The effects of three metabolic inhibitors (acetylene, methanol, and allylthiourea [ATU]) on the pathways of N2 production were investigated by using short anoxic incubations of marine sediment with a 15N isotope technique. Acetylene inhibited ammonium oxidation through the anammox pathway as the oxidation rate decreased exponentially with increasing acetylene concentration; the rate decay constant was 0.10+/-0.02 microM-1, and there was 95% inhibition at approximately 30 microM. Nitrous oxide reduction, the final step of denitrification, was not sensitive to acetylene concentrations below 10 microM. However, nitrous oxide reduction was inhibited by higher concentrations, and the sensitivity was approximately one-half the sensitivity of anammox (decay constant, 0.049+/-0.004 microM-1; 95% inhibition at approximately 70 microM). Methanol specifically inhibited anammox with a decay constant of 0.79+/-0.12 mM-1, and thus 3 to 4 mM methanol was required for nearly complete inhibition. This level of methanol stimulated denitrification by approximately 50%. ATU did not have marked effects on the rates of anammox and denitrification. The profile of inhibitor effects on anammox agreed with the results of studies of the process in wastewater bioreactors, which confirmed the similarity between the anammox bacteria in bioreactors and natural environments. Acetylene and methanol can be used to separate anammox and denitrification, but the effects of these compounds on nitrification limits their use in studies of these processes in systems where nitrification is an important source of nitrate. The observed differential effects of acetylene and methanol on anammox and denitrification support our current understanding of the two main pathways of N2 production in marine sediments and the use of 15N isotope methods for their quantification.  相似文献

Quantification of microbial contaminant biodegradation based on stable isotope fractionation analysis (SIFA) relies on known, invariable isotope fractionation factors. The microbially induced isotope fractionation is caused by the preferential cleavage of bonds containing light rather than heavy isotopes. However, a number of non-isotopically sensitive steps preceding the isotopically sensitive bond cleavage may affect the reaction kinetics of a degradation process and reduce the observed (i.e., the macroscopically detectable) isotope fractionation. This introduces uncertainty to the use of isotope fractionation for the quantification of microbial degradation processes. Here, we report on the influence of bacterial cell density on observed stable isotope fractionation. Batch biodegradation experiments were performed under non-growth conditions to quantify the toluene hydrogen isotope fractionation by exposing Pseudomonas putida mt-2(pWWO) at varying cell densities to different concentrations of toluene. Observed isotope fractionation depended significantly on the cell density. When the cell density rose from 5 × 105 to 5 × 108cells/mL, the observed isotope fractionation declined by 70% and went along with a 55% decrease of the degradation rates of individual cells. Theoretical estimates showed that uptake-driven diffusion to individual cells depended on cell density via the overlap of the cells’ diffusion-controlled boundary layers. Our data suggest that biomass effects on SIFA have to be considered even in well-mixed systems such as the cell suspensions used in this study.  相似文献

Removal of biologically available nitrogen (N) by the microbially mediated processes denitrification and anaerobic ammonium oxidation (anammox) affects ecosystem N availability. Although few studies have examined temperature responses of denitrification and anammox, previous work suggests that denitrification could become more important than anammox in response to climate warming. To test this hypothesis, we determined whether temperature responses of denitrification and anammox differed in shelf and estuarine sediments from coastal Rhode Island over a seasonal cycle. The influence of temperature and organic C availability was further assessed in a 12‐week laboratory microcosm experiment. Temperature responses, as characterized by thermal optima (Topt) and apparent activation energy (Ea), were determined by measuring potential rates of denitrification and anammox at 31 discrete temperatures ranging from 3 to 59 °C. With a few exceptions, Topt and Ea of denitrification and anammox did not differ in Rhode Island sediments over the seasonal cycle. In microcosm sediments, Ea was somewhat lower for anammox compared to denitrification across all treatments. However, Topt did not differ between processes, and neither Ea nor Topt changed with warming or carbon addition. Thus, the two processes behaved similarly in terms of temperature responses, and these responses were not influenced by warming. This led us to reject the hypothesis that anammox is more cold‐adapted than denitrification in our study system. Overall, our study suggests that temperature responses of both processes can be accurately modeled for temperate regions in the future using a single set of parameters, which are likely not to change over the next century as a result of predicted climate warming. We further conclude that climate warming will not directly alter the partitioning of N flow through anammox and denitrification.  相似文献

