Phrynocephalus erythrurus of the Qiangtang Plateau occupies the highest regions of any reptile on earth. Here, we report mitochondrial DNA haplotypes sampled throughout the distribution of P. erythrurus and analyze patterns of genetic divergence among populations. The species diverged into two major lineages/subspecies at 3.67 mya corresponding to the Northern and Southern Qiangtang Plateau. The Northern Qiangtang lineage diverged into two subpopulations at 2.76 mya separated by the Beilu River Region and Wulanwula Mountains. Haplotypes from the southern Qiangtang lineage diverged 0.98 mya as a star-shaped pattern. Analyses of molecular variance indicated that most of the observed genetic variation occurred among populations/regions implying long-term interruptions to gene flow. There was no evidence of sudden recent range expansions within any of the clades/lineages. NCPA infers allopatric fragmentation and restricted gene flow as the most likely mechanisms of population differentiation. Our results also indicate the presence of at least three refugia since the Hongya glaciation. Mountain movement and glaciations since mid-Pliocene are considered to have shaped phylogenetic patterns of P. erythrurus. P. erythrurus parva is suggested as a valid subspecies of P. erythrurus. Using four calibration points, we estimate an evolutionary rate of 0.762% divergence per lineage per million years for a mitochondrial genomic segment comprising the genes encoding ND2, tRNATrp and tRNAAla.  相似文献

Giant petrels Macronectes spp. are not thought to be important predators of albatross chicks, although they are known to kill pre-fledging Thalassarche and Phoebetria albatrosses. We report the first records of predation of healthy great albatross Diomedea spp. chicks, killing wandering albatrosses D. exulans at night on sub-Antarctic Marion Island. Breeding success of this species has decreased markedly in the area where attacks occurred, suggesting that giant petrel predation events are a recent phenomenon. Mouse attacks on wandering albatross chicks may have contributed to the development of this hunting technique. We also report the first observations of giant petrel predation on pre-fledging grey-headed albatross T. chrysostoma chicks as well as additional records of sooty albatross P. fusca chicks being targeted. Only adult northern giant petrels M. halli have been confirmed to kill albatross chicks on Marion Island. Given the threatened status of wandering albatrosses, and the importance of Marion Island for this species, monitoring of their breeding success is necessary to assess whether the predation of chicks by giant petrels spreads around the island.  相似文献

Fat dormouse is a squirrel-like rodent which is closely tied to deciduous forest ecosystem in southwestern Eurasia. As such it is a valuable indicator of forest survival in refugia during glacial-interglacial periods. Previous phylogeographic analyses uncovered divergent fat dormouse lineages in southern refugia in Italy and the Balkans, but retrieved a surprisingly low overall genetic diversity across the majority of the species’ range. We explored 812 bp long fragment of a cytochrome b (cyt b) gene in ten fat dormice from refugial Hyrcanian forests in northern Iran. We identified 10 new cyt b haplotypes, which generated a total dataset of 28 fat dormouse haplotypes. The phylogenetic reconstruction clustered the new haplotypes into the Iranian lineage which hold a sister position against all other fat dormouse haplotypes from Europe and Asia Minor. The divergence between these lineages suggests a fragmentation event of an ancestral population at 5.76 mya (95% HPD = 3.21–8.92). This early evolutionary divergence was possibly triggered in the Middle East by dramatically divergent environmental conditions at the Messinian Salinity Crisis. The divergence clearly exceeds the intraspecific divergence, and is well within the range between congeneric rodent species. We suggest a long-term persistence of the Iranian lineage in the Hyrcanian refugium which is consitent with a high number of endemics along the southern Caspian coastal areas.  相似文献

Cytochrome b5 is the main electron acceptor of cytochrome b5 reductase. The interacting domain between both human proteins has been unidentified up to date and very little is known about its redox properties modulation upon complex formation. In this article, we characterized the protein/protein interacting interface by solution NMR and molecular docking. In addition, upon complex formation, we measured an increase of cytochrome b5 reductase flavin autofluorescence that was dependent upon the presence of cytochrome b5. Data analysis of these results allowed us to calculate a dissociation constant value between proteins of 0.5 ± 0.1 μM and a 1:1 stoichiometry for the complex formation. In addition, a 30 mV negative shift of cytochrome b5 reductase redox potential in presence of cytochrome b5 was also measured. These experiments suggest that the FAD group of cytochrome b5 reductase increase its solvent exposition upon complex formation promoting an efficient electron transfer between the proteins.  相似文献

