The reductase component (MhpP) of the Sulfobacillus acidophilus TPY multicomponent phenol hydroxylase exhibits only 40 % similarity to Pseudomonas sp. strain CF600 phenol hydroxylase reductase. Amino acid sequence alignment analysis revealed that four cysteine residues (Cys-X 4 -Cys-X 2 -Cys-X 29-35 -Cys) are conserved in the N terminus of MhpP for [2Fe-2S] cluster binding, and two other motifs (RXYS and GXXS/T) are conserved in the C terminus for binding the isoalloxazine and phosphate groups of flavin adenine dinucleotide (FAD). Two motifs (S/T-R and yXCGp) responsible for binding to reduce nicotinamide adenine dinucleotide phosphate (NADPH) are also conserved in MhpP, although some residues differ. To confirm the function of this reductase, MhpP was heterologously expressed in Escherichia coli BL21(DE3) and purified. UV-visible spectroscopy and electron paramagnetic resonance spectroscopy revealed that MhpP contains a [2Fe-2S] cluster. MhpP mutants in which the four cysteine residues were substituted via site-directed mutagenesis lost the ability to bind the [2Fe-2S] cluster, resulting in a decrease in enzyme-specific oxidation of NADPH. Thin-layer chromatography revealed that MhpP contains FAD. Substrate specificity analyses confirmed that MhpP uses NADPH rather than NADH as an electron donor. MhpP oxidizes NADPH using cytochrome c, potassium ferricyanide, or nitro blue tetrazolium as an electron acceptor, with a specific activity of 1.7 ± 0.36, 0.78 ± 0.13, and 0.16 ± 0.06 U/mg, respectively. Thus, S. acidophilus TPY MhpP is a novel NADPH-dependent reductase component of phenol hydroxylase that utilizes FAD and a [2Fe-2S] cluster as cofactors.  相似文献

A novel metalloprotein containing a unique [S2MoS2CuS2MoS2]3? cluster, designated as Orange Protein (ORP), was isolated for the first time from Desulfovibrio gigas, a sulphate reducer. The orp operon is conserved in almost all sequenced Desulfovibrio genomes and in other anaerobic bacteria, however, so far D. gigas ORP had been the only ORP characterized in the literature. In this work, the purification of another ORP isolated form Desulfovibrio alaskensis G20 is reported. The native protein is monomeric (12443.8 ± 0.1 Da by ESI–MS) and contains also a MoCu cluster with characteristic absorption bands at 337 and 480 nm, assigned to S–Mo charge transfer bands. Desulfovibrio alaskensis G20 recombinant protein was obtained in the apo-form from E. coli. Cluster reconstitution studies and UV–visible titrations with tetrathiomolybdate of the apo-ORP incubated with Cu ions indicate that the cluster is incorporated in a protein metal-assisted synthetic mode and the protein favors the 2Mo:1Cu stoichiometry. In Desulfovibrio alaskensis G20, the orp genes are encoded by a polycistronic unit composed of six genes whereas in Desulfovibrio vulgaris Hildenborough the same genes are organized into two divergent operons, although the composition in genes is similar. The gene expression of ORP (Dde_3198) increased 6.6 ± 0.5 times when molybdate was added to the growth medium but was not affected by Cu(II) addition, suggesting an involvement in molybdenum metabolism directly or indirectly in these anaerobic bacteria.  相似文献

