In the last few years, continuous progress in instrumental analytical methodology has been achieved with a substantial increase in the number of new, more specific and more flexible methods for ligand-protein assays. In general, the methods used for drug-protein binding studies can be divided into two main groups: separation methods (enabling the calculation of binding parameters, i.e. the number of binding sites and their respective affinity constants) and non-separation methods (describing predominantly qualitative parameters of the ligand-protein complex). This review will be focussed particularly on recent trends in the development of drug-protein binding methods including stereoselective and non-stereoselective aspects using chromatography, capillary electrophoresis and microdialysis as compared to the “conventional approach” using equilibrium dialysis, ultrafiltration or size exclusion chromatography. The advantages and limitations of various methods will be discussed including a focus on “optimal” experimental strategies taking into account in vitro, ex vivo and/or in vivo studies. Furthermore, the importance of some particular aspects concerning the drug binding to proteins (covalent binding of drugs and their metabolites, stereoselective interactions and evaluation of binding data) will be outlined in more detail.  相似文献

The nature of protein folding pathways: The classical versus the new view   总被引:12,自引:0,他引:12  
Summary Pulsed hydrogen exchange and other studies of the kinetic refolding pathways of several small proteins have established that folding intermediates with native-like secondary structures are well populated, but these studies have also shown that the folding kinetics are not well synchronized. Older studies of the kinetics of formation of the native protein, monitored by optical probes, indicate that the folding kinetics should be synchronized. The model commonly used in these studies is the simple sequential model, which postulates a unique folding pathway with defined and sequential intermediates. Theories of the folding process and Monte Carlo simulations of folding suggest that neither the folding pathway nor the set of folding intermediates is unique, and that folding intermediates accumulate because of kinetic traps caused by partial misfolding. Recent experiments with cytochrome c lend support to this new view of folding pathways. These different views of the folding process are discussed. Misfolding and consequent slowing down of the folding process as a result of cis-trans isomerization about prolyl peptide bonds in the unfolded protein are well known; isomerization occurs before refolding is initiated. The occurrence of equilibrium intermediates on the kinetic folding pathways of some proteins, such as -lactalbumin and apomyoglobin, argues that these intermediates are not caused by kinetic traps but rather are stable intermediates under certain conditions, and this conclusion is consistent with a sequential model of folding. Folding reactions with successive kinetic intermediates, in which late intermediates are more highly folded than early intermediates, indicate that folding is hierarchical. New experiments that test the predictions of the classical and the new views are needed.  相似文献

Red cell morphology in alcoholics: a new test for alcohol abuse   总被引:1,自引:0,他引:1  
Scanning electron microscopy has shown that the blood of alcoholics contains a large number of morphologically abnormal red cells. In two groups of alcoholics, the number of morphologically abnormal red cells ranged from 23.1% to 89.3% and 27.4% to 57.3% of total red cells compared to values in healthy controls of 4.5%-12.6% and 27.7%-79.5% in nonalcoholic liver disease patients. A characteristic finding was the presence of triangulocytes: these ranged from 1.2% to 18.0% of total red cells in the alcoholics as compared to 0-0.5% in healthy controls, and 0-1.3% in patients with nonalcoholic liver disease. The presence of elevated numbers of triangulocytes in blood appears to be specific to alcohol abuse. It is not, for example, elevated in nonalcoholic liver disease. No correlation was found between the number of triangulocytes or the number of morphologically abnormal red cells in blood and either the duration of alcohol abuse or the amount of alcohol consumed. Both parameters tended, however, to return to normal values during withdrawal. The mechanism by which alcohol abuse causes the morphologic abnormalities is not known. Preliminary in vitro experiments indicate that it is unlikely to arise as an effect of alcohol on circulating red cells. Based on the data presented, the measurement of the number of triangulocytes in a blood sample, although slow and laborious, may provide a highly specific test for alcohol abuse.  相似文献

We present a new formulation of the steady state, isothermal, nonlinear reaction-diffusion problem involving nth order reaction kinetics for slab geometry. This results in tractable expressions for the effectiveness factor as a function of the Thiele modulus, the Thiele modulus as a function of the centerline concentration, and the concentration profiles in the slab. The expressions are valid asymptotically in the limit of large orders n. We compare these results with the exact numerical solutions obtained by transforming the nonlinear differential equation into an integral form, using Green's function methods, and solving by successive approximations. The formulation for a membrane is also given, and the nature of the asymmetrical solution discussed. The analysis is facilitated through the introduction of pseudo-reaction orders. A comparison of the asymptotic Thiele modulus obtained herein with a previously given expression shows the present theory to be an improvement.  相似文献

