Helicoverpa armigera has been controlled effectively with chemical insecticides in the major cotton crop production areas of northern Greece for many years. However, a resurgence of the pest was observed in 2010, which significantly affected crop production. During a 4‐year survey (2007 – 2010), we examined the insecticide resistance status of H. armigera populations from two major and representative cotton production areas in northern Greece against seven insecticides (chlorpyrifos, diazinon, methomyl, alpha‐cypermethrin, cypermethrin, gamma‐cyhalothrin and endosulfan). Full dose‐response bioassays on third instar larvae were performed by topical application. Lethal doses at 50% were estimated by probit analysis and resistance factors (RF) were calculated, compared to a susceptible laboratory reference strain. Resistance levels were relatively moderate until 2009, with resistance ratios below 10‐fold for organophosphates and carbamates and up to 16‐fold for the pyrethroid alpha‐cypermethrin. However, resistance rose to 46‐ and 81‐fold for chlorpyrifos and alpha‐cypermethrin, respectively in 2010, when the resurgence of the pest was observed. None of the known pyrethroid resistance mutations were found in the pyrethroid‐resistant insects. The possible association between resistance and H. armigera resurgence in Greece is discussed.  相似文献

The quick detection of domestic and peridomestic triatomines in their environments becomes difficult without the use of dislodgement substances that flush them out from their shelters. At present, tetramethrin 0.2% is being widely used in control programs. Although it is an efficient dislodging agent, its toxicity might affect the health of captured triatomines, of other insects and, to a lesser extent, of other animals, including humans. Here, we tested if semiochemicals released by disturbed adults of Triatoma infestans and/or Rhodnius prolixus can make larvae of the same species exit from their refuges. In a walking olfactometer we found that: 1) larvae of T. infestans were repelled by the odors released by disturbed adults of their own species and of R. prolixus, 2) larvae of R. prolixus did not change their behavior in the presence of odors released by adults of both species, and 3) activity levels were not modulated by these odors in any of both species. Besides, in pseudo‐natural conditions we found an increased flushing‐out activity of larvae of T. infestans when their shelters were sprayed with isobutyric acid or 3‐pentanol, and of larvae of R. prolixus when sprayed with 3‐methyl1butanol. We succeeded in this work to dislodge larvae of triatomines from artificial shelters using natural volatile compounds, allowing the capture of live bugs for further investigations (e.g., xenodiagnosis or genetic studies) and favoring ecological aspects (e.g., minimizing environmental insecticide‐contamination and non‐targeted mortality).  相似文献

Thirty‐two Aedes aegypti populations collected throughout Thailand and five populations of Aedes albopictus from southern Thailand were subjected to standard WHO contact bioassays to assess susceptibility to three commonly used synthetic pyrethroids: permethrin, deltamethrin, and lambda‐cyhalothrin. A wide degree of physiological response to permethrin was detected in Ae. aegypti, ranging from 56.5% survival (Lampang, northern Thailand) to only 4% (Kalasin in northeastern and Phuket in southern Thailand). All 32 populations of Ae. aegypti were found to have evidence of incipient resistance (62.5%) or levels of survival deemed resistant (37.5%) to permethrin. Four populations of Ae. albopictus were found with incipient resistance (97 – 80% mortality) and one with resistance (< 80%) to permethrin. The majority of Ae. aegypti populations (68.7%) was susceptible (> 98% mortality) to deltamethrin, with incipient resistance (observed 97–82% mortality) in other localities. In contrast, all populations of Ae. aegypti were completely susceptible (100% mortality) to the recommended operational dosage of lambda‐cyhalothrin. All five populations of Ae. albopictus were found completely susceptible to both deltamethrin and lambda‐cyhalothrin. Evidence of defined incipient or resistance to synthetic pyrethroids mandates appropriate response and countermeasures to mitigate further development and spread of resistance. In light of these findings, we conclude that routine and comprehensive susceptibility monitoring of dengue mosquito vectors to synthetic pyrethroids should be a required component of resistance management policies and disease control activities.  相似文献

