The lethal and mutagenic effect of N-nitrozo-N-methylbiuret (NMB) on the organisms producing levorin, amphotericin B and mycoheptin was studied. The mutagen effect depended on the dose, culture and physiological state of the spores. NMB had a low mutagenic effect on the levorin-producing organism characterized by high activity and genetic homogenicity with respect to the colony morphology and antibiotic production. As for the organisms producing amphotericin B and mycoheptin characterized by high genetic heterogenicity, significant variation of all the features studied was observed on their exposure to the mutagen. Inspite of diverse reaction of the organisms producing levorin, amphotericin B and mycoheptin to the effect of NMB mutants with increased antibiotic production were obtained from the three cultures. The lethal and mutagenic effect of NMB on the mycoheptin-producing organism depended on the process of the spore DNA replication. The spores during the DNA replication period were least sensitive to the lethal effect of the mutagen and most mutable with the respect to the colony morphology. For selection of highly active and stable strains exposure to NMB of the spores of the mycoheptin-producing organism during replication of DNA proved to be more effective than that of the spores during the lag-phase.  相似文献

The effect of N-nitrozo-N-methylbiuret (NMB) on various features of Didymocladium ternatum, an organism producing termatin was studied. It was found that the lethal effect of NMB depended on the exposure time, prolongation of which resulted in increased numbers of the plus variants, morphological mutations and reverses into the populations. Variants possessing 8 times higher activity as compared to the initial culture were obtained as a result of selection of active variants.  相似文献

从自然感病的温室桃蚜上分离到一株球孢白僵菌Bb21,测定了该菌株对桃蚜的致病性及其对两种捕食性天敌的影响.结果表明:Bb21菌株对桃蚜的致病力强,LD5o为97孢子·mm-2,95%置信区间为45~191孢子·mm-2;对草蛉2龄幼虫有较弱的致病性,LD50为1089孢子·mm-2,是桃蚜的11.2倍;对异色瓢虫致病性极小,高浓度处理(5×108孢子·mL-1)的平均感染率仅为l3%.该菌株低浓度处理对两种捕食性天敌的发育历期和生殖力均无显著影响,但高浓度处理(5×108孢子·mL-1)使异色瓢虫的幼虫期平均缩短1.4d,羽化率降低33%,产卵量减少14%,使普通草蛉的幼虫期平均缩短0.7d,羽化率降低24%,产卵量减少11%.该菌株对桃蚜的半致死剂量远低于对两种捕食性天敌的半致死剂量,并且在防治桃蚜使用浓度下对两种捕食性天敌成虫羽化率和繁殖力的影响极小,可作为温室桃蚜的生物控制因子在有害生物综合治理中应用.  相似文献

Aerial conidia of Beauveria bassiana in an emulsifiable formulation germinated by >95% after 24 h exposure to the regimes of 20, 25 and 30 degrees C with 51%, 74% and 95% RH. Ovicidal activities of the formulation towards two-spotted spider mite, Tetranychus urticae, were assayed at the concentrations of 0, 18, 160 and 693 conidia mm(-2) sprayed separately onto fava bean leaves including 39 (25-76) eggs per capita. All the sprayed eggs on the leaves were directly exposed to the different regimes for hatch after 24 h maintenance in covered Petri dishes. Generally, hatched proportions increased over post-spray days and decreased with the elevated fungal concentrations; no more eggs hatched from day 9 or 10 onwards. Based on the counts of the hatched/non-hatched eggs in the different regimes, the final egg mortalities were 15.0-40.4%, 48.9-66.6% and 62.9-87.5% at the low, medium and high concentrations, respectively, but only 5.6-11.3% in blank controls. The RH effect on the fungal action was significant at 20 and 25 degrees C but not at 30 degrees C whereas the effect of temperature was significant at 51% and 74% RH but not at 95% RH. Probit analysis of the egg mortalities versus the fungal sprays generated median lethal concentrations (LC(50)) of 65-320 conidia mm(-2) at all the regimes, and of only 65-78 conidia mm(-2) at 25-30 degrees C with 74-95% RH. The results highlight ovicidal activities of the emulsifiable formulation against the mite species at the tested regimes and its potential use in spider mite control.  相似文献

