The results of the study of the inhibiting effect of neutral salts upon the clotting tendency of fibrinogen by thrombin may be summarised as follows: Salts like NaCl and KCl inhibit only weakly. Salts of the same cation (K) with monovalent anions of different ionic radius are the more active the larger the anion (Cl'',Br'',I''). Salts of the same cation with anions of different valency are the more active the higher the charge of the anion (1–1 <1–2 <1–3 <1–4). Salts with the same anion with cations of different valency show stronger inhibition in the case of cations of higher charge (K,Na < Mg••, Ca••, Sr••, Ba••). Salts with the same anion and cations of the same charge, but of different radius, are the more active the larger the cation (but with an inversion between Mg•• and Ca•• in the series of the alkali earths, which is not infrequent in biocolloids). These results show that the clotting of fibrinogen with thrombin is, at least partly, caused by a coacervation process, due to electrostatic attraction between positive and negative groups. Background

Cytoplasmic pH homeostasis in Escherichia coli includes numerous mechanisms involving pH-dependent catabolism and ion fluxes. An important contributor is transmembrane K+ flux, but the actual basis of K+ compensation for pH stress remains unclear. Osmoprotection could mediate the pH protection afforded by K+ and other osmolytes.

Methods and Principal Findings

The cytoplasmic pH of E. coli K-12 strains was measured by GFPmut3 fluorimetry. The wild-type strain Frag1 was exposed to rapid external acidification by HCl addition. Recovery of cytoplasmic pH was enhanced equally by supplementation with NaCl, KCl, proline, or sucrose. A triple mutant strain TK2420 defective for the Kdp, Trk and Kup K+ uptake systems requires exogenous K+ for steady-state pH homeostasis and for recovery from sudden acid shift. The K+ requirement however was partly compensated by supplementation with NaCl, choline chloride, proline, or sucrose. Thus, the K+ requirement was mediated in part by osmolarity, possibly by relieving osmotic stress which interacts with pH stress. The rapid addition of KCl to strain TK2420 suspended at external pH 5.6 caused a transient decrease in cytoplasmic pH, followed by slow recovery to an elevated steady-state pH. In the presence of 150 mM KCl, however, rapid addition of another 150 mM KCl caused a transient increase in cytoplasmic pH. These transient effects may arise from secondary K+ fluxes occurring through other transport processes in the TK2420 strain.

Conclusions

Diverse osmolytes including NaCl, KCl, proline, or sucrose contribute to cytoplasmic pH homeostasis in E. coli, and increase the recovery from rapid acid shift. Osmolytes other than K+ restore partial pH homeostasis in a strain deleted for K+ transport.  相似文献

1. The question of the critical pore diameter for streaming potential is discussed. 2. The surface charge is calculated for cellulose in contact with solutions of K3PO4, K2CO3, K2SO4, KCl, and ThCl4. 3. The surface charge of cellulose in contact with a solution of 2 x 10–4 N NaCl is calculated as a function of temperature and is found to show a sharp break at 39°. This is interpreted in terms of the change of the specific heat of water. 4. A marked ion antagonism is found in NaCl:KCl, KCl:MgCl2, NaCl:MgCl2, NaCl:CaCl2, KCl:CaCl2, CaCl2:MgCl2 mixtures when the surface charge is calculated as a function of concentration.  相似文献

Atriplex prostrata was grown for one month in nutrient solutions with NaCl, KCl, Na2SO4, and K2SO4 (at osmotic potentials of 0, –0.75, –1.00, and –1.50 MPa). Plants treated with K2SO4 had less glycinebetaine at –1.0 and –1.50 MPa than those treated with Na+ salts, probably due to the inhibitory effects of K+ on glycinebetaine accumulation.  相似文献