Agricultural activities lead excessive emission of ammonia nitrogen in the environment and can profoundly interfere the equilibrium of the natural ecosystems leading to their contamination. Actually, the biological purification of wastewaters is the most adopted technique thanks to its several advantages such as high performance and low energy consumption. For this reason, two novel strains of Alcaligenes sp. S84S3 and Proteus sp. S19 genus were isolated from an activated sludge and applied in the treatment of ammonium and nitrite in aqueous solution. Under the optimum operating conditions of temperature (30 °C), pH (7), carbon substrate (2 g/L of glucose) and duration of incubation time (69 h), the strain Alcaligenes sp. S84S3 could oxidize 65 % of the ammonium as high as 272.72 mg-NH4 +/L. Moreover, during 48 h, the nitrate reduction rate performed by the strain Proteus S19 was about 99 % without production of nitrite intermediate (negligible concentration). Moreover, the coculture of the strains Alcaligenes sp. S84S3 and Proteus sp. S19 could eliminate 65.83 % of the ammonium ions without production of toxic forms of nitrogen oxides during a short time of incubation (118 h) at the same operational conditions with providing the aeration in the first treatment phase. The coculture of our isolated strains is assumed to have a good potential for nitrification and denitrification reactions applied in the treatment of wastewater containing ammonium, nitrite and nitrate. As a result, we can consider that the mixed culture is a practical method in the treatment of high-strength ammonium wastewater with reducing of sludge production.  相似文献

The simultaneous partial nitrification, anammox and denitrification (SNAD) process for treating domestic wastewater was investigated in a sequencing batch reactor (SBR). The SBR was operated with air flow rate of 500 L h?1 at 30 °C. Domestic wastewater was used as influent and Kaldnes rings were used as biomass carriers. In the beginning, long aeration condition was implemented to cultivate nitrification biofilm. Afterwards, intermittent aerobic condition was conducted during the cycle operation. The influent organic matter loading rate was improved by reducing the aeration and mixing times. Consequently, when the SNAD biofilm reactor was fed with the organic matter loading rate of 0.77 (kg COD m?3 d?1), the bio-bubbles appeared in the reactor and the total inorganic nitrogen (TIN) removal efficiency decreased. After the organic matter loading rate decreased to 0.67 (kg COD m?3 d?1), the reactor showed excellent nitrogen removal performance. The TIN removal efficiency varied between 80 and 90 %, and the average TIN removal loading rate was 0.22 (kg TIN m?3 d?1). Additionally, the scanning electron microscope (SEM) observation confirmed that the anammox bacteria located in the inner part of the carriers. Finally, the microbial community analysis of 16S rRNA gene cloning revealed that the anammox bacteria on the carriers consisted of three main genuses: Candidatus Brocadia sp., Candidatus Brocadia caroliniensis and Candidatus Brocadia fulgida.  相似文献