Fifty-seven fecal samples were collected from giant pandas (Ailuropoda melanoleuca) in the China Conservation and Research Centre for the Giant Panda (CCRCGP) in Sichuan and examined for Cryptosporidium oocysts by Sheather's sugar flotation technique. An 18-year-old male giant panda was Cryptosporidium positive, with oocysts of an average size of 4.60 × 3.99 μm (n = 50). The isolate was genetically analyzed using the partial 18S rRNA, 70 kDa heat shock protein (HSP70), Cryptosporidium oocyst wall protein (COWP) and actin genes. Multi-locus genetic characterization indicated that the present isolate was different from known Cryptosporidium species and genotypes. The closest relative was the Cryptosporidium bear genotype, with 11, 10, and 6 nucleotide differences in the 18S rRNA, HSP70, and actin genes, respectively. Significant differences were also observed in the COWP gene compared to Cryptosporidium mongoose genotype. The homology to the bear genotype at the 18S rRNA locus was 98.6%, which is comparable to that between Cryptosporidium parvum and Cryptosporidium hominis (99.2%), or between Cryptosporidium muris and Cryptosporidium andersoni (99.4%). Therefore, the Cryptosporidium in giant pandas in this study is considered as a new genotype: the Cryptosporidium giant panda genotype.  相似文献

In alkaline media (pH 12) a catalytic peroxidase activity of cytochrome b5 was found associated to a different conformational state. Upon incubation at this pH, cytochrome b5 electronic absorption spectrum was altered, with disappearance of characteristic bands of cytochrome b5 at pH 7.0. The appearance of new electronic absorption bands and EPR measurements support the formation of a cytochrome b5 class B hemichrome with an acquired ability to bind polar ligands. This hemichrome is characterized by a negative formal redox potential and the same folding properties than cytochrome b5 at pH 7. The acquired peroxidase-like activity of cytochrome b5 found at pH 12, driven by a hemichrome formation, suggests a role of this protein in peroxidation products propagation.  相似文献

Hybridization in natural populations provides an opportunity to study the evolutionary processes that shape divergence and genetic isolation of species. The emergence of pre-mating barriers is often the precursor to complete reproductive isolation. However, in recently diverged species, pre-mating barriers may be incomplete, leading to hybridization between seemingly distinct taxa. Here we report results of a long-term study at Bird Island, South Georgia, of the extent of hybridization, mate fidelity, timing of breeding and breeding success in mixed and conspecific pairs of the sibling species, Macronectes halli (northern giant petrel) and M. giganteus (southern giant petrel). The proportion of mixed-species pairs varied annually from 0.4–2.4% (mean of 1.5%), and showed no linear trend with time. Mean laying date in mixed-species pairs tended to be later than in northern giant petrel, and always earlier than in southern giant petrel pairs, and their breeding success (15.6%) was lower than that of conspecific pairs. By comparison, mixed-species pairs at both Marion and Macquarie islands always failed before hatching. Histories of birds in mixed-species pairs at Bird Island were variable; some bred previously or subsequently with a conspecific partner, others subsequently with a different allospecific partner, and some mixed-species pairs remained together for multiple seasons. We also report the first verified back-crossing of a hybrid giant petrel with a female northern giant petrel. We discuss the potential causes and evolutionary consequences of hybridization and back-crossing in giant petrels and summarize the incidence of back-crossing in other seabird species.  相似文献