Twenty days’ exposure to 50 or 100 mM NaCl in the rooting medium substantially increased fresh and dry weights of seedling shoots of the recretohalophyte Limonium sinense while 200 or 300 mM were increasingly inhibitory. KCl treatment was only slightly stimulating (50 mM) or strongly inhibitory (100–300 mM). Lesser effects on leaf area were also seen. Diameter of foliar salt glands was significantly larger than that of controls in 100 and 200 mM NaCl with the effect being reversed at higher concentrations. Gland enlargement was also observed in the presence of 100 mM KCl, while larger concentrations reduced gland size. Generally, gland diameter was larger in the presence of NaCl than in KCl. NaCl and KCl also increased gland number per leaf and secretion rate per gland. At 100 and 200 mM NaCl or KCl, Na+ secretion per leaf from NaCl-treated plants exceeded K+ secretion rate from KCl-treated plants while at 200 mM, Na+ secretion per gland was significantly higher for Na+ than for K+. Evidence of cell death in leaves of salt-treated plants using Evans blue staining indicates that release of cell contents through loss of membrane integrity contributed to the secretion values. We conclude that the greater tolerance of L. sinenseto to NaCl compared to KCl is linked to the more effective secretion of Na+ than of K+ and, in turn, to a greater stimulation of salt gland formation and activity and larger gland diameter.  相似文献

The present study aimed to determine the mechanism of cation-selective secretion by multicellular salt glands. Using a hydroponic culture system, the secretion and accumulation of Na+ and K+ in Tamarix ramosissima and T. laxa under different salt stresses (NaCl, KCl and NaCl+KCl) were studied. Additionally, the effects of salt gland inhibitors (orthovanadate, Ba2+, ouabain, tetraethylammonium (TEA) and verapamil) on Na+ and K+ secretion and accumulation were examined. Treatment with NaCl (at 0–200 mmol L−1 levels) significantly increased Na+ secretion, whereas KCl treatment (at 0–200 mmol L−1 levels) significantly increased K+ secretion. The ratio of secretion to accumulation of Na+ was higher than that of K+. The changes in Na+ and K+ secretion differed after adding different ions into the single-salt solutions. Addition of NaCl to the KCl solution (at 100 mmol L−1 level, respectively) led to a significant decrease in K+ secretion rate, whereas addition of KCl to the NaCl solution (at 100 mmol L−1 level, respectively) had little impact on the Na+ secretion rate. These results indicated that Na+ secretion in Tamarix was highly selective. In addition, Na+ secretion was significantly inhibited by orthovanadate, ouabain, TEA and verapamil, and K+ secretion was significantly inhibited by ouabain, TEA and verapamil. The different impacts of orthovanadate on Na+ and K+ secretion might be the primary cause for the different Na+ and K+ secretion abilities of multicellular salt glands in Tamarix.  相似文献

Summary Simultaneous measurements of net ion and water fluxes were made in the stripped intestine of the seawater eel, and the relationship between Na+, K+, Cl and water transport were examined in the presence of mucosal KCl and serosal NaCl Ringer (standard condition). When Cl was removed from both sides of the intestine, net K+ flux from mucosa to serosa was reduced, accompanied by complete blockage of water absorption. Since it has been shown that net Cl and water fluxes depend on K+ transport under the standard condition (Ando 1983), the interdependence of K+ and Cl transport suggests the existence of a coupled KCl transport system, while the parallelism between the net Cl and water fluxes suggests that water absorption is linked to the coupled KCl transport. The coupled KCl and water transport were inhibited by treatment with ouabain or with Na+-free Ringer solutions, suggesting the existence of a Na+-dependent KCl transport system and linkage of water absorption to the coupled Na+–K+–Cl transport. Since ouabain blocked the active Na+–K+–Cl transport almost completely, the permeability coefficients for K+ and Na+ through the paracellular shunt pathway were estimated as PK=0.076 and PNa=0.058 cm/h, and PCl was calculated as 0.005 cm/h. Although Na+-independent K+ and Cltt- fluxes were observed again in the present study, these fluxes were not inhibited by CN, ouabain or diuretics, and evoked even after blocking the Na+–K+–Cl transport completely with ouabain. These results indicate that the Na+-independent K+ and Cl fluxes are distinct from the active Na+–K+–Cl transport and are not themselves active.  相似文献

