The life cycle of Leishmania alternates between two main morphological forms: intracellular amastigotes in the mammalian host and motile promastigotes in the sand fly vector. Several different forms of promastigote have been described in sandfly infections, the best known of these being metacyclic promastigotes, the mammal-infective stages. Here we provide evidence that for Leishmania (Leishmania) mexicana and Leishmania (Leishmania) infantum (syn. chagasi) there are two separate, consecutive growth cycles during development in Lutzomyia longipalpis sand flies involving four distinct life cycle stages. The first growth cycle is initiated by procyclic promastigotes, which divide in the bloodmeal in the abdominal midgut and subsequently give rise to non-dividing nectomonad promastigotes. Nectomonad forms are responsible for anterior migration of the infection and in turn transform into leptomonad promastigotes that initiate a second growth cycle in the anterior midgut. Subsequently, leptomonad promastigotes differentiate into non-dividing metacyclic promastigotes in preparation for transmission to a mammalian host. Differences in timing, prevalence and persistence of the four promastigote stages were observed between L. mexicana and L. infantum in vivo, which were reproduced in cultures initiated with lesion amastigotes, indicating that development is to some extent governed by a programmed series of events. A new scheme for the life cycle in the subgenus Leishmania (Leishmania) is proposed that incorporates these findings. 相似文献

Lutzomyia spp. are New World phlebotomine sand flies, many of which are involved in the transmission of human diseases, such as leishmaniases and bartonellosis. The systematic classification of the approximately 400 species in the genus has been based on morphological characters, but the relationships within the genus are still very much in question. We have inferred phylogenies of 32 species of phlebotomine sand flies belonging to seven sub-genera and two species groups, by using fragments of the mitochondrial small subunit (12SrRNA) and of the nuclear large subunit (28SrRNA) ribosomal gene sequences. The subgenus Helcocyrtomyia and the Verrucarum species group, prominent representatives of the Peruvian sand fly fauna, were represented by 11 and 7 species, respectively. Although based on a limited number of taxa, the resulting phylogenies, based on 837 characters, provide an initial phylogenetic backbone for the progressive reconstruction of infrageneric relationships within Lutzomyia. 相似文献

An expression library was created and 2304 clones sequenced from a monogyne colony of Solenopsis invicta. The primary intention of the project was to utilize homologous gene identification to facilitate discovery of viruses infecting this ant pest that could potentially be used in pest management. Additional genes were identified from the ant host and associated pathogens that serve as an important resource for studying these organisms. After assembly and removal of mitochondrial and poor quality sequences, 1054 unique sequences were yielded and deposited into the GenBank database under Accession Nos. EH412746 through EH413799. At least nine expressed sequence tags (ESTs) were identified as possessing microsatellite motifs and 15 ESTs exhibited significant homology with microsporidian genes. These sequences most likely originated from Thelohania solenopsae, a well-characterized microsporidian that infects S. invicta. Six ESTs exhibited significant homology with single-stranded RNA viruses (3B4, 3F6, 11F1, 12G12, 14D5, and 24C10). Subsequent analysis of these putative viral ESTs revealed that 3B4 was most likely a ribosomal gene of S. invicta, 11F1 was a single-stranded RNA (ssRNA) virus contaminant introduced into the colony from the cricket food source, 12G12 appeared to be a plant-infecting tenuivirus also introduced into the colony as a field contaminant, and 3F6, 14D5, and 24C10 were all from a unique ssRNA virus found to infect S. invicta. The sequencing project illustrates the utility of this method for discovery of viruses and pathogens that may otherwise go undiscovered. 相似文献

Simple sequence repeats (SSRs) markers were developed for Betula maximowicziana using 2698 expressed sequence tags (ESTs) from the NCBI database. Out of 112 designed primer pairs, 54 showed clear PCR amplification
and 14 of these revealed polymorphism in eight individuals sampled across the species’ range. The number of alleles detected
and the expected heterozygosity ranged from 1 to 3 and 0.000 to 0.570, respectively, when these 14 loci were examined in 49
individuals from a single population. In the cross species transferability test, eight of the 14 loci were also polymorphic
in all four of the diploid, tetraploid and hexaploid Betula species examined. These results showed high transferability of the developed EST-SSRs and that these markers are likely to
be useful in studies of the population genetics of species in the genus Betula. 相似文献

Midgut pH profile and protein digestion in the larvae of Lutzomyia longipalpis (Diptera: Psychodidae) 总被引:1,自引:0,他引:1
The sand fly Lutzomyia longipalpis is the vector of Leishmania infantum, the etiological agent of American visceral leishmaniasis. Despite its importance, until now the internal anatomy of the immature forms has never been described and little is known about their digestive processes. In nature, sand fly larvae feed on organic detritus in the soil, constantly ingesting large amounts of material. The objective of this study was to describe the anatomy of the gut and the pH of the gut lumen, as well as to investigate the proteases responsible for protein digestion. The larvae have a short gut with a prominent, well-developed midgut. Ingestion of food containing indicator dyes permitted the gut pH to be measured. A pH gradient was observed, varying from >9 in the anterior midgut to 6.5-7.0, in the posterior midgut. The endoproteolytic enzymes are secreted in the anterior midgut and are able to digest azocasein over a large pH range, specially at pH 11. Studies with various inhibitors indicated that the digestive endoproteases are trypsin- and chymotrypsin-like enzymes. These results were confirmed by using the substrates BApNA and N-CBZ-L-PpNA, specific for trypsin and chymotrypsin, respectively. Aminopeptidases were also investigated with p-nitroaniline-derived substrates. These enzymes are located in the posterior midgut, bound to the membranes and functioning at an optimal pH of 6.5-8.0. The results presented here are consistent with the current proposal that proteins are digested to peptides in the anterior midgut inside the endoperitrophic space and subsequently undergo digestion in the ectoperitrophic space of the posterior midgut. 相似文献