We have found the following differences in the male meiosis among three triatomine species: (1) The three largest autosomal bivalents ofTriatoma infestans are heterochromatic.Rhodnius prolixus has two autosomal bivalents with heterochromatic blocks.Triatoma rubrovaria does not show any heteropycnotic autosomes. (2) Sex chromosomes inT. infestans form a chromocenter. At early prophase terminal associations are seen between sex chromosomes inT. rubrovaria, and they maintain a close association until diakinesis. An intimate association between the X and Y chromosomes is observed during early prophase inR. prolixus, but a distant association is maintained by the sex chromosomes at diffuse and diplotene stages in this species. (3) Polyploid nuclei of the nutritive cells are quite distinct. Numerous chromocenters of different shapes and sized are seen in those ofT. infestans. InT. rubrovaria one chromocenter having two positively heteropycnotic elements is observed surrounded by homogeneous chromatin. Only one compact chromocenter is found amongst unevenly distributed chromatin, inR. prolixus.  相似文献

Randomness of mating between susceptible and resistant individuals is a major factor that closely relates to the refuge strategy of resistance management for Helicoverpa armigera (Hübner) to Bacillus thuringiensis cotton. The mating behaviour of Cry1Ac‐susceptible and Cry1Ac‐resistant strains of H. armigera was compared to investigate the randomness of their mating. The percentage of mating was lower for Cry1Ac‐resistant H. armigera compared with that of the susceptible strain under both no‐choice and multiple‐choice conditions. The low percentage of mating in the resistant strain indicates a reduced incidence of successful mating. The percentage of spermatophore‐containing mated female H. armigera in the crossing of susceptible females × resistant males was significantly lower than in the crossing of resistant females × susceptible males, but the observed mating frequencies of these two types of cross were similar to each other. This indicates that resistant males reduce the incidence of mating paternity more than they do their mating frequency. The percentages of heterogametic matings (susceptible females × resistant males, resistant females × susceptible males) in the multiple‐choice experiment were lower than those of homogametic matings (susceptible × susceptible, resistant × resistant) on peak mating nights. However, the difference between heterogametic and homogametic mating was not significant, indicating that there was a random mating between susceptible and resistant strains. The results presented here do not reflect reality in mating associated with Cry1Ac resistance but can provide insight into variable expression.  相似文献

Dialelic crosses and backcrosses of pyrethroid resistant (RR) and susceptible (SS) Rhipicephalus (Boophilus) microplus tick strains were carried out and the substitution (Phe-Ile) within the sodium channel gene was monitored in order to analyze the effects of the genotype on the pyrethroid resistance phenotype as measured by the larval packet test (LPT). Parental strains: susceptible (SS) and resistant (RR); dialelic crosses: RS (♂RR × ♀SS), and SR (♂SS × ♀RR); and backcrosses: RS × SS, RS × RR, SR × SS and SR × RR were infested on 280 kg calves. Resistance type (monogenic or polygenic) and effective dominance were determined based on the discriminant concentration (DC) for cipermethrine (0.5%), deltamethrine (0.09%) and flumethrine (0.01%). Allele specific PCR (AS-PCR) was used for genotyping, looking at a sodium channel mutation (Phe-Ile substitution). The mortality rates and allele frequency of susceptible and pyrethroid resistant reference strains were 0% mortality and 90% RR alleles for resistant strain, and 100% mortality and 0% RR alleles as measured by the larval packet test (LPT) and allele specific PCR (AS-PCR) respectively. Backcrossed strain SR × RR showed an effective dominance (DML) of 0.605 for cypermethrin, 0.639 for deltamethrin and 0.498 for flumethrin, while survival of backcrosses RS × SS, RS × RR and SR × SS showed a significant tendency to recesivity. Backcrossed strain SR × RR (69.4%) also showed a higher RR genotype frequency with regards to RS × SS (25.5%), RS × RR (36.7%) and SR × SS (32.0%), however, susceptible allele was inherited in general as an incomplete dominant trait. Monogenic inheritance hypothesis was tested and the results showed monogenic inheritance for cypermethrin and flumethrin (P < 0.05) but not for deltamethrin (P > 0.05). However, significant correlation was found between RR genotype and the survival rate for all three pyrethroids used (P < 0.05), suggesting that a single substitution on the sodium channel gene can be responsible for resistance to pyrethroids as a class, due to the high frequency for RR genotypes. Combination with different mutations or metabolic resistance mechanisms cannot be excluded.  相似文献