Summary Ehrlich ascites tumor cells resuspended in hypotonic medium initially swell as nearly perfect osmometers, but subsequently recover their volume within 5 to 10 min with an associated KCl loss. 1. The regulatory volume decrease was unaffected when nitrate was substituted for Cl, and was insensitive to bumetanide and DIDS. 2. Quinine, an inhibitor of the Ca2+-activated K+ pathway, blocked the volume recovery. 3. The hypotonic response was augmented by addition of the Ca2+ ionophore A23187 in the presence of external Ca2+, and also by a sudden increase in external Ca2+. The volume response was accelerated at alkaline pH. 4. The anti-calmodulin drugs trifluoperazine, pimozide, flupentixol, and chlorpromazine blocked the volume response. 5. Depletion of intracellular Ca2+ stores inhibited the regulatory volume decrease. 6. Consistent with the low conductive Cl permeability of the cell membrane there was no change in cell volume or Cl content when the K+ permeability was increased with valinomycin in isotonic medium. In contrast, addition of the Ca2+ ionophore A23187 in isotonic medium promoted Cl loss and cell shrinkage. During regulatory volume decrease valinomycin accelerated the net loss of KCl, indicating that the conductive Cl permeability was increased in parallel with and even more than the K+ permeability. It is proposed that separate conductive K+ and Cl channels are activated during regulatory volume decrease by release of Ca2+ from internal stores, and that the effect is mediated by calmodulin.  相似文献

The hexa-coordinate heme in the H2S-generating human enzyme cystathionine β-synthase (CBS) acts as a redox-sensitive regulator that impairs CBS activity upon binding of NO or CO at the reduced iron. Despite the proposed physiological relevance of this inhibitory mechanism, unlike CO, NO was reported to bind at the CBS heme with very low affinity (Kd = 30–281 μm). This discrepancy was herein reconciled by investigating the NO reactivity of recombinant human CBS by static and stopped-flow UV-visible absorption spectroscopy. We found that NO binds tightly to the ferrous CBS heme, with an apparent Kd ≤0.23 μm. In line with this result, at 25 °C, NO binds quickly to CBS (kon ∼ 8 × 103 m−1 s−1) and dissociates slowly from the enzyme (koff ∼ 0.003 s−1). The observed rate constants for NO binding were found to be linearly dependent on [NO] up to ∼ 800 μm NO, and >100-fold higher than those measured for CO, indicating that the reaction is not limited by the slow dissociation of Cys-52 from the heme iron, as reported for CO. For the first time the heme of human CBS is reported to bind NO quickly and tightly, providing a mechanistic basis for the in vivo regulation of the enzyme by NO. The novel findings reported here shed new light on CBS regulation by NO and its possible (patho)physiological relevance, enforcing the growing evidence for an interplay among the gasotransmitters NO, CO, and H2S in cell signaling.  相似文献

Osmotic adjustment of cultured tobacco (Nicotiana tabacum L. var Wisconsin 38) cells was stimulated by 10 micromolar (±) abscisic acid (ABA) during adaptation to water deficit imposed by various solutes including NaCl, KCl, K2SO4, Na2SO4, sucrose, mannitol, or glucose. The maximum difference in cell osmotic potential (Ψπ) caused by ABA treatment during adaptation to 171 millimolar NaCl was about 6 to 7 bar. The cell Ψπ differences elicited by ABA were not due to growth inhibition since ABA stimulated growth of cells in the presence of 171 millimolar NaCl. ABA caused a cell Ψπ difference of about 1 to 2 bar in medium without added NaCl. Intracellular concentrations of Na+, K+, Cl, free amino acids, or organic acids could not account for the Ψπ differences induced by ABA in NaCl treated cells. However, since growth of NaCl treated cells is more rapid in the presence of ABA than in its absence, greater accumulation of Na+, K+, and Cl was necessary for ion pool maintenance. Higher intracellular sucrose and reducing sugar concentrations could account for the majority of the greater osmotic adjustment of ABA treated cells. More rapid accumulation of proline associated with ABA treatment was highly correlated with the effects of ABA on cell Ψπ. These and other data indicate that the role of ABA in accelerating salt adaptation is not mediated by simply stimulating osmotic adjustment.  相似文献