将球孢白僵菌(Beauveriabassiana)BBSG8702的未干燥孢子粉(含水量58.9±1.6%)和真空冷冻干燥孢子粉(含水量7.4±0.9%)置于4℃和20℃下贮存1个月,每隔5d取样测定活孢率和孢子内贮总糖和蛋白含量,发现含水量和贮存温度交互影响孢子的活力以及内贮总糖和蛋白质的代谢水平,各组合中的活孢率一般与内贮总糖和蛋白质代谢水平均存在显著或极显著相关性.在1个月的贮存期间,4℃下冻干粉总糖含量下降13.4%,蛋白质含量下降39.2%,清水中的萌发率下降32.0%,营养液中的萌发率仅下降6.7%,而未干燥孢子粉的相同指标分别下降42.4%、66.3%、96.4%和99%;在20℃下,冻干粉的上述指标分别下降了14.1%、38.2%、55.8%和 10.4%,而未干燥孢子粉则分别下降了 43.2%、65.4%、99.4%和98.4%. 显然,含水量影响活孢率和内贮营养衰变的幅度,而温度影响衰变的速度,但内贮营养的耗尽并不立即引起孢子失活,在供给外源营养之后孢子仍能萌发.将含水量降至4.0±0.9%的冻干粉贮存1年,4℃下活孢率由初始的99.0%下降至90.2%,而20℃下贮存的前165d活孢率下降较为缓慢,但此后急剧下降,至第240d时几乎全部失活.模拟分析表明,低含水量冻干粉在4℃和20℃下贮存的半衰期(即活孢率减少一半所需的时间)分别为1006d和197d.这些结果说明,白僵菌纯孢粉的含水量  相似文献

[背景[白僵菌是一种应用最广泛的虫生真菌,已被用于工厂化大量生产,可防治多种农林害虫。[方法]试验设置17、21、25、29和33℃5个温度水平,采用喷雾法,将浓度为1×105和1×108个·mL-1的白僵菌孢子悬浮液分别感染红火蚁工蚁,以测试不同温度下白僵菌菌株对红火蚁工蚁的致病力。[结果]当浓度为1×108个·mL-1时,在21、25和29℃条件下,红火蚁工蚁在15 d的累计死亡率均达到了100%,在17和33℃下分别为99.36%和98.74%。当浓度为1×105个·mL-1时,在21、25、29和33℃下,红火蚁工蚁在15 d的累计死亡率分别为29.42%、36.18%、33.17%和27.21%,显著高于17℃的累计死亡率。2个浓度处理在17~25℃时,白僵菌对红火蚁工蚁的致死中时(LT50)随着温度的升高而缩短,当温度为29和33℃时,红火蚁的LT50不减少反而增加。25℃时白僵菌Bb04菌株对红火蚁工蚁的LT50最短,死亡速度最快,致病力最强。[结论与意义]该研究可为制定田间利用白僵菌防治红火蚁的最佳时期奠定基础。  相似文献

The entomogenous filamentous fungus, Beauveria bassiana expresses two hydrophobin genes, hyd1 and hyd2, hypothesized to be involved in cell surface hydrophobicity, adhesion, virulence, and to constitute the protective spore coat structure known as the rodlet layer. Targeted gene inactivation of hyd1 resulted in seemingly 'bald' conidia that contained significantly altered surface fascicles or bundles. These cells displayed decreased spore hydrophobicity, loss of water mediated dispersal, changes in surface carbohydrate epitopes and β-1,3-glucan distribution, lowered virulence in insect bioassays, but no effect on adhesion. In contrast, Δhyd2 mutants retained distorted surface bundles, but truncated/incomplete rodlets could be seen within the bundles. Δhyd2 conidia displayed both decreased cell surface hydrophobicity and adhesion, but the mutant was unaffected in virulence. The double Δhyd1Δhyd2 mutant was distinct from the single mutants, lacking both bundles and rodlets, and displaying additively decreased cell surface hydrophobicity, reduced cell attachment and lowered virulence than the Δhyd1 mutant. Epitope tagged constructs of the proteins were used to examine the expression and distribution of the proteins and to demonstrate the continued presence of Hyd2 in the Δhyd1 strain and vice versa. The implications of our results with respect to fascicle and rodlet assembly on the spore surface are discussed.  相似文献