Understanding the relationship between geographic range limits and physiological traits of vector species is under increasing demand to predict the potential effects of global warming, not only in terms of geographic distribution of vector species but also in terms of the risk of disease transmission. Like in many other insect species, the geographical distribution of Chagas’ disease vectors is affected by temperature. This study examines, for the first time, the relationship between the limits of geographic distribution and thermo‐tolerance of the most important vectors of Chagas disease, Triatoma infestans in southern South America and Rhodnius prolixus in northern South America and Central America, to test the climatic variability hypothesis (CVH). We applied species distribution modeling (SDM) using bioclimatic variables and identified the most important limiting factors of the habitat suitability. Then, we measured and compared: the critical thermal maximum (CTmax) and the upper lethal temperature (ULT) (measured by thermo‐limit respirometry), chilled coma recovery (i.e. the time to recovery from 4 h at 0°C) and the critical thermal minimum (CTmin). For both species the minimum temperature of the coldest month was the most important abiotic factor restricting their geographic distribution. By taking a correlative approach and testing predictions with thermal tolerance traits, it was possible to explain the southern limit distribution for both species in terms of physiological constraints. The greater temperature tolerance of T. infestans compared to R. prolixus supports the CVH.  相似文献

Pyrethroid resistance has been detected in Triatoma infestans (Klug) (Hemiptera: Reduviidae) specimens from different areas of Argentina and Bolivia. Genes conferring resistance can have a pleiotropic effect with epidemiological and evolutionary consequences. This research studied excretion/defecation patterns in deltamethrin‐resistant T. infestans in order to elucidate its biological performance, adaptive consequences and role in the transmission of Chagas' disease. One deltamethrin‐susceptible strain and two deltamethrin‐resistant strains were used. Fifth‐instar nymphs were fed ad libitum and their defecations recorded during and after the first or second feeding in the stadium. Resistant insects began to defecate later, defecated less, showed a lower proportion of defecating individuals and lower defecation indices compared with susceptible insects during the first hour after feeding. The number of bloodmeals in the stadium did not affect the main variables determining the pattern of defecation. The present study suggests that alterations in the excretion/defecation pattern in resistant insects entail an adaptive cost and, considering only this pattern, determine a lower capacity for transmission of Trypanosoma cruzi (Kinetoplastida: Trypanosomatidae) compared with susceptible insects.  相似文献

Triatomines (Hemiptera: Reduviidae: Triatominae) are nocturnal blood‐sucking insects. During daylight hours they remain in an akinetic state inside their shelters, whereas at dusk they become active and move outside. When they are outside their shelters during the photophase, triatomines are vulnerable to diurnal predators and the period just before dawn is critical to their survival. This work analyses the existence of competitive interactions involved in the occupancy of shelters by triatomines. Behavioural assays were performed in which nymphs of different stages, nutritional status or species were released in an experimental arena containing a space‐limited artificial shelter. The proportions of individuals occupying the shelter during the photophase were quantified to estimate the competitive abilities of each stage and species. Intraspecific comparisons showed higher levels of shelter occupancy for fourth over fifth instars and fed over unfed nymphs of Triatoma infestans. Interspecific comparisons showed higher rates of shelter occupancy for Triatoma sordida in comparison with T. infestans, and for T. infestans over Rhodnius prolixus. Arrival order was also relevant to determining shelter occupancy levels: early arrival was advantageous in comparison with later arrival. The study of intra‐ and interspecific competitive interactions for shelter occupancy provides relevant information about colonization and recolonization processes in the natural environments of triatomines.  相似文献

Euschistus heros (Fabricius, 1798) is one of the most harmful insect pests damaging Brazilian soybean crops and has become a major problem due to its high population density and resistance to insecticides. Currently, there are no data on whether alterations of testicular morphology and chromosomal behavior are associated with the resistance mechanisms related to the action of insecticides. This study integrated analyses of the testicular morphology, meiocyte cell division, and chromosomal structure and behavior in the process of spermatogenesis in E. heros. We compared these features among wild‐caught individuals, insecticide‐susceptible and ‐resistant strains. The resistant strain was established through a selection experiment exposing the bugs to insecticides (thiamethoxam + lambda‐cyhalothrin) for 15 generations. No differences were detected in the examined features among the three groups of experimental individuals: the testis comprised six lobes, with the fifth lobe thinner than the others; the karyotype was 2n = 14 (12 + XY), with no evident changes in chromosomal breakage, rearrangement or behavior in the meiosis; and abundant spermatozoa were observed in all testicular lobes. Thus, any effects of the long‐term (15 generations) experimental selection by exposure to the insecticides were not detected on the male germinal tissue and chromosomes, suggesting the irrelevancy of the examined features to insecticide‐resistance mechanisms in E. heros.  相似文献