Summary Autotrophic Thiobacillus sp. CH11 and heterotrophic Pseudomonas putida CH11 were isolated from piggery waste water. Extensive tests, including removal action, removal efficiency, removal capacity, and adaptability to perturbed conditions were conducted on autotrophic and heterotrophic biofilters. The upflow bioreactor system without biomass-liquid separation unit was evaluated for its efficacy in sustaining autotrophic nitrification and denitrification (AND). The bioreactor system was capable of sustaining AND by means of carefully controlled oxygenation to achieve the maximum NH(4)(+)-N removal rate of 0.054 g N gVSS(-1) day(-1) (38% removal efficiency) at the oxygen influx and nitrogen loading rate of 3.68 mg O(2) h(-1) L-bioreactor(-1) and 182 mg N day(-1) L-bioreactor(-1), respectively. Additional nitrogen removal was achieved in a two-stage bioreactor configuration due to endogenous denitrification under long mean cell residence time. Quiescent conditions maintained in the bioreactor provided stable hydrodynamic environments for the chemoautotrophic biomass matrix, which revealed porous, loosely-structured, and mat-like architecture. More than 95% of the total biomass holdup (1.3-1.5 g VSS) was retained, thereby producing low biomass washout rate ( approximately 40 mg VSS day(-1)) with VSS < 11 mg VSSL(-1) in the effluent.  相似文献

An autotrophic denitrification process using reduced sulfur compounds (thiosulfate and sulfide) as electron donor in an activated sludge system is proposed as an efficient and cost effective alternative to conventional heterotrophic denitrification for inorganic (or with low C/N ratio) wastewaters and for simultaneous removal of sulfide or thiosulfate and nitrate. A suspended culture of sulfur-utilizing denitrifying bacteria was fast and efficiently established by bio-augmentation of activated sludge with Thiobacillus denitrificans. The stoichiometry of the process and the key factors, i.e. N/S ratio, that enable combined sulfide and nitrogen removal, were determined. An optimum N/S ratio of 1 (100% nitrate removal without nitrite formation and low thiosulfate concentrations in the effluent) has been obtained during reactor operation with thiosulfate at a nitrate loading rate (NLR) of 17.18 mmol N L(-1) d(-1). Complete nitrate and sulfide removal was achieved during reactor operation with sulfide at a NLR of 7.96 mmol N L(-1) d(-1) and at N/S ratio between 0.8 and 0.9, with oxidation of sulfide to sulfate. Complete nitrate removal while working at nitrate limiting conditions could be achieved by sulfide oxidation with low amounts of oxygen present in the influent, which kept the sulfide concentration below inhibitory levels.  相似文献

Autotrophic denitrification coupled with sulfide oxidation represents an interesting alternative for the simultaneous removal of nitrate/nitrite and sulfide from wastewaters. The applicability of such bioprocess is especially advantageous for the post treatment of effluents from anaerobic reactors, since they usually produce sulfides, which can be used as endogenous electron donor for autotrophic denitrification. This study evaluated the effect of sulfide concentration on this bioprocess using nitrate and nitrite as electron acceptors in vertical fixed-bed reactors. The results showed that intermediary sulfur compounds were mainly produced when excess of electron donor was applied, which was more evident when nitrate was used. Visual evidences suggested that elemental sulfur was the intermediary compound produced. There was also evidence that the elemental sulfur previously formed was being used when sulfide was applied in stoichiometric concentration relative to nitrate/nitrite. Nitrite was more readily consumed than nitrate. For both electron acceptors and sulfide concentrations tested, autotrophic denitrification was not affected by residual heterotrophic denitrification via endogenic activity, occurring as a minor additional nitrogen removal process.  相似文献

Summary A strain of Thiobacillus denitrificans was isolated after enrichment under anaerobic conditions by the continuous culture technique using thiosulfate as energy source and nitrate as electron acceptor and nitrogen source. The isolate was an active denitrifyer, the optimal conditions being 30°C and pH 7.5–8.0. Denitrification was inhibited by sulfate (the reaction product) above 5 g SO 4 = /l, whereas high concentrations of the substrates nitrate and thiosulfate were less harmful; nitrite affected denitrification above 0.2 g NO 2 /l. During the time course of denitrification in a batch culture growth and substrate consumption slowed down already after only half the substrate was utilized due to product inhibition. The following parameters were determined in continuous culture under nitrate limitation: max=0.11 h–1, K S=0.2 mg NO 3 /l, maximum denitrification rate=0.78 g NO 3 /g cells·h, g cells/g NO 3 , g cells/g S2O 3 = . Nitrite did not accumulate during steady state denitrification; the denitrification gas was almost pure N2. The concentrations of N2O and NO were below 1 ppm.  相似文献