In the past years, late summer blooms of the bioluminescent dinoflagellate Alexandrium ostenfeldii have become a recurrent phenomenon in coastal waters of the central and Northern Baltic Sea. This paper reports exceptionally high cell concentrations (105 to 106 cells L?1) of the species found during bioluminescent blooms in 2003 and 2004 in a shallow embayment of the Åland archipelago at the SW coast of Finland. Clonal cultures were established for morphological, molecular, toxicological and ecophysiological investigations to characterize the Finnish populations and compare them to other global A. ostenfeldii isolates. The Finnish isolates exhibited typical morphological features of A. ostenfeldii such as large size, a prominent ventral pore and an orthogonally bent first apical plate. However, unambiguous differentiation from closely related Alexandrium peruvianum was difficult due to considerable variation of sulcal anterior plate shapes. The Finnish strains were genetically distinct from other isolates of the species, but phylogenetic analyses revealed a close relationship to isolates from southern England and an A. peruvianum morphotype from the Spanish Mediterranean. Together these isolates formed a distinct clade which was separated from a clade containing other Northern European, North American and New Zealand populations. Toxin analyses confirmed the presence of the PSP toxins GTX2, GTX3 and STX in both Finnish isolates with GTX3 being the dominant toxin. Total relative PSP toxin contents were moderate, ranging from approximately 6 to 15 fmol cell?1 at local salinities of 5 and 10 psu, respectively. Spirolides were not detected. Salinity tolerance experiments showed that the Finnish isolates were well adapted to grow at the low salinities of the Baltic Sea. With a salinity range of approximately 6 to 20–25 psu, Baltic populations are physiologically distinct from their marine relatives. Vigorous production of different cyst types in the cultures suggest that cysts may play a crucial role in the survival and retainment of A. ostenfeldii populations in the Baltic Sea.  相似文献

Phylogeographic relationships, the timing of clade diversification, and the potential for cryptic diversity in the Slender Madtom, Noturus exilis, was investigated using mitochondrial Cyt b, nuclear RAG2, shape analysis, and meristic and pigmentation data. Three well-supported and deeply divergent clades were recovered from analyses of genetic data: Little Red River (White River drainage) clade, Arkansas + Red River (Mississippi River) clade, and a large clade of populations from the rest of the range of the species. Recovered clades showed little to no diagnostic morphological differences, supporting previous hypotheses of morphological conservatism in catfishes, and indicating morphology may commonly underestimate diversity in this group of fishes. The Little Red River clade is the most distinct lineage of N. exilis with 11 POM pores (vs. 10 in other populations) and unique Cyt b haplotypes and RAG 2 alleles. However, treating it as a species separate from N. exilis would imply that the other major clades of N. exilis are more closely related to one another than they are to the Little Red River clade, which was not supported.The UCLN age estimate for Noturus was 23.9 mya (95% HPD: 13.49, 35.43), indicating a late Oligocene to early Miocene origin. The age of N. exilis was estimated as late Miocene at 9.7 mya (95% HPD: 5.32, 14.93). Diversification within the species spanned the late Miocene to mid-Pleistocene. The largest clade of N. exilis, which dates to the late Miocene, includes populations from the unglaciated Eastern and Interior Highlands as well as the previously glaciated Central Lowlands. Diversification of this clade coincides with a drastic drop in sea-level and diversification of other groups of Central Highlands fishes (Centrarchidae and Cyprinidae). Sub-clades dating to the Pleistocene show that northern populations occurring in previously glaciated regions resulted from dispersal from populations in the Ozarks up the Mississippi River following retreat of the Pleistocene glaciers. Pre-Pleistocene vicariance, such as drainage pattern changes of the Mississippi River, also played a prominent role in the history of the species. The incorporation of a temporal estimate of clade diversification revealed that in some instances, phylogeographic breaks shared with other aquatic species were best explained by different or persistent vicariant events through time, rather than a single shared event.  相似文献

A lone petrel was shot from the decks of an Italian warship (the ‘Magenta’) while it was sailing the South Pacific Ocean in 1867, far from land. The species, unknown to science, was named the ‘Magenta petrel’ (Procellariiformes, Procellariidae, Pterodroma magentae). No other specimens of this bird were collected and the species it represented remained a complete enigma for over 100 years. We compared DNA sequence of the mitochondrial cytochrome b gene from the Magenta petrel to that of other petrels using phylogenetic methods and ancient DNA techniques. Our results strongly suggest that the Magenta petrel specimen is a Chatham Island taiko. Furthermore, given the collection location of the Magenta petrel, our finding indicates that the Chatham Island taiko forages far into the Pacific Ocean (near South America). This has implications for the conservation of the taiko, one of the world's rarest seabirds.  相似文献

