The aluminium-formaldehyde (ALFA) histofluorescence method reveals an extensive plexus of brilliant greenish monoaminergic elements in the glandular zones of the Mytilus foot, while only scanty nerve fibres are acetylcholinesterase-positive. By electron microscopy, bundles of nerve fibres can be seen i) in close connection with the intrinsic musculature located in the connective septa among the glands, and ii) near the cell bodies and necks of all the byssus glands. The nerve fibres show varicosities containing three types of vesicles: small clear (50-60 nm), small granular (80-90 nm), and large granular (160-200 nm). The regions of close apposition between nerve terminals and muscle or gland cells generally do not show typical pre- or postsynaptic specializations. Along the pedal groove, mainly in the proximal two thirds of the foot, peripheral bipolar neurons can be detected, both by fluorescence and electron microscopy.  相似文献

The structure of the nerve fibres in the chronically isolated cat ileum was studied by fluorescence and electron microscopy after imipramine group compounds, parachlorophenylalanine, and 5-hydroxytryptamine treatment. Following treatment with parachlorophenylalanine and imipramine group compounds in the nerve fibres of the small intestine, specific granules were selectively decreased (p less than 0.001) in number. In concordance with ultrastructural observations, a marked diminution of fluorescence intensity was demonstrable in the small intestine. In addition, the number of the granular vesicles was significantly increased following 5-hydroxytryptamine treatment, and yellow fluorescent neurones and processes were observed in the myenteric and submucous plexuses. On the basis of these observations, the serotoninergic nature of certain nerve fibres could be demonstrated.  相似文献

The ultrastructure of the innervation of the human ureterovesical junction was studied. Three different nerve terminals were distinguished among the smooth muscle cells. 1. Nerve processes containing predominantly small granular vesicles (40--60 nm in diameter). 2. Other nerve fibres contained predominantly small round agranular vesicles (30--50 nm in diameter). 3. Processes with large granulated vesicles (80--120 nm in diameter). The first type may be adrenergic, the second cholinergic and the third may originate from the local nerve cells. The gap between the nerve fibres and muscle cells was 300 to 500 nm wide and no synaptic thickenings were observed. This suggests that the transmitter may influence several muscle cells, and the different nerve fibres may directly innervate the smooth muscle cells.  相似文献

Summary The stomach, small intestine and large intestine of the toad, Bufo marinus, were processed for formaldehyde-induced fluorescence histochemistry. After extrinsic denervation or pretreatment with 6-hydroxydopamine to remove catecholamine fluorescence, yellow fluorescence typical of 5-hydroxytryptamine was observed in neurones in the small intestine only. The cell bodies and their processes were confined to the myenteric plexus. Additional pretreatment with 5-hydroxytryptamine enhanced the fluorescence of neurones in the small intestine and revealed yellowfluorescent nerve fibres, but not cell bodies, in the longitudinal and circular muscle layers and myenteric plexus of the large intestine. No fluorescent neurones were observed in the stomach. Following reserpine treatment, which removed native yellow fluorescence in the small intestine, exposure to 5-hydroxytryptophan produced yellow fluorescence in axons in both small and large intestine; exposure to tryptophan never restored fluorescence. The neurotoxin, 5,7-dihydroxytryptamine had no effect on the distribution of yellow-fluorescent neurones in the small and large intestine. No 5-HT-containing mast cells were present in either the small or large intestine. Thin layer chromatography with three different mobile phases showed a 5-hydroxytryptamine-like compound in extracts of mucosa-free small and large intestine but not of stomach.  相似文献

Summary Fluorescence histochemistry has been used to study the effects of 6-hydroxydopamine (6-OHDA) (100 mg/kg injected into the dorsal lymph sac) on adrenaline-containing nerves in the large intestine, mesentery, lung, bladder and heart atria of the toad Bufo marinus. A gradual decrease both in fluorescence intensity and in number of detectable fibres during the first 4 hours after 6-OHDA was accompanied by a build-up of fluorescence in the nonterminal regions. These phenomena have been discussed in relation to the time course of the degeneration produced by 6-OHDA in noradrenergic nerves of higher vertebrates. Almost complete chemical sympathectomy was seen after one day, and it was not till 13 days that regenerating nerve fibres were seen in any organ. In the large intestine, however, re-innervation was slower, being first noted after 39 days. The time course of regeneration has been compared with that following sympathectomy in various mammalian organs.  相似文献

