50% of total organic particulate carbon. Picoplankton biomass was greater in the summer than in the autumn. At the halocline layer this biomass can attempt ca. 390 gC l–1during the summer cruise. In the Rhône delta, a lower picoplankton biomass (6–39 gC l–1) was observed at the end of the winter. These biomass represented between 0.4 and 22% of the particulate organic carbon, which could reach 71% of the total photosynthetic plankton biomass at the marine station. In the Lena-Laptev system, picoplankton biomass varied between 6 and 56 gC l–1 in surface waters. Picoplankton biomass decreased with depth, but picoeukaryotes were still observed in deep samples (20, 30 m) in the Laptev Sea, showing a considerable autotrophic activity in spite of low temperatures (0–1 °C). Although the widely dispersed estuary geographic distribution and their different estuarine characteristics, the data point out that these small organisms can also play an important role in the transfer of organic carbon from rivers to oceans and that flow cytometry can be able to detect these small cells in turbid systems.  相似文献

The levels of certain essential amino acids, in particular cysteine, lysine and methionine, in the seed storage protein of a commercial spring variety of rape, Brassica napus, have been increased by the introduction of an antisense gene for cruciferin, which is the most abundant storage protein in rapeseed. The antisense construct contained part of the cruA gene in an inverted orientation, and the gene was driven by the 5 flanking region of the gene for napin such that antisense RNA was expressed in a seed-specific manner. The construct was introduced by Agrobacterium-mediated gene transfer. In self-pollinated seeds (T1 seeds) of transgenic plants there was a reduction in the levels of the 11 and 2/32/3 subunits of cruciferin, whereas the level of the 44 subunit was unchanged. The total protein and lipid contents of transgenic seeds did not differ significantly from that of normal seeds. Seeds with reduced amounts of cruciferin accumulated higher amounts of napin than non-transformed seeds, but the level of oleosin was unaffected. Amino-acid analysis of the seed storage protein revealed that T1 seeds with reduced amounts of cruciferin contained higher relative levels of three essential amino acids, namely, lysine, methionine and cysteine, with increases of 10%, 8% and 32% over the respective levels in non-transgenic seeds (B. napus cv Westar).  相似文献

Summary A novel cyclic dipeptide —cyclo[(S)-His-(S)-NorArg] — has been prepared which catalyzes an enantioselective version of the Strecker amino acid synthesis. The catalyst, when present in 2 mol % quantity in methanol solution, catalyzes the addition of hydrogen cyanide toN-alkylimines to afford-amino nitriles in high yield and high enantiomeric excess. Furthermore, acid hydrolysis ofN-benzhydryl--amino nitriles afforded the corresponding-amino acids directly. This methodology affords a variety of arylglycines in exceptionally high enantiomeric excess, but aliphatic amino acids were obtained with low enantioselectivity. Current efforts are underway to expand the scope of this reaction, as well as to elucidate the mechanism of catalysis and the roles played by substrate and catalyst in determining the stereochemical outcome of the reaction.A preliminary communication on this work has recently appeared in: Iyer MS, Gigstad KM, Namdev ND, Lipton MA (1996) J Am Chem Soc 118: 4910–4911.  相似文献

Partitioning of water resources among plants of a lowland tropical forest   总被引:3,自引:0,他引:3  
Source water used by plants of several species in a semi-evergreen lowland tropical forest on Barro Colorado Island, Panama, was assessed by comparing the relative abundance of deuterium, D, versus hydrogen, H (stable hydrogen isotope composition, D) in xylem sap and in soil water at different depths, during the dry season of 1992. Ecological correlates of source water were examined by comparing xylem water D values with leaf phenology, leaf water status determined with a pressure chamber, and rates of water use determined as mass flow of sap using the stem heat balance method. Soil water D values decreased sharply to 30 cm, then remained relatively constant with increasing depth. Average D values were-13, for 0–30 cm depth and-36.7 for 30–100 cm depth. Soil water D values were negatively associated with soil water content and soil water potential. Concurrent analyses of xylem water revealed a high degree of partitioning of water resources among species of this tropical forest. Xylem water D of deciduous trees (average=-25.3±1.4) was higher than that of evergreen trees (average=-36.3±3.5), indicating that evergreen species had access to the more abundant soil water at greater depth than deciduous species. In evergreen shade-tolerant and high-light requiring shrubs and small trees, D of xylem water was negatively correlated with transpiration rate and leaf water potential indicating that species using deeper, more abundant water resources had both higher rates of water use and more favorable leaf water status.  相似文献