The International Union for the Conservation of Nature (IUCN) Red List of ecosystems and Red List of threatened species are global standards for assessing risks of ecosystem collapse and species extinction. However, misconceptions of the Red List assessment process, along with its technically demanding nature, can result in the misapplication of their criteria, leading to inconsistent and potentially unreliable assessments. To address this problem, we developed redlistr, an R package aiding in the production of consistent species and ecosystem Red List assessments. Redlistr's features include methods to calculate 1) area from spatial data, 2) range size metrics, 3) rates of change of distributions or populations, and 4) distribution or population at another time from these rates. A key feature of the package is the systematic approach used to eliminate geometric uncertainty when estimating area of occupancy. Here, we develop two case studies to demonstrate the functionalities of redlistr with typical workflows for both species and ecosystems. Redlistr was developed to be accessible to users with a broad range of experience in programming for spatial and temporal data analysis, and sufficiently flexible to allow users to parameterise functions and select equations to fit their purposes. The package specifically aims to assist researchers and conservation practitioners to conduct robust and transparent risk assessments of ecosystems and species under the IUCN Red List criteria but is also useful for other studies requiring analyses of range size, area change and calculations of rates of change.  相似文献

Mass spectrometric methods for the quantitative and qualitative analyses of algal biotoxins are often complicated by co-eluting compounds that present analytically as interferences. This issue is particularly critical for organic polyamines, where co-eluting materials can suppress the formation of cations during electrospray ionization. Here we present an extraction procedure designed specifically to overcome matrix-derived ion suppression of algal toxins in samples of Lyngbya wollei, a filamentous benthic algae known to produce several saxitoxin analogues. Lyngbya wollei samples were collected from a large, persistent harmful algal bloom in Lake Wateree, SC. Six known Lyngbya wollei-specific toxins (LWT1–6) were successfully resolved and quantified against saxitoxin using hydrophilic interaction liquid chromatography coupled with triple quadrupole and quadrupole time-of-flight mass spectrometry. The parent ions [M2+ – H+]+ were observed for LWTs 1–6 and the [M]2+ ion was observed for LWT5. High resolution mass spectra and unique fragmentation ions were obtained for LWTs 1–6. A dilution factor of 50 resulted in a linear calibration of saxitoxin in the algae matrix. Ion suppression was resolved by sample dilution, which led to linear, positive correlations between peak area and mass of the extracted sample (R2 > 0.96). Optimized sample extraction method and instrument parameters are presented.  相似文献

A substantial and increasing number of reports have documented dramatic changes and continuing declines in Caribbean coral reef communities over the past 2 decades. To date, the majority of disease reports have focused on scleractinian corals, whereas sponge diseases have been less frequently documented. In this study, we describe Aplysina red band syndrome (ARBS) affecting Caribbean rope sponges of the genus Aplysina observed on shallow reefs in the Bahamas. Visible signs of disease presence included 1 or more rust-colored leading edges, with or without a trailing area of necrotic tissue, such that the lesion forms a contiguous band around part or all of the sponge branch. Microscopic examination of the leading edge of the disease margin indicated that a cyanobacterium was consistently responsible for the coloration. Although the presence of this distinctive coloration was used to characterize the diseased state, it is not yet known whether this cyanobacterium is directly responsible for disease causation. The prevalence of ARBS declined significantly from July to October 2004 before increasing above July levels in January 2005. Transmission studies in the laboratory demonstrated that contact with the leading edge of an active lesion was sufficient to spread ARBS to a previously healthy sponge, suggesting that the etiologic agent, currently undescribed, is contagious. Studies to elucidate the etiologic agent of ARBS are ongoing. Sponges are an essential component of coral reef communities and emerging sponge diseases clearly have the potential to impact benthic community structure on coral reefs.  相似文献

15 lipoxygenase (15LO) is a lipid-oxidizing enzyme that is considered to contribute to the formation of oxidized lipids in atherosclerotic lesions. Monocyte-macrophages are the key cells that express 15LO in atherosclerotic lesions. In this review, we examine the evidence for 15LO involvement in atherogenesis and explore and evaluate the potential mechanisms whereby 15LO may contribute to the oxidation of LDL by monocyte-macrophages. We also describe several possible pro- versus anti-atherogenic functions that may be mediated by various products of 15LO lipid oxidation. Central pathways involved in regulating 15LO expression and activity that may serve as future targets for intervention and regulation of this enzyme are also reviewed.  相似文献