Impact of different levels of elevated CO 2 on the activity of Frankia (Nitrogen-fixing actinomycete) in Casuarina equisetifolia rooted stem cuttings has been studied to understand the relationship between C. equisetifolia, Frankia and CO2. The stem cuttings of C. equietifolia were collected and treated with 2000 ppm of Indole Butyric Acid (IBA) for rooting. Thus vegetative propagated rooted stem cuttings of C. equisetifolia were inoculated with Frankia and placed in the Open top chambers (OTC) with elevated CO2 facilities. These planting stocks were maintained in the OTC for 12 months under different levels of elevated CO2 (ambient control, 600 ppm, 900 ppm). After 12 months, the nodule numbers, bio mass, growth, and photosynthesis of C. equisetifolia rooted stem cuttings inoculated with Frankia were improved under 600 ppm of CO2. The rooted stem cuttings of C. equisetifolia inoculated with Frankia showed a higher number of nodules under 900 ppm of CO2 and cuttings without Frankia inoculation exhibited poor growth. Tissue Nitrogen (N) content was also higher under 900 ppm of CO2 than ambient control and 600 ppm levels. The photosynthetic rate was higher (17.8 μ mol CO2 m?2 s?1) in 900 ppm of CO2 than in 600 ppm (13.2 μ mol CO2 m?2 s?1) and ambient control (8.3 μ mol CO2 m?2 s?1). This study showed that Frankia can improve growth, N fixation and photosynthesis of C. equietifolia rooted stem cuttings under extreme elevated CO2 level conditions (900 ppm).  相似文献

Objectives

To prepare (R)-phenyl-1,2-ethanediol ((R)-PED) with high enantiomeric excess (ee p) and yield from racemic styrene oxide (rac-SO) at high concentration by bi-enzymatic catalysis.

Results

The bi-enzymatic catalysis was designed for enantioconvergent hydrolysis of rac-SO by a pair of novel epoxide hydrolases (EHs), a Vigna radiata EH3 (VrEH3) and a variant (AuEH2A250I) of Aspergillus usamii EH2. The simultaneous addition mode of VrEH3 and AuEH2A250I, exhibiting the highest average turnover frequency (aTOF) of 0.12 g h?1 g?1, was selected, by which rac-SO (10 mM) was converted into (R)-PED with 92.6% ee p and 96.3% yield. Under the optimized reaction conditions: dry weight ratio 14:1 of VrEH3-expressing E. coli/vreh3 to AuEH2A250I-expressing E. coli/Aueh2 A250I and reaction at 20 °C, rac-SO (10 mM) was completely hydrolyzed in 2.3 h, affording (R)-PED with 98% ee p. At the weight ratio 0.8:1 of rac-SO to two mixed dry cells, (R)-PED with 97.4% ee p and 98.7% yield was produced from 200 mM (24 mg/ml) rac-SO in 10.5 h.

Conclusions

Enantioconvergent hydrolysis of rac-SO at high concentration catalyzed by both VrEH3 and AuEH2A250I is an effective method for preparing (R)-PED with high ee p and yield.

In this study, a novel engineering Escherichia coli strain (CBMG111) with the expression of mgtCB gene was constructed for the enhanced fermentative production of succinic acid by utilizing the synergetic effect of mgtC gene to improve the growth of strains at the environment of low Mg2+ concentration and mgtB to enhance the transport of Mg2+ into cells. After the effect of the expression of the individual genes (mgtA, mgtB, mgtC) on the growth of E. coli was clarified, the fermentative production of succinic acid by CBMG111 was studied with the low-price mixture of Mg(OH)2 and NH3·H2O as the alkaline neutralizer and the biomass hydrolysates as the carbon sources, which demonstrated that the expression of mgtCB gene can significantly increase the productivity of succinic acid (2.97 g L?1 h?1) compared with that by using the engineering strain with the overexpression of mgtA gene.  相似文献

This study evaluated the ability of a hydrothermal time model (HTT) to describe the kinetics of watermelon (Citrullus vulgaris cv. ‘Crimson sweet’) seed germination under different temperatures (T) and water potentials (ψ) and also to determine the cardinal temperatures of watermelon. Results indicated that ψ influenced germination rate and germination percentage. For this seed lot, cardinal temperatures were 10 °C for T b, 28.34 °C for T o and 40.8 °C for T c in the control (0 MPa) treatment. There was a decrease in hydrotime constant (θ H) when T was increased to T o and then remained constant at supra-optimal temperatures (30 MPah?1). Also, at temperatures above T o, ψ b(50) values increased linearly with T. The k T value (the slope of the relationship between ψ b(50) and T exceeds T o) of this seed lot was calculated as 0.076 MPa°Ch?1. Results this study show that when the HTT model is applied, it can accurately describe ψ b(g) and the course of germination around Ts (R 2 = 0.82). Moreover, the ψ b(50) was estimated to be ?0.96 MPa based on this model. Consequently, the germination response of watermelon for all Ts and ψs can be adequately described by the HTT model and enabling it to be used as a predictive tool in watermelon seed germination simulation models.  相似文献