Survival of Botrytis cinerea conidia was studied after storage without pretreatments at different temperatures (-80 degrees C, -20 degrees C, 4 degrees C and 21 degrees C). Germination tests performed during 3 years showed that viability at 21 degrees C was completely lost after 1 month. Conidia stored for 30 months at -80 degrees C, -20 degrees C and 4 degrees C were able to germinate, respectively, at 79%, 8% and 0.2%. Changes in adenylate level, energy charge and respiration (O(2) consumption) made on each set of conidia were correlated to the germination rate. The 30-month-old stored conidia showed differences in pathogenicity tests on apples. While the pathogenic aggressiveness of conidia stored at -80 degrees C was almost the same as for fresh conidia, it decreased with increasing temperature of storage. An ultrastructural study made on conidia stored for 30 months at -80 degrees C has shown the emergence of a new wall layer in a retraction zone of the cytoplasm by comparison to fresh conidia. However, the integrity of the cytoplasmic content was maintained. The effects of low temperature storage, maintenance of cell integrity and pathogenicity of conidia of B. cinerea are discussed.  相似文献

AIM: To evaluate the activity against fungal phytopathogens of two synthetic peptides derived from the protein bovine lactoferricin: the antibacterial active core of six amino acid residues (LfcinB(20-25)) and an extension of 15 amino acids (LfcinB(17-31)). METHODS AND RESULTS: In vitro activity against fungal pathogens was determined and compared with that against model micro-organisms. Activity was demonstrated against fungi of agronomic relevance. Distinct antimicrobial properties in vitro were found for the two peptides. LfcinB(17-31) had growth inhibitory activity higher than LfcinB(20-25). However, LfcinB(17-31) was not fungicidal to quiescent conidia of Penicillium digitatum at the concentrations assayed, while LfcinB(20-25) killed conidia more efficiently. Microscopical observations showed that the mycelium of P. digitatum treated with LfcinB(17-31) developed alterations of growth, sporulation and chitin deposition, and permeation of hyphal cells. In experimental inoculations of mandarins, both peptides showed limited protective effect against the disease caused by P. digitatum. CONCLUSIONS: LfcinB(20-25) and LfcinB(17-31) peptides were shown to have antimicrobial activity against plant pathogenic filamentous fungi, with distinct properties and mode of action. SIGNIFICANCE AND IMPACT OF THE STUDY: LfcinB(20-25) and LfcinB(17-31) peptides offer novel alternatives to develop resistant plants by molecular breeding.  相似文献

Selection of a phage-stable strain of a new species of the rifamycin-producing organism was carried out. The phage-stable mutants were selected with respect to the virulent phage 2739 isolated from a lysogenic culture of the rifamycin-producing organism. Spontaneous phage-stable mutants formed at a rate of 0.8 per cent. Most of them belonged to the morphological colony type with a decreased activity level. No shifts in variation with respect to the property of the antibiotic production were noted under the action of phage 2739. 62 per cent of the phage-stable variants isolated from the secondary growth colonies after infection with the phage were lysogenic and liberated phage 2739 to the culture fluid. Induction of mutations with MNNG, UV and gamma(Co30) rays increased the frequency of the phage-stable mutanta by 1.5 times. Active phage-resistant mutants stable to the phage because of its adsorption and liberating no phage 2739 into liquid media during its cultivation were selected.  相似文献

Investigations of the temperature tolerances of Metarhizium flavoviride conidia, of different moisture contents, in an oil formulation were carried out. The conidia were dried to 5, 9, 15 or 19% moisture contents and exposed to a range of temperatures, -15, 12, 25, 38, 48 and 55 C. Viability declined due to high temperatures and high moisture contents. After 4 months of storage, the conidia were at over 80% viability in all but the 19% moisture content group of the 25 C treatment and all the samples stored at lower temperatures. In a second experiment, conidia dried to 4-5% moisture content showed greater temperature tolerance than conidia with higher moisture contents. Short-term storage under adverse conditions could be feasible with conidia dried to a suitable degree.  相似文献