Pollen beetle, Meligethes aeneus (Fabricius) (Coleoptera: Nitidulidae), and cabbage seed weevil, Ceutorhynchus assimilis (Paykull) (Coleoptera: Curculionidae), are important pests in the production of European winter oilseed rape, Brassica napus L. (Brassicaceae), which is grown on several million hectares in Europe. Insecticide treatments are common to control both pests once they exceed economic damage thresholds; however, not many chemical classes are available for their control in European oilseed rape. Particularly pollen beetles recently developed high levels of pyrethroid resistance impairing field control at recommended rates in many countries, whereas no resistance is yet reported to another important insecticide, thiacloprid. The major objective of this study was to investigate the spatio‐temporal susceptibility status of pollen beetle against the recently introduced insecticide thiacloprid. From 2009 to 2012 more than 630 populations of pollen beetle collected in 13 countries were monitored for resistance to thiacloprid by using an adult vial test. No shifting to lower susceptibility of pollen beetle to thiacloprid has been observed between 2009 and 2012. Furthermore, we were able to show that pollen beetle larvae are extremely susceptible to thiacloprid, whereas within strains larvae are significantly more resistant than adults to pyrethroids such as lambda‐cyhalothrin. Dose‐response data for thiacloprid against cabbage seed weevil populations collected in 2011 in Germany, Sweden, and Ukraine showed a 10‐fold higher intrinsic sensitivity compared to pollen beetle, and showed only a low variation in response. In addition, we also tested 17 cabbage seed weevil populations collected in five countries against lambda‐cyhalothrin with low variation in response (three‐fold), suggesting full baseline susceptibility and no resistance to pyrethroids. The implications of the data presented for resistance management in coleopteran pests in winter oilseed rape are discussed.  相似文献

Insecticides cause a range of sub‐lethal effects on targeted insects, which are frequently detrimental to them. However, targeted insects are able to cope with insecticides within sub‐lethal ranges, which vary with their susceptibility. Here we assessed the response of three strains of the maize weevil Sitophilus zeamais Motschulsky (Coleoptera: Curculionidae) to sub‐lethal exposure to the pyrethoid insecticide cypermethrin. We expected enzyme induction associated with cypermethrin resistance since it would aid the resistant insects in surviving such exposure. Lower respiration rate and lower activity were also expected in insecticide‐resistant insects since these traits are also likely to favor survivorship under insecticide exposure. Curiously though, cypermethrin did not affect activity of digestive and energy metabolism enzymes, and even reduced the activity of some enzymes (particularly for cellulase and cysteine‐proteinase activity in this case). There was strain variation in response, which may be (partially) related to insecticide resistance in some strains. Sub‐lethal exposure to cypermethrin depressed proteolytic and mainly cellulolytic activity in the exposed insects, which is likely to impair their fitness. However, such exposure did not affect respiration rate and walking behavior of the insects (except for the susceptible strain where walking activity was reduced). Walking activity varies with strain and may minimize insecticide exposure, which should be a concern, particularly if associated with (physiological) insecticide resistance.  相似文献

Extensive use of insecticides on cotton has prompted resistance development in the cotton aphid, Aphis gossypii (Glover) in China. A deltamethrin‐selected population of cotton aphids from Xinjiang Uygur Autonomous Region, China with 228.59‐fold higher resistance to deltamethrin was used to examine how carboxylesterase conferred resistance to this pyrethroid insecticide. The carboxylesterase activity in the deltamethrin‐resistant strain was 3.67‐, 2.02‐ and 1.16‐fold of the susceptible strain when using α‐naphthyl acetate (α‐NA), β‐naphthyl acetate (β‐NA) and α‐naphthyl butyrate (α‐NB) as substrates, respectively. Carboxylesterase cDNA was cloned and sequenced from both deltamethrin‐resistant and susceptible strains. The cDNA contained 1581 bp open reading frames (ORFs) coding a 526 amino acid protein. Only one amino acid substitution (Val87‐Ala) was observed between deltamethrin‐resistant and susceptible strains but it is not genetically linked to resistance by the catalytic triad and signature motif analysis. The real‐time polymerase chain reaction analysis indicated that the resistant strain had a 6.61‐fold higher level of carboxylesterase mRNA than the susceptible strain. The results revealed that up‐regulation of the carboxylesterase gene, not modified gene structure, may be responsible for the development of resistance in cotton aphids to deltamethrin.  相似文献