The coccinellid beetle Anovia punica Gordon (Coleoptera: Coccinellidae: Noviini) is an important predator of the Colombian fluted scale, Crypticerya multicicatrices Kondo & Unruh (Hemiptera: Monophlebidae). In order to gather information on the biological traits of A. punica, we conducted a series of studies, including of the developmental time of each life history stage, estimation of life table parameters, and predation rates under laboratory conditions [25.1 ± 1.6°C, with 70.5 ± 7.3% RH, and natural light regime, approx. 12:12 (L:D) h]. Developmental stages of A. punica were categorized as follows: egg stage, four larval instars, prepupal instar, pupal instar, and adult. Developmental time from egg to adult emergence averaged 29.41 ± 1.85 days, and 47.6% of the eggs developed to adulthood. Female and male survival was 94.42 and 90 days, respectively. Life table parameters show that one female of A. punica is replaced by 86 females (R 0), the intrinsic growth rate (r m ) was 0.1115, the average generation time (T) was 40 days, and the doubling time (D t ) was 6.2 days. The life table parameters suggest that A. punica can be used as a potential predator of C. multicicatrices and, more importantly, provided baseline information for a mass-rearing protocol. This is the first detailed study on the biology of A. punica that reports the potential of this predator as a biological control agent for scale insects of the tribe Iceryini.  相似文献

The aims of the present study were to develop a parameterization of a one-year-long observed PAR time-series, apply the PAR parameterization in a primary production relation, and compare calculated and observed time-series of primary production. The PAR parameterization was applied in the generally used relation for the primary production (P d): P d = a(BI 0 Z 0) + b with observed photic depth (Z 0) and Chl-a concentrations (B). It was tested whether the PAR parameterization in combination with this simple relation for primary production was able to describe the actual measured primary production. The study is based on a one year long time-series of PAR, CTD-casts (n = 45), and primary production measurements (n = 24) from Århus Bay (56°09′ N; 10°20′ E), south west Kattegat. Results showed a high and positive correlation between observed and calculated primary production in the bay, as based on the present PAR parameterization combined with the simple primary production relation. The developed PAR parameterization, which calculates total daily surface irradiance per day (M photons m?2 d?1), can be applied in any ecological application taking into account that it was developed for the latitude of 56° N.  相似文献

Helicobacter pylori is an infectious agent commonly associated with gastrointestinal diseases. The use of probiotics to treat this infection has been documented, however, their potential antimicrobial metabolites have not yet been investigated. In the present study, the effect of reuterin produced by Lactobacillus reuteri on H. pylori growth and virulence gene expression was evaluated. It was observed that reuterin caused significant (P < 0.05) H. pylori growth inhibition at concentrations from 0.08 to 20.48 mM, with minimal inhibitory concentrations (MICs) of 20.48 mM for H. pylori ATCC700824 and 10.24 mM for H. pylori ATCC43504. In a reuterin bacterial killing assay, it was observed that half of the MIC value for H. pylori (ATCC700824) significantly (P < 0.01) reduced colony numbers from 5.65 ± 0.35 to 3.78 ± 0.35 Log10 CFU/mL after 12 h of treatment and then increased them to 5.25 ± 0.23 Log10 CFU/mL at 24 h; at its MIC value (20.48 mM), reuterin abrogated (P < 0.01) H. pylori (ATCC700824) growth after 20 h of culture. In addition, reuterin significantly (P < 0.01) reduced H. pylori (ATCC 43504) colony numbers from 5.65 ± 0.35 to 4.1 ± 0.12 Log10 CFU/mL from 12 to 24 h of treatment and abrogated its growth at its MIC value (10.24 mM), after 20 h of treatment. Reuterin did not alter normal human gastric Hs738.St/Int cell viability at the concentrations tested for H. pylori strains. Furthermore, 10 μM reuterin was shown to significantly (P < 0.01) reduce mRNA relative expression levels of H. pylori virulence genes vacA and flaA at 3 h post-treatment, whose effect was higher at 6 h post-treatment, as measured by RT-qPCR. The observed direct antimicrobial effect and the downregulation of expression of virulence genes on H. pylori by reuterin may contribute to the understanding of the mechanisms of action of probiotics against H. pylori.  相似文献