Abstract: Intracisternal administration of 100 μg 6-OHDA to newborn rats causes permanent defects, not only of the monoaminergic neuron system, but also of extraneuronal tissue elements. The long noradrenergic fibre tracts are irreversibly destroyed, while the short projections recover and regenerate after a transient period of injury. In the major noradrenergic cell group, the locus coeruleus, most of the cells in the caudal and middle parts degenerate, while a small dorsorostral group survives and forms the source of the regenerating fibres. Dopaminergic and serotonergic fibre tracts are also affected. The 6-OHDA treatment also damages granule and dial cells of the cerebellar cortex as well as the mesenchymal cells of the pial coverings of the cerebellum, leading to primitive foliation, absence of fissuration, and defective migration of granule cells and resulting in a marked reduction of cerebellar size, area, and granule cell number.  相似文献

Summary The occurrence and distribution of supra-ependymal nerve terminals storing serotonin (5-HT) are described for the fourth ventricle of the rat brain. The nerve terminals were identified as monoaminergic 1) fluorescence-histochemically, by the presence of a varicose, formaldehyde-induced fluorescence (FIF) on the free surface of the ependyma, 2) electron microscopically, by the presence of electron dense (chromaffin) cores in small (50 nm) and large (100 nm) vesicles found within the varicose regions of supra-ependymal nerve fibres, and 3) by the absence of both the FIF and chromaffin dense cores after treatment with reserpine. Moreover, the serotonergic nature of these nerve fibres could be concluded from 1) the yellow colour of the FIF, 2) the increased FIF after treatment with nialamide or reserpine+nialamide, 3) the diminished FIF and absence of chromaffin dense cores after treatment with p-CPA, and finally 4) the persistence of the FIF and chromaffin dense cores after treatment with -MPT.A high density of 5-HT nerve terminals occurred throughout the floor of the fourth ventricle and on the floor and roof of the lateral recess. Few 5-HT nerve terminals occurred only on the roof of the fourth ventricle (velum medullare, lamina epithelialis of the tela chorioidea), and the surface of the choroid plexus epithelia was devoid of such nerves. Summary Synaptic regulation of arginine vasopressin (AVP)-containing neurons by neuropeptide Y (NPY)-containing monoaminergic neurons was demonstrated in the paraventricular nucleus of the rat hypothalamus. NPY and AVP were immunolabeled in the pre- and the post-embedding procedures, respectively, and monoaminergic fibers were marked by incorporating 5-hydroxydopamine (5-OHDA), a false neurotransmitter. The immunoreaction for NPY was expressed by diaminobenzidine (DAB) chromogen, and that for AVP by gold particles. The DAB chromogen was localized on the surface of the membrane structures, such as vesicles or mitochondria, and on the core of large cored vesicles. Gold particles were located on the core of the secretory granules within the AVP cell bodies and processes. The incorporated 5-OHDA was found as dense cores within small or large vesicular structures. From these data, three types of nerve terminals were discernible: NPY-containing monoaminergic, NPY-containing non-aminergic, and monoaminergic fibers. The AVP cell bodies appeared to have synaptic junctions formed by these nerve terminals as well as by the unlabeled nerve terminals which have small clear vesicles and large cored vesicles. These different types of nerve terminals were frequently observed in a closely apposed position on the same AVP cell bodies. The functional relationships of these three types of neuronal terminals are discussed.  相似文献

Calcitonin-gene-related-peptide (CGRP)-like immunoreactivity was localized in nerve fibres, neuronal somata and in mucosal endocrine cells of the human small intestine. Immunoreactive enteric neurons were more numerous in the submucous plexuses than in the myenteric plexus. Morphologically, they predominantly had the appearance of type II neurons. The majority of the CGRP-like immunoreactive nerve fibres ran within the ganglionic nerve plexuses. Only a small proportion could be observed in the lamina propria, the lamina muscularis mucosae, or the circular and longitudinal outer smooth muscle layer. These findings suggest that within the wall of the human small intestine neuronal CGRP of either extrinsic or intrinsic origin exerts its effect chiefly on other enteric neurons, and might be indirectly involved in the regulatory functions of the human small intestine.  相似文献

