The period (per) locus of Drosophila melanogaster has a fundamental role in the expression of biological rhythms. A DNA sequence homologous to a short region of the Drosophila per locus was detected in the chloroplast of Acetabularia mediterranea. A 1175 bp DNA fragment containing the sequence was used as a probe in 'Northern' hybridization experiments. It was found that this DNA was not transcribed or only marginally transcribed in A. mediterranea, at least at the developmental stage just prior to cap formation. It seems that the 1175 bp ctDNA fragment is not involved in the Acetabularia biological rhythm mechanism.  相似文献

Abstract. The level of peroxidase activity utilizing ascorbic acid changes during the development of the green alga, Acetabularia mediterranea. During development almost parallel levels of peroxidase activity and ascorbic acid content are detectable: both steadily decrease as algae progress from very young, slowly growing cells to the rapid growth stage and then to cells exhibiting differentiation into primordium and cap. Changes in the levels of the enzyme and its substrate in the cytoplasm and periplasm were demonstrated using biochemical and cytochemical procedures. Concomitant with these developmental changes, we also observed changes in the stage-specific patterns of ascorbic acid concentration: growing algae exhibit a pronounced negative apicobasal gradient of ascorbic acid. Acetabularia cultivated at 1,200 lux (the normal intensity in a 12-h-light/12-h-dark cycle) and at 700 lux (intensity at which growth is reduced, and cap formation is delayed) were also compared. The higher light intensity induced a moderate decrease in the ascorbic acid content without noticeable changes in the compartmental distribution in the cytoplasm and periplasm, and an increase in the level of periplasmic peroxidase activity with little change in the total peroxidase activity. Catalase was found to be present at very low levels and is unlikely to play a role in H2O2 catabolism. Possible roles for ascorbic acid and peroxidase in the development of Acetabularia are discussed.  相似文献

Polyadenylated RNA was isolated from whole cells and from anucleate cytoplasm of Acetabularia mediterranea. It is synthesised in nucleate but not in anucleate cells. This RNA has a high molecular weight ranging from 0.5 to 3.0 × 106 daltons. In contrast to the RNA of 80 S ribosomes the synthesis of polyadenylated RNA is only slightly enhanced in nucleated cell fragments during the initial phase of regeneration. Newly synthesised polyadenylated RNA migrates from the nucleus through the stalk at about 5 mm a day. The data suggest that polyadenylated RNA migrates independently of 80 S ribosomes.  相似文献

Nuclei from Acetabularia mediterranea were isolated, microinjected with simian virus 40 (SV40) DNA and fused with cytoplasts from the same species. Various times after fusion of the injected nuclei the fusion products were screened for expression of the T-antigen by indirect immunofluorescence. One and two days after injection a bright fluorescence could be observed in the nuclei of Acetabularia. On the basis of this immunofluorescence we conclude that in Acetabularia cells the T-antigen is expressed and accumulated in the nucleus. Moreover, evidence is presented that the Acetabularia cell recognizes the SV40 enhancer sequence. The expression product of the SV40 DNA appears significantly earlier than the expression products of other foreign genes in Acetabularia. The results suggest that the well characterized SV40 can be used as a vector system for the introduction and expression of foreign genes in Acetabularia.  相似文献

The process of induction of polarity in enucleated fragments (EF) of Acetabularia mediterranea has been studied by the method of local illumination. A device is described which allows to illuminate one part of EF at 2, 500 lx and another at 5 lx. The local illumination of a part of EF during 10-24 hrs was shown to polarize with a high probability the subsequent growth and formation of caps at the subsequent uniform illumination. Using the method of successive illumination and darkening of apical and basal portions of EF, it was shown that the initial stages of induction were partially reversible and the complete determination of polarity took place after the initiation of the cap anlage. After the short-term induction by light (2500 lx), the darkening of the growth zone inhibits completely the process of cap initiation, but already at 100 lx the EF previously induced at 2 500 lx are initiated and grow. While using the local illumination of grafts of EF from two species, A. crenulata and A. mediterranea, with their subsequent fragmentation and the analysis of cap morphology, it was shown that the induction of polar growth did not provide the directed transport of the factors controlling morphogenesis to the future growth zone.  相似文献