GTP hydrolysis catalyzed in the ribosome by a complex of two polypeptide release factors, eRF1 and eRF3, is required for fast and efficient termination of translation in eukaryotes. Here, isothermal titration calorimetry is used for the quantitative thermodynamic characterization of eRF3 interactions with guanine nucleotides, eRF1 and Mg2+. We show that (i) eRF3 binds GDP (Kd = 1.9 μM) and this interaction depends only minimally on the Mg2+ concentration; (ii) GTP binds to eRF3 (Kd = 0.5 μM) only in the presence of eRF1 and this interaction depends on the Mg2+ concentration; (iii) GTP displaces GDP from the eRF1•eRF3•GDP complex, and vice versa; (iv) eRF3 in the GDP-bound form improves its ability to bind eRF1; (v) the eRF1•eRF3 complex binds GDP as efficiently as free eRF3; (vi) the eRF1•eRF3 complex is efficiently formed in the absence of GDP/GTP but requires the presence of the C-terminus of eRF1 for complex formation. Our results show that eRF1 mediates GDP/GTP displacement on eRF3. We suggest that after formation of eRF1•eRF3•GTP•Mg2+, this quaternary complex binds to the ribosomal pretermination complex containing P-site-bound peptidyl-tRNA and the A-site-bound stop codon. The guanine nucleotide binding properties of eRF3 and of the eRF3•eRF1 complex profoundly differ from those of prokaryotic RF3.  相似文献

Astroglial cells are known to release taurine in response to stimulation by a variety of stimuli including -adrenergic receptor agonists such as isoproterenol (IPR). The effects of changing osmolarity and extracellular [K+] on IPR-stimulated taurine release were studied with LRM55 cells, a continuous astroglial cell line. IPR-stimulated taurine release decreased almost 8% for each 1% increase in osmolarity, indicating that IPR-stimulated release is highly regulated by the osmolarity of the medium. IPR-stimulated taurine release was greatly enhanced when external [K+] was increased isosmotically by substituting KCl for NaCl but was strongly suppressed when external [K+] was increased hyperosmotically by adding KCl to the medium. Both IPR-stimulated and K+-stimulated taurine release depended on external [Cl]; IPR-stimulated release declined approximately in parallel to K+-stimulated release as [Cl] in the medium was reduced. The high sensitivity of IPR-stimulated release to factors that change cell volume (osmolarity, external [K+], external [Cl]) is consistent with the idea that IPR, elevated [K+], and reduced osmolarity all elicit taurine release via a single tension-controlled mechanism.Special issue dedicated to Dr. Claude Baxter.  相似文献

The role of cation and anion uptake in salt-stimulated growth of light-grown, GA3-treated lettuce (Lactuca sativa L.) hypocotyl sections was investigated. Potassium chloride (10 mm) causes a 2-fold increase in the growth rate of GA3-treated hypocotyl sections without affecting the growth rate of sections incubated in the absence of GA3. Salt uptake is the same in both treatments, and furthermore the uptake of cation and anion is stoichiometric during the first 24 hours under all incubation conditions. The importance of the anion for cation uptake is demonstrated in experiments with benzenesulfonate and iminodiacetate2−. When K+ and Na+ are supplied only as the benzenesulfonate and iminodiacetate salts, growth and cation uptake are markedly reduced compared to KCl and NaCl. Calculation of the osmotic potential of salt-treated sections based on measurement of K+ and Cl uptake suggests that the observed increase in tissue osmolality is a result of salt uptake. Similarly, uptake of ions can account for the shift in water potential when sections are incubated in 10 mm KCl. We conclude that the change in growth rate of light-grown, GA3-treated sections caused by the addition of KCl or NaCl to the incubation medium results solely from decreased water potential of the tissue due to ion uptake.  相似文献