Hydrogen sulfide is highly toxic and fatal to benthic organisms as well as causing depletion of dissolved oxygen and generating blue tide in eutrophic coastal seas. The purposes of this study are to reveal adsorption characteristics of hydrogen sulfide onto crushed oyster shell, and to evaluate removal efficiency of hydrogen sulfide from pore water in organically enriched sediments using container experiment in order to develop a coastal sediment amendment. The crushed oyster shell was mainly composed of CaCO3 with calcite and CaO crystal phase. The batch experiment showed removal kinetics of hydrogen sulfide can be expressed as the first order equation and Langmuir plot fitted well in describing the adsorption behavior with the adsorption maximum at 12 mg-S g−1. The container experiments suggested the oyster shell adsorbs hydrogen sulfide in pore water effectively and reduces oxygen consumption in the overlying water. Furthermore, oxidation–reduction potential of the sediment was higher with addition of crushed oyster shell than the control without oyster shell. Thus, it is concluded that crushed oyster shell can be an effective amendment to remediate organically enriched sediments in eutrophic coastal seas.  相似文献

A combined bioelectrochemical and sulfur autotrophic denitrification system (CBSAD) was evaluated to treat a groundwater with nitrate contamination (20.9-22.0mgNO(3)(-)-N/L). The reactor was operated continuously for several months with groundwater to maximize treatment efficiency under different hydraulic retention times (HRT) and electric currents. The denitrification rate of sulfur autotrophic part followed a half-order kinetics model. Moreover, the removal efficiency of bioelectrochemical part depended on the electric current. The reactor could be operated efficiently at the HRT ranged from 4.2 to 2.1h (corresponding nitrogen volume-loading rates varied from 0.12 to 0.24 kg N/m(3)d; and optimum current ranged from 30 to 1000 mA), and the NO(3)(-)-N removal rate ranged from 95% to 100% without NO(3)(-)-N accumulation. The pH of effluent was satisfactorily adjusted by bioelectrochemical part, and the sulfate concentration of effluent was lower than 250 mg/L, meeting the drinking water standard of China EPA.  相似文献

The simultaneous ammonium and sulfate removal was detected in an anammox reactor, consisted of ammonium oxidization with sulfate deoxidization, and subsequently traditional anammox process, in via of middle medium nitrite with solid sulfur and N2 as the terminal products. The pure anammox bacteria offered a great biotechnological potential for the completely autotrophic reaction indicated by batch tests. Denaturing gradient gel electrophoresis (DGGE) analysis further revealed that a new organism belonging to Planctomycetales was strongly enriched in the defined niche: the redox of ammonium and sulfate. The new species "Anammoxoglobussulfate" was so considered as holding a critical role in the ammonium oxidization with sulfate deoxidization to nitrite. Afterwards, the Planctomyces existing in the bacteria community performed the anammox process together to achieve the complete nitrogen and sulfate removal. The potential use of sulfate as electron acceptor for ammonium oxidizing widens the usage of anammox bacteria.  相似文献

We measured denitrification and nitrate removal rates in cold seep sediments from the Gulf of Mexico. Heterotrophic potential denitrification rates were assayed in time-series incubations. Surficial sediments inhabited by Beggiatoa exhibited higher heterotrophic potential denitrification rates (32 μ N reduced day−1) than did deeper sediments (11 μ N reduced day−1). Nitrate removal rates were high in both sediment horizons. These nitrate removal rates translate into rapid turnover times (<1 day) for the nitrate pool, resulting in a faster turnover for the nitrate pool than for the sulfate pool. Together, these data underscore the rigorous nature of internal nitrogen cycling at cold seeps and the requirement for novel mechanisms to provide nitrate to the sediment microbial community.  相似文献