Zusammenfassung In Acetabularia mediterranea, Acetabularia crenulata und Polyphysa cliftonii wurde das Vorhandensein von Ribonuclease-Aktivität nachgewiesen. Es wurde gezeigt, daß das Enzym hitzestabil ist und bei pH 7,2 und im Bereich von pH 5 Maxima aufweist. Selbst durch eine Zentrifugation nit 105000 x g über 120 min wurde die Enzym-Aktivität nicht sedimentiert.In kernlosen Zellen von Acetabularia nimmt die Ribonuclease-Aktivität zu, allerdings in geringerem Maße als in kernhaltigen Zellen.Die Stabilität der genetischen Information im Cytoplasma von kernloser Acetabularia kann nicht durch das Fehlen von Ribonuclease-Aktivität erklärt werden.Fräulein A. Grohs sei für die geschickte und sorgfältige Hilfe bei der Durchführung der Experimente gedankt.

Summary In Acetabularia mediterranea, Acetabularia crenulata and Polyphysa cliftonii ribonuclease activity was demonstrated. The enzyme is heat stabile and exhibits maxima at pH 7.2 and at the region of pH 5. Even centrifugation with 100000 x g for 120 minutes does not result in sedimentation of enzyme activity.Anucleate cells of Acetabularia are capable of increasing their ribonuclease activity, but to a smaller extent than nucleate cells do.The stability of the genetic information in the cytoplasm of anucleate Acetabularia cannot be explained by a lack in ribonuclease activity.

The 3',5'-adenosine monophosphate (cyclic-AMP) content of the unicellular alga Acetabularia has been examined at various developmental stages. It has been found that very young algae, less than 10 mm in length, have a high cAMP content [more than 7 pmoles per 100 mg wet weight (WW)], but that with the growth of the algae, the cAMP content decreases rapidly, reaching the low level of 0.5–1.0 pmoles per 100 mg WW. The cAMP content remains at this level until cap differentiation, after which an increase in cAMP content accompanies cap enlargement. It has been shown that these results are unlikely to be affected by changes in the cAMP content induced by variations in circadian rhythm.
Treatment with theophylline (2.10−3 M), a phosphodiesterase inhibitor, results in an increase in the cAMP content and delays growth and cap formation. Experiments on the effects of theophylline upon the circadian rhythm of oxygen evolution have shown that the continuous presence of theophylline in the culture medium does not induce a phase shift in the rhythm.
The cAMP content of anucleate Acetabularia shows development stage variations parallel to that of the whole algae.  相似文献

Abstract. Crude enzymatic extracts from Acetabularia exhibit very low peroxidase activity after a lag period. Starch gel electrophoresis of extracts from growing algae shows a single, extremely anodic band. Extracts of small, slow-growing or cap-bearing algae, which do not grow any more, do not exhibit any peroxidase band. Cytochemical staining with benzidine reveals changes in both the quantity and distribution of peroxidase along the polarized Acetabularia cell. The homogenous staining of small algae becomes distributed along a negative apico-basal gradient when the algae initiate their rapid growth phase. This polarized pattern is repeated on the hair whorls. A similar developmental sequence directs cap growth, with an initial intense staining reaction of the primordium, which later leaves only the corona inferior stained blue. Finally, the Acetabularia cell remains slightly blue at the edges of the rhizoidal out-growths and cap rays. Crude extracts of Acetabularia induce a lag in standard horseradish peroxidase (HRP) activity. The inhibitor is always present in small and growing algae; it is sometimes absent or less active in cap-bearing algae. In no case does it change the kinetics of the HRP reaction with guaïacol. The lag is completely suppressed by pretreatment with either H2O2 or ascorbate oxidase. The changes in peroxidase activity, correlated with developmental stage and according to a polarized gradient, suggest that the enzyme could be involved in some way in the control of morphogenesis in Acetabularia . An inhibitor of peroxidase activity, which disappears as the cap matures, might, in turn, exert a regulatory function.  相似文献