Over this aw range, glucose and sorbitol had similar effects on recovery, whereas at an aw of 0.82 to 0.92, NaCl had a detrimental effect. Colony diameters were generally largest on media containing sorbitol and smallest on media containing NaCl. Maximum colony size and viable population of heat-stressed conidia were observed on media at an aw of ca. 0.92. When the recovery incubation temperature was 20 degrees C, the number of uninjured conidia detected at an aw of 0.82 was reduced compared with the number detected at 25 degrees C, while at 30 degrees C, the number recovered at an aw of 0.97 was reduced. The effect on heat-stressed conidia was magnified. This suggests that W. sebi conidia may be more tolerant of aw values higher than the optimum 0.92 when the incubation temperature is decreased from the near optimum of 25 degrees C and less tolerant of aw values greater than 0.92 when the incubation temperature is higher than 25 degrees C. The sensitivity of heat-stressed conidia increased as the pH of the recovery medium was decreased from 6.55 to 3.71. The aim of the present investigation was to determine the impact of relative humidity (RH) and temperature on conidial germination, nuclear position and effect of important fungicides on growth and conidial germination of Cercospora canescens. Germination of conidia was observed at RH range 92–100% at 5–35°C. Significant interaction between temperature and RH indicated that higher humidity and high temperature promoted quick germination both in the presence and absence of free moisture. Although in absence of free moisture at 92–95% RH higher temperatures 25–35°C promoted quick evaporation of moisture and no conidial germination. Number of germtube was increased significantly at the optimum temperature 25–30°C and higher humidity (98–100%). But higher temperature 25–35°C with lower RH did not support the conidial germination. This finding is very important for disease forecasting using meteorological data. The spray of Carbendazim as contact fungicide may not be useful since it is not effective against the conidia of C. canescens. Triadimefon did not inhibit the conidia germination but completely inhibited mycelium development at 50 μg/ml. Propriconazole inhibited both conidia germination and mycelial development. Therefore, Propiconazole may be taken as protective as well as curative spray. In non-systemic fungicide, Copper oxychloride gave anticipated result by inhibiting both conidial germination and mycelium development. Therefore, copper oxychloride can be used as protectant fungicides for Cercospora leaf spot caused by C. canescens.  相似文献

Changes in both respiratory pathways and mitochondrial structure of Neurospora crassa occurred under conditions of microcycle conidiation. Upon heat-treatment at 46°C, conidia developed a highly cyanide-insensitive, hydroxamate-sensitive respiration associated with morphological alterations in mitochondrial membranes; such changes were time-dependent. When heat-treated conidia were shifted down to 25°C, the alternate, hydroxamate-sensitive respiration decreased significantly, paralleling the recovery of well-cristated mitochondria with an electron-dense matrix in the germ tubes. The decrease in hydroxamate-sensitivity was associated with two periods of increase in cyanide sensitivity corresponding to the events of germination and precocious proconidial budding.  相似文献

We studied conidiogenesis and adhesive knob formation (maturation) by newly developed conidia of the nematophagous fungusDrechmeria coniospora. Upon conidiogenesis on infected nematodes or during saprophytic growth of the fungus in axenic cultures compact clusters of conidia developed. Less than 10% of such clustered conidia matured; mature conidia were invariably located on the periphery of the clusters.The kinetics and rate of maturation of conidia were studied inin vitro systems and in soil. In both cases adhesive knobs were formed; the rate at which knobs were formed appeared to be determined by the age of the conidia, the temperature and the soil moisture. In addition, knob formation was suppressed at increasing conidial densities. Under favorable conditions, however, over 90% of the conidia matured within a period of 3 days. The rate of knob formation was neither influenced by the presence of nematodes nor by that of exogenous nutrients, which suggests that maturation is an autonomous process. Electron-microscopical analysis indicated that budding of the conidia at the initial stage of maturation occurred simultaneously with the deposition of the sticky, adhesive layer around the wall of the developing knob.The ecological significance of the time- and spatially separated maturation of conidia after conidiogenesis is discussed with respect to survival of the conidia.  相似文献