We have performed microsecond molecular dynamics (MD) simulations to characterize the structural dynamics of cation-bound E1 intermediate states of the calcium pump (sarcoendoplasmic reticulum Ca2+-ATPase, SERCA) in atomic detail, including a lipid bilayer with aqueous solution on both sides. X-ray crystallography with 40 mM Mg2+ in the absence of Ca2+ has shown that SERCA adopts an E1 structure with transmembrane Ca2+-binding sites I and II exposed to the cytosol, stabilized by a single Mg2+ bound to a hybrid binding site I′. This Mg2+-bound E1 intermediate state, designated E1•Mg2+, is proposed to constitute a functional SERCA intermediate that catalyzes the transition from E2 to E1•2Ca2+ by facilitating H+/Ca2+ exchange. To test this hypothesis, we performed two independent MD simulations based on the E1•Mg2+ crystal structure, starting in the presence or absence of initially-bound Mg2+. Both simulations were performed for 1 µs in a solution containing 100 mM K+ and 5 mM Mg2+ in the absence of Ca2+, mimicking muscle cytosol during relaxation. In the presence of initially-bound Mg2+, SERCA site I′ maintained Mg2+ binding during the entire MD trajectory, and the cytosolic headpiece maintained a semi-open structure. In the absence of initially-bound Mg2+, two K+ ions rapidly bound to sites I and I′ and stayed loosely bound during most of the simulation, while the cytosolic headpiece shifted gradually to a more open structure. Thus MD simulations predict that both E1•Mg2+ and E•2K+ intermediate states of SERCA are populated in solution in the absence of Ca2+, with the more open 2K+-bound state being more abundant at physiological ion concentrations. We propose that the E1•2K+ state acts as a functional intermediate that facilitates the E2 to E1•2Ca2+ transition through two mechanisms: by pre-organizing transport sites for Ca2+ binding, and by partially opening the cytosolic headpiece prior to Ca2+ activation of nucleotide binding.  相似文献

Programmed cell death (PCD) is an integrated cellular process occurring in plant growth, development, and defense responses to facilitate normal growth and development and better survival against various stresses as a whole. As universal toxic chemicals in plant and animal cells, reactive oxygen or nitrogen species (ROS or RNS), mainly superoxide anion (O2−•), hydrogen peroxide (H2O2) or nitric oxide (NO), have been studied extensively for their roles in PCD induction. Physiological and genetic studies have convincingly shown their essential roles. However, the details and mechanisms by which ROS and NO interplay and induce PCD are not well understood. Our recent study on Cupressus lusitanica culture cell death revealed the elicitor-induced co-accumulation of ROS and NO and interactions between NO and H2O2 or O2- in different ways to regulate cell death. NO and H2O2 reciprocally enhanced the production of each other whereas NO and O2−• showed reciprocal suppression on each other''s production. It was the interaction between NO and O2- but not between NO and H2O2 that induced PCD, probably through peroxynitrite (ONOO). Four selected NaCl-tolerant cell lines of Sour orange (Citrus aurantium) were compared with the nonselected cell line in their growth and internal ion content of Na+, K+, and Cl when exposed to increasing NaCl concentrations. No difference was found among the various NaCl-tolerant cell lines in Na+ and Cl uptake, and all these cell lines took up similar or even larger amounts of Na+ and Cl than the NaCl-sensitive cell line. Exposure of cells of NaCl-sensitive and NaCl-tolerant lines to equal external concentrations of NaCl, resulted in a greater loss of K+ from the NaCl-sensitive cell line. This observation leads to the conclusion that growth and ability to retain high levels of internal K+ are correlated. Exposure of the NaCl-tolerant cell lines to salts other than NaCl resulted in even greater tolerance to Na2SO4, but rather poor tolerance to K+ introduced as either K2SO4 or KCl; the latter has a stronger inhibitory effect. The NaCl-sensitive cell line proved to be more sensitive to replacement of Na+ by K+. Analyses of internal Na+, K+, and Cl concentrations failed to identify any particular internal ion concentration which could serve as a reliable marker for salt tolerance.  相似文献