The c-AMP content has been found to double when Acetabularia develop from 5–10 mm long to grown or almost full-grown algae.
The biological significance of this fact has been approached by studying the effects of drugs known to influence the intracellular c-AMP content on the development of Acetabularia. When grown in the presence of theophyllin or papaverin, inhibitors of phosphodiesterase, the Acetabularia display a striking response during the exponential growth period; the final length, however, is not affected. Both substances increase the c-AMP content of the algea. Isoproterenol, which activates adenylate cyclase in many systems, also influences Acetabularia during the exponential growth period and, in addition, slightly affects cap formation.
The change in c-AMP content in the course of development and the effects of drugs influencing (theophyllin and papaverin) or likely to influence (isoproterenol) the c-AMP content of the algae suggest that this nucleotide plays a role at the time of intense growth.
The same phosphodiesterase activity has been found in the 5–10 mm and the 19–25 mm long algae, whereas two enzymes were found in cap-bearing Acetabularia.
The results are discussed as well as the involvement of c-AMP in the development of this alga.  相似文献

Formation of three germ layers is the most important event in early vertebrate development. Animal cap assays can be used to reproduce the in vivo induction of amphibian tissues in order to investigate the differentiation processes that occur in normal embryonic development. Activin treatment strongly and dose-dependently induces various types of mesodermal and endodermal tissue in cultured animal caps. Beating heart, pronephros, pancreas and cartilage can be induced by microsurgical manipulation and simultaneous treatment with activin and other factors. These in vitro induction systems will be helpful for elucidating the mechanisms of tissue induction and organ formation in vertebrate development.  相似文献

The isozymes of malate dehydrogenase (MDH) were studied by means of electrophoresis in polyacrilamide gel in Acetabularia crenulata and A. mediterranea. The isozyme profile of MDH was shown to be variable in different parts of the plant. Distinct differences in isozyme profiles of MDH between A. crenulata and A. mediteranea were found when studying the cell fractions which consisted mainly of chloroplasts. The chloroplast fraction of A. mediterranea contained 8 isozymes which form 2 groups with different electrophoretic mobility. The chloroplast fraction of A. crenulata contained 9 isozymes. All the isozymes of the first group were common for both the species under study.  相似文献

Summary Rifampicin (10g/ml) strongly inhibits the incorporation of [5-3H]-uridine into the chloroplast RNA of anucleate cells of the green alga Acetabularia mediterranea, whereas incorporation into nuclear RNA is hardly affected.Furthermore, at a concentration range of 1–10g/ml rifampicin has only a small effect on stalk- and cap formation in nucleate posterior parts of the stalk. As has already been shown, the morphogenesis of such cell segments depends on the synthesis of new RNA in the nucleus. Similarly rifampicin only slightly inhibits the synthesis of the enzyme UDPG-pyrophosphorylase, which is coded by nuclear DNA.These slight inhibitions are interpreted as secondary effects arising from a blockage of plastid RNA synthesis, since both nucleate and anucleate cells respond in a similar manner and to the same degree.In contrast the increase in the chlorophyll content in nucleate and anucleate cells is severely impaired by the antibiotic. These findings indicate that the nucleus and the plastids contain different DNA-dependent RNA-polymerases.  相似文献

In addition to the endogenous production of reactive oxygen species (ROS) as a result of normal development, amphibian external development often forces embryos to deal with oxidative stress-producing agents present in the environment. Embryos should therefore develop protective systems to reduce ROS toxicity and achieve successful development. The present work was aimed to characterize the effects produced by the widespread-used ROS-generator pesticide Paraquat during early embryonic development in the toad Chaunus arenarum, as well as to get insights into the defense response elicited by amphibian embryos. The approach consisted in generating a sharp and brief oxidative stress condition early during embryonic development to stimulate the cellular mechanisms involved in ROS-antioxidant response. Results revealed that Paraquat-treatment reduced the ability of embryos to develop normally, leading to arrests of development and severe malformations such as tail abnormalities, abdominal edema, reduced head development and curved dorsal structures. Although Paraquat effects were morphologically evident from gastrula stage on, alterations such as chromatin condensation were observed even at blastula stage by histological examinations. Regarding detoxifying enzymes, a significant induction of Mn-superoxide dismutase activity was detected at stages beyond gastrula in embryos surviving Paraquat treatment, suggesting a major role of this enzyme in the antioxidant response during early embryonic development.  相似文献