The compilation of the Red Lists of butterflies in Flanders and the Netherlands was based on two criteria: a trend criterion (degree of decline) and a rarity criterion (actual distribution area). However, due to the large difference in mapping intensity in the two compared periods, a straightforward comparison of the number of grid cells in which each species was recorded, appeared inappropriate. To correct for mapping intensity we used reference species that are homogeneously distributed over the country, that have always been fairly common and that did not fluctuate in abundance too much during this century. For all resident species a relative presence in two compared periods was calculated, using the average number of grid cells in which these reference species were recorded as a correction factor. The use of a standardized method and well-defined quantitative criteria makes national Red Lists more objective and easier to re-evaluate in the future and facilitates the comparison of Red Lists among countries and among different organisms. The technique applied to correct for mapping intensity could be useful to other organisms when there is a large difference in mapping intensity between two periods.  相似文献

More than 12 % of bird species are threatened with extinction. Numerous anthropogenic activities and processes are considered responsible for such declines, including tourism related activities. These activities often occur in global biodiversity hotspots but few studies consider the potential risks associated with tourism. The relative importance of tourism as a threat to birds was quantified using a global analysis of the threats facing critically endangered and endangered birds in the hotspots. Sixty-three critically endangered and endangered bird species are reportedly threatened by tourism. Among those 63 species, marine, coastal and aquatic birds are threatened more by tourism than was expected. Hotspots with the most species threatened by tourism are Polynesia–Micronesia and the Mediterranean Basin. This study uses individual threatening processes in a new way to characterise hotspots for conservation action, advancing previous identification criteria. Analysing hotspots in terms of the relative presence of individual threatening processes may help to more effectively direct future research in these priority regions.  相似文献

A new method for the preperative and analytical electrophoresis of cells   总被引:1,自引:0,他引:1  
In this paper, a new method is described for the horizontal electrophoresis of cells on a density cushion under near-isopycnic conditions. When cell sedimentation is minimized, the electrophoresis of red blood cells (RBC) used as model cells within an anti-convective porous matrix (with pores over 300 μm in diameter) was capable of separating a mixture of human and chicken RBC according to their electrophoretic mobilities. Samples taken from the separated RBC bands show over 90% purity for each species. The simultaneous electrophoresis of several RBC samples carried out under identical conditions permitted the use of comparative data based on the electrophoretic mobility of cells which differ in their surface properties. We believe that this relatively simple system, in which cell sedimentation and convection are minimized, has the potential to be modified and adapted for the separation of other cell types/organelles.  相似文献

目的 建立一种新的糠秕马拉色菌药敏试验方法.方法 在ATBF2半固体培养基中添加不同种类和含量的脂质,用ATB Fungus 3药敏板条对ATCC14521和临床分离的糠秕马拉色菌进行了MIC测试.结果 孵育时间为72 h时对照孔中糠秕马拉色菌生长充分,吐温40浓度为1%时糠秕马拉色菌生长充分,且对药敏结果的影响最小.脂质的种类和含量对实验结果有影响.结论 用改良ATB Fungus 3药敏试验方法对糠秕马拉色菌进行抗真菌药敏试验,方法操作简便、结果易观察、试验结果重复性好.临床分离菌株与糠秕马拉色菌ATCC14521在ATB Fungus 3对照孔中的生长状况相同,结果易于判断.  相似文献

The question if ant behaviour and biological limitations should be considered before generalisations about the ant’s defensive capabilities in ant–plant relationships was explored through a new experimental manipulation. In the Brazilian tropical savanna, we tested the protective action of Cephalotes pusillus Klug on the extrafloral nectar-bearing plant Ouratea spectabilis Engl. (Ochnaceae). Three treatments were performed: control (free ant access), Cephalotes-treatment (access permitted only to C. pusillus), and ant free treatment (no ants). No difference was found in the levels of leaf herbivory among experimental stems. Visitation by different ant species to control stems translated into significantly greater fruit and seed production by this stem category than by ant-free and Cephalotes-treated stems. Thus, results showed that an investigation of system’s natural history, ant’s morphological traits, defensive capabilities and behaviour are needed before a protective role is inferred to each associated ant species.  相似文献

The potential effects of 'escape' of genetically modified material (transgenes) into natural communities is a major concern in their use. These effects may be limited in the first instance by limiting the proportion of transgene-carrying plants in the natural community. We previously presented an analytical model of the ecological processes governing the relative abundance and persistence of insect resistance (IR) transgenes in a natural community. In that paper, we illustrated the case in which the transgene is input into the community in a single season using data from oilseed rape (OSR) and its known herbivore, Plutella macropennis. We found that the transgene is unlikely to have a great impact on the natural community. Here, we extend the model for repeated input of crop pollen carrying the transgene. We show the model output, again using OSR, for continuous input of the transgene over 10 years, the projected commercial lifetime of a transgene without associated undesirable agronomic effects. Our results do not change our original conclusion that the IR transgene need not have a large impact on the natural community and our suggestions for assessing and mitigating any threat still stand.  相似文献