Summary Single- and dual-labelling immunohistochemistry were used to determine the distribution and coexistence of neuropeptides in perivascular nerves of the large arteries and veins of the snake, Elaphe obsoleta, using antibodies for vasoactive intestinal polypeptide, substance P, calcitonin gene-related peptide, neuropeptide Y, galanin, somatostatin, and leu-enkephalin. Blood vessels were sampled from four regions along the body of the snake: region 1, arteries and veins anterior to the heart; region 2, central vasculature 5 cm anterior and 10 cm posterior to the heart; region 3, arteries and veins in a 30-cm region posterior to the liver; and region 4, dorsal aorta and renal arteries, renal and intestinal veins, 5–30 cm cephalad of the vent. A moderate to dense distribution of vasoactive intestinal polypeptide-like immunoreactive fibres was found in most arteries and veins of regions 1–3, but fibres were absent from the vessels of region 4. The majority of vasoactive intestinal polypeptide-like immunoreactive fibres contained colocalized substance P-like immunoreactivity, and these fibres were unaffected by either capsaicin or 6-hydroxydopamine (6-OHDA) pretreatment. In the anterior section of the snake, the vagal trunks contained many cell bodies with colocalized vasoactive intestinal polypeptide and substance P-like immunoreactivity. It is suggested that the vasoactive intestinal polypeptide/substance P-like immunoreactive cell bodies and fibres are parasympathetic postganglionic nerves. Neuropeptide Y-like immunoreactive fibres were observed in all arteries and veins, being most dense in regions 3 and 4. The majority of these fibres also contained colocalized galanin-like immunoreactivity, and were absent in tissues from 6-OHDA pretreated snakes, suggesting that neuropeptide Y and galanin are colocalized in adrenergic nerves. A small number of neuropeptide Y-like immunoreactive fibres contained vasoactive intestinal polypeptide but not galanin, and were unaffected by 6-OHDA treatment. All calcitonin gene-related peptide-like immunoreactive fibres contained colocalized substance P-like immunoreactivity, and these fibres were observed in all vessels, being particularly dense in the carotid artery and jugular veins. All calcitonin gene-related peptide/substance P-like immunoreactive fibres appeared damaged after capsaicin treatment suggesting they represent fibres from afferent sensory neurons. A sparse plexus of somatostatin-like immunoreactive fibres was observed in the vessels only from region 4. No enkephalin-like immunoreactive fibres were found in any blood vessels from any region. This study provides morphological evidence to suggest that there is considerable functional specialization within the components of the rat snake peripheral autonomic system controlling the circulation, in particular the regulation of venous capacitance.  相似文献

The nerve elements in the urinary bladder of the cat were studied by electron microscopy. According to their ultrastructure, nerve cell somata can be classified into three types: the large cells with a cytoplasm rich in organelles, several processes and numerous synaptic contacts on their surface; the cytoplasm contained 80- 120-nm granulated vesicles. The second type is poor in cytoplasmic organelles and has very few processes and virtually no synaptic contacts on the soma. The third type contains numerous large 160- to 220-nm 'neurosecretory' vesicles in the cytoplasm. According to the morphology of the vesicle population, four types of nerve processes could be distinguished: Type a, with a dominant population of small (40-60 nm) agranular vesicles. These are thought to be sacral parasympathetic fibres. Type b, with small (40-60 nm) granular vesicles, which may be the noradrenergic sympathetic fibres. Type c, with 80- to 120-nm granulated vesicles, probably of local origin. Typed d, with large 160- to 220-nm 'neurosecretory' vesicles also of local origin. Different types of nerve fibres are converging on the local nerve cells. This suggests that the local circuits can play an important role in coordinating the function of the bladder. Therefore, ganglia may be considered as an elementary functional unit.  相似文献

The myocardium of rats kept on DeCarli and Lieber's liquid diet containing alcohol to 36% of the total calories for 8 weeks were examined with Falck-Hillarp's method for the histologic demonstration of catecholamines. Relative fluorescence intensity of the myocardial cells and nerves was measured with the Zeiss-Fluoval microphotometer. Under the effect of alcohol, density of the noradrenergic innervation (number of visualized nerve fibres) decreases and fluorescence of myocardial cells is intensified. The increased fluorescence of myocardial cells can be eliminated with reserpine. It is assumed that chronic alcohol intake induces continuous catecholamine release from monoaminergic nerve fibres of the heart.  相似文献