In this study, our working hypothesis was to examine whether temperature alters biomass and metabolite production by microalgae according to strain. We also addressed whether it is possible to choose a strain suitable for growing in each season of a given region. A factorial experiment revealed a significant interaction between chlorophylls a and b (Chl a and Chl b), carotenoid/Chl (a?+?b) ratio, biomass and total lipid productivity of six green microalgae (four Chlorella spp., Chlorella sorokiniana and Neochloris oleoabundans) after 15 days at four temperatures. At 39/35 °C, two Chlorella sp. strains (IPR7115 and IPR7117) showed higher total carotenoids/Chl (a?+?b) (0.578 and 0.830), respectively. N. oleoabundans had the highest Chl a (8210 μg L?1) and Chl b (1909 μg L?1) at 19/15 °C and highest maximum dry biomass (2900 mg L?1), specific growth rate (0.538 day?1) and total lipids (1003 mg L?1) at 15/8 °C. We applied a method to infer the growth of these six green microalgae in outdoor ponds, as based on their response to changing temperatures and by combining with historical data on day/night air temperature occurrence for a given region. We conclude that the use of regionalized maps based on air temperature is a good strategy for predicting microalgal cultivation in outdoor ponds based on their features and tolerance to changing temperature.  相似文献

A new strain of Chlorella sp. (Chlorella-Arc), isolated from Arctic glacier melt water, was found to have high specific growth rates (μ) between 3 and 27 °C, with a maximum specific growth rate of 0.85 day?1 at 15 °C, indicating that this strain was a eurythermal strain with a broad temperature tolerance range. To understand its acclimation strategies to low and high temperatures, the physiological and biochemical responses of the Chlorella-Arc to temperature were studied and compared with those of a temperate Chlorella pyrenoidosa strain (Chlorella-Temp). As indicated by declining F v/F m, photoinhibition occurred in Chlorella-Arc at low temperature. However, Chlorella-Arc reduced the size of the light-harvesting complex (LHC) to alleviate photoinhibition, as indicated by an increasing Chl a/b ratio with decreasing temperatures. Interestingly, Chlorella-Arc tended to secrete soluble sugar into the culture medium with increasing temperature, while its intracellular soluble sugar content did not vary with temperature changes, indicating that the algal cells might suffer from osmotic stress at high temperature, which could be adjusted by excretion of soluble sugar. Chlorella-Arc accumulated protein and lipids under lower temperatures (<15 °C), and its metabolism switched to synthesis of soluble sugar as temperatures rose. This reflects a flexible ability of Chlorella-Arc to regulate carbon and energy distribution when exposed to wide temperature shifts. More saturated fatty acids (SFA) in Chlorella-Arc than Chlorella-Temp also might serve as the energy source for growth in the cold and contribute to its cold tolerance.  相似文献