本文研究碳源、氮源、温度、湿度、pH值和光照等对蝉拟青霉LB菌株生长、产孢和孢子萌发的影响.结果表明,适合该菌株菌落生长和产孢的最佳碳源是可溶性淀粉和蔗糖,最佳氮源为蛋白胨;菌丝生长和孢子萌发的最适温度范围是25℃~27℃,产生分生孢子的最适温度是25℃;分生孢子萌发所需湿度范围是RH 90%~100%,当RH低于90%时很难萌发;在pH值4~10的范围内该菌能生长和产孢,菌丝生长最适pH为6,产生分生孢子和孢子萌发最适pH范围为6-7;光照处理对该菌产孢有一定的影响;分生孢子的致死条件为55℃ 10min.生物学特性显示,蝉拟青霉LB菌株是一株对营养要求不高、对环境适应能力较强的昆虫病原真菌.  相似文献

Variation of the janthinellin-producing organism P. janthinellum was induced by nitrozomethylurea (NMU). The following concentrations of NMU were tested: 0.5; 0.25, 0.125 per cent at the exposure time of 15 minutes, 1 and 10 hours. The conidia survival had back dependence on the concentration and exposure time. The morphological variation was evident from the presence of forms with changed colour of the colony surface mycelium, light green, brown, light yellow, yellow-pink and white conidia and forms differing in the rate of the mycelium development and sporulation. The reverse surface of the colonies of both cultures was light brown to brown because of pigmentation. The feature of pigmentation was stable and no apigmented froms were obtained under the effect of NMU. Formation of variants producing higher yields of janthinellin (400, 2000 units) as compared to the activity of the control variant was noted. Plus variants were detected among the morphologically unchanged cultures.  相似文献

A study was undertaken to evaluate optimum concentrations of chitin in sodium alginate pellet formulations to enhance conidia production. Chitin concentrations tested were 0, 0.5, 1, 2, 3 and 4% (w/v), with (2%, w/v) or without wheat bran. The different chitin-wheat bran pellet combinations were prepared with Beauveria bassiana isolate Qu-B306 at 108 conidia mL-1. After 21 days of incubation in a humid chamber at 28°C, conidia production was assessed. Improvements up to three times the initial conidia number were achieved using 2% chitin and 2% wheat bran. Higher levels of chitin decreased the number of conidia per pellet. For all chitin concentrations, conidia number increased with the addition of wheat bran (P≤0.05). Contamination by saprophytic fungi was reduced by the incorporation of chitin in the pellet formulation.  相似文献

Adult female western flower thrips (Frankliniella occidentalis) were exposed 12-24h to bean (Phaseolus vulgaris) and impatiens (Impatiens wallerana) leaf disks treated with Beauveria bassiana conidia and then transferred to clean bean or impatiens at various times post-treatment. Significantly greater levels of fungal infection were observed when thrips were treated on bean versus impatiens, but exposure to impatiens following treatment had no effect on fungal infection (percent mortality). This result, combined with observations of no inhibition of germination of conidia exposed to intact or macerated impatiens foliage, indicated that the negative effect of the impatiens host plant was not due to plant chemical compounds (antibiosis). Further observations revealed that insects acquired (picked-up) 75% more conidia from treated bean disks than from treated impatiens disks. This difference in dose acquisition was determined to account for the observed difference in percent mortality (15%) following treatment on the two host plants. Median lethal doses (LD(50)) of B. bassiana were not significantly different on the two host plants, but median lethal concentrations were nearly 7-fold greater on impatiens. This difference was explained by disproportionate rates of conidial acquisition at measured rates of conidial deposition (an inverse relationship was observed between application rate expressed as conidia/mm(2) and the number of conidia acquired). The mechanism underlying the differential rates of conidial acquisition from bean versus impatiens was not determined.  相似文献