Colletotrichum gloeosporioides causes anthracnose, the most severe foliar disease of field-grown water yam (Dioscorea alata). The inheritance of resistance to a moderately virulent (FGS) strain of the pathogen was investigated in crosses between tetraploid D. alata genotypes: TDa 95/00328 (resistant)×TDa 95–310 (susceptible) (cross A), and TDa 85/00257 (resistant)×TDa 92–2 (susceptible) (cross B). Segregation of F1 progeny fitted genetic ratios of 3:1, 5:1 (crosses A and B) and 7:1 (cross A) resistant:susceptible when inoculated with the FGS strain, indicating that resistance is dominantly inherited and suggesting that more than one gene controls the inheritance of resistance to this strain in the accessions studied. When parental and progeny lines of cross A were inoculated with an aggressive (SGG) strain of the pathogen, all plants expressed a susceptible phenotype, indicating strain-specific resistance in TDa 95/00328. Screening of 20 cultivars/landraces confirmed the high susceptibility of D. alata accessions to the SGG strain and revealed the presence of apparent strain non-specific resistance in TDa 85/00257. TDa 85/00257 and TDa 87/01091 which were resistant to the SGG strain, will be useful both as sources of resistance and in the development of a host differential series for D. alata. Among peridomestic structures, chicken coops are sites of major importance for the domestic ecology of Triatoma infestans (Hemiptera: Reduviidae). The aim of this study was to evaluate in an experimental context the effects of a cypermethrin pour‐on formulation applied to chickens on blood intake, moulting and mortality in T. infestans, under the natural climatic conditions of a region endemic for Chagas' disease. Experimental chicken huts were made of bricks and covered with plastic mosquito nets. Ninety fourth‐instar nymphs were maintained in each hut. The study used a completely random design in which chickens in the experimental group were treated with a cypermethrin pour‐on formulation. Five replicates (= huts) of the experimental and control groups were conducted. The number of live T. infestans, blood intake and moults to fifth‐instar stage were recorded at 1, 5, 20, 35 and 45 days after the application of cypermethrin. Cumulative mortality was higher in nymphs exposed to treated chickens (> 71%) than in control nymphs (< 50%) (P < 0.01). Blood intake and moulting rate were lower in nymphs fed on treated chickens than in control nymphs (P < 0.05). Pour‐on cypermethrin was able to cause significant mortality, although it did not eliminate the experimental population of T. infestans.  相似文献

Responses to artificial selection on body mass in the maize weevil Sitophilus zeamais (Coleoptera: Curculionidae) were investigated to determine whether changes in body mass are associated with insecticide susceptibility, rate of population growth, and metabolic rate. Two strains of the maize weevil differing in susceptibility to pyrethroid insecticides were subjected to bidirectional selection on body mass. The susceptible strain responded to selection resulting in individuals with lower or higher body mass, but the resistant strain responded significantly only to selection for lower body mass. The resistant strain selected for low body mass increased its level of deltamethrin resistance in 44 × . In contrast, selection for low body mass in the susceptible parental strain led to increased deltamethrin susceptibility (50 × ) and selection for high body mass increased deltamethrin resistance (4 × ). Thus, the correlated response of insecticide resistance to selection for body mass differed between strains, a likely consequence of their distinct genetic background. Regardless, body mass was positively correlated with fitness (reproductive output) (r = 0.79; P < 0.001), while such correlation with respiration rate was significant only at P = 0.07 (r = 0.44). Therefore, the association between body mass and deltamethrin resistance is population‐dependent in the maize weevil, and the confluence of deltamethrin resistance and high body mass in a given strain will likely favour its energy metabolism and lead to the mitigation of fitness costs usually associated with insecticide resistance. The genetic background and selection history of insecticide resistant populations should not be neglected since they may favour the confluence of insecticide resistance with mitigation mechanisms of its associated fitness costs limiting the tactics available to their management.  相似文献