Guidelines for visitors to sub-Antarctic Marion Island recommend 15 and 100 m minimum approach distances for breeding Northern (Macronectes halli) and Southern (Macronectes giganteus) Giant Petrels, respectively. Using artificial eggs containing FM transmitters, we measured the heart rate responses of incubating Northern Giant Petrels to pedestrian approaches. The mean resting heart rate was 80 beats per minute. Heart rates increased upon the detection of a person approximately 40 m away, and continued to increase during the approach to 5 m. Maximum increases over resting heart rate in response to natural disturbances and human approach were 97 and 204%, respectively. Northern Giant Petrels appear at least as sensitive to human disturbance as their congenerics. While low-key disturbance is unlikely to affect this solitary breeder as severely as it would the colonial Southern Giant Petrel, improved protection from disturbance could be achieved by restricting human passage through breeding colonies of Northern Giant Petrels to defined paths.  相似文献

The Neotropical knifefish genus Gymnotus is the most broadly distributed and the most diverse (34 + species) gymnotiform genus. Its wide range includes both Central and South American drainages, including the Amazon, Orinoco, and La Plata Basins. Like all gymnotiforms, Gymnotus species produce weak electric fields for both navigation and communication, and these fields exhibit interspecific variation in electric waveform characteristics. Both biogeography and electric signal evolution can profitably be analyzed in a phylogenetic context. Here, we present a total evidence phylogeny for 19 Gymnotus species based on data from the mitochondrial cytochrome b and 16S genes (1558 bp), the nuclear RAG2 gene (1223 bp), and 113 morphological characters. Our phylogenetic hypothesis resolves five distinct Gymnotus lineages. In a previous morphology-based analysis, the Central American Gymnotus cylindricus lineage was hypothesized as the sister group to all other Gymnotus species. In our analysis, the G. cylindricus lineage is nested within South American species, and molecular age estimates support a relatively recent origin for the clade in Central America. Phylogenetic optimization of electric signal waveforms indicate that the ancestral state in Gymnotus is a multiphasic (4 + phases of alternating polarity) condition, and independent phase loss has occurred in multiple lineages. Gymnotus is a model group for understanding Neotropical diversification and the evolution of communication at a continental scale.  相似文献

To investigate the phylogenetic position of Blakiston’s fish owl (Bubo blakistoni), we sequenced the mitochondrial (mt) DNA control region and cytochrome b (cyt b) for nine Bubo species. Maximum-likelihood analyses of combined control region and cyt b sequences, and cyt b sequences alone, showed that species formerly placed in genus Ketupa comprise a monophyletic group. Unexpectedly, we discovered a long cluster of 20–25 tandem repeat units 77 or 78 bp long in the third control region domain in four of the nine Bubo species for which the control region was sequenced (B. blakistoni, B. flavipes, and B. ketupu in the Ketupa clade; B. lacteus), leading to overall control region lengths of 3.0–3.8 kpb estimated from agarose gel electrophoresis. The control region in B. lacteus is the longest (3.8 kbp) reported to date in vertebrates. Sequencing of eight repeat units at each end of the cluster in 20 B. blakistoni individuals detected several types of repeat units 77 or 78 bp long, and six patterns in the order of unit types. The occurrence of a repeat cluster in all three species examined in the Ketupa clade suggests their common ancestor also had a cluster, whereas a maximum parsimony tree showed repeat-unit types grouping by species, rather than by paralog groups, suggesting independent origins of the clusters. We reconcile these results with a turnover model, in which the range in cluster-length variation and unit types at the 5′ end are hypothetically functionally constrained by the protein-binding function of the control region, but otherwise there is a continual turnover of units in evolutionary time, with new unit types arising through mutations, proliferating by duplication of single and double repeat blocks, and being lost through deletion. Estimated free energies for reconstructed secondary structures of single and especially pairs of repeat units were higher than for homologous single-unit blocks in species lacking a repeat cluster, supporting slipped-strand mispairing as the mechanism of cluster turnover.  相似文献