Stress conditions cause increases in ROS and polyamines levels, which are not merely collateral. There is increasing evidence for the ROS participation in signaling as well as for polyamine protective roles under stress. Polyamines and ROS, respectively, inhibit cation channels and induce novel cation conductance in the plasma membrane. Our new results indicate that polyamines and OH also stimulate Ca2+ pumping across the root plasma membrane. Besides, polyamines potentiate the OH-induced non-selective current and respective passive K+ and Ca2+ fluxes. In roots this synergism, however, is restricted to the mature zone, whereas in the distal elongation zone only the Ca2+ pump activation is observed. Remodeling the plasma membrane ion conductance by OH and polyamines would impact K+ homeostasis and Ca2+ signaling under stress.  相似文献

Debaryomyces nepalensis NCYC 3413, a food spoiling yeast isolated from rotten apple, has been previously demonstrated as halotolerant yeast. In the present study, we assessed its growth, change in cell size, and measured the intracellular polyol and cations (Na+ or K+) accumulated during growth in the absence and presence of different concentrations of salts (NaCl and KCl). Cells could tolerate 2 M NaCl and KCl in defined medium. Scanning electron microscopic results showed linear decrease in mean cell diameter with increase in medium salinity. Cells accumulated high amounts of K+ during growth at high concentrations of KCl. However, it accumulated low amounts of Na+ and high amounts of K+ when grown in the presence of NaCl. Cells grown in the absence of salt showed rapid influx of Na+/K+ on incubation with high salt. On incubation with 2 M KCl, cells grown at 2 M NaCl showed an immediate efflux of Na+ and rapid uptake of K+ and vice versa. To withstand the salt stress, osmotic adjustment of intracellular cation was accompanied by intracellular accumulation of polyol (glycerol, arabitol, and sorbitol). Based on our result, we hypothesize that there exists a balanced efflux and synthesis of osmolytes when D. nepalensis was exposed to hypoosmotic and hyperosmotic stress conditions, respectively. Our findings suggest that D. nepalensis is an Na+ excluder yeast and it has an efficient transport system for sodium extrusion.  相似文献

Summary The patch-clamp technique in whole-cell configuration was used to study the electrical properties of the tonoplast in isolated vacuoles fromAcer pseudoplatanus cultured cells. In symmetrical KCl or K2 malate solutions, voltage- and time-dependent inward currents were elicited by hyperpolarizing the tonoplast (inside negative), while in the positive range of potential the conductance was very small. The specific conductance of the tonoplast at –100 mV, in 100mm symmetrical KCl was about 160 S/cm2. The reversal potentials (E rev) of the current, measured in symmetrical or asymmetrical ion concentrations (cation, anion or both) were very close to the values of the K+ equilibrium potential. Experiments performed in symmetrical or asymmetrical NaCl indicate that Na+ too can flow through the channels. NeitherE rev nor amplitude and kinetics of the current changed by replacing NaCl with KCl in the external solution. These results indicate the presence of hyperpolarization-activated channels in tonoplasts, which are permeable to K+ as well as to Na+. Anions such as Cl or malate seem to contribute little to the channel current.  相似文献