Assays on eggs of Panonychus ulmi held on leaf discs of Myrobalan plum were used to characterise strains (1) multi-resistant to organophosphates (OPs) and binapacryl (strain BR), (2) multi-resistant to OPs and tetradifon (strain TR), and (3) susceptible to OPs, binapacryl and tetradifon (strain EM). By crosses of EM females and BR males, and of EM females and TR males, and by repeated backcrossing with EM males under selection pressure of the acaricides concerned, two strains were produced in which the specific genes for resistance to binapacryl and tetradifon were isolated in the genome of the EM strain. Reciprocal crosses of the resulting backcrossed strains with EM, assays on the F1 eggs and the F2 haploid eggs, confirmed the presence of single major genes for resistance to binapacryl and tetradifon, and the absence of maternal effects. The expression of gene BR was incompletely dominant in response to binapacryl, whilst gene TR was effectively dominant in response to tetradifon. Eggs of the field-derived strain BR showed c. 50× resistance to binapacryl, 8–19 × to dinobuton, but only c. 2× to dinocap and dinoseb. The backcrossed strain was less resistant (26 ×) to binapacryl implying that resistance in strain BR was augmented by the effects of minor genes. Resistance to tetradifon (150–200 × on eggs) involved cross-resistance to tetrasul (23 ×) and to chlorfensulphide (27×); resistance in the field-derived strain appeared to depend on the major gene alone. Resistance mechanisms were highly specific in their effects, so that there was no cross-resistance to unrelated acaricides, including amitraz, dicofol, cyhexatin, formetanate, chlordimeform, and quinomethionate. Fecundity of strain BR was 44% less than that of EM; laboratory and field evidence suggest that multi-resistant mites tend to be less fit than susceptible mites. Triatomines display most of their activities during the night. Before sunrise, they search, select and occupy adequate shelters to stay during the photophase, avoiding exposure to diurnal predators. In this work, we first explored the interactions between individuals of the same or different species in the shelter selection process of Triatoma infestans (Klug 1834) and Rhodnius prolixus (Stål 1859). When two groups of insects (either of different nutritional status, nymphal instar or species) were released together over an experimental arena containing two identical shelters, all nymphs were distributed randomly, suggesting the absence of intra- and inters-pecific interactions. Secondly, we analysed their preferences for particular features of shelters by releasing one group of insects (either T. infestans or R. prolixus) over an arena containing two different refuges. Nymphs exhibited preferences for darker shelters with a vertical orientation of its substrate and elevated from the ground, highlighting the importance of such features in a shelter selection context. We conclude that these species disregard the presence of other individuals but evaluate certain characteristics of the shelters to choose them. This information may contribute to understanding the colonization/recolonization dynamic processes of these Chagas disease vectors.  相似文献

Jaburetox is a recombinant peptide derived from one of the Canavalia ensiformis urease isoforms. This peptide induces several toxic effects on insects of different orders, including interference on muscle contractility in cockroaches, modulation of UDP-N-acetylglucosamine pyrophosphorylase (UAP) and nitric oxide synthase (NOS) activities in the central nervous system of triatomines, as well as activation of the immune system in Rhodnius prolixus. When injected, the peptide is lethal for R. prolixus and Triatoma infestans. Here, we evaluated Jaburetox toxicity to Nauphoeta cinerea cockroaches, exploring the effects on the central nervous system through the activities of UAP, NOS, acid phosphatases (ACP), and acetylcholinesterase (AChE). The results indicated that N. cinerea is not susceptible to the lethal effect of the peptide. Moreover, both in vivo and in vitro treatments with Jaburetox inhibited NOS activity, without modifying the protein levels. No alterations on ACP activity were observed. In addition, the enzyme activity of UAP only had its activity affected at 18 hr after injection. The peptide increased the AChE activity, suggesting a mechanism involved in overcoming the toxic effects. In conclusion, our findings indicate that Jaburetox affects the nitrinergic signaling as well as the AChE and UAP activities and establishes N. cinerea as a Jaburetox-resistant model for future comparative studies.  相似文献