The South American sea lion (Otaria byronia) (SSL) is a widespread opportunistic predator that inhabits waters ranging from Southern Ecuador to Southern Chile in the Pacific Ocean and from Southern Brazil to Southern Argentina in the Atlantic. SSL abundance estimates, as for many pinniped species, have relied on shore censuses, with the uncertainty that an indeterminate number of individuals at sea might remain uncounted. The proportion of the population that remains at sea during censuses and their distribution patterns are not clear and has been scarcely assessed. We used line transect sampling to gather information about at-sea abundance, density and spatial distribution of SSL in coastal waters off the Chilean Northern Patagonia. A total of 123 groups were sighted while on-effort, with an estimated density of 0.393 ind./km2 (95%CI = 0.262–0.591) and a total abundance, for the surveyed area, of 13,721 individuals (95%CI = 9127–20,627). Even when our survey was rather restricted in spatial range, generalized additive model results showed that depth, distance to SSL rockeries, coastline complexity and geographical coordinates had a significant influence on SSL spatial distribution. The results on abundance and spatial distribution of SSL at sea are discussed in terms of current population estimates. Our substantial at-sea abundance estimate suggests that shore censuses might have been historically biased. Several aspects should be taken into account in further research on SSL abundance, such as including pelagic waters and undertaking simultaneously on-rockery and marine SSL abundance estimates. The results presented here provide valuable insights for revisiting and possibly improve population estimates of SSL and other pinnipeds species worldwide. This is particularly relevant when management and conservation actions need to be taken on those species that have strong interactions with fisheries and aquaculture.  相似文献

Phylogenetic analyses using up to 1532 base pairs (bp) of mitochondrial DNA from 106 specimens of Neotropical Mabuya, including 18 of the 19 recognized South American and Mesoamerican species, indicate that most species of the genus are monophyletic, including M. nigropunctata that had previously been reported to be paraphyletic. The present results shows that this species includes three highly divergent and largely allopatric lineages restricted to occidental, meridional, and oriental Amazonia. Our dataset demonstrates that previous claims regarding the paraphyletic status of M. nigropunctata and the phylogenetic relationships within this species complex based on the analysis of three mitochondrial and four nuclear genes (approx. 5000 bp) were erroneous and resulted from two contaminated cytochrome b sequences.The phylogenetic results indicate that diversification in the Neotropical genus Mabuya started approximately in the Middle Miocene (15.5–13.4 Ma). The divergence dates estimated for the Mabuya nigropunctata species complex suggest that the major cladogenetic events that produced the three main groups (occidental (oriental + meridional)) occurred during the Late Miocene. These estimations show that diversification within the M. nigropunctata species complex was not triggered by the climatic changes that occurred during the Pleistocene, as has been suggested by several authors. Rather, our data support the hypothesis that the late tertiary (essentially Miocene epoch) was a period that played a very important role in the generation of biological diversity in the Amazonian forests.Speciation between Mabuya carvalhoi, endemic to the coastal mountain range of Venezuela, and M. croizati, restricted to the Guiana Shield, occurred during the Middle Miocene and may have been as the result of a vicariant event produced by the formation of the present day Orinoco river drainage basin and the consequent appearance of the Llanos del Orinoco, which acted as a barrier to dispersal between these two species. The split between M. bistriata and M. altamazonica and between the occidental and (meridional + oriental) clades of M. nigropunctata fits very well with the biogeographic split between the eastern and western Amazon basins reported for several other taxa.  相似文献