To quantify both temperature (T) and water potential (ψ) effects on sesame (Sesamum indicum L.) seed germination (SG) and also to determine the cardinal T s for this plant, a laboratory experiment was carried out using hydrothermal time model (HTT). For this purpose, four sesame cultivars (‘Asbomahalleh’, ‘Darab’, ‘Dashtestan’ and ‘Yellowhite’) were germinated at seven constant T s (20, 25, 30, 35, 37, 39 and 43 °C) at each of the following ψ s (0, ? 0.12, ? 0.24 and ? 0.36 MPa; provided by PEG 8000). Germination rate (GR) and germination percentage (GP) significantly influenced by ψ, T and their interactions in all cultivars (P ≤ 0.01). There was no significant difference, based on the confidence intervals of the model coefficients, between cultivars, so an average of cardinal T s was 14.7, 35.4 and 47.2 °C for the minimum (T b), optimum (T o) and maximum (T c) T s, respectively, in the control condition (0 MPa). Hydrotime values in all cultivars decreased when T was increased to T o and then remained constant at T s > T o (15 MPa h?1). An average value of ψ b(50) was estimated to be ? 1.23 MPa at T s ≤ T o and then increased linearly (0.1041 MPa°Ch?1, the slope of the relationship between ψ b(50) and supra-optimal T s) with T when T s increased above T o and finally reached to zero at T c. The T b and T o values were not influenced by ψ, but T c value decreased (from 47.2 for zero to 43.5 °C for ? 0.36 MPa) at supra-optimal T s as a result of the effect of ψ on GR. Based on our findings, this model (as a predictive tool) and or the estimated parameter values in this study can easily be used in sesame SG simulation models to quantitatively characterize the physiological status of sesame seed populations at different T s and ψ s.  相似文献

Life history strategies of batoid fishes have evolved within dynamic marine ecosystems. Adaptations in reproductive and developmental biology are paramount to the survival of species, and therefore knowledge of growth rates to maturity is fundamental for identifying constraints on the conservation of populations. The butterfly rays (Myliobatiformes: Gymnuridae) are highly derived batoids with generally low reproductive potentials for which age and growth information remains unknown. In this study we applied high-resolution X-ray computed tomography (HRXCT) to vertebral centra from a stingray for the first time to estimate age, and used a multimodel approach to investigate growth of spiny butterfly ray, Gymnura altavela. Estimated ages of the oldest male and female were 11 and 18 yrs. at disk widths (WD) 1355 mm and 2150 mm, respectively. Disk width-at-age data were analyzed using three growth models (von Bertalanffy, logistic, Gompertz), and the most parsimonious and empirically supported model was the logistic function with sex treated as a fixed effect on asymptotic disk width (WD ) and k parameters. Model parameter estimates were (males) WD  = 1285.46 ± 67.27 mm, k = 0.60 ± 0.10, and (females) WD  = 2173.51 ± 129.78 mm, k = 0.27 ± 0.04. Results indicated sexually dimorphic growth patterns, with males growing faster and reaching asymptotic size at earlier ages than females. These age and growth results are the first reported for the genus, and suggest that G. altavela grows at a similar rate as some teleosts and batoids, and relatively fast among chondrichthyans.  相似文献

We examined changes in suspended-sediment yields (SSY) after a 50 % strip thinning in headwater streams draining a Japanese cedar (Cryptomeria japonica) and cypress (Chamaecyparis obtusa) plantation forest. We applied a paired-catchment analysis to treated (K T : 17.1 ha) and control (K C : 8.9 ha) catchments. Annual suspended-sediment yield (SSYan) in the prethinning period in the K T and K C catchments was 110.0 and 142.1 kg/ha per year. For the postthinning period, SSYan in the K T catchment became 5055.6 kg/ha per year, whereas that in the K C catchment increased 893.2 kg/ha per year. The paired-catchment analysis revealed that SSY in the K T catchment increased 17.0-fold compared with the prethinning period. However, the dominant hysteresis pattern remained clockwise in both pre- and postthinning periods. Sequences of large storm events in the postthinning period elevated SSY in both catchments. Increase in suspended sediment in the K T catchment was associated with the combined impacts of thinning and sequences of storm events during the period of thinning operation.  相似文献