The fluorescent Ca2+ indicator, quin 2, has been used in isolated striated muscle fibres. There is a distinct quin 2 fluorescence peak at lambda 500 nm upon excitation at lambda 339 nm after axial injection of the potassium salt of quin 2, pH 7.1. Single voltage-clamp or current clamp electrical stimulation resulted in a distinct transient change in the fluorescence at lambda 500 nm which was not observed at lambda 400 nm, the peak of the fibre autofluorescence. Ca2+ buffering is marked at high quin 2 concentrations (greater than or equal to 400 microM) producing a slow decay of force and fluorescence. At lower concentrations (8-30 microM) of quin, the decay of force is within the range observed in non-injected control fibres. A Kd of 457 nM at 5 mM free Mg2+ suggests an upper resting free Ca2+ concentration of 310 nM at 12 degrees C.  相似文献

Summary The frontal ganglion, part of the stomatogastric nervous system, contains about 60 to 80 neurons, 25 to 30 m in diameter. A well developed Golgi system, producing dense-core vesicles, lysosomes, multivesicular bodies and dense bodies are abundant. Glia elements are sparsely distributed. Many nerve fibres contain granules of different size and electron density. Five groups of fibres can be distinguished: Fibres with granules of about 200 nm (type A), fibres with granules of about 160 to 170 nm (type B), fibres with granules of about 80 to 100 nm (type C) and those with synaptic vesicles of 50 nm (type D) respectively. A fifth very small type contains neither vesicles nor granules. Special attention was paid to synaptic contacts. The divergent dyad seems to be the main type in the frontal ganglion. Frequently, neurosecretory endings are observed in presynaptic position. Immunocytochemical staining of neurosecretory material closely corresponds to the distribution of type A fibres, as observed electron microscopically. Immunoelectrophoresis of extracts from frontal ganglia with polyspecific anti-neurosecretion-serum reveals a single precipitation line, indicating that the immunocytochemical localization of neurosecretory material is due to reaction with a specific as well as a crossreagent antibody.Supported by the Ministerium für Wissenschaft und Technik der DDRThe authors wish to thank Mrs. B. A comparative study of the quantitative data of the frog extraocular muscles and the cranial nerves that innervate them was performed. Oculorotatory muscles contain muscle fibres of at least 4 types which are arranged in heterogeneous layers. The zonal arrangement of the muscles does not occur on the cross-sections in the vicinity of muscle insertions. In these regions only two muscle fibre types are present and the total number of fibres is smaller by 70% than in the central region of the muscle. Most numerous are muscle fibres in the rectus inferior muscle, while the smallest number of fibres is found in rectus interior muscle. Three distinct types of nerve fibres are distinguished according to the following criteria: occurrence and thickness of myelin sheath, fibre diameter and ratio "g". The fibres with thin myelin sheaths indicate small diameters (1-5--6- mum) and their ratio "g" equals 0-82 +/- 0-08. They constitute about 30% of the myelinated fibres in the nerve supply of the oculorotatory muscles and about 14% in the supply of the retractor bulbi muscle. Both the value of the ratio "g" and a greater number of these fibres in the nerve supply of the muscles that contain slow contracting muscle fibres indicate that they are rather slow conducting nerve fibres. The range of the diameters of the fibres with thick myelin sheaths is greater (3-5--13-5 mum) and their "g" equals 0-66 +/- 0-06. These fibres constitute about 70% of the myelinated ones in the nerve supply of the oculorotatory muscles and 86% in the supply of the retractor bulbi muscles. The value of the ratio "g" in these fibres indicates that they are fast contracting ones. The smallest diameters are found in the myelinated fibres (0-5--1-7 mum). These fibres occur frequently in all the examined nerves; they constitute 36--47% of the total number of all the nerve fibres. The frog extraocular muscles are characterized by an abundal nerve supply which is reflected in the low innervation ratio (1:4--1:5). On the distal cross-section of nerves the number of nerve fibres is greater than on the proximal ones. Ganglionic neurons occur sporadically around the nerves; in the nerve III synaptic contacts between two neurons were observed.  相似文献