A late stage during the biological cycle of the unicellular alga Acetabularia is the differentiation of a cap at the apical end of the stalk. A minimal model of the spatio-temporal regulation of this event is proposed on the basis of biological data available and current hypotheses. This involves the interaction between a diffusing inhibitor specific to the translation of cap mRNAs and a graded distribution of these messengers. The model accounts for delayed protein synthesis which occurs preferably at the apex and is likely to initiate the formation of the cap. Accumulation of peroxidase was demonstrated by light and electron microscopy to occur in Acetabularia in certain regions of the cap rays in relation to the development of the gametangia (cysts). Peroxidase was found to be incorporated into special, cell wall-like obstructions that separate the cap rays from the stalk when the secondary nuclei have settled in the cap rays. It is assumed that peroxidase acts as an anti-microbial protectant of the gametangia.  相似文献

Many amphibians are known to suffer embryonic die-offs as a consequence of Saprolegnia infections; however, little is known about the action mechanisms of Saprolegnia and the host-pathogen relationships. In this study, we have isolated and characterized the species of Saprolegnia responsible for infections of embryos of natterjack toad (Bufo calamita) and Western spadefoot toad (Pelobates cultripes) in mountainous areas of Central Spain. We also assessed the influence of the developmental stage within the embryonic period on the susceptibility to the Saprolegnia species identified. Only one strain of Saprolegnia was isolated from B. calamita and identified as S. diclina. For P. cultripes, both S. diclina and S. ferax were identified. Healthy embryos of both amphibian species suffered increased mortality rates when exposed to the Saprolegnia strains isolated from individuals of the same population. Embryonic developmental stage was crucial in determining the sensitivity of embryos to Saprolegnia infection. The mortalities of P. cultripes and B. calamita embryos exposed at Gosner stages 15 (rotation) and 19 (heart beating) were almost total 72 h after challenge with Saprolegnia, while those exposed at stage 12 (late gastrula) showed no significant effects at that time. This is the first study to demonstrate the role of embryonic development on the sensitivity of amphibians to Saprolegnia.  相似文献

The activity of ATP: polyphosphate phosphotransferase was detected in free-cellular extracts of Acetabularia mediterranea. The enzyme activity in cells originally deficient in phosphorus and subsequently transferred into the phosphate-containing medium increases 5-10-fold as compared to normal. Polyphosphate degradation in A. mediterranea is probably produced by polyphosphatase, which was also detected in the free-cellular extract. It was shown that the polyphosphatase activity has two pH optima, i.e. 4.5 and 7.5, and is considerably increased when the cells are transferred into the phosphate-free medium. It is assumed that high-molecular polyphosphates involved in A. Mediterranea metabolism are responsible for regulation of orthophosphate and ATP level in the cells by ATP: polyphosphate phosphotransferase and polyphosphatase.  相似文献

Cap formation, a major developmental process in the alga Acetabularia, is influenced by a single perturbation of the entraining light-dark schedule and thus, presumably, of the circadian rhythms. This perturbation is brought about several weeks before cap formation, the most conspicuous expression of morphogenesis in Acetabularia. The effect is more pronounced on cap formation than on growth. It varies in importance with the circadian time at which the perturbation was brought about. The effect is dependent on the developmental state of the alga: transfer carried out during the logarithmic phase of growth produces a delay whose importance decreases with time. When carried out during the phase of slow terminal growth, the transfer induces a transitory acceleration of cap formation. When the algae approach their final length, no effect is elicited. Photoperiodism seems to be involved.  相似文献

We investigated the effects of temperature on the growth and development of embryonic and early larval stages of a western North American amphibian, the rough-skinned newt (Taricha granulosa). We assigned newt eggs to different temperatures (7, 14, or 21 °C); after hatching, we re-assigned the newt larvae into the three different temperatures. Over the course of three to four weeks, we measured total length and developmental stage of the larvae. Our results indicated a strong positive relationship over time between temperature and both length and developmental stage. Importantly, individuals assigned to cooler embryonic temperatures did not achieve the larval sizes of individuals from the warmer embryonic treatments, regardless of larval temperature. Our investigation of growth and development at different temperatures demonstrates carry-over effects and provides a more comprehensive understanding of how organisms respond to temperature changes during early development.  相似文献