Raspberry breeding is a long, slow process in this highly heterozygous out-breeder. Selections for complex traits like fruit
quality are broad-based and few simple methodologies and resources are available for glasshouse and field screening for key
pest and disease resistances. Additionally, the timescale for selection of favourable agronomic traits requires data from
different seasons and environmental locations before any breeder selection can proceed to finished cultivar. Genetic linkage
mapping offers the possibility of a more knowledge-based approach to breeding through linking favourable traits to markers
and candidate genes on genetic linkage maps. To further increase the usefulness of existing maps, a set of 25 polymorphic
SSRs derived from expressed sequences (EST-SSRs) have been developed in red raspberry (Rubus idaeus). Two different types of expressed sequences were targeted. One type was derived from a root cDNA library as a first step
in assessing sequences which may be involved in root vigour and root rot disease resistance and the second type were ESTs
from a gene discovery project examining bud dormancy release and seasonality. The SSRs detect between 2 and 4 alleles per
locus and were assigned to linkage groups on the existing ‘Glen Moy’ × ‘Latham’ map following genotyping of 188 progeny and
examined for association with previously mapped QTL. The loci were also tested on a diverse range of Rubus species to determine transferability and usefulness for germplasm diversity studies and the introgression of favourable alleles. 相似文献

A thorough understanding of the transmission mechanism of any infectious agent is crucial to implementing an effective intervention strategy. Here, our current understanding of the mechanisms that Leishmania parasites use to ensure their transmission from sand fly vectors by bite is reviewed. The most important mechanism is the creation of a "blocked fly" resulting from the secretion of promastigote secretory gel (PSG) by the parasites in the anterior midgut. This forces the sand fly to regurgitate PSG before it can bloodfeed, thereby depositing both PSG and infective metacyclic promastigotes in the skin of a mammalian host. Other possible factors in transmission are considered: damage to the stomodeal valve; occurrence of parasites in the salivary glands; and excretion of parasites from the anus of infected sand flies. Differences in the transmission mechanisms employed by parasites in the three subgenera, Leishmania, Viannia and Sauroleishmania are also addressed. 相似文献

Flow cytometry analysis showed variation of nuclear DNA content among different species of Spartina. Spartina alterniflora had the biggest genome (1763.9 Mbp) and S. cynosuroides had the smallest genome (756.35 Mbp), whereas the genomes of S. patens (969.36 Mbp) and S. spartinae (979.78 Mbp) were comparable. Mining simple sequence repeats (SSR) from 1227 expressed sequence tags (EST) generated from salt stressed S. alterniflora showed an abundance of di- and tri-nucleotide repeats. Of 100 ESSR (EST-derived SSR) loci with five or more repeats, 81 loci were successfully amplified in eight S. alterniflora genotypes and 15 (22.2%) ESSR markers were polymorphic. Eleven of the 15 polymorphic ESSRs showed amplification across six different species of Spartina while 100% cross transferability was observed with at least one species of Spartina. The average number of alleles per marker was 3.9 and 5.8 within S. alterniflora and among Spartina species, respectively. Geographic and ecological distributions of three Lutzomyia sand flies that are cutaneous leishmaniasis vectors in South America were analysed using ecological niche modelling. This new tool provides a large-scale perspective on species' geographic distributions, ecological and historical factors determining them, and their potential for change with expected environmental changes. As a first step, the ability of this technique to predict geographic distributions of the three species was tested statistically using two subsampling techniques: a random-selection technique that simulates 50% data density, and a quadrant-based technique that challenges the method to predict into broad unsampled regions. Predictivity under both test schemes was highly statistically significant. Visualisation of ecological niches provided insights into the ecological basis for distributional differences among species. Projections of potential geographic distributions across scenarios of global climate change suggested that only Lutzomyia whitmani is likely to be experiencing dramatic improvements in conditions in south-eastern Brazil, where cutaneous leishmaniasis appears to be re-emerging; Lutzomyia intermedia and Lutzomyia migonei may be seeing more subtle improvements in climatic conditions, but the implications are not straightforward. More generally, this technique offers the possibility of new views into the distributional ecology of disease, vector, and reservoir species. 相似文献

Phlebotomine sandflies are vectors of important pathogens world-wide, including Leishmania spp. in the Neotropics. Entomoparasites have been described from phlebotomines, including virus, bacteria, protozoa, fungi, nematodes, and mites, some of which are capable of killing the host. In the present study, interference, fluorescence, and scanning electron microscopies were used for the first time to detect and morphologically characterize a new entomoparasite infecting Lutzomyia longipalpis. Several filiform larvae and eggs in different stages were encountered in the abdomen of female and male insects. Pairs of large egg-bearing nematodes found within cyst-like structures or free in the hemocel accompanied by larvae could be the adult sexual stages. This entomoparasite infects sand flies naturally in the field. We believe that stress caused by the colonization procedure produced an increase in the infection rate among sand flies affecting their development. These findings could be applied to future biological control studies of sand fly vectors. 相似文献