Summary The pattern of noradrenergic innervation of guinea-pig hepatic tissue was examined by using the fluorescence method of Falk et al. (1962). The distribution of monoaminergic nerve fibres in untreated animals was clearly evident at the level of the portal vein, the hepatic artery and the bile duct. The hepatic artery was the most densely innervated. Mast cells and Kupffer cells had no connection with the adrenergic fibres. In daunomycin-pretreated animals, the hepatic cells appeared to be deep red in colour owing to the presence of the antibiotic. Some noradrenergic nervous fibres, never previously described, were unexpectedly distinguishable in the liver parenchyma against the background red colour of daunomycin-pretreated tissue. They appeared to be contiguous to the hepatocytes and were of different shapes: comma-like, V-like, or like a string of pearls. The nature and the functional role of these elements is briefly discussed.  相似文献

Summary The ciliary body of the cat was investigated by fluorescence histochemistry and electron microscopy in an attempt to clarify its sympathetic innervation. Subconjunctival doses of 5-hydroxydopamine (5-OHDA) or 6-hydroxydopamine (6-OHDA) were given to establish the precise location of the sympathetic nerve terminals. The distribution of noradrenergic fibers and terminals was shown by fluorescence histochemistry to be sparse in the trabecular meshwork and the anterior portion of the ciliary muscle, but dense in the subepithelial tissue. The small and large dense core vesicles which occur in many nerve endings of the subepithelial tissue adjacent to the pigmented epithelial layer increased in electron density following the administration of 5-OHDA. Many degenerating nerve endings were found in the same region of animals treated with 6-OHDA. In contrast, there were few noradrenergic terminals in the ciliary muscle except for a portion of the smooth muscle which was shown to be dually innervated. The noradrenergic fibers in the subepithelial region and the trabecular meshwork may play an important role in aqueous secretion and outflow.This work was supported in part by a research grant from the Ministry of Education, Japan  相似文献

Summary The projections of nerve fibres with immunoreactivity for the peptides enkephalin (ENK), gastrin-releasing peptide (GRP), neuropeptide Y (NPY), somatostatin (SOM), substance P (SP) and vasoactive intestinal peptide (VIP) were studied in canine small intestine by analysing the consequences of lesions of intrinsic and extrinsic nerves. Of peptides present in fibres supplying myenteric ganglia, GRP, SOM and VIP were in anally directed nerve pathways, whereas ENK and NPY were in orally directed pathways. Pathways ran for up to about 30 mm. SP fibres ran for short distances in both directions in the myenteric plexus. The circular muscle was supplied with ENK, NPY, SP and VIP fibres arising from the myenteric ganglia, whereas most mucosal SP and VIP fibres were deduced to arise from submucous ganglia. There were projections of fibres reactive for ENK, GRP, SOM, SP and VIP from myenteric ganglia to submucous ganglia. Antibodies to tyrosine hydroxylase were used to locate noradrenaline nerve fibres supplying the intestine; these fibres all disappeared when extrinsic nerves running through the mesentery to the small intestine were cut. It is deduced that there is an ordered pattern of projections of peptide-containing fibres in the canine intestine.  相似文献

The myenteric plexus of the domestic fowl (Gallus domesticus) small intestine was studied by means of silver staining, glyoxylic acid-induced fluorescence, the modified Koelle-Friedenwald method for the detection of acetylcholinesterase, NADH-diaphorase techniques and the unlabelled antibody method involving the use of an antiserum raised against GABA conjugated by glutaraldehyde to bovine serum albumin. The majority of the perikarya were in the ganglia, with an average density of 3370 +/- 942 nerve cells/cm2. Cholinesterase-positive and a few GABA-immunoreactive nerve cell bodies were seen in the myenteric ganglia, while fluorescent ganglion cells were not observed. In addition to AChE and GABA-positive nerve fibres, a rich fluorescent network of varicose and nonvaricose nerve fibres was detected, pointing to the presence of an extrinsic aminergic system in the domestic fowl myenteric plexus. Electron microscopic observations on nerve cells, axon profiles and varicosites with various vesicle populations were in good agreement with the histochemical findings.  相似文献