Fundulus grandis (Baird and Girard), the Gulf Killifish, is an abundant species throughout the marshes of the northern Gulf of Mexico. Its wide distribution and high site fidelity makes it an ideal indicator species for brackish and salt marshes, which experience a variety of anthropogenic disturbances. Despite the ecological, commercial, and scientific importance of F. grandis, age determination methods have not been validated and little is known of its growth pattern. By combining a tag-recapture study with a chemical marker to stain otoliths, we validated an ageing method for F. grandis adults (49–128 mm TL) using whole sagittal otoliths and determined growth rates of recaptured individuals in winter (n = 58) and summer (n = 36) in Louisiana. Mean somatic growth in length was significantly greater during the winter (0.085 mm d?1) than summer (0.054 mm d?1). In contrast, mean otolith growth was significantly greater in summer (1.37 μm d?1) than winter (0.826 μm d?1). The uncoupling of somatic and otolith growth may be primarily attributed to warm summer temperatures, which led to enhanced otolith growth while simultaneously reducing somatic growth. Fundulus grandis was aged to a maximum of 2.25 years. The parameters of the von Bertalanffy growth model were estimated as: L  = 87.27 mm, k = 2.43 year?1, and t 0 = ?0.022. These findings reveal essential age and growth information for F. grandis and provide a benchmark to evaluate responses to environmental disturbances.  相似文献

This paper identifies the potential molecular markers predicting the impact of nTiO2 on plants and explores the new statistical correlations between the biomarkers and growth parameters. The quantitative mRNA expression of the three genes involved in DNA mismatch repair (MLH1) and cell division (PCNA1 and PCNA2) in Zea mays and Triticum aestivum seedlings were related to the growth parameters measured in response to five nTiO2 treatments. The results indicated that the higher concentrations were harmless to Z. mays but not to T. aestivum. nTiO2 treatments increased the total protein levels in both species and significantly inhibited the percentage of DPPH radical scavenging in Z. mays compared with T. aestivum seedlings. The exposure to both 50 μg/ml and 30 μg/ml concentrations of nTiO2 significantly induced the expression of MLH1 and PCNA1 genes in both species; however, the exposure to 30 μg/ml of nTiO2 also significantly induced the expression of PCNA2 genes in T. aestivum. The exposure to 50, 70 and 140 μg/ml significantly inhibited the expression of PCNA2 in both species, while 70 and 140 μg/ml repressed the expression of MLH1 and PCNA1 in the seedlings of Z. mays. The induction and repression of the expression of the three genes were correlated with some growth parameters and biological indices in both species. This key finding suggests that the above genes may play a vital role in mediating plant stress response to nTiO2 and could be used as sensitive molecular biomarkers indicative of the oxidative stress of nTiO2 exposure.  相似文献

Insertion/deletion (InDel) markers are valuable for genetic applications in plant species, and the public databases of expressed sequence tags (ESTs) have facilitated the development of genic InDel markers. In this study, we developed a novel set of 144 InDel markers in an important tree genus Eucalyptus L’Hérit. using the ESTs of GenBank. Amplicon sequencing against two parents of a mapping population (Eucalyptus urophylla S. T. Blake × E. tereticornis Smith) revealed that the InDel size ranged from 2 to 44 bases, and the dinucleotide type was the most abundant (37.3 %). The cross-species/subgenus amplification rate ranged from 62.5 % in E. tessellaris F. Muell. (subgenus Blakella) to 99.3 % in E. grandis Hill ex Maiden (subgenus Symphyomyrtus) with an average of 85.4 %. There were 121 EST-InDels (84.0 %) polymorphic among 12 individuals of E. grandis, and the mean number of alleles per polymorphic locus (N a), observed heterozygosity (H o), expected heterozygosity (H e) and polymorphic information content (PIC) were 4.0, 0.278, 0.538 and 0.465, respectively. Physical positions of 143 EST-InDels were predicted on the E. grandis genome sequence. A total of 81 EST-InDels were incorporated into prior dense genetic maps of E. urophylla and E. tereticonis, and extensive synteny and colinearity were observed between E. grandis genome sequence and the mapped EST-InDel markers. These EST-InDels will provide a valuable resource of functional markers for genetic diversity evaluation, genome comparison, QTL mapping and marker-assisted breeding in Eucalyptus.  相似文献