Biological communities are shaped by competition between and within species. Competition is often reduced by inter‐ and intraspecific specialization on resources, such as differencet foraging areas or time, allowing similar species to coexist and potentially contributing to reproductive isolation. Here, we examine the simultaneous role of temporal and spatial foraging segregation within and between two sympatric sister species of seabirds, Northern Macronectes halli and Southern Macronectes giganteus Giant Petrels. These species show marked sexual size dimorphism and allochrony (with earlier breeding by Northern Giant Petrels) but this is the first study to test for differences in foraging behaviours and areas across the entire breeding season both between the two species and between the sexes. We tracked males and females of both species in all breeding stages at Bird Island, South Georgia, to test how foraging distribution, behaviour and habitat use vary between and within species in biological time (incubation, brood‐guard or post‐brood stages) and in absolute time (calendar date). Within each breeding stage, both species took trips of comparable duration to similar areas, but due to breeding allochrony they segregated temporally. Northern Giant Petrels had a somewhat smaller foraging range than Southern Giant Petrels, reflecting their greater exploitation of local carrion and probably contributing to their recent higher population growth. Within species, segregation was spatial, with females generally taking longer, more pelagic trips than males, although both sexes of both species showed unexpectedly plastic foraging behaviour. There was little evidence of interspecific differences in habitat use. Thus, in giant petrels, temporal segregation reduces interspecific competition and sexual segregation reduces intraspecific competition. These results demonstrate how both specialization and dynamic changes in foraging strategies at different scales underpin resource division within a community.  相似文献

Genetic diversity and population structure of two species of South American pinnipeds, Otaria flavescens and Arctocephalus australis, from colonies located along the south-eastern coast of South America, were analysed using mitochondrial DNA haplotypes and compared with two populations of these species from the Pacific coast. A 445 base-pair segment, that included the tRNA-Glu gene (31 bp) and the adjacent cytochrome b gene (414 bp), was amplified using the polymerase chain reaction and sequenced directly. O. flavescens and A. australis showed six and seven haplotypes with 12 and 20 polymorphic sites, respectively. In the Atlantic Ocean there was an individual of A. australis that showed an haplotype that was highly divergent from the others. If this haplotype is excluded, the pattern of haplotype differentiation obtained for both species indicated a possible bottleneck that would have occurred 110,000 years ago, which also affected other pinnipeds. Colonies of the Atlantic and the Pacific did not share haplotypes. This result, based on a limited number of samples for the comparisons between oceans, suggests that populations from both oceans correspond to different evolutionarily significant units. O. flavescens on the Atlantic coast shows two clusters of breeding colonies in Uruguay and Patagonia, separated by a thousand kilometres. Colonies within clusters did not show significant differences in haplotype frequencies, but the difference between the clusters was significant, suggesting that they correspond to different conservation stocks.  相似文献

A gene encoding an unusually small dynein heavy chain homologue, hDYHH, was cloned from the genome of a free-living diplomonad, Hexamita inflata (Hi). The open reading frame (ORF) of hDYHH is 867 bp and encodes a polypeptide of 289 amino acids (aa), hDYHH. hDYHH is homologous to the region around the third P-loop ATP-binding site of several dynein heavy chain polypeptides that are around 4000 aa. Northern blot analysis showed that hDYHH is expressed in vivo and that the mRNA length (approximately 1.8 kb) is consistent with the gene length (1.67 kb). Southern blot analysis indicated that there are hDYHH homologues within the Hi genome, possibly including a longer dynein heavy chain gene. An hDYHH homologue was also identified in Hexamita pusilla (Hp). hDYHH is the first full-length protein-encoding gene cloned from Hexamita.  相似文献

The phylogeography of the Thomomys bottae–umbrinus complex in the United States and Mexico was assessed with sequences of the mitochondrial cytochrome b gene. These sequences were obtained from 225 individuals representing 108 locations over the range, including 56 sequences from GenBank. 110 (500 bp) sequences were used for Bayesian inference and neighbor-joining analyses, and 34 (1140 bp) specimens from the main clades obtained from the Bayesian inference were used in maximum-parsimony and maximum-likelihood analyses. The different analyses indicate significant variation within the species complex that averages 13% among major groups of genetic differences among Thomomys bottae–umbrinus. The overall pattern of geographic variation is not concordant with the current taxonomy. To the contrary, eight monophyletic groups are supported by all analyses and can be considered phylogenetic species. Overall divergence among these groups appears influenced by historical biogeographic events active during the Pliocene and Pleistocene.  相似文献