The thermal sensitivities of organisms regulate a wide range of ecological interactions, including host–parasite dynamics. The effect of temperature on disease ecology can be remarkably complex in disease systems where the hosts are ectothermic and where thermal conditions constrain pathogen reproductive rates. Amphibian chytridiomycosis, caused by the pathogen Batrachochytrium dendrobatidis (Bd), is a lethal fungal disease that is influenced by temperature. However, recent temperature studies have produced contradictory findings, suggesting that our current understanding of thermal effects on Bd may be incomplete. We investigated how temperature affects three different Bd strains to evaluate diversity in thermal responses. We quantified growth across the entire thermal range of Bd, and beyond the known thermal limits (T max and T min). Our results show that all Bd strains remained viable and grew following 24 h freeze (?12 °C) and heat shock (28 °C) treatments. Additionally, we found that two Bd strains had higher logistic growth rates (r) and carrying capacities (K) at the upper and lower extremities of the temperature range, and especially in low temperature conditions (2–3 °C). In contrast, a third strain exhibited relatively lower growth rates and carrying capacities at these same thermal extremes. Overall, our results suggest that there is considerable variation among Bd strains in thermal tolerance, and they establish a new thermal sensitivity profile for Bd. More generally, our findings point toward important questions concerning the mechanisms that dictate fungal thermal tolerances and temperature-dependent pathogenesis in other fungal disease systems.  相似文献

Nitrogen (N) is the key factor limiting photosynthetic processes and crop yield. Little is known about the response of leaf gas exchange of spring triticale (Triticosecale Wittm.) to N supply. The effect of N fertilizers on different gas exchange variables, i.e., photosynthetic rate (A), transpiration rate (E), stomatal conductance (g s), instantaneous water use efficiency (WUE) and maximum quantum yield of photosystem II (PSII) (F v/F m), chlorophyll index (SPAD, soil–plant analysis development), and the relationship of these variables with yield were studied in spring triticale grown under field conditions. Six treatments of N—0, 90, 180, 90 + 30, 90 + 30 + 30 kg ha?1 (applied as ammonium nitrate, AN) and one treatment of N 90 + 30 + 30 kg ha?1 (applied as urea ammonium nitrate solution, UAN) were compared. The analysis of variance showed that throughout the triticale growing season, N fertilization had significant effects on A, WUE, g s and SPAD. On average, N fertilizer application increased A values by 14–70%. E and F v/F m values were not influenced by N fertilization levels. The effect of growth stage and year on gas exchange variables and F v/F m and SPAD was found to be significant. At different growth stages, A values varied and maximum ones were reached at BBCH 31–33 (decimal code system of growth stages) and BBCH 59. With aging, values of A decreased independently of N fertilization level. The gas exchange variables were equally affected by both fertilizer forms. The interplay among grain yield, leaf gas exchange variables, F v/F m and SPAD of spring triticale was estimated. The statistical analysis showed that grain yield positively and significantly correlated with A and SPAD values throughout the growing season.  相似文献

The biodegradation of furfuryl alcohol (FA) in shake flask experiments using a pure culture of Pseudomonas putida (MTCC 1194) and Pseudomonas aeruginosa (MTCC 1034) was studied at 30 °C and pH 7.0. Experiments were performed at different FA concentrations ranging from 50 to 500 mg/l. Before carrying out the biodegradation studies, the bacterial strains were acclimatized to the concentration of 500 mg/l of FA by gradually raising 100 mg/l of FA in each step. The well acclimatized culture of P. putida and P. aeruginosa degraded about 80 and 66% of 50 mg/l FA, respectively. At higher concentration of FA, the percentage of FA degradation decreased. The purpose of this study was to determine the kinetics of biodegradation of FA by measuring biomass growth rates and concentration of FA as a function of time. Substrate inhibition was calculated from experimental growth parameters using the Haldane equation. Data for P. putida were determined as µ max ?=?0.23 h?1, K s ?=?23.93 mg/l and K i ?=?217.1 mg/l and for P. aeruginosa were determined as µ max ?=?0.13 h?1, K s ?=?21.3 mg/l and K i ?=?284.9 mg/l.
